将生后2天、2周、4周、8周、12周、16周、20周、24周的家兔胫骨前肌分成前、后亚体,分别得到腓得神经主要肌分支的支配,腓深神经分支的神经分布型式随年龄增长而递降.每个肌亚体组织化学特征经由乙酰胆碱碘溶液孵育后的运动终板而确定.肌纤维可分成SO、FG、FOG和FO型.除外生后2天龄之外,前、后亚体内SO、FG、FOG肌纤维在生后2周龄、4周龄、8周龄分别约占30%左右,生后12周龄以后,在前亚体内的SO型与FOG型纤维比率下降和FG型纤维升高要超过后亚体.全部肌纤维型在生后2周龄、4周龄、8周龄、12周龄的每个肌亚体的深、中、浅部都是均匀性分布.然而,不同部位的肌纤维类型差别明显,深部以氧化型为主,而浅部则以糖酵解型为主.这种差别在生后16、20、24周龄是最典型的.两个亚体生后发育期间的每条肌纤维毛细血管数(NCF)表明SO>FO>FOG>FG型纤维和毛细血管数与肌纤维横切面积比(CCA)显示FO>SO>FOG>FG型纤维,由此指出氧供较大需求的获得是通过减小肌纤维的面积而不仅仅是依赖于增加毛细血管数量.  相似文献

The middle gluteal muscle of five, two-year-old untrained trotters was investigated by repeated needle biopsy sampling over a training period of six months. A second group of five, three-year-old untrained horses was included to examine the effect of growth. After the training period increases were found in the relative distribution of slow twitch (ST) fibres from 18 per cent to 25 per cent and fast twitch (FTa) fibres from 36 per cent to 45 per cent, and a decrease in FTb fibres from 46 per cent to 30 per cent. A proportionally equal reduction (approximately 18 per cent) in the cross sectional area of all fibre types was observed after the first two months of training succeeded by an increase to approximately pretraining levels at the end of the period. The number of capillaries per fibre was enhanced from 1.7 to 2.4. Proliferation of capillaries occurred around fibres of all types. The metabolic adaptations showed increases in the activities of 3-hydroxy-acyl-CoA dehydrogenase (HAD) (50 per cent) and citrate synthase (31 per cent). Growth had no effect on the relative fibre type distribution nor the capillary per fibre ratio, but as the mean fibre area increased 36 per cent (primarily because of increases in FT fibres) the number of capillaries/mm2 was lower in the older untrained horses (350 capillaries/mm2) compared with the younger untrained ones (460 capillaries/mm2). Increase with growth was found in the activity of phosphorylase and HAD and a decrease was seen in the activity of hexokinase. It is concluded that the training programme exclusively induced alterations which improved the aerobic capacity of the muscle.  相似文献

