A collection formed from 53 accessions of white clover stolonswas carried out in naturalized populations from Argentina betweenparallels 26° and 37° South latitudes and 58° to 62°West longitude. Almost half of them (43%) wereobtained from grazed sites. Collected samples were grown in the fieldat Pergamino Experimental Station and compared with a local cultivar.Attributes scored or measured were petiole length, width and lengthof central leaflet, relation width/length, height, growing habit,leaf mark, stolon thickness and density, days to flowering,cyanogenesis, seed yield and winter herbage yield. There weresignificant correlations among seven pairs of characters. Allpopulations were cyanogenic within a range from 40 to 100%.Populations from grazed sites had prostrate habit and lower canopyheight than those from non grazed sites. The collection was clusteredin seven groups. Differences among groups were attributed toagroecological differences in sites of collection. Results obtainedencouraged in Argentina further collection missions for naturalizedpopulations of white clover.  相似文献

Detailed karyotypes and 4C DNA amounts have been studied in five cultivars of Cajanus cajan and 20 species belonging to Cajanus, Rhynchosia, Dunbaria, Flemingia and Paracalyx. C. cajan shows intraspecific variability and its karyotype is most similar to that of C. cajanifolius (sect. Cajanus) in the morphology and number of satellite chromosomes and the lack of any correlation between chromosome size within the complement and asymmetry. Karyotypes of C. lineatus and C. sericeus belonging to sect. Atylia are similar with respect to maximum r-index and the ratio of longest and shortest chromosomes in their respective complements. C. acutifolius (sect. Frutocosa) is distinct in having a very low ratio (1.44) between the longest and the shortest chromosomes, while in C. albicans, C. goensis, C. scarabaeoides (sect. Cantharospermum) there is no chromosome pair with r-index >2.0. C. mollis and C. volubilis (sect. Volubilis) show similarity with regard to ratio between the longest and the shortest chromosomes and a distinct chromosome pair with an arm ratio of 2.7 and C. platycarpus is distinct (sect. Rhynchosoides) in having the smallest ratio (1.36) between the longest and the shortest chromosomes. 4C DNA amounts in Cajanus and Rhynchosia vary between 3.28 pg to 11.69 pg and 4.92 pg to 11.13 pg respectively, while in Dunbaria, Flemingia and Paracalyx these vary between 4.70 and 7:06 pg. In the genus Cajanus karyotypic features and 4C DNA amounts agree with the sectional classification and bring out a close relationship between C. cajan and C. cajanifolius. This is supported by studies based on seed protein patterns, isozyme analysis, trypsin and chymotrypsin inhibitor patterns, RFLP and RAPD data and crossability relationships. A clear relation between DNA amount and abnormalities effecting the extent of pairing and recombination in interspecific hybrids shows the importance of this study in developing future breeding programme involving C. cajan and its wild relatives that are of potential value.  相似文献

Changes in microbial C, N, and P were investigated for 1 year in two soils with similar physicochemical properties but supporting different crops under subtropical conditions. One was cropped with palmarosa (Cymbopogon martinii L.) and the other with Japanese mint (Mentha arvensis L.). Both the season and the type of cropping had a significant influence on changes in the soil microbial biomass. In general, soil microbial biomass C, N, and P were highest in summer months and lowest in midwinter. Soil microbial biomass levels and microbial C:N and C:P ratios were higher and N:P ratios lower under palmarosa soil than under mint.  相似文献

