Five colostrum deprived hysterectomy-derived piglets were inoculated by the intratracheal route with a Porcine Respiratory Coronavirus (PRCV). The clinical effects and growth performances were compared with those in 5 similar piglets which were inoculated with Minimum Eagle Medium only. A transient hyperthermia, mild to severe dyspnoea, polypnoea and an obvious cessation of growth were recorded on infected pigs only whereas the control group did not show any adverse reaction after inoculation. The reasons for the variability of effects of experimental infection observed in previous trials, are discussed.  相似文献

An experiment was conducted to determine the effects of yeast culture (YC) and modified yeast culture [YC + cell wall product (CWP) containing mannan oligosaccharides] in pig diets on the performance, gut integrity, and blood cell composition of weanling pigs and to determine whether these dietary supplements could replace antimicrobial growth promoters (AGP) in pig diets. A total of 480 weanling pigs (27 d old and 7.8 +/- 0.1 kg of BW) were assigned to 1 of 4 experimental treatments: 1) diets without AGP or YC (control diet); 2) control + AGP; 3) control + 0.125% YC; and 4) control + 0.125% YC + 0.2% CWP. Piglets were fed experimental diets for 5 wk after weaning. Blood samples were collected from 8 piglets at weaning and from 8 piglets per treatment on d 14 and 35 after weaning for blood cell composition. These piglets were slaughtered for measurement of villous length and crypt depth in the jejunal mucosa and microbial profiling on the intestinal digesta. Average daily gain (P = 0.06) and G:F (P = 0.02) were improved for piglets that were fed the supplemented diets compared with piglets that were fed the control diet. Average daily feed intake was unaffected by dietary treatment. Performance was similar in piglets fed diets supplemented with AGP, YC, and YC + CWP. Blood cell composition, villous length, crypt depth, and microbial composition in the gut were unaffected by dietary treatment, but they were affected by time after weaning. Red blood cells, hemoglobin, hematocrit value, mean cell volume, mean cell hemoglobin, percentage of lymphocytes in the leukocyte population, villous length, and crypt depth were greater (P < 0.05) at 5 wk after weaning than at 2 wk after weaning. Eosinophils (P = 0.06) in the leukocyte population tended to be greater at 5 wk after weaning. Concentration of neutrophils in the leukocyte population and percentages of CD4 and CD8 cells were lower (P < 0.02) at 5 wk after weaning. The CD4:CD8 ratio (P = 0.07) tended to be lower at 5 wk after weaning. Results suggest that yeast culture could be an alternative to AGP in the diets of weanling pigs and that addition of CWP to diets containing YC would not improve the performance or health of weanling pigs above that of YC alone. Thus, more insight into the mode of action of YC is needed.  相似文献

Haemophilus parasuis and Streptococcus suis are both major causes of losses during the nursery period, especially in herds using the segregated early weaning system. In this system, only a few piglets may be colonized with the herd's prevalent systemic strain, which results in infection of naive penmates late in the nursery. In view of these factors, the objectives of this study were: (1) to evaluate the early colonization of piglets with the farm's prevalent systemic strain of H. parasuis and S. suis as an alternative method for disease prevention; and (2) to evaluate 2 different protocols for experimental colonization: direct colonization of piglets and colonization of piglets through nose-to-nose contact with inoculated sows. Haemophilus parasuis and S. suis isolates recovered from diseased nursery pigs were characterized by the rep-PCR technique and the herd's prevalent strains were used for colonization. Piglets in the experimentally colonized groups were inoculated at 5 days of age by the oral route using a spray pump. Sows were colonized at 2 weeks prior to farrowing using a similar protocol. Although both colonization protocols were successful in getting the piglets colonized, direct inoculation of 5-day-old piglets with the herd's systemic strains of H. parasuis and S. suis tended to be more effective in reducing the morbidity and the mortality than the colonization of piglets by nose-to-nose contact with inoculated sows.  相似文献

