This study aimed at isolating and genotyping Toxoplasma gondii from serologically positive free-range chickens from Argentina, and to evaluate the use of sentinel animals during a short time period of exposure to determine environmental contamination with T. gondii oocysts. Two groups of chickens on six farms were compared in this study: (i) young, 2-3 month-old broiler-type chickens reared as sentinel animals on the farms and (ii) adult chickens reared on the same farms for more than one year. Seroconversion rates of 7.0% or 5.7% were observed in sentinel broiler chickens reared for a period of 74 days (January-April 2010) or 88 days (August-November 2010) respectively, as shown by a T. gondii specific immunofluorescent antibody test. Fifty-three percent (17 of 32) of adult chickens were positive and showed higher titres than sentinel animals. Isolation of T. gondii from tissues (brain and heart) of serologically positive chickens was achieved from six of seven free-range adult birds with IFAT titres of 200 and higher. The isolated parasites were analysed by multi-locus polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The isolated T. gondii showed three different genotypes: two genotypes consisted in atypical allele combinations, and the remaining genotype had exclusively clonal type II alleles. All isolates obtained at a single farm, corresponded to the same genotype. The T. gondii genotypes observed are identical to those described in cats, dogs, chickens and capybaras elsewhere in South America. Two isolates, which showed different allele combinations in PCR-RFLP, were characterized in a mouse virulence assay. While one isolate showed a low virulence a second isolate was of intermediate virulence to mice.  相似文献

The prevalence of Toxoplasma gondii in free-ranging chickens is a good indicator of the prevalence of T. gondii oocysts in the soil because chickens feed from the ground. The prevalence of T. gondii in 85 free-range chickens (Gallus domesticus) from Chile was determined. Antibodies to T. gondii were assayed by the modified agglutination test (MAT), and found in 47 of 85 (55.3.9%) chickens with titers of 1:5 in six, 1:10 in four, 1:20 in four 1: 40 in three, 1: 80 in nine, 1: 160 in four 1:320 in nine, and 1: 640 or higher in eight. Hearts and brains of 47 chickens with titers of 1:5 or higher were pooled for each chicken and bioassayed in mice. Tissues from 16 seronegative (MAT<1:5) chickens were pooled and fed to one T. gondii-free cat. Feces of the cat were examined for oocysts but none was found based on bioassay of fecal floats in mice. Hearts and brains from seven seronegative (<1:5) were pooled and bioassayed in mice; T. gondii was not isolated. T. gondii was isolated by bioassay in mice from 22 chickens with MAT titers of 1:20 or higher. Genotyping of these 22 isolates using polymorphisms at the loci SAG1, SAG2, SAG3, BTUB and GRA6 revealed three genotypes. Seventeen isolates had type II alleles and four isolates had type III alleles at all loci. One isolate contained the combination of type I and III alleles. This is the first report of genetic characterization of T. gondii isolates from Chile, South America.  相似文献

The aim of the present study was to analyze the effect of range enrichment (trees or tall grass stand) on the performance and herbage intake of free-range chicken. This trial was conducted in 2 different seasons (winter and summer) comparing a standard free-range system with 2 kinds of range enrichment: a stand of sorghum (a tall grass) and olive trees. Two hundred fifty male naked neck chickens were used in each season and system; performance, behavior, forage intake, and body lesions were recorded. Productive performance was not affected by range enrichment. However, in the presence of trees or tall grass stand, no predation losses were experienced; whereas, in the standard paddock, cases of predation by raptors or crows were registered, mainly during the first period of rearing. Consequently, mortality rate was significantly lower in the enriched environment. Control chickens stayed indoors more rather than forage in the pasture, whereas, in enriched environments, birds spent more time outdoors and widely exploited the available pasture. Forage intake was significantly influenced by rearing system and season; chickens reared under olive trees had higher herbage ingestion, exploring the available area up to almost 50 m from the hut. With the olive trees, chickens also showed the lowest frequency of foot and breast damage.  相似文献

Toxoplasma gondii is widely distributed in humans and other animals including domestic poultry throughout the world, but little is known of the prevalence of T. gondii in chickens and ducks in People's Republic of China. In the present study, antibodies to T. gondii were investigated in 349 domestic ducks (Anas spp.), 361 free-range, and 244 caged chickens (Gallus domesticus) raised in commercial flocks in Southern China's Guangdong Province using the modified agglutination test (MAT). Antibodies to T. gondii (MAT titer of 1:5 or higher) were found in 56 (16%) of 349 ducks, 41 (11.4%) of 361 free-range, and 10 (4.1%) of 244 caged chickens. The results indicate soil contamination due to T. gondii oocysts because free-range chickens feed from the ground, and suggest that the meat from the domestic poultry may be an important source for human infection by T. gondii in People's Republic of China.  相似文献

