Treatment of chronic bovine endometritis and factors for treatment success   总被引:1,自引:0,他引:1  
In a controlled field trial, 178 dairy cows with chronic endometritis and at least 21 days in lactation were randomly assigned to four different treatment groups: prostaglandin F2alpha intramuscularly (PG, 5 mg dinoprost (5 ml Dinolytic), n = 51), intrauterine antibiotics (AB; 400 mg ampicillin + 800 oxacillin (20 ml Totocillin), n = 49), intrauterine antiseptics (AS; 100 ml 4% Lotagen, n = 50); control (C, no initial treatment, n = 28). Before treatment, uterine swabs for bacteriologic examination and blood samples for determination of serum progesterone concentrations were collected. Two weeks following the first treatment, cows were reexamined. In case no clinical cure was diagnosed, treatment was repeated and control cows were treated for the first time with one of the three treatments mentioned above. The four treatment groups did not differ with respect to the clinical cure or reproductive performance. Therefore, factors that might have an influence on clinical cure and fertility were evaluated. With increasing duration of lactation, the clinical cure after a single treatment increased significantly over all treatment groups from 59.5% (treatment before day 42 postpartum) to 79.6% (treatment following day 42 postpartum) (P < 0.05). Within the PG group, a statistically significantly higher cure rate after a single treatment and first service conception rate and a lower pregnancy index were obtained when the treatment was performed following day 42 postpartum (P < 0.05). This was not the case in the other treatment groups. A retarded involution of the uterus based on the size had a negative effect on clinical cure over all groups (first treatment clinical cure: 68.2% (small uteri) vs 44.4% (large uteri); P < 0.05). Within groups, this effect was also detected, but only as a trend (P > 0.05). Isolation of Arcanobacterium (A.) pyogenes negatively influenced first treatment clinical cure over all treatment groups (79.0% vs 31.5%) and within treatment groups (P < 0.05). In the AB group, the pregnancy index, days open and the interval from first insemination to conception increased compared with the other treatment groups, when A. pyogenes was detected. Isolation of unspecific bacteria and the presence or absence of a corpus luteum only had minor effects over all and within the PG, AS and C group. Within the AB group, presence of luteal tissue was connected with a higher pregnancy index and increased days open and interval from first insemination to conception (P < 0.05). A multivariate analysis revealed that the presence of A. pyogenes and no initial treatment (control group) had a statistically significant independent and negative influence on the first treatment clinical cure. Ill-smelling vaginal discharge and no treatment had a negative independent effect on first service conception rate. Furthermore, days open were negatively influenced by the interval from calving to first examination and the presence of ill-smelling vaginal discharge. In conclusion, under the conditions of this field study, it seems preferrable to treat chronic endometritis. However, under the condition of a herd health program with continuous observation of sick and healthy animals, a wait-and-see attitude might also be justified.  相似文献

A study of bovine cutaneous dermatophilosis was carried out on a total of 6530 animals under different management systems. Of the 6530 animals examined, 334 (5.1%) were found to be positive. A significantly (P > 0.01) higher breed susceptibility was found in Holstein-Friesian cattle (12.8%) than in the local zebu (4.8%). The prevalence of the disease on feed-lots, a ranch, traditional farms and urban dairy farms was 2.9%, 4.2%, 7.1% and 12.8% respectively. Management and breed differences were found to be very important risk factors associated with the disease. However, other factors, including ticks, were also found to be apparently associated with the disease, indicating a complex interplay of several factors in the pathogenesis of dermatophilosis. Possible measures to be taken into account in the control of the disease are suggested.  相似文献

This study examined the effect of single IU administration of cephapirin on clinical recovery, clearance of uterine bacteria and reproductive performance of postpartum buffaloes with subclinical endometritis (SCE). Buffaloes (n = 86) at 35 days postpartum (DPP) with >10% polymorphonuclear (PMN) cells in endometrial cytosmears were designated as positive (SCEP, n = 29), and buffaloes with ≤10% PMN cell were designated as negative (SCEN, n = 57) for SCE. Out of 29 positive buffaloes, 15 were administered a single intrauterine dose of cephapirin benzathine on 40 DPP (SCEP‐CB), while the remaining 14 animals were kept as untreated control (SCEP‐C). All animals were observed regularly for oestrous signs and were again subjected to cytobrush sampling on the first postpartum (FPP) oestrus. Buffaloes positive for SCE at 35 DPP were later considered “recovered” if their PMN cells dropped to ≤5% on the FPP oestrus. Presence of Escherichia coli, Arcanobacterium pyogenes and Fusobacterium necrophorum in uterus was detected based upon PCR amplification of genes related to bacteria‐specific virulence factors. A total of 66.7% of SCEP‐CB group buffaloes recovered as compared to 28.6% in SCEP‐C (χ2 = 4.21; p < 0.05). Rate of bacterial clearance did not differ between treated (38.5%) and untreated buffaloes (8.3%) (χ2 = 1.67; p > 0.05). The median days to first service did not differ significantly (p > 0.05) among the three groups, whereas cephapirin administration reduced (p < 0.05) the days open by 14 days in SCEP‐CB compared to SCEP‐C buffaloes. SCEP‐CB buffaloes were as likely to conceive as SCEN, whereas SCEP‐C had 0.28 hazard ratio for pregnancy. In conclusion, a single treatment with cephapirin benzathine at 40 DPP improved the reproductive performance of buffaloes with subclinical endometritis.  相似文献

