以异育银鲫(Carassius auratus gibelio)幼苗[(136.70±7.98)g]为实验对象,监测4种环境源性胁迫(高pH 9.2、低溶氧2 mg·L~(-1)、高亚氮2 mg·L~(-1)和高氨氮4 mg·L~(-1))下第3、7、10、15天血浆内皮质醇(COR)、葡萄糖(GLU)、I型干扰素(IFN)、白细胞介素-2(IL-2)、超氧化物歧化酶(SOD)、免疫球蛋白M(Ig M)、补体3(C3)以及丙二醛(MDA)的变化规律。结果显示:COR经高pH、高亚氮和高氨氮胁迫时显著升高,而SOD显著降低,Ig M经高亚氮和高氨氮胁迫时亦显著升高,表明COR、SOD和Ig M可能是广源性的胁迫状态指示指标,COR和Ig M的升高以及SOD活力的降低指示异育银鲫可能处于被胁迫状态。GLU、IFN和C3只经高氨氮胁迫时显著升高,且响应幅度剧烈,表明GLU、IFN和C3可能是灵敏且狭源性的应激指示指标,高水平指示机体可能处于氨氮应激状态。IFN和MDA经高亚氮和高氨氮胁迫时,表现出不同的响应趋势:氨氮胁迫时IFN和MDA显著升高,但亚氮胁迫时则显著降低,表明IFN和MDA响应模式可能具有特异性,不适合指示综合应激状态。  相似文献

洪湖碘泡虫(Myxobolus honghuensis)是引起异育银鲫(Carassius auratus gibelio Bloch)“喉孢子病”的重要病原, 每年导致养殖苗种和成鱼大量死亡。本研究通过隐性感染异育银鲫母本人工受精、实验室条件下受精卵孵化和幼鱼培育, 采用单管半巢式 PCR、荧光定量 PCR 和寡核苷酸荧光原位杂交等检测手段进行亲本、卵和幼鱼等环节的检测分析, 探究异育银鲫寄生洪湖碘泡虫是否存在经卵传播途径。结果表明, 所采用的 34 尾异育银鲫母本(A1~A22, B1~B12)的洪湖碘泡虫隐性感染率达 50%~75%, 其中, 卵和伪鳃检出率高于卵巢组织样品; 特异性寡核苷酸探针荧光原位杂交在隐性感染母本的卵巢、伪鳃、肾、脾组织检测到洪湖碘泡虫前孢子生成阶段营养体; 实验室条件下阳性母本所产的卵经孵化和培育出的幼鱼 15 dph 和 30 dph 样品可以检出阳性(A1、A18、B8 和 B9); 荧光原位杂交显示 15 dph 幼鱼在伪鳃、鳃和肾脏组织检测出阳性信号。本研究进一步揭示了异育银鲫寄生洪湖碘泡虫存在经鱼卵传播途径; 研究结果可为相关疾病制定防控措施奠定重要的理论基础。  相似文献

Two 8‐week feeding trials were conducted to evaluate dietary carbohydrate utilization by omnivorous gibel carp (Carassius auratus gibelio) (2.4 ± 0.1 g) and herbivorous grass carp (Ctenopharyngodon idellus) (6.5 ± 0.1 g). Five isonitrogenous (370 g kg?1) and isolipid (70 g kg?1) diets were formulated with increasing corn starch levels (60, 140, 220, 300 and 380 g kg?1). Results showed that specific growth rate (SGR), feed efficiency (FE) and protein retention efficiency (PRE) of gibel carp significantly increased from dietary starch of 60 to 300 g kg?1 and then decreased from 300 to 380 g kg?1, but those of grass carp showed no significant differences between treatments. Independent of dietary starch levels, grass carp gained significantly higher FE and PRE than gibel carp. Feeding rate (FR) of gibel carp was significantly higher than that of grass carp. In two fish species, high dietary starch (300 and 380 g kg?1) tended to obtain higher hepatosomatic index (HSI), serum triglyceride, hepatic lipid and body lipid contents. Serum glucose concentration of grass carp was not affected, while that of gibel carp fed the starch of 300 g kg?1 diet was significantly lower than those of the fish fed other four diets (60, 140, 220 and 380 g kg?1). Grass carp showed high tolerance to dietary starch while dietary corn starch should be no more than 300 g kg?1 for gibel carp. High starch contents may cause lipid accumulation in the liver and body.  相似文献

