The effect of fasting and refeeding on total antioxidant status (TAS), glutathione peroxidase (GSH-Px) activity and concentration of some non-enzymatic antioxidant compounds was studied in cockerels and pullets. Blood was collected before and after 48-h fasting and 24 h after refeeding. In cockerels, fasting resulted in a significant decrease of TAS and uric acid concentration. After refeeding, the concentration of TAS remained significantly lower as compared to the control level. At the same time, blood plasma level of total lipids increased in comparison to the control and post-fasting values. In pullets, fasting resulted in a significant decrease of whole blood haemolysate GSH-Px activity and blood plasma concentrations of albumin and uric acid. Simultaneously, a significant increase in total lipids and cholesterol was obtained. In pullets, refeeding resulted in a further decrease of TAS to undetectable values, a significant decrease of blood plasma cholesterol, and a significant increase of GSH-Px in the whole blood haemolysate and in blood plasma uric acid content. The results indicate that fasting has a negative impact on the antioxidant defence system of the blood, which leads to a reduced resistance to oxidative stress in both cockerels and pullets. However, pullets seem to be more susceptible to fasting-provoked oxidative stress than cockerels.  相似文献

The influence of fish oil (highly unsaturated) and beef tallow (highly saturated) with vitamin E (100 IU/kg) supplementation on the antioxidant status of broiler chicken cockerels was investigated. Chicks were fed a control diet with no added fat, 40 g/kg each of fish oil and beef tallow diets, respectively, from 11 to 42 days of age. Tocopherol concentration and the rate of lipid peroxidation, thiobarbituric acid reactive substance (TBARS) in liver, fatty acid composition of the liver lipids, blood serum total antioxidant status (TAS), and reduced glutathione (GSH) content were determined. Vitamin E supplementation of the diet increased liver alpha-tocopherol content in chicks regardless of the type of dietary fat. Fish oil diet resulted in higher liver TBARS value while beef tallow diet showed lower values compared to the control diet. Vitamin E supplementation reduced liver TBARS as well as serum GSH, and raised serum TAS for all diets. Serum GSH was the same for vitamin E supplemented diets regardless of the fat supplement. Fish oil diets resulted in a significant increase in hepatic lipid n-3 PUFA content. A significant positive correlation was found between liver TBARS and n-3 PUFA content. No relationships were established, however, between liver TBARS and n-6 PUFA or saturated fatty acids. The results suggest that feeding oils rich in n-3 PUFA increases tissue concentration of these fatty acids, consequently increasing tissue lipid peroxidation and reducing the antioxidative status of broiler chickens. Supplementing high levels of vitamin E with such oils may increase tissue oxidative stability. Serum TAS or GSH may be used as a measure of antioxidative status in chickens.  相似文献

We studied the influence of an embryonal application of antiestrogen tamoxifen on gonad differentiation and sex dimorphism during the growth of Slovgal chickens. A single application of 500 micrograms tamoxifen to the incubated eggs before the onset of gonad differentiation induced in pullets the growth of the right gonad, which usually develops only in cockerels. Comparing the size of both gonads it is clear that the above dose of antiestrogen had the 22.31% masculinizing effect. Morphological changes observed just after hatching correlated with the testosterone concentration in plasma. The pullets masculinized by tamoxifen had a significantly higher level of male hormone. The changes in sex differentiation were reflected in the altered growth of pullet bodies. With respect to the higher growth rate and more effective feed conversion, their growth can be appreciated as masculinized. The higher body weight at the end of seven-week fattening (4.25% in comparison with the control group of pullets) was due to the higher trunk weight. The growth was stimulated by tamoxifen application also in cockerels (5.57% increase in body weight) but it was not accompanied by any changes in feed conversion.  相似文献

1. Heterozygous cockerels which inherited the sex‐linked recessive dwarfing gene (dw) from a normal dam had significantly different contents of sodium and potassium in the blood serum compared with heterozygous cockerels from dwarf dams.