Brassica juncea (2n = 36, AABB) is an amphidiploid derived from its diploid progenitor species B. rapa (2n = 20, AA) and B. nigra (2n = 16, BB). Resynthesis of B. juncea by exploiting the wider gene pool of the present day diploids offers a powerful tool for obtaining novel genetic variation. Hybridization between the two species is difficult because of the presence of crossability barriers. Barriers to hybridization were identified using Aniline Blue Fluorescence (ABF) method. Crosses were classified as having pre- or post-fertilization barriers based on whether less than or more than one per cent ovules showed pollen tube entry. Of the 23 crosses studied, seven crosses all with B. rapa as the female parent showed pre-fertilization barriers. Only 0 to 0.3 per cent ovules were fertilized in these crosses. The remaining 16 crosses, four with B. rapa as the female parent and 12 with B. nigra as the female parent, showed post-fertilization barriers. In crosses with pre-fertilization barriers use of bud pollination and stump pollination ensured or increased fertilization. These two simple techniques were used in combination with ovary-ovule culture to ensure recovery of hybrids. In crosses with post-fertilization barriers, ovary-ovule culture alone helped to obtain hybrids. Hybrids were obtained at higher frequency with the use of direct ovule culture compared to ovary-ovule culture. The F1 plants obtained in vitro were multiplied on MS medium + 2 mg/l Kn + 0.2 mg/l IAA. These F1 plants were confirmed as true hybrids through morphology, leaf isozymes and cytology. The root-knot nematode, Meloidogyne javanica (Treub) Chitwood is an important parasite of chickpea (Cicer arietinum L.). Four chickpea genotypes were evaluated for tolerance to M. javanica in naturally infested fields at three locations. Each genotype was evaluated for number of galls, gall size, root area covered with galls and number of egg masses produced. All the cultivars were susceptible or highly susceptible. Seed yield, weight of 100 undamaged seeds, total dry matter and plant height were compared with checks. Chickpea cultivar Annigeri and a local check were used as nematode susceptible checks in all locations. The four promising nematode tolerant genotypes produced significantly greater yield and total dry matter than the checks in fields naturally infested with M. javanica at three locations. These M. javanica tolerant lines represent new germplasm and they are available in the chickpea genebank at the International Crops Research Institute for the Semi-Arid Tropics (ICRISAT) bearing the identification numbers ICC 8932, ICC 11152, ICCV 90043 and ICCC 42.  相似文献

Detection of DNA polymorphism in cultivated pigeonpea (Cajanus cajan) and two of its wild relatives Cajanus volubilis and Rhynchosia bracteata is reported here for the first time using amplified fragment length polymorphism (AFLP) fingerprinting. For this purpose, two EcoRI (three selective nucleotides) and 14 MseI (three selective nucleotides) primers were used. The two wild species shared only 7.15% bands with the pigeonpea cultivars, whereas 86.71% common bands were seen among cultivars. Similarly, 62.08% bands were polymorphic between C. volubilis and pigeonpea cultivars in comparison to 63.33% polymorphic bands between R. bracteata and pigeonpea cultivars, and 13.28% polymorphic bands among pigeonpea cultivars. The cluster analysis revealed low polymorphism among pigeonpea cultivars and very high polymorphism between cultivated pigeonpea and its wild relatives. The AFLP analysis also indicated that only one primer combination (EcoRI + ACT and MseI + CTG), at the most any four primer pair combinations, are sufficient for obtaining reliable estimation of genetic diversity in closely related cultivars like pigeonpea material analyzed herein. AFLP analysis may prove to be a useful tool for molecular characterization of pigeonpea cultivars and its wild relatives and for possible use in genome mapping.  相似文献

A rhizosphere application of NO inf3 sup- and/or naringenin affected the Pisum sativum — Rhizobium leguminosarum biovar viciae symbiosis. NO inf3 sup- (5 mM) lowered while naringenin raised the nodulation status (nodule numbers and weight) and nodule efficiency (C2H2 reduction activity). However, the inhibitory effect of NO inf3 sup- was to some extent alleviated when applied in combination with naringenin. The plant biomass was increased by the application of NO inf3 sup- and naringenin, either alone or in combination, while a higher root: shoot ratio was observed only in the naringenin-treated plants. Root flavonoids are known to regulate the expression of nod genes; their high-performance liquid chromatography profile was influenced in different ways by NO inf3 sup- and naringenin.  相似文献

