河南省玉米茎基部镰刀菌的形态和分子鉴定   总被引:7,自引:0,他引:7  
 为明确河南省玉米茎基部镰刀菌(Fusarium spp.)的种群组成及分布, 2011和2012年, 我们采集了河南省14个地市42个县(区)的玉米茎基腐病病样, 分离得到163个镰刀菌单孢菌株。首先对菌株进行了形态学鉴定, 在此基础上使用镰刀菌种特异性引物进行了PCR检测;对于部分PCR检测未能确认的菌株进行了Translation elongation factor 1-α(TEF-1α)基因片段测序及BLAST分析, 最终将163个菌株鉴定到种。结果表明在163个菌株中, 禾谷镰刀菌F. graminearum为优势种, 占44.2%(72株), 其次为层出镰刀菌F. proliferatum和轮枝镰刀菌F. verticillioides, 分别占28.8%(47株)和27.0%(44株)。豫西北的焦作(47.6%)、洛阳(50.0%)及豫中的许昌(45.5%)均以F. verticillioides为主, 豫东的商丘(42.9%)、开封(57.1%)以F. proliferatum为主, 豫北的安阳(66.7%)、濮阳(71.4%)、新乡(62.5%)和豫南的南阳(57.1%)、驻马店(45.0%)以及中部的郑州(57.1%)、漯河(66.7%)则均以F. graminearum为优势种。  相似文献

A large collection (= 539) of Fusarium graminearum species complex (FGSC) isolates was obtained from Brazilian maize, and collections formed according to geography and maize part: (i) kernel (= 110) from south and south‐central Brazil; (ii) stalk (= 134) from Paraná state (south); and (iii) stubble (= 295) from Rio Grande do Sul state (south). Species composition, identified using a multilocus genotype approach, was assessed separately in each collection due to differences in geographic sampling. Overall, three species were found: F. meridionale (Fmer; 67% prevalence) with the nivalenol (NIV) genotype, F. graminearum (Fgra; 19%) with the 15‐acetyl (A) deoxynivalenol (DON) genotype, and F. cortaderiae (Fcor; 14%) with the NIV (49/74) or the 3‐ADON (25/74) genotype. In kernels, Fmer was spread across all locations and Fgra and Fcor were found mostly at high elevation (>800 m a.s.l.). The majority (97·8%) of stalk isolates was assigned to Fmer; three were assigned to Fgra. In the stubble, Fmer was less dominant (53%), with a shift towards Fcor as the most frequent species at high elevation sites (>600 m a.s.l.). No differences in the mycelial growth rate were observed among isolates from each species grown at 15°C. Fgra grew faster at 25°C and Fmer showed the widest range of variation across the isolates at both temperatures. The survey data suggest that Fmer may outcompete other species on ears and stalks in comparison to stubble. Additional sampling that controls for other factors, as well as direct testing of aggressiveness on ears and stalk tissue, will be needed to fully evaluate this hypothesis.  相似文献

This study aimed to assess the extent and distribution of Fusarium graminearum species complex (FGSC) diversity in rice seeds produced in southern Brazil. Four species and two trichothecene genotypes were detected among 89 FGSC isolates, based on a multilocus genotyping assay: F. asiaticum (69·6%) with the nivalenol (NIV) genotype, F. graminearum (14·6%) with the 15‐acetyldeoxynivalenol (ADON) genotype, and F. cortaderiae (14·6%) and F. meridionale (1·1%), both with the NIV genotype. Seven selected F. asiaticum isolates from rice produced NIV in rice‐based substrate in vitro, at levels ranging from 4·7 to 84·1 μg g?1. Similarly, two F. graminearum isolates from rice produced mainly 15‐ADON (c. 15–41 μg g?1) and a smaller amount of 3‐ADON (c. 6–12 μg g?1). One F. meridionale and two F. cortaderiae isolates did not produce detectable levels of trichothecenes. Two F. asiaticum isolates from rice and two from wheat (from a previous study), and one F. graminearum isolate from wheat, were pathogenic to both crops at various levels of aggressiveness based on measures of disease severity in wheat spikes and rice kernel infection in a greenhouse assay. Fusarium asiaticum and the reference F. graminearum isolate from wheat produced NIV, and deoxynivalenol and acetylates, respectively, in the kernels of inoculated wheat heads. No trichothecene was produced in kernels from inoculated rice panicles by any of the isolates. These findings constitute the first report of FGSC composition in rice outside Asia, and confirm the dominance of F. asiaticum in rice agroecosystems.  相似文献

采集了河南省12个地区的玉米弯孢菌叶斑病标样,分离获得了34个菌株,其中3个是画眉草弯孢(Curvu-laria eragrostidis),其余均为新月弯孢(C.lunata)。利用含KClO3的培养基对其中14个新月弯孢菌株和1个画眉草弯孢菌株(XZ3c)进行诱导,在MM培养基上鉴定均获得了Nit突变体。对获得的190个Nit突变体在MO2和MH培养基上进行了生理表型的划分,并用于菌株内、菌株间的营养体亲和性研究,发现有不少配对组合中均出现了明显的亲和现象,表明Nit突变体技术可以用于玉米弯孢病菌营养体亲和性的研究。根据亲和性测定的结果将这15个菌株初步分为2个营养体亲和群。  相似文献