2. Hemizygous dwarf pullets had a significantly lower packed cell volume than normal pullets, while mean corpuscular haemoglobin concentration was significantly lower in the normal pullets.

3. Total serum calcium values were significantly higher in normal pullets. Sodium concentration was higher in dwarf pullets. It is suggested that the better adaptation of dwarf pullets to higher temperatures might be due to their capacity to retain more water in body tissue under a stressful environment.  相似文献

One hundred 6-week-old susceptible cockerels were inoculated with a pathogenic strain of infectious bursal disease virus (IBDV) and kept in the same pen as 100 each of 6-week-old pullets, local chickens and broilers. The cockerels developed depression and diarrhoea on day 3 post inoculation (PI) and most of the pullets and some of the local chickens and broilers showed similar signs on day 4 PI. Loss in weight was severe and similar in the pullets and local chickens, being significantly greater than that in the broilers from days 3–11 PI. The total mortality was 85%, 66.7%, 30% and 20% for the pullets, cockerels, local chickens and broilers, respectively. The lesions were more severe in the pullets and local chickens than in the broilers. IBDV antigen and antibody were detected, respectively, in all the bursal and serum samples from the infected chickens tested. The contact exposure method used in this study simulates better what happens in nature than inoculation with IBDV. The reduced mortality observed among the local chickens, compared with that (61.5%) seen in earlier studies using intraocular inoculation of IBDV, may have been due to behavioural differences that tend to result in their ingesting a relatively low dose of the virus.  相似文献

1. We examined the changes in plasma IGF-I concentration and tissue IGFBP-2 gene expression of young fasted chickens refed a commercial diet or administered bovine insulin intravenously. 2. Plasma IGF-I concentration was decreased by fasting for 2 d. Although plasma IGF-I concentration was increased by refeeding, it didn't recover to the level of chickens fed a commercial diet ad libitum. 3. Insulin administration lowered plasma IGF-I concentration compared to other groups. 4. Hepatic IGFBP-2 mRNA was increased by fasting for 2 d and decreased by refeeding for 6 h. Insulin administration also decreased hepatic IGFBP-2 gene expression stimulated by fasting to the level of refed chickens. 5. IGFBP-2 mRNA in the gizzard was increased by fasting for 2 d and tended to decrease after refeeding for 6 h. Insulin administration decreased gizzard IGFBP-2 gene expression to less than that in refed chickens. 6. There was no between-treatment difference in IGFBP-2 mRNA in the brain and kidney. 7. These results suggest that the changes in IGFBP-2 gene expression in the liver and gizzard by fasting and refeeding might be partly regulated by the alteration in plasma insulin concentration.  相似文献

1. We examined the influence of refeeding after 2 d of fasting on plasma insulin-like growth factor-I (IGF-I) concentration and hepatic IGF-I gene expression in chickens at 6 weeks of age. 2. Hepatic IGF-I mRNA was measured by ribonuclease protection assay and plasma IGF-I concentration was determined by radioimmunoassay. 3. Plasma IGF-I concentration decreased following fasting, increased to the level of fed controls after 2 h of refeeding but then fell back to the level of fasted chickens after 6 h of refeeding. 4. Fasting reduced hepatic IGF-I mRNA concentrations to less than half of those in the fed controls. Refeeding increased IGF-I mRNA sharply at 2 h after refeeding, but by 6 h after refeeding they had taller back again to levels significantly lower than at 2 h. 5. A significant correlation between plasma IGF-I concentration and hepatic IGF-I gene expression was found, suggesting that when chicks are refed after 2 d of fasting, the short-term increase in plasma IGF-I concentration may be partly regulated by the alteration in hepatic IGF-I mRNA.  相似文献