To investigate the response of gibel carp to dietary lysine levels in zero fish meal (FM) diets, 8 experimental diets were designed. Among them, diet 1 and diet 2 with 150 g/kg FM, methionine, lysine and threonine were supplemented in diet 2. Diet 3 was without FM but matched the amino acid profile of diet 2 except for lysine. Diets 4–8 were based on diet 3, with graded levels of lysine. After 8‐week feeding trail, final body weight, weight gain (WG), specific growth rate, feed conversion ratio (FCR) and feed intake were significantly improved by diets 1–2 and 6–8 compared with diet 3. Plasma glucose, triglyceride, immunoglobulin G and aspartate aminotransferase levels in diets 4–8 were higher than those in diet 1. Apparent digestibility coefficients (ADCs) of protein, energy, methionine, lysine, valine, histidine and phenylalanine in diet 1 were significantly lower than those in diet 6. The highest ADC of isoleucine was found in diet 3. TOR, IGF‐1, JAK3, STAT4, STAT6 and PepT2 mRNA levels were significantly increased with increasing dietary lysine up to a certain level in zero FM diet and lower than diets 1–2. Based on WG and FCR, the optimal lysine requirement of juvenile gibel carp was determined to be 24.4 g/kg and 24.2 g/kg (65.9 g/kg and 65.4 g/kg of dietary protein) in zero FM diet, respectively, using quadratic regression analysis.  相似文献

A 12‐week feeding trial was carried out in fibreglass tanks to examine partial replacement of fish meal (FM) with poultry by‐product meal (PBM), meat and bone meal (MBM) and blood meal (BM) in practical diets for gibel carp Carassius auratus gibelio (Bloch). Triplicate groups of fish (initial body weight 15.3 g fish?1) were fed eight isonitrogenous (crude protein: 37.5%) and isolipidic diets (crude lipid: 7%). The control diet is the commercial diet of gibel carp, which used 18% of FM as the sole animal protein source. In the other seven diets (Diet 2–Diet 8), 17–83% of FM protein was substituted by a blend of PBM and BM or a blend of PBM, MBM and BM. The final body weight and thermal‐unit growth coefficient of fish fed the feeds in Diet 8 in which 83% of FM protein was replaced by the blend of 3% BM, 10% PBM and 5% MBM is significantly lower than Diet 1 (the control). The feed efficiency ratio in Diet 8 group is significantly lower than Diet 1, 2, 4 and 7 groups. The results of the present study indicated that a combination of PBM, BM and MBM can replace most of the FM protein and the FM level can be reduced to about 6% with satisfactory growth and feed utilization in practical diets for gibel carp.  相似文献

A 10‐week feeding trial was conducted in a flow‐through system to determine dietary choline requirement for juvenile gibel carp (Carassius auratus gibelio) (5.5 ± 0.1 g). Purified basal diet was formulated using vitamin‐free casein as protein source. Choline chloride was supplemented to the basal diet to formulate seven diets containing 76.1, 163, 356, 969, 1457, 2024 and 4400 mg kg?1 choline. Dietary methionine was 0.58%, less than the requirement (0.69%). The results indicated that specific growth rate (SGR) was higher in the fish fed 2024 mg kg?1 diet than the control group. Feeding rate and feed efficiency were not significantly affected. Protein productive value increased as dietary choline increased from 76.1 to 2024 mg kg?1 diet and was lower in the fish fed the diet containing 4400 mg choline kg?1 diet. Serum high‐density lipoprotein cholesterol (HDL‐C) and total cholesterol significantly increased with increasing dietary choline up to 1457 mg kg?1, and no differences were found with further increase. Fish carcass fat contents decreased significantly with increased dietary choline. Hepatic lipid contents increased with dietary choline up to 1457 mg kg?1 and then decreased. Quadric regression of SGR and plasma HDL‐C indicted dietary choline requirement was 2500 and 2667 mg kg?1 diet, respectively.  相似文献

