Copper, zinc and iron levels were measured in cerebrospinal fluid (CSF) of normal and copper poisoned sheep. Copper values were within the range recorded for humans but zinc levels were six to 20 times and iron levels were 20 to 30 times higher than values reported for humans. The amount of copper, zinc and iron in the CSF did not increase in sheep dosed with copper and hence it is unlikely that the level of copper in the CSF is related to the changes in the nervous system that have been reported in copper poisoned sheep.  相似文献

The effect of a systemic disease on the dynamics of iron, zinc, and copper in chickens fed ad libitum was examined by infecting 10-day-old specific pathogen-free chickens with infectious bursal disease virus (IBDV). Liver, bursa of Fabricius, pancreas, spleen, and serum were sampled in 10 controls and 10 challenged chickens at 3-day intervals postinfection (PI) for 15 days. The samples were analyzed using atomic absorption spectroscopy. Serum levels were similar to that reported in the literature. Concentrations of iron and zinc did not change significantly in the pancreas, but there was an increase in copper in infected pancreatic tissue on days 9 and 15 PI. Iron concentration in the spleen showed a significant increase on days 6, 9, and 15 PI, whereas zinc was only significantly increased on day 15 PI. There was no significant change in copper concentrations in the spleens of infected chickens vs. controls. This finding is in line with previously reported data. The results showed that the liver was not a major tissue where iron and zinc were sequestered, as previous data have shown in mammals. Instead, the bursa of Fabricius had significantly increased levels of both iron and zinc in infected tissue vs. control tissue from 9 days PI on. Furthermore, the bursa had increased levels of copper in the latter portion of the study. These findings suggest that the bursa of Fabricius rather than the liver is the major organ for metallic ion sequestering during IBDV infection.  相似文献

The effect of zinc deficiency on trace metals in the liver, spleen, kidney, pancreas and duodenum was investigated in the control and zinc-deficient rats at 17 days and 20 days of pregnancy. Zinc-deficient rats fell into limosis after 5 days of pregnancy. The contents of zinc, iron, copper and manganese in the maternal tissues were measured by colorimetry with 5NPPF. The morphological changes of the liver were observed by light and electron microscopy. The contents of zinc in the pancreas and duodenum were less in the zinc-deficient group than in the control at both 17 days and 20 days of pregnancy. The contents of the copper and manganese in the liver, kidney, pancreas, duodenum and spleen, however, were not significantly different from the control. The contents of iron in the liver, spleen and kidney in the deficient group increased greatly at 17 days and 20 days of pregnancy compared with the control group. The combination rate of transferrin with iron also increased significantly in the deficient group. Staining with Berlin blue or Turnbull's blue showed intense reaction to iron around the interlobular connective tissue of the liver in the zinc deficient group at 20 days of pregnancy. Ultrastructurally, the liver of the zinc-deficient rats showed the decrease of glycogen granules and increase of lipid-like granules and lysosomes with various sizes and electron densities. These findings suggest that zinc deficiency causes the increase of iron contents in the various organs during pregnancy, and that there is an intimate interrelationship between zinc and iron in the metabolism of iron during pregnancy.  相似文献

Concentrations of iron, copper, and zinc were measured in livers of 95 dogs that were suspected of having liver disease. Iron concentrations ranged from 177 to 7,680 ppm (dry weight basis); 54 dogs had iron concentrations greater than the normal concentration of 1,200 ppm. Iron stores were present in Kupffer cells and macrophages but not hepatocytes. The dogs did not have lesions of hemochromatosis. Dogs with high liver iron tended to have high liver copper and inflammatory lesions. High liver copper concentrations usually were associated with hepatocellular necrosis and fibrosis. High liver zinc was found in only 5 animals and was accompanied by histologic inflammatory lesions in one. In humans, increased iron concentration in the liver exacerbates liver damage caused by a variety of insults, and the same may be true for dogs.  相似文献

The concentrations of copper, zinc and molybdenum were measured in the liver of goats from western and south-eastern Norway. The mean copper concentrations in the liver of goats from these two districts were 23±19 µg Cu/g and 59±31 µg Cu/g wet weight, respectively. As for zinc and molybdenum, no difference was found between the two groups of animals. No correlations were detected between copper and zinc, zinc and molybdenum, or copper and molybdenum. The copper levels in Norwegian goat liver are considerably lower than in sheep liver, and the ranges are significantly more narrow. The concentrations of molybdenum in goat liver are at the same levels as in sheep and swine, while the levels of zinc are somewhat lower.  相似文献

1. Zinc, iron and copper interactions were examined in chicks (New Hampshire X Single Comb White Leghorn) grown individually in stainless steel cages and fed on purified diets with three concentrations of zinc: control (37 mg Zn/kg), moderate (100 mg Zn/kg) and excess (2000 mg Zn/kg) in three studies. 2. Chicks given either moderate or excess zinc excreted more than twice as much 65Zn from a test diet as those fed on the control diet. Thus chicks given the moderate amount of zinc had concentrations of zinc in tissues similar to those of controls but chicks given excess zinc accumulated zinc in their tissues. 3. The specific activity of 65Zn in the tissues, especially in the bursa of Fabricius, of chicks given excess zinc was not reduced as much as would be predicted. Thus, ingestion of excess zinc appeared to reduce tissue turnover of 65Zn, especially in the bursa. However, humoral immune responses, as measured by antibody titres to sheep red blood cells, were not affected. 4. Chicks given excess zinc or pair-fed on the control diet retained less 59Fe from a test diet than chicks given control or moderate concentrations of zinc ad libitum. The iron concentrations in tibiae of chicks given excess zinc were depressed.  相似文献

