This study examined the effects and optimal conditions of double heat‐shock treatment (HST) for the suppression of cleavage in fertilized willow minnow (Gnathopogon caerulescens) eggs. In addition, we performed a cytological analysis of the nucleus and mitotic apparatus dynamics before and after HST. Tetraploidy occurred at the highest frequency (45.4%) when the first HST was delivered 25 min after fertilization and the second 15 min later. In this group, the spindle atrophied immediately after completion of the second HST; subsequently, three distinct patterns were observed in the nucleus and mitotic apparatus of the first cell cycle. In the first pattern, one pole disappeared and a monopolar spindle formed; chromosomes were unable to separate and accumulated at one pole where they formed a large nucleus. In the second pattern, the poles were dispersed or localized to one side of the nucleus, resulting in the failure of chromosomes to separate and the formation of a single nucleus. In the third pattern, the first cleavage was completed normally. Thus, double HST can block the first cleavage division by eliminating one centrosome or bringing two poles close together, giving rise to a subset of tetraploid embryos. 相似文献

ABSTRACT: The sex ratios of the offspring of males from broods of honmoroko Gnathopogon caerulescens that displayed thermolabile sex determination (TSD), and those from eggs collected from the wild, strongly suggest that the combination of parents plays an important supplementary role in TSD in this species. The proportion of females in all broods from eight pairings of fish captured in the field decreased significantly at 30°C compared to 20°C, and one brood was entirely female at the lower temperature. These results suggest that phenotypic males (XX-males) exist in nature, probably as a result of sex change from genetic females caused by TSD. Sex ratios in relation to water temperature fall into five patterns, two of which seem to result from normal pairings of XX-females and XY-males, and from pairings of XX-males and XX-females with non-thermosensitivity. Two other patterns are thought to result from the same types of pairing, but with thermosensitivity. The last pattern, which shows male bias at both low and high temperature, is not explained only by the combination of genotypic sex determination (XX/XY) and TSD. Fluctuating temperatures close to natural conditions showed little potential to masculinize broods laid in the field between April and early June. However, the sex ratios of fish spawned in early June showed male bias at low and high temperatures. The sex determination mechanism in G. caerulescens may involve the interaction between temperature and sex-determining genetic factors, and the relative importance of each component differs with breeding season. 相似文献

Feeding incidence or number of larvae with preys (FIC) and intensity or number of prey per larvae (FIT) at first feeding of Pacific red snapper ( Lutjanus peru) larvae was investigated under different conditions: prey type (rotifer and copepod nauplius) and density, nauplii size, light intensity, water temperature, salinity and microalgae concentration. Rotifers were not consumed at any prey density and FIC increased significantly when a high nauplii density (10 > 1, 0.1 mL?1) and light intensity (2000 > 1000, 500, 0 lx) were supplied. In a multifactorial experiment where light intensity (2000, 2500, 3000 lx), tank colour (grey and black) and prey type (nauplii and a mixed diet: rotifers and nauplii) were tested, a significant difference was found only for light intensity and prey type with a significant interaction between these factors. FIC was significantly higher with nauplii stage I–III than IV–VI and also at 25 °C than at 28 °C. Green water (0, 0.3 × 106 or 1 × 106 cells mL?1) and salinity (25, 30, 35 gL?1) did not affect FIC. FIT was not affected by any variables tested except in the density experiment where it was significantly higher at 10 nauplii mL?1. 相似文献

Southern bluefin tuna, Thunnus maccoyii, are cultured in Australia following collection of wild juveniles. Hatchery culture from egg is in the experimental stage. High early mortality has hindered the production of quality juveniles in the hatchery. This study investigated the visual capacity of T. maccoyii during early larval ontogeny in order to describe the best larval rearing conditions to produce high‐quality seed stock. Functional visual ability, determined through behavioural experimentation, identified the effect of light intensity, prey density, turbidity, tank colour and turbulence on the feeding response. Larvae were visually challenged to feed under a range of conditions in short‐duration (4 h) feeding experiments. Feeding performance was measured as the proportion of larvae feeding and the intensity of feeding. First‐feeding performance was positively affected by increasing prey density and lower turbidities and unaffected by light intensity, tank colour, turbulence, prey size and larval density. The key findings from feeding experiments on 6 and 9 dph larvae was that as T. maccoyii aged, lower light intensities and higher prey densities significantly increased feeding performance. In addition, the study has identified that high light intensity and high air‐driven turbulence induced significant mortality. The proficient first‐feeding response indicated that early mortality common in culture is unlikely to be associated with a failure to initiate feeding. Our findings show the use of low light intensity has the potential to significantly improve survival and feeding response during the first two critical weeks of culture, when the major bottleneck in hatchery production is currently experienced. 相似文献

