ST-segment changes in the ECG, which are an indication of acute myocardial ischaemia, are obtained when a small quantity of an extract from dried Pachystigma pygmaeum is injected intravenously in sheep. When the fraction was injected subcutaneously, animals reached a crisis after about 5 h, with low values of stroke volumes and high values for pulmonary arterial pressures, pulmonary vascular resistances and heart rates. The haemodynamic changes are an indication of the development of pump failure of the heart. In sheep, injected subcutaneously with the toxic fraction, as well as for sheep dosed with plant material through rumen fistula, increased serum levels for thromboxane and increased or decreased levels for prostacycline were observed. The experimental results are interpreted as being an indication that these prostaglandines may be involved in the development of gousiekte by impeding cardiopulmonary function as a result of coronary and pulmonary vasoconstriction. The sudden death observed in some gousiekte sheep may be due to myocardial ischaemia and associated arrhythmias.  相似文献

Twenty sheep were dosed with either Pachystigma pygmaeum or Fadogia homblei belonging to the Rubiaceae. The experimentally-induced cardiotoxicoses were monitored by various clinical pathological parameters and heart function tests. Elevated AST (aspartate transaminase) activity in the serum proved to be a more reliable indicator of cardiac damage in gousiekte than either LD (lactate dehydrogenase) or CK (creatine kinase). Persistent increases of AST activity were recorded from c. 14 days after commencement of dosing, and this activity sometimes peaked as late as 30 days after the dosing had ceased. Tachycardia and diminished heart function were registered only terminally. Lesions of gousiekte were present in all the sheep that were exposed to the plants. In a field outbreak of P. pygmaeum, where 60 out of 90 sheep died, 14 out of the 15 animals examined had increased AST levels compared with none of the 15 controls. These results indicated that increased enzyme levels can be of use to identify affected animals during latency in a natural outbreak of gousiekte.  相似文献

Lesions in 4 field cases (3 sheep and 1 goat) of 'waterpens' or water belly, caused by the plant Galenia africana, are described. The clinical pathological and pathological findings in 7 sheep which were drenched with toxic plant material are also reported. Inappetence, ruminal stasis and apathy as well as tachycardia were noticed in some of the sheep towards the end of the dosing period. The most prominent clinical pathological change in the experimental animals was an increase in the activity of gamma-glutamyltransferase which in some animals occurred within days after commencement of dosing. This indicates liver involvement in the early stages of the intoxication, and at this stage no heart abnormalities were detected clinically, clinical pathologically or with cardiac function tests. Decrease in cardiac function were recorded in 2 sheep towards the end of the dosing period. Liver and heart lesions were present in all the animals. In some cases hepatic changes were mild and characterized by dilation of central veins and sinusoids and, less commonly, centrilobular fibrosis. More advanced lesions included centrilobular fibrosis and bridging between neighbouring lobules with adjacent areas of coagulative necrosis, lysis and ballooning degeneration of hepatocytes. Myocardial changes occurred in the free ventricular walls and interventricular septum and comprised hypertrophy of myocytes with consequent degeneration and necrosis and fibrosis. In cases of longer duration myocytes were diffusely atrophic with scattered groups of remaining hypertrophic fibres. The clinical pathological and pathological features suggest that G. africana is primarily hepatotoxic with myocardial involvement occurring only in the terminal stages of the intoxication.  相似文献

Three African buffalo (Syncerus caffer) that died after capture and translocation from Mutirikwe Recreational Park in southern Zimbabwe showed macroscopic and microscopic lesions of cardiomyopathy compatible with a diagnosis of gousiekte. The buffalo had had access to Pavetta schumanniana, a plant that is known to cause gousiekte. Death was attributed to cardiac failure as a result of previous consumption of the plant, exacerbated by the stress of translocation.  相似文献