This experiment was conducted to study the effects of dietary carbohydrate on glucose metabolism in gibel carp (Carassius gibelio) as well to investigate if different clones had different responses to carbohydrate intake. Unselected Dongting strain (DT strain), selected gibel carp “CAS III” (A strain) and gibel carp “CAS V” (F strain) were fed with no corn starch diet (0C), 30% corn starch diet (30C) and 45% corn starch diet (45C) for 8 weeks. The results showed that selected F strain showed higher growth performance. F strain and A strain showed better control on postprandial glucose regulation by increasing plasma triglyceride, plasma cholesterol, liver and muscle glycogen contents and also enhancing glycolysis and restraining gluconeogenesis. DT strain had higher body lipid and lipid utilization. In conclusion, gibel carp could tolerate 45% dietary carbohydrate and F strain showed better growth by increasing glycolysis while decreasing glycogenesis.  相似文献

在(28±1)℃水温条件下,以4种循环饥饿的投喂方式饲养初始体重为1.58 g的异育银鲫(Carassius auratus gibelio)幼鱼,研究其补偿生长和部分生理生化指标的变化。实验分为四组,第一组每天投喂,为对照组,第二组投喂4 d后饥饿1 d,第三组投喂2 d后饥饿1 d,第四组投喂1 d后饥饿1 d,4种方式分别以C、R1/4、R1/2和R1/1表示,试验持续60 d。结果显示,3个试验组的实际摄食率均显著高于对照组(P<0.05),但饲料转化效率均显著低于对照组,导致3个试验组的特定生长率显著下降,体重未赶上对照组,而且饥饿时间越长,体重增长率越低。鱼体的蛋白质、灰分含量未受到投喂方式的显著影响,干物质、脂肪和能量含量随饥饿时间增加显著下降;肥满度、肝体比、ATP酶、SOD酶活性以及谷丙转氨酶和谷草转氨酶活性也随饥饿时间的增加显著下降。  相似文献

采用高效液相色谱法检测甲苯咪唑(MBZ)及其代谢物氨基甲苯咪唑(MBZ-NH2)和羟基甲苯咪唑(MBZ-OH)在银鲫(Carassius auratus gibelio)血浆及组织中的浓度,数据经3P97药代动力学程序分析.结果表明:(25±1)℃的水温条件下,银鲫单剂量口灌MBZ20mg·kg-1,血药经时过程符合二室开放式模型.主要药代动力学参数为:吸收速率常数恐Ka 0.235h-1;消除半衰T(1/2)β 52.26h;药时曲线下面积AUC 180.07μg·h·mL-1;表观容积分布Vd/F 2.465L·kg-1;清除率CL(s)0.111 mL·h-1·kg-1;达峰时间T(peak) 6.20 h;质量浓度Cmax 4.14 μg·mL-1.与哺乳类相比,MBZ在银鲫体内吸收较慢,消除半衰期明显延长,药代动力学参数也有较大差异.建议在做到合理用药的同时加强药物检测力度.  相似文献

A Thelohanellus species was encountered during a survey on Thelohanellus diversity of Carassius auratus gibelio (Bloch) in China. The infection is characterized by the presence of large cysts of 1.4–3.2 cm in diameter in the skin of host. Mature spores were ampullaceous in frontal view and testudinate in lateral view, measuring 19.7 ± 0.7 (18.6–20.8) μm long, 7.6 ± 0.4 (6.6–8.4) μm wide and 7.3 ± 0.5 (6.6–8.8) μm thick. The single polar capsule was elongated pyriform, with 11.1 ± 0.5 (10.0–11.9) μm long and 5.3 ± 0.3 (4.3–5.8) μm wide. Polar filaments coiled with 7–8 turns. Scanning electron microscopy revealed a smooth spore surface with flat side and convex side. The sutural line was straight or ‘S’ like, running near the middle of the valves. Histologically, the large cysts consisting of numerous small plasmodia developed in the dermis of the skin. The BLAST search indicated that the newly obtained ssrRNA gene sequences did not match any available sequences in GenBank and phylogenetic analysis placed it in the Thelohanellus clade. Based on morphology and molecular differences with reported Thelohanellus spp., this parasite was described as a new species of genus Thelohanellus.  相似文献