We investigated the first‐feeding success of two species: southern bluefin tuna (Thunnus maccoyii) and yellowtail kingfish (Seriola lalandi) to determine if similar culture parameters can be used for both, especially when S. lalandi are held in the same tanks as prey for T. maccoyii. The feeding performance (proportion and intensity) was examined in three short‐duration (4 h) experiments: prey density, prey size and larval density. Increasing prey density from 0.5 to 25 rotifers mL?1 increased the proportion of T. maccoyii and S. lalandi larvae feeding. Prey size alone did not affect feeding in either species. Seriola lalandi had a decreased proportion of larvae feeding when larval density reached 50 larvae L?1 concurrent with a gradual increase in feeding intensity between 2 and 50 larvae L?1. In T. maccoyii, there was no pattern to the effect of larval density on the proportion of larvae feeding. The overall feeding performance of larvae was higher in T. maccoyii than S. lalandi. Increased prey density improved the first‐feeding ability of T. maccoyii and S. lalandi larvae. The effect of larval density on S. lalandi feeding requires further investigation, to ensure that they remain feeding when provided as prey in T. maccoyii culture. The identification of factors in this study, which increase first‐feeding success, will improve the culture of both species. 相似文献

Based on existing models of fish vision and turbulence-mediated ingestion rates, we develop a model of ingestion rates in larval fish that combines several physical properties of the environment (turbulence, irradiance, light attenuation) and visual characteristics of predators and prey. The model of visual range was calibrated with observed estimates in larval herring ( Clupea harengus ) and cod ( Gadus morhua ). The improved visual ability with length of larvae was predicted to be the most sensitive part of the model. Both turbulence and light had strong impacts on the ingestion rate of larval fish. The optimal level of turbulence increased with larval length. Also, due to the exponential decay of light with depth, it was predicted that larvae will have higher ingestion rates near the surface, even at high wind velocities. It is also proposed that larvae (particularly the smallest larvae) should concentrate on larger prey in turbulent environments. We suggest that biophysical models of larval growth and survival in field situations should include these factors to account for environmental effects on growth, survival and recruitment processes in the early life stages. 相似文献

This study aimed to evaluate the performance of Pyrrhulina larvae, Pyrrhulina brevis, under different photoperiods and feeding frequencies. A completely randomized design was used, with four replicates in a 5 × 2 factorial, with five photoperiods (6L:18D, 12L:12D, 16L:8D, 20L:4D, 24L:0D) and two feeding frequencies (two or four times a day). Four hundred larvae with an initial length of 4.6 ± 0.09 mm were randomly distributed in 40, 1‐L containers at a density of 10 larvae/L. Feeding was performed with Artemia nauplii in the ratio of 150 Artemia nauplii/larva/feeding. After 15 days, all larvae were euthanized for measurements of length (mm), final weight (mg), survival rate (%) and larvae uniformity in weight an length (%). The data were subjected to analysis of variance and subsequently, in case of significance, a Tukey test was performed to compare means. There was no interaction between photoperiod and feeding frequency on studied variables (P > 0.05). Likewise, there was no influence of feed rate on the same variables (P > 0.05). There was only influence of photoperiod, where the larvae subjected to longest photoperiods (24L:0D and 20L:4D) showed improved weight and length (P < 0.05). ABSTRACT: The abundance of food organisms and feeding habits of larval and juvenile Japanese flounder were examined during the period from May to August in 1999, 2000 and 2001 at the sandy Ohama beach, the central Seto Inland Sea. The food organisms collected with a sledge net consisted of 40 families from 18 orders, dominated by mysids, crangonids and gammarids. The mean densities of mysids, crangonid shrimp ( Crangon spp.), gammarids and fish were 2.74, 6.74, 2.91 and 0.15 individuals/m2 , respectively. The main prey of the flounder ( n = 202; range of total length 9.80–75.95 mm) was mysids and small crangonid shrimp (<14 mm in body length). Prey fish availability was low, as the density of fish was low. The small crangonid shrimp was abundant, and the large crangonid shrimp, which could prey on larval flounder, was not abundant. The crangonid shrimp was important not as a predator for the flounder but as prey. The flounder preferred epifaunal mysids, Nipponomysis ornata and Anisomysis ijimai , to sand-burrowing mysids, Iiella oshimai , and avoided crangonid shrimp. 相似文献

