The relative accuracies of various commercially available detection procedures in the evaluation of pregnancy status were evaluated in 51 sows of mixed parity. Sows were mated 1 to 3 times during estrus to one or more boars. A blood sample was obtained from each sow between 19 and 23 days after estrus (first day of estrus = day 1) and again between 26 and 30 days and was analyzed by radioimmunoassay for content of progesterone and estrone sulfate, respectively. Sows were considered pregnant if progesterone concentrations were greater than 5 ng/ml and if estrone sulfate concentrations were greater than 0.5 ng/ml. At daily intervals, sows were exposed to mature, sexually aggressive boars and observed for signs of estrus. Between 31 and 35 days of gestation, sows were evaluated for pregnancy by use of the following mechanical devices: a Doppler ultrasound pregnancy detector fitted with an abdominal probe, the Doppler detector with a rectal probe, an amplitude-depth ultrasound pregnancy detector (diode ruler), and another amplitude-depth ultrasound detector from a different manufacturer (oscilloscope). Relative to the proportion of sows that farrowed, the overall accuracies of the techniques were returns to estrus, 98%; estrone sulfate, 92%; progesterone, 92%; Doppler with rectal probe, 86%; Doppler with abdominal probe, 86%; diode ruler amplitude-depth ultrasound, 73%; and oscilloscope amplitude-depth ultrasound, 86%. Estrus detection resulted in the lowest prevalence of sows that were predicted to farrow, but did not subsequently farrow (false-positive diagnoses) and the lowest prevalence of sows that were not predicted to farrow, but subsequently did farrow (false-negative diagnoses).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

This investigation characterized serum concentrations of luteinizing hormone (LH), estradiol-17 beta (E2), progesterone (P4) and cortisol (C) in anestrous sows. Twenty-two sows that had not returned to estrus within 45 days after weaning (anestrous sows), and ten sows that had returned to estrus within seven days following weaning (cyclic sows) were nonsurgically fitted with indwelling jugular vein cannulae. Blood samples were collected at 6 h intervals for seven days and at 15 min intervals for 8 h on the fifth day after cannulation. Serum LH concentrations were determined in all samples, while C, E2 and P4 levels were quantitated in serum collected at 6 h intervals. Serum P4 concentrations in anestrous sows were consistently less than 0.5 ng/mL, and E2 levels ranged from 10 to 19 pg/mL. Concentrations of LH remained less than 1.0 ng/mL in anestrous sows, whereas a preovulatory LH surge was observed in five of ten cyclic sows. There was a circadian rhythm in mean C levels with C peaks occurring at 0600 or 2400 h and nadir levels observed at 1200 and 1800 h. Few differences in C levels were detected between anestrous and cyclic sows. It was evident that anestrous sows did not exhibit cyclic or predictable variations in steroid hormone concentrations. Unfortunately, the results of this study failed to elucidate the endocrine pathogenesis of the anestrous sow.  相似文献

The accuracy of urinary estrone sulphate (E1S) concentrations as an indication of pregnancy was compared to that of plasma E1S concentration. Urine and plasma were collected from 145 sows 18 to 30 days after mating. Concentrations of E1S in urine were indexed to creatinine concentration (CR) and specific gravity (SG) to correct for urine dilution. There was no significant difference in E1S concentration between urine corrected for CR or SG and uncorrected urine (UN) from pregnant sows. Urinary E1S concentrations were 20 to 100 times higher than plasma E1S concentrations. The E1S test was accurate in the detection of pregnancy in sows using both plasma and urine (test sensitivity, 98.8% vs. 96.4%, respectively) during the optimal sampling period of 20 to 30 days post mating. The test was slightly more accurate (NS) using plasma than urine in detecting non-pregnant sows (test specificity, 100.0% vs. 91.9%, respectively) 20-30 days after mating. Urinary E1S concentrations could be used to predict litter size, but the precision was poor ( +/- 3 piglets), and thus could only be used to predict small (less than 5 piglets), medium (6-10 piglets) and large (greater than 11 piglets) litters.  相似文献