Although Escherichia coli is a commensal bacterium of the bovine vaginal microbiota, it is an important pathogenic bacterium that causes diseases of the reproductive tract and sub-fertility. Recent studies have focused on virulence factors (VFs) of intrauterine E. coli; however, actual endometrial VFs have not been clearly identified. The purpose of this study was to identify the VFs of E. coli associated with clinical metritis and endometritis. Thirty-two strains of E. coli and four mixed Trueperella pyogenes (TP) strains were detected in the uterus of 19 Holstein dairy cows with obvious clinical signs (between 8 and 66 days postpartum). The presence of six E. coli VFs (fimH, fyuA, kpsMTII, hra1, csgA, and astA) was examined by PCR, and clinical signs and reproductive performance (mixed TP, the percentage of polymorphonuclear neutrophils [PMN%], days to uterine involution, etc.) were evaluated. Four VFs (fimH, hra1, csgA, and astA) were detected in all E. coli strains, whereas fyuA and kpsMTII were detected in 94% and 50% of strains, respectively. Cows with E. coli strains harboring kpsMTII exhibited significantly severe clinical scores (vaginal discharge score, PMN%, uterine involution), suggesting that kpsMTII is a key VF for progression of clinical metritis and endometritis. In the present study, we clearly identified six VFs associated with clinical metritis and endometritis. In addition, E. coli strains with kpsMTII probably play a crucial role in the progression of clinical metritis and endometritis.  相似文献

We evaluated the efficacy of intrauterine plasma infusion in mares as a treatment for infertility caused by endometritis and distinguished the effects of intrauterine infusion of plasma vs saline solution. Forty-three subfertile mares were randomly assigned to 1 of 3 treatment groups: untreated controls (n = 14), those treated by saline infusion (n = 14), and those treated by plasma infusion (n = 15). Reproductive status was assessed daily by transrectal ultrasonography. Uterine aspirates and biopsy specimens were obtained 8 days after ovulation for cytologic and histologic evaluation, and mares were treated on days 12 to 16. Uterine aspirates and biopsy specimens were obtained again on day 8 of the next estrous cycle, and the mares were bred at the subsequent estrus. A postovulation intrauterine infusion of either plasma or saline solution was administered to mares in their respective treatment groups. Biopsy specimens were scored from 1 (no indications of inflammation) to 6 (severe inflammation). The pregnancy rate was lower (P less than 0.005) for mares with scores 5 and 6 (0/5) than for those with scores 1 to 4 (17/35). There was no significant effect of treatment nor a treatment by biopsy score interaction on pregnancy rate; however, the pregnancy rate for mares treated with plasma or saline solution (9/27) tended to be lower than for the control (untreated) mares (8/13). There was no change in mean biopsy score between specimens obtained before treatment and those obtained after treatment for the control group and the group treated with saline solution; however, there was a significant increase (P less than 0.05) in scores in the group treated with plasma.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Calves affected with citrullinaemia are clinically normal immediately after birth. In the majority of calves the clinical course of the disease was similar. Within 24 hours of birth they become depressed; then within 3 to 4 days were observed to wander aimlessly or stand with their head pressed against a wall or fence. By day 4 to 5 they become recumbent, developed convulsions, followed by collapse and death. Oedema of the cerebral cortex is a consistent histological lesion. Citrulline concentration in blood, cerebrospinal fluid, eye fluid and cerebral tissue is greatly elevated. Information gathered from pedigrees of affected calves indicate that the defect is widely disseminated throughout the Australian Friesian population.  相似文献