Reasons for performing study: The trotting speed of Norwegian‐Swedish Coldblooded Trotters (NSCTs) is, although inferior to Standardbred speeds, gradually increasing. This development is mainly attributed to breeding progresses. Characteristics of the middle gluteal muscle have recently been described in a small number of these horses but the actual spread in muscle parameters within the population is unknown. Objectives: To describe the characteristics of the middle gluteal muscle and their naturally occurring variability in young NSCT horses and investigate possible association with body size, individual breeding index and early training. Methods: Biopsies of the middle gluteal muscle of 144 NSCT horses born in 2005 were collected at age 17–24 months. The horses were randomly selected by stratified sampling with gender and paternal progeny as stratification factors. The muscle samples were analysed for fibre type composition, fibre type areas, capillary supply and the activity of the key metabolic enzymes citrate synthase, 3‐OH‐acyl‐CoA‐dehydrogenase and lactate dehydrogenase. Correlation analysis including muscle parameters and body size, individual breeding index and early training was performed. Results: Large interindividual differences were identified in muscle characteristics. Male horses had higher percentages of type IIA fibres and lower percentages of type IIX fibres than age‐matched females. Fibre type areas followed the order type IIX>IIAX>IIA>I, while the capillary supply per fibre type area increased in the opposite order. No gender‐related differences were found in fibre type areas or capillary supply. Height was correlated to the relative area of type I fibres. Early training was associated with increased oxidative capacity and increased percentage of type IIA fibres. The lowest breeding index was found in untrained horses. Conclusions: Muscle characteristics differ among young NSCT horses, but the innate oxidative capacity seems to be lower than in Standardbreds of comparable age. Skeletal muscle constitutes the largest organ system in the mammalian body and is essential for movement and force generation. Muscle tissue has the unique ability to adapt and remodel with regular exercise. Adaptation of equine contractile apparatus to exercise training with a different character occurs at the structural to the cellular and molecular levels and depends on age, breed, and sex. In Andalusian and Arabian horses from 3 months to 24 years of age, it was found that the mean cross-sectional area occupied by IIA and IIX fibers was greater in stallions than in mares. In muscle of Dutch Warmblood foals from birth to 1 year of age, a significant number of fibers coexpress either developmental and type IIa or cardiac–alpha and type I myosin heavy chain (MyHC) isoforms. Endurance training results in increased mitochondrial density, capillary supply, changes in key metabolic enzymes, and increased maximal oxygen uptake and promotes a transition from type II to type I muscle fiber. In horses, prolonged aerobic exercise training has been shown to induce a further decline in the percentage of type IIx MyHC isoform expression and an increase of type I and IIa MyHC isoform expression. Short-duration, high-intensity exercise training stimulates type IIA and hybrid (IIA/IIX) fibers. Therefore, intensive high-speed trotting facilitates muscle fiber hypertrophy and increases the oxidative capacity of type IIX fibers.  相似文献

The effect of animal maturity on fiber cross-sectional area, percentage of fiber types, activities of isocitrate dehydrogenase (ICDH) and lactate dehydrogenase (LDH), total and insoluble collagen and lipid concentration was investigated in the longis-simus thoracis (LT), semitendinosus (ST), and triceps brachii (TB) muscles. The analysis considered 2,642 muscle samples from bulls, steers, and cows of Aubrac, Charolais, Limousin, Montbéliard, and Salers breeds. For the bulls, the fiber cross-sectional area, percentage of slow oxidative fibers, and ICDH activity showed a quadratic relationship (P < 0.05), and the percentage of fast oxidative-glycolytic and fast glycolytic fibers and LDH activity showed a cubic relationship (P < 0.05) with increased maturity. A linear relationship was observed for the collagen and lipid muscle characteristics. The response equation coefficients for different muscles indicate that development of muscle characteristics is different for each muscle. Compared with the other muscles, ST muscle had a greater fiber cross-sectional area, proportion of fast glycolytic fibers, LDH activity, and collagen content. The LT muscle had a greater proportion of slow-oxidative fibers and lipid (P < 0.05). Within the ST muscle, all characteristics except lipid concentration showed different development between the breeds. Steers showed greater changes in muscle fiber cross-sectional area, percentage of fast oxidative-glycolytic and fast glycolytic fibers, and total lipid in the muscle with increasing maturity compared with bulls. The mean fiber cross-sectional area and percentage of fast glycolytic fibers was greater and the mean lipid concentration was less in bulls compared with steers (P < 0.05). Data for cows were from more mature animals. Muscle characteristics in cows did not show large changes with increasing degree of maturity. Muscle type accounts for a greater proportion of the variation in the muscle characteristics than breed and sex of the animal.  相似文献