The efficacy of a molybdate formulation and a zinc oxide bolus as prophylactic agents for enzootic icterus was evaluated in sheep. Before copper loading, liver biopsies were performed on 12 male, 6-month-old, Mutton Merino sheep to determine hepatic copper (Cu) and zinc (Zn) concentrations. The animals were restrictively randomised according to liver copper concentrations to 3 treatment groups (n = 4) to achieve similar mean liver copper concentrations per group. All sheep received 4 ml/kg of a 0.5 % aqueous solution of CuSO4 5H2O intraruminally 7 days per week for 10 weeks. On Day 0 the sheep in the Mo-group were injected subcutaneously with 42 mg molybdenum (Mo) contained in a commercial molybdate formulation. The animals in the Zn-group each received a zinc oxide bolus, containing 43 g zinc oxide, via a rumen cannula. Treatment was repeated on Day 42. Four animals served as untreated controls. Urinary copper excretion, plasma copper concentration, haematocrit and glutamate dehydrogenase (GLDH) activity were determined throughout the trial. The animals were sacrificed after 10 weeks and liver samples were submitted for histopathological examination. Liver and kidney copper and zinc concentrations were determined. Neither the molybdate treatment nor the zinc oxide boluses prevented hepatic copper accumulation. The urinary copper excretion, plasma copper concentration, haematocrit and GLDH activity were not significantly different (P > 0.05) from the controls.  相似文献

Changes in copper and zinc concentrations in the blood serum of sheep were studied in clinical conditions for sixty days in relation to the intramuscular application of zinc oxide at a dose of 10 mg Zn per kg liveweight; zinc oxide was administered in a developmental preparation. Serum concentrations of zinc started increasing significantly the eighth day after administration (p less than 0.01), the maximum values were found out on the twelfth day (21.19 +/- 3.89 mumol X l-1). Significantly higher concentrations of zinc in the blood serum of test sheep were observed as soon as on the eighth day (p less than 0.01), in comparison with the untreated group, and they persisted till the end of investigation (p less than 0.01). Simultaneously with the changes in serum zinc concentrations, copper metabolism was influenced when the statistically higher values of copper concentrations in the blood serum of the test group were recorded the eighth and twelfth days (p less than 0.01), in comparison with the control animals. In the remaining period, serum copper concentrations did not show any mathematical dependence and the equalization to the original values in the treated group occurred at the end of the experiment. The above-mentioned results from the sheep treated with zinc oxide refer to a possibility of influencing mutually the copper and zinc metabolism also parenterally; this could be utilized during the treatment of secondary deficiencies, or toxicosis.  相似文献

Reference values were established for some haematological and serum biochemical constituents in Bactrian camels (Camelus bactrianus) in China. The contents of seven trace elements in the blood, hair, liver, kidney, heart, spleen, lung, cerebrum, cerebellum, rib, ovary, pancreas and gluteal muscle of Bactrian camels were also measured. Some of these values are reported for the first time for Bactrian camels in China. Most haematological and serum biochemical values were similar to those of cattle, yaks, sheep and dromedary camels, but the mean serum albumin concentration and the albumim/globulin ratio were significantly higher than those in other ruminants and the mean thyroxine concentration was half that in dromedary camels. The liver contained the highest concentrations of copper, zinc, manganese and iron, and the renal cortex contained the highest concentration of selenium. The concentrations of selenium, cobalt, zinc, manganese and molybdenum in the tissues were within the reference ranges for other ruminants, but the mean iron and copper concentrations in the liver were significantly higher than those in other ruminants.  相似文献

The contents of manganese, zinc, copper, and iron in the liver and kidney of laying hens (Shaver), kept on modern large-capacity poultry farms, were determined under the conditions of a normal and increased supply of these microelements to the organism. The content of the mentioned trace elements was determined by means of atomic absorption spectrophotometry, flameless technique, and with the use of graphite cell. In the layers fed a diet with a normal content of trace elements the average concentrations of the microelements in the liver were the following: manganese: 15.8 +/- 8.8 mg kg-1, zinc: 35.0 +/- 5.1 mg kg-1, copper: 19.9+/- 2.8 mg kg-1, and iron: 761.6 +/- 245.8 mg kg-1. When increased amounts of microelements were added to the diet, their average content generally increased just in the kidneys, whereas their average concentrations in the liver remained the same, with the exception of iron, the content of which showed an increase.  相似文献

Merino wether weaners were exposed to toxic lupin stubbles for periods of one, two and six to nine weeks, and the effect on their liver copper, selenium and zinc concentrations studied. After the one week period there was a slight decrease in liver copper and selenium concentrations in sheep severely affected with lupinosis. This was attributed to loss of these elements from necrotic cells, and greatly increased quantities of fat in the liver. In the same period liver zinc concentrations declined and were negatively correlated with the degree of liver injury. After two weeks or more of exposure to toxic lupins a positive correlation existed between both the liver copper and selenium concentrations, and the degree of liver injury. Furthermore, total liver copper and selenium levels were also positively correlated with the degree of liver injury. Liver zinc concentrations were negatively correlated with the degree of liver injury. It is suggested that when the liver is under the influence of the toxins causing lupinosis for more than two weeks, it stores copper and selenium, and loses zinc.  相似文献

The distribution of elements in the cells of the liver and kidney of normal sheep, of sheep chronically intoxicated with copper and in sheep given copper and thiomolybdate was studied by atomic absorption spectrophotometry, electron microscopy and electronprobe X-ray microanalysis. Copper concentrations were increased in the liver and kidney during the pre-haemolytic and haemolytic stages of the disease. In addition iron concentrations were markedly increased in the kidney during haemolysis. Copper and molybