In the present study, the in vivo effects of λ-cyhalothrin and cypermethrin on the activity of acetylcholinesterase (AChE, EC 3.1.1.7) were evaluated for 96 h in brain, muscle and gills of Channa punctatus. Both compounds exhibited tissue specific as well as dose dependent decrease in the activity of AChE. The treated fish showed a significant decrease in the activity of AChE in brain and a lesser inhibition in muscle and gills in response to the increasing concentrations of λ-cyhalothrin as well as cypermethrin. Our results indicated that the brain was the main target organ for both insecticides, followed by muscle and gills, as determined by AChE inhibition study. However, these organs showed variations in the degree of AChE inhibition for separate treatments of both insecticides. The λ-cyhalothrin was a more potent AChE inhibitor as compared to cypermethrin. These findings indicated that apart from the established mechanism of delayed closure of sodium ion channels, these pyrethroids inhibit the activity of AChE in C. punctatus which could further aggravate their neurotoxic effects.  相似文献

The assessment of human health risks resulting from the presence of metabolites in groundwater and food residues has become an important element in pesticide authorisation. In this context, the evaluation of mutagenicity is of particular interest and a paradigm shift from exposure‐triggered testing to in silico‐based screening has been recommended in the European Food Safety Authority (EFSA) Guidance on the establishment of the residue definition for dietary risk assessment. In addition, it is proposed to apply in silico predictions when experimental mutagenicity testing is not possible due to a lack of sufficient quantities of the pesticide metabolite. This, combined with animal welfare and economic considerations, has led to a situation where an increasing number of in silico studies are submitted to regulatory authorities. Whilst there is extensive experience with in silico predictions for mutagenicity in the chemical and pharmaceutical industry, their suitability in pesticide regulation is still insufficiently considered. Therefore, we herein discuss critical issues that need to be resolved to successfully implement (Quantitative) Structure‐Activity Relationship ((Q)SAR) as an accepted tool in pesticide regulation. For illustration purposes, the results of a pilot study are included. The presented study highlights a need for further improvement regarding the predictivity and applicability domain of (Q)SAR systems for pesticides and their metabolites, but also raises other questions such as model selection, establishment of acceptance criteria, harmonised approaches to the combination of model outputs into overall conclusions, adequate reporting and data sharing. © 2020 The Authors. Abstract

Permethrin applied as a spray to cattle at 0.025% or 0.05% effectively controlled Boophilus microplus (Canestrini) by the third day post‐treatment, but only provided < 60% control within 1 day post‐treatment with both concentrations. Percentage control did not drop below 90% until day 28 for 0.025% and day 31 for 0.05% permethrin; percentage control did not fall below 92% for 0.025% fenvalerate until after day 24–25; and at 0.05%, fenvalerate provided 95% control until day 28. A multiple treatment (spray applied every 2 weeks) with permethrin (0.05%) provided greater than 96% control of B. microplus after the initial treatment and 100% control by day 56. We concluded that although both chemicals have potential in an eradication program, they are not satisfactory for use at a quarantine line because both acaricides provided less than 100% control within 1 day post‐treatment.  相似文献

It has been reported previously that the major resistance mechanism to pyrethroid insecticides by the cotton bollworm Helicoverpa armigera (Hübner) in Australia is a consequence of overproduction of esterase isoenzymes. This paper reports structure-activity relationships that support such a view, based on in vivo bioassays conducted with a range of pyrethroid structures containing a variety of acid and alcohol moieties and the correlation with in vitro esterase inhibition assays against the same structures, and identifies the critical regions of the molecule with regard to esterase inhibition, and hence resistance. The implications of this work in terms of possible resistance management are evaluated and discussed.  相似文献

Twelve tundra swans, Cygnus columbianus (Ord), from Nevada and one from New Mexico were collected and examined for schistosomes. Mature worms, determined as Allobilharzia visceralis, were found in 92% of the swans, in the inferior mesenteric vein of the large intestine and its branches. In 12 cases, there was endophlebitis of the inferior mesenteric vein. The morphology of the worms is consistent with the recently described genus Allobilharzia. Placement in this genus was confirmed also by phylogenetic analysis of nuclear 28S, 18S and, internal transcribed spacer (ITS) ribosomal DNA (rDNA), and mitochondrial cytochrome oxidase I (CO1) sequences. Data further suggest the worms are con-specific with the European A. visceralis, the only described species of the genus and which was found to be the sister taxon to the most diverse avian schistosome genus, Trichobilharzia. This is the first report of a schistosome infection from native swans in North America.  相似文献