The objective of this study was to investigate the changes in muscle fiber bundles of cattle of different breeds during growth. Different numbers of muscle fibers are surrounded by connective tissue to form bundles macroscopically visible as meat fibers or meat grain, a common meat quality trait. To determine the influence of breed and age on morphological characteristics of muscle fiber bundles, 4 cattle breeds with different growth impetus and muscularity were reared and slaughtered under experimental conditions. German Angus, a typical beef cattle; Galloway, a smaller beef type; Holstein Friesian, a dairy type; and double-muscled Belgian Blue, an extreme type for muscle growth, were used. Between 5 and 15 bulls of each breed were slaughtered at 2, 4, 6, 12, or 24 mo of age, and slices of semitendinosus muscle were removed. Muscle structure characteristics were determined by computerized image analysis. During growth, the muscle cross-sectional area enlarged (P < 0.001) about 5-fold in double-muscled Belgian Blue bulls and about 4-fold in the other breeds. This was a result of the enlargement (P < 0.001) of primary bundles and muscle fibers. The bundle size was similar (P > or = 0.15) in bulls of German Angus and Galloway in all age groups and was doubled (P < 0.001) in double-muscled Belgian Blue animals from 4 mo of age on. The Holstein Friesian bulls had the smallest (P < 0.001) muscle fiber bundles at 24 mo of age. The number of muscle fibers per bundle and the number of bundles per muscle remained nearly constant (P > 0.05) during growth. This supports the existing view that the structure of the muscle is already fixed in prenatal life. The double-muscled Belgian Blue bulls showed a more than 2.5-fold greater (P < 0.001) number of muscle fibers per primary bundle compared with the other breeds investigated. The larger muscle fiber bundles led to a smaller amount of connective tissue per muscle area in double-muscled cattle. The coarser grain of meat in double-muscled Belgian Blue bulls and in older animals was not related to greater shear force values.  相似文献

旨在探究大足黑山羊妊娠早期胎儿胎盘血管发育、分布和血管生成相关因子表达状况,并对血管生成相关因子表达之间和血管发育进行相关性分析。本研究随机选取15只8月龄、体格相近、身体健康的国家级畜禽遗传资源—大足黑山羊青年母羊,经同一种公羊自然交配后(以末次配种当天记为0 d),经剖腹产手术采集妊娠第20、25和30天的胎儿和胎儿胎盘以及经屠宰采集妊娠第45和60天的胎儿和胎儿胎盘作为研究对象,通过免疫组化法评估胎盘血管发育和分布情况,采用qRT-PCR技术检测主要血管生成相关基因的mRNA表达水平,分析血管生成相关基因之间以及与胎盘血管发育分布的相关性。结果显示,胎儿体重和体长随着妊娠的进行逐渐增加,且血管内皮细胞阳性率处于较高水平(>26%)。山羊胎儿胎盘毛细血管总面积/组织区域面积(CAD)逐渐升高;毛细血管总数/单位组织区域面积(CND)在妊娠25~30 d增加,30~60 d降低,毛细血管总面积/毛细血管根数(APC)呈相反趋势;毛细血管总周长/单位组织区域面积(CSD)无显著变化。妊娠早期胎儿胎盘VEGFA的表达量随着妊娠的进行逐渐升高,且与血管分布存在一定的相关性,其受体FLT1和KDR表达量呈先增高后降低的趋势,分别在30和45 d达到峰值。血管生成相关基因ANGPT1/2及其受体TEK和FGF2及其受体FGFR2的表达均在妊娠30 d时较高。综上表明,在妊娠30 d前,胎儿胎盘主要通过毛细血管形成分支促进血管面积的增加;而在30~60 d,主要通过单位血管的增粗促进血管面积的增加,并且血管生成相关基因的表达在妊娠30 d时较高,这可能和子叶开始形成有关。山羊妊娠早期胎儿胎盘中VEGFA可能通过其受体结合与其他血管生成因子共同作用调控胎儿胎盘血管形成。  相似文献

