Tropospheric ozone (O3) is considered harmful to agriculture production and soil community. Wheat cultivars HD 2987 (O3 sensitive) and Kharchiya 65 (O3 tolerant) were grown under ambient and elevated (ambient + 30 ppb) levels of O3 (EO) using open top chambers, and microbial biomass and enzymatic activities were investigated in soil rhizosphere. Elevated O3 declined soil enzymatic activities related to carbon cycling viz. β-glucosidase, cellobiohydrolase, cellulase and amylase more in sensitive cultivar compared to tolerant. Enzymatic activities linked to nitrogen cycling like N-acetyl-glucosaminidase and urease decreased while protease and glycine aminopeptidase increased. Microbial biomass carbon and nitrogen declined more in sensitive cultivar by 11.1 and 21.2%, respectively. Root biomass reduced in wheat cultivars with an increase of their phenolics contents by 34.3 and 10.2% in HD 2987 and Kharchiya 65, respectively at 60 days after germination. Non-significant changes were observed in soil organic carbon and total nitrogen in both the cultivars. Redundancy analysis suggested that soil enzymatic activities were predominantly affected by O3 induced changes in microbial biomass carbon and root biomass. Study also showed that rhizosphere of sensitive cultivar HD 2987 was affected more under EO as compared to tolerant Kharchiya 65.  相似文献

Tropospheric O3 (ozone) stress can negatively affect forest productivity and crop yields. Yet, relatively little attention has been paid to the effects of O3 stress on belowground system. Here, a pot experiment was conducted in open top chambers to monitor the response of physico-chemical properties, main microbial groups, and potential enzyme activities of a soil cropped to soybean (Glycine max; a highly sensitive species to O3) and exposed to background O3 concentration (45?±?5 ppb, control) and O3 stress (80?±?10 ppb, O3+ and 110?±?10 ppb, O3++) with sampling at branching, flowering, and podding stages. The growth of soybean was significantly inhibited by O3 stress, which showed significant effects on soil microbial biomass C and pH during the whole growth of soybean at the highest concentration. The O3++ stress significantly decreased soil pH at flowering stage, and increased soil pH at podding stage; the O3+ stress and growth stage?×?O3+ stress showed significant influences on the potential activities of acid phosphomonoesterase, invertase, and amylase. The O3 stress significantly reduced the abundances of total PLFAs (phospholipid fatty acid), bacterial PLFAs, and AMF (arbuscular mycorrhizal) PLFAs at branching and podding stages. Our results suggest that the main soil microbial groups might be indirectly affected by the O3 stress through the alteration of soil physico-chemical properties with changes in the potential enzyme activities of soil.  相似文献

Land‐use change and soil management play a vital role in influencing losses of soil carbon (C) by respiration. The aim of this experiment was to examine the impact of natural vegetation restoration and long‐term fertilization on the seasonal pattern of soil respiration and cumulative carbon dioxide (CO2) emission from a black soil of northeast China. Soil respiration rate fluctuated greatly during the growing season in grassland (GL), ranging from 278 to 1030 mg CO2 m?2 h?1 with an average of 606 mg CO2 m?2 h?1. By contrast, soil CO2 emission did not change in bareland (BL) as much as in GL. For cropland (CL), including three treatments [CK (no fertilizer application), nitrogen, phosphorus and potassium application (NPK), and NPK together with organic manure (OM)], soil CO2 emission gradually increased with the growth of maize after seedling with an increasing order of CK < NPM < OM, reaching a maximum on 17 August and declining thereafter. A highly significant exponential correlation was observed between soil temperature and soil CO2 emission for GL during the late growing season (from 3 August to 28 September) with Q10 = 2.46, which accounted for approximately 75% of emission variability. However, no correlation was found between the two parameters for BL and CL. Seasonal CO2 emission from rhizosphere soil changed in line with the overall soil respiration, which averaged 184, 407, and 584 mg CO2 m?2 h?1, with peaks at 614, 1260, and 1770 mg CO2 m?2 h?1 for CK, NPK, and OM, respectively. SOM‐derived CO2 emission of root free‐soil, including basal soil respiration and plant residue–derived microbial decomposition, averaged 132, 132, and 136 mg CO2 m?2 h?1, respectively, showing no difference for the three CL treatments. Cumulative soil CO2 emissions decreased in the order OM > GL > NPK > CK > BL. The cumulative rhizosphere‐derived CO2 emissions during the growing season of maize in cropland accounted for about 67, 74, and 80% of the overall CO2 emissions for CK, NPK, and OM, respectively. Cumulative CO2 emissions were found to significantly correlate with SOC stocks (r = 0.92, n = 5, P < 0.05) as well as with SOC concentration (r = 0.97, n = 5, P < 0.01). We concluded that natural vegetation restoration and long‐term application of organic manure substantially increased C sequestration into soil rather than C losses for the black soil. These results are of great significance to properly manage black soil as a large C pool in northeast China.  相似文献