Abstract: Two hundred and seventeen captive great apes (150 chimpanzees, Pan troglodytes; 14 bonobos, Pan paniscus; 53 western gorillas, Gorilla gorilla) and 20 personnel from thirteen European zoos and two African sanctuaries were sampled and examined in order to determine the occurrence ofEnterocytozoon bieneusi and species of Encephalitozoon in faecal specimens and to compare the epidemiological situation between zoos and sanctuaries. Microsporidia were detected at all sampling sites. Sequence analyses of ITS amplicons generated by using microsporidia-specific primers determined the presence ofmicrosporidia in 87 samples including 13 humans; since two cases of simultaneous occurrence of Encephalitozoon cuniculi and Enterocytozoon bieneusi were identified, 89 full-length ITS sequences were obtained, namely 78 Encephalitozoon cuniculi genotype I, five E. cuniculi genotype II, two E. hellem 1A and four Enterocytozoon bieneusi. No Encephalitozoon intestinalis-positive samples were identified. This is the first report of Encephalitozoon species and Enterocytozoon bieneusi genotypes in captive great apes kept under various conditions and the first record of natural infection with E. hellem in great apes. A comparison of zoos and sanctuaries showed a significantly higher prevalence of microsporidia in sanctuaries (P<0.001), raising a question about the factors affecting the occurrence of microsporidia in epidemiologically and sanitarily comparable types of facilities.  相似文献

Using a recently developed model pathosystem involving Medicago truncatula and Mycosphaerella pinodes, causal agent of Mycosphaerella blight on pea to understand host molecular response to a fungal suppressor, we applied the suppressor to leaves of M. truncatula and identified 151 nonredundant cDNA fragments as newly expressed genes. These included genes encoding lipoxygenase (LOX) and enoyl-CoA hydratase, which are presumably involved in jasmonic acid (JA) synthesis. Potential genes encoding plastidic enzymes, including allene oxide synthase (AOS) and allene oxide cyclase (AOC), and other peroxisomal enzymes involved in β-oxidation were predicted from the Medicago Gene Index EST database and tested for altered expression by semiquantitative RT-PCR. The coordinated expression of genes encoding both plastidic and peroxisomal enzymes showed that the suppressor likely conditions certain cellular process(es) through the JA synthesis in M. truncatula. To explore the role of JA or JA-regulated cellular process(es) in conditioning susceptibility, we used an Apple latent spherical virus (ALSV)-based virus-induced gene silencing (VIGS) technology to silence pea genes including LOX, AOS, AOC and 12-oxo-phytodienoic acid reductase (OPR). In LOX-, AOS-, AOC- or OPR-silenced pea plants, disease development induced by M. pinodes was remarkably reduced. Similarly, silencing of mRNA for LOX, AOS, AOC or OPR reduced the sensitivity to a phytotoxin, coronatine, which is believed to act through a JA-dependent process. On the basis of these results, it is conceivable that M. pinodes has evolved a strategy to condition susceptibility by manipulating the physiology of host cells, in particular JA-regulated cellular process(es), to promote disease development in pea.  相似文献

Detecting near-surface soil freeze-thaw cycles in high-altitude cold regions is important for understanding the Earth's surface system, but such studies are rare. In this study, we detected the spatial-temporal variations in near-surface soil freeze-thaw cycles in the source region of the Yellow River(SRYR) during the period 2002–2011 based on data from the Advanced Microwave Scanning Radiometer for the Earth Observing System(AMSR-E). Moreover, the trends of onset dates and durations of the soil freeze-thaw cycles under different stages were also analyzed. Results showed that the thresholds of daytime and nighttime brightness temperatures of the freeze-thaw algorithm for the SRYR were 257.59 and 261.28 K, respectively. At the spatial scale, the daily frozen surface(DFS) area and the daily surface freeze-thaw cycle surface(DFTS) area decreased by 0.08% and 0.25%, respectively, and the daily thawed surface(DTS) area increased by 0.36%. At the temporal scale, the dates of the onset of thawing and complete thawing advanced by 3.10(±1.4) and 2.46(±1.4) days, respectively; and the dates of the onset of freezing and complete freezing were delayed by 0.9(±1.4) and 1.6(±1.1) days, respectively. The duration of thawing increased by 0.72(±0.21) day/a and the duration of freezing decreased by 0.52(±0.26) day/a. In conclusion, increases in the annual minimum temperature and winter air temperature are the main factors for the advanced thawing and delayed freezing and for the increase in the duration of thawing and the decrease in the duration of freezing in the SRYR.  相似文献