肌纤维的分类建立在它们所含有的不同肌球蛋白重链(MHC)异构体基础上,对大鼠和家兔生后发育各年龄阶段趾浅屈肌采用标准的肌动球蛋白ATP酶和琥珀酸脱氢酶组织化学染色。在生后2周龄至24周龄的大鼠和家兔,I型和ⅡⅩ型肌纤维百分比例趋于减少,而ⅡA型和ⅡB型纤维百分比例则增加。进行大量单肌纤维的组织化学特征的比较和肌纤维间相关性探讨,结果表明,动物平均体重与趾浅屈肌的平均湿重随着生后发育逐渐增加。Ⅰ,ⅡⅩ,ⅡA型和ⅡB型纤维均在生后各年龄组的全部肌肉内被发现,但生后2日龄组是个例外。在生后发育期间,雄性大鼠和家兔ⅡB型纤维的平均肌纤维型构成要大于雌性大鼠和家兔,而雄性大鼠和家兔Ⅰ,ⅡⅩ型和ⅡA型3种氧化组织化学分类的肌纤维型构成均小于雌性大鼠和家兔。大鼠Ⅰ,ⅡⅩ,ⅡA型和ⅡB型纤维的平均横切面积明显小于家兔的同类型肌纤维,在大鼠和家兔可见明显的性别差异。在大鼠和家兔的ⅡⅩ型纤维横切面积是最小的,Ⅰ型和ⅡA型纤维呈中等大小横切面积,ⅡB型纤维横切面积最大。此研究有助于我们尝试研究啮齿类动物快肌纤维生理特征的适应性。  相似文献

研究旨在通过测定豁眼鹅、皖西白鹅、浙东白鹅肌肉纤维直径、横切面积和肌纤维的密度,探讨不同鹅品种的肌纤维特性差异.结果表明,豁眼鹅胸肌肌纤维直径、横切面积显著大于浙东白鹅、皖西白鹅(P< 0.05),而肌纤维密度表现为相反的趋势.豁眼鹅的腿肌肌纤维直径和横切面积显著大于皖西白鹅(P< 0.05),而豁眼鹅的腿肌肌纤维密度显著小于皖西白鹅(P< 0.05).品种内不同性别之间部分指标也存在显著差异,主要表现在皖西白鹅和浙东白鹅的肌纤维横切面积和直径上.相关性分析表明,肌纤维的直径与肌纤维横切面积呈极显著正相关(P< 0.01),肌纤维直径和横切面积与肌纤维密度呈极显著负相关(P< 0.01).由此可知,不同品种间肌纤维特性存在差异,同品种间不同性别间的肌纤维特性也存在差异.  相似文献

An ontogeny study of the porcine semitendinosus muscle was conducted to meet two objectives: 1) to evaluate enzyme histochemistry, lectin cytochemistry and actin staining for usefulness as quantitative markers of muscle capillaries and 2) to describe the ontogeny of capillary density changes in developing porcine muscle. Muscle samples were obtained from three to eight crossbred pigs at each of the following ages: newborn, 1 to 2 d and 1, 2, 3, 4.5 and 24 wk. Cryostat sections were stained or reacted for alkaline and neutral phosphatase, dehydrogenase, actin, a panel of nine plant lectins (fluorescein isothiocyanate conjugated) and routine cytochemical tests for muscle fiber typing. Capillaries were quantitatively marked by reactions for dehydrogenase activity in the young pigs but not in the oldest animals. The lectins, soybean agglutinin, Bauhinia purpuria agglutinin, Ulex europeus agglutinin-I and Griffonia simplicifolia agglutinin-I (GS-I) quantitatively stained capillaries at all ages. Histological observations of thin sections of epon-embedded tissues served to validate the lectin and cytochemical capillary staining. Micrographs of sections stained with the lectin GS-I were used to count capillaries and muscle fibers so the capillary:fiber ratio (C/F) could be calculated. Deep (red) and superficial (white) aspects of muscle sections had different C/F at birth, 2, 4 and 24 wk. The deep aspects (DA) had higher C/F than superficial (SA) aspects (at all four ages), and there were age-dependent increases (P less than .001) in C/F of DA and SA. This study demonstrates the usefulness of lectin staining for determining C/F in porcine muscle.  相似文献

