AIM:To approach the relationship between the expression of intercellular adhesion (ICAM-1 mRNA) and acute lung injury (ALI) as well as the mechanisms of rhubarb in the prevention and treatment of the lung injury. METHODS:ALI animal model was performed by Lipopolysaccharide (LPS). The rats were divided into 4 groups: LPS group, control group, rhubarb+LPS group and dexamethasone+LPS group. Histopathological examination and biological markers were measured for the lung specimens. Molecular hybridization method was used to determine the expression of ICAM-1 mRNA. RESULTS:The ICAM-1 mRNA expression in the lung tissues of LPS group significantly increased compared with control group (P<0.01), rhubarb and dexamethasone had the action of decreasing the ICAM-1 mRNA expression (P<0.05, P<0.01); pathologic changes and the biological markers of ALI significantly decreased or ameliorated. CONCLUSION:The increase in the expression of ICAM-1 mRNA in the lung tissues of ALI is involved in the formation of ALI. Rhubarb and dexamethasone can ameliorate the lung damage, mechanism of which may be related to the inhibition of ICAM-1 mRNA expression.  相似文献

AIM: To study the production of intercellular adhesion molecule-1(ICAM-1), E-selectin and P-selectin in serum, lung tissues and bronchoalveolar lavage fluid(BALF)of acute lung injury(ALI) model and to observe the effects of ambroxol combined with low-dose heparin on the changes of the 3 factors above.METHODS: Twenty-four healthy rabbits were randomly divided into 3 groups: normal saline control group (NC), oleic acid injury group (OA), ambroxol+ heparin treatment group (AH). The rabbit ALI model was induced by oleic acid injection through auricular vein. Partial pressure of O2 in artery(PaO2) was analyzed.The concentrations of ICAM-1 and E-selectin were detected by ELISA.The apoptosis index(AI) was measured by TUNEL method.The expression of P-selectin was determined by immunohistochemical method.The ultrastructural changes of the lung tissues were observed under electron microscope, and the lung wet/dry ratio(W/D) was calculated.RESULTS: PaO2 in AH group and OA group was significantly lower (P<0.01) than that in NC group, and PaO2 in AH group was significantly higher than that in OA group (P<0.01). The concentrations of ICAM-1 and E-selectin in serum, lung tissues and BALF, and AI and W/D in lung tissues in AH group were higher (P<0.05 or P<0.01) than those in NC group, and was lower than those in OA group (P<0.05 or P<0.01). In NC group, no significant change of the above parameters at all time points was observed (P>0.05). In OA group, PaO2 was significantly decreased (P<0.01) with the pathological process developed, and the concentrations of ICAM-1 and E-selectin were significantly increased. In AH group, PaO2 was decreased (P<0.05),and the concentrations of ICAM-1 and E-selectin were increased with the process of ALI developed. The P-selectin expression in lung tissues of OA group was distributed mainly in inflammatory cells, capillary endothelial cells and plasma. From low to high levels, the order was NC group < AH group < OA group in the expression of P-selectin. The most obvious apoptosis was observed in OA group. No apoptosis or occasional positive cells were found in NC group. The apoptotic rate in AH group was significantly reduced compared with that in OA group.CONCLUSION: In ALI induced by OA, ICAM-1, E-selectin and P-selectin are significantly increased and are involved in the occurrence and development of ALI. Ambroxol combined with low-dose heparin reduces the levels of ICAM-1, E-selectin and P-selectin, the pulmonary edema and the lung injury, improves pulmonary functions, and plays an important role in the prevention and treatment of acute lung injury.  相似文献