A model stress situation was created by nutrition deprivation. The experiment was performed with 130 cockerels of the initial breed White Leghorn. The experimental cockerels of the age of 18 to 24 days fasted for 24, 48 and 72 hours. Uric acid was determined in the supernatant of liver homogenate by means of a colorimetrically reductometric method with phospho-wolframic agent. A significant increase in the uric acid concentration in the liver of cockerels was found out during the model stress: the concentration of uric acid increased progressively from the initial 70.25 +/- 8.94 mg per 100 g to 93.15 +/- 9.66 mg per 100 g after 24-hour fasting, to 90.23 +/- 7.84 mg per 100 g after 48-hour fasting and even to 137.04 +/- 15.59 mg per 100 g after three-day fasting of 72 hours. After re-alimentation, the concentrations of uric acid were reduced to physiological values already within 24 hours.  相似文献

Serial blood samples were obtained from 16 Standardbred foals from time of birth to postpartum day 28. Sera were obtained and analyzed for gamma-glutamyltransferase (GGT), aspartate transaminase, and immunoglobulin (Ig) G. Presuckle colostrum from the respective mares of these foals was analyzed for GGT activity. Mean serum aspartate transaminase activities were significantly increased above presuckle values by postpartum hour 48 (P less than 0.01) and increased gradually over the first 14 days. Mean serum IgG concentrations were significantly greater than presuckle values by 5 hours after foals first suckled (P less than 0.01) and remained significantly increased during the 28-day sampling period. Serum GGT activity did not differ significantly over the period sampled. The SD were large, since there was a large degree of interindividual variation. Serum GGT activity in the foals was significantly increased over that in the mares throughout the period of the study. The profile of serum GGT activity over time in each foal did not show a pattern of change. There was no postsuckle increase in serum GGT activity nor a correlation between serum GGT activities and IgG concentrations at 24 hours after foals first suckled. Evidence was not obtained to support a colostric source of GGT involved in the increase of serum GGT activity in foals. Serum GGT activity seems to be increased in foals due to endogenous sources.  相似文献

本试验旨在研究高钙饲粮对青年蛋鸡血清一氧化氮(NO)浓度和抗氧化功能的影响。将 35日龄 100羽伊沙蛋鸡随机均分为对照组(含钙 1.00%)和高钙组(含钙 3.78%),每组 5个重复,每个重复 10羽。每天观察鸡的精神状态、食欲和粪便情况。在饲喂高钙饲粮第 0、8、16、24和 32天时,分别测定 2组鸡的血清中 NO浓度和丙二醛含量及抗氧化酶系统的变化。结果表明:1)高钙饲粮能引起青年蛋鸡长期腹泻,在饲喂高钙饲粮的第 8天就发生高血钙症。2)与对照组相比,高钙组在饲喂高钙饲粮第 8天和第 16天血清 NO浓度无显著变化(P>0.05),第24天和第 32天显著升高(P<0.05);第 8天和第 24天血清超氧化物歧化酶活性、总抗氧化能力无显著变化(P>0.05),第 16天显著升高(P<0.05),第 32天显著降低(P<0.05);第 8、16、24天血清丙二醛含量无显著变化(P>0.05),第 32天显著升高(P<0.05);第 24天和第 32天黄嘌呤氧化酶活性有升高趋势(P>0.05)。结果提示,饲喂高钙饲粮可导致青年蛋鸡发生长期腹泻,引起高血钙症,导致其机体内自由基的产生与清除的动态平衡遭到破坏,使自由基的产生远远超出清除能力。  相似文献

