 菊花白色锈病(chrysanthemum white rust,CWR)是影响菊花生产效益的重要病害,明确菊花高抗品种‘C029’的抗病基因CmWRKY15-1响应菊花白色锈病的分子机制,对于培育菊花抗病新品种具有重要的理论和现实意义。本研究以菊花高抗品种‘C029’为试验材料,构建沉默载体pTRV2-CmWRKY15-1与过表达载体pBI121-CmWRKY15-1,利用农杆菌介导法转化菊花‘C029’,分别获得8株沉默株系,表达效率约为正常水平的30%~60%,和3株过表达株系,其中一株的表达水平为野生型(WT)的3.4倍,命名为OE-1。在病原菌侵染条件下,qRT-PCR测定WT与转基因株系中病程相关基因的表达量,探究CmWRKY15-1在菊花抵抗白色锈病过程中的功能。病原菌侵染后,在WT与OE-1中,NPR1和PR5的表达水平呈先上升后下降趋势,其表达量在OE-1中早于WT(分别在24和38 h)达到最高峰,PR1的表达量在OE-1于38 h达到峰值。在沉默株系中,NPR1和PR5的表达则受到抑制,且一直低于WT与OE-1株系。结果表明:CmWRKY15-1通过调控病程相关基因参与菊花抵抗白色锈病的反应。综上,CmWRKY15-1对菊花‘C029’抗白色锈病起到调控作用。  相似文献

Puccinia horiana is the causal agent of chrysanthemum white rust or Japanese rust. This microcyclic autoecious rust has a quarantine status and can cause major damage in the commercial production of Chrysanthemum x morifolium. Given the international and often trans-continental production of planting material and cut flowers of chrysanthemum and the decreasing availability of registered fungicides in specific regions, breeding for resistance against P. horiana will gain importance and will need to involve the appropriate resistance genes for the pathotypes that may be present. As pathotypes have not been well characterized in this system, the main objective was to build an international collection of isolates and screen these on a large collection of cultivars to identify different pathotypes. Using a robust and high throughput bioassay, we tested 36 selected cultivars with 22 individual single-pustule isolates of P. horiana. The isolates originated from three different continents over 4 different collection years and included some isolates from cultivars previously reported as resistant. In most cases the bioassays resulted in a clear scoring of interaction phenotypes as susceptible or resistant, while in several cases consistent intermediate phenotypes were found, often on specific cultivars. Twenty-four of the cultivars gave a differential interaction phenotype profile. All isolates produced a unique profile, infecting a minimum of 4 and a maximum of 19 differential cultivars. Based on the Person analysis of these profiles, this pathosystem contains at least seven resistance genes (and seven avirulence genes), demonstrating the highly complex race structure in this pathosystem.  相似文献

Resistance to the first successful anticoagulant rodenticide, warfarin, was detected in the Norway rat (Rattus norvegicus Berk.) in 1958, and it has since expanded to cover other commensal rodent species (R. rattus L., Mus musculus L.) and the majority of later-developed anticoagulant compounds. The purpose of this paper is to give an up-to-date picture of the current distribution of anticoagulant resistance in Europe, as revealed by the questionnaire dispatched by EPPO (European and Mediterranean Plant Protection Organization) to its member countries in 1992. Replies were received from 13 countries (43% of EPPO members), and it was found that broad-spectrum resistance occurred in practically all those countries that reported testing activity. A likely conclusion based on this finding is that true distribution of resistance is certainly more extensive than documented by the questionnaire replies. Another conclusion drawn by the EPPO Rodent Control Panel was that the existing resistance detection protocols were no longer adequate to cover the whole array of existing anticoagulant rodenticides. Consequently, a new guideline including novel methods particularly for detecting resistance to single-dose anticoagulant compounds was urgently needed. EPPO started work on biological control agents (BCAs) in 1996, and the joint EPPO/IOBC Panel was established in 1997. The history of the Panel is provided and EPPO Standards developed by the Panel are described. These Standards are: PM 6/1 ‘First import of exotic biological control agents for research under contained conditions’, PM 6/2 ‘Import and release of non‐indigenous biological control agents’ and PM 6/3 ‘List of biological control agents widely used in the EPPO region’. The last of these has been annually updated as a ‘Positive List’ of BCAs for which EPPO recommends its member countries to use a simplified procedure for import and releases. EPPO activities in biological control have been focused on the safety aspects of the introduction of invertebrate BCAs. However, the scope of EPPO's work in this area is now under review, and a number of issues are being considered by the Working Party on Phytosanitary Regulations. These include the remit of the Panel and Standards in relation to micro‐organisms, the evaluation of potential environmental benefits as well as potential risks from releases and the potential for use of BCAs against regulated pests and those recommended for regulation.  相似文献