A greenhouse study was performed to examine the ability of the earthworms Microscolex dubius and Aporrectodea trapezoides to transport Rhizobium meliloti L5-30R through soil. When R. meliloti L5-30R was inoculated into either ezi-mulch (a pelleted formulation of cereal-pea straw), oat hay, pea hay, or sheep dung and placed on the soil surface together with either A. trapezoides or M. dubius, >104 colony-forming units (CFU) of R. meliloti L5-30R g-1 soil were detected at 90 mm soil depth after 18 days. In the absence of earthworms, R. meliloti L5-30R was not detected at 90 mm soil depth after this time. In a second experiment using ezi-mulch as the inoculant material and in the presence of A. trapezoides (in a number equivalent to 471 or 785 m-2), ca. 103 CFU of R. meliloti L5-30R per 10 mm of alfalfa root were detected at 0–30, 30–60, and 60–90 mm soil depth after 18 days, while <3 CFU were detected per 10 mm of root in the absence of A. trapezoides. In a third experiment in which R. meliloti L5-30R was distributed evenly through soil at the start of the experiment, A. trapezoides (in a number equivalent to 157, 471, or 785 m-2) significantly decreased the survival of L5-30R in soil after 40 days of incubation at 15°C, but not after 20 days. The decrease in survival of R. meliloti L5-30R was correlated with the density of A. trapezoides. These results demonstrate that A. trapezoides can increase root colonization of alfalfa by R. meliloti L5-30R, but may also reduce the ability of R. meliloti L5-30R to survive in soil.  相似文献

Effects of soil amendment with crabshell chitin on the growth of white clover (Trifolium repens L.) and perennial ryegrass (Lolium perenne L.), and on populations of soil bacteria, fungi, and plant-parasitic and free-living nematodes were investigated in a pot trial. Five soil samples were collected from Te Puke (Paengaroa Shallow Sand, a Typic Hapludand) and five from Hamilton (Bruntwood silt loam, an Aquic Hapludand), New Zealand. Subsamples of each soil were either amended with chitin or unamended and planted with white clover and ryegrass. The ryegrass shoot weight in amended soil was greater (P<0.01), most probably due to N mineralised from chitin. A significantly lower (P<0.01) root: shoot ratio of ryegrass in the amended soil also suggested improved N availability, and therefore less root mass was needed to support a given shoot mass. A reduction in nodulation was observed in 12-day-old white clover seedlings (P<0.05) and also in 6-week-old seedlings (P<0.01). The shoot weight of white clover was significantly lower (P<0.05) in amended soil, possibly due to phytotoxic effects of chitin. Chitin increased (P<0.01) the populations of bacteria and fungi by 13-fold and 2.5-fold, respectively. The cyst nematode of white clover, Heterodera trifolii, was significantly reduced in chitin-amended soil, possibly due to increased levels of chitinase produced by rhizosphere microorganisms. Two other plant-parasitic nematodes, Pratylenchus spp. and Tylenchus spp., were also reduced in ryegrass roots and in soil as a result of the chitin amendment. However, the total number of free-living nematodes increased 5.4-fold in amended soil.  相似文献