Foot-and-mouth disease virus was detected during two periods in the air of looseboxes which housed susceptible, vaccinated or recovered pigs, cattle or sheep exposed to infection. The first was 30 min to 22 h after exposure and occurred in all animals. The second was two to seven days after exposure and occurred with those susceptible and vaccinated animals which developed clinical lesions, and with vaccinated and recovered pigs and sheep, which did not develop clinical lesions. Vaccination of animals before exposure resulted in less or no virus being detected. The virus during the first period was attributed to virus trapped on the animal during exposure, and the virus during the second period to limited multiplication in the respiratory tract. Control of movement for two weeks after contact with infection is suggested as a means of preventing spread of foot-and-mouth disease in areas that contain vaccinated animals.  相似文献

SUMMARY Salmonellosis in sheep was studied on 7 commercial voyages from Australia to the Middle East and in one animal house study. Faecal excretion of salmonellas was not a good indicator of pathological changes in the gastrointestinal tract. The ratio of the proportion of sheep excreting salmonellas to the proportion with histological lesions of salmonellosis ranged from 1:1 to 23:1. Increasing excretion rates in our studies indicated that most sheep would be exposed to infection, although deaths from salmonellosis were not spatially or temporally clustered as would be expected if challenge alone was a sufficient cause. We considered that inappetence predisposed to death from salmonellosis, and that sheep with inappetence were likely to be randomly distributed aboard ship. In the animal house study, histological lesions of salmonellosis were exclusively in inappetent sheep although almost all sheep excreted salmonellas in faeces. Lesions of salmonellosis were found only in sheep that were seriously ill or had died, which suggested that, under the conditions of lot-feeding and sea transport, most sheep with enteric lesions are likely to die. Lesions were not found in feeding controls. Adrenal gland weights, an indicator of stress duration and severity, were used to examine the temporal sequence of events in the development of salmonellosis. Weights were greater in sheep that died of inanition than in controls, suggesting that inappetent sheep were already stressed for that reason. The presence of enteric lesions of salmonellosis was associated with further increases in adrenal gland weights. In a small proportion of cases, septicaemic salmonellosis developed rapidly and without adrenal gland hypertrophy suggesting that the sheep succumbed before the adrenal glands had time to enlarge. We concluded that sheep were predisposed to salmonellosis by inappetence and that stress was involved in lesion development. We considered that the development of enteric lesions of salmonellosis, in the live export system, inevitably led to death.  相似文献

Lymphoproliferative response (LPR) was studied in 19 lambs orally infected (Group I) with Mycobacterium avium subsp. paratuberculosis (MAP) with in vitro lymphocyte stimulation test using MTT dye reduction assay. The non-specific LPR against Con A and specific LPR against sonicated antigen and johnin PPD (purified protein derivatives) were estimated on preinfection (0 day) and various days postinfection period (15 to 330 dpi) in the animals, which were classified according to histological and bacteriological evidence of paratuberculosis infection. Of the two antigens used, johnin PPD was found to be superior in terms of consistency and uniformity of response over an observation period of about a year. Significantly (P < 0.05) higher LPR were observed in the infected sheep during postinfection period, as compared with preinfection values and values from uninfected control sheep. It was evident from the present study that the LPR in histologically infected animals fluctuated during the long course of infection and had a definite relationship with the gut pathology and the mycobacterial load. The LPR were stronger but variable in sheep with grades 1, 2 and 3 lesions (paucibacillary) and increased progressively from 30 dpi onwards. The sheep with the advanced lesions (grade 4, multibacillary) showed progressive decline in LPR till 120 dpi after initial stronger response at 30 dpi. Most of the animals were detected by LPR before initiation of faecal shedding of MAP. The results suggested that repeated testing was required while screening an infected flock for detecting most of the positive animals.  相似文献

Summary

The feel of horses, sheep, and goats of different breeds and from many different localities were examined for Chorioptes bovis. In horses, mites were mainly found in the Belgian and Frisian breeds (40% and 62% infected, respectively). In sheep and goats, respectively 63% and 86% were infected. In horses as well as in sheep and goats, mange‐lesions were rarely seen. A number of sheep and goats were examined for mites and lesions quantitatively. In sheep all mites were restricted to the region close to the accessory digits and the claws. In goats the average number of mites was higher than in sheep, and mites could be found on all locations of the feet at least as far as the carpal and tarsal joint. Both in sheep and goats the biggest density of mites was found just below the accessory digits. When crusts were present, they were generally small and hidden under the coat. In sheep, which were housed for a long period, crusts were seen more often and were more distinct than in pastured animals. A negative correlation between the number of mites and the presence and extensiveness of crusts was observed A possible explanation for this phenomenon is suggested. From the results of this study it is clear that there is no necessity to list chorioptic mange in sheep and goats as a notifiable disease.  相似文献