Quinone outside inhibitors (QoIs) and succinate dehydrogenase inhibitors (SDHIs) are major groups of agricultural fungicides. However, resistance to some of these fungicides has been reported in a Japanese population of Puccinia horiana, the causal agent of chrysanthemum white rust disease. Because their mechanisms are not well understood, we investigated the existence of mutations in QoI and SDHI target protein-encoding genes. Eight out of nine isolates from cultivated chrysanthemum carried L275F and L299F amino acid substitutions in cytochrome b, the target protein of QoIs. These isolates showed 23- and 17-fold higher EC50 values for the QoI fungicides azoxystrobin and kresoxim-methyl, respectively, in basidiospore germination inhibitory tests, while they were hypersensitive to another QoI, famoxadone. All nine isolates were resistant to SDHI oxycarboxin and carried the I88F substitution in SdhC. This substitution was orthologous to the SdhC-I86F substitution found in some Brazilian isolates of the soybean rust fungus, Phakopsora pachyrhizi, showing reduced sensitivity to some SDHIs. Although the rarity of wild-type sensitive isolates, the subsequent limited number of comparisons between wild types and mutants, and a difficulty in applying reverse genetic analysis to this obligate parasite, are obstacles in making definitive conclusions, L275F and L299F in cytochrome b and SdhC-I88F are suspected to be responsible for the different patterns of sensitivity to QoI and for oxycarboxin-resistance in P. horiana, respectively.  相似文献

In 2011, EPPO conducted a survey among its member countries to re‐evaluate the status of 14 pests currently listed on the EPPO A2 List of pests recommended for regulation as quarantine pests: Cacoecimorpha pronubana, Cacyreus marshalli, Ciborinia camelliae, Dickeya (Erwinia) chrysanthemi pv. dianthicola, Frankliniella occidentalis, Impatiens necrotic spot virus, Leptinotarsa decemlineata, Phialophora cinerescens, Puccina horiana, Quadraspidiotus perniciosus, Raspberry ringspot virus, Tomato spotted wilt virus, Viteus vitifoliae, Xanthomonas fragariae. The Japanese flower thrips, Thrips setosus, is a polyphagous insect which in Europe has mainly been observed on hydrangeas (Hydrangea spp.). The first report of T. setosus in the EPPO region was in the Netherlands (2014). Since then it has been observed in four other countries inside the EPPO region: Germany (2015), the United Kingdom (2016), France (2016) and Croatia (2016). Its dispersal is most likely to be related to trade. Because of its polyphagous nature, there is a risk of spread to other economically important crops. Therefore, T. setosus was included in the national survey project ‘STATREGO’, during which the status of several invasive plant pests and diseases in Belgium was determined.  相似文献

The European and Mediterranean Plant Protection Organization (EPPO), in the framework of its activities in the Mediterranean Basin, launched a questionnaire to gather information on eradication actions and control actions for invasive alien plants. Thirty‐four (34) eradication actions were reported, 16 occurring in Spain, 7 in Italy, 7 in France, 1 in Portugal, 1 in Malta, and 2 on the southern rim of the Mediterranean, with 1 in Israel and 1 in Tunisia. Twenty‐two (22) species were targeted, and the habitats where such actions were undertaken most frequently were coastal dunes, sandy shores and aquatic ecosystems. Most of the species that have been the object of an eradication action were known invasive species recorded in the EPPO lists, such as Ailanthus altissima, Buddleia davidii, Carpobrotus spp., Eichhornia crassipes, Ludwigia peploides, Pennisetum setaceum, Pistia stratiotes, Solanum elaeagnifolium and Sicyos angulatus. Other eradicated species have a limited distribution in the EPPO region and may represent emerging invasive plants, such as Ambrosia confertifolia, Cylindropuntia rosea, Leersia oryzoides, Leptochloa fusca subsp. uninervia and Marsilea drummondii. Control actions concerning 90 species were also reported. A large number of eradication actions, as well as management actions, are therefore happening in Mediterranean countries. At present countries appear to operate with complete autonomy, without consulting neighbouring countries or other stakeholders within the country. There is therefore a need for coordination of new eradication actions.  相似文献