AIM: Effect of endothelial cell on the development of acute lung injury and the prevention of dexamethasone in acute lung injury were observed.METHODS:Rats were divided into three groups:1.Control group.2.LPS group:Venous injection with LPS(5mg/kg body weight),execute respectively at 1 h,2 h,6 h and 24 h after LPS injection. 3.dexamethasone group:intraperitoneal injection with dexamethasone ,1 h before LPS injection,execute after 2 hours after LPS injection.RESULTS: Serum NO,TNF-α levels,lung iNOS activity and lung ICAM-1mRNA expression were increased( P <0.05, P <0.01, vs control group),but serum ACE was decreased( P <0.01).Dexamethasone could improve all the changes above mentioned.CONCLUSION:Endothelial cell played a vital role in the development of acute lung injury and dexamethasone could prevent acute lung injury.  相似文献

AIM: To study the change of intercellular adhesion molecule-1(ICAM-1) expression in intestine tissues of mice induced by LPS and regulatory effect of p38 mitogen-activated protein kinase(p38 MAPK) on ICAM-1 expression. METHODS: Protein and mRNA of ICAM-1 were measured using Western blotting and RT-PCR respectively in intestine tissue of BALB/c mice treated by lipopolysaccharide(LPS) or LPS plus SB203580, a specific inhibitor of p38 MAPK. RESULTS: Compared with control group, the expression of ICAM-1 protein and mRNA was increased significantly by LPS stimulation in dose- and time-dependent manner. ICAM-1 expression reached peak value at 12-36 h after LPS stimulation. 20.0 mg/kg of LPS could induce the maximum of ICAM-1 expression. Pretreatment of mice with SB203580 for 30 min could inhibit significantly LPS-induced expression of ICAM-1 protein and mRNA expression in mouse intestine tissues. CONCLUSIONS: These data highlight that LPS could up-regulate ICAM-1 protein and mRNA expression in intestine tissue of mice in dose- and time-dependent manner, and p38 MAPK signal pathway plays an important role in ICAM-1 expression induced by LPS. It suggests that inhibition of p38 MAPK might be a useful principle for the prevention and treatment of intestine damage of endotoxic shock.  相似文献

AIM:To observe the inhibitory effects of vinpocetine injection on lipopolysaccharide (LPS)-induced acute lung injury (ALI) in the rats and to explore the underlying mechanisms.METHODS:Male Wistar rats (n=50) were randomly divided into 5 groups:control group,ALI model group,and low,medium and high doses of vinpocetine treatment groups.The rats in control group were injected with 0.9% NaCl at 5 mL/kg through femoral vein.The rats in ALI model group received LPS at 10 mg/kg through femoral vein.After injected with LPS,the rats in vinpocetine treatment groups received vinpocetine at 0.2 mg/kg,0.7 mg/kg or 1.2 mg/kg via intraperitoneal injection.The pathological changes of the lung tissues were observed under microscope with HE staining.The cell apoptosis in the lung tissues was detected by TUNEL staining.Myeloperoxidase (MPO) activity was measured by the method of spectrophotometry.The protein expression of NF-κB,ICAM-1,VCAM-1,Bax and Bcl-2 was determined by Western blot.RESULTS:Compared with ALI group,administration of vinpocetine significantly attenuated the structural injury of the lung and the infiltration of inflammatory cells.Moreover,vinpocetine decreased cell apoptosis and MPO activity in the lung tissues of ALI rats.In addition,the protein expression of NF-κB,ICAM-1,VCAM-1 and Bax was inhibited after vinpocetin treatment,whereas Bcl-2 expression was increased.CONCLUSION:Vinpocetine attenuates LPS-lung injury by reducing MPO activity and regulating NF-κB,ICAM-1,VCAM-1,Bax and Bcl-2 protein expression.  相似文献

AIMTo investigate the effects of cucurbitacin E on airway inflammation and the signaling pathways of MAPKs and NF-κB in asthmatic mice. METHODSHealthy mice (n=40) were randomly divided into control group, model group, low-dose cucurbitacin E group, high-dose cucurbitacin E group and dexamethasone group. Ovalbumin sensitization was used to induce asthma in the mice. The protein levels of p-JNK, p-ERK1/2, p-p38 MAPK and p-p65 in the lung tissues were determined by Western blot. RESULTSCompared with control group, the numbers of inflammatory cells, such as eosinophils, lymphocytes and neutrophils, were significantly increased in model group, and the activity of MAPKs and NF-κB signaling pathway-related proteins was significantly enhanced. Cucurbitin E at high dose attenuated airway inflammation in asthmatic mice, and significantly inhibited the activity of MAPKs and NF-κB signaling pathway-related proteins. Histopathological results showed proliferation of goblet cells and bronchial mucosal epithelial cells, infiltration of inflammatory cells in the alveoli, and narrow alveolar cavity in model group, while the pathological changes were significantly alleviated in cucurbitin E treatment groups. CONCLUSION Cucurbitin E improves airway inflammation in asthmatic mice, and its mechanism may be related to the inhibition of MAPKs and NF-κB signaling pathways.  相似文献

AIM:To investigate the effect of inhalation of inactivated Mycobacte-rium phlei on the expression of nuclear factor-kappa B (NF-κB), intercellular adhesion molecule (ICAM)-1 and vascular cell adhesion molecule (VCAM)-1 in the lung tissues of asthmatic mice. METHODS:Male BALB/c mice (n=24) were randomly divided into normal control group (A), asthmatic model group (B), and inactivated Mycobacterium phlei inhalation group (C). Asthmatic model was made by inhalation of chicken ovalbumin. The mice in group C were treated with inactivated Mycobacterium phlei for 5 d. The lung tissues and bronchoalveolar lavage fluid (BALF) were harvested. HE and AB-PAS staining were used to measure the lung inflammation and mucus production. The inflammatory cells in the BALF were counted. The mRNA expression of NF-κB, ICAM-1 and VCAM-1 was detected by real-time fluorescence quantitative PCR. RESULTS:Mycobacterium phlei treatment alleviated lung inflammation, attenuated mucus production, and reduced the percentage of eosinophils in the BALF. The mRNA levels of NF-κB and ICAM-1 were significantly decreased after treated with Mycobacterium phlei. However, no significant difference of VCAM-1 mRNA expression was found before and after treatment. The correlation between NF-κB mRNA and ICAM-1 mRNA, and between NF-κB mRNA and VCAM-1 mRNA was not found. CONCLUSION:Inhalation of inactivated Mycobacterium phlei attenuates asthmatic airway inflammation. NF-κB participates in the pathogenesis of asthma. NF-κB signal pathway may be associated with the therapeutic mechanism. Another important mechanism is the reduction of adhesion molecule expression.  相似文献

AIM: To investigate the effects of cannabinoids WIN55, 212-2 (WIN) and O-1602 on the expression of heat-shock proteins (HSPs), such as HSP27, HSP60 and HSP70, in the inflammatory tissues of mice with experimental colitis or acute pancreatitis (AP). METHODS: Mouse colitis was induced by feeding the C57/BL with 4% dextran sulfate sodium (DSS) for 7 d, and AP was induced by intraperitoneal injection of ceruline in the mice (50 μg/kg hourly, with a total of 6 times). The mice were intraperitoneally administered with WIN or O-1602 for the therapeutic evaluation by observing the following parameters: pathological changes of the tissues, plasma activity of amylase, plasma levels of IL-6 and cytokine-induced neutrophil chemoattractant-1 (CINC-1), and HSP expression in the colonic and pancreatic tissues. RESULTS: Compared with normal control mice, the colonic tissues from colitis mice and the pancreatic tissues from AP mice appeared obvious signs of inflammation and injury. The plasma levels of IL-6 and CINC-1 significantly increased in colitis mice and AP mice. In DSS group of colitis mice, HSP27 expression increased, but HSP60 and HSP70 were reduced in the colonic tissues. WIN showed anti-inflammatory effects on the pathological changes of colonic tissues and the plasma cytokine levels. WIN also improved the expression of HSP70 (P<0.05). In AP group, the expression of HSP27 and HSP70 in pancreatic tissues increased, and HSP60 decreased. O-1602 also showed some anti-inflammatory effects on the changes of the pathological tissues and the plasma parameters, but had no obvious effects on the HSP expression (P>0.05). CONCLUSION: The experimental colitis and acute pancreatitis in mice were induced by feeding DSS and injection of cerulein, respectively. The changes of HSP expression were different in the inflammatory tissues. WIN and O-1602 show anti-inflammatory effects and increase the expression of several HSPs to some extent.  相似文献

AIM: To investigate the effect of siduqing decoction, a Chinese medicine, on survival rate and multiple organ dysfunction in mice challenged with LPS. METHODS: Mice were administered intragastrically with Siduqing decoction or distilled water (0.2 ml/10 g) twice a day for 3 days, two hours after Chinese herbal medicine treatment on day 3, LPS or normal saline was injected intraperitoneally, and survival rates in each group were recorded at 12-h intervals. In another experiment, mice were sacrificed at 12 h after LPS, lung, liver, kidney and small intestine were collected and processed for the H & E staining. In addition, Blood was collected at 10 h after LPS injection for determining alanine aminotransferase (ALT) activity, blood urea nitrogen (BUN) and creatinine (Cr) contents. RESULTS: At 96 h after LPS injection, the survival rate (27%, n=34) was lower in LPS group than Siduqing treatment group (65%, n=31, P<0.05). ALT activity, BUN and Cr contents in serum were higher in LPS group than control group, Siduqing treatment significantly attenuated a increase in ALT activity, BUN and Cr content in serum induced by LPS. Histological examination showed inflammatory injury in the lung and intestine, hemorrhage in the lung and kidney, degeneration, necrosis in the liver and kidney, while Siduqing treatment attenuated pathological changes induced by LPS. CONCLUSION: These data indicate that Siduqing has a protective effect against LPS-induced multiple organ injury and increases survival rate in mice challenged with LPS.  相似文献

AIM: To investigate the effects of antioxidant N-acetylcysteine (NAC) on the lipopolysaccharide (LPS)-induced MAPK phosphorylation in mouse liver. METHODS: 54 male mice were divided into three groups: control (n=6), 0.9% sodium chloride 0.2 mL ip; LPS group (n=24): LPS 5 mg ip; NAC+LPS group (n=24): NAC 150 mg·kg-1·d-1 ip, for 3 d; LPS 5 mg ip after 1 h of NAC administration at 3rd day. The liver was excised with carbrital anesthesia after LPS or 0.9 % sodium chloride injection at 0.5 h, 1 h, 2 h and 6 h for GSH and MDA assays. The protein extracted from liver was assayed for the phosphorylation of MEK1/2, ERK1/2, p38 MAPK by Western blotting. TNF-α in liver was assayed by radioimmunoassay. RESULTS: MDA in the liver was decreased remarkably and the GSH in the liver was increased significantly by NAC pretreatment. The phosphorylation of MEK1/2, ERK1/2 and p38 MAPK in liver were inhibited significantly by NAC pretreatment after LPS challenge. Meanwhile, TNF-α in liver was decreased markedly. CONCLUSION: Reactive oxygen species plays a critical role in MAPK signaling during the LPS induced acute liver injury. NAC partially inhibits LPS-induced MAPK signaling by antioxidant effect and decreases TNF-α production.  相似文献