为研究crh (Corticotropin-releasing hormone)基因对鱼类摄食的调控作用,首次克隆了银鲫(Carassius auratus gibelio) crh基因c DNA全长序列。运用实时荧光定量PCR(RT-qPCR)技术检测crh基因m RNA在不同组织的表达及餐前、餐后和禁食对其表达的影响。结果显示,银鲫crh基因的c DNA序列全长为920 bp,其中,5’-UTR为53 bp,3’-UTR为378 bp,开放阅读框(Open reading frame,ORF)为489 bp。推导银鲫crh基因的ORF区编码蛋白由162个氨基酸组成,其中,含有11个氨基酸的保守区,24个氨基酸的信号肽,41个氨基酸的成熟肽。氨基酸序列的多重比较分析显示,银鲫crh基因与金鱼(Carassiusauratus)的同源性为99%,与鲤(Cyprinuscarpio)的同源性为96%。荧光定量PCR表达分析显示,银鲫crh基因在下丘脑中的表达量最高,其次是端脑和心脏。crh基因的表达量在餐前与餐后没有显著变化(P0.05),在禁食第5天、第7天出现极显著降低(P0.01),而在复投喂后第9天、第11天出现极显著升高(P0.01)。以上结果显示,crh基因在银鲫摄食调控方面可能扮演着重要角色。  相似文献

2008年10月江苏盐城大丰精养异育银鲫(Carassius auratus gibelio) 发生严重疾病,从病鱼肝脏、血液及腹水中分离到优势生长的细菌。对分离做纯培养的4株菌(JY081016-1至JY081016-4)进行形态特征、理化特性等表型生物学性状检验;测定了菌株(JY081016-1)的16S rRNA和gyrB基因序列,分析了16S rRNA和gyrB两种基因序列的同源性,并构建了系统发生树。结果表明分离菌对供试健康异育银鲫有强致病性,菌株(JY081016-1)所扩增的16S rRNA基因序列长度为1448bp(GenBank 登录号:GQ232759),所扩增的gyrB基因序列长度为1177bp(GenBank 登录号:GQ232760),其16S rRNA和gyrB基因序列与GenBank数据库中维氏气单胞菌和维氏气单胞菌温和生物型的16S rRNA和gyrB基因序列相似性均在97%以上;根据分离菌的表型及分子特征,判定分离鉴定的4株菌为温和气单胞菌(Aeromonas sobria)。胞外酶及溶血活性检测表明分离菌均能产生蛋白酶、卵磷脂酶、脂酶,在含7%家兔脱纤血液营养琼脂培养基上呈β型溶血;设计的特异性引物可扩增出溶血素基因。  相似文献

A ten‐week feeding trail was conducted to investigate the effects of increasing DL‐methionine (Met) supplementation on the success of fish meal (FM) replacement with plant proteins in practical diets for juvenile gibel carp, Carassius auratus gibelio. Twelve isoenergetic diets were formulated including two 150 g kg?1 FM diets (Diet 1—positive control 1 reflecting a commercial diet and Diet 2—positive control 2 reflecting a commercial diet but with balanced essential amino acid (EAA) profile) and ten 50 g kg?1 FM diets (negative controls) supplemented with graded levels (0–3.0 g kg?1) of DL‐Met (Diets 3–12). Each diet was fed to triplicate groups of gibel carp, near satiation four times daily for 10 weeks. Diet 2 with balanced EAA profile produced better final weight, specific growth rate (SGR) and feed conversion ratio (FCR) than the negative control diet containing no supplemental Met (Diet 3), but did not significantly differ from Diet 1. However, DL‐Met supplementation (0.5–3.0 g kg?1) in the negative control diets (Diets 4–12) produced growth performances similar to those fed the positive control diets (Diets 1 and 2). Based on quadratic regression analysis, the optimal dietary Met level with 5.2 g kg?1 of dietary cysteine (Cys) was found to be 7.1 g kg?1 dry diet for SGR and FCR. The corresponding total sulphur amino acid requirements (Met + Cys) of this species were calculated to be 12.3 g kg?1 dry diet for SGR and FCR. DL‐Met supplementation in 50 g kg?1 FM diets showed a decreasing trend in plasma cholesterol contents (< .05). No significant differences were observed in whole‐body composition, plasma protein, triglyceride and free EAA contents among dietary treatments, while plasma aspartate transaminase, albumin and ammonia contents were significantly influenced by dietary Met levels. Juvenile gibel carp grew equally well on 150 g kg?1 FM diet or 50 g kg?1 FM diets balanced for EAA profile with supplemental amino acids. The results of this study overall indicate that balancing dietary amino acid levels with DL‐Met supplementation is a key strategy in successfully reducing FM levels in the diets of gibel carp.  相似文献

