Risks associated with mycotoxin contamination of cereals, that are included in the ten major staple foods and greatly contribute to the dietary energy intake, are of worldwide relevance. In small grain cereals, mycotoxins are produced by fungi such as Aspergillus, Penicillium, Alternaria and Fusarium that colonize the plant in the field and can grow during the post-harvest period, producing several classes of mycotoxins. The identification of mycotoxigenic fungal species and strains is essential for developing effective strategies for control. For this purpose, genetic traceability has proved to be a valuable tool that can be applied along the whole production chain, starting in the field for early diagnosis of FHB (Fusarium Head Blight) disease to the final processing steps, such as malting or pasta making. In this paper, DNA-based analytical tools that are currently available for the identification and quantification of mycotoxigenic fungal species and strains are reviewed, with particular emphasis on Fusarium, and their possible applications in mycotoxin control in small grain cereal chains are discussed.  相似文献

The genus Aspergillus represents a diverse group of fungi that are among the most abundant fungi in the world. Germination of a spore can lead to a vegetative mycelium that colonizes a substrate. The hyphae within the mycelium are highly heterogeneous with respect to gene expression, growth, and secretion. Aspergilli can reproduce both asexually and sexually. To this end, conidiophores and ascocarps are produced that form conidia and ascospores, respectively. Marine fungi represent a huge potential for new natural products and an increased number of new metabolites have become known over the past years, while much of the hidden potential still needs to be uncovered. Representative examples of biodiversity studies of marine fungi and of natural products from a diverse selection of marine fungi from the author’s lab are highlighting important aspects of this research. If one considers the huge phylogenetic diversity of marine fungi and their almost ubiquitous distribution, and realizes that most of the published work on secondary metabolites of marine fungi has focused on just a few genera, strictly speaking Penicillium, Aspergillus and maybe also Fusarium and Cladosporium, the diversity of marine fungi is not adequately represented in investigations on their secondary metabolites and the less studied species deserve special attention. In addition to results on recently discovered new secondary metabolites of Penicillium species, the diversity of fungi in selected marine habitats is highlighted and examples of groups of secondary metabolites produced by representatives of a variety of different genera and their bioactivities are presented. Special focus is given to the production of groups of derivatives of metabolites by the fungi and to significant differences in biological activities due to small structural changes.  相似文献

Aspergillus section Flavi is able to produce aflatoxins (AFs) in vitro down to 0.85 aw with a potential maximum occurring between 0.95 and 0.99 aw, while in the field AFs increased significantly with kernel humidity below 0.95 aw. In order to clarify this apparent discrepancy, a 3-year field trial with artificial inoculation of maize ears with Aspergillus flavus strains was organised. The co-occurrence of Aspergillus section Flavi and Gibberella fujikuroi species complex (Gfsc) was observed. The incidence of A. flavus was significantly influenced by the year and negatively related to Gfsc incidence. In 2012, when the highest temperature and the lowest rain were registered, aflatoxin B1 (AFB1) content was the highest and aw < 0.95 in kernels was measured early throughout the growing season. In 2013 and 2014, the temperature was lower and rain more abundant, and aw decreased below 0.95 only close to harvest and AFB1 contamination was limited. The possibility of describing/predicting reasonably well aw dynamic based on temperature, (degree day) was confirmed. With aw > 0.95, a positive correlation between AFB1 production rate and aw was found, but a negative correlation resulted with aw < 0.95. Other factors than aw play a role, but aw = 0.95 should be considered as an indicator of suitable conditions for rapid aflatoxin accumulation.  相似文献