Purpose

Rhizosphere and fertilization might affect soil microbial activities, biomass, and community. This study aimed to evaluate the impacts of Phyllostachys edulis (moso bamboo) rhizospheres on soil nutrient contents and microbial properties in a moso bamboo forest with different fertilizer applications and to link soil microbial activities with abiotic and biotic factors.

Materials and methods

The experiment included three treatments: (1) application of 45% slag fertilizer (45%-SF); (2) application of special compound fertilizer for bamboos (SCF); and (3) the control without any fertilizer application (CK). Simultaneously, bulk soils and 0.5, 2.5, 4.5, and 6.5-year-old (y) bamboo rhizosphere soils were selected. Soil nutrient contents were analyzed. Microbial activities were evaluated based on the activities of soil enzymes including β-glucosidase, urease, protease, phosphatase, and catalase. The total microbial biomass and community were assessed with the phospholipid fatty acids (PLFAs) method.

Results and discussion

In the CK and SCF treatments, organic matter contents of rhizosphere soils were significantly higher than those of bulk soils. Soil β-glucosidase, urease, protease, phosphatase, and catalase activities in rhizosphere soils were higher than those of bulk soils, with the sole exception of β-glucosidase of 0.5 y rhizosphere soil in the 45%-SF treatment. Compared with the CK treatment, fertilizer applications tended to increase soil total PLFAs contents and changed soil microbial community. Moso bamboo rhizospheres did not significantly increase the total microbial biomass. In the SCF treatment, the Shannon index of bulk soil was significantly lower than those of rhizosphere soils.

Conclusions

Our results suggested that both rhizospheres and fertilizer applications could change the soil microbial community structures and that moso bamboo rhizosphere could increase microbial activity rather than biomass in the forest soils with different fertilizer applications.

Biochar application to arable soils could be effective for soil C sequestration and mitigation of greenhouse gas (GHG) emissions. Soil microorganisms and fauna are the major contributors to GHG emissions from soil, but their interactions with biochar are poorly understood. We investigated the effects of biochar and its interaction with earthworms on soil microbial activity, abundance, and community composition in an incubation experiment with an arable soil with and without N-rich litter addition. After 37 days of incubation, biochar significantly reduced CO2 (up to 43 %) and N2O (up to 42 %), as well as NH4 +-N and NO3 ?-N concentrations, compared to the control soils. Concurrently, in the treatments with litter, biochar increased microbial biomass and the soil microbial community composition shifted to higher fungal-to-bacterial ratios. Without litter, all microbial groups were positively affected by biochar × earthworm interactions suggesting better living conditions for soil microorganisms in biochar-containing cast aggregates after the earthworm gut passage. However, assimilation of biochar-C by earthworms was negligible, indicating no direct benefit for the earthworms from biochar uptake. Biochar strongly reduced the metabolic quotient qCO2 and suppressed the degradation of native SOC, resulting in large negative priming effects (up to 68 %). We conclude that the biochar amendment altered microbial activity, abundance, and community composition, inducing a more efficient microbial community with reduced emissions of CO2 and N2O. Earthworms affected soil microorganisms only in the presence of biochar, highlighting the need for further research on the interactions of biochar with soil fauna.  相似文献