AIM: To investigate the effects of caveolin-1 (Cav-1) scaffolding domain peptide, cavtratin, on lipopolysaccharide (LPS)-induced mouse acute lung injury and heme oxygenase-1 (HO-1) activity. METHODS: Adult male BALB/c mice were randomly divided into 6 groups (n=8 to 10):control, Antennapedia internalization sequence (AP), LPS, LPS+hemin, LPS+ hemin+cavtratin and LPS+hemin+cavtratin+zinc protoporphyrin IX (ZnPP) groups. After LPS administration for 24 h, the lung pathological changes, the wet/dry weight (W/D) ratio of lung tissues, total cell number in bronchoalveolar lavage fluid and serum lactate dehydrogenase activity were measured. The co-localization of HO-1 and Cav-1 was displayed by immunofluorescence, and the HO-1 activity were detected. The mRNA expression of pro-inflammatory cytokines IL-1β, IL-6, TNF-α, MCP-1 and iNOS was detected by real-time PCR. RESULTS: The mice in LPS+hemin+cavtratin group had the decreased interaction between HO-1 and Cav-1, and the increased HO-1 activity compare with LPS group (P<0.05). Compared with LPS group, the pulmonary damage was attenuated in LPS+hemin+cavtratin group, and the injury indexes, including W/D ratio, total cell number in bronchoalveolar lavage fluid and lactate dehydrogenase activity in the serum, and the mRNA expression of inflammatory cytokines all decreased (P<0.05). HO-1 activity inhibitor ZnPP abolished the above protective effect of cavtratin on the lung tissues with LPS-induced acute lung injury. CONCLUSION: Cavtratin has beneficial effects on the lung with LPS-induced acute injury by restoring the HO-1 activity.  相似文献

AIM:To investigate the effects of rosiglitazone (ROSI),an agonist of peroxisome proliferator-activated receptor γ (PPARγ),on the lung expression of intercellular adhesion molecule-1 (ICAM-1) and cytokine-induced neutrophil chemoattractant (CINC) in rats with acute lung injury.METHODS:Thirty-six male Wistar rats were randomly divided into six groups:control group,ROSI group,GW9662 (a PPARγ antagonist) group,lipopolysaccharide (LPS,6 mg/kg,iv) group,ROSI-LPS group (0.3 mg/kg ROSI iv 30 min prior to LPS) and GW9662-ROSI-LPS group (0.3 mg/kg GW9662,iv,20 min before ROSI).Four hours after LPS injection,wet/dry weight (W/D) ratio,myeloperoxidase (MPO) activity,malondialdehyde (MDA) and CINC-1 concentrations were assayed in the lung tissues.Immunohistochemical analysis of ICAM-1 expression was also studied.RESULTS:Pretreatment with ROSI significantly attenuated LPS-induced increases in W/D ratio,MPO activity,MDA and CINC-1 concentrations as well as ICAM-1 expression in the lung tissues.The specific PPARγ antagonist GW9662 antagonized the effects of ROSI.CONCLUSION:Pretreatment with ROSI reduces LPS-induced lung injury in rats.The mechanism involves inhibition of the lung expression of ICAM-1 and CINC-1 by the activation of PPARγ.  相似文献

AIM: To study the protective effect of glutamine (Gln) against endotoxemia by observing the effect of glutamine on heat shock proteins (HSPs) and tumor necrosis factor-α (TNF-α) in endotoxemic rats. METHODS: The rats were randomly divided into 3 groups, lipopolysaccharide group (LPS), glutamine-treated group (Gln) and control group (C). The blood was drawn from lateral tail vein for analysis of cytokine levels at 0, 2, 4 and 6 h post-lipopolysaccharide (LPS) challenge. TNF-α was measured by radioimmunity assay. Multiple tissues were harvested from the rats, and HSP70 was detected by immunohistochemistry. At the same time, lung, liver, and ileum tissue section were stained with hematoxylin and eosin. RESULTS: Gln treatment resulted in marked attenuation of TNF-α expression at 2 h post-LPS injection (P<0.01). Gray gradients of HSP70 in lungs, liver and ileum tissue in group Gln were much lower than those of group LPS (P<0.05), This suggested that HSP70 content in these tissues of group Gln was higher than that of group LPS. Tissue sample from lung, liver and ileum revealed significantly less evidence of endotoxin-induced tissue damage in Gln-treated animals. CONCLUSION: Gln can significantly enhance HSP70 expression in multiple tissues of endotoxin-treated rats. A single dose of intravenous Gln given concomitantly with an endotoxin injury can markedly reduce organ histological damage, and attenuate pro-inflammatory cytokine release.  相似文献