The prevalence of Toxoplasma gondii in free-ranging chickens is a good indicator of the prevalence of T. gondii oocysts in the soil because chickens feed from the ground. The prevalence of T. gondii in 77 free-range chickens (Gallus domesticus) from Colombia, South America was determined. Antibodies to T. gondii were assayed by the modified agglutination test (MAT), and found in 32 (44.4%) of 72 chickens with titers of 1:5 in 4, 1:10 in 3, 1:20 in 1, 1:40 in 1, 1:80 in 8, 1:160 in 8, 1:320 in 3, and 1:640 or higher in 4. Hearts and brains of 31 seropositive chickens were pooled and bioassayed in mice. Tissues from 32 (16+16) seronegative chickens were pooled and fed to two, T. gondii-free cats, and tissues from nine chickens without matching sera were fed to one T. gondii-free cat. Feces of cats were examined for oocysts. T. gondii oocysts were excreted by a cat that was fed tissues of 16 seronegative chickens. T. gondii was isolated by bioassay in mice from 23 chickens with MAT titers of 1:20 or higher. All infected mice from 16 of the 23 isolates died of toxoplasmosis. Overall, 82 (81.1%) of 101 mice that became infected after inoculation with chicken tissues died of toxoplasmosis. Genotyping of these 24 isolates using polymorphisms at the SAG2 locus indicated that seven T. gondii isolates were Type I, 17 were Type III, and none was Type II. Phenotypically, T. gondii isolates from chickens from Colombia were similar to isolates from Brazil but different from the isolates from North America; most isolates from chickens from Brazil and Colombia were lethal for mice whereas isolates from North America did not kill inoculated mice. Genetically, none of the T. gondii isolates from Colombia and Brazil was SAG2 Type II, whereas most isolates from chickens from North America were Type II. This is the first report of genetic characterization of T. gondii isolates from Colombia, South America.  相似文献

The prevalence of Toxoplasma gondii in free-ranging chickens is a good indicator of the prevalence of T. gondii oocysts in the soil because chickens feed from the ground. The prevalence of T. gondii in 98 free-range chickens (Gallus domesticus) from Nicragua was determined. Antibodies to T. gondii were assayed by the modified agglutination test (MAT), and found in 84 (85.7%) of 98 chickens with titers of 1:5 in 10, 1:10 in eight, 1:20 in seven, 1:40 in nine, 1:80 in 11, 1:160 in one, 1:200 in 27, 1:400 in six, 1:800 four, and 1:3200 in one bird. Hearts and brains of 32 chickens with titers of 1:10 or less were pooled and fed to three T. gondii-free cats. Hearts and brains of 66 chickens with titers of 1:20 or higher were bioassayed in mice. Feces of cats were examined for oocysts. The cat fed tissues from eight chickens with titers of 1:10 shed T. gondii oocysts. The two cats fed tissues of 24 chickens with titers of 1:5 or less did not shed oocysts. T. gondii was isolated by bioassay in mice from 47 chickens with MAT titers of 1:20 or higher. All infected mice from six isolates died of toxoplasmosis. Overall, 41 of 170 (24.1%) mice that became infected after inoculation with chicken tissues died of toxoplasmosis. Genotyping of these 48 isolates (47 from mice and 1 from pooled tissues) using polymorphisms at the loci SAG1, SAG2, SAG3, BTUB and GRA6 revealed eight genotypes. Six isolates had Type I alleles, three isolate had Type II alleles and six isolates had Type III alleles at all loci. Four isolates had mixed infections. Two isolates have a unique allele at SAG1 locus and combination of I and III alleles at other loci. The rest 27 isolates contained the combination of Type I and III alleles and were divided into four genotypes. More than one genotypes were often isolated in chickens from the same household, indicating multiple genotypes were circulating in the same environment. This may explain the high frequency of mixed infections observed. High rate of mixed infection in intermediate hosts such as chickens may facilitate genetic exchange between different parasite lineages in definitive feline hosts. This is the first report of genetic characterization of T. gondii isolates from Nicragua, Central America.  相似文献