The study was carried out on 50 goats of Polish White breed. Blood samples were taken on day 60 after mating from V. jugularis. Pregnancy diagnosis was made using transcutan ultrasound scanning (Pie Medical Scanner 2000 with a 5 MHz curved array probe). The estrone sulphate level was determined by RIA method after extraction and hydrolysis with arylsulphatase/glucuronidase from Helix pomatia. As a threshold value for differentiation of pregnant from nonpregnant goats 1 ng/ml was assumed. In eight nonpregnant goats the estrone sulphate level was 0.61 +/- 0.21 ng/ml. In 42 pregnant goats the level was 6.1 +/- 3.5 ng/ml. The diagnosis was correct in all cases. The number of single, twin and triplet pregnancies was six, 34 and two, respectively. Estrone values in goats with twin pregnancies were significantly higher compared to those with single pregnancies (7.2 +/- 4.1 ng/ml vs. 4.5 +/- 1.8 ng/ml; p < or = 0.01). The results of this study indicate, that the determination of estrone sulphate in blood of goats may be useful for pregnancy diagnosis. The estrone sulphate level in goats with twins is higher, but due to high variation between animals the number of fetuses cannot be exactly predicted.  相似文献

Early and accurate diagnosis of nonpregnant sows and gilts has the potential to increase reproductive efficiency and the financial income in pig production by reducing non-productive days per sow per year. The objectives of this study were to compare the efficiency of pregnancy diagnosis between Doppler Echo+ and real time ultrasonography (RTU) and to compare the efficiency by using RTU at different days post-mating (days 17 to 24) under commercial conditions. In the 1st study, using crossbreed sows and gilts (n=107), pregnancy diagnoses were done with Doppler Echo+ and then with RTU. Between 28 and 65 days of gestation, Doppler Echo+ had 85% sensitivity and 32% specificity, and efficiency was 73%. In the 2nd study, sows (n=142) were scanned for pregnancy diagnosis between 17 and 24 d post-mating (PD1) and reconfirmed between 38 and 45 days of gestation (PD2). After 21 days of gestation, RTU had over 90% sensitivity and 45% specificity, and 70% efficiency. Accuracy between PD1-farrowing was 75.5% and between PD1 and PD2 was 80.6%. In the 3rd study, sows were diagnosed pregnant by RTU (n=151) at 17 to 24 days of gestation or A-mode ultrasound (n=172) at 28 to 30 days of gestation. There were no significant differences in conception rate (P > 0.09) and farrowing rate (P > 0.67) between both groups. Hence, there was no improvement in fertility and farrowing rate by using RTU instead of A-mode ultrasound under commercial conditions.  相似文献

