In an effort to optimize the efficiency of high‐density incubation of Atlantic cod Gadus morhua and haddock Melanogrammus aeglefinus eggs, the per cent hatch of eggs treated with four disinfectants (3% hydrogen peroxide, 1% polyvinylpyrrolidone iodine, 0.1% sodium hypochlorite and a 0.005% antibiotic solution – penicillin/streptomycin) was compared in both species. The per cent hatch of eggs of each species was greatest after a 24 h treatment with the antibiotic solution. The hatching success of eggs treated within the different disinfectant treatments depended upon the embryonic developmental stage in both species. The sodium hypochlorite treatment had the lowest % coverage of colony growth after disinfected haddock eggs were plated onto sterile agar media, highest survival rates to the end of the embryonic period, but the lowest per cent hatch.  相似文献

The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. We showed that sperm concentration could be assessed using spectrophotometry at 260 nm. Sperm motility significantly decreased after a 168‐h storage at 4 °C. A 1:9 dilution of sperm in a non‐activating medium (1/3 seawater and 2/3 freshwater, osmotic pressure: 360 mOsm kg?1) improved sperm storage. Sperm concentration, sperm velocity and storage capacity at 4 °C peaked during the medium period of the spawning season and then decreased to values close to those observed at the beginning of the reproductive period. The measured values of osmotic pressure, pH, protein, Na+, Cl? and Ca2+ concentrations of the seminal fluid were modified along the spawning period. Cell damage was noted at the end of the spawning period: local blebs were observed on the flagellum but also loops at its distal part. On the other hand, spermatocrit did not vary with the sampling date. In conclusion, cod sperm quality is modified during the spawning period, the highest‐quality samples being collected during the medium part of this season.  相似文献

Two year‐classes of Atlantic cod were reared in indoor tanks from November 2002 to November 2004. The average annual mortality was 21% and 36% for males and females respectively. The highest monthly mortality rates in females were observed around and during the spawning periods from March to July, maximally 7% and 20% for year‐class 2001 and 18% and 34% for year‐class 2000. The mortality in males was less dependent on season, reaching a maximum of 7% and 5% month?1 for year‐class 2001 and 2000 respectively. The maximum mortality during spawning increased with age from: 7%, 19% and 34% per month for 2‐, 3‐ and 4‐year‐old females respectively. The average gonadosomatic indices in 3‐ and 4‐year‐old mature females that died during the spawning periods were 22% and 26% respectively and most of them were spawning when they died. The results suggest that egg‐retention may have been the main cause of death in females during spawning.  相似文献

Commercial intensive fry production of Atlantic cod will be dependent on production of viable eggs independent of season. This can only be done by manipulation of maturation by photoperiod, but little is known about potential effects on egg characteristics and larval viability. In two cod broodstocks, maturation was successfully advanced or delayed 6 months compared with normal spawning season (March–April) by manipulation of photoperiod. The advanced broodstock spawned both in spring and autumn the same year. In two of the spawning tanks during autumn, ambient temperature was reduced after reaching 13.7°C during the first half of the spawning period. Egg quality and viability were monitored, and several egg batches were incubated, hatched and start‐fed for examination of growth and survival. Temperatures above 9.6°C resulted in significant reductions in fertilization and normal egg development. Concurrently, fractions of dead and unfertilized eggs increased with elevated temperature. Actual relative fecundity was not affected by temperature. Egg characteristics improved when temperature was controlled and lowered below 9.6°C. Occurrence of irregular spawners suggests that handling of broodstock fish should be avoided during maturation and spawning. Cod larvae originating from eggs of the advanced or delayed broodstocks were successfully reared beyond metamorphosis. Survival was 9.0–46.6% and 29.3% in green and clear water respectively. Survival correlated with both initial and average feeding conditions, but growth rate did not correlate with either of survival and feeding conditions. Specific growth rates (8.3–13.6% day−1) is comparable with other intensive rearing trials with cod, but were lower than reported from nature‐like systems.  相似文献

The potential effects of anthropogenic noise on the physiology of Atlantic cod have not been well described. The aim of the present study was to investigate the impact of anthropogenic noise on Atlantic cod stress response using cortisol as a biomarker as well as on broodstock spawning performance. Results showed that artificial noise consisting of a linear sweep from 100 to 1000 Hz can induce a transient and mild cortisol elevation with a clear noise intensity dose response. In all cases plasma levels returned to baseline levels <1 h post sound exposure. Daily exposure to a similar intensity and frequency noise range applied habitually to a broodstock population during the spawning window resulted in a significant reduction in total egg production and fertilisation rates thus reducing the total production of viable embryos by over 50%. In addition, a significant negative correlation between egg cortisol content and fertilisation rate was observed. These results confirm that cod can perceive noise generated within a frequency range of 100–1000 Hz and display a heightened cortisol plasma level. In addition, anthropogenic noise can have negative impacts on cod spawning performances.  相似文献