Clavibacter michiganensis subsp. sepedonicus has a clearly restricted geographical distribution in the EPPO region, being present almost exclusively in the north and east. Countries where it is absent take strong phytosanitary measures to prevent its introduction. Within the European Union, countries where it is present aim to eradicate the disease. Although invasive alien plants are gaining increased attention within EPPO countries, there is no existing widely agreed method to identify those alien plants that are considered invasive and represent the highest priority for pest risk analysis. In the framework of the ad hoc Panel on Invasive Alien Species, EPPO proposes a prioritization process for invasive alien plants designed (i) to produce a list of invasive alien plants that are established or could potentially establish in the EPPO region and (ii) to determine which of these have the highest priority for an EPPO pest risk analysis. The process consists of compiling available information on alien plants according to pre‐determined criteria, and can be run at the EPPO region level, or at a country or local area level. These criteria examine whether the species is alien in the area under study, and whether it is established or not. The criteria used primarily rely on observations in the EPPO region but, if the species is not established, the invasive behaviour of the species in other countries should be investigated, as well as the suitability of the ecoclimatic conditions in the area under consideration. The spread potential, the potential negative impacts on native species, habitats and ecosystems, as well as on agriculture, horticulture or forestry are considered. If the species qualifies as an invasive alien plant of major concern through this first set of questions, the process then investigates the efficiency of international measures (to be justified through a pest risk analysis) to prevent the entry and spread of the species. The second set of questions are designed to determine whether the species is internationally traded or enters new countries through international pathways for which the risk of introduction is superior to natural spread, and whether the species still has a significant suitable area for further spread. If used by several EPPO countries, this prioritization process represents an opportunity to provide consistent country lists of invasive alien plant species, as well as a tool for dialogue and exchange of information.  相似文献

An EPPO Contingency Planning Workshop for a Forestry Pest was held in Zlatibor, Serbia, between 27 and 29 November 2018. In total, 55 experts from 21 EPPO member countries attended the 3‐day workshop. Participants were split into four groups for the exercise and each group acted as an ‘outbreak management team’ for a scenario based on the finding of a quarantine pest for forestry. Only when the exercise had started were participants informed that the outbreak scenario centred around the fictitious discovery of pinewood nematode (Bursaphelenchus xylophilus) in the Tara National Park in Serbia. As the scenario developed, each team had to organize themselves to manage a number of issues designed to mimic the development of an outbreak over the first month following its detection. It was clear from the feedback from participants that that contingency planning workshop was a valuable exercise for EPPO to conduct and participants highlighted the importance of it to prepare themselves for real‐life situations.  相似文献

为明确不同花色菊花品种上西花蓟马Frankliniella occidentalis发生情况及种群变化规律,利用蓝色粘虫板调查了大棚种植的7种单色(黄色、橙色、白色、绿色、粉色、紫色、红色)以及4种双色(黄绿、黄红、白绿、黄白)菊花品种上不同时期西花蓟马的种群密度及雌雄性比。结果表明,根据四分位法得到西花蓟马的发生早期与菊花苗期重合,主要发生期与菊花开花期重合,发生晚期为切花后生长期。在所有单色菊花品种中,西花蓟马在黄色菊花品种上的种群密度最高,主要发生期虫口密度达19.98头/板;且雌雄性比最大,为5.85,显著高于其它单色菊花品种上的雌雄性比。在所有双色菊花品种中,黄绿菊花品种上的西花蓟马种群密度最高,主要发生期虫口密度达26.89头/板,显著高于其它双色菊花品种上数量,且雌雄性比也最大,为6.87。菊花开花是导致西花蓟马种群迅速上升和种群中雌虫比例迅速增加的主要原因,且西花蓟马偏好为害与黄色花色相关的菊花品种。  相似文献

Walnut (Juglans regia L.) is traditionally present in most European countries as an ornamental tree, and in Southern Europe in particular it is grown for both fruit and wood. Since the 1980s, to supply the increasing demand for walnut timber, large areas of southern and central Europe, from France to Hungary, have been planted with black walnut (Juglans nigra L.) to provide wood for furniture production. The fungus Geosmithia morbida and its vector Pityophthorus juglandis, causing the thousand cankers disease of walnut in the USA in the last 2 decades, were recently reported in Europe (in Italy) on both walnut species. Thousand cankers disease can have a high negative impact on the landscape and economy of many agricultural and forest areas. Following a detailed pest risk analysis performed by EPPO in 2015, both organisms were included in the EPPO A2 List of pests recommended for regulation as quarantine pests. The main biological, epidemiological and monitoring aspects of thousand cankers disease and its status in Europe are reported.  相似文献