The prevalence of Toxoplasma gondii in free-ranging chickens is a good indicator of the prevalence of T. gondii oocysts in the soil because chickens feed from the ground. The prevalence of T. gondii in free-range chickens (Gallus domesticus) from 11 Bio-farms in Austria was determined. Antibodies to T. gondii assayed by the modified agglutination test (MAT) were found in 302 of 830 (36.3%) chickens with titers of 1:10 in 50, 1:20 in 69, 1:40 in 53, 1:80 in 40, 1:160 or higher in 90. Hearts of 218 chickens with MAT titers of 10 or higher were bioassayed individually in mice. Tissues from 1183 chickens were pooled and fed to 15, T. gondii-free cats. Feces of the cats were examined for oocysts; 11 cats shed T. gondii oocysts. T. gondii was isolated from 56 chickens by bioassay in mice. Thus, there were 67 isolates of T. gondii from these chickens. Genotyping of these 67 isolates using the SAG2 locus indicated that all 33 were Type II. Phenotypically and genetically these isolates were different from T. gondii isolates from Brazil. None of the isolates was virulent for mice. This is the first report of isolation of T. gondii from chickens from Austria.  相似文献

We conducted a cross-sectional study to determine the prevalence and species of gastro-intestinal helminths and haemoparasites in 100 chickens kept under extensive management systems in Ghana, West Africa. All the examined chickens (100%) were infected with gastro-intestinal helminths; a total of 18 species were detected. The species and their prevalences were: Acuaria hamulosa (25%), Allodapa suctoria (20%), Ascaridia galli (24%), Capillaria spp. (60%), Choanotaenia infundibulum (13%), Gongylonema ingluvicola (62%), Heterakis gallinarum (31%), H. isolonche (16%), Hymenolepis spp. (66%), Raillietina cesticillus (12%), R. echinobothrida (81%), R. tetragona (59%), Strongyloides avium (2%), Subulura strongylina (10%), Tetrameres fissispina (58%), Trichostronygylus tenuis (2%), and finally one unidentified acanthocephalan (1%) and one unidentified trematode (1%). Thirty-five per cent of the chickens were infected with the haemoparasites Aegyptinella pullorum and Plasmodium juxtanucleare (prevalences 9% and 27%, respectively). Association between chicken sex and prevalences was not significant. An over-dispersed distribution was seen for most of the helminth species.  相似文献

Background Since 2005, H5N1 avian influenza (AI) has spread from South-East Asia to over 60 different countries, resulting in the direct death or slaughter of over 250,000,000 poultry. Migratory waterfowl have been implicated in this spread and in Australia there have been numerous isolations of low-pathogenicity AI virus from wild waterfowl and shorebirds. The Department of Human Services, Victoria maintains 10 sentinel free-range chicken flocks in the Riverland at locations that are populated by large numbers of waterfowl known to carry a range of strains of AI. Objective This study analysed historical samples collected in 1991–94 and 2003–06 from the library of serum samples for antibodies against AI to assess the potential for transfer of AI virus from wild waterfowl to free-range poultry. Results Of the 2000 serum samples analysed, 17 were positive for antibodies against AI and 87 were suspect, with a clustering of positive and suspect results in the years 1994, 2003 and 2004. There was also a clustering of positive samples at the site of the Barmah flock. Nine sequential sets of sera from individual chickens with at least one positive result were identified. Analysis of these sequential sets showed that infection was acquired on site but that the antibody response to AI infection was short-lived and was no longer detectable at 8 weeks after the positive finding. Conclusion The surveillance of sentinel chickens is a potential avenue for monitoring the circulation of AI viruses and could provide an early warning system for the commercial poultry industries.  相似文献

ABSTRACT

1. The aim of this study was to determine the nutritional status of backyard chickens reared under 4 different agro-climatic conditions (tropical, humid subtropical, cool-temperate and semi-arid) of India during different seasons of the year and to compare their nutritional status with those reared under confined feeding system (CFS). In each season, 15 adult and 15 grower chickens were slaughtered for collecting content of crop and gizzard to assess the nutritional status from different agro-climatic conditions.

2. The dry matter content of crop and gizzard was higher (P < 0.01) in chickens reared under backyard conditions compared to those reared under CFS.

3. Higher (P < 0.01) crop and gizzard contents were recorded during winter compared to the rainy season. The proportion of grains in the crop during summer and winter season was higher compared to rainy season. Further, there were more insects present in the crop during the rainy season and winter compared to summer.

4. The proportion of insects was higher in chickens reared in humid, subtropical and cool temperate regions compared to semi-arid conditions. The consumption of grasses was higher in cool-temperate regions as compared to those reared under other areas.

5. Crude protein (CP), Ca, P and gross energy (GE) content of the crop were higher in chickens reared under CFS compared to those reared in backyard systems and CP in the crop contents was lower in backyard chickens reared in humid subtropical and semi-arid regions compared to other areas. Similarly, lower (P < 0.01) Ca levels were seen in crop and gizzard contents from birds reared in semi-arid Rajasthan, and P and GE were lower in tropical wet and dry Telangana (TWT) compared to the chickens reared in the backyard conditions in other regions.