为了明确不同氮素浓度和水分对土壤硝化作用和微生物特性的影响,特别是高氮素浓度下的响应特异性,以红壤水稻土为供试土壤,设置4个硫铵用量水平[0(CK)、120 mg(N).kg-1(A1)、600 mg(N).kg-1(A2)、1 200 mg(N).kg-1(A3)],调节土壤水分为饱和持水量(WHC)的40%、60%和80%,研究了短期内不同氮素浓度和不同水分条件下土壤硝化作用、微生物生物量碳和微生物功能多样性的变化。结果表明:在40%、60%和80%WHC水分条件时,硫铵A2、A3浓度处理土壤硝化率和硝化速率普遍较低,硫铵A1浓度处理硝化率和硝化速率随土壤含水量的升高而升高;同含水量时随硫铵用量的升高而显著降低。在40%、60%和80%WHC水分条件时,微生物生物量碳随硫铵浓度的升高而降低;同浓度硫铵用量水平时,微生物生物量碳的变化基本表现为:60%WHC80%WHC40%WHC。分析发现不同水分和硫铵处理之间存在交互作用。BIOLOG分析显示:不同氮素浓度和不同水分处理,60%WHC下A1处理的平均吸光值(AWCD)和Shannon、Simpson、McIntosh指数最大,其次为60%WHC的硫铵CK处理,而不同水分下硫铵A2、A3处理,其AWCD值和Shannon、Simpson、McIntosh多样性指数都较低,进一步说明过量施肥导致微生物活性降低。不同氮素浓度和水分条件下土壤微生物和生化性状不同,过量施用化肥后将有可能造成土壤微生物性状和生化功能衰减。  相似文献

Purpose

Evergreen broad-leaved forest ecosystems are common in east China, where they are both ecologically and economically important. However, nitrogen (N) addition over many years has had a detrimental effect on these ecosystems. The objective of this research was to evaluate the effect of 4 years of N addition on microbial communities in an evergreen broad-leaved forest in southern Anhui, China.

Materials and methods

Allochthonous N in the form of aqueous NH4NO3 and phosphorus (P) in the form of Ca(H2PO4)2·H2O were applied at three doses with a control (CK, stream water only without fertilizer): low-N (50 kg N ha?1 year?1), high-N (100 kg N ha?1 year?1) and high-N+P (100 kg N ha?1 year?1 + 50 kg P ha?1 year?1). Quantitative PCR analysis of microbial community size and Illumina platform-based sequencing analysis of the V3-V4 16S rRNA gene region were performed to characterize soil bacterial community abundance, structure, and diversity.

Results and discussion

Bacterial diversity was increased in low-N and high-N treatments and decreased in the high-N+P treatment, but α-diversity indices were not significantly affected by N additions. Proteobacteria, Acidobacteria, and Actinobacteria were the predominant phyla in all treatments, and the relative abundance of different genera varied among treatments. Only soil pH (P = 0.051) showed a weak correlation with the bacterial community in CK and low-N treatment.

Conclusions

The composition of the bacterial community and the abundance of different phyla were significantly altered by N addition. The results of the present study indicate that soil bacterial communities in subtropical evergreen broad-leaved forest are, to a certain extent, resilient to changes derived from N additions.

Purpose

Changes of nitrogen (N) cycle caused by N fertilization and precipitation regimes have affected the key ecosystem structure and functions in temperate steppe, which may modify the structure of soil microbial communities involved in N transformation. This paper was designated to examine the response of soil ammonia oxidizers and denitrifiers to the N fertilization and precipitation regimes in a semi-arid steppe where N and water contents are major limiting factors of the grassland productivity.

