The uterine effects of oxytocin, the prostaglandins dinoprost and cloprostenol as well as clenbuterol, ergometrin, xylazine and Utrorale were investigated in 8 cows during late pregnancy, parturition and early puerperium (until 4th day p. p.). Uterine motility was measured by means of pressure microsensors and electrodes which were surgically implanted 3 to 4 weeks before parturition. Hysterograms were characterized by means of pressure amplitude, frequency and duration of uterine contractions and also by electromyography. Oxytocin (2-5 IE) given intravenously always provoked strong uterine contractions until the 4th day p.p. From the prostaglandins examined during early puerperium only dinoprost (15 mg i.v.) produced uterotonic effects, while the synthetic analogue cloprostenol (0.25 mg i.v.) had a weak stimulatory activity only on day 1 p.p. Both prostaglandins were ineffective when injected intramuscularly. Clenbuterol (0.3 mg i.v.) a beta 2-mimetic compound effectively induced long lasting tocolysis during parturition, which could be abolished by oxytocin. Xylazine (10 mg i.v.) was able to significantly increase uterine motility during late gestation. Following intravenous administration of ergometrin (1 and 10 mg), bunitrolol (1-16 mg) and Utrorale (0.1-4 ml) including its compounds oleum sabinae, oleum terebinthinae, balsamum copaivale and Styrax no uterokinetic activity was recorded at any time.  相似文献

The aim of this study was to test the functional status of uterine oxytocin receptors in cows in vivo around parturition. The animals received consecutive, intra‐arterial injections of 800, 1600 and 3200 mU of oxytocin at three different stages: during late gestation (days 260–274), at 12 h and at 24 h after intramuscular injection of a prostaglandin F analogue at day 275 to induce parturition. Cows (n=6) had been provided with myometrial electrodes and a catheter had been installed in the aorta and in a branch of the uterine vein (UV). Regular blood samples were obtained from the UV from 5 min before until 45 min after each oxytocin injection to measure plasma levels of prostaglandin F (PGF) and oxytocin. Uterine electromyographic (EMG) activity was registered continuously during each experiment. The increase of oxytocin levels in UV plasma after intra‐arterial injections was dose dependent (p < 0.02). Pre‐ and post treatment oxytocin levels at 24 h after induction of parturition were significantly increased (p=0.0313). Both during late pregnancy and at 12 h after induction of parturition, oxytocin caused a significant increase in EMG activity (p=0.022). After the 3200 mU dose the increase was significantly higher than with the other 2 doses (p=0.004). After each dose, EMG activity returned to baseline levels within some 15 min. At 24 h after induction of parturition, the pre‐treatment level of EMG activity had increased. Doses of 800 mU and 1600 mU of oxytocin produced a significant (p=0.022) increment of EMG activity, which was of the same magnitude as during the preceding stages; after 3200 mU of oxytocin the response was significantly higher than before (p=0.008). No significant increases of PGF levels in UV plasma could be measured after oxytocin injections at any of the three stages. It is concluded that the myometrium of the pregnant cow responds in vivo to physiological doses of oxytocin. At 24 h after induction of parturition, when luteolysis has occurred and a parturient pattern of parturient myometrial activity has already started to develop, the response is enhanced. Physiological doses of oxytocin did not evoke a spurt release of PGF in uterine venous blood during the peripartal period.  相似文献