The first cases of scrapie were detected in Greece in a flock of sheep in October 1986. All the animals of the affected flock and all sheep in two flocks that were in contact were killed and buried. A systematic investigation of all available cases with signs indicating a neurological disease started in sheep and goats in late 1986, as well as in cattle in 1989. The investigation was based on clinical examination, necropsy or macroscopical examination of the brain and viscera, and histological examination of the brain in all animals except those with coenurosis. Histological examinations of specimens from the spinal cord and other tissues, and if considered necessary bacteriological, toxicological and serological examinations were also carried out. In October 1997, scrapie was diagnosed in sheep of a second flock (a mixed flock of sheep and goats), grazing in a pasture close to the place where scrapie was initially detected. All animals of the second flock were also killed and buried. Diagnosis in the first flock was based on clinical signs and histological lesions, and in the second immunoblotting was also used. Distinctive lesions of scrapie were found in the brain and/or the spinal cord of eight sheep with clinical signs from the two flocks. The lesions were revealed in the brain stem and/or in the cervical spinal cord, and tended to be symmetrical. In one sheep, severe lesions in the cortex of cerebral hemispheres and of the cerebellum were also found. In the brain of two sheep from the second flock the pathological isoform of PrP protein was detected. Despite the eradication scheme applied, scrapie in sheep reappeared after 11 years in a place close to where it occurred initially. This may indicate that the effectiveness of the eradication scheme implemented was not adequate and additional approaches may be needed.  相似文献

The synergistic hemolysis-inhibition (SHI) test, a serologic test for the detection of infection with Corynebacterium pseudotuberculosis, was applied to serum samples from 196 goats and 76 sheep, including animals both with and without C pseudotuberculosis abscesses. Fifty-one of 52 (98%) goats and 27 of 28 (96%) sheep with abscesses caused by C pseudotuberculosis had seropositive titers. Seropositivity continued on subsequent samplings, even after superficial lesions were completely healed. The SHI test may detect subclinically infected animals, as well as animals with clinically recognizable lesions. Of the animals without abscesses, 53 of 186 (28%) goats and 4 of 41 (10%) sheep were seropositive. Either the SHI test is lacking in specificity or these titers are a reflection of a past or a current infection without any grossly visible abscesses.  相似文献

A particular variant of the maedi visna virus (MVV) that although present in blood causes no clinical signs in infected sheep has been described. This variant carries a 13-14 nucleotide deletion in the R region of the proviral long terminal repeats. The hypothesis that this specific deletion may be associated with low pathogenicity has been investigated by comparing the distribution of proviral sequences, the histopathological lesions and the expression of viral proteins in the brain, lungs and udders of sheep naturally infected with viral strains carrying the deletion. Provirus could be demonstrated in most of the tissues examined from sheep infected with either type of virus, and the tissue-derived virus carried the typical deletion in the study flock animals. Histopathological analysis revealed that the lungs were significantly less affected in the animals infected with virus carrying the deletion. Concomitantly, viral expression was significantly reduced in the lungs of these animals. The findings suggest that the reduced pathogenicity of MVV with the specific deletion in the R region is not due to a restriction in the availability of specific tissues to infection, but is associated with a reduced capacity for viral expression in the lungs.  相似文献