We conducted a study to evaluate the effects of holding water temperature on the fecundity and egg quality of photo‐manipulated Atlantic cod broodstock. Adult cod Gadus morhua were distributed among several 25‐m3 tanks. Three separate photoperiods were used for each group to obtain three spawning per year (in May, July and December). While photoperiod was controlled, the temperature was not controlled and followed the ambient seasonal pattern. Due to the uncontrolled seasonal temperature used, each spawning group experienced different temperatures at a particular gonadal developmental stage. Otherwise, all groups were treated the same. Results showed that the May and July spawning groups released significantly more eggs per kg of fish (four and three times more respectively) than the December spawning group. The egg fertilization success and proportion of normal eggs were also higher in the May and July groups. Possible reason for the difference may be that the May and July groups experienced lower temperatures (4–7°C) during oocyte maturation and ovulation than the December group (7–9°C). Our results show the importance of using lower/natural temperatures during oocyte maturation and ovulation to obtain good quality eggs in photo‐manipulated cod broodstock.  相似文献

Piscine mycobacteriosis causes losses in a number of fish species both in the wild and in aquaculture worldwide. Mycobacterium salmoniphilum infections have on several occasions been reported in farmed Atlantic salmon, Salmo salar L. The present study tested and confirmed the susceptibility of Atlantic cod, Gadus morhua L., an important yet relatively novel aquaculture species, to infection with M. salmoniphilum. Atlantic cod injected intraperitoneally with a suspension of this bacterium were maintained together with cohabitant (COH) fish in a flow-through marine water system at 10-11 °C. The fish were supervised daily and samples taken at 2, 7, 14, 23, 34 and 53 weeks post-infection and examined pathologically, bacteriologically and using molecular biology. Injected mycobacteria were re-isolated in high concentrations from both injected and COH fish groups. Death attributable to mycobacterial infection was observed in both injected (47%) and COH (28%) fish groups. Extensive development of granuloma in visceral organs, mainly the mesenteries, spleen, kidney and liver (lesser extent) and at later stages of the infection in heart tissues and gills, was observed in both injected and COH fish. Granulomas underwent a temporal progression of distinct morphological stages, culminating in well-circumscribed lesions surrounded by normal or healing tissue. Acid-fast bacilli were detected in both granulomas and non-granulomatous tissues. This study confirms that Atlantic cod is highly susceptible to M. salmoniphilum infection and that this bacterial species may be a threat to cod both in the wild and in the aquaculture.  相似文献

Farmed Atlantic cod with a mean weight of 4.8 kg were maintained for 9 weeks in sea cages and fed diets where the dietary fishmeal component was substituted with increasing proportions (0%, 22%, 63% and 100%) of meal from Antarctic krill, Euphausia superba. Wild‐caught cod were included in the study as external control. At termination of feeding, all fish were slaughtered and muscle pH was immediately recorded. The fish were then stored on ice for three days and assessed for muscle pH and objective (skin and fillet colour, and fillet texture) and subjective (sensory evaluation) quality criteria. Replacement of fishmeal with krillmeal in the diets resulted in the skin colour above and below the lateral line to be more red than the control group without krillmeal substitutions, even though this difference was not significant, and with more yellow hue. Additions of krillmeal increased the muscle whiteness and yellow hue compared with the control group and wild fish. There was no difference in red hue between the groups. Muscle pH, texture or sensory attributes were unaffected by dietary inclusion level of krillmeal. Wild‐caught cod deviated in several aspects from the farmed cod. It is concluded that the replacement of fishmeal with Antarctic krillmeal in the diets two months before slaughter did not move the sensory attributes more towards wild fish.  相似文献

We carried out an experiment to determine how rapidly the early incubation temperature of Atlantic cod eggs can be increased without affecting normal embryonic development and hatching. Atlantic cod eggs were incubated at a constant low temperature (4.5 ± 0.5°C; T5 – control) and four temperature increment treatments where the temperatures were increased stepwise from 4.5°C at zygote stage to 9.5 ± 05°C (T1‐8 h, T2‐32 h, T3‐64 h and T4‐96 h). Embryonic cell symmetry, embryonic mortality, hatching success and larval skeletal abnormalities, length and yolk sac volume were recorded. Larval samples were also taken at hatch for histological analysis. Except for higher egg mortality and lower hatching success in the T1, the differences among experimental groups were minor. Cell asymmetries and embryo mortalities were not significantly different between the control and T2–T4 treatment groups. Control larvae were significantly longer and had smaller yolk reserves at hatch than T1–T4 larvae and larvae from T2 had the largest yolk reserves. Tissue and organ histology of hatched larvae were similar. Considering embryonic cleavage pattern, hatching success and larval morphology and histology, a gradual increment of temperature in 32 h seems to be the better choice for future developmental programming studies in Atlantic cod.  相似文献