Summary The kinetics of Zn absorption were studied in mycorrhizal (Glomus macrocarpum) and non-mycorrhizal roots of corn (Zea mays L.) at pH 6.0 at Zn concentrations of 75 mol to 1.07 mol m-3. Five concentration-dependent phases of Zn absorption were recognized; phase 0 (1.5–4.0 mmol m-3) was linear but the other four phases (4.0 mmol to 1.07 mol m-3) obeyed Michaelis-Menten kinetics. At low concentrations (less than 4 mmol m-3), sigmoidal kinetics of Zn absorption were observed. The absorption of Zn by mycorrhizal maize was greater at low concentrations but decreased at higher levels. This appeared to be a result of a higher maximal uptake rate in phase 1 and lower K m values in the subsequent phases. Genetic and phenotypic diversity among randomly selected 36 downy-mildew-resistant sorghum accessions were assessed, the former using 10 simple sequence repeat (SSR) marker loci and the latter using 20 phenotypic traits. The number of alleles (a j ) at individual loci varied from five to 14 with an average of 8.8 alleles per locus. Nei's gene diversity (H j ) varied from 0.59 to 0.92 with an average of 0.81 per locus. High gene diversity and allelic richness were observed in races durra caudatum (H j = 0.76, a j = 4.3) and guinea caudatum (H j = 0.76, a j = 3.8) and in east Africa (H j = 0.78, a j = 7.2). The regions were genetically more differentiated than the races as indicated by Wright's F st. The pattern of SSR-based clustering of accessions was more in accordance with their geographic proximity than with their racial likeness. This clustering pattern matched little with that obtained from phenotypic traits. The inter-accession genetic distance varied from 0.30 to 1.00 with an average of 0.78. Inter-accession phenotypic distance varied from 0.01 to 0.55 with an average of 0.33. Eleven accession-pairs had phenotypic distance of more than 0.50 and genetic distance of more than 0.70. These could be used as potential parents in a sorghum downy mildew resistance-breeding program.  相似文献

The domestication of cultivated flax (Linum usitatissimum L. subsp.usitatissimum) is briefly discussed. Using data documented as a matter of routine in genebank work, 63 accessions of cultivated flax from the flax germplasm collection of the Gatersleben Genebank are compared with 73 accessions of its wild progenitor pale flax (subsp.angustifolium (Huds.) Thell.), which have been observed in systematic field trials. Range of variation, genetically based variation, heritability and correlation of several characters are considered, especially with respect to the influence of domestication. Cultivated flax shows higher variation in the characters of generative plant parts, while pale flax varies more in the vegetative parts of the plant. The character correlations are similar in both subspecies. Of the 29 characters studied in pale flax the parameters describing tillering, height of plant, weight of seeds, width of petals and width of leaves are especially suitable for distinguishing between different accessions. In spite of the high heritabilities of most of the characters, the influence of the environment is significant in most cases.  相似文献

Summary There was no correlation between the quantity of exopolysaccharide produced and acetylene reduction activity by Rhizobium spp. or by Bradyrhizobium spp. (Cajanus). The exopolysaccharide-defective mutants of Rhizobium sp. strain P 116 either failed to nodulate or showed a decrease in effectiveness. The deficiency in exopolysaccharide production was corrected by the addition of purified exopolysaccharide from the parent strain, or from Bradyrhizobium sp. strain P 149 or S24 isolated from pigeonpea (Cajanus cajan) and mungbean (Vigna radiata), respectively. However, the nodules so formed were not fully effective compared to those formed by the parent strain.  相似文献

Genetic diversity was investigated in 348 accessions and subaccessions of grasspea (Lathyrus sativus L.) from 10 geographical regions. Polymorphism for 20 isozymes of 13 enzyme systems was studied to estimate the genetic diversity. The Near East and North Africa regions included the most variability for these isozyme systems, suggesting that the center of diversity (center of origin) for grasspea is in this general area. The lowest variability was found in accessions and subaccessions from South America, followed by those from Sudan–Ethiopia. Diversity was measured for individual loci over regions and EST-1 and SKDH had the highest genetic diversity. The closest genetic diversity was observed for LAP-2, followed by AAT-1 and PGM. The closest genetic distance existed between populations from the Near East and North Africa. Populations from South Asia and Sudan–Ethiopia, though geographically widely separated, exhibited a closer genetic distance from each other than from other regions.  相似文献

Genetic diversity in 12 landraces of bambara groundnut (Vigna subterranea), an indigenous African legume, was evaluated using Random Amplified Polymorphic DNA (RAPD) markers. DNA from individuals of each landrace was also analysed to determine the level of heterogeneity within landraces. RAPDs revealed high levels of polymorphism among landraces. The percentage polymorphism ranged from 63.2% to 88.2% with an average of 73.1% for the 16 RAPD primers evaluated. The construction of genetic relationships using cluster analysis groups the 12 landraces in two clusters. RAPDs are useful for the genetic diversity studies in V. subterranea and can identify variation within landraces.  相似文献