Delaying the onset of estrus after weaning and adding fat to the postweaning diet were studied for their effects on estrus and fertility in 232 crossbred primiparous sows on a commercial swine farm. Sows were assigned randomly to the following treatments after weaning in June, July, August, or September, 1983: 1) altrenogest (20 mg/d) was fed for 7 d after weaning (n = 76), 2) altrenogest was fed for 7 d plus .53 kg dried animal and vegetable fat product (.45 kg actual fat/d) for 14 d after weaning (n = 78), or 3) no treatment (controls, n = 78). While a similar proportion of sows came into heat after weaning (lactation length = 4 wk), sows fed altrenogest (14.4 +/- .2 d) returned to estrus about 9 d later (P less than .01) than controls (5.6 +/- .2 d). Serum progesterone concentration was assayed in blood samples collected from a subgroup (74%) of the cows not observed in estrus by 3 wk after weaning to determine possible causes of anestrus. If serum progesterone (greater than 5 ng/ml) was elevated, we assumed that sows had ovulated without expressing estrus (behavioral anestrus) or ovulated with undetected estrus (less overt estrus or error in estrous detection), whereas low progesterone (less than or equal to 5 ng/ml) indicated that sows were anovulatory. About 53% of the sows not observed in estrus across all treatments had luteal function, probably resulting from post-weaning ovulation. Incidence of anovulation without estrus was 47%. Lactating, primiparous Landrace x Yorkshire sows were used to characterize LH secretion during lactation in sows that experienced an early (less than 9 d; n = 14) or late (greater than 15 d; n = 9) return to estrous postweaning and to evaluate the relationship between LH secretion and blood metabolites. Twenty-three sows were fed one of nine corn-soybean meal diets to achieve a matrix of lysine (15 to 45 g/d) and energy (6.5 to 16.5 Mcal of ME/d) intakes and a range in metabolite concentrations and return-to-estrus intervals. Blood samples for LH analysis were collected every 15 min for 6 h on d 0, 7, 14, 21, and 28 of lactation. Circulating concentrations of glucose, amino acids, insulin, triglycerides, urea N, and nonesterified fatty acids also were measured on d 7 and 21. Mean LH concentrations were .27 and .42 ng/mL at farrowing for sows with an early and late return to estrus, respectively, but decreased (P less than .01) to .12 ng/mL by d 7 in both early and late groups. Mean LH and number of LH peaks per 6 h increased linearly (P less than .01) from d 7 to 28 for early sows. Early sows had a higher LH mean and more LH peaks per 6 h on d 14, 21, and 28 than did late sows (P less than .05). Early sows had higher serum insulin on d 7 (P less than .05) and d 21 (P less than .01) than did late sows. The present study was carried out to measure fecal progestagen and estrone concentrations during pregnancy in a giraffe and examine the possibility of utilizing this assay system for pregnancy diagnosis. Fecal samples were collected from a giraffe during her third and fourth parities and mixed with methanol to prepare a fecal solution. Diluted fecal solution was used for direct enzyme immunoassay for progestagen and estrone. The newborn calf from the third parity was viable, although that from the fourth parity died 5 days after calving. In the third parity, the giraffe's progestagen and estrone concentrations increased transiently from days 30 to 120 of pregnancy. Then, they decreased and remained low until day 330. This was followed by a drastic rise in both concentrations as parturition approached. Parturition caused a reduction in the progestagen and estrone concentrations of the feces. In the fourth parity, the progestagen concentration increased gradually after mating until day 320. This was followed by a reduction in the concentration until parturition. However, the estrone concentration fluctuated, and the duration and extent of the prepartum rise in concentration were shorter and lower than those of the third parity. The hormone dynamics of the third parity suggest the possibility of early pregnancy diagnosis by measuring progestagen or estrone between days 30 and 120 after mating.  相似文献

Female guinea pigs were tested to determine whether they could serve as a model of zearalenone (ZEN) toxicosis during early pregnancy, as observed in domestic swine. Only 1 of 4 female guinea pigs that were given 21 mg of ZEN/kg of body weight orally during the first 8 days after mating was pregnant when examined 22 days after mating. Guinea pigs that were given 7 or 14 mg of ZEN/kg had normal fetal development. Serum concentrations of progesterone were less than 12 ng/ml in all guinea pigs 8 and 15 days after mating. Serum concentrations of progesterone were greater than 100 ng/ml in pregnant guinea pigs on day 22, but remained less than 12 ng/ml in nonpregnant guinea pigs. Three of 5 guinea pigs treated with 20 mg of ZEN/kg and only 1 of 4 guinea pigs given 30 mg of ZEN/kg on days 1 to 3 after mating were pregnant 22 days after mating. Female guinea pigs treated with 20 or 30 mg of ZEN/kg on days 4 to 5 or 6 to 8 after mating and female guinea pigs treated with 60 or 90 mg of ZEN/kg on days 4 and 5 after mating had normal pregnancies. In four Kenyan pig breeding units the pregnancy diagnosis of sows has been carried out in two groups: Group 1 (n = 1911): the sows were transrectaly pregnancy tested between Days 17-22 post-mating by ultrasound. Sows testing non-pregnant immediately received one dose of 400 IU pregnant mare serum gonadotropin (PMSG) (equine chorion gonadotropin, eCG) and 200 IU human chorion gonadotropin (hCG). On showing signs of oestrous, the animals were subsequently artificially inseminated (AI). Group 2 (n = 1923): sows were pregnancy tested by serum progesterone (P4)-based enzyme-linked immunosorbent assay (ELISA) on Day 17 post-breeding. P4 concentrations were categorized as positive (> 5 ng/ml) or negative (< 5 ng/ml). Sows testing nonpregnant immediately received one dose of 400 IU PMSG and 200 IU hCG by injection, and were subsequently artificially inseminated. The following parameters were evaluated: sows diagnosed non-pregnant, days from first post-weaning insemination until the sows were inseminated at their first return to oestrus; farrowing rate and total piglets born and number of live-born piglets in litters. The percentage of sows diagnosed non-pregnant in the two groups, as well as the totals of born piglets and of live-born piglets in litters did not differ significantly between the two groups. The number of days from the first post-weaning mating until the sows were artificially inseminated at their first return to oestrus and the administration of eCG and hCG was shorter (P < 0.01) and farrowing rate was higher (P< 0.01) in the ELISA-tested sows.  相似文献