Materials and methods

This study was based on a long-term N fertilization and precipitation regimes experiment in Inner Mongolia (116° 17′ 20″ E, 42° 2′ 29″ N). The treatments including CK (control), R (reduced precipitation), W (30% increase in precipitation), N (10 g N m?2 y?1), RN (reduced precipitation and 10 g N m?2 y?1), and WN (30% increase in precipitation and 10 g N m?2 y?1). Soil basic chemical properties and microbial activities were analyzed. Molecular methods were applied to determine the abundance, structure and diversity of ammonia oxidizers and denitrifiers. Statistical analysis detected the main and interactive effect of treatments on soil microbial communities and revealed the relationship between soil microbial community structures and environmental factors.

Results and discussion

N fertilization significantly increased ammonia-oxidizing bacteria (AOB) abundance. Ammonia-oxidizing archaea (AOA) community structure was markedly changed in N fertilizer treatment and strongly affected by soil pH, while soil nitrate and water content correlated with AOB community structure. Soil nitrate was the key factor influencing nirK gene community structure, while soil pH and water content explained much of the variations of nosZ gene community. AOB-amoA and nosZ gene community diversities were influenced by precipitation regimes and interaction of N fertilization and precipitation regimes, respectively.

Conclusions

N fertilization and precipitation regimes had significant influences on the changes of soil properties and microbial functional communities. Soil nitrification was mainly driven by AOB in the semi-arid grassland. Changes of substrate content and soil pH were the key factors in shifting functional microbial communities. The non-synergistic effects of N fertilization and precipitation regimes on the microbial functional groups indicated that the negative effect of lower pH induced by N fertilization would be alleviated by precipitation regimes, which should be well considered in grassland restoration.

Extreme flood events are predicted to have a negative impact on soil quality. Currently, there is a lack of information about the effect of agricultural practices on soil functioning and microbial processes under these events. We hypothesized that the impact of flooding on soil quality will be exacerbated when crop residues are present in the soil as they will induce more extreme anaerobicity. A spring extreme flood event (10 °C, 9 weeks) was simulated in mesocosms containing an arable sandy-loam soil low in nutrients. The main treatments were (1) with and without flooding and (2) with and without maize residue addition (8 Mg ha?1). We monitored changes in soil chemical quality indicators (e.g. pH, salinity, Fe3+, P, C, NH4 +, NO3 ? and organic N), greenhouse gas (GHG) emissions (CO2, CH4, N2O) and soil microbial community composition (PLFAs) during a prolonged flood period (9 weeks) and an 8-week “recovery” period after flooding. In comparison to the other treatments, flooding in the presence of crop residues resulted in a dramatic drop in soil redox potential. This was associated with the enhanced release of Fe and C into solution and an increase in CH4 emissions. In contrast, maize residues reduced potential nitrate losses and N2O emissions, possibly due to complete denitrification and microbial N immobilization. Both flooding and maize residues stimulated microbial growth and promoted a shift in microbial community composition. Following floodwater removal, most of the soil quality indicators returned to the levels of the control treatment within 5 weeks. After this short recovery phase, no major impact of flooding could be observed on plant growth (maize pot-grown). Overall, we conclude that both extreme flooding and management regime negatively impact upon a range of soil quality indicators (e.g. redox, GHG emissions); however, the soil showed high resilience and recovered quickly after floodwater removal. Further work is required to investigate the impact of repeated extreme flood events on soil quality and function over longer timescales.  相似文献