Experimental airborne transmission of Streptococcus suis type 2 was studied in specific pathogen free piglets. Forty piglets were allotted to five groups of eight 7-week-old animals and housed in three separated units. Negative control pigs (group 1) were housed in unit A and infected batches were housed in units B (group 2) and C (groups 4). In units B and C, non-inoculated groups (groups 3 and 5, respectively), 40 cm distant from the respective inoculated group and without any physical contact between them, also took place. Six animals of groups 2 and 4 were inoculated intravenously with 2 x 10(8) colony forming units (cfu) of a mild and a high virulent S. suis strains, respectively. The remaining animals in these groups and pigs from groups 1, 3, 5 received broth medium in the same way. Differences among virulence of S. suis capsular type 2 were observed in inoculated pigs of groups 2 and 4. Pigs from group 2 became carriers, showing only mild symptoms. By contrast, animals from group 4 presented an acute form of the disease. All the indirect contact pigs in groups 3 and 5 had S. suis in palatine tonsils from day 6 after the infection and they presented clinical manifestations similar to those observed in experimentally infected pigs. Two direct contact animals were also contaminated in the upper respiratory tract but surprisingly they did not show any symptoms. Airborne transmission of S. suis in experimentally pigs was demonstrated in the present study. Indirect infections, as described in this study, are a more realistic way to infect pigs than other experimental procedures and may be used to further study the pathogenesis of the infection caused by this important pathogen.  相似文献

The pathogenicity and immunogenicity of vaccinia virus were examined in order to evaluate the possibility of its application as a recombinant viral vaccine in pigs. Following scarification inoculation with vaccinia virus, a mild reddish papulation developed only on the scarified part of the skin. No symptoms of illness such as fever or stunting were noted. Vaccinia virus was recovered in titers from scarified skin 4 and 7 days after inoculation. Control piglets cohabited with inoculated animals remained normal for the whole 5 week observation period. Hemagglutination inhibition and indirect immunofluorescence tests detected antibodies against vaccinia virus in the inoculated piglets, whereas no anti-vaccinia virus antibodies were detected in the contact control animals. Antigen-induced blastogenic tests of peripheral blood lymphocytes from animals, revealed that lymphocytes obtained from inoculated donors 5 weeks after inoculation, had a higher stimulation index (P less than 0.05) than did those from uninoculated piglets. These results suggested that vaccinia virus would be useful as a recombinant viral vector for pigs.  相似文献

Epidermal growth factor (EGF) promotes gastrointestinal mucosal recovery by stimulating the mitogenic activity of intestinal crypt epithelial cells. The aim of this study was to determine the effects of EGF on atrophic enteritis induced in piglets by experimental infection with porcine epidemic diarrhoea virus (PEDV) strain Dr13. Two groups of 12 conventional, colostrum-deprived, 1-day-old, large White-Duroc cross breed piglets were inoculated orally with PEDV (3 x 10(5) 50% tissue culture infective doses), with or without EGF (10 microg/kg/day, intraperitoneally once daily for 4 days after infection) and compared to 12 uninfected, untreated control piglets. PEDV+EGF piglets had less severe clinical signs than PEDV only piglets at 48 and 60 h post-infection (hpi). Histologically, the ratio of villous height:crypt depth of PEDV+EGF piglets was significantly higher than PEDV only piglets at 36 and 48 hpi. Immunohistochemistry for Ki67 demonstrated increased proliferation in intestinal crypt epithelial cells of PEDV+EGF piglets compared to PEDV only piglets at 36, 48 and 60 hpi. EGF stimulates proliferation of intestinal crypt epithelial cells and promotes recovery from atrophic enteritis in PEDV-infected piglets.  相似文献

猪圆环病毒2型(PCV2)是引起PMWS的必需病原,与其他病原混合感染或受到外界刺激时表现PMWS临床症状。本试验将16头商品化断奶仔猪(32日龄)随机分为4组(每组4头),即对照组、钥匙孔血蓝蛋白刺激组、PCV2攻毒组和PCV2攻毒后钥匙孔血蓝蛋白刺激组,用上海某猪场分离株PCV2-SH、钥匙孔血蓝蛋白(KLH)反复刺激断奶仔猪以复制PMWS。其中对照组、钥匙孔血蓝蛋白刺激组未出现症状;PCV2攻毒组症状较轻,出现增重缓慢,伴有短暂的体温升高;而PCV2攻毒后钥匙孔血蓝蛋白刺激组出现明显的临床症状,2头猪在攻毒后第11天濒临死亡,其中1头在第12天死亡,攻毒猪宰杀后脏器出现明显的病理变化。PCV2攻毒组和PCV2攻毒后钥匙孔血蓝蛋白刺激组于攻毒后第4天出现病毒血症,宰杀后肺脏、淋巴结均检测到PCV2的DNA。以上结果说明,PCV2-SH感染结合免疫刺激可以引起商品化仔猪发生PMWS,为PCV2致病机理和免疫学研究提供了动物发病模型。  相似文献