The objectives of the present study were to measure oestrone derivatives [oestrone, oestrone sulphate (E1S) and oestrone glucuronide] in sow faeces by direct enzyme-linked immunosorbent assay (ELISA) and to explore the changes of oestrone derivative profile in faeces of miniature pig for demonstrating the possibility of pregnancy diagnosis. Faecal samples (1.5 g) were put into 6 ml of buffer, shaken and centrifuged. Then supernatant was added to the wells of multi-plate without extraction and incubated with anti-oestrone antibody and horseradish peroxidase-labelled oestrone. Standard solution was prepared at various concentrations of E1S. Sensitivity was estimated as 0.035 ng/ml (0.14 ng/g). Intra- and inter-assay coefficient variations were 3.5-7.7% and 10.9-15.3%, respectively. When 1-5 ng/ml E1S were added to a faecal solution, recovery rates ranged between 80.0 and 103.3%. There is a temporal increase in the E1S equivalent concentration of miniature pig faeces from day 25 to 31 after mating. From day 35 to 70, the E1S equivalent concentrations remained low. Thereafter its concentrations increased again towards farrowing. On day 27 and 29 after mating pregnancy diagnosis by the faecal E1S equivalent concentrations agreed with the results of farrowing (seven of seven animals). These results suggest that the present direct ELISA is practical and suitable as a routine assay for measuring the faecal concentration of oestrone derivatives and that this assay might be usable for pregnancy diagnosis in sows at day 27-29.  相似文献

Modern sows are younger and leaner at time of mating and probably have poorer appetites than sows of 10 to 15 years ago. Therefore, feeding strategies should aim to minimize weight loss and maintain a sow's body condition throughout her reproductive life. The efficiency with which gilts are introduced into the breeding herd is as important in economic terms as is the efficiency with which the sow returns to estrus after weaning. Gilts should be selected at 50 to 60 kg, and fed a 16% protein diet ad libitum until mated at their second estrus, when they weigh 115 to 120 kg and have 17 to 20 mm backfat. Flushing gilts before the onset of second or third estrus increases ovulation rate of restricted gilts to the levels achieved by gilts fed ad libitum. During gestation, maintenance represents 75 to 85% of total energy requirements. The aim should be to achieve 20 to 25 mm backfat at farrowing. Increased feed intake from day 2 to 3 after mating will not increase embryo mortality. Feeding an extra 1 kg feed/sow/day for the last 10 days of gestation increases piglet birth weight slightly and prevents a loss of 1.5 to 2.0 mm of sow backfat. Wherever possible, sows should be fed ad libitum from the day after farrowing until weaning. Reduced feed intake by lactating sows, for whatever reason, results in excessive weight and condition loss. Excessive weight loss in lactation causes extended remating intervals, a lower percentage of sows returning to estrus within 10 days of weaning, reduced pregnancy rate, and reduced embryo survival. Ovulation rate is not affected by level of feed intake in lactation. It has been suggested that sows will have minimum weaning-to-service intervals when they weigh 150 kg or more at weaning. It is likely that the sow must be anabolic for about 10 days before she will exhibit postweaning estrus. The decision when to rebreed is made some time prior to weaning and is mediated by a host of substrates, hormones, and neurotransmitters. Sows with a delayed return to estrus also have a lower pregnancy rate and smaller subsequent litters. If sows lose considerable weight or condition during lactation, a high level of feeding in the postweaning period will improve embryo survival.  相似文献