Biodiversity of the marine world is only partially subjected to detailed scientific scrutiny in comparison to terrestrial life. Life in the marine world depends heavily on marine fungi scavenging the oceans of lifeless plants and animals and entering them into the nutrient cycle by. Approximately 150 to 200 new compounds, including alkaloids, sesquiterpenes, polyketides, and aromatic compounds, are identified from marine fungi annually. In recent years, numerous investigations demonstrated the tremendous potential of marine fungi as a promising source to develop new antivirals against different important viruses, including herpes simplex viruses, the human immunodeficiency virus, and the influenza virus. Various genera of marine fungi such as Aspergillus, Penicillium, Cladosporium, and Fusarium were subjected to compound isolation and antiviral studies, which led to an illustration of the strong antiviral activity of a variety of marine fungi-derived compounds. The present review strives to summarize all available knowledge on active compounds isolated from marine fungi with antiviral activity.  相似文献

Groundnut (Arachis hypogaea L.) is susceptible to pre- and post-harvest infections by Aspergillus spp. Aflatoxin B1 (AFB1), is the contaminant produced by the fungus in infected grains posing a threat to human and animal health. This paper reports of a study undertaken in Malawi to determine the occurrence and distribution of Aflatoxigenic Aspergilli in the soil and AFB1 contamination in groundnuts. A total of 1397 groundnut samples collected from farm homesteads, local markets, warehouses and shops in 2008 and 2009 were analyzed for AFB1 contamination using the enzyme linked immunosorbent assay (ELISA), and A. Aspergilli population densities in 1053 soil samples collected from the same sites were estimated using serial dilutions plated on A. Aspergilli medium. Farmer socio-economic profile information was also collected to determine relationships to AFB1 contamination. The results revealed 46% and 23% of the total samples, from 2008 to 2009, respectively, had AFB1 contamination levels greater than 4 ppb, and those above 20 ppb were 21% for 2008 and 8% for 2009, respectively. Fitted smooth curve relationships show that there is a clear increase in the chance of groundnut contamination when the population density of A. Aspergilli in the soil increased beyond 3000 (log (cfu) > 8). The measured level of A. Aspergilli in soil varied by location, as well as ecologies within location. Low-altitude ecologies, which were warmer and experienced low precipitation levels, had the highest densities of A. Aspergilli, whereas cooler high-altitude ecologies had the lowest density of these fungi. Similarly high AFB1 contamination, was recorded across the country with 11–28% of all samples collected from the warm low to mid-altitude ecologies recording contamination ≥20 ppb and low contamination (2–10% of samples) in the mid to high altitude cool ecologies. From a crop management perspective, this study also suggests that both less experienced and older farmers were more likely to produce groundnuts contaminated with aflatoxin. These findings have implications in the design of intervention strategies to avoid short- and long-term human health effects from aflatoxin exposure.  相似文献

为探明西藏麦类作物中真菌毒素污染程度及产毒真菌种类与分布,以青稞、小麦为研究对象,对其籽粒中多种毒素进行分析,并对毒素污染阳性样品进行真菌分离鉴定。结果表明,2018年毒素总检出率为20.13%;杂色曲霉毒素检出率最高,为10.74%;OTA超标率最高,为3.36%;小麦的毒素检出率和超标率都高于青稞,二者的污染毒素种类以OTA、ZEN、ST为主。西藏高原42份被真菌毒素污染的农作物籽粒中共分离到199份真菌,分属23属67种真菌,青霉属为优势属,曲霉属次之。  相似文献

Filamentous fungi are widely used for enzyme production for the biofuel industry. The ascomycetous fungus Chrysosporium lucknowense C1 was isolated as a natural producer of neutral cellulases. It is at present an attractive alternative to well known fungi like Aspergillus sp. and Trichoderma reesei for protein production on a commercial scale. Besides many cellulases, a large number of hemicellulases (particularly xylanases and arabinofuranosidases) and esterases (acetyl xylan esterases and ferulic acid esterases) encoding genes have also been identified in the C1 genome. Many of these extracellular enzymes have been selectively expressed in C1 and then purified and characterized. Four arabinofuranosidases, two acetyl xylan esterases, two ferulic acid esterases, an α-glucuronidase and four xylanases have been purified and characterized. All these enzymes were found to be active towards arabinoxylans, demonstrating the high potential of C1 as a producer of hemicellulolytic enzymes.  相似文献