Caseous lymphadenitis (CLA) infection was established experimentally in a total of 30, 2-tooth merino wethers. In the first experiment a broth culture of the causative organism, Corynebacterum pseudotuberculosis was placed directly on to a freshly shorn area on the right shoulder of 21 sheep after pretreatment with a defatting agent, water or 4 different commercial sheep dips. Within 6–13 days focal skin lesions appeared on 7 sheep and these were followed by abscessation of the right prescapular lymph node. Lung lesions of CLA developed in 6 sheep; 5 of which had skin lesions. In the second experiment C. Pseudotuberculosis broth culture was applied over the right shoulder region of 14 sheep, 24 hours post shearing. The 7 animals had only 1 application of the culture and the remaining 7 had 4 applications on consecutive days. A total of 9 of the 14 sheep developed skin lesions and subsequent CLA in the right prescapular lymph nod but there was no significant difference between the 2 methods of application. As part of the second experiment a broth culture of C. pseudotuberculosis was added to a commercial arsenical sheep dip and 2 hours later sprayed on to the right side of sheep shorn 24 hours or 2 weeks previously. Within 6–13 days all 14 sheep treated in this way developed skin lesions from which C. pseudotuberculiosis was recovered in dense pure growth. Healing time of skin lesions in all affected sheep ranged from 17–72 days post inoculation. A serological test to detect the presence of C. pseudotuberculosis antitoxin was used at regular intervals on all experimental sheep. The earliest positive reaction in an affected sheep was recorded 4 weeks post inoculation. In 23 known affected sheep tested at 35 weeks post inoculation, 10 were positive to the Zaki test. No false positive reactors were recorded. The localisation of focal lung lesions in the interstitial tissue suggests that pulmonary abscessation caused by C. pseudotuberculosis develops via the haematogenous route.  相似文献

The transmission of capripoxvirus to sheep, using an aerosol suspension of a Yemen isolate of the virus, was demonstrated. Capripoxvirus was also transmitted by contact to sheep and goats kept with animals infected with virus isolates from the Yemen, Sudan, India and Nigeria. The incubation period for capripoxvirus infection in sheep and goats was approximately eight to 12 days. Animals that had well developed clinical signs transmitted capripoxvirus more rapidly than animals which died of peracute disease or animals that had only mild clinical signs.  相似文献

An accidental infection from a vaccine was suggested as the explanation for the sudden increase in outbreaks of scrapie in Italy in 1997 and 1998. This paper describes a recent outbreak of scrapie in sheep and goats which were exposed to the same vaccine. No ewes or goats had been imported into the herd since 1992, but a vaccine against Mycoplasma agalactiae had been administered twice, in 1995 and 1997. High rates of crude mortality and scrapie incidence were experienced by both species, all birth cohorts were involved and a large proportion of aged animals was affected. A pattern of brain lesions was observed, with slight differences between the sheep and goats, which was very similar to the pattern observed in animals previously exposed to the same vaccine but clearly different from that observed in the brains of sheep with scrapie in a flock not exposed to the vaccine. Regardless of their exposure status, genotype analysis of the sheep showed the presence of polymorphism only at codon 171. The patterns of both incidence and brain lesions provide evidence that the epidemic of scrapie was due to the use of the vaccine.  相似文献

Sheep scab mites, Psoroptes ovis, collected from a Merino donor sheep, were used to infest Merino and Dorper sheep, and Angora and Boer goats. Mites were placed on the sheep on 1 or 2 occasions and on 5 occasions on the goats. All the animals were examined at regular intervals for the presence of scab lesions and living mites. Both sheep breeds developed lesions, but those on the Merino sheep were always larger than those on the Dorper sheep at the same intervals after infestation. None of the goats developed lesions or showed signs of irritation, or harboured any mites.  相似文献