A greenhouse experiment was performed to evaluate effectiveness of goosegrass (Eleusine indica) in phytoremediation of soil contaminated with 8,247 mg kg?1 of total petroleum hydrocarbons (TPH). We determined seed germination toxicity, soil microbial viable counts, catalase activity, dehydrogenase activity (DHA), and the concentrations of TPH and 14 polycyclic aromatic hydrocarbons (PAHs) in soil and plant tissue. After 5 months, the initial level of contamination was reduced by 47% in planted soil, whereas it was only reduced by 11% in nonplanted soil. Bacterial numbers were 72 times greater in the rhizosphere treated soil than in the unvegetated treatment at the end of the study. There was no correlation between microbial counts or DHA and catalase activity, and the correlation between microbial counts and DHA was weak. Significant chemical reduction of H2O2, caused by the soil fabric, was observed in the determination of catalase activity. In case of vegetated treatment, 32% of PAHs was removed, but only 5% of PAHs was dissipated in the unvegetated pots. Gas chromatography/mass spectrometry analysis of plant tissue indicated that a low amount of PAHs (25.50 mg kg?1 dry biomass) was detected in goosegrass roots growing in the contaminated soil, and no uptake into the shoots was occurring.  相似文献

The effects of root activity on microbial response to cadmium (Cd) loading in the rhizosphere are not well understood. A pot experiment in greenhouse was conducted to investigate the effects of low Cd loading and root activity on microbial biomass and community structure in the rhizosphere of pakchoi (Brassica chinensis L.) on silty clay loam and silt loamy soil. Cd was added into soil as Cd(NO3)2 to reach concentrations ranging from 0.00 to 7.00 mg kg-1. The microbial biomass carbon (MBC) and community structure were affected by Cd concentration, root activity, and soil type. Lower Cd loading rates (〈 1.00 mg kg-1) stimulated the growth of pakchoi and microorganisms, but higher Cd concentrations inhibited the growth of microorganisms. The content of phospholipid fatty acids (PLFAs) was sensitive to increased Cd levels. MBC was linearly correlated with the total PLFAs. The content of general PLFAs in the fungi was positively correlated with the available Cd in the soil, whereas those in the bacteria and actinomycetes were negatively correlated with the available Cd in the soil. These results indicated that fungi were more resistant to Cd stress than bacteria or actinomycetes, and the latter was the most sensitive to Cd stress. Microbial biomass was more abundant in the rhizosphere than in the bulk soil. Root activity enhanced the growth of microorganisms and stabilized the microbial community structure in the rhizosphere. PLFA analysis was proven to be sensitive in detecting changes in the soil microbial community in response to Cd stress and root activity.  相似文献

为探明使用沼液对土壤质量的影响,明确土壤微生物功能多样性对沼液使用量和使用年限的响应,在甘肃祁连葡萄观光园进行等氮量替代和增施沼液处理,采用Biolog方法测定土壤微生物功能多样性。结果表明:(1)100%等氮量沼液处理的土壤微生物群落代谢活性最小,替代33%氮量处理的土壤微生物群落代谢活性最高,连续两年使用沼液的处理间差异大于一年。(2)试验地土壤微生物利用的主要碳源为碳水化合物类,使用沼液能够显著提高土壤微生物对氨基酸类和羧酸类碳源的利用率。连续两年使用沼液条件下,增施沼液处理(133%和167%)效果优于等氮量替代处理。(3)与单施化肥对照相比,使用沼液处理仅在提高土壤微生物均一度指数方面存在优势,且增施沼液处理(133%和167%)效果优于等氮量替代处理。(4)主成分分析表明,沼液使用量和使用年限对土壤微生物碳源利用水平以及代谢多样性的影响有差异,且使用年限增加能使各处理间变异程度增大。(5)碳水化合物类和氨基酸类碳源是引起不同沼液用量处理土壤微生物功能群分异的主要碳源,其中碳水化合物类是使用年限增加后土壤微生物群落变化的敏感碳源。(6)Pearson相关性分析表明,该试验地土壤微生物功能多样性与土壤有机质、碱解氮呈正相关关系,与速效磷、速效钾呈负相关关系。综上,长期使用沼液(尤其是沼液与化肥配施)有利于提高土壤微生物代谢活性和土壤微生物群落功能多样性,且当沼液使用量较大时能达到完全取代化肥的效果。  相似文献