A certain level of endometrial bacterial infection and inflammation is involved in bovine uterine involution during the puerperal period. Factors that hamper normal uterine involution expose the uterine environment to pathological conditions, causing different endometritis levels. The lack of proper diagnostic tools extends the time to conception. Efforts have been made to elucidate the postpartum uterine environment, including bacterial flora, changes in transient endometrial inflammation, and the pathophysiology of endometritis, to improve bovine reproductive performance. E. coli and Trueperella pyogenes in the uterus are likely to cause persistent infection, and Mycoplasma bovigenitalium infection is associated with dystocia and cytological endometritis in postpartum dairy cows. Due to the widespread use of cytobrush as a diagnostic tool for bovine subclinical endometritis (SE) that enables quantification of the degree of inflammation, we found that endometritis at week 5 postpartum was associated with delayed first ovulation. Approximately 30% of open cows have SE during the postpartum period, and cows with low blood glucose during prepartum have a high risk of developing SE. Additionally, cows with purulent vaginal discharge do not always have endometritis but only vaginitis and/or cervicitis. Intrauterine infusion of polyvinylpyrrolidone-iodine (PVP-I) improves fertility and promotes endometrial epithelial cell regeneration after inducing transient uterine inflammation, suggesting that PVP-I could be a good alternative to antibiotics. In conclusion, prepartum management to prevent glucose deficiency, prompt diagnosis to identify causative agents and intrauterine inflammation levels, and appropriate treatment to minimize antimicrobial resistance is beneficial for tackling endometritis and improving reproductive performance in bovine herds.  相似文献

Non-methylated CpG motifs, present in viral and bacterial DNA, are one of many pathogen-associated molecular patterns (PAMP) recognized by the mammalian innate immune system. Recognition of this PAMP occurs through a specific interaction with toll-like receptor 9 (TLR9) and this interaction can induce cytokine responses that influence both innate and adaptive immune responses. Previous investigations determined that both the flanking sequences in synthetic CpG oligodeoxynucleotides (CpG ODN) and the cellular pattern of TLR9 expression can influence species-specific responses to CpG ODN. Therefore, the structure, function and cellular distribution of bovine TLR9 were compared with what is known for mice and human. Analysis of the bovine TLR9 gene revealed greater sequence homology between cattle and humans than cattle and mice Similar CpG motifs induced optimal activation of both human and bovine leukocytes and these motifs were distinct from those which activated mouse leukocytes. Functional analyses with CpG ODN stimulated bovine blood leukocytes revealed that class A CpG ODN were more potent inducers of interferon-alpha (IFN-alpha) than class B CpG ODN. Furthermore, magnetic activated cell sorting of bovine blood leukocyte subpopulations implicated dendritic cells but not monocytes in the regulation of CpG ODN-induced IFN secretion. Thus, the cellular pattern of CpG ODN-induced responses in cattle shared many similarities with human leukocytes. Collectively, these analyses revealed substantial conservation of TLR9 structure and TLR9 function in blood leukocytes of humans, cattle and other domestic species.  相似文献

The objectives of this study were to determine the risk factors for postpartum endometritis by evaluating several reproductive factors in individual cows, and to determine the effect of endometritis on the subsequent reproductive performance in dairy herds in Korea. The data, including health status, parity and body condition score (BCS) of cows, and calving date, were collected from 320 cows in eight dairy herds calving from January 2001 to October 2002. We used logistic regression to evaluate the effects of these factors on endometritis. A stepwise procedure, used to obtain the appropriate model with alpha=0.05, revealed that retained placenta, metabolic disorders and parity are the important risk factors for endometritis. The mean intervals from calving to first service and conception were prolonged (P<0.01) by 23 and 36 days, respectively, in the endometritis group compared to the non-endometritis group. The first service conception rate was lower (P<0.05) in the endometritis group (47.6%) than in the non-endometritis group (62.6%). The number of services per conception was higher (P<0.05) in the endometritis group (1.9) than in the non-endometritis group (1.6). We conclude that retained placenta, metabolic disorders and cow parity are strongly correlated with the development of postpartum endometritis, which decreases reproductive performance in dairy herds in Korea.  相似文献

Data from 3276 cows with 6598 lactations in 85 herds participating in a herd health monitoring programme were selected for an observational study. Cases of endometritis were diagnosed based on a clinical examination after 21 days postpartum. The cases were grouped by the severity of endometritis (mild, severe) and by the treatment performed (intrauterine infusion, prostaglandin injection, no treatment). The conception rates and the calving-to-conception intervals of these groups were compared statistically. Conception rate was lower in cows with severe endometritis compared to cows with mild endometritis while it did not differ between treatment groups. In cows with mild endometritis a significantly longer calving-to-conception interval was observed if intrauterine infusions were performed compared to the other two treatment options. The occurrence of other fertility problems during the same service period had a significant effect on conception rate and calving-to-conception intervals. In cows with severe endometritis intrauterine or prostaglandin treatments appeared to be beneficial, whereas intrauterine infusions in cows with slight endometritis had a negative effect on reproductive efficiency.  相似文献