Two types of transmission experiments were performed to estimate the basic reproduction ratio R(0), indicating the level of encephalomyocarditis virus (EMCV) transmission among pigs. In a first experimental set-up with nine separate pairs, one randomly chosen piglet per pair was inoculated with a Belgian (myocardial) EMCV strain (B279/95, 10(3)TCID(50)/ml oronasally) and placed back into the pen. In the second experiment with two separate groups of five piglets, two piglets in each group were inoculated at the start. During the experiments, viraemia in blood and excretions was measured as well as the serological response against EMCV antigen. After death or euthanasia, the piglets were checked for heart lesions and virus isolation was done on various tissues. In both the experiments, the majority of the inoculated piglets either died with typical heart lesions (five out of nine and three out of four resp.), or produced high levels of neutralising antibody. EMC virus was isolated from the hearts of all piglets that died during either one of the experiments. The pairwise experiment revealed a point estimate for R(0) of 2.0 (95% confidence interval (CI)=0.37-10.74), while the group experiment resulted in a R(0)-value of 0.71 (95% CI=0.08-4.93). Combining the information from both experiments results in an estimate for R(0) of 1.24 (95% CI=0.39-4.35). Since R(0) has values around the threshold value of 1, the spread of EMCV due to contacts between pigs will in most cases be limited, but due to chance processes may lead to large outbreaks as well.  相似文献

Due to almost identical lipopolysaccharide (LPS) O-antigens, infections with Yersinia enterocolitica serotype O:9 (YeO:9) cause false positive serological reactions (FPSR) in tests for Brucella and thus cause problems in National Brucella surveillance programs. As LPS are strong inducers of antibody responses it was hypothesized that cell-mediated immune responses to non-LPS antigens of the two bacteria can be used to separate immune responses to these two biologically very different infections. Following subclinical experimental infections with Brucella suis biovar 2, high interferon-gamma (IFN-gamma) assay responses with a commercial Brucella melitensis antigen preparation (Brucellergene OCB) preceded the development of antibodies. High IFN-gamma responses in the seven B. suis inoculated pigs with serological evidence of infection were consistent throughout a 20-week post-inoculation observation period. In contrast, IFN-gamma responses in two B. suis inoculated pigs without bacteriological or serological evidence of infection were below a cut-point of 25pg/ml at all samplings. IFN-gamma responses in repeated samplings from 5 uninfected control pigs and 18 pigs experimentally infected with YeO:9 were all negative, except for solitary false positives in 3.7% of the samples from both the experimentally YeO:9 infected pigs and control pigs. Skin tests using the same commercial Brucella antigen confirmed the ability of cell-mediated immune responses to differentiate between the two infections. In addition, a field evaluation of the diagnostic use of cell-mediated immune responses by IFN-gamma assay and skin test to resolve serological suspicions of Brucella was conducted in an YeO:9 infected pig herd. Following a screening of 200 pigs 39 pigs were identified with false positive serological Brucellosis reactions. While 36 of the 39 FPSR pigs were also FPSR in a second test, none of the pigs were test positive in whole blood IFN-gamma assay or Brucellergene OCB skin test. In conclusion, use of IFN-gamma assay and skin test as measurements of cell-mediated immune responses to non-LPS Brucella antigens were specific and sensitive in discriminating subclinical experimental infections with B. suis from both natural and experimental infections with YeO:9.  相似文献

蓝耳病是影响规模化猪场繁殖性能的主要传染性疾病,笔者在江苏省6个年产万头商品猪的规模化猪场猪群中随机采集297头种猪及仔猪血清进行蓝耳病血清学调查,使用IDEXX酶联免疫吸附诊断试剂盒,检测蓝耳病病毒抗体。结果表明,2个未免疫接种老猪场猪群抗体阳性率52%,断奶仔猪群阳性率39.1%;2个免疫接种老猪场猪群抗体阳性率94%,断奶仔猪群阳性率89.8%。而较新猪场的种猪群阳性率28.6%,断奶仔猪群阳性率5%。  相似文献