The soil physicochemical properties, soil denitrification rates (PDR), denitrifiers via nitrite reductases (nirK and nirS) and nitrous oxide reductase (nosZ), abundance and community composition of denitrifiers in both the rhizosphere and bulk soil from a long-term (32 year) fertilizer field experiment conducted during late rice season were investigated by using the MiSeq sequencing, quantitative PCR, terminal restriction fragment polymorphism (T-RFLP). The experiment including four treatments: without fertilizer input (CK), chemical fertilizer alone (MF), rice straw residue and chemical fertilizer (RF), and organic manure and chemical fertilizer (OM). The results showed that the application of rice straw residue and organic manure increased soil organic carbon (C), total nitrogen (N), and NH4+-N contents. The nirK, nirS, and nosZ copy numbers with OM and RF treatments were significant higher than that of the MF and CK treatments in the rhizosphere and bulk soil (p < 0.05). The principal coordinate analysis (PCoA) analysis showed that the different parts of root zone are the most important factors for the variation of denitrifying bacteria community, and the different fertilization treatments is the second important factors for the variation of denitrifying bacteria community. The MiSeq sequencing result showed that nirK, nirS and nosZ-type denitrifiers communities within bulk soil had lower species diversity compared with rhizosphere soil, and were dominated by Rhizobiales, Rhodobacterales, Burkholderiales, and Pseudomonadales. As a result, the application of fertilization practices had significant effects on soil N and PDR levels, and affected the abundance and community composition of N-functional microbes.  相似文献

With the advent of glyphosate [N-(phosphonomethyl)glycine] tolerant crops, soils have now been receiving repeated applications of the herbicide for over 10 years in the Midwestern USA. There is evidence that long-term use of glyphosate can cause micronutrient deficiency but little is known about plant potassium (K) uptake interactions with glyphosate. The repeated use of glyphosate may create a selection pressure in soil microbial communities that could affect soil K dynamics and ultimately K availability for crops. Therefore, the objectives of this study were to characterize the effect of foliar glyphosate applied to GR (glyphosate resistant) soybeans on: (1) rhizosphere microbial community profiles using ester linked fatty acid methyl ester (EL-FAME) biomarkers, (2) exchangeable, non-exchangeable, and microbial K in the rhizosphere soil, and (3) concentrations of soybean leaf K. A greenhouse study was conducted in a 2 × 2 × 3 factorial design with two soil treatments (with or without long-term field applications of glyphosate), two plant treatments (presence and absence of soybean plants), and three rates of glyphosate treatments (0×, 1× at 0.87, and 2× at 1.74 kg ae ha?1, the recommended field rate). After each glyphosate application, rhizosphere soils were sampled and analyzed for microbial community structure using ester linked fatty acid methyl ester biomarkers (EL-FAME), and exchangeable, plant tissue and microbial biomass K. Glyphosate application caused a significant decrease in the total microbial biomass in soybean rhizosphere soil that had no previous exposure to glyphosate, at 7 days after glyphosate application. However, no significant changes were observed in the overall microbial community structure. In conclusion, the glyphosate application lowered the total microbial biomass in the GR soybean rhizosphere soil that had no previous exposure to glyphosate, at 7 days after glyphosate application; caused no changes in the microbial community structure; and did not reduce the plant available K (soil exchangeable or plant tissue K).  相似文献