Contents: Uterine activity, i.e. myometrial contractions, were recorded in 11 sows prior to and during parturition by using strain gauges placed onto uterine horns about 15 to 20 cm cranial of the bifurcation. Tocographic patterns observed prior to and during spontaneous and alfaprostol induced parturitions were normal. Administration of 20, 10 or 5 I.U. oxytocin 22 hours after alfaprostol quickly initiated abnormal periparturient contraction patterns with 20 and 10 I.U. oxytocin and in one of two animals receiving 5 I. U. These were characterized initially by prolonged tetanic contractions resulting in delayed deliveries, and later by the premature occurrence of patterns seen normally only during the phase of placental expulsions, although not all piglets had been delivered. To quantify the pattern of myometrial activity during gestation and parturition, one bipolar electrode was implanted on each uterine horn of four bilaterally pregnant pygmy goats between days 102 and 120 of gestation. After a recovery period, electromyographic recordings were made for at least six hours per day and continuously when parturition was judged to be imminent. During late gestation myometrial activity occurred as discrete episodes of myometrial electrical activity (EMEA) with a mean duration ranging from 6.2 +/- 2.07 to 8.3 +/- 1.60 minutes. The mean interval between two successive EMEAs ranged from 45.8 +/- 19.95 to 74.7 +/- 42.27 minutes. In three of the four goats these characteristics were not significantly different from the two uterine horns. Parturition was preceded by a prolonged period (eight to 12 hours) of myometrial quiescence. It was only from 19 to 15 hours before expulsion of the first kid that total duration of EMG activity increased. This finally resulted in the regular occurrence of bursts which occupied 25 to 30 per cent of the recording time. The results demonstrate that, as in the sheep, cow and pig, the myometrium is active during late pregnancy. It is postulated that luteolysis coincides with the prolonged period of myometrial quiescence which precedes the onset of the parturient pattern of uterine activity.  相似文献

The objective of this study was to confirm in various breeds of dogs the efficacy and safety of a parturition induction treatment described to be successful in Beagle dogs. Parturition was induced in seven various sized pregnant bitches of different breeds, with 15 mg aglepristone per kg at day 59–61 post-estimated ovulation day, followed 24 h later by 0.15 IU oxytocin per kg subcutaneous injections every 2 h. Two bitches were small-sized bitches (<10 kg), three bitches were large-sized bitches (30–40 kg) and two bitches were giant bitches (>40 kg). The results were compared to a control group (n = 6), in which bitches underwent a natural delivery in the same environmental conditions as the induced group. In the induced group, parturition was successfully induced in 7/7 bitches. The first pup in a litter was born on average 25.9 ± 3.29 h after aglepristone administration (21–30 h). Two of seven bitches from the small-sized group delivered some of their pups before the first administration of oxytocin. The mean duration of parturition was 9.6 ± 5.4 h vs 8.0 ± 4.8 h in the control group. The mean interval between two successive pups being delivered was 115.6 ± 82.8 min (34–265) vs 68.8 ± 24.5 min in the control group (p < 0.03). The mean weight at parturition did not differ significantly between the two groups. One litter of four Yorkshire Terrier pups in the induced group were premature at the time of birth and died between 19 and 29 h post-delivery. This study, although on a very limited number of dogs, confirms the efficacy of the aglepristone/oxytocin protocol to induce parturition in dogs.  相似文献

This paper will examine the augmentation of oxytocin (OT) secretion in the final day before parturition in the pig, and review the key endocrine systems of influence. Recent experiments have shown that increases in plasma OT above baseline secretion were not seen until 7 hours prior to the birth of the first piglet. Progesterone release begins to decline some days before birth and is unlikely to provide such acute control of OT release immediately before birth. The involvement of relaxin is complex but it is possible that the prepartum relaxin surge, which ends close to the OT rise, attenuates OT secretion to prevent excessive myometrial activity until cervical softening has occurred. Increasing myometrial electromyographic activity appears to be seen as relaxin concentrations decline and OT increase. Endogenous opioid peptides within the hypothalamus exert a profound influence upon OT secretion during birth, and some effects are also seen prepartum. It has been proposed that this system may allow parturition to be interrupted or slowed in response to changes in environment. The therapeutic choice of exogenous injections of OT to control the onset time of parturition will be discussed in the light of current physiological knowledge.  相似文献