Brenneria quercina has been reported as one of the causal agents of oak decline in Spain. To investigate the bacterial variability of this pathogen from different Spanish oak forests, a collection of 38 bacterial isolates from seven geographic locations and from different oak species was analysed by sequencing 16S rDNA and rep-PCR fingerprinting. All Spanish isolates of B. quercina were grouped by rep-PCR into a homogenous cluster that differed significantly from B. quercina reference strains from California. 16S rDNA analysis revealed that 34 out of 38 isolates were Brenneria . However, four isolates belonged to the genus Serratia , suggesting that this bacterium could cause cankers in oak trees. The information obtained by rep-PCR fingerprint analysis was used to develop PCR primers for the sensitive and specific detection of B. quercina from infected plant tissues. Pathogenicity tests performed with Brenneria and Serratia isolates showed that both were able to grow and cause cankers in oak trees.  相似文献

西瓜种传镰刀菌形态和分子鉴定及其对种子发芽的影响   总被引:3,自引:0,他引:3  
 采用洗涤检验法和PDA培养基法,从京欣一号和新京欣一号西瓜种子的种壳上分离得到镰刀菌,通过形态学观察和分子检测进行了鉴定,并研究了其对种子发芽的影响。通过菌落形态、色素颜色、菌落生长速率、大型分生孢子、小型分生孢子、厚垣孢子以及分生孢子梗等形态学观察,初步认为其为尖孢镰刀菌;采用真菌rDNA ITS区的通用引物、镰刀菌属特异性引物和尖孢镰刀菌特异性引物对其DNA进行PCR扩增,PCR产物经连接转化并验证后测序,将测序结果登录GenBank进行BLAST分析,分子检测结果与形态学观察结果一致,表明西瓜种子种壳携带的镰刀菌为尖孢镰刀菌,此结果为国内首次报道。用种传尖孢镰刀菌孢子悬浮液处理西瓜种子后,发芽势、发芽率和发芽指数均显著降低。  相似文献

Botrytis spp. associated with neck rot disease were isolated from New Zealand onions. The fungi were identified using molecular sequences of the ribosomal internal transcribed spacer (ITS) and intergenic spacer (IGS) regions, and the glyceraldehyde-3-phosphate dehydrogenase (G3PDH) gene. Analyses of the sequences showed that the majority of the isolates gathered in 2005–07 were B. aclada. A new high resolution melting analysis (HRMA) assay was developed that allowed fast and simple discrimination between B. aclada and other Botrytis spp. causing onion neck rot in New Zealand. To further verify these results, Botrytis isolates from New Zealand onions, stored in the International Collection of Microorganisms from Plants (ICMP), were also examined. Only a single isolate from the ICMP collection was B. aclada while two isolates were B. byssoidea, one B. squamosa and another closely related to Botryotinia porri. Identification of the remaining Botrytis isolates was more difficult; while IGS and ITS sequences indicated a close relationship to B. allii or B. byssoidea, a previously unreported intron insertion was observed at the 3′ end of the ribosomal small subunit gene in these isolates. No evidence of heterogeneity was observed in the G3PDH gene sequences, as might have been expected of the allodiploid B. allii, but the G3PDH sequence ruled out B. byssoidea as the identity of these isolates.  相似文献

Yellowing disease is one of the most important diseases of black pepper (Piper nigrum L.). To characterize the pathogen(s) responsible for yellowing disease of black pepper in Malaysia, 53 isolates of Fusarium were collected from the roots of diseased black pepper plants and from rhizosphere soils from major growing areas in Sarawak and Johor. A total of 34 isolates of F. solani and 19 isolates of F. proliferatum were obtained and identified based on morphological characteristics and molecular techniques. DNA sequencing of the internal transcribed spacers (ITS1 and ITS2) and 5.8S ribosomal DNA regions was conducted to identify Fusarium species. Nucleotide sequence analysis of the ITS regions revealed that this molecular technique enabled identification of Fusarium at the species level as F. solani and F. proliferatum. In a pathogenicity test on 3-month-old black pepper plants, F. solani was pathogenic, but F. proliferatum was not. On the basis of morphology, DNA sequences and pathogenicity of the fungal isolates from the diseased plants, we showed that yellowing disease on black pepper is caused by F. solani  相似文献

Fusarium fujikuroi is a species of the Gibberella fujikuroi species complex (GFSC) and the causal agent of bakanae disease on rice. Even if F. fujikuroi is the most abundant Fusarium species found on rice, other species can also be isolated from rice, such as F. proliferatum. Multiple alignment of translation elongation factor (TEF) gene sequences of different Fusarium spp., showed a deletion of six nucleotides in F. fujikuroi sequence and a two nucleotide polymorphism in the same region of F. proliferatum sequence. These elements of variability were used to develop a conventional and Real-Time PCR assay for diagnosis. The species specific primer pairs (Fuji1F/TEF1R and Proli1F/TEF1R) gave a product of 179 and 188?bp for F. fujikuroi and F. proliferatum respectively. Primer specificity was confirmed by analyzing the DNA of the most representative species of the GFSC and 298 strains of Fusarium spp. isolated from rice plants and seeds in Italy. The specific primers were also successfully used to detect fungal presence directly from infected rice tissues and seeds, providing a rapid tool for the early detection of pathogen contamination.  相似文献

