从引进的含亨得拉病毒(Hendra virus,HeV)基因的克隆质粒中扩增得到该病毒核蛋白基因(N),将其克隆到pET-28a(+)原核表达栽体,转化大肠杆菌后成功地表达了N蛋白,SDS-PAGE电泳分析结果表明目的基因在JM109(DE3)大肠杆菌菌株中得到了良好的表达,目的蛋白量占菌体总蛋白的14.6%。Western-blotting检测表明,表达的蛋白可以被兔抗亨得拉全病毒阳性血清识别,表明该蛋白具有良好的反应性。该研究结果可进一步应用于诊断试剂和单克隆抗体的制备以及流行病学调查,以防范该病在我国的流行。  相似文献

CIDR和PMSG诱导东北梅花鹿同期发情的初步研究   总被引:6,自引:2,他引:4  
应用阴道内孕酮释放装置 (CIDR)结合不同来源和不同剂量的PMSG处理 ,观察东北梅花鹿同期发情效果。 2 6只试验鹿分成 5组 ,将CIDR放置母鹿阴道内 15d后取出 ,肌注不同剂量 (10 0~ 5 0 0IU)孕马血清促性腺激素 ,放入带试情布的公鹿试情 ,持续观察 72h。结果表明 ,取出CIDR后试验鹿在 (33.3± 7.0 )h发情。其中含 10 0IUPMSG的“Folligon”处理的试验鹿同期发情率达 10 0 % ;国产PMSG处理的梅花鹿随着PMSG剂量的增加 ,发情率有增加的趋势 ,但各组间差异不显著 (χ2 检验P >0 .0 5 )。同时 ,不同剂量的PMSG处理没有显著影响CIDR取出后至发情时的平均时间长度。  相似文献

Smutsvingergras‐ en oulandsgrashooi is by drie bemestingspeile, naamlik 30 kg N + 2,8 kg P ha‐1; 90 kg N + 8,3 kg P ha‐1 en 150 kg N + 13,8 kg P ha‐1 geproduseer. Die hooi is in alle voorgenoemde gevalle op twee verskillende stadia gesny, naamlik gedurende die pypstadium en gedurende die vroeë volwasse stadium (sodra bloeiwyses oopgegaan het).

Die hooi wat in alle voorgenoemde behandelings gesny is, is aan groepe skape op ‘n kafeteriastelsel gevoer om voorkeurinname te bepaal. Soortgelyke hooi is ook aan individuele skape gevoer om inname te bepaal.

Die resultate het aangedui dat skape deurgaans ‘n voorkeur aan Smutsvingergrashooi, bo oulandsgrashooi gegee het. Die verband tussen bemesting, snystadium en inname kon nie duidelik uit die resultate bepaal word nie. ‘n Verband tussen hoër in‐vitro verteerbaarheid en hoër fosforinhoud met inname kon uit die resultate afgelei word.

Die gebruik van oulandsgras as hooigewas is ‘n algemene praktyk, maar Smutsvingergras kan ook op grond van bevindinge in hierdie ondersoek as hooigewas vir skape aanbeveel word.  相似文献

ABSTRACT: An experimental infection with highly pathogenic avian influenza virus (HPAIV) and low pathogenic avian influenza virus (LPAIV) was carried out in red-legged partridges (Alectoris rufa) in order to study clinical signs, gross and microscopic lesions, and viral distribution in tissues and viral shedding. Birds were infected with a HPAIV subtype H7N1 (A/Chicken/Italy/5093/1999) and a LPAIV subtype H7N9 (A/Anas crecca/Spain/1460/2008). Uninoculated birds were included as contacts in both groups. In HPAIV infected birds, the first clinical signs were observed at 3 dpi, and mortality started at 4 dpi, reaching 100% at 8 dpi. The presence of viral antigen in tissues and viral shedding were confirmed by immunohistochemistry and quantitative real time RT-PCR (qRRT-PCR), respectively, in all birds infected with HPAIV. However, neither clinical signs nor histopathological findings were observed in LPAIV infected partridges. In addition, only short-term viral shedding together with seroconversion was detected in some LPAIV inoculated animals. The present study demonstrates that the red-legged partridge is highly susceptible to the H7N1 HPAIV strain, causing severe disease, mortality and abundant viral shedding and thus contributing to the spread of a potential local outbreak of this virus. In contrast, our results concerning H7N9 LPAIV suggest that the red-legged partridge is not a reservoir species for this virus.  相似文献