Male broiler chickens were killed at various times up to 36 h after withdrawal of food; hepatic and muscle glycogen concentrations and pH were compared with those of control birds given continuous access to food and water. 2. Liver glycogen was reduced to negligible concentrations (less than 1 mg/g) within 6 h of food deprivation and the initial pH was elevated. 3. Withdrawal of food had no effect on glycogen concentrations in the m. pectoralis superficialis (PS) but reduced them in the m. biceps femoris (BF); the reduction was significant after 12 h. 1. Plasma concentrations of luteinising hormone (LH), comb size and body weight were measured between 46 and 208 d of age in intact and castrated cockerels from lines of selected and control Australorp domestic chickens. The selected line had been selected for increased rate of lay by reducing oviposition intervals within sequences. The cockerels were reared and maintained on 15.25 h light/d. 2. Concentrations of plasma LH in the intact control cockerels were low in the 'immature' phase (less than 100 d old) and increased during the 'mature' phase (older than 175 d) with a peak occurring when the testes are beginning to grow at the onset of the 'mature' phase. In comparison with the control line of cockerels, the selected line had significantly higher plasma LH concentrations at the onset of the 'mature' phase. Selection had no effect on the mean concentration of plasma LH in either the immature phase or a few weeks after the onset of the mature phase. 3. There was no effect of selection in sibling females for higher rates of lay on changes in comb size or body weight during sexual maturation in the intact cockerels. 4. Castration at 45-46 d of age resulted in increased plasma LH concentrations and no comb growth. Plasma LH concentration increased progressively between 40-208 d. There was no difference between the two lines of cockerels in plasma LH concentration or rate of body growth after castration. 5. It is concluded that selection of females for a change in the rate of egg production has resulted in increased plasma LH concentrations in sibling males around the onset of sexual maturation. Selection appears to have caused this effect by altering an unidentified component of the inhibitory feedback mechanism which controls the tonic secretion of LH.  相似文献

1. Indirect calorimetry was used to study the effects on the energy metabolism of cockerels and hens after peak lay of a hybrid layer strain of regulating food or energy intake to 80% of ad libitum for an extended period of time. 2. Regulation of food intake decreased fasting and fed heat productions per bird and per unit metabolic body weight (kg0.75). 3. Maintenance energy requirements (per kg0.75) of both sexes were reduced about 20% by regulation; maintenance requirements of cockerels were about 30% lower per kg0.75 than hens on the same treatments. 4. Reducing the intake of all nutrients by 20% had the same effect on the energy metabolism of cockerels as restricting energy alone by the same amount. 5. Gross efficiency of egg production, in terms of both mass and energy, was higher when intake was regulated. During weeks 21 to 52 of restriction, daily egg mass output was significantly higher in hens fed ad libitum. 6. After 52 weeks of food regulation, restricted hens weighed 20% less and their total carcass energy was 36% less than hens fed ad libitum; similar effects were found in the cockerels.  相似文献

The change in the rate of protein synthesis of different muscles, concentrations of plasma insulin, plasma insulin-like growth factor-I (IGF-I) and other plasma components were investigated after refeeding in fasted chicks. 5.2 g of the complete diet was refed. This was the maximum that could be force-fed with water. 2. The fractional synthesis rates (FSR) of breast (M. pectoralis major) and leg (M. gastrocnemius) muscles were measured after injection of L-[2, 6-(3)H]phenylalanine. Plasma insulin and IGF-I concentration were determined by radioimmunoassay. 3. In the breast muscle, FSR was significantly reduced by 2-d fasting. The FSR had recovered completely after 1 h of refeeding and was maintained until 6 h. The change in FSR after refeeding was associated with the change in ribosomal efficiency (K(RNA); absolute synthesis rate per unit RNA), while no change in ribosomal capacity (C(S); RNA: protein ratio) was observed. 4. In the leg muscle, FSR was decreased by 2-d fasting and increased gradually toward 6 h after refeeding but did not reach the level of the fed control. In contrast to the breast muscle, no significant changes in Cs and K(RNA) in the leg muscle were observed. 5. Plasma glucose concentration increased significantly at 1 h after refeeding but returned to the fasted level after 24 h. Plasma insulin concentration in chicks refed for 1 h was higher than in the fasted group. There was no significant change in plasma IGF-I concentration. 6. These results suggest that the FSR of breast muscle was more sensitive to refeeding than that of leg muscle which may be explained, in part, by differences in sensitivity to the change in circulating plasma insulin concentration after refeeding.  相似文献