The effects of larval density, rearing media, and protein additions on fish larval survival were tested by rearing larvae in small containers. Yolk-sac larvae of Japanese flounder, Paralichthys olivaceus, were transferred 1 day after hatching to plastic dishes and cell-culture microplates by Pasture pipette at densities ranging from 0.3 to 30 individuals/ml and incubated at 16 °C without food. Dead larvae were removed and counted every 12 h. Three peaks of larval mortality were observed: the first peak appeared just after the transfer, the second corresponded to the completion of yolk absorption, and the third occurred 10–12 days after the transfer. Since about 50% of larvae survived until the third peak, it was clear that mortality at the second peak (completion of yolk absorption; equivalent to the first feeding) was not due to starvation. At the first mortality peak, better survival was unexpectedly recorded at higher larval densities (30 individuals/ml), suggesting that some material beneficial for survival is derived from the larvae. This idea was further supported by the improved survival in media previously used to rear larvae at high densities. The addition of bovine serum albumin (BSA) at 10 μg/ml effectively reduced mortality in microplates and in 30-l tanks. To reduce the larval mortality just after handling, addition of egg white at a dilution rate of 1/30,000 (vol/vol) or less is recommended as a practical alternative to BSA. Since the addition of polyethylene glycol 6000 (1 μg/ml) also effectively reduced mortality, reduction in mortality could not be attributed to a specific mechanism. The precise determination of the magnitude and timing of larval mortality was attained through the use of small containers, and a method to prevent mortality due to handling was identified in this study. These types of tests may allow the logical evaluation and establishment of better rearing procedures for seed production in aquaculture. 相似文献

Successful natural spawning of Pomacanthus semicirculatus in captivity from 11 September to 18 October, 2006 is described for the first time. Each female laid an average of 230 000 eggs during the spawning period. Fertilized eggs were spherical, transparent and buoyant and had a mean diameter of 0.61 ± 0.03 mm (mean ± SD). Embryonic development lasted 18–21 h at 28.5 °C. Newly hatched larvae were 1.35 ± 0.02 mm in total length (TL) with 27 (12+15) myomeres and had an oil globule in the ventroposterior area of the yolk sac. Larvae completed yolk absorption within 3 days post hatching at 2.37 ± 0.05 mm TL. Larvae were fed either 100% microalgae ( Nannochloropsis sp.), 100% s-type rotifers ( Brachionus rotundiformis ), 100% dinoflagellates ( Gonyaulax sp.) or different combinations of the three (50%:50%:0%, 30%:35%:35%) to determine the effect of live feed on the survival rate. The survival was significantly ( P <0.001) better in larvae fed a combination of diets (30%: 35%: 35%) than others. These results indicate that P. semicirculatus is a potential species for captive-breeding programmes and the use of a combination of diets (microalgae plus s-type rotifers and dinoflagellates) may be a suitable first food for fish larvae. 相似文献

Using vertically stratified data of the abundance of silver hake (Merluccius bilinearis) larvae and concentrations of copepods collected in the field, we examine relationships among the vertical distribution of larval fish, their potential prey, feeding success and water column turbulence. Water column turbulence and associated stratification parameters were estimated from: (i) in situ measures of turbulent kinetic energy dissipation (?) provided by an EPSONDE profiler; (ii) in situ wind speed; (iii) the Richardson number (Ri); and (iv) the buoyancy frequency (N2). Small (< 5 mm total length) silver hake were more abundant in the least turbulent waters (i.e. at a minimum in the rate of dissipation of turbulent kinetic energy, ? < 10–7 W kg?1; Ri > 0.25; N2 > 0.001 (rad s–1)2). Partial correlations amongst ?, N2 and small hake larvae were significant only for N2. The abundance of larger (> 5 mm total length) hake larvae was positively correlated with depth and was not associated with either ? or N2. Vertical distributions of three potential prey (classified by stage) were variable. Early stage copepodids were positively correlated with N2 and negatively correlated with ?. We found no evidence of diel distribution patterns for small (< 5 mm total length) hake larvae or for any of the developmental stages of the copepods examined. Neither estimate of water column turbulence inferred from wind speed nor from Ri was meaningfully related to in situ estimates of ? or to larval fish abundance. Feeding success, measured either as prey items (gut)–1, average prey length, or total prey volume (gut)–1, was not related to predicted encounter rates between days. However, the average prey length (gut)–1 was significantly (P < 0.01) related to water column turbulence. These conflicting results suggest that the relationship between larval feeding and the environment is more complicated than assumed. We conclude that without substantial high resolution in situ examination of the relationship between the vertical distributions of turbulence, larvae and their prey, the growing acceptance in the secondary literature that turbulence has a positive and biologically meaningful effect on trophic interactions between fish and their zooplankton prey (a generalization based largely on modelling and laboratory experiments) is premature. 相似文献