This study investigated whether injections of ACTH for 48 h, from the onset of the second standing estrus after weaning, had any impact on time of ovulation and patterns of progesterone, estradiol, luteinizing hormone (LH), and inhibin alpha. The studied sows (n=15) were fitted with jugular vein catheters and randomly divided into a control (C group) and an ACTH group. From the onset of standing estrus, the sows were injected (NaCl or synthetic ACTH, 5 microg/kg) every 4h; blood samples were collected immediately before and 45 min after each injection. Ovulation was monitored using ultrasonography. The ACTH-group sows stopped displaying signs of standing estrus sooner after ovulation in their second estrus, but no impact was found on time of ovulation. There were no significant differences in the intervals between LH peak, estradiol peak, and the onset of standing estrus between the C and ACTH groups. The cortisol and progesterone concentrations were significantly elevated (p<0.001) in samples taken 45 min after ACTH injection. There were minor differences in estradiol and LH concentrations between the groups. Overall inhibin alpha concentrations were significantly higher during the treatment period in the ACTH than in the C group, but there were no significant differences between samples taken either 45 min or 4h after injection. In conclusion, injections of synthetic ACTH during estrus in the sow apparently disturb the duration of signs of standing estrus and the hormonal pattern of progesterone, and possibly of inhibin alpha, estradiol and LH.  相似文献

Serum and urinary estrone sulfate concentrations were determined in 7 pregnant mares before and after prostaglandin-induced abortion (n = 4) or surgical removal of the fetus (n = 3) to determine the source of estrogen during early pregnancy (gestation days [GD] 44 to 89). Estrone sulfate concentrations also were determined in serum samples (stored frozen for 2 years) from 3 mares that had been ovariectomized between GD 51 and 58. Estrone sulfate concentrations decreased in serum and urine after expulsion or removal of the fetus (urinary patterns were more definitive than were patterns for serum), whereas a transient decrease in serum estrone sulfate concentration was observed after ovariectomy. Seemingly, products of conception are the major source of estrone sulfate during early pregnancy, although there appears to be some ovarian contribution. Serum or urinary estrone sulfate measurements provide a simple and accurate test for fetal viability after GD 44 in the mare.  相似文献

An experiment was conducted to study the effects of major dietary energy source fed from weaning to ovulation or from ovulation to d 35 of pregnancy on reproductive traits in primiparous sows. Dietary energy sources were used to manipulate the plasma insulin concentration. One hundred thirteen sows were used in a split-plot design. From weaning to ovulation sows were fed at two times maintenance either a diet with tallow (Fat) or maize starch plus dextrose (Starch) as the major energy source. From ovulation onward, sows within each dietary group were alternately reassigned to either the Fat or the Starch diet and were fed at 1.25 times maintenance. Estrus detection was performed three times a day from d 3 to 9 after weaning and sows were inseminated each day of standing estrus. On d 35 of pregnancy, the sows were slaughtered and their reproductive tracts were removed. Plasma insulin concentration was higher in sows fed the Starch-rich diet than in sows fed the Fat-rich diet on d 4 after weaning (1.30 vs 0.97 ng/mL, P = 0.08) and on d 32 of pregnancy (1.20 vs 0.51 ng/mL, P < 0.001). Plasma glucose and IGF-I concentration on d 4 after weaning and d 32 of pregnancy did not differ between sows on the two dietary energy sources. The percentage of sows exhibiting estrus within 9 d after weaning was 52 and 67% for the Fat and Starch diet before ovulation, respectively (P = 0.11), whereas the weaning-to-estrus interval was 134 vs 123 h, respectively (P = 0.12). Survival analysis showed that sows fed the Fat-rich diet had a 1.6 times higher risk to remain anestrous until d 9 after weaning than sows fed the Starch-rich diet (P = 0.04). No effect of dietary energy source, either before or after ovulation, on uterine, placental, or embryonal development on d 35 of pregnancy was found. It can be concluded that the dietary energy source provided after weaning can affect the risk of sows to remain anestrous but does not affect uterine, placental, or embryonic traits.  相似文献

