Summary Optimum storage conditions to identify resistance to silver scurf among commercially grown potato cultivars were determined in a series of experiments. Inoculation of field-grown tubers with a conidial suspension ofH. solani (concentration 104 conidia per ml of water) and incubation at 15°C with 95% RH for 1 month followed by 2 months at 85% RH produced the greatest differences in disease severity between potato cultivars. Results with glasshouse-grown tubers were comparable with those from field-grown tubers, indicating that the test could be used to identify resistance in wild species of potato and to understand the inheritance of this resistance.  相似文献

Two shade house and six field experiments were undertaken to evaluate fungicides and timing of application for the control of pink rot of potatoes caused byPhytophthora erythroseptica. Pink rot developed in up to 60% of tubers grown in artificially inoculated soil and 21% of the tubers in naturally infected soil. Ridomil (metalaxyl) and Ridomil Gold (mefenoxan) granules applied to the soil at planting completely or nearly completely controlled pink rot throughout the season in both pot and field experiments. Foliar applications of Ridomil Gold MZ (mefenoxan plus mancozeb) applied when tubers were around 20 mm diameter and another one or two sprays 12 to 28 days later also provided more than 80% control of pink rot in most experiments. Foliar applications of Shirlan (fluazanim) or Acrobat MZ (dimethomorph plus mancozeb) showed some activity against pink rot, but were significantly less effective than Ridomil Gold MZ. Foli-R-Fos 200 (phosphonic acid) or Dithane M45 (mancozeb) applied to the canopy showed little activity against pink rot in most experiments. Yield increases of between 6 and 17 tons ha?1 resulted from the application of either Ridomil granules at planting or two to three Ridomil Gold MZ foliar sprays around “tuber set.”  相似文献

Treating seed potatoes with contact fungicides prevents infection of Phytophthora infestans from tuber to tuber during handling but does not prevent field infections in the young plant. Dimethomorph (DMM) 9% + mancozeb 60% (Acrobat MZ, BASF) can cover both aspects due to the contact and systemic action of mancozeb and DMM, respectively. Experiments were performed under greenhouse and field conditions (three seasons and two locations) with cvs Kennebec, Shepody, Spunta, Russet Burbank, and Ranger Russet. Immediately after cutting, seed pieces were treated with Acrobat MZ at doses equivalent to 2, 3, and 4 kg ha−1. At any dose, Acrobat MZ protected healthy seed tubers against P. infestans infections. Acrobat MZ (4 kg ha−1) also protected the foliage of all cultivars against artificial inoculation of P. infestans, assessed by the detached-leaf method. These effects were observed up to 30 days after emergence in greenhouse experiments and up to 28 days after crop emergence under field conditions in two growing seasons. In cv Spunta, significant foliage protection was observed up to 35 days after field emergence whereas in seed tubers of cv Kennebec planted in sandy soils, significant foliage protection persisted longer. Additional evidence for acropetal translocation of Acrobat MZ in field crops was also found. With this strategy, there is no need to apply contact or systemic fungicides early in the season, and spraying costs and environmental risks are also reduced.  相似文献

Summary Formulae which determined tuber surface area from tuber dimensions and weight were tested by using samples from three potato cultivars. Surviving populations of a labelled isolate ofErwinia carotovora ssp.atroseptica following planting, and populations of a fluorescent pseudomonad following dusting with a bacterial powder formulation were used to demonstrate the application of these formulae to the calculation of bacterial density on the tuber surface. Results obtained by washing whole tubers using the formulae to calculate the surface area were shown to be comparable in sensitivity with those obtained by dilution-plating of periderm discs of 1cm2.  相似文献

Summary TwoPythium spp. were isolated from diseased tubers exhibiting rots at harvest, in summer stores and also in refrigerated storage. Symptoms shared features described for both pink rot caused byPhytophthora erythroseptica and leak caused byPythium spp. The causal agents were identified asPythium aphanidermatum andP. ultimum by morphological and physiological observations and by fingerprinting using oomycete specific primers to amplify the internal transcribed spacer (ITS1) within ribosomal DNAs. The optimum temperatures for infection were 30°C forP. aphanidermatum and 25°C forP. ultimum, corresponding to the optimum temperatures for growth in vitro. The optimum concentration for infection of tubers inoculated by dipping in a suspension of oospores or hyphal swellings after wounding was found to be 103 reproductive organs/ml, whereas the infection threshold was 10 reproductive organs/ml.  相似文献