Application of new procedures in the sphere of the control of sexual functions requires an extension of present knowledge of postparturient endocrinium or endogenic factors comprised in postparturient physiology of sexual activity. According to recent data, oxytocin, besides its uterotonic and luteolytic activity, acts as an ovarian factor in the local intrafollicular regulation of stereidogenesis and as a modulator of uterine secretion of prostaglandines. Based on present knowledge of oxytocin effects, this study was aimed at investigation of the influence of repeated carbetocin (Depotocin inj. Spofa) administration on the dynamics of changes in thyroxine (T4), triiodothyronine (T3), 17 beta-estradiol (E2), progesterone (P4) concentrations and their mutual correlations from the 36th hour till the 51st day after parturition. Simultaneous study of a possible delayed influence of applied carbetocin on conception of ewes after oestrus evocation on day 51 after lambing was carried out. Nineteen ewes of the Slovak Merino breed, lambed in the first decade of February, were assigned to the experimental (n = 9) and to the control group (n = 10). Experimental ewes were subjected to repeated postparturient carbetocin treatment at the dose 0.07 mg per animal. The first dose was applied i. m. in 24 hours, and the second in 72 hours after parturition. On day 51 oestrus was induced in nine ewes of each group by combined treatment with chlorsuperlutin (Agelin, vaginal pessaries, Spofa) and PMSG (500 I.U./animal). On the day of PMSG application ewes were housed together with rams for the period of the next six days. Samples of blood were taken 24 hours before parturition (-1st day), up to 36 h after parturition and on days 4, 7, 14, 17, 21, 25, 34, 42 and 51 after parturition. Concentrations of T4, T3, E2 and P4 were determined by commercial kits RIA-test-T4; RIA-test-T3; RIA-test-ESTRA and RIA-test-PROG (URVJT Kosice). Animal of the control group showed variations of T4 concentrations (Tab. I, Fig. 1) at the level of original values (59.4 +/- 9.69 nmol.l-1) up to the 21st day with the exception of temporary drop on day 4 and rise on day 7, insignificant compared to the -1st day. T4 concentrations of the control group displayed an intermittent increasing trend with the statistically insignificant peak after 36 h and on day 17, compared to the -1st day. The influence of the suckling stimulus and ovarian secretions on LH response to naloxone was studied in 16 postpartum anestrous beef cows that were assigned randomly to one of four groups (n = 4/group): intact suckled (IS), intact nonsuckled (IN), ovariectomized suckled (OS) or ovariectomized nonsuckled (ON). Ovariectomy (OS + ON) and calf removal (IN + ON) were performed on d 2, 3 or 4 after parturition. Jugular venous blood was collected at 15-min intervals for 4 h before and 4 h after administration of naloxone (1 mg/kg BW, i.v.) on d 14 and d 28 after parturition. Gonadotropin-releasing hormone (5 micrograms, i.v.) was given 3 h after naloxone. Both IN and OS increased (P less than .05) mean pretreatment LH above IS values (mean +/- SE, ng/ml; IS 1.6 +/- .1 vs IN 2.5 +/- .3 and OS 2.7 +/- .4; P less than .01), whereas ON increased (P less than .01) LH (3.7 +/- .3 ng/ml) even further. Mean LH increased (P less than .05) after naloxone administration in all treatment groups. However, magnitude of this response was variable and dependent on ovarian status. Amplitude of the naloxone-induced LH response was greater (P less than .05) for ovariectomized (5.9 +/- 1.1 ng/ml) than for intact groups (2.7 +/- .5 ng/ml). Gonadotropin-releasing hormone increased mean LH concentrations in all groups. The effects of nutrition during