Microvascular changes in the mammary gland in mice during pregnancy and lactation were investigated by scanning electron microscopy (SEM) with a corrosion cast method, transmission electron microscopy (TEM) and morphometry. By SEM, duct-associated capillary plexuses were sparsely distributed to branch into adipocytes during virgin period. With advance in pregnancy, both branches from the capillary plexuses and branches from the vessels surrounding adipocytes extended further to form capillary networks. The basket-like architecture was completed by day 18 of pregnancy. These findings may indicate that angiogenesis occurs frequently during this period. During lactation, the basket-like architecture still remained and the capillaries surrounding alveoli meandered. After weaning, the regression of microvasculature followed the degeneration of alveoli. By TEM and morphometry, the density of pinocytotic vesicles (PVs) (number of PVs per microns 2 of endothelium cytoplasm) increased twofold from day 18 of pregnancy to day 5 of lactation, furthermore increased threefold from days 10 to 20 of lactation, and subsequently decreased after weaning. Marginal folds and microvillous processes gradually increased in length with advance in pregnancy, reached the maximum from days 5 to 15 of lactation, and thereafter decreased. In addition, the capillaries with thinner walls were in close contact with alveoli during the late stage of pregnancy and during lactation. Furthermore, the alveolar epithelial cells had well-developed basal infoldings during lactation. These findings suggest that the capillaries play an important role in transporting materials necessary for milk production.  相似文献

OBJECTIVE: To determine whether postnatal development of oxidative capacity and capillary supply of skeletal muscle is affected by various movement regimens in horses. ANIMALS: 35 foals. PROCEDURES: Dutch Warmblood foals were allocated into 3 groups (box stall rest, box stall rest with training, and free pasture exercise). Training comprised an increasing number of gallop sprints from 1 week after birth to 22 weeks of age. From 22 to 48 weeks, the 3 groups were combined and allowed to exercise freely. Capillary supply (diffusion index [ie, area supplied by 1 capillary]), citrate synthase (CS) activity, and succinate dehydrogenase (SDH) activity were measured in biopsy specimens of deep gluteus medius muscle. RESULTS: During the first 22 weeks, diffusion index increased in all 3 groups (the training and pasture groups had a smaller increase, compared with the box stall rest group), total SDH activity increased in the training and pasture groups and decreased in the box stall rest group, and CS activity decreased in all groups. The effect of the various movement regimens on the diffusion index remained after the groups were combined. CONCLUSIONS AND CLINICAL RELEVANCE: Withholding of exercise had a negative effect on the capillary supply (ie, diffusion index increased) that remained after box stall rest was discontinued and on oxidative capacity. Box stall rest with training prevented the negative effects and eventually had the same positive effect as pasture exercise.  相似文献

Introduction and Aim:  The corpus luteum is one of the most intensely vascularized tissues. Luteal angiogenesis is strictly controlled and blood vessels regress completely within a short period of time. The aim of this study was to investigate vascular dynamics in relation to cellular and molecular mechanisms of luteal angiogenesis and anti-angiogenesis.
Material and Methods:  Endothelial cells of blood vessels in paraffin sections of bovine corpora lutea from different stages were examined by labelling with the lectin Bandeiraea simplicifolia agglutinin I. Angiogenesis was studied by morphometry of the capillaries, and immunolocalization of the angiogenic factor VEGF and VEGF-receptor 2. Presence of apoptotic luteal and endothelial cells was investigated using the TUNEL test and transmission electron microscopy.
Results:  During development of the corpus luteum (day 3–8 of the oestrous cycle) a dense capillary network (8–12% area ratio) is established and maintained until day 17. Early regression (day 18–24) is characterized by a remarkable decrease of capillaries (1% area ratio). In the regressing corpus luteum the number of apoptotic luteal cells is closely correlated ( r  = 0.9) to the number of apoptotic endothelial cells. VEGF is immunolocalized in luteal cells (day 3–17), smooth muscle cells and endothelial cells of arterioles of the regressing corpus luteum. During late luteal regression, a moderate increase of capillaries (2.5% area ratio) is obvious.
Conclusions:  The dynamic changes of the capillarity during development and regression of the cyclic corpus luteum correlate with VEGF and VEGF-R2 activities. In contrary to expectations the late stage of luteal regression is accompanied by angiogenesis. One reason for this phenomenon may be an increase in metabolic activity resulting in re-organization of blood vessels already regressed.  相似文献