6. Based on the results, the nutrient intake for birds utilising the scavenging feed resource base (SFRB) in the TWT region was below the requirements of backyard chickens from the other regions selected in the present study. GE and CP were critically deficient in the SFRB of the TWT region. Therefore, evolving location-specific supplementary diets using locally available feed resources to meet nutrient requirement of backyard chickens is required for optimising production potential.  相似文献

The prevalence of T. gondii in 144 free-range chickens (Gallus domesticus) from Costa Rica was determined. Antibodies to T. gondii were assayed by the modified agglutination test (MAT), and found in 60 (40.1%) of 144 chickens with titers of 1:5 in 16, 1:10 in 5, 1:20 in 2, 1:40 in 3, 1:80 in 5, and 1:160 or higher in 29. Tissues of all chickens were bioassayed for T. gondii in mice or cats. Hearts and brains of 52 chickens with titers of 1:5 or higher and 16 chickens with doubtful titers were pooled and bioassayed in mice. Tissues from 76 chickens with MAT titers of 1:10 or less were pooled and fed to three T. gondii-free cats. Fecal floats of cats were bioassayed orally in mice but were negative for T. gondii oocysts. T. gondii was isolated by bioassay in mice from 32 chickens with MAT titers of 1:10 or higher. All infected mice from 4 of the 32 isolates died of toxoplasmosis. Genotyping of these 32 isolates using polymorphisms at the loci SAG1, SAG2, SAG3, BTUB and GRA6 revealed five genotypes. Five isolates had type I alleles and one isolate had type III alleles at all loci. The rest 26 isolates contained the combination of type I and II or I and III alleles and were divided into three genotypes. None was found to have genotype II alleles at all five loci. Neospora caninum and Toxoplasma gondii are biologically and morphologically similar coccidians with canids as definitive hosts for N. caninum and felids for T. gondii. Feral chickens have been used as indicators of soil contamination with T. gondii oocysts because they feed from ground. In the present study we studied seroprevalence of N. caninum in free range chickens from different countries in America as an indicator of soil contamination due to N. caninum oocysts. Antibodies to N. caninum were found in sera of 524 (39.5%) of 1324 chickens using indirect fluorescent antibody test (IFAT, titer 1:25 or higher). Seropositive chickens from different countries were: 18.5% of 97 from Mexico, 7.2% of 97 from USA, 39.5% of 144 from Costa Rica, 71.5% of 102 from Grenada, 44% of 50 from Guatemala, 83.6% of 98 from Nicaragua, 58.1% of 55 from Argentina, 34.3% of 358 from Brazil, 62.3% of 85 from Chile, 11.2% of 62 from Colombia, 38.7% of 80 from Guyana, 18% of 50 from Peru and 21.7% of 46 from Venezuela. The results indicate widespread exposure of chickens to N. caninum.  相似文献

Toxoplasma gondii is widely distributed in humans and other animals including domestic poultry throughout the world, but the data on prevalence of T. gondii in free-ranged (FR) chickens in People's Republic of China (PRC) are limited. In the present study, the seroprevalence of T. gondii infection in FR chickens was investigated in 13 provinces/municipalities of China during the period from January to June 2010. A total of 1173 serum samples were collected and assayed for T. gondii circulating antigens (TCA) and antibodies (TCAb) using enzyme linked immunosorbent assay (ELISA) technique. Out of this number, 199 samples were TCA positive (16.97%), 226 samples were TCAb positive (19.27%), 69 samples were positive for both TCA and TCAb (5.88%), and the total seropositive rate was found in 356 of 1173 (30.36%). The results of the present survey indicated that infection with T. gondii in FR chickens is widely spread in China.  相似文献

1. The effects of rearing chickens using standard, maize-fed, free-range or organic production systems on meat quality and sensory characteristics were evaluated. The standard system used either Ross or Cobb birds with a slaughter age of approximately 40 d, the other systems in the trial used Hubbard birds with considerably older slaughter ages, up to 72 d in the case of organic systems. 2. Paired breast fillets from 120 birds, 30 from each rearing system, were used. The meat quality variables, ultimate pH, colour coordinates L*, a*, b* and water-holding capacity were measured and taste panel assessments were made using 8-point category scales of texture, juiciness, abnormal flavour, flavour liking and overall flavour. 3. There were significant differences between rearing systems, with fillet muscles from birds grown under the standard system having a higher ultimate pH. Differences were also seen in colour with fillets from birds reared under a standard system having a smaller hue angle than those grown using the maize-fed system which had the highest. 4. Fillets from birds reared in the standard system were rated by the taste panel as more tender and juicy. There were no significant differences in chicken flavour. Based on hedonic assessments of flavour liking and overall liking, by a small panel of assessors, meat from birds produced in the standard system was most preferred and that from organic systems the least preferred. Meat from free-range and maize-fed systems was intermediate in preference. This result reveals a trend, but does not infer consumer acceptance.  相似文献