The point of no return (PNR) and disappearance of the oil droplet were measured in Chirostoma estor larvae as a function of the time of first feeding. In a separate trial, growth and survival of larvae fed rotifers enriched with Chlorella sp., cod liver oil and corn oil were assessed. Fatty acid and lipid composition of eggs, oil droplets, egg yolk, feed and larvae were also evaluated. The PNR was found between 7 and 8 days posthatching (dph). Total oil droplet depletion occurred between 7 and 11 dph, depending on the time of first feeding. Best growth and survival were obtained in larvae fed with Chlorella‐enriched rotifers, followed by those fed cod liver oil‐enriched rotifers. In larvae fed corn oil, Chlorella and cod liver oil‐enriched rotifers, total oil droplet depletion took place on days 9, 10 and 11, respectively. There was a direct relationship between presence and duration of oil droplets and the survival of larvae under different starvation conditions. The feed source could prolong the existence of the oil droplet depending on particular dietary supply of essential fatty acids; the time of its disappearance could be a useful indicator of larval vigour and health status. 相似文献

A 8 week growth trial of three feeding frequencies (2, 4 and 6 meals day?1) and three dietary protein levels (30%, 34% and 38%) was conducted to investigate the influence of feeding frequency and dietary protein level on growth, feed utilization, serum free essential amino acids (EAA) dynamics and nitrogen (N), phosphorus (P), carbon (C) retention in juvenile allogynogenetic gibel carp (Carassius auratus gibelio) ‘CAS III’. The results showed that feeding frequency and dietary protein level increased feed intake (FI), specific growth rate (SGR) and feed efficiency (FE). SGR increased significantly with the increase in feeding frequency and dietary protein level. But, the increase in dietary protein level from 30% to 38% in group 6 meals day?1 did not result in better growth. Compared with the group of 4 meals day?1 × 38%, the fish fed the diet containing 30% crude protein at 6 meals day?1 obtained similar SGR but showed lower FIp (protein intake). Compared with 2 meals day?1, NREs (nitrogen retention efficiency) of 4 and 6 meals day?1 were relatively higher. PRE (phosphorus retention efficiency) and CRE (carbon retention efficiency) increased significantly with feeding frequency while TNW (total nitrogen waste output), TPW (total phosphorus waste output) and TCW (total carbon waste output) decreased significantly. Feeding frequency and dietary protein level increased trypsin activity. Serum free EAA concentrations varied more synchronously at 6 meals day?1 than 2 meals day?1. It could be concluded that increasing feeding frequency could improve growth by increasing feed intake and feed efficiency for juvenile gibel carp. Twenty‐four archival tags were recovered from Pacific bluefin tuna previously released off Tsushima Island in the East China Sea. By analysis of the time‐series data of the pressure and the ambient and internal temperature from the 24 tags, we examined the relationship between the tuna's pattern of diving and the thermocline depth. In the East China Sea, diving and feeding events occurred throughout almost the entire day in both winter and summer, suggesting that the purpose of diving is for feeding. In summer, the feeding frequency was greater than that in winter, which corresponds to the fact that growth is more rapid in summer than in winter. During summer in the Kuroshio–Oyashio transition region, on the other hand, feeding events were much more frequent than those in the East China Sea, in spite of a lower diving frequency. The mean horizontal distance traveled was also significantly higher and it seems that in this area they may move horizontally to feed on prey accumulated at the surface. We conclude that, in addition to the ambient temperature structure, the vertical and horizontal distribution of prey species plays an important role in the feeding behavior of Pacific bluefin tuna. One bluefin tuna migrated to the Oyashio frontal area, where both the horizontal and the vertical thermal gradients are much steeper. The fish spent most of the time on the warmer side of the front and often traveled horizontally to the colder side during the day, perhaps to feed. This implies that there is a thermal barrier effect, in this case from the Oyashio front, on their behavior. The frequency of feeding events was low, although all the monitored fish dived every dawn and dusk, irrespective of the seasons or location. It is possible that these twice‐daily diving patterns occurred in response to the change in ambient light at sunrise and sunset. 相似文献