The heart of 12 adult llamas, one female and 11 males, 2-7 years of age, was studied gross-anatomically (absolute and relative heart weight) and microscopic-anatomically (left and right ventricular wall, left auricle). Comparing the 9 younger animals (2-3 years old) with the 3 older llamas (5-7 years old) a significant increase of the absolute heart weight and of the number of capillaries per mm2 and a significant decrease of the intercapillary distance in the left ventricular wall was found. This results in an increase of capillarity in the studied heart regions (5.5% compared to 6.9%). The simultaneous increase of the capillary-to-myocyte-ratios from 0.9 to 1.1 shows a significant correlation to the cross-sectional area of the myocytes (r = 0.79) and an also significant but negative correlation to the number of myocytes per mm2 (r = -0.82). In both groups the cross-sectional area of the myocytes and their diameter differs significantly in the three regions of the heart (left ventricular wall--214 micron 2 resp. 17.5 microns; right ventricular wall--156 micron 2 resp. 15.5 microns; left auricle--96 micron 2 resp. 11.5 microns). It is to be supposed, that the morphological conditions of the cardiac vascular supply of llamas living in an altitude of 4000 m are improving with age.  相似文献

The objective of this study was to investigate the growth- and breed-related changes of muscle fiber characteristics in cattle and their importance to meat quality. Four cattle breeds with different growth impetus and muscularity were reared and slaughtered under experimental conditions. German Angus as a beef type, Galloway as a hardy type, Holstein Friesian as a dairy type, and double-muscled Belgian Blue as an extreme type for muscle growth were used. Between 5 and 17 bulls of each breed were slaughtered at 0, 2, 4, 6, 12, 18, and 24 mo of age. Muscle fiber traits were determined and classified by computerized image analysis, and several measures of meat quality were also determined, including shear force value, meat color, and i.m. fat content. The postnatal growth of semitendinosus muscle in cattle was characterized by a nearly 10-fold increase of muscle fiber area from birth to 24 mo of age. In the first few months after birth, a transformation of type IIA fibers into IIB fibers was found, whereas type I fibers were nearly unaffected by age. The apparent total muscle fiber number of semitendinosus muscle did not increase during postnatal life. These results confirm that the fiber number is determined in embryonic development. Throughout the study, the double-muscled Belgian Blue (BBDM) bulls had almost twice the fiber number of the other breeds, emphasizing a more extensive hyperplasia of muscle fibers during embryonic development in BBDM compared with the other three breeds. The apparent number of type I fibers was, however, not affected by breed, which suggests that the additional fibers found in BBDM postnatally were type IIB and IIA fibers. We did not find significant differences in muscle fiber total number, muscle fiber type frequencies, or meat quality characteristics among breeds, with the exception of BBDM. Having pooled the four breeds, paler meat was related to a higher frequency of type IIB fibers, a lower area of type IIA and type I fiber, and a higher total muscle fiber number. These findings based on data of double muscling give us some hints for biological causes for the variation of meat quality. Further investigation, in particular within each breed, is necessary to identify the superior fiber traits for bovine meat production.  相似文献