Summary Undamaged and damaged tubers of 10 cultivars were inoculated withPhoma foveata by two methods at five test centres over three years. Damaging tubers before inoculation increased the discrimination between cultivars. While this increase was small with tubers rolled in cornmealsand cultures, it was often substantial with those dipped in a suspension of comminuted agar cultures. Correlations between centres for the four tests were significant (P<0.05) and were highest for tubers inoculated with cornmeal-sand with or without damage. The gangrene scores of the cultivars in the four tests were correlated (P<0.05) with their field susceptibility ratings but there was a strong interaction between cultivar reaction and year of test. When tubers grown in Cumbria and Midlothian were tested at one centre over two years, the gangrene scores were also correlated (P<0.05).  相似文献

Summary Potato tubers were inoculated with two biotypes ofPhytophthora infestans then stored at 3,7, 10 and 15°C. Image analysis quantified average reflective intensity (ARI) of diseased tissue from cut surfaces of sample tubers. Tuber tissue infection and infection rate were measured by calculating Mean ARI of samples. Average tuber tissue infection and infection rate was minimal at 3°C (P.i.-US8 orP.i.-US1). Tuber tissue infection increased at temperatures >3°C, from 220 Mean ARI seven days after inoculation (dai) to 190–150 Mean ARI 50 dai (depending on cultivar and biotype ofP. infestans). Rate of tuber tissue infection caused byP.i.-US1 at 7°C was about zero in cv. Snowden but greater than −0.2 ARI day−1 (cvs Russet Burbank and Superior). Rate of late blight infection in tuber tissue generally increased with temperature from −0.2 ARI day−1 (at 7°C) in all cultivars to a maximum of −0.8 ARI day−1 (10°C).  相似文献

Summary The effects of biological, chemical and integrated control on the formation of selerotia ofRhizoctonia solani on new potato tubers were studied in experimental fields. Sprouts of seed tubers, sprouted in daylight, were inoculated withVerticillium biguttatum, an ecologically obligate mycoparasite ofR. solani. Fungicides were mixed with the top soil. Biological control byV. biguttatum increased the percentage of harvests with less than 5% loss due to grading from 24% (non-inoculated) to 56%. Fungicides at the recommended rates usually gave good results but lower doses were less effective in sand than in loam soils. When fungicides were combined with biological control, the results were often, better: harvests with less than 5% sorting loss increased from 56 to 81%. Integrated control with pencycuron at 25% of the recommended rate was about equal to chemical control with pencycuron at full rate. The advantages of integrated control are discussed. Summary Potato plants, after inoculation with a South African strain of biovar 2 or a strain of biovar 3 ofPseudomonas solanacearum were grown in growth chambers at temperatures ranging from 14/16°C (dark/light) up to 25/30°C. When grown at 14/16°C or higher, plants inoculated with the biovar 2 strain developed wilting symptoms and internal symptoms on progeny tubers and the disease was transmitted by the progeny tubers. When the plants were inoculated with the biovar 3 strain, wilting symptoms developed from 18/20°C, internal symptoms on progeny tubers from 20/22°C and the disease was transmitted by progeny tubers from plants grown at 16/18°C or higher.  相似文献

Summary The effect of the moisture level of potting compost at harvest on the susceptibility of tubers of three cultivars to late blight was investigated in the glasshouse in three successive years. Three moisture regimes (dry, moist, wet) were imposed at flowering time, the tubers harvested 3 weeks later and immediately dipped in a zoospore suspension ofPhytophthora infestans. Tubers from dry compost were significantly more susceptible than those from wet or moist compost, which showed similar levels of susceptibility.  相似文献

