羊毛是由蛋白质组成的天然纤维,被广泛应用于纺织等行业,然而随着经济的发展,人们对羊毛品质的要求越来越高,传统的研究方法已满足不了人们对羊毛蛋白研究的需要。蛋白质组学作为后基因组时代的一门新兴学科,可大规模、高通量、系统化地从整体水平研究羊毛蛋白质的组成、功能及其蛋白之间的相互作用,在羊毛研究领域已得到广泛应用。文章从羊毛蛋白质组学的研究方法,已知的羊毛蛋白质以及蛋白组学在羊毛蛋白质研究中的应用进展进行了综述,以期为蛋白质组学在羊毛中的应用提供参考。 相似文献

乳蛋白是生命最初阶段中最为重要的营养功能成分,并且种类多样、含量丰富.通过一系列的遗传改变、丰富的蛋白质翻译后修饰,使乳蛋白的构成变得非常复杂、数量异常庞大.采用蛋白质组学方法对乳蛋白进行系统、整体地研究,可以认识乳蛋白质在构成以及调控一些生命活动中的规律.这样能够非常全面、深入地表明乳蛋白质的表达水平.本文阐述了蛋白质组学中的主要技术,包括双向电泳技术、高效液相色谱技术、同位素标记相对和绝对定量(isobaric tags for relative and absolute quantitation,iTRAQ)技术、质谱技术以及生物信息学技术.分别讨论了蛋白质组学技术在人乳、牛乳中的应用,并对人乳与牛乳的差异蛋白质组学进行讨论.利用蛋白质组学技术分析人乳与牛乳蛋白,能够更有目的地加强对人乳与牛乳的认识,旨在为婴幼儿食品以及乳制品提供理论依据. 相似文献

蛋白质组学以蛋白质组为研究对象,从整体上对生命载体进行研究,已成为后基因组时代的研究热点。目前,蛋白质组学技术主要包括双向凝胶电泳、生物质谱及生物信息学。双向凝胶电泳根据蛋白质的等电点和分子质量分离蛋白质,而质谱技术已成为鉴定蛋白质的极为灵敏而迅速的工具。由此得到的肽质量图谱结合准确全面的数据库技术,就使得新的蛋白质或多肽得以鉴定。近年来,蛋白质组研究技术已应用到多种生命科学领域中,在兽医学研究领域中尤其是兽药开发方面也将会有很广阔的应用前景。 相似文献

Proteomics is a discipline which study proteome using the technology of protein isolation,identification and quantitative.Proteomics technology also has greatly developed,and proteomics technologies based on mass spectrometry have been widely used in the related field.In this review,we mainly introduced the advantages and disadvantages of some proteomic technologies based on mass spectrometry on the studies of protein-protein interactions,differential proteomics and quantitative proteomics,reviewed the advance of hair follicles and focused on the proteomic technologies application in wool-sheep and cashmere-goat.Finally,some prospects of proteomics technologies application in the hair follicles study of wool-sheep and cashmere-goat were presented. 相似文献

Proteomics experiments have the ability to simultaneously identify and quantify thousands of proteins in one experiment. The use of this technology in veterinary/animal science is still in its infancy, yet it holds significant promise as a method for advancing veterinary/animal science research. Examples of current experimental designs and capabilities of proteomic technology and basic principles of mass spectrometry are discussed. Proteomics holds significant promise as a method for advancing animal science research. The use of this technology in animal science is still in its infancy. The ability of proteomics to simultaneously identify and quantify potentially thousands of proteins is unparalleled. In this review, we will discuss basic principles of doing a proteomic experiment. In addition, challenges and limitations of proteomics will be considered, stressing those that are unique to animal sciences.