AIM: To investigate the protective effects of berberine against liver injury induced by lipopolysaccharide in mice and the mechanisms underlying its protective effect. METHODS: The male mice were divided randomly into control, berberine group, LPS group and berberine treatment group. Mice were administered intragastrically with distilled water (0.01 mL/g) or 5 g/L neutral sulfate berberine (0.01 mL/g) once a day for 5 days and injected intraperitoneally with normal saline or LPS (0.02 mL/g,28 mg/kg)at 1 h after gavage on day 5. Blood was collected for determining alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities, the content of tumor necrosis factors-α (TNF-α) at 10 h and 2 h after LPS or normal saline injection, respectively. Furthermore, the liver tissue was processed, and histological changes and ultrastructure in liver were observed with light and electron microscopy, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity in liver were also detected. RESULTS: Both ALT and AST activities in serum in LPS group were higher than those in control and berberine treatment group. LPS increased the serum TNF-ɑ content at 2 h after injection, which was reversed by berberine pretreatment. The histological examination showed that LPS caused severe hepatic cell edema, degeneration, apoptosis and even necrosis, and ultrastructure observation demonstrated that LPS induced mitochondrial swelling, condensation and margination of chromatin, irregular nuclear envelope in hepatocytes. The above pathological changes produced by LPS were attenuated by berberine pretreatment. Moreover, MDA contents in liver tissue were higher in LPS group than control and berberine treatment group, but there were no significant difference in SOD activity between berberine treatment and LPS group. CONCLUSION: Berberine has a protective effect on LPS-induced liver injury in mice, the mechanisms may be related to its decreasing the production of TNF-α, inhibiting lipid peroxidation and protecting mitochondria.  相似文献

AIM:To investigate the effect of platelet inhibitor from Agkistrodon halys venom (AHV-PI) on lipopolysaccharide (LPS) induced human umbilical vein endothelial cells (HUVECs) injury in vitro, and to explore its mechanism. METHODS:Cultured HUVECs were induced inflammatory injury by LPS (1 mg/L). The experiment was divided into blank control group, LPS group, AHV-PI group and AHV-PI+LPS group. The viability of HUVECs was measured by MTT assay. The morphological changes of HUVECs were observed under inverted microscope. The optimum concentration of AHV-PI at 5 mg/L was selected. Flow cytometry was used to detect apoptosis of HUVECs. Immunohistochemical method was used to observe the expression of tissue type plasminogen activator (t-PA) and plasminogen activator indhibitor-1 (PAI-1) of HUVECs. ELISA was used to detect the concentrations of intercellular adhesion molecule-1 (ICAM-1) and tissue factor (TF) in the supernatant. The activation and translocation of NF-κB subunit p65 were observed by immunofluorescence staining. RESULTS:The HUVECs were spindle shaped, the ratio of length to width was increased, the cells were fibroblast-like, and granular substance appeared in the cytoplasm in LPS group. The viability and morphological changes of HUVECs were not significantly affected as treated with AHV-PI at concentration of 0~5 mg/L, but the viability of HUVECs induced by LPS was inhibited and the morphological changes were alleviated. Compared with the blank control group, the levels of TF and ICAM-1 in the supernatant increased, and the expression of t-PA and PAI-1 in the HUVECs was decreased in LPS group. Compared with the LPS group, the contents of TF and ICAM-1 in the supernatant were significantly decreased, the expression of t-PA and PAI-1 in the HUVECs was increased and the expression of nucleus NF-κB p65 was decreased in AHV-PI+LPS group (P<0.05). CONCLUSION:AHV-PI reduces HUVECs damage. The protective mechanism is related to the inhibition of cytokine secretion and NF-κB activation.  相似文献