A phytoplasma infecting alfalfa crops was detected and characterised in Argentina, the Argentinean Alfalfa witches′-broom (ArAWB) phytoplasma. Typical witches′-broom symptoms were observed in diseased alfalfa plants from fields of the ‘Cuyo’ region in the Andean midwest. Pleomorphic bodies were observed by electron microscopy in sieve tubes of the diseased plants. The results obtained from the sequence homology, similarity coefficients derived from RFLP of the 16S rDNA and phylogenetic analysis led us to include this phytoplasma in the 16Sr VII (Ash Yellows) group. However, the ArAWB phytoplasma showed several differences when compared to other members of group 16Sr VII. The RFLP analysis of partial 16S rRNA gene of two ArAWB isolates, digested with 16 restriction enzymes, showed differences between the ArAWB and the reference strain (AshY1T) in six enzyme patterns. Restriction patterns unique for the group and an exclusive HinfI restriction site were found in the ArAWB phytoplasma rDNA. Moreover, the similarity coefficients (0.92–0.86) were lower than those obtained among other group members. The significant differences detected suggested that this phytoplasma belonged to a subgroup different from those described so far. We propose therefore, that the ArAWB phytoplasma should be included in a new VII-C subgroup, closely related to the EriWB phytoplasma (VII-B) described in Brazil.  相似文献

Abstract

Very large breeding sites of the onchocerciasis vector, Simulium sirbanum were discovered along the Rivers Niger, Mafou, Kouya, Niandan and Milo in the Upper Niger Basin of Guinea. Flies reared from these breeding sites and caught biting man nearby were found to be in the same size range as those collected in the south‐eastern Mali invasion zone of the Onchocerciasis Control Programme Area. The periodicity of the Mali invasion appeared to be related to the hydrological conditions in the Upper Niger Basin during the early monsoon. During this period, rising water levels greatly increase vector breeding in the principal rapids of the Niger and its tributaries which lie upwind and to the south‐west of the invasion sites in south‐eastern Mali. The Mali invasion ceases towards the height of the rainy season when many of these rapids are flooded. It was concluded that the very large breeding sites associated with the principal rapids in the Upper Niger Basin were the main sources of the S. sirbanum invading south‐eastern Mali.  相似文献

The wide natural incidence of Cherry leaf roll virus (CLRV) in deciduous forest trees and nurseries in northern Europe is believed to have occurred, apart from occasional mechanical spread and transmission through grafting, mainly by seed transmission. The mode of the vertical transmission and its role in the epidemiology of the virus has not been investigated, basically due to the inconvenient host-pathogen combinations studied to date. With the aim of obtaining an appropriate system for identification of viral genes and products participating in infection processes and seed transmission of CLRV, we performed infection and seed transmissibility tests with CLRV in Arabidopsis thaliana plants. Two phylogenetically and serologically different CLRV isolates were tested. Both of them were found able to infect A. thaliana plants, exhibited clear symptoms of the infection and spread systemically in the plants. Infection of the seeds and of a remarkable number of seedlings generated from infected seeds was possible for two consecutive generations. These results, for first time, report seed transmission of CLRV in the model plant A. thaliana and allow the assumption to be made of embryo invasion during seed transmission. Furthermore, first indications are given that genetically diverse CLRV isolates exhibit different abilities for vertical transmission in A. thaliana. The CLRV-A. thaliana model system is suitable for investigating viral invasion of developing plant organs and meristematic tissue, a prerequisite for successful virus dissemination via vertical transmission through seed.  相似文献
