AIM:To construct a subtracted cDNA library of differentially expressed genes in human unstable angina lymphocytes.METHODS:Suppression subtractive hybridizations (SSH) were performed between the patients with unstable angina pectoris and stable angina pectoris. Lymphocyte RNA, the obtained forward and reverse cDNA fragments were directly inserted into T/A cloning vector and transformed into E.coli JM 109 to construct a subtractive cDNA library. The inserting fragments were screened by blue and while blot screening and bacterium liqulid PCR.RESULTS:Each subtractive cDNA library contained more than 2000 positive bacteria clones. Most of them distributed between 200-600 bp inserts.CONCLUSION:The library is efficient and lays solid foundation for screening and cloning new and specific expressed genes in unstable angina lymphocyte RNA.  相似文献

AIM: To Screen and identify differentially expressed genes that involved in apoptosis model in rat cerebellar granule neurons (CGNs).METHODS: The rat cerebellar granule neurons were isolated and primarily cultured.Fluorescent differential display RT-PCR (FDD RT-PCR) was performed to screen differentially expressed ESTs in the apoptosis model of primarily cultured rat CGNs.ESTs were subcloned into pGEM-T EasyTM vector and then sequenced.Alignment assay in non-redunant database was applied for encoding information.Reverse Northern blotting was used to appraise the results from DDRT-PCR.RESULTS: 164 pieces of differentially expressed ESTs were obtained by FDDRT-PCR.17 of them were subcloned and sequenced.5 ESTs of 17 were confirmed to be positive results by reverse Northern blotting.CONCLUSION: DD-PCR is a rapid,simple-operation and sensitive method for screening differentially expressed genes,which would contribute to the molecular mechanisms of apoptosis/survive of CGNs.  相似文献

AIM: To observe activity of scavenger receptor of macrophage derived from monocytes, the change of inflammation factor in plasma (including CRP, sICAM-1, sVCAM-1) and the influence of atorvastatin on activity of scavenger receptor in patients with coronary heart disease. METHODS: 75 patients with coronary heart disease (CHD), whose plasma lipid levels were normal, were divided into three groups: stable angina pectoris, unstable angina pectoris and acute myocardia infarction. 29 healthy persons were served as control. The level of C-reactive protein (CRP), soluble intercellular adhesion molecule-1 (sICAM-1) and soluble vascular cell adhesion molecule-1 (sVCAM-1) in plasma were determined in all subjects. Monocytes in peripheral blood were dissevered and cultured to transform into macrophage. The influence of atorvastatin on activity of scavenger receptor in macrophage was observed. RESULTS: The level of CRP, sICAM-1, sVCAM-1 and activity of scavenger receptor of macrophage in acute myocardia infarction group were higher than that in stable angina pectoris, unstable angina pectoris and control. Atorvastatin lowered the activity of scavenger receptor of macrophages derived from monocytes in patients with coronary heart disease. Activity of scavenger receptor of macrophages derived from monocytes in patients with coronary heart disease was correlated positively with CRP, sICAM-1 and sVCAM-1. CONCLUSION: Activity of scavenger receptor may be taken as index for monitoring the degree of active vulnerable atherosclerosis plaque. Atorvastatin may inhibit activity of scavenger receptor in macrophages derived from monocytes in patients with coronary heart disease.  相似文献

AIM: Centrifuge training can improve forward acceleration (+Gz) endurance. This study was to analyzed the gene expression of rat heart affected by centrifuge, and to research the molecular mechanism of improving+Gz endurance by centrifuge training. METHODS: Differential expressed genes between high+Gz endurance (+16Gz) rats, of test group after trained12 d and control were screened using suppression subtractive hybridization (SSH) and dot blot hybridization. The obtained expressed sequence tags (ESTs)were used as probes to perform RNA slot hybridization with heart total RNA isolated from each gruop of centrifuge training and high+Gz endurance and low+Gz endurance (+12Gz) rats, respectively. The positive ESTs were sequenced and analyzed using BLAST(nr) at NCBI.RESULTS: Three down-regulated ESTs were obtained from heart samples, all of them are new, and their expression levels were decreasing during centrifuge training. CONCLUSION: Centrifuge training can significantly affect the special gene expressions of rat heart, and the expression changes of these genes may be ralated to the mechainism that+Gz endurance can be improved by centrifuge training.  相似文献