A 100‐day growth trial was executed to determine the dietary selenium (Se) requirement of juvenile gibel carp (Carassius auratus gibelio). Selenomethionine was supplemented to casein‐gelatin diets at 0, 0.1, 0.25, 0.5, 1.0, 2.5 and 5 mg Se kg?1, respectively. Each of these seven semi‐purified diets containing 0.34, 0.47, 0.66, 0.82, 1.23, 2.77 and 5.13 mg Se kg?1 was fed to triplicate groups of gibel carp (2.74 ± 0.02 g) in a flow‐through system. No behaviour abnormalities and no mortality were found in fish exposed to dietary Se concentrations. With the increasing dietary Se supplemented concentrations, weight gain of fish remarkably increased at the levels of ≤1 mg Se kg?1 diet and then showed no significant difference above 1 mg Se kg?1 levels. Although growth performances (weight gain, hepatosomatic index, condition factor and survival) were not impaired in gibel carp fed at above the levels of 2.5 mg Se kg?1, indicators of oxidative stress were changed significantly. Serum superoxide dismutase (SOD) activities significantly declined, hepatic glutathione peroxidase (GPx) activities significantly increased and the tissue Se concentrations significantly raised at the highest supplemented level of 5 mg Se kg?1. A clear linear relationship between Se‐depended GPx activities and hepatic Se concentrations was observed. The present results indicated that the dietary Se requirement for gibel carp is 1.18 mg Se kg?1 diet based on weight gain, GPx activities and tissue accumulation.  相似文献

The objectives of the study were to investigate the effect of a feeding stimulant on feeding adaptation of gibel carp (Carassius auratus gibelio Bloch) fed diets with replacement of fish meal by meat and bone meal (MBM), and whether or not the juvenile gibel carp could adapt to higher MBM level in the diet. Juvenile and adult gibel carp were tested. Two and one replacement levels were used for juvenile and adult fish respectively. Each group of diets was set as two types with or without a unique rare earth oxide: Y2O3, Yb2O3, La2O3, Sm2O3, Nd2O3 or Gd2O3 (only the first four rare earth oxides were used in adult diets) for four adaptation periods of 3, 7, 14 and 28 days respectively. After mixing, an equal mixture of all six diets for juvenile or four diets for adult was offered in excess for 2 days. During the last 2 days of each experiment, no feed was offered and faeces from each tank were collected. Feeding preference was expressed as relative feed intake of each diet, which was estimated based on the relative concentration of each marker in the faeces. Given some adaptation period, such as 3–28 days, the effects of MBM and squid extract inclusion on the preference to each diet were reduced. After 28 days adaptation, the preferences between groups were not significantly different.  相似文献

以纯种方正鲫作母本、以方正鲫(♀)与鳞鲤(♂)交配获得的雌性后代经性反转的生理雄性个体作父本进行育种试验,在3个组配中,发现有一组F_1表现出新的形态特征。经过对F_1及其后代作定向培育和观测,证实F_1形态指标与方正鲫差异显著,且其后代遗传性状稳定,可作为鲫鱼属的一个新品系。  相似文献