Five Merino sheep were dosed 3 g/kg of dry, finely-milled Homeria glauca (Natal yellow tulp) plant material. An electrocardiogram was recorded and the arterial and central venous blood pressure, blood gases, haematological variables, plasma electrolytes (Na+, K+, Ca2+, Mg2+, Cl- and PO4(2-) ) and a variety of serum enzymes and chemical constituents were measured hourly until death (3 sheep) or until sheep were in extremis (2 sheep). Heart rate rose progressively as a result of sinus and, later, ventricular tachycardia. Systolic blood pressure rose, but there was little change in the mean and diastolic arterial pressures and central venous pressure. There was progressive hypoxaemia, hypercarbia and acidaemia with depletion of plasma bicarbonate. Haemoconcentration, hyperkalaemia and hypochloraemia were found along with rising serum creatinine and plasma glucose. Rises in serum enzymes indicated widespread tissue damage. Electrocardiographic recordings were being made at the moment of death in 3 of the 5 sheep. In these 3 sheep the cause of death was ventricular fibrillation.  相似文献

Two hundred and twelve dog cadavers belonging to different breeds were examined, to investigate the formation of the femoral, obturator and the sciatic nerve. Besides the commonly described formation patterns of the mentioned nerves, some variations were also found. These variations were not gender-related, but on the other hand we discovered a certain correlation between the variations appearing in the formation of the femoral, obturator and the sciatic nerve. In 74.05% of cases, the femoral nerve was formed from ventral branches of the 4th, 5th and 6th lumbar nerve, and 16.98% of the dogs had the nerve formed from ventral branches of the 3rd, 4th and 5th lumbar nerve. Many dogs (i.e. 2.30%) had the femoral nerve formed from the ventral branches of the 5th, 6th and 7th, the 3rd, 4th, 5th and 6th or the 4th, 5th, 6th and 7th lumbar nerve, respectively. In 1.88% of dogs in particular, the femoral nerve was formed from ventral rami of the 4th and 5th lumbar nerve. In 66.98% of the examined dogs, the obturator nerve was formed from the ventral branches of the 4th, 5th and 6th lumbar nerve, followed by 16.59% of the dogs with the obturator nerve formed from the ventral rami of the 4th and 5th lumbar nerve and 9.43% of dogs in which the nerve was formed from the ventral branches of the 5th and 6th lumbar nerve. In 4.71% of dogs, the obturator nerve was formed from the ventral rami of the 4th, 5th, 6th and 7th lumbar nerve, while only 2.30% of the examined dogs had the same nerve formed from the ventral branches of the 5th, 6th and 7th lumbar nerve. The sciatic nerve was formed from ventral branches of the 6th and 7th lumbar nerve and the 1st sacral nerve in 86.79% of the dogs. In 5.18% of cases, the same nerve was formed from a junction of the ventral branches of the 7th lumbar and the 1st and 2nd sacral nerve, and, in the same percentage of cases, it was formed from a junction of ventral branches of the 6th and 7th lumbar nerve and the 1st and 2nd sacral nerve. In 2.83% of the dogs, it was formed from a junction of the ventral branches of the 6th and 7th lumbar and the 1st sacral nerve. The correlation of variations established in the formation of the femoral, obturator and the sciatic nerve was not statistically significant.  相似文献