对石榴休眠枝段、成熟叶片及当年生新梢离体培养 ,建立其再生体系。结果表明 ,以休眠枝段、成熟叶片为外植体均能形成愈伤组织并分化出不定芽 ;当年生新梢茎段培养 ,以MS +BA 2 0mg·L-1+NAA0 3mg·L-1对茎段腋芽的增殖最适宜。诱导生根 ,用 1/2MS为基本培养基附加NAA 0 5mg·L-1+活性炭0 1mg·L-1+蔗糖 2 0g·L-1,生根率达 95 8%。培养基中附加 0 1%活性炭 ,对促进生根均有显著效果。  相似文献

 采用SSH技术以紫外照射的梁平柚[Citrus grandis(L.)Osbeck]果皮作为实验方(tester),以未被照射的正常果皮作为驱动方(driver),构建了一个梁平柚果皮紫外诱导基因的正向差减文库。经菌液PCR检测后随机挑取200个阳性克隆测序,获得168条表达序列标签(ESTs)。比对这168条ESTs,发现有分属于57个基因的114条ESTs与已知基因高度同源,54条ESTs同源性较低或没有同源性。功能分析发现,这些ESTs主要参与抗逆防御、生长发育、细胞凋亡、转录与翻译、细胞分化、信号传导、能量代谢、糖类及氨基酸代谢以及次生代谢等。  相似文献

AIM:To detect the role of fibrinogen activity (Fa) in the progress of coronary artery disease (CHD).METHODS:Fa was measured with hemorheology methods in patients with CHD stable phase (n=30) and angina pectoris (n=27).RESULTS: (1) Levels of plasmatic fibrinogen and plasmatic viscosity in patients with CHD were higher than that of control group (P<0.01,P<0.05).(2)Fa and platelet aggregation activity (Pt max, Pt H, Pt K) in patients with CHD angina pectoris were very much higher than that of control group (P<0.01, respectively).(3)There was a negative correlation between PT max, Pt H and Fa(r=-0.8379,P<0.01;r=-0.8784,P<0.01 respectively) in patients with CHD angina pectoris.CONCLUSION: Fa may play a role in the progress of CHD.  相似文献

AIM:To investigate clinical effect of tongxinluo capsule in treating stable angina pectoris patients,and its influence on endothelial function,superoxide dismutase (SOD) and malondialdehyde (MDA).METHODS:One hundred and twenty-four stable angina pectoris patients were divided into three groups,isosorbide treatment group (41 cases),tongxinluo capsule treatment group (40 cases),tongxinluo and isosorbide treatment group (combined treatment group,43 cases).The serum concentrations of nitric oxide (NO),endothelin-1 (ET-1),SOD and MDA were determined before and after treatment.The data in traetment groups were compared with that in normal control.RESULTS:The symptoms of 3 groups were significantly improved,and the total effective rate of tongxinluo capsule group and combined treatment groups were better than that in isororbide treatment group (85.00% and 88.37% vs 73.17%,P<0.05).Before treatment,the levels of serum NO and activity of SOD in angina patients were lower than that in control group.The serum MDA and ET-1 levels were higher than those in control.The levels of serum NO and SOD activity were increased remarkably after tongxinluo capsule or tongxinluo combined treatment.However,besides the concentration of NO increased after isosorbide treatment,the levels of serum ET-1 and MDA and SOD activity were not changed.CONCLUSIONS:The results suggest that tongxinluo capsule could effectively improve the symptoms of stable angina pectoris,and it is important for tongxinluo capsule to increase NO level and decrease ET-1 product,scavenge free radical and prevent lipid peroxidation.  相似文献