Effect of season and diet on muscle composition were evaluated in farmed Atlantic cod (Gadus morhua L.), fed varying levels of macro‐nutrients, and kept at two different light regimes during 1 year grow‐out in sea‐cages. The cod were fed seven different diets varying in protein, lipid and starch, in a mixture design. The diets spanned 4–20% starch, 8–26% lipid and 36–66% protein. Each dietary regime was subjected to two different light regimes: continuous light (24 h), or natural light (August 2001 to June 2002). Fish subjected to natural light started to mature in December/January and spawning was more or less completed during March/April. No maturation was registered in the continuous light groups at this point. No variation was found in muscle dry matter, protein or lipid concentration as a consequence of the dietary or light regime variations, except for the groups spawning in March. Glycogen varied from 1 to 6.5 mg g?1 wet weight, without any correlation to the present dietary variations. At the June 2002 sampling all groups given a natural light regime showed almost twice the concentration of muscle glycogen compared with fish subjected to continuous light. Such clear results were not measured at the December or March samplings. Increased dietary lipid resulted in lowered muscle vitamin E concentration. A strong covariation between muscle vitamins C and E was found at all samplings, and these showed a negative correlation towards eicosapentaenoic (EPA) and docosahexaenoic acid (DHA), and the ratio n‐3/n‐6 in muscle. The highest dietary vitamin E resulted in the lowest muscle vitamin C concentrations. All dietary lipid added was identical and of marine origin; in addition, the wheat added as a starch source summed up parts of the dietary lipid fraction, resulting in slightly decreased sum of polyenes in the diets holding the highest levels of wheat. The lean cod muscle consists almost solely of membrane lipids. These were, however, highly influenced by the dietary lipid composition, especially as concerned the different monoenes. Two of the diets showed tendencies to increased thiobarbituric acid‐reactive substances (TBARS). This was not reflected in the muscle concentrations of vitamins E or C, and did not result in any changes in muscle TBARS values at any of the samplings, except for the fish spawning in March. The water‐soluble fraction of the muscle protein, pH range from 4.5 to 6, did show a difference in structure when comparing our experimental cod to wild cod. We could also identify a differential pattern between some of the experimental groups. The method used to identify this was, however, not quantitative, and further studies are needed. Taste panel evaluation and shear force measurements after final sampling in June concluded with minor differences between muscles from the different diet groups, except for fish given one diet with relatively high protein, intermediate lipid and low starch levels. Fish kept at continuous light was described as less firm (texture) compared with fish kept at natural light, explained partly by the different size of these two categories of fish. In conclusion, both diet and season (spawning or not) did influence several of the measured muscle parameters.  相似文献

Sequential ichthyoplankton surveys were used to determine the spatial and temporal distribution of eggs and larvae over coastal spawning grounds of Atlantic cod (Gadus morhua) in Smith Sound, Trinity Bay, Newfoundland, during the spring and summer of 2006 and 2007. Egg densities showed similar patterns for both years with two peaks in abundance in spring (March–April) and late summer (late July). A clear progression of development stages (1–4) was observed in spring and summer in 2006 and summer in 2007, suggesting retention of eggs within the Sound during these periods. Modelled predictions of vertical egg distributions indicated eggs were broadly distributed near the surface during spring (March–April), but were concentrated below the pycnocline (>10 m) within the inner portions of the Sound during the summer months (July–August). Back‐calculated peaks in spawning based on water temperatures were estimated at 11 and 4 April for 2006 and 2007, respectfully, with late season peaks centred on 21–24 July for both years. Environmental data indicated cooler water temperatures and periods of high wind stress in spring, and warmer, calmer periods late summer, consistent with higher retention and faster development times on the spawning grounds later in the season. We conclude that spring and summer spawning events result in different distributions of early life stages and may lead to different distributions of juvenile and adult fish.  相似文献