Summary Soil in a potato field naturally infested with black dot (Colletotrichum coccodes) was fumigated with methyl bromide at 126 g m−2 or left unfumigated. Potato seed tubers (cv. BP1) uninfected, lightly infected (1–25% surface affected), severely infected (26–100% surface affected) and severely infected withC. coccodes but dusted with prochloraz manganese chloride as Octave 2.5% DP at 750 g per 100 kg seed were planted in fumigated and unfumigated soil. When harvested, the incidence of black dot on the progeny of infected seed planted in unfumigated soil was twice that of progeny in fumigated soil, with progeny of uninfected seed having a 68.5 times higher disease incidence in unfumigated soil. Black dot on progeny tubers was reduced by pre-treatment of seed with prochloraz in fumigated soil only. With black dot infested fields, planting disease-free seed or treating seed with fungicides would not decrease disease on progeny tubers.  相似文献

Summary High grade potato seed tubers of three cultivars were inoculated in Scotland by vacuum infiltration in suspensions ofErwinia carotovora subsp.carotovora and subsp.atroseptica in different combinations, and transported to Valencia, Spain. They were cut into two to three pieces and planted in January 1984, 1985 and 1986 in fields furrow irrigated before and after emergence. High incidences of blanking and blackleg were associated with seed inoculated with high numbers ofE.c. atroseptica alone or together withE.c. carotovora and not with high or low numbers ofE.c. carotovora alone.E.c. atroseptica predominated at emergence in March in rotting seed pieces which failed to produce a plant, regardless of the seed treatment, and in diseased stems in April but it was replaced byE.c. carotovora in May. The levels of blanking and blackleg after inoculating 16 cultivars with both erwinias were not correlated between years nor were they significantly different within years among most of the cultivars.  相似文献

A plant systemic acquired resistance inducer, acibenzolar-S-methyl (ASM), was evaluated to determine the efficacy for suppression of Phytophthora blight of squash caused by Phytophthora capsici under field conditions. ASM was applied as foliar sprays before and after transplanting at rates of 17.5, 8.8, and 4.4 g a.i. ha−1. Application of ASM did not significantly reduce final Phytophthora blight incidences in 2 out of 3 field experiments; however, area under the disease progress curve (AUDPC) values were reduced significantly by ASM in all experiments conducted. Disease suppression by the three application rates of ASM was not significantly different. To determine the effect of application of ASM in conjunction with standard chemical fungicides, ASM was applied with mefenoxam (Ridomil Gold), copper hydroxide, and mandipropamid (Revus). AUDPC values and final disease incidences were consistently lower in field plots treated by the combination of ASM and standard fungicides than applications of these chemicals alone. Application of ASM resulted in significantly higher squash yield than the non-treated control in 2 of 3 experiments and plots treated with the combined use of ASM and standard fungicides produced the highest yields. These results suggest that ASM may induce plant resistance under field conditions, providing suppression of Phytophthora blight of squash, and that there may be some benefit to the integration of ASM and standard chemical fungicides.  相似文献

Summary The susceptibility of 21 potato cultivars to leak (watery wound rot) caused byPythium aphanidermatum was compared. Whole tubers were inoculated after wounding by dipping into a water suspension of 103 oospores/ml and incubated for 3 days at 25°C. The mean penetration of tissue calculated from the recording of lesion width and depth and the qualitative assessment of rot extension were highly correlated. For 10 of 12 cultivars tested at least twice, consistent reactions were obtained: cvs Korrigane. Superstar, Safrane and Yesmina were moderately susceptible, whereas Atlas, Diamant, Mondial, Obélix, Spunta and Timate were susceptible to highly susceptible. Tubers originated from two locations and/or two cropping seasons: thus, the performance of these cultivars under different production conditions reveals a satisfactory level of agreement with the test. Moreover, ratings assigned are supported by field or store observations.  相似文献