 以辣椒(Capsicum annuum L. ) 细胞质雄性不育系23A、121A, 和其相应的近等基因恢复系23C、121C为试验材料, 利用抑制消减杂交( SSH) 技术成功构建了CMS恢复基因诱导表达的消减cDNA文库。结合高密度点阵膜杂交差异筛选, 获得了282个阳性克隆。通过测序, 除去重复序列共得到175个Unique ESTs。在GenBank上进行BLAST分析, 55个EST片段未找到对应的同源序列, 可能代表了新基因;120个EST片段找到了对应的同源序列, 包括103个已知功能基因和17个未知功能基因。按照MIPS功能分类法, 将其分为14个功能组, 涉及代谢、胁迫应答、蛋白活性、转录因子、信号转导等多方面的功能。  相似文献

AIM: To investigate the plasma neurotensin(NT) concentration in patients with coronary heart disease: acute myocardial infarction(AMI),unstable angina(UA),stable angina pectoris(SAP), old myocardial infarction (OMI), and to study the relationship between the plasma NT level and the myocardial ischemia . METHODS:The plasma NT concentration of 30 patients with AMI,32 pat ients with UA,35 patients with SAP,31 pat ients with OMI and 32 normal controls were determined by radioimmunoassay(RIA). RESULTS: The plasma NT level in patients with AMI(24 h),in patients with UA when angina attacked is significantly higher than that of healthy controls. The plasma NT level in patients with SAP, in patients with OMI is not significantly different from that of healthy controls. The NT level of patients with UA when angina attacked is significantly higher than that after 2 weeks treatment (P<0.01). The NT level of AMI patients rose from early period and reached peak value at 24 h .It began to decline at 48h,and restored to normal level at 72h. CONCLUSION: When acute myocardial ischemia occured in patients with coronary heart disease, the plasma NT level was elevated. This suggests NT may have participated in the pathophysiological course of myocardial ischemia.  相似文献

 为分析柑橘果实贮藏过程中枯水相关特异基因的表达情况,以纽荷尔脐橙(Citrus Sinensis Osbeck)采摘后贮藏至略见枯水果实的果肉为测验方(Tester),以刚采摘果实的果肉为驱动方(Driver),采用抑制性差减杂交(Suppression Subtractive Hybridization,SSH)技术成功构建了贮藏果实差减cDNA文库,共获得656个阳性克隆。随机挑取克隆进行菌液PCR分析,插入片段长度主要集中在100 ~ 1 000 bp。成功对213个克隆进行测序,获得有效序列143条,经序列分析,共得到53条uniESTs序列。在NCBI基因库中对这53个uniESTs进行BLAST分析,比对结果参照MIPS的分类标准,按功能分为14类,其中参与代谢、能量及蛋白合成的ESTs最多,分别占到了全部ESTs的17%、11%和13%。  相似文献

AIM:To determine differentially expressed genes associated with cell adhesion and immune regulation in peripheral blood eosinophils from asthmatic patients. METHODS:Peripheral blood eosinophils were isolated from the asthmatic patients at the time of exacerbation and after improvement. Total RNA was extracted. Super SMART PCR cDNA was synthesized, suppression subtractive hybridization (SSH) and PCR-select differential screening technology were used to detect expressed genes. The differentially expressed genes were sequenced. RESULTS:High efficiency subtractive cDNA library was constructed successfully. Differential screening identified 15 differentially expressed genes, which were Charcot-Leyden crystal protein (CLC protein;galectin-10), putative pre-mRNA splicing regulator female-lethal (2D), aquaporin 9 (AQP9), IL-8, slingshot 2L (SSH-2L), PP1 catalytic subunit, beta isoform, helicase with zinc finger domain (HELZ), β2-microglobin (β2-MG) and a gene associated with mitochondrion. CONCLUSION:Increased expression of these genes might be associated with eosinophil migration, adhesion and immune regulation. Intervention of these pathways may provide a theoretical base for future new targeting treatment for asthma.  相似文献