Argan (Argania spinosa (L.)Skeels) flowering was observed at Ait Melloul, Ait Baha andArgana, three sites in the south-west of Morocco during twoseasons, 1994–95 and 1995–96. Glomerule count wasmonitored on first-year and on last-year growth shootevery twenty days. Season was the primary source of variation ofglomerule count on argan first-year and last-yeargrowth shoots. The first season was very dry when flowering beganlate April, while the second was very humid and flowering startedearly December. In both cases flowering precocity was associated toprecipitation precocity, i.e. flowering would commence whenapproximately 100 mm rain was accumulated. High positivecorrelation between glomerule count and precipitation were observed.But, considering the two seasons, glomerule count always attained itspopulation maximum in spring even if differences were observedbetween sites. However, intensity and duration of the peak were alsodistinct depending on shoot type: on first-year growth shoot,glomerules were abundant but would last for a short period, whereason last–year growth shoot, they were less frequent but wouldlast for a longer period. Not all trees flowered during a season,although some trees flowered every season, showing the extremes ofthe large variability observed for glomerule count abundance andduration within argan populations.  相似文献

Wheat endosperm storage proteins, namely gliadins and glutenins, are the major components of gluten. They play an important role in dough properties and in bread making quality in various wheat varieties. In the present study, the different alleles encoded at the 6 glutenin loci and at 3 -gliadin loci were identified from a set of 200 hexaploid wheat cultivars grown primarily in France using SDS PAGE. At Glu-A1, Glu-B1 and Glu-D1, encoding high molecular weight glutenin subunits (HMW-GS), 3, 8 and 5 alleles were observed respectively. Low molecular weight glutenin subunits (LMW-GS) displayed similar polymorphism, as 5 and 11 alleles were identified at loci Glu-A3 and Glu-B3 respectively. Four alleles were observed at Glu-D3 loci. Omega-gliadin diversity was also very high, as 7, 13 and 9 alleles were found at Gli-A1, Gli-B1 and Gli-D1, respectively. A total of 147 (or 149) patterns resulted from the genetic combination of the alleles encoding at the six glutenin loci (or Glu-1 and Gli-1 loci). Although Glu-1 and Glu-3 loci were located on different chromosome arms and were theoretically independent, some associations were revealed due to pedigree relatedness between some French wheat cultivars. The usefulness of allelic identification of LMW-GS together with HMW-GS and gliadins for future genetic and technological wheat improvement is discussed.  相似文献

Summary We tested the response of the wetland rice cultivar Prakash to inoculation with ten vescular-arbucular mycorrhizal (VAM) fungi (three selected from the first screening and seven isolated from local paddy fields) in a pot experiment under flooded conditions in order to select the most efficient mycorrhizal fungi to inoculate the rice nursery. A sandy clay loam soil was used as the substrate, fertilized with the recommended N and K levels (100 kg N ha–1 as ammonium sulphate and 50 kg K ha–1 as muriate of potash) and half the recommended level of P (25 kg ha–1 as super phosphate). The inoculation was made into dry nursery beds and the beds were flooded when the seedlings were about 25 cm high, in 15 days. Twenty-eight-day old seedlings were transferred to pots filled with well puddled soil flooded with 5 cm of standing water. Based on the increase in grain yield and total biomass, Glomus intraradices and Acaulospora sp. were considered efficient and suitable for inoculation into rice nurseries.  相似文献

New Potentilla synonyms of the Fragaria species names are discussed and considered to be dispensable. Further unnecessary nomenclatural changes based on the argument to recognize monophyletic but not paraphyletic taxa should be avoided to maintain nomenclatural stability for global communication about plant genetic resources. The new combination Fragaria×rosea (Mabb.) K. Hammer et Pistrick is presented for the hybrid Potentilla palustris (L.) Scop. ×Fragaria×ananassa Duchesne ex Rozier.  相似文献