In the present study, a total of 380 samples of 5 rice varieties were investigated for the presence of aflatoxins (AFs) and tocopherols (Vitamin E) using HPLC, equipped with a fluorescence detector. The data revealed that super basmati rice has the highest mean level of total AFs (12.45 μg/kg) followed by KS-282 (11.2 μg/kg), basmati PK-385 (9.8 μg/kg), Irri-6 (9.7 μg/kg) and the lowest mean level was found in Irri-9 (8.9 μg/kg). Furthermore, the results have shown that the variety with lowest level of AFs has the highest level of tocopherol content; i.e. Irri-9 (53.2 mg/100 g), basmati PK-385 (45.9 mg/100 g), Irri-6 (45.3 mg/100 g), KS-282 (40.4 mg/100 g) and super basmati rice (40.2 mg/100 g). The data has shown correlation (r = −0.62, p < 0.05) between vitamin E content and AFs concentration in different rice varieties. The results are interesting and need further study to investigate the mechanistic background of vitamin E content and its effect on aflatoxins contamination.  相似文献

Colorectal cancer is one of the most common cancers diagnosed in the world. Chemotheraphy is one of the most common methods used for the pharmacological treatment of this cancer patients. Nevertheless, the adverse effect of chemotherapy is not optimized for improving the quality of life of people who are older, who are the most vulnerable subpopulation. This review presents recent updates regarding secondary metabolites derived from marine fungi and actinobacteria as novel alternatives for cytotoxic agents against colorectal cancer cell lines HCT116, HT29, HCT15, RKO, Caco-2, and SW480. The observed marine-derived fungi were from the species Aspergillus sp., Penicillium sp., Neosartorya sp., Dichotomomyces sp., Paradendryphiella sp., and Westerdykella sp. Additionally, Streptomyces sp. and Nocardiopsis sp. are actinobacteria discussed in this study. Seventy one compounds reviewed in this study were grouped on the basis of their chemical structures. Indole alkaloids and diketopiperazines made up most compounds with higher potencies when compared with other groups. The potency of indole alkaloids and diketopiperazines was most probably due to halogen-based functional groups and sulfide groups, respectively.  相似文献

To identify the susceptibility of long-horned grasshoppers to entomopathogenic fungi, the effect of infection with the fungi Beauveria bassiana (Balsamo) Vuillemin (Hypocreales: Clavicipitaceae) and Metarhizium anisopliae (Metchnikoff) Sorokin (Hypocreales: Clavicipitaceae) on food consumption by Uvarovistia zebra (Uvarov) (Orthoptera: Tettigoniidae) was investigated. Preliminary results showed that both fungi had a negative effect on food consumption of the insects. For both fungi a significant reduction of food consumption and faeces production by insects were observed between the highest spore concentration (5 × 106 spores/ml) and other treatments. Compared with control insects, the insects treated with 5 × 106 spores/ml of B. bassiana and M. anisopliae showed 60 and 63% reduction in mean food consumption/insect, respectively. The corrected cumulative percent mortality of the insects treated with the highest concentration of B. bassiana and M. anisopliae were 57.7 and 55.5%, respectively. This was the first account of these entomopathogenic fungi being used against a species from this family, therefore based on the results obtained from this research, it could be said that the fungi have pathogenicity effect on U. zebra as a long-horned grasshopper.  相似文献

Fungal growth is a major problem of food storage in humid environments, as occur in South Pacific countries for parts of the year. Major crops, including edible nuts, copra and root crops, are susceptible toAspergillus growth and therefore potential contamination with aflatoxin. Liver cancer occurs in Fiji and Tonga, with the occurrence in Fijians being significantly higher than in the Indian population.Thirty-three peanut samples from farmers were analysed for aflatoxin and 50% of the samples from Fiji were positive but only 9% from Tonga, reflecting different storage practices. Local copra, cassava, and maize samples were found contaminated, with only the maize at a serious level. Twenty-five plate food samples from Fiji showed low contamination. When starch foods from the Fijian diet left after cooking were analysed to follow potential aflatoxin development only sweet potatoes showed some contamination.  相似文献