the last two trimesters of gestation on GnRH-induced LH release were assessed in crossbred heifers. Heifers (n = 58) were allotted at 90 d gestation to one of three levels of an experimental diet fed at 1, 1.5 or 2% of BW to attain maternal BW loss, BW maintenance or BW gain, respectively, at parturition. Twenty-two heifers were injected (i.m.) once with 100 micrograms GnRH between d 14 and 1 before parturition, and 32 heifers were injected (i.m.) once with 100 micrograms GnRH between d 8 and 21 after parturition. Jugular blood samples were collected before and at 30-min intervals after GnRH for 4 h. Least squares means for BW change differed (P less than .01) among BW loss (-17.6%), BW maintenance (-6.0%) and BW gain (7.0%) heifers. Basal plasma LH concentration was not influenced by nutritional treatment and was similar before and after parturition for all groups. However, in response to GnRH, peak plasma LH concentration was greater (P less than .10) for prepartum than for postpartum heifers. Mean LH peak amplitude in prepartum heifers was approximately twofold greater (P less than .10) in the BW loss and maintenance groups compared with the BW gain group. Prepartum LH release was related inversely (r = -.64) to change in heifer BW and increased (P less than .01) as BW loss increased during gestation. Objectivisation of the physiological course of puerpery and control of involution processes of uterus represent topical questions of an increased frequency of lambing. Our study was directed at the determination of the weight, length of body and horns of uterus as well as of the weight, length, width and height of ovaries, following the administration of carbetocin (Depotocin inj. Spofa), in the puerperal period of Slovak Merino sheep. The study was aimed at obtaining objective biometric data on uterus and ovaries and recording the influence of carbetocin on the involution of uterus and ovaries of sheep. Thirty ewes, divided into two groups, were included in the experiment. The animals of the experimental group (n = 15) were subjected to treatment with carbetocin (Depotocin inj. Spofa) at the amount 2 x 0.07 mg. The first dose was applied intramuscularly (i. m.) on the 2nd day after parturition and the second dose was injected subcutaneously (s.c.) on the 4th day after lambing. The second group (n = 15) served as a control. Three ewes (n = 3) were bled on each of the following days: 1, 7, 17, 25 and 34 after parturition. After bleeding the sexual organs were removed by excision and their weight and the length of the body and horns of uterus and of ovaries was determined. The number and size of follicles were determined on the surface of ovaries. The weight of uterus body showed a declining tendency in sheep of the control group with a statistically significant decrease from the 17th till the 34th day after parturition (P less than 0.01). Its weight varied from 56.0-14.6 to 5.2-0.3 g in the above-mentioned time interval. The experimental group showed a statistically significant decrease in weight as early as on the 7th day after lambing compared to the 1st day after parturition, down to the values 185.66-38.91 g (P less than 0.05). The decrease in the weight of uterus body recorded on day 17 (106.56-41.08 g) was less pronounced than in the control group. The final average weight of the body of uterus on the 34th day after parturition was 41.36-8.52 g, that means higher than in the control group (5.2-0.3 g). The weight of gravid horns in the experimental group declined significantly as early as on the 7th day after parturition to the average weight of 60.89-9.45 g and reached 9.86-3.49 g on the 34th day.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