Hormonal changes, lesions, and virus isolation studies were determined in sows after uterine artery inoculation with porcine parvovirus [( PPV], strain NADL-8) in early pregnancy. Two sows were given PPV on days 14 or 16 and were euthanatized and necropsied on day 35 after twice daily plasma collection for hormone measurement. Parvovirus was given to 4 sows on day 14 and to 4 sows on day 21 with 5 times daily plasma samples collected for 1 week. Sows were examined on days 21 and 28, respectively. Four control sows in each group on days 14 and 21 were given a placebo injection and were similarly studied. All embryos in all but 1 sow given PPV were in various stages of resorption at necropsy. Normal embryos were present in all control sows. Estrone sulfate values increased logarithmically, progesterone values remained stable, and concentrations of 13, 14-dihydro-15-keto-prostaglandin (PG) F2 alpha (PGFM), a PGF2 alpha metabolite, were less than 200 ng/ml for sows given a placebo. In contrast, sows with resorbing embryos did not have an increase in estrone sulfate values. A decrease in plasma progesterone values occurred in 9 of 10 sows inoculated with PPV; this decrease was accompanied by greater than or equal to 1 marked increase in PGFM concentrations. Quantitative assessment of the uterus revealed significantly greater cytoplasmic density in endometrial and glandular cell (P less than 0.01), a greater glandular epithelium height (P less than 0.05), and twice the number of glands (P less than 0.05) in control sows, compared with values in sows inoculated with PPV.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

选用5头妊娠后期母猪,于妊娠111d上午9∶00肌肉注射氯前列烯醇(PG)0.20mg。5头母猪皆于次日白天分娩。从注射前至分娩结束,在整个诱导过程中多次采血,监测了血清中孕酮(P4)、硫酸雌酮(E1S)、催乳素(PRL)和皮质醇(cortisol)4种激素浓度的变化。结果表明,诱导分娩过程中各种激素的变化和自然分娩基本一样。这就解释了用国产前列腺素诱导母猪分娩,只要处理时间合适(妊娠111d或112d),就会有良好效果,不会产生不良副作用。  相似文献

AIMS: To investigate the utility of faecal oestrone sulphate (OS) concentrations for detecting pregnancy in mares during behavioural studies of feral horses, in which the collection and preservation of samples is not immediate. METHODS: Oestrone sulphate concentrations were measured in fresh dung samples collected from 153 free-roaming Kaimanawa mares throughout the year. In addition, multiple samples were taken from the same pile to investigate the reliability of diagnosis from a single sample, as well as the influence of time until preservation on OS concentrations. Samples were also taken before and after a 10mm simulated rainfall event to test for dilution of OS concentrations by rain. Oestrone sulphate concentrations in all samples were measured using an enzyme immunoassay. RESULTS: From approximately 150 to 250 days of gestation, OS concentrations were consistently >80 ng/g in mares which subsequently foaled. Mares which did not foal and had low faecal OS concentrations in multiple samples throughout the year had faecal OS concentrations of 31+/-13 ng/g (mean+/-s.d.) with an upper 95% confidence limit of 57 ng/g. Mares sampled from 1 week before to 1 month after behavioural oestrus, and that did not foal in the previous and subsequent seasons, had OS concentrations of 37+/-32 ng/g (mean+/-s.d.) with an upper 95% confidence limit of 100 ng/g. The standard error of oestrone sulphate concentrations in multiple samples from the same dung pile ranged from 1 to 37% of the mean. This large within-pile variation, however, did not result in incorrect diagnoses from single samples unless mares were within 18 days of parturition. Keeping samples at ambient temperatures for up to 16 hours did not affect OS concentrations. Simulated rainfall caused a 17% mean reduction in OS concentrations, but did not change pregnancy diagnoses. CONCLUSIONS: Faecal OS concentrations >100 ng/g were indicative of pregnancy in Kaimanawa mares. For mares more than 150 days post-mating, OS concentrations <57 ng/g were indicative of non-pregnancy, while concentrations between 57 and 100 ng/g provided an inconclusive diagnosis. A single sample from each dung pile collected within 16 hours of defecation was sufficient to accurately diagnose pregnancy in mares 150-250 days post conception. CLINICAL RELEVANCE: Measurement of OS concentrations in dung samples was a reliable and robust indicator of pregnancy status in feral mares 150-250 days post mating. This corresponds approximately to the period from May to August, given the seasonal breeding pattern in this population. This method of determining pregnancy status is suitable for field use in behavioural and demographic studies of wild horse populations.  相似文献