The objective of this study was to characterize genetic, environmental, and economic factors related to the incidence of bovine respiratory disease (BRD) in feedlot calves. Records from 18,112 calves representing 9 breeds (Angus, Braunvieh, Charolais, Gelbvieh, Hereford, Limousin, Pinzgauer, Red Poll, and Simmental) and 3 composite types (MARC I, MARC II, and MARC III) over a 15-yr period (1987 to 2001) were evaluated. Disease incidence was observed and recorded by station veterinary and technical staff. The incidence of BRD varied across years, with the annual observed incidence ranging from 5 to 44%. From 1987 to 1992, the annual average incidence generally exceeded 20%. However, in later years the annual incidence did not exceed 14%. The epidemiological pattern indicated that BRD infection increased dramatically after 5 d on feed and remained high until approximately 80 d on feed. Previous BRD infection during the preweaning period did not influence subsequent BRD infection in the feedlot. Steers were more likely to become sick with BRD than heifers; castration before entry in the feedlot may be a predisposing cause. Few significant differences among breeds were detected for BRD incidence. Adjusted solutions from mixed model analyses indicated that Herefords were generally more susceptible to BRD infection (P < 0.05) than MARC I and III composite types. Composite breed types had similar susceptibility compared with other purebred breeds. Mortality associated with BRD was greatest in Red Poll calves (9%) compared with the average over all breeds (4%). Estimates of heritability for resistance to BRD ranged from 0.04 to 0.08 +/- 0.01. When the observed heritability was transformed to an underlying continuous scale, the estimate increased to 0.18. Selection for resistance to BRD could be effective if phenotypes for BRD resistance were known. Thus, development of an inexpensive and humane method of challenging animals with BRD to determine resistance would be an important step in reducing the incidence of BRD. This study also demonstrated that producer-collected field data could be used for selection against this disease. The economic loss associated with lower gains and treatment costs for BRD infection in a 1,000-cattle feedlot was estimated as dollar 13.90 per animal, not including labor and associated handling costs.  相似文献

Based on our clinical experience on bovine mastitis, we hypothesized that subtypes of Staphylococcus aureus (S. aureus) exist which differ in their contagious and pathogenic properties. In order to investigate this hypothesis, we analyzed strains of S. aureus isolated from spontaneous intramammary infection (IMI) with their virulence gene patterns and genotypes obtained by PCR amplification of the 16S–23S rRNA intergenic spacer (RS–PCR). The genotypes were then associated with epidemiological and clinical data including 26 herds. The results demonstrated a high association between genotypes and virulence gene patterns as well as between epidemiological and pathogenic properties of S. aureus. In particular, genotype B was related to high contagiosity and increased pathogenicity whereas the other types (C, OG) were found with infection of single cows. Because of the high clinical relevance, our results indicate the need to subtype the IMI-associated strains of S. aureus in the future.  相似文献

Bovine herpes mammillitis was diagnosed in a dairy herd with udder and teat skin lesions. Clinical symptoms seen in 6 cows consisted of round dry areas at the teats as well as large red and painful areas with crust formation at the teats, the teat basis and the udder. Diagnosis was verified by demonstrating numerous virus particles with the typical herpes structure and by BHV-2 serum neutralization test. Prevalence of BHV-2 in the herd was determined by using BHV-2 SNT at 7 occasions during a period of 15 months. The relatively low BHV-2 SNT-titres as well as the seasonal increase of BHV-2 titres and seroprevalence in the month of September were indicative of a chronic and latent BHV-2 infection in the herd.  相似文献

The aim of the study was to evaluate phagocytic and killing activity of phagocytic cells in blood and uterine flush of cows with endometritis before and after intrauterine (i.u.) administration of cephapirin and methisoprinol. The research was carried out on 28 cows with clinical endometritis. Animals were divided into four groups, each composed of seven cows, depending on the i.u. treatment used: Group A—cephapirin; Group B—methisoprinol; Group C—cephapirin and methisoprinol at the same time; and a control group—without medication. Using flow cytometry technique, the phagocytic activity of granulocytes and monocytes was identified, as well as the oxidative burst activity of neutrophils in the peripheral blood and uterine washings. Summarizing the results of the research, i.u. infusion of cephapirin caused a reduction in the phagocytic and killing activity of phagocytes. The i.u. use of methisoprinol increased phagocytic and killing activity of phagocytes in the uterus. Administering both listed substances simultaneously showed a decrease in phagocytosis, presumably due to the dominating inhibitor effect of the antibiotic. However, also an increase of mean fluorescence intensity was observed, presumably caused by the methisoprinol. Intrauterine use of immunostimulatory substances, can improve the effectiveness of the treatment of endometritis in cows.  相似文献