Aflatoxin is a strong carcinogenic and toxic fungal toxin produced by Aspergillus flavus and other Aspergillus species,and can seriously threaten the health of consumers,thus becoming a global concern.Corn,as an important oil and economic crop,is highly susceptible contaminated by aflatoxin.In this study,antagonistic bacteria with strong inhibitory effect on aflatoxin were screened to provide support for the treatment aflatoxin contamination control in corn.Ten strains which have strong antagonistic effects against A.flavus were isolated from healthy corn from different corn producing areas in China.Among them,the antagonistic bacteria JTFM1001 through corn kernels in vivo and field experiment,the inhibition effect of aflatoxin contamination reached above 70%and 55%,respectively.And the strain was identified as Bacillus subtilis based on its morphological,physiological and biochemical characteristics and phylogenetic analysis of 16S rDNA.In addition,our data showed that it can colonize in the rhizosphere and survive for a long time,forming the dominant flora,with broad application prospect.Finally,we were surprised to find that the antibacterial metabolites secreted by the antagonistic bacteria was one of the mechanisms of its inhibition of A.flavus and aflatoxin.This will provide us with new ideas and perspectives on the effective prevention and control of aflatoxin contamination in corn and corn oil.  相似文献

The deep sea has been proven to be a great treasure for structurally unique and biologically active natural products in the last two decades. Cold seeps and hydrothermal vents, as typical representatives of deep-sea extreme environments, have attracted more and more attention. This review mainly summarizes the natural products of marine animals, marine fungi, and marine bacteria derived from deep-sea cold seeps and hydrothermal vents as well as their biological activities. In general, there were 182 compounds reported, citing 132 references and covering the literature from the first report in 1984 up to March 2022. The sources of the compounds are represented by the genera Aspergillus sp., Penicillium sp., Streptomyces sp., and so on. It is worth mentioning that 90 of the 182 compounds are new and that almost 60% of the reported structures exhibited diverse bioactivities, which became attractive targets for relevant organic synthetic and biosynthetic studies.  相似文献

Marine-derived fungi constitute an interesting source of bioactive compounds, several of which exhibit antibacterial activity. These acquire special importance, considering that antimicrobial resistance is becoming more widespread. The overexpression of efflux pumps, capable of expelling antimicrobials out of bacterial cells, is one of the most worrisome mechanisms. There has been an ongoing effort to find not only new antimicrobials, but also compounds that can block resistance mechanisms which can be used in combination with approved antimicrobial drugs. In this work, a library of nineteen marine natural products, isolated from marine-derived fungi of the genera Neosartorya and Aspergillus, was evaluated for their potential as bacterial efflux pump inhibitors as well as the antimicrobial-related mechanisms, such as inhibition of biofilm formation and quorum-sensing. Docking studies were performed to predict their efflux pump action. These compounds were also tested for their cytotoxicity in mouse fibroblast cell line NIH/3T3. The results obtained suggest that the marine-derived fungal metabolites are a promising source of compounds with potential to revert antimicrobial resistance and serve as an inspiration for the synthesis of new antimicrobial drugs.  相似文献

Artichoke seeds suffer attacks of various fungi that result in a decreased yield. Coatings based on chitosan were used as an antifungal agent and for enhancing the germination and quality of artichoke seeds. The effect of formulation (chitosan molecular weight, presence or absence of Span 80, pH), and thickness (number of coating layers) on seed germination (G%), fungi activity and vegetative growth were studied. Results indicated significant differences between treatments on seed germination. It was observed that all chitosan treatments reduced the number of type of fungi and increased plant growth. Chitosan with lower Mw gave better results both from microbial and germination point of view. In addition results indicated that treatments including chitosan and TMTD increased significantly G% and plant growth of artichoke seeds and produced a decrease in fungi contamination.  相似文献