There is increasing evidence that microorganisms participate in soil C sequestration and stabilization in the form of resistant microbial residues. The type of fertilizers influences microbial activity and community composition; however, little is known about its effect on the microbial residues and their relative contribution to soil C storage. The aim of this study was to investigate the long-term impact (21 years) of different fertilizer treatments (chemical fertilizer, crop straw, and organic manure) on microbial residues in a silty clay loam soil (Udolls, USDA Soil Taxonomy). Amino sugars were used to indicate the presence and origin of microbial residues. The five treatments were: CK, unfertilized control; NPK, chemical fertilizer NPK; NPKS1, NPK plus crop straw; NPKS2, NPK plus double amounts of straw; and NPKM, NPK plus pig manure. Long-term application of inorganic fertilizers and organic amendments increased the total amino sugar concentrations (4.4–8.4 %) as compared with the control; and this effect was more evident in the plots that continuously received pig manure (P?<?0.05). The increase in total amino sugar stock was less pronounced in the straw-treated plots than the NPKM. These results indicate that the accumulation of soil amino sugars is largely influenced by the type of organic fertilizers entering the soil. Individual amino sugar enrichment in soil organic carbon was differentially influenced by the various fertilizer treatments, with a preferential accumulation of bacterial-derived amino sugars compared with fungal-derived glucosamine in manured soil.  相似文献

Photosynthetically derived rhizodeposits are an important source of carbon (C) for microbes in root vicinity and can influence the microbial community dynamics. Pulse labeling of carbon dioxide (13CO2) coupled with stable isotope probing techniques have potential to track recently fixed photosynthate into rhizosphere microbial taxa. Therefore, the present investigation assessed the microbial community change associated with the rhizosphere and bulk soil in Jatropha curcas L. (a biofuel crop) by combining phospholipid fatty acid (13C-PLFA) profiling using a stable isotope 13CO2 labeling approach. The labeling (13C) took place after 45 days of germination, PLFAs were extracted from both soils (rhizosphere and bulk) after 1 and 20 days pulse labeling and analyzed by gas chromatography-isotope ratio mass spectrometry. There was no significant temporal effect on the PLFA profiles in the bulk soil, but significantly increased abundance of Gram positive (i15:0) and Gram negative (16:1ω7c and 16:1ω5c) biomarkers was observed in the rhizosphere soil from day 1 to day 20 after labeling. The Gram negative (16:1ω7c) decreased and fungal (18:2ω6,9c) increased significantly in rhizospheric soil compared to bulk soil after day 1 of labeling. Whereas, after 20 days of labeling, the Gram negative biomarker (16:1ω7c and 18:1ω7c) decreased and Gram positive (a15:0) increased significantly in rhizospheric soil compared to bulk soil. One day following labeling, i15:0, a15:0, i16:0, 16:1ω5c, 16:0, i17:0, a17:0, 18:2ω6,9c, 18:1ω9c, and 18:0 PLFAs were significantly more enriched in δ13C in the rhizosphere than in the bulk soil. Twenty days after labeling, 16:1ω5c (Gram negative) and 18:2ω6,9c (fungal) were significantly more enriched in δ13C in the rhizosphere than in the bulk soil. These results shows the effectives of PLFA coupled using the pulse chase labeling technique to examine the microbial community changes in response to recently fixed photosynthetic C flow in rhizodeposits.  相似文献

Bacterial wilt caused by Ralstonia solanacearum is one of the most serious tobacco diseases worldwide. Brevibacillus brevis (L-25) and Streptomyces rochei (L-9) with strong inhibitory effects on R. solanacearum in vitro were isolated from the rhizosphere of a healthy tobacco plant in a severely wilt-diseased field. Pot and field experiments were conducted to evaluate the biocontrol effect of the isolated antagonists alone and in combination with organic fertilizer. In pot experiment, the control efficacy was 92.3–100 % in the treatments applied with L-25 and L-9 alone or together with organic fertilizers. When bioorganic fertilizer containing L-9 and L-25 was applied to the soil in field condition, the control efficacies were 95.4 and 30.0 in the Anhui and Guizhou field plots, respectively. The counts of bacteria and actinomycetes in rhizosphere soil were significantly increased (p?≤?0.05) under all antagonist applications compared with CK (PR). In contrast, fungal and R. solanacearum densities in the rhizosphere soil applied with antagonists were much lower than the CK (PR) rhizosphere. Combined application of the two antagonists had better effect than single antagonist treatments. The antagonists were more effective when they were combined with organic fertilizer as compared with the antagonistic strains only. These results allow us to conclude that a combination of the biocontrol agents, L-25 and L-9, together with organic fertilizers can effectively control bacterial wilt by affecting soil microbial structure.  相似文献

Purpose

Chlorothalonil (CTN) has received much attention due to its broad-spectrum antifungal function and repeated applications in agriculture production practice. An incubation experiment was conducted to study the accumulating effects of CTN repeated application on soil microbial activities, biomass, and community and to contrast the discrepancy of effects in contrasting soils.