Triplicate groups of gibel carp Carassius auratus gibelio Bloch (initial body weight: 4.89 g) were fed for 8 weeks at 24.8–30.8°C with nine isonitrogenous and isoenergetic diets. The control diet (F1) used white fishmeal (FM) as the sole protein source. In the other eight diets (F2–F9), 40.5–100% of FM protein was substituted by poultry by‐product meal (PBM) at 8.5% increments. The specific growth rate (SGR), feed efficiency ratio, protein efficiency ratio, protein retention efficiency and energy retention rate for fish fed PBM diets (F2–F9) were all higher, but not always significantly, than those for fish fed F1. All apparent digestibility coefficients for fish fed PBM diets were lower than those for fish fed F1. Fish fed F1 had a significantly higher hepatosomatic index value than fish fed PBM diets (P<0.05). No significant (P>0.05) effect of diet was found in whole‐body moisture and fat content. Whole‐body protein and energy content for fish fed PBM diets were slightly higher than that for fish fed F1. The optimal replacement level of FM by PBM was estimated by second‐order polynomial regression to be 66.5% in protein.  相似文献

An 84‐day growth trial was designed to investigate effects of dietary replacements fish oil with pork lard (PL) or rapeseed oil (RO) on growth and quality of gibel carp (Carassius auratus gibelio var. CAS III) (initial body weight: 158.2 ± 0.2 g), and responses of the fish refed fish oil (FO) diet. Three isonitrogenous (crude protein: 30%) and isolipid (crude lipid: 10%) diets were formulated containing 7.73% FO, PL or RO. Five experimental treatments including FO group (FO), PL group (PL), RO group (RO), group fed PL for 42 days and refed FO for 42 days (PL+rFO), RO and refed FO group (RO+rFO) was tested. At the end of first 42 days, the fish fed PL and RO had higher mortality than that of the control (P > 0.05). At the end of whole experiment, fish fed PL and RO showed higher plasma cortisol than FO fish (P < 0.05). RO+rFO fish showed higher lysozyme activity than RO fish (P < 0.05). Fish growth and feed utilization, composition of whole body and muscle, free amino acids, texture, off‐flavour substances or sensory attributes were not affected by dietary treatments (P > 0.05). PL and RO diet decreased muscle EPA, DHA and n‐3/n‐6 ratio (P < 0.05), while FO‐refeeding had recovery effect. It can be concluded that the replacement of FO by PL and RO does not affect the growth, feed utilization or fish tasting quality in gibel carp. Fish muscle fatty acids modified by dietary PL and RO can be recovered by refeeding with FO diet.  相似文献

This study was designed to use sucrose as carbon source to investigate the effect of biofloc technology on water quality and feed utilization in the cultivation of gibel carp (Carassius auratus gibelio var. CAS III). Three isolipidic and isoenergetic diets were formulated containing graded levels of dietary protein (257.4, 300.7 and 353.4 g/kg). The control group (P34) was fed with 353.4 g/kg protein diet without biofloc. Three biofloc treatments (P24 + B, P29 + B and P34 + B) were fed with the three diets with daily sucrose supplementation and continuous air inflation. The results found that the sucrose supplementation could generate biofloc and increased biofloc volume (BFV,< 0.05). The concentration of total ammonia nitrogen (TAN), nitrate nitrogen (NO3‐N) and the total phosphorus (TP) of all tanks decreased in biofloc groups, and the total nitrogen (TN) of P24 + B treatment tanks was significantly lower than that of the control group (< 0.05). The growth of P24 + B and P29 + B group was similar with that of control group (> 0.05). Feed conversion ratio (FCR) of P29 + B was lower than P24 + B and P34 + B (< 0.05). Protein retention efficiency (PRE) of P24 + B group was significantly higher than other treatments (< 0.05). Phosphorus retention efficiency (PhRE) of P24 + B group was the highest among all treatments. Compared with the control group, P29 + B group had an improved total antioxidant capacity (T‐AOC), superoxide dismutase (SOD) activity and contained the lowest malondialdehyde level (MDA). The present results indicated that low‐protein diet is more suitable for gibel carp in zero‐water exchange biofloc system compared with high‐protein diet.  相似文献