The effects of the 5‐HT1A receptor agonist 8‐OH‐DPAT on food and water intake in male broiler chickens were investigated. The injection of 25 or 50 μg/kg of 8‐OH‐DPAT 15 min before refeeding in fasted animals produced a decrease in food intake. No effect was observed in drinking. The injection of 25 or 50 μg/kg of the 8‐OH‐DPAT 60 min after the start of refeeding did not produce any significant modification in food intake. No effect on drinking was recorded. The agonist 8‐OH‐DPAT injected 15 min before water presentation in water‐deprived chickens, produced an increased drinking 60 min after the presentation of water. No effect on food intake was observed. The results show that the effect on food intake of the agonist 8‐OH‐DPAT in fasted–refed broiler chickens was similar to those observed in mammals and layer‐strain chickens. However, the agonist did not alter significantly the food intake when the broilers were fed 60 min before the injection. These results are contrary to the observed effects in mammals and in layer‐strain chickens. Probably, the selection for rapid growth rate in broilers causes modifications in the feeding control pattern. The comparison between broilers and layers strain may be a useful tool to elucidate the complex mechanisms involved in food and water intake regulation in chickens.  相似文献

The current study was designed to examine the effects of intracerebroventricular injections of SHU9119 [a nonselective melanocortin receptor (McR) antagonist] and MCL0020 (a selective McR antagonist) on the serotonin-induced eating and drinking responses of broiler cockerels deprived of food for 24 h (FD24). For Experiment 1, the chickens were intracerebroventricularly injected with 2.5, 5, and 10 µg serotonin. In Experiment 2, the chickens received 2 nmol SHU9119 before being injected with 10 µg serotonin. For Experiment 3, the chickens were given 10 µg serotonin after receiving 2 nmol MCL0020, and the level of food and water intake was determined 3 h post-injection. Results of this study showed that serotonin decreased food intake but increased water intake among the FD24 broiler cockerels and that these effects occurred in a dose-dependent manner. The inhibitory effect of serotonin on food intake was significantly attenuated by pretreatment with SHU9119 and MCL0020. However, the stimulatory effect of serotonin on water intake was not altered by this pretreatment. These results suggest that serotonin hypophagia and hyperdipsia were mediated by different mechanisms in the central nervous system, and that serotonin required downstream activation of McRs to promote hypophagia but not hyperdipsia in the FD24 chickens.  相似文献

Previous studies in avian species have reported time-dependent losses in muscle glycogen with prolonged feed withdrawal (FW). However, cervical dislocation was used to collect tissues, a method that results in significant involuntary muscle convulsions. In this study, cervical dislocation alone was found to reduce muscle glycogen by 23%, therefore, barbiturate overdose was used to collect tissue samples before and after FW, at the end of refeeding, and from continuously fed controls at each interval. Additionally, plasma samples from 6-wk-old male chickens were taken at the initiation and end of a 24-hr feed withdrawal, and at various times during refeeding. After 24 hr of FW, liver glycogen decreased markedly (77%; P < 0.05), whereas muscle glycogen decreased slightly and transiently, such that it returned to and remained at control levels, even after prolonged (72 hr) FW. Plasma glucose was decreased, whereas glucagon was elevated after a 24-hr feed withdrawal (P < 0.05), when compared with control concentrations. Muscle glycogen levels were not significantly increased over control levels after refeeding, but liver glycogen was increased by 380% (P < 0.05). Feed deprivation followed by refeeding resulted in increased circulating insulin and glucose levels when compared with control levels. Therefore, by using methods of tissue collection that ensure that muscle glycogen determinations are not confounded by artifactual degradation, these results verify that regulation of avian muscle glycogen stores is similar to that in mammals.  相似文献

Broiler chicken flocks were studied to determine the mortality from sudden death syndrome occurring in the flocks. The difference in the incidence of the syndrome in pullets and cockerels, and the age at which the most birds are affected were also studied. The weight of sudden death syndrome birds was compared with the flock average and the effect of continuous lighting as opposed to intermittent lighting was examined.The results suggest that; continuous lighting produces more sudden death syndrome deaths than intermittent lighting; that the incidence of sudden death syndrome is higher in cockerels than pullets; that the highest death rate occurred during the third and fourth weeks of life, and that sudden death syndrome birds on the average were heavier than the flock average.  相似文献