Campylobacter species are leading agents of human bacterial gastroenteritis and consumption of food of animal origin is a major source of infection. Although pigs are known to frequently exhibit high counts of Campylobacter in their faeces, more information is needed about the dynamics of this excretion. An experimental trial was conducted to evaluate the faecal excretion of Campylobacter by 7-week-old specific pathogen-free piglets inoculated per os with three Campylobacter strains (one C. coli isolated from a pig, one C. coli and one C. jejuni from chickens) alone or simultaneously (5x10(7)CFU/strain). Non-inoculated pigs were housed in adjacent pens. Pigs were monitored for 80 days for clinical signs and by bacteriological analysis of faeces. Pigs inoculated with porcine C. coli or with a mix of the three strains excreted from 10(3) to 10(6)CFU/g of faeces with a slight decrease at the end of the trial. Animals inoculated with poultry C. coli or C. jejuni strain excreted a lower quantity and some of them stopped excreting. At the end of the trial, only C. coli was detected in the faeces of pigs inoculated simultaneously with the three bacteria. Moreover, the transmission of Campylobacter was noticed between pens for the two C. coli strains and all the neighbouring animals became shedders with a level of excretion similar to the inoculated pigs. Intermittence in the Campylobacter excretion was also observed. Finally, our study highlighted a host preference of Campylobacter, namely C. coli seems to have a higher colonization potential for pigs than C. jejuni.  相似文献

Sepsis with subsequent multisystem organ failure after translocation of bacteria from the gut is a serious risk associated with stress situations. We showed that intestinal bacterial translocation could be one of the pathways for pathogenic Streptococcus suis infections in the pig. In 24 piglets weighing 10-14 kg, free of the extracellular factor (EF+) producing phenotype of S. suis serotype 2, a silicon canula was placed in the proximal jejunum to enable intestinal inoculation and bypassing the upper alimentary tract. The pigs were individually housed. After stress induction in 18 pigs by means of a truck drive in individual cages for 1h, pigs were inoculated through the intestinal canula either with S. suis type 2 EF+ or with growth medium only, and put back in their original housing. The six not transported pigs were also inoculated with the same strain. To prevent oral self-infection, faeces were collected in a bag that was glued around the anus. Clinical and behavioral symptoms were recorded for 72 h post inoculation, and then the animals were sacrificed for pathological and bacteriological examination. In three animals, the inoculation strain was re-isolated from mesenterial lymph nodes and typically affected organs. No S. suis type 2 EF+ was detected by specific polymerase chain reaction (PCR) in any of the tonsil-swabs and -homogenates. We concluded that infection of the organs had taken place after bacterial translocation out of the gut and that the intestinal tract can be a porte d'entree for S. suis type 2 EF+.  相似文献

A standardized model of Streptococcus suis type 2 infection in specific-pathogen-free piglets, housed in high-security barns, was used to compare the virulence of 3 French field strains of S. suis serotype 2 isolated from tonsils of a healthy pig (strain 65) or from diseased pigs (meningitis, strain 166', or septicemia, strain 24). In one of the 2 trials, 7-week-old pigs, in 3 groups of 8, were inoculated intravenously with 2 x 10(8) colony-forming units of S. suis type 2. In each group, 1 uninfected animal was a sentinel. Eight animals were also used as negative control group. The experiment was repeated under similar conditions with strains 65 and 166'. Virulence differed markedly among these S. suis strains when clinical signs, zootechnical performances, lesions, and bacteriological data were analyzed. Strain 65 did not induce clinical signs in inoculated pigs. In contrast, pigs infected with the other 2 strains exhibited clinical signs and typical lesions of S. suis type 2 infections. Differences in virulence were also observed between the 2 virulent strains. Sentinel animals exhibited the same manifestations as those recorded in inoculated piglets. Results were similar in the second trial, indicating that under the present experimental conditions, results were reproducible. The standardized conditions described in this study could be a useful tool to further study about the S. suis infection.  相似文献