The present trials aimed to investigate the effects of replacing fish meal with plant proteins in diets for cod, using a regression design where fish meal constituted the control. The plant protein diets were formulated to meet the amino acid requirements according to NRC (1993) and contained corn gluten meal, soybean meal, a mixture of these, or a mixture of wheat gluten meal and soybean concentrate. The plant protein fraction constituted up to 440 g kg−1 of the extruded diet. Two feeding experiments were conducted, one at high (11 °C) and one at low (6.5 °C) temperature. High growth and feed utilization were obtained in all diet groups at both temperatures. However, only in Exp.1 (11 °C), growth and feed utilization were linearly reduced by increases in dietary soybean and corn gluten meal, while no such effect was detected in Exp.2 (6.5 °C). Reductions in protein retention were seen at both temperatures with use of all evaluated plant protein sources. Due to large amounts of plant protein ingredients in diet, differences in diet amino acid composition were seen. These were partly reflected in the muscle free amino acid pool. Dietary plant ingredients did not affect whole body, liver or muscle proximate compositions, or liver indices. In both experiments, blood parameters were within the range of earlier reported normal values and indicated, together with low mortality, good fish health status. The results show that there is high potential to use protein‐rich plant ingredients in diets for Atlantic cod.  相似文献

During the development of breeding programme for Atlantic cod Gadus morhua L., in Iceland, genetic parameters were estimated for 1402 individuals, which were assigned with DNA profiling to 140 dams and 70 sires. The cod was reared in cages on the eastern and western coasts of Iceland from 2004 to 2005. At the average body weight of 1.8 kg, the estimated heritability (h2 ± SE) for body weight, gutted weight and the condition factor (CF) were 0.31 ± 0.06, 0.34 ± 0.04 and 0.24 ± 0.06 respectively. Genetic correlation (rG) in body weight between the two rearing locations was estimated as 0.95, which reflects a low G × E interaction. The estimated heritability for hepatosomatic index (HSI) and fillet yields was 0.061 ± 0.04 and 0.04 ± 0.04 respectively. The HSI and fillet yields were highly genetically correlated with body weight or 0.67 and 0.82 respectively. The genetic correlation between the CF and body weight was estimated as 0.31. There appears to be substantial amount of additive genetic variation for body weight suggesting that selection is likely to be successful. Low heritability for fillet yields and the HSI indicates less promise of genetic improvement. Assigning of parentage to individuals with DNA profiling was 80% successful.  相似文献

An industrial and experimental electrical method for stunning farmed Atlantic cod in air and seawater (SW), respectively, were compared. The impacts of sedation with AQUI‐S? and exercise to exhaustion before electrical stunning were also assessed to monitor the possible depletion of rested muscle energy levels by electrical stunning. Stress (blood glucose, haematocrit, muscle pH, muscle excitability, high‐energy phosphates and rigor mortis) and flesh quality (fillet texture, colour, liquid leakage (LL), gaping, residual blood and K‐value) were assessed. For the industrial stunning method, an average of 41 V, 0.2 A dc was applied to individual cod for 18–27 s. For the SW method, a bipolar square wave current (170 Hz, 33% duty cycle) was applied for 5 s. After stunning, recovery was prevented by exsanguination in chilled SW. There were no differences (P>0.05) between the two stunning methods except for a higher ultimate fillet pH for cod stunned in air 8 days postmortem. Exercise before stunning depleted muscle energy levels at slaughter, increased LL and fillets had redder and darker flesh after storage on ice for 8 days. Electrical stunning (in air) of AQUI‐S?‐treated fish partly depleted muscle energy levels (pH 7.3, ATP 18.7 μmol g?1, PCr 70.1 μmol g?1). However, flesh quality was not affected. Unless pre‐rigor filleting is the chosen processing strategy, electrical stunning of cod seems to be a promising stunning method.  相似文献

Lipids and essential fatty acids, particularly the highly unsaturated fatty acids, 20:5n‐3 (eicosapentaenoic acid; EPA), 22:6n‐3 (docosahexaenoic acid; DHA) and 20:4n‐6 (arachidonic acid, AA) have been shown to be crucial determinants of marine fish reproduction directly affecting fecundity, egg quality, hatching success, larval malformation and pigmentation. In Atlantic cod (Gadus morhua L.) culture, eggs from farmed broodstock can have much lower fertilization and hatching rates than eggs from wild broodstock. The present study aimed to test the hypothesis that potential quality and performance differences between eggs from different cod broodstock would be reflected in differences in lipid and fatty acid composition. Thus eggs were obtained from three broodstock, farmed, wild/fed and wild/unfed, and lipid content, lipid class composition, fatty acid composition and pigment content were determined and related to performance parameters including fertilization rate, symmetry of cell division and survival to hatching. Eggs from farmed broodstock showed significantly lower fertilization rates, cell symmetry and survival to hatching rates than eggs from wild broodstock. There were no differences in total lipid content or the proportions of the major lipid classes between eggs from the different broodstock. However, eggs from farmed broodstock were characterized by having significantly lower levels of some quantitatively minor phospholipid classes, particularly phosphatidylinositol. There were no differences between eggs from farmed and wild broodstock in the proportions of saturated, monounsaturated and total polyunsaturated fatty acids. The DHA content was also similar. However, eggs from farmed broodstock had significantly lower levels of AA, and consequently significantly higher EPA/AA ratios than eggs from wild broodstock. Total pigment and astaxanthin levels were significantly higher in eggs from wild broodstock. Therefore, the levels of AA and phosphatidylinositol, the predominant AA‐containing lipid class, and egg pigment content were positively related to egg quality or performance parameters such as fertilization and hatching success rates, and cell symmetry.  相似文献