AIM: To use disease modelling to inform a response team about the number of animals per herd/flock to be examined, and the start date and duration of clinical surveillance required to be confident that foot-and-mouth disease (FMD) was not present on an island in New Zealand with a population of approximately 1,600 cattle, 10,000 sheep and a small number of pigs, goats and alpacas. METHODS: Because the probability of detecting clinical disease in (the) primary case(s) in larger herds and flocks was extremely low, deterministic and stochastic mathematical SLIR (susceptible, latent, infectious, recovered) models for the transmission of infection were constructed to estimate the date when clinical lesions in herds and flocks would be detected with 95% confidence. Surveillance targeted the first wave of infections following a suspect index case. RESULTS: If 70 cattle in herds of about 400 cattle were examined it was estimated it would take approximately 13 (90% stochastic range 9-19) days from first exposure before it would be possible to achieve 95% confidence for detecting clinical signs for a low-virulence virus, and 9 (7-14) days for a high-virulence virus. The duration of sufficiently accurate clinical detection was 17 (15-19) days and 13 (12-14) days for low- and high-virulence viruses, respectively. A sample of 70 sheep from flocks of >1,000 would be required to achieve clinical detection at about the same time but with a shorter period of detection than for cattle. The duration of effective detection could be increased by examining a larger sample in most sheep flocks, however the small size of many cattle herds in the study population limited the confidence of detecting group-level disease in cattle, therefore necessitating repeated herd inspections. The model suggested that group-level detection was not feasible if it was based on elevated body temperature alone because of short durations of fever in infected animals. CONCLUSION AND CLINICAL RELEVANCE: Simulation modelling is a useful and powerful tool for informing ongoing surveillance activities in the face of an exotic disease incursion. Results of modelling suggested to start clinical inspection activities at 4 days and to continue regular inspection twice a week for about 35 days after the date of first exposure, to satisfy the required 95% confidence threshold of clinical detection of FMD in cattle herds and sheep flocks.  相似文献

The response of the supramammary lymph node of seven sheep to secondary infection with orf virus was examined by cannulating the efferent lymphatic before infecting the drainage area of the node. All animals developed typical orf lesions and responded after an initial lag period with an increase in total cell output paralleled by a rising proportion of lymphoblast cells. Most lymphoblast contained immunoglobulin with a predominance of the IgG class. When cultured, these cells produced measurable amounts of virus-specific antibody. The proportion of cells which stained with a T-cell-specific monoclonal antibody was measured in two of the sheep and found to decrease as the response developed. These data suggest that the nodal response is directed mainly towards the production of virus-specific antibody. The extent of T-cell involvement remains unclear.  相似文献

Maedi-visna, a multisystemic disease of adult sheep, was first described in Spain in 1984. To get an idea of the seroprevalence of the disease locally and to estimate the number of seropositive animals with lesions, samples of blood, lungs and mammary glands were taken from 124 randomly selected sheep killed in the main slaughterhouse of Zaragoza. In the agar gel immunodiffusion test, 74 (59.7 per cent) of the sheep were positive and 50 were negative. Among the 74 seropositive animals, 19 (25.6 per cent) had no lesions in any organ, 12 (16.2 per cent) had lesions in the lungs only, 15 (20.2 per cent) had lesions in the mammary glands and 28 (37.8 per cent) had lesions in both organs. In the lungs hyperplasia of lymphoid follicles was more evident than an interstitial infiltrate but in the mammary glands this relationship was not observed. Even when the lesions occurred in both organs, they did not show the expected proportion in terms of either type or severity. Among the 50 seronegative sheep, eight (16 per cent) showed maedi-like lesions, formed exclusively by the hyperplasia of lymphoid follicles.  相似文献

This report describes the clinical and laboratory findings for 5 sheep from 3 different flocks with extensive proliferative skin lesions grossly resembling warts on the distal limbs. The lesions affected the front and rear extremities in all sheep, and 2 sheep also had lesions around the head. The sheep exhibited signs of pain when the lesions were touched, and most sheep were reluctant to move. Various empirical treatments, including systemic antibiotics, topical antibiotics, and antifungal ointments, were administered without clinical improvement. Diagnostic tests including skin biopsy and histopathology, examination of skin scrapings, bacteriology, mycology, electron microscopy of lesions, and immunohistochemical analysis demonstrated that the lesions were the result of parapoxvirus infection. All 5 animals were euthanized either because of the lack of resolution of clinical signs or a decision by the owner. These animals illustrate an atypical presentation of parapoxvirus infection in sheep (orf, contagious ecthyma, and scabby mouth). The infection appeared to be minimally contagious; however, the lesions did not spontaneously resolve. This appears to be the 1st report of such lesions in multiple sheep in North America, although similar lesions have been reported in Israel and the United Kingdom.  相似文献