西南地区玉米纹枯病病菌融合群鉴定和UP-PCR分析   总被引:1,自引:1,他引:1  
为明确西南地区玉米纹枯病病菌组成结构,从西南地区(四川、贵州、云南和重庆)玉米纹枯病样上分离获得285个疑似丝核菌菌株,根据培养性状、菌丝形态和细胞核染色对其进行鉴定,通过载玻片配对培养法对鉴定出的菌株进行菌丝融合群鉴定,并采用UP-PCR方法从各融合群中选取代表性菌株进行遗传变异分析。结果显示,供试疑似菌株中共鉴定出253个丝核菌,分别为224株立枯丝核菌Rhizoctonia solani和29株玉蜀黍丝核菌R.zeae。立枯丝核菌包括5个融合群,其中AG1-ⅠA共201株,出现频率为79.4%;AG1-ⅠB共2株,出现频率为0.8%;AG2-2ⅢB共3株,出现频率为1.2%;AG4-HGⅠ共10株,出现频率为3.9%;AG-5共8株,出现频率为3.2%。29株玉蜀黍丝核菌融合群均为WAG-Z,出现频率为11.5%。遗传变异分析中,当相似系数为0.78时,按照出现频率从立枯丝核菌和玉蜀黍丝核菌选取的不同融合群的20株菌株被划分为6个类群,与菌丝融合分析结果完全吻合,其中AG2-2ⅢB融合群首次从西南地区玉米上分离到。  相似文献

A sample of 140 Fusarium graminearum isolates from Rio Grande do Sul, Brazil, representing three populations at least 150 km from one another, were examined for trichothecene genotype based on PCR amplification of portions of the Tri3 and Tri12 genes and a species‐specific (Fg16F/R) primer pair. Genetic diversity was assessed in a sample of 103 F. graminearum lineage 7 (F. graminearum sensu stricto) isolates using amplified fragment length polymorphism (AFLP) markers. The 15‐ADON genotype was dominant, followed by the NIV genotype (2–18% prevalence), across all three populations. All NIV‐type isolates were in lineage 2 (F. meridionale) and all 15‐ADON‐type isolates were in lineage 7. Isolates with the same haplotype were rare and genotypic diversity was uniformly high (≥98% of the count), suggesting that recombination has played a significant role. The number of migrants (Nm) was estimated between 5 and 6 across all loci and all populations, but the high frequency of private alleles (up to 30%) suggests a historical, rather than contemporary, gene flow. Regarding linkage disequilibrium, 0·8, 1·5 and 2·2% of the locus pairs from the three populations were in disequilibrium, which is lower than values reported in other locations. Thus, Brazilian populations differ from those found in Europe, North America and most of Asia in the presence of a significant frequency (7·8%) of isolates of the NIV genotype in lineage 2.  相似文献

对一批引进后隔离培养中患病的红掌种苗,进行了根系线虫检测,通过病症分析、线虫形态测量、rDNA-ITS区序列分析等鉴定,结果表明:检测的线虫形态和香蕉穿孔线虫基本一致,rDNA-ITS区测序结果序列一致性为98%~100%.本报道再次证明香蕉穿孔线虫在红掌上的分布和危害,应该对种苗的跨区引进和种植过程进行严格的检疫.  相似文献

Brazilian isolates of Colletotrichum spp. from citrus orchards affected by postbloom fruit drop were examined for colony colour, mycelial growth, benomyl-resistance, pathogenicity, and genetic variability by random amplified polymorphic DNA (RAPD) analysis. All isolates were obtained from flowers and persistent calyxes from different citrus hosts from Sao Paulo, Brazil. DNA polymorphisms detected after amplification with random 10-mer primers were used to classify the isolates into two groups. Group I isolates grew rapidly on potato-dextrose agar (PDA) and were sensitive to benomyl, and group II isolates grew slowly on PDA and were benomyl-resistant. Colletotrichum acutatum was analyzed by RAPD and had high genetic similarity with group II isolates of Colletotrichum from citrus. Probably, the group I is C. gloeosporioides and group II is C. acutatum.  相似文献

从宁波口岸进境的日本鸡爪槭(Acer palmatum)景观树根际介质中分离到1种胞囊线虫属的2龄幼虫,通过形态鉴定和分子特征分析,鉴定其为旱稻胞囊线虫(Heterodera elachista Ohshima,1974)。该日本群体的2龄幼虫体长374~421μm;唇区有3个唇环;口针粗壮,长16.5~19.2μm,基部球圆形或前端略凹陷;侧线3条;尾长圆锥形,末端细圆,透明尾长22.9~30.7μm,约占尾长的50%。基于核糖体DNA的28S-D2/D3区和ITS区序列构建的系统发育树以及ITS序列酶切分析证实该鉴定结论,这是我国首次截获旱稻胞囊线虫。  相似文献