AIM To explore the repair effect of purple sweet potato anthocyanin on intestinal barrier injury of ulcerative colitis mice induced by dextrin sulfate sodium (DSS). METHODS The mice were randomly divided into normal drinking group, DSS model group, different doses of purple sweet potato anthocyanin (12.5, 25, 50 and 75 mg/kg) groups, and 5-aminosalicylic acid (5-ASA) positive drug control group. Except using normal drinking water for control group, the mice in the other groups were treated with 2.5% DSS in drinking water for 7 days to induce the ulcerative colitis model. The mice in purple sweet potato anthocyanin treatment group and the 5-ASA positive drug control group were given the drug by intragastric gavage on the first day of modeling. The body weight of the mice and the hematocheziawere recorded every day. After continuous administration for 8 days, the mice in each group were killed and colon tissue was retained. Immunohistochemical technique (IHC) was used to detect the expression of tight junction protein ZO-1 and occludin and inflammatory factors tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in the colon of mice. The expression of mucin in goblet cells of colon tissue was observed by glycogen PAS staining. The protein expression of ZO-1, occludin, TNF-α were determined by Western blot, Sirius red staining was used to detect colonic fibrosis in mice. RESULTS Compared with control group, the disease activity index and histological injury of DSS model mice were significantly increased(P<0.01). Compared with model group, the disease activity index scores of the mice in different dose groups of purple sweet potato anthocyanin were decreased. The expression and distribution of ZO-1 and occludin in colon tissues were increased, and the expression and distribution of TNF-α and IL-6 in colon tissues were decreased. Glycogen PAS staining showed a significant increase in the distribution and expression of mucin in goblet cells of colon tissues in the purple sweet potato anthocyanin treatment group. Sirius red staining also showed that the degree of fibrosis in the purple sweet potato anthocyanin treatment groups was lower than that in model group. CONCLUSION Purple sweet potato anthocyanins has therapeutic effect on ulcerative colitis in mice induced by DSS, mainly through up-regulating the expression of tight junction proteins ZO-1 and occludin, to protect the integrity of intestinal barrier, inhibiting the expression of inflammatory cytokines TNF-α and IL-6 and intestinal fibrosis, to suppress the development of colonic inflammation.  相似文献

AIM:To investigate whether the panaxadiol saponins (PDS) and dexamethasone (DEX) have similar effects on lipopolysaccharide (LPS)-induced acute kidney injury (AKI). METHODS:C57BL/6 mice were randomly divided into 4 groups: the control mice received intraperitoneal injection of normal saline; in LPS group, the mice were subjected to intraperitoneal injection of LPS (10 mg/kg); in PDS + LPS group and DEX + LPS group, the mice were injected intraperitoneally with PDS (25.0 mg/kg) and DEX(2.5 mg/kg) 1 h before LPS injection, respectively. The blood was collected from the hearts, and the kidneys were collected for the biochemical and Western blotting analysis 12 h after LPS injection. RESULTS:LPS induced AKI, evidenced by markedly increased blood urea nitrogen (BUN) and creatinine (CREA) contents compared with control group (P<0.01). However, serum contents of CREA and BUN obviously reduced in PDS + LPS group and DEX + LPS groups compared with LPS group (P<0.05). Both PDS and DEX decreased the production of TNF-α and IL-6 by inhibiting renal NF-κB signaling activation. PDS and DEX also down-regulated the expression of inducible nitric oxide synthase, up-regulated the expression of manganese superoxide dismutase and reduced oxidative stress in the kidneys of LPS-challenged mice. In addition, treatment with PDS and DEX significantly increased the nuclear glucocorticoid receptor in the kidneys of LPS-treated mice. CONCLUSION:PDS and DEX have inhibitory effects on LPS-induced AKI mice. However, it is unclear whether PDS reduces LPS-induced AKI via direct action on glucocorticoid receptor.  相似文献