Aflatoxins, produced by the fungi Aspergillus flavus Link ex Fries and Aspergillus parasiticus Speare, are the major toxins affecting the quality of groundnuts (Arachis hypogaea L.) meant for human consumption. Groundnuts can be infected with aflatoxin-producing fungi pre-harvest, at harvest and post-harvest. This survey was conducted in Gujarat province in India in order to assess farmers’ and other stakeholders’ (extension staff and traders) perceptions and knowledge of aflatoxin contamination of groundnuts and to evaluate the agronomic and market practices used to manage it. The survey investigated the effects of the socioeconomic background of the farmers. The results showed that the socioeconomic and psychological characteristics, viz. education, caste, farm size, social participation, extension participation, market orientation, economic motivation, innovativeness and perception had positive and significant associations with farmers’ knowledge. The extension staff and traders had a good understanding of the problem and of the importance of managing aflatoxin contamination but farmers did not. Farmers’ who practised effective crop husbandry in order to increase production were unwittingly managing aflatoxin contamination to some extent. Their marketing practises showed that the problem of aflatoxin contamination was neglected at both the production and marketing stages. We suggest that extension agencies need to train farmers in the use of biological control agents, post-harvest management and identification of aflatoxin contamination. Partnerships need to be forged between research institutions, the departments of agriculture of various states, marketing agencies, NGOs, farmers’ groups, consumer groups, agrochemical manufacturers and other stakeholders in order to develop strategies for addressing the problem of aflatoxin contamination.  相似文献

Arsenic contamination of shallow groundwater and related health problems are threats for the millions in endemic regions of West Bengal. Contamination of rice grain creates the food chain pathway of mineral arsenic besides drinking water contamination. Present study concentrated on association of arsenic concentration in irrigated water, paddy field soil and rice with the cropping seasons. Irrigated ground water arsenic concentration decreased significantly (p = 0.007) from summer (median 0.42 mg l?1) to winter (median 0.35 mg l?1). Carried over effect created significant decrease (p = 0.03) of paddy field soil arsenic concentration from summer (median 8.35 mg kg?1) to winter (median 6.17 mg kg?1). Seasonal variation was observed in rice straw (p = 0.03) but not in husk (p = 0.91). Arsenic concentration decreased significantly (p = 0.05) in the rice grains collected in winter season (median 0.23 mg kg?1) than the samples collected in the summer season (median 0.30 mg kg?1). In conclusion, seasonal effects need to be considered in case of human health risk assessment from arsenic consumption.  相似文献

Nowadays, the logarithmic production of existing well-known food materials is unable to keep up with the demand caused by the exponential growth of the human population in terms of the equality of access to food materials. Famous local food materials with treasury properties such as mangrove fruits are an excellent source to be listed as emerging food candidates with ethnomedicinal properties. Thus, this study reviews the nutrition content of several edible mangrove fruits and the innovation to improve the fruit into a highly economic food product. Within the mangrove fruit, the levels of primary metabolites such as carbohydrates, protein, and fat are acceptable for daily intake. The mangrove fruits, seeds, and endophytic fungi are rich in phenolic compounds, limonoids, and their derivatives as the compounds present a multitude of bioactivities such as antimicrobial, anticancer, and antioxidant. In the intermediary process, the flour of mangrove fruit stands as a supplementation for the existing flour with antidiabetic or antioxidant properties. The mangrove fruit is successfully transformed into many processed food products. However, limited fruits from species such as Bruguiera gymnorrhiza, Rhizophora mucronata, Sonneratia caseolaris, and Avicennia marina are commonly upgraded into traditional food, though many more species demonstrate ethnomedicinal properties. In the Middle East, A. marina is the dominant species, and the study of the phytochemicals and fruit development is limited. Therefore, studies on the development of mangrove fruits to functional for other mangrove species are demanding. The locally accepted mangrove fruit is coveted as an alternate food material to support the sustainable development goal of eliminating world hunger in sustainable ways.  相似文献