Materials and methods

Different dosage CTN (5 mg kg?1, T1, and 25 mg kg?1, T5) was applied into two contrasting soils at 7-day intervals. Soil samples were taken 7 days after each application to assess soil enzyme activities and gene abundances. At the end of incubation, the soil samples were also taken to analyze microbial communities in the two test soils.

Results and discussion

Soil fluorescein diacetate hydrolysis (FDAH) and urease activities were inhibited by CTN repeated applications. After 28 days of incubation, bacterial 16S rRNA gene abundances in T1 and T5 treatments were significantly lower than those in the CK treatments (46.4 and 36.6 % of the CK treatment in acidic red soil, 53.6 and 37.9 % of the CK treatment in paddy soil). Archaeal 16S rRNA gene abundances of T1 and T5 treatments were observed the similar trends (56.1 and 40.8 % of the CK treatment in acidic red soil, 45.6 and 43.7 % of the CK treatment in paddy soil). Repeated applications at 25 mg kg?1 exerted significantly negative effects on the Shannon-Weaver, Simpson and McIntosh indices.

Conclusions

Microbial activity, biomass, and functional diversity were significantly inhibited by repeated CTN application at the higher dosage (25 mg kg?1), but the inhibitory effects by the application at the recommended dosage (5 mg kg?1) were erratic. More emphasis needs to be placed on the soil type and cumulative toxicity from repeated CTN application when assessing environmental risk.

The effect of an elevated concentration of atmospheric CO2 and the application rate of nitrogen fertilizers on the microbial biomass and maximum specific growth rate of microorganisms in the soil and rhizosphere was studied in a long-term field experiment involving the growing of sugar beets and winter wheat. It was shown that the treatment of field plots with carbon dioxide at a concentration higher than that in the atmosphere (550 ppm) for three-four years resulted in the formation of a microbial community with a higher maximum specific growth rate and a larger share of R-strategy microorganisms as compared to the soil under the control plants. No reliable differences in the total microbial biomass in the soil under the winter wheat were revealed between the treatments with the ambient and elevated CO2 concentrations; in the soil under the beet plants, a reliable increase in the total microbial biomass at the elevated CO2 concentration was noted only in the close vicinity of the plant roots.  相似文献

通过2年定位试验研究了有机无机肥料配施对南方旱坡地果园柑桔(沙糖桔)产量和品质的影响,并比较了土壤可培养微生物数量、土壤微生物碳、氮、柑桔根际土壤活性铝含量及柑桔根系铝含量的变化。结果表明,与100%化肥处理(CK)相比,20%有机肥+80%化肥(T1)、40%有机肥+60%化肥(T2)、100%有机肥(T3)处理的细菌、放线菌、真菌数量均显著提高,平均分别是对照的1.30、1.40、1.46倍;土壤微生物量碳、氮显著增加,其中T3处理的土壤微生物量碳、氮分别增加45.6%6、3.3%。施用有机肥后,活跃了土壤中的微生物种群,改善了土壤的生物学质量。施用有机肥可缓解旱坡地柑桔园根际土壤铝毒的危害,显著减少柑桔根系对土壤铝的吸收累积。尤其是T3处理,根际土壤活性铝含量较CK有大幅度的降低,降幅达到77.4%。施用有机肥明显地促进了柑桔根系的生长发育,提高果实产量和品质,沙糖桔平均增产率18.5%,增产效果显著。  相似文献