The aim of this study was to test whether low dose oxytocin i.v. injection once a day to mares diagnosed as being ready for birth by mammary secretion calcium strip test measurements could be used as a reliable method to induce parturition and/or predict the mare would not foal during the following night if parturition did not occur within 2 h of treatment. Fifty-one near-term Haflinger mares were used and a single injection of 2.5 iu oxytocin was given between 1700 and 1900 h, including 10 mares used as controls which were administered a placebo. Administration of oxytocin resulted in the delivery of a normal foal within 120 min in 95% of mares. Twenty-four out of 38 (63%) treated animals foaled in response to the first oxytocin injection, 9 out of 38 (24%) in response to the second injection and 3 out of 38 (8%) in response to the third treatment. Two out of 38 (5%) treated mares foaled during the night irrespective of treatment whereas 7 out of 10 (70%) control mares foaled during the night. It was concluded that the major advantage of injecting a daily low dose of oxytocin appears to be that such a low dose induces delivery only in mares carrying a mature fetus and which are ready to foal.  相似文献

Nest building is an important part of maternal behavior in domestic pigs. The aim of the study was to assess the effect of nesting material sawdust vs. straw on sow behavior 24 h before and after birth of the first piglet (BFP) and piglet production. Sows, housed in farrowing crates, were randomly divided into 2 treatments: sawdust (n = 12) and straw (n = 13). Sawdust and straw were provided during the pre- and parturient period; after parturition, straw was given to both experimental groups. The prepartum nesting period (the time interval between the first and last nest-building records, including all other activity and resting before BFP), the nesting records (number of nesting records), nesting duration (duration of all nesting records), the start and termination of nesting, and the frequency of prepartum postural changes were collected 24 h before BFP. After BFP, number of nesting records and time to first sucking of the litter were collected. Frequency of postural changes and duration of udder access were collected 24 h after BFP during 3 time periods (during parturition, from the end of parturition to 12 h after BFP, and 12 to 24 h after BFP) and the frequency of nursing during 2 time periods (from the end of parturition to 12 h after BFP, and 12 to 24 h after BFP). Piglet BW gain and mortality were estimated 24 h after BFP. Data were analyzed using PROC MIXED and the probability of the piglet mortality using PROC GENMOD in SAS. Nesting material did not affect (P > 0.10) most of sow prepartum nesting behavior and had no effect (P > 0.10) on the prepartum frequency of postural changes. Sows from the sawdust treatment had a longer nesting period (P < 0.05), and nest building tended to start sooner (P < 0.10) than in the straw treatment. Nesting material had only a small effect on later maternal behavior. Sows from the straw treatment tended to have more nesting records after BFP (P < 0.10). The frequency of postural changes was affected by the interaction (P < 0.01) between treatments and time period: sow from the straw treatment had more postural changes during parturition compared with other time periods and sawdust treatment. No effect (P > 0.10) of the nesting material on piglet BW gain and mortality was found. The results suggest that sawdust compared with straw as nesting material provided to sows before and through parturition does not negatively affect maternal behavior during the 24 h before and after parturition or piglet production. Therefore, sawdust can be recommended as a suitable nesting material for farrowing sows when straw is not available.  相似文献

The aims of the present study were to further examine the efficacy of different doses and routes of R-cloprostenol administration on the parturition response in sows. Fifty crossbred multiparous sows (Landrace x Yorkshire) with an average parity number of 4.7 +/- 2.4 were allocated to induce farrowing by one of the following treatments: Group I (control, n = 10) injection with normal saline 2 ml administered intramuscularly (i.m.); Group II (n = 10) injection with 75 microg of R-cloprostenol administered i.m. (at 7 AM); Group III (n = 10) injection with 75 microg of R-cloprostenol (at 7 AM) together with 10 IU of oxytocin (24 h after injection of R-cloprostenol) administered i.m.; Group IV (n = 10) injection with 37.5 microg of R-cloprostenol (at 7 AM) administered into perivulva region; Group V (n = 10) injection with 37.5 microg of R-cloprostenol (at 7 AM) administered into perivulva region together with 10 IU of oxytocin (24 h after injection of R-cloprostenol) administered i.m. The following parameters: pre-farrowing maternal behaviour, restless behaviour, R-cloprostenol or oxytocin injection to farrowing interval, expulsion intervals, duration of farrowing, total number of piglets born, litter birthweight, umbilical cord morphology and the degree of meconium staining were record. There were no significant differences among groups for the pre-farrowing maternal behaviours. In all the sows, the restless behaviour was not observed. There were no significant effect of oxytocin administration (10 IU, i.m.) on the percentage of umbilical cord morphology and the degree of meconium staining in different groups. There were no significant effect of route and dose of administration on the number of total piglet born, piglet born alive, stillbirth, mummy and litter birthweight. No significant effects of the different groups were found on the R-cloprostenol and oxytocin injection to farrowing interval, expulsion interval and farrowing duration. In conclusion, the present results demonstrated that a half dose (37.5 microg) of R-cloprostenol administered into the perivulva region was effective for inducing farrowing as the full recommended dose (75 microg) administered into the neck region (i.m.) and with no restless behaviour.  相似文献