Twenty egg batches spawned naturally from 17 different females over two spawning seasons were used to evaluate the egg quality of cobia Rachycentron canadum. A reduction in egg size was observed towards the end of the spawning season for both years. The proportion of floating eggs demonstrated a positive linear relationship with both yolk‐sac larval survival (r2=0.91, P<0.05) and batch larval production (r2=0.80, P<0.01). Viable egg batches (i.e. fertilization success >50%) were of higher batch fecundity, had larger eggs and a higher proportion of floating eggs than non‐viable batches (i.e. 0% fertilization success). Also, biochemical analyses revealed that these viable eggs had significantly higher protein and amino acid contents. A multiple regression model based on the proportion of floating eggs, batch fecundity and fertilization success provided the most accurate predictions of batch larval production (r2=0.95, P<0.001). Similarly, using the egg content of arginine/glycine and methionine significantly increased the correlation coefficient in the multiple regression model predicting larval deformity (r2=0.92, P=0.002). This study reveals that accurate determination of egg quality in cobia can be improved using a combination of several variables rather than a single variable.  相似文献

Wild and farmed Atlantic salmon ( Salmo salar L.) and Atlantic cod ( Gadus morhua L.) were collected to assess changes in mercury with size in wild vs. farmed fish. Mercury concentrations were compared with Health Canada and United States Environmental Protection Agency consumption guidelines. Lipid dilution of mercury was examined by comparing lipid-extracted (LE) and non-lipid-extracted (NLE) flesh samples in both farmed and wild fish. Mercury concentrations in the flesh and liver of farmed salmon were significantly lower than concentrations in wild salmon of similar fork length ( P <0.001), possibly due to growth dilution in rapidly growing farmed fish. Mercury concentrations were higher in LE tissue compared with NLE ( P <0.05), suggesting lipid dilution of mercury in farmed fish with a high lipid content. Farmed cod, which do not grow more rapidly than wild cod, did not have significantly different flesh and liver concentrations compared with wild cod of similar fork length ( P >0.05). Between species of farmed fish, cod had significantly higher mercury concentrations than salmon ( P <0.05), but neither farmed nor wild salmon mercury concentrations exceeded federal consumption guidelines. These results suggest that rapid growth rates and a high lipid content may play important roles in regulating concentrations of contaminants such as mercury.  相似文献

Broodstock of Atlantic cod were fed with three diets; astaxanthin supplemented (100 ppm kg?1) hydrated diet (30% water) (HWHAs), no additional astaxanthin supplemented (50 ppm kg?1) hydrated diet (30% water) (HWLAs) and a control diet (50 ppm kg?1 astaxanthin and 10% water). Throughout the spawning period, eggs were collected daily and fertilization success and proportion of embryos with normal cleavage were recorded. Larvae were reared using standard culture methods. Larval growth was recorded every 10 days and larval survival was recorded at 50 days post hatch. In general, broodstock fed with diets containing higher water content (HWHAs and HWLAs) and higher astaxanthin (HWHAs) had higher egg production and efficiency of egg output, lower egg incubation mortality and higher larval growth and survival. Broodstock fed with higher water content diets consumed less feed compared to control group. Also, broodstock fed with HWHAs and HWLAs recovered faster in terms of body condition than the control group. The egg bound mortality of female fish was significantly higher in control broodstock compared to broodstock fed with HWHAs and HWLAs. Broodstock fed with higher astaxanthin (HWHAs) had significantly higher fertilization success than other two groups however; no differences were found in proportion embryos with normal cleavage among different groups. Larval survival was significantly higher in HWHAs broodstock compared to other two groups. Our results showed that increased water content and astaxanthin levels in the diet could improve the condition and fecundity of Atlantic cod broodstock.  相似文献