OBJECTIVE: To determine histochemical and biochemical properties of muscle during adaptation to training on a flat or sloped track. ANIMALS: 22 Thoroughbreds. PROCEDURE: Samples were obtained from the middle gluteus muscle before and after training programs were conducted, using a needle-biopsy technique. Training programs consisted of horses running 1,600 m on a flat or sloped track for 16 weeks. Amplitude of middle gluteus muscle activity per burst was calculated. Muscle fiber composition and area were examined on serial cross sections processed by standard histochemical staining procedures (ATPase stain after prior incubation with an acid or base, followed by succinate dehydrogenase [SDH] stain). Furthermore, SDH and phosphofructokinase activities were determined biochemically, and composition of myosin heavy chain (MHC) isoforms was analyzed electrophoretically. RESULTS: Training resulted in substantial adaptations. Relative contribution of muscle fiber with high SDH activity (type-I and -IIa fibers) to total cross-sectional area, SDH activity, and composition of MHC-IIa isoforms were increased. Gel electrophoresis revealed a large amount of MHC-IIx isoform and a small amount of MHC-IIb isoform in the muscle. Although root mean square of muscle activity for training on a sloped track was 7.6% higher than the value obtained while training on a flat track, muscle histochemical and biochemical properties did not differ significantly between groups training on flat and sloped tracks. CONCLUSIONS: Training adaptations for contractile and metabolic properties of the middle gluteus muscle were evident for the 2 types of training. Carcass, muscle and meat characteristics of lean and obese pigs   总被引:1,自引:0,他引:1  
Six pigs obtained from a lean selected strain and six pigs obtained from an obese selected strain were slaughtered at about 110 kg live-animal weight. Carcasses were evaluated; hams were dissected into bone, skin, fat and lean, and loin samples were obtained for fiber type characteristics, percentage of fat and moisture, collagen analysis, sensory characteristics, textural properties and objective color analysis. Carcasses from lean pigs were longer, had less backfat and larger longissimus muscle cross-sectional areas than carcasses obtained from obese pigs. Hams from lean pigs had less fat, more bone and more lean than hams from carcasses of obese pigs. The percentages and cross-sectional areas of red and white muscle fibers of the longissimus muscle from lean and obese pigs were not different. However, lean pigs had intermediate fibers that were only 79% as large (P less than .10) as intermediate muscle fibers from obese pigs. Intermediate fibers represented only 7 and 10% of total fiber area, whereas white fibers represented 84 and 79% of total fiber area in longissimus muscle of lean and obese pigs, respectively. Overall, lean pigs tended to possess fewer fibers (-16%) per unit of area than obese pigs, indicating that total muscle fiber hypertrophy was partially responsible for the increased longissimus muscle area of the lean strain. Sensory properties of longissimus meat samples from lean and obese strains were not different. However, the shear force requirement of the longissimus samples from the lean strain were slightly, but significantly (P less than .10), higher than those from the obese strain. No differences were observed in meat color.  相似文献

Biopsy samples were obtained from the middle gluteal muscle of 10 Thoroughbred horses undergoing a commercial race-training program. Samples were obtained before the program began and again after 6 and 12 weeks of training. All horses had raced at least once by the 12th week of training. Serial sections of muscle were examined histochemically for myosin adenosinetriphosphatase after either acid (pH 4.3 and 4.6) or alkaline (pH 10.3) preincubation, and then muscle fibers were identified as types I, IIA, IIB, or IIC. The oxidative capacity of individual fibers was assessed, using the reduced nicotinamide dinucleotide tetrazolium-reductase stain, and the number of intermyofibrillar capillaries adjacent to each fiber were counted after staining, using the alpha-amylase-periodic acid-Schiff technique. Biochemical analyses involved the fluorometric measurement of 3 enzymes--citrate synthase, 3-hydroxy-acyl coenzyme A dehydrogenase, and lactate dehydrogenase--as markers of end terminal oxidative, beta-oxidative, and glycolytic potentials, respectively. Changes in fiber-type percentages did not occur in response to training. There was a significant (P less than 0.01) increase in the percentage of type IIB fibers, having high nicotinamide dinucleotide-tetrazolium reductase staining after 12 weeks of training. Alterations in the number of capillaries adjacent to each fiber type did not occur during the training period. There were increases in the activities of both citrate synthase and 3-hydroxy-acyl coenzyme A dehydrogenase after 6 weeks (P less than 0.05) and 12 weeks (P less than 0.001) of training. Alterations in the activity of lactate dehydrogenase did not occur in response to training.  相似文献