Various parenteral treatment forms of oxytocin, as often used under praxis circumstances, may act differently on contractility of the uterus during the first days of the puerperium. Various patterns of such induced uterotonic responses may lead to alterations in the emptying characteristics of the uterine lumen, thus influencing, as a late consequence, the process of involution. Therefore, this study was designed to test whether two different parenteral administration forms of oxytocin induce changes in peripheral plasma concentrations of 15-ketodihydro-prostaglandin F(2α) (PGF(2α) metabolite) in early post-partum cows. Between 13 and 15 h after uncomplicated calving, healthy dairy cows without retained foetal membranes were treated with 50 IU oxytocin, either intramuscularly (OT-IM group; n = 15) or intravenously (OT-IV group; n = 16). Saline solution was administered intramuscularly as controls (CON group; n = 15). Jugular blood samples were taken at 10-min intervals from 1 h before to 2 h after treatment. Plasma PGF(2α) metabolite levels were measured by radioimmunoassay. No significant differences in peripheral plasma PGF(2α) metabolite concentrations occurred in the OT-IM and CON groups, but mean values significantly increased in the OT-IV group, peaking at 20 min after treatment and reaching pre-treatment baseline values again at 120 min. Although the source of prostaglandins was not investigated in this study, our results suggest that exogenous oxytocin may enhance secretion of prostaglandins by the uterus during the first day after normal calving. These prostaglandins might contribute, by an endocrine or paracrine route, to the stimulation of myometrial contractility when exogenous oxytocin is given during this early post-partum stage.  相似文献

The effect of pre-treatment with a selective platelet-activating factor (PAF) antagonist, WEB 2086, on the actions of low-dose endotoxin was evaluated in ponies prepared with gastrointestinal strain gauges. Endotoxin (0.1 microgram/kg i.v.) produced a marked reduction in gastric contraction amplitude and rate, and an increased frequency and reduced duration of jejunal phase III activity fronts (AFs). WEB 2086 (6.6 mg/kg) administered i.v. 10 min before the endotoxin, produced significant antagonism (P less than 0.001) of the effect of endotoxin on gastric contraction amplitude and rate. The combination of WEB 2086 and endotoxin produced gastric contractions of significantly (P less than 0.01) higher frequency than in the control studies. WEB 2086 also reduced endotoxin-induced abnormal phase III AFs in the jejunum and increases in heart rate and packed cell volume. These results provide evidence that endogenous PAF plays a role in mediating the acute effects of endotoxin on equine gastrointestinal motility.  相似文献

The influence of exposure to exogenous estradiol on the interval from parturition to first ovulation, luteinizing hormone (LH) secretion and luteal function was examined in cows. Cows were assigned at parturition to one of three treatments. Cows received either a 3.0 (1-E; n = 30) or .75 cm (1/4-E; n = 28) implant containing 17 beta-estradiol or served as untreated control animals (C; n = 33). Implants were administered within 2 days following parturition and removed on day 40 postpartum (day 0 = day of parturition). Single blood samples were collected twice weekly and analyzed for progesterone to determine length of postpartum anestrus and duration of the initial increase in progesterone. Sequential blood samples were collected on day 35 +/- .1 postpartum (15 min intervals for 18 hrs) from 5 cows in each treatment and analyzed for LH. Concentrations of estradiol were higher (P less than .01) in the 1-E (5.3 +/- .24) than in C (3.9 +/- .23) or 1/4 E (3.9 +/- .25) cows on day 35 postpartum. The interval from parturition to the first estrous cycle of normal duration was similar for cows in the C and 1-E treatment (53 +/- 2.4 and 56 +/- 2.4 days, respectively). Cows in the 1/4-E treatment had a longer (P less than .05) interval (68 +/- 2.5 days). Secretion of LH was similar among treatments on day 35 postpartum. The first normal luteal phase after parturition was preceded by a transient rise in progesterone in 81, 64 and 85% of the cows in the C, 1-E and 1/4-E treatments, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

On day 64 after artificial insemination, a six-year-old primiparous briard bitch whelped three live pups between 05.00 and 08.00. It was presented at 11.00 on the same day with failure to complete parturition. On ultrasound examination, a normal live fetus was observed and the bitch was treated with oxytocin three times during the day (1.0, 2.0 and 2.0 iu intramuscularly), with no effect. The following day, a higher dose of oxytocin (5.0 iu) was administered intramuscularly at 11.00, after a uterine ultrasound examination confirmed viability of the fetus. At 18.00 of the same day, the bitch whelped the fourth normal live pup, 37 hours after initiation of parturition and 34 hours after expulsion of the last fetus. Effectiveness of oxytocin and normal versus prolonged parturition due to uterine inertia are discussed.  相似文献