AIM: To examine the relation between serum concentrations of interleukin-18, interleukin-10, interleukin-6 and acute coronary syndrome (ACS). METHODS: Serum concentrations of IL-18, IL-10, IL-6 were measured in 17 patients with acute myocardial infarction (AMI), 30 patients with unstable angina pectoris (UAP), 15 patients with stable angina pectoris (SAP) and 20 controls by enzyme-linked immunosorbent assay (ELISA) and radioimmunoassay (RIA).The relation between IL-18, IL-6 and IL-10 was compared. RESULTS: Serum concentrations of IL-18, IL-6 were significantly increased in the AMI and UAP groups in comparison with the SAP and control groups. Conversely, serum concentrations of IL-10 were significantly decreased in the AMI and UAP groups in comparison with the SAP and control groups. The correlation of concentrations of IL-18 and IL-6 had no significance; but the levels of IL-18 and IL-6 were negatively correlated with IL-10. CONCLUSION: Serum IL-18, IL-6 concentrations increase while serum IL-10 concentration decreases in patients with acute coronary syndromes. The inflammatory imbalance between IL-18, IL-6 and IL-10 may play an important role in the instability of atherosclerotic plaque.  相似文献

AIM To explore novel genes closely related to the prognosis of colorectal cancer through bioinformatics analysis of RNA sequencing (RNA-Seq) data of gene expression profile in large samples of colorectal cancer tissues from The Cancer Genome Atlas (TCGA) database, and to validate the new genes and study their functions. METHODS The RNA-Seq data were downloaded from TCGA database to screen differentially expressed genes, R-survival package was used to carry out survival analysis of differentially expressed genes to screened out genes related to prognosis of colorectal cancer, and the most significantly up-regulated genes that have not been reported in cancer studies were selected for further verification. RT-qPCR was employed to detect mRNA expression of these novel genes in human colon cancer cell lines and human colorectal cancer tissues. The colon cancer cell line with stable silencing of the novel gene expression was constructed by transfection of lentivirus vector, and its viability, migration and invasion were detected by CCK-8 assay and Transwell assay. RESULTS (1) A total of 4 017 differentially expressed genes were found in the gene expression profile of the colorectal cancer samples. Kaplan-Meier survival analysis showed that 69 genes were closely related to poor prognosis in patients with colorectal cancer, 36 of which were up-regulated, and 11 of these 36 genes have not been reported in cancer studies. (2) The mRNA expression of the top 3 up-regulated genes CCDC78, PGGHG and TSPEAR of the 11 genes was significantly increased in colon cancer cell lines, and the expression of CCDC78 mRNA was also up-regulated in colorectal cancer tissues. (3) CCDC78 gene silencing significantly suppressed the viability, migration and invasion of colon cancer cells (P<0.01). CONCLUSION Bioinformatics analysis based on TCGA database is helpful to discover nov?el genes related to prognosis of colorectal cancer, and CCDC78 may be a novel oncogene associated with poor prognosis of colorectal cancer.  相似文献

AIM:To explore the serum levels of certain adhesion molecules and its significance in acute coronary syndrome(ACS). METHODS:The subjects included 40 patients with acute myocardial infarction(AMI) and 40 patients with unstable angina pectoris (UAP). Among the 80 patients, 60 patients accepted a follow-up 4 months. At the same time we selected 40 controls from people who attended a routine health check in the university. Serum levels of E-selectin, sICAM-1, sVCAM-1 were measured by ELISA.RESULTS:Serum levels of E-selectin, sICAM-1, sVCAM-1 were significantly higher in the ACS group(AMI or UAP) than in the control group. Four months later, the levels of E-selectin, sICAM-1 became significantly lower in the follow-up group than in the ACS group, while sVCAM-1 showed no significant difference. CONCLUSION:Serum levels of E-selectin, sICAM-1 may have certain diagnostic value for ACS, and can be a useful marker reflecting the stability of the disease.  相似文献