Combined rotavirus and K99 Escherichia coli infection in gnotobiotic pigs   总被引:2,自引:0,他引:2  
Fifty nine 3-day-old gnotobiotic pigs were randomly assigned to 4 experimental groups: 14 pigs were orally inoculated with rotavirus (RV), 14 were orally inoculated with enterotoxigenic Escherichia coli (ETEC), 18 were orally inoculated with both agents, and 13 were controls. Pigs inoculated with RV plus ETEC were given the RV inoculum at 3 days of age and then, 24 hours later, were given the ETEC inoculum. Three pigs inoculated only with RV, 3 pigs inoculated only with ETEC, 4 pigs inoculated with RV plus ETEC, and 3 pigs in the control group were euthanatized at 5 and 7 days of age. Two pigs in each of the 4 experimental groups also were euthanatized at 9 days of age. Intestinal segments from 6 sites in the small intestine were examined by virologic, bacteriologic, and histologic procedures. For 10 days after inoculation, the remaining pigs in each group were observed clinically to monitor severity and duration of diarrhea, mortality, and shedding of RV or ETEC. Pigs inoculated with the combined RV plus ETEC inoculum developed more severe diarrhea, compared with pigs inoculated with the single agents; all dually inoculated pigs died between 3 and 6 days after inoculation. There was no mortality in pigs inoculated with either RV or ETEC. Lesions were restricted to the small intestine in pigs inoculated with RV plus ETEC and in pigs inoculated with RV or ETEC. 试验研究了日粮中添加凝结芽孢杆菌对仔猪血浆抗氧化指标和肠黏膜生长的影响。选取30头体重相近的21日龄健康断奶仔猪随机分为3组(对照组、试验Ⅰ组和试验Ⅱ组),每组10个重复,每个重复1头猪。对照组饲喂基础日粮,试验Ⅰ、Ⅱ组在基础日粮的基础上分别添加2.0×10~6、2.0×10~7 CFU/g的凝结芽孢杆菌。正试期21 d,试验结束时前腔静脉采血后屠宰取样。结果显示:与对照组相比,试验Ⅰ和Ⅱ组断奶仔猪血浆超氧化物歧化酶(SOD)活性、空肠和回肠绒毛高度与隐窝深度的比值、回肠总蛋白(TP)含量及RNA与DNA的比值均显著提高(P<0.05);试验Ⅱ组仔猪血浆丙二醛(MDA)含量显著降低(P<0.05);试验Ⅰ组空肠TP含量及TP/DNA显著提高(P<0.05)。在本试验条件下,日粮中添加凝结芽孢杆菌可以提高断奶仔猪的抗氧化能力,促进仔猪肠道黏膜细胞的增殖与更新,从而改善了肠道黏膜形态结构。  相似文献

为了评定无针注射器接种猪瘟活疫苗的免疫效果,将300头非免仔猪随机分为6个组,其中3组用无针注射器分别免疫1头份、1/2头份剂量的瘟倍安以及1头份剂量的STTM猪瘟活疫苗,剩下的3组将无针注射器替换成传统的有针注射器进行免疫。30日龄进行首次免疫,55日龄进行二次免疫,免疫后一周内观察猪有无不良反应。首免后第14天、二免后第25天分别采血、分离血清,ELISA试剂盒检测CSFV抗体,计算每组猪抗体阻断率及免疫合格率,统计分析各组之间的差异。实验结果显示,无针注射器与有针注射器接种疫苗均未对猪精神状态、采食、运动造成影响,无针注射器接种部位炎症发生率也低于有针注射器;无针注射器免疫组抗体滴度明显高于有针注射器接种组,即使疫苗使用剂量减半也能达到很好的免疫效果。  相似文献

The enteric pathogenicity of the ovine C. psittaci serotype 1 isolate S26/3 was assessed using a litter of gnotobiotic piglets. In one group, eight piglets were inoculated at 3 days of age; at 10 days, two of these were re-inoculated. In a second group, six animals were mock-inoculated at 3 days of age as negative controls; subsequently, at 10 days, three of these piglets were inoculated with C. psittaci. The animals were observed for clinical signs, killed and necropsied sequentially between 4 and 17 days of age. At necropsy, specimens were collected for histopathology, immunohistochemistry and serology. Clinical manifestations consisted of sporadic slight softening of faeces observed between 8 and 12 days post inoculation (d.p.i.) in pigs inoculated at 3 days of age and between 4 and 6 d.p.i. in those inoculated at day 10. Histopathological changes were minimal and inconsistent and occurred almost exclusively in the small intestine in pigs of 15 days of age and older; they consisted of a slight shortening of villi, of a small number of tongue-shaped villi and of villous fusions. Immunohistochemistry revealed small numbers of chlamydial inclusions in the small intestinal enterocytes of only five pigs, all killed within 5 d.p.i. An ELISA run on faecal samples collected daily after inoculation from six of the pigs showed that chlamydial antigen was excreted in the faeces. In pigs inoculated at 3 days, chlamydial antigen was detected inconsistently before, and consistently after 9 d.p.i. Pigs inoculated at 10 days excreted antigen consistently after inoculation until the end of their observation period (8 d.p.i.). Infective chlamydiae were detected from the faeces of inoculated piglets using Vero cell cultures. Sera of all pigs were negative for anti-chlamydial antibodies using a complement fixation test. In conclusion, enteric pathogenicity of C. psittaci serotype 1 in a litter of gnotobiotic piglets proved minimal. The results, therefore, indicate that serotype 1 C. psittaci is not likely to cause enteric disease in conventionally reared pigs. Nevertheless, a potential role of swine in the epidemiology of this agent should be considered with regard to spread of Chlamydia to other species.  相似文献