Although parturition commonly is induced in swine with prostaglandin F2 alpha or one of its analogs, variation in response to the treatment is large. The purpose of this investigation was to more precisely terminate parturition. Two experiments were conducted. Sows all received prostaglandin analog followed, 20 h later, by different doses of the beta-adrenergic blocking agent carazolol and(or) oxytocin. Treatment with 3 mg of carazolol alone caused parturition to commence within 2.6 +/- .23 (SEM) h after injection. Parturition was complete in 2.0 +/- .13 h and proceeded smoothly without the necessity of manual assistance. Adding 2.5 I.U. of oxytocin to the 3 mg of carazolol speeded the onset of labor (1.7 +/- .17 h). However, in some cases parturition was interrupted and the time passing between the birth of the first and the last piglet was prolonged to 2.5 +/- .28 h. We conclude that by applying carazolol 20 h after a prostaglandin analog, parturition in swine may be terminated very effectively.  相似文献

Changes in numbers of ovarian follicles and coincident secretion of pituitary gonadotropins were characterized in suckled, anovulatory beef cows injected iv with 500 ng of luteinizing hormone-releasing hormone (LHRH) every 2 h for 48 or 96 h, starting 21.4 +/- .4 d after parturition. Two hours after the last injection, all cows were ovariectomized. Compared with saline-injected controls, LHRH had no effect on baseline or overall concentrations of luteinizing hormone (LH) in serum (P greater than .10), but increased (P less than .05) frequency and decreased (P less than .05) amplitude of LH pulses. Luteinizing hormone-releasing hormone increased (P less than .05) baseline concentration of follicle stimulating hormone (FSH) in serum and frequency of FSH pulses, but decreased (P less than .05) pulse amplitude. Overall concentrations of FSH increased 20% (P less than .10). Exogenous LHRH did not affect diameter of the two largest follicles or numbers of follicles 1.0 to 3.9 mm, 4.0 to 7.9 mm or greater than or equal to 8.0 mm in diameter. These data suggest that increasing the frequency of episodic LH and FSH pulses in postpartum cattle by intermittent administration of LHRH did not increase mean circulating levels of LH, or alter size and numbers of ovarian follicles within the 96-h period of injections. Thus, induction of ovulation in anovulatory cows treated with low-dose injections of LHRH cannot be explained on the basis of an increase in mean concentrations of LH or numbers of antral follicles within 96 h after initiation of injections.  相似文献

We evaluated effects of three concentrations of phenylephrine, ergonovine, oxytocin and norepinephrine (myometrial stimulants) on viability of spermatozoa when they were included in a seminal extender. Using a split ejaculate technique, ejaculates from each of 10 bulls were extended in egg-yolk citrate to a final concentration of 35 x 10(6) sperm/ml, including 20 mg/ml, 2 mg/ml and .2 mg/ml, of phenylephrine or ergonovine, 20 IU/ml, 2 IU/ml and .2 IU/ml oxytocin or 200 micrograms/ml, 20 micrograms/ml and 2 micrograms/ml norepinephrine prior to freezing. Extended semen without a myometrial stimulant served as the control. Percentage of intact acrosomes was determined prior to freezing for all treatments. Motility and percentage of intact acrosomes were determined immediately after thawing (0 h) and again after 4 h incubation at 37 degrees C. Percentage of intact acrosomes was reduced (P less than .05) prior to freezing by phenylephrine (20 mg/ml) and ergonovine (20 mg/ml) (phenylephrine = 56%; ergonovine = 63%; control = 74%). The same doses of phenylephrine and ergonovine reduced (P less than .05) post-thaw motility and percentage of intact acrosomes at both 0 and 4 h compared with controls. Sperm exposed even to the intermediate concentration (2 mg/ml) of ergonovine had lower (P less than .05) motility 4 h post-thaw. No other compound or concentration of compound or concentration of compound affected percentage of intact acrosome or motility.