Summary The viability of five pathogens was decreased by treatment with hot water when tested in vitro.Polyscytalum pustulans was most sensitive andRhizoctonia solani least sensitive. Potato tubers were exposed to 55°C for 5 min in a commercial continuous hot water treatment plant using naturally contaminated seed tubers and tubers which had been inoculated by dipping in comminuted cultures. The frequency of eyes colonised byP. pustulans, Helminthosporium solani, andR. solani was reduced to virtually zero and the effect persisted on tubers subsequently stored at 4°C and at 15°C for up to 16 wk. Results withColletotrichum coccodes were inconclusive. Treatment suppressedPenicillium spp. which, however, rapidly recolonised the eyes during storage, leading to higher contamination levels in the treated than in the untreated tubers. With tubers inoculated withPhoma foveata, good control was achieved when the incubation period before treatment was 10 d but not when the period was 42 d.  相似文献

Summary Field-grown potato plants of cvs King Edward, Record, Maris Piper and Désirée were inoculated on seven different dates during the growing season of 1987 and 1988 with either potato virus YO (PVYO) or PVYN, using three viruliferous peach-potato aphids (Myzus persicae) per plant. In each cultivar, the proportion of progeny tubers infected with PVYO or PVYN was high in plants inoculated during the four weeks following emergence, the proportion declining to zero or close to zero in the subsequent 4–6 wks.  相似文献

Summary A single blighted tuber released on average 1.39 mg ofP. infestans mycelia and sporangia during simulated washing. In subsequent experiments up to 100% of tubers were infected when healthy tubers were washed in a suspension ofP. infestans equivalent to a concentration of 1.65 μg ml−1, i.e. that calculated to be in a commercial washing plant after washing a potato stock with approximately 1% of tubers infected with blight for 4 hours. Immature tubers were more prone to infection during washing than mature tubers. Damaging tubers, irrespective of their maturity, increased the incidence of tuber infection. The viability ofP. infestans isolates was significantly reduced byin vitro immersion in water at 44°C for 5 minutes. The infection of daughter tubers dipped in a suspension ofP. infestans for 3 minutes was prevented when the suspension temperature was 44°C. Summary The influence of freeze dried potato powder derived from tubers (cvs Désirée and Epicure) exposed to light as a culture medium on the growth, number and spore size of nine fungal pathogens was determined in vitro with particular emphasis on the role of glycoalkaloids. Greater reductions in pathogen growth reflected increased exposure to light of tubers prior to freeze drying. High correlations (% R2 adj) were obtained between glycoalkaloid concentration and growth rate of the majority of pathogens tested. Where spores were produced, numbers were in general significantly less when cultured on freeze dried powder derived from tubers exposed to light. No effects on spore size ofFusarium species were recorded, but spore length of remaining pathogens was reduced and spore width increased, with the exception ofC. coccodes where spore length and width was increased following culture onto freeze dried powder derived from tubers exposed to light.  相似文献

Summary In 1983 and 1984 there were reports in Norway of more severe late blight infection than usual on tubers of cv. Pimpernel, a cultivar without R-genes known to be somewhat resistant to both haulm and tuber infection. Results from routine tests confirmed earlier experience that cv. Pimpernel shows stable resistance to tuber late blight, although the ranking of cultivars may vary between years. In experiments during three harvest seasons tubers of different cultivars were inoculated with isolates ofP. infestans obtained from stocks of cv. Pimpernel with severe tuber late blight and with other isolates. In these experiments the isolates from cv. Pimpernel caused more rot on tubers of Pimpernel than did other isolates.  相似文献

Summary Various methods of screening for resistance to root-knot nematodes were compared and evaluated. Seedling populations ofSolanum spp., grown in clay pots and plastic tubes with silver sand and inoculated with juveniles ofMeloidogyne chitwoodi andM. hapla, showed large differences in the number of egg masses on roots 7 weeks after inoculation. The differences were reproducible when re-testing was done with cuttings and plants from tubers. No resistance toMeloidogyne spp. was observed with ten potato cultivars when grown in clay pots, plastic tobes or closed containers. Plants from tubers in growth pouches developed a large two-dimensional root system, and after inoculation with juveniles the infection process could be observed over 8 weeks. A method of infecting potato tuber tissue withMeloidogyne is described, using tuber slices in Petri dishes as a potential screening test for tuber resistance.  相似文献