12 Large-White-Landrace piglets were subdivided in four groups of 3 and housed in separate units. The piglets of three groups were inoculated with the 86/27V 6C2 thymidine kinase negative (TK-) mutant of pseudorabies virus (PRV), by different routes. A second inoculation with the same mutant was given to the pigs 21 days later. The animals of a fourth group were left as uninoculated controls. 21 days following the second inoculation with the TK- mutant all pigs were challenge infected with the virulent PRV. On post challenge day (PCD) 30 all pigs were killed and samples for virus detection and histology were taken from several organs. The inoculated TK- mutant of PRV did not induce any ill effects in the pigs except a transient febrile reaction in some animals. Virus was recovered from nasal swabbings from one pig 2 days after the first inoculation of the mutant. After challenge exposure with virulent PRV, the TK- mutant-inoculated pigs were apparently protected, whereas the control pigs all were severely affected and recovered very slowly over 3 weeks. Virus was isolated from the nasal swabbings from the TK- mutant-inoculated pigs on PCDs 2 and 4, whereas the nasal swabbings from the control piglets were all positive for virus from PCD 2 through PCD 10. DNA analysis of the virus recovered showed a pattern identical to that of the virulent PRV. Histologic lesions were found in the respiratory and the central nervous systems, however, the lesions in the TK- mutant-inoculated pigs were much milder compared to those registered for the control pigs. Virus was not isolated from any of the tissue samples that were tested, but viral DNA with sequences typical of PRV genome was detected by PCR in all samples of trigeminal ganglia from either the TK- mutant-inoculated pigs or from the controls.  相似文献

The aim of this study was to compare the development of Glasser's disease in sow-reared and colostrum-deprived piglets. Ninety piglets from a commercial pig farm in Spain were used. The farm was positive for Haemophilus parasuis. Fifty-two pigs were sow-reared (SR) and 38 were colostrum-deprived (CD) piglets. The animals were intratracheally inoculated with H. parasuis serovar 5 and sacrificed at 1, 2 and 3 days post-infection. To assess the development of disease, antibody titers, clinical signs, pathological lesions, microbiological isolation and PCR amplification were compared between the groups. Inoculation of SR pigs did not cause clinical signs or lesions of Glasser's disease. In SR pigs, H. parasuis isolation and specific PCR amplification from tissues showed a very low number of positive samples. In contrast, in CD pigs, inoculation resulted in the typical signs and lesions of Glasser's disease. Positive microbiological isolation and specific PCR products were obtained from the majority of the tissues tested, and no antibodies against H. parasuis were detected. The experimental infection using CD pigs describes a successful method to study this microorganism and confirms the important role that maternal antibodies play in protection against clinical signs and disease.  相似文献

Virulence of enterotoxigenic Escherichia coli (ETEC) is associated with fimbrial adhesins and enterotoxins such as heat-labile (LT) and/or heat-stable (ST) enterotoxins. Previous studies using a cell culture model suggest that exclusion of ETEC from attachment to epithelial cells requires expression of both an adhesin such as K88 (F4) fimbriae, and LT. To test the ability of non-pathogenic E. coli constructs to exclude virulent ETEC sufficiently to prevent clinical disease, we utilized a piglet ETEC challenge model. Thirty-nine 5-day-old piglets were inoculated with a placebo (control), or with either of the three K88(+)E. coli strains isogenic with regard to modified LT expression: 8017 (pBR322 plasmid vector control), non-toxigenic mutant 8221 (LT(R192G)) in pBR322, or 8488, with the LT gene fused to the STb gene in pBR322 (LT(R192G)-STb). Piglets were challenged with virulent ETEC Strain 3030-2 (K88(+)/LT/STb) 24h post-inoculation. K88ac receptor-positive piglets in the control group developed diarrhea and became dehydrated 12-24h post-challenge. Piglets inoculated with 8221 or 8488 did not exhibit clinical signs of ETEC disease; most piglets inoculated with 8017 showed diarrhea. Control pigs exhibited significant weight loss, increased blood total protein, and higher numbers of colony-forming units of 3030-2 E. coli in washed ileum and jejunum than treated pigs. This study shows for the first time that pre-inoculation with an avirulent strain expressing adhesive fimbriae and a non-toxic form of LT provides significant short term protection from challenge with a virulent ETEC strain that expresses the same fimbrial adhesion and enterotoxin.  相似文献