AIM:To determine the role of LOX-1/PPAR pathway in regulating expression of adhesion molecules elicited by oxidizing low density lipoprotein(Ox-LDL) through Lectin-like oxidized low-density lipoprotein receptor-1(LOX-1) in human umbilical vein endothelial cells(HUVECs). METHODS:HUVECs were incubated with Ox-LDL,poly(I),carrageenan or 15-deoxy-△12,14-prostaglanding J2 (15d-PGJ2 ). PPAR mRNA and protein were examined by real time RT-PCR and Western blotting. ICAM-1 and E-selectin were detected by RT-PCR and Western blotting respectively. RESULTS:Ox-LDL increased PPAR expression in HUVECs,which was inhibited by pretreatment of HUVECs with LOX-1 blockers. Preincubation of HUVECs with 15d-PGJ2 attenuated the expression of intercellular adhesion molecule-1(ICAM-1) and E-selectin in response to Ox-LDL. Upregulation of ICAM-1 and E-selectin mediated by Ox-LDL were suppressed more significantly by the combination of 15d-PGJ2 and polyinosonic acid as compared to either 15d-PGJ2 or polyinosonic acid alone. CONCLUSION:The results indicate that Ox-LDL exerts a biphasic effects on inflammatory response. It evokes harmful effects by inflammatory injury on one side and protective effects by triggering the LOX-1/ PPAR signaling pathway on the other hand.  相似文献

AIM: To investigate the mechanisms by which paeoniflorin (Pae) attenuates lipopolysaccharide(LPS)-induced acute lung injury in mice. METHODS: Male BALB/c mice were randomly divided into 4 groups: control,LPS, Pae+LPS, and Pae. Mice were administered intragastrically with double distilled water or Pae (20 mg/kg) once a day for 3 days. One hour after intragastrical treatment on the third day, LPS (20 mg/kg) or normal saline was injected intraperitoneally. Twelve hours after LPS challenge, the histological changes of the lung were observed, and histology score was also assessed. The myeloperoxidase (MPO),cytosolic phospholipase A2 (cPLA2) and phosphorylated cytosolic phospholipase A2 (phospho-cPLA2) in lung tissues were detected by Western blotting.RESULTS: LPS challenge resulted in acute lung injury, activated cPLA2 and increased MPO content in lung. Pretreatment with paeoniflorin significantly attenuated lung injury induced by intraperitoneal injection of LPS. The levels of MPO and phospho-cPLA2 in the lung tissues of the mice in Pae+LPS group were lower than those in LPS group (P<0.05).CONCLUSION: Pretreatment with paeoniflorin remarkably reduces LPS-induced acute lung injury through inhibiting phosphorylation of cPLA2 and decreasing neutrophil infiltration in the lung. These findings provide a new strategy for the prevention and treatment of LPS-induced acute lung injury.  相似文献

AIM: To establish the two-hit rat model with hemorrhage and lipopolysaccharide (LPS) and to study the effect of panaxadiol saponins (PDS) against acute lung injury.METHODS: Forty Wistar rats were divided randomly into 4 groups: sham operational group(S);two-hit groups with hemorrhagic shock-LPS (HL);dexamethasone pretreatment group (HLD) and PDS pretreatment group (HLP).The mean arterial blood pressure (MABP) was monitored dynamically by 4-channel physiological meter RM-6000,and pathological alteration of lung tissues was also observed.The levels of various serum enzymes,TNF-α and IL-6 were detected.RESULTS: MABP decreased in two-hit rat model with hemorrhage-LPS.The serum levels of aspartate aminotransferase,alanine aminotransferase,alkaline phosphatase,lactate dehydrogenase,creatine kinase,TNF-α and IL-6 increased significantly.Severe inflammatory change of pulmonary interstitium in HL group was also observed.CONCLUSION: Endotoxin injection following hemorrhage can be used to establish the animal model of acute lung injury.Similar with dexamethasone,PDS prevents lung tissue from seriously damage through increasing MABP and decreasing the level of serum enzymes,TNF-α and IL-6 levels.  相似文献