Cryptosporidiosis acutely impacts the digestive and/or respiratory tract of the birds in many species of various orders. More importantly, it is also well known as a significant zoonotic disease, which can lead to diarrhea in humans and livestock. Regarding increasing demand for free-range products and increasing the number of free-range poultry farms, the present paper evaluated histopathological and molecular detection of Cryptosporidium baileyi and Cryptosporidium parvum in free-range and commercial broiler chickens in the north part of Iran. For this purpose, 100 fecal and tissue samples of the chickens in Guilan province were collected. After microscopic examination using Ziehl-Neelsen staining, molecular analyses of the fecal samples were processed by Nested-PCR targeting the 18S rRNA gene followed by sequencing of the amplicons and phylogenetic analyses. Eventually, the tissue samples were studied for histological lesions. Findings demonstrated the presence of Cryptosporidium baileyi and Cryptosporidium parvum in 6 % and 2 % of fecal samples, respectively. This is the first identification of C.parvum in avian hosts in Iran, and for the first time, C.baileyi and C.parvum are shown in native free-range chickens in Iran. All of the PCR positive birds with clinical symptoms showed gross lesions of respiratory infections. There was no significant difference between infection rate in free-range and commercial broiler chickens; however, the infection rate was significantly higher in chickens <25 days old. To conclude, we present here a notable Cryptosporidium infection rate in the free-range chicks in Iran, which notify the role of this host as a reservoir and should be more noted due to the economic and zoonotic importance.  相似文献

Thirteen adult indigenous chickens from Oodi, Kgatleng district, Botswana, were examined for helminth parasites. Two species of nematodes, Ascaridia galli and Heterakis gallinarum, and species of the cestode genus Raillietina, were recovered. A. galli and H. gallinarum were the most commonly seen parasites. Little is known of the prevalence of Toxoplasma gondii in commercially raised chickens. In the present study, the prevalence of T. gondii in 96 free-range chickens (Gallus domesticus) from a commercial farm in Israel was assessed. Blood, heart, and brain from each chicken were examined for T. gondii infection. Antibodies to T. gondii, assayed with the modified agglutination test (MAT > or = 1:5), were found in 45 of the 96 chickens. Hearts and brains of seropositive (MAT > or = 1:5) chickens were bioassayed in mice. Additionally, hearts and brains of 51 seronegative (MAT < 1:5) chickens were bioassayed in two T. gondii-free cats. T. gondii was isolated from 19 of the 45 (42.2%) seropositive chickens by bioassay in mice. Both the cats fed tissues pooled from seronegative chickens shed T. gondii oocysts. Tachyzoites and tissue cysts of all 21 isolates of T. gondii from chickens were avirulent for mice. Seventeen of the 19 isolates genotyped were found to be type II, and 2 were type III. Understanding of the sources of infection on such farms could be the key to the development of better prevention strategies.  相似文献

A prospective study of risk factors associated with outbreaks of Newcastle disease (ND) in indigenous free-range chickens was carried out in three agro-ecological zones in eastern Uganda. Sixty households keeping chickens were randomly selected and studied from March 2004 to February 2005, covering rainy and dry seasons. Data on ND outbreaks, risk factors and flock dynamics were collected using an interviewer-administered questionnaire, while ND outbreaks were confirmed by haemagglutination inhibition test. Multivariate survival analyses were performed to identify the risk factors for Newcastle disease outbreaks. Although the survival time against ND was longer (>100 days) in the dry compared to the rainy season, incidence in the two seasons was not significantly different (p > 0.05). None of the factors investigated was significantly associated with ND. However, purchasing of restocking chickens from the market and neighbourhood (hazard ratio [HR] = 1.79; 95% confidence interval [CI] 0.89–3.60), the presence of migratory wild birds (HR = 1.70; 95% CI 0.65–4.48) and being in agro-ecological zone 1 (HR = 1.48; 95% CI 0.66–3.36) showed a positive but non-significant association with risk for ND. To understand the roles of other domestic poultry species in the epidemiology of ND in rural free-range chickens, virus isolation and molecular characterization of isolates in addition to more prospective cohort studies with a larger sample size and similarly long period of follow-up are needed.  相似文献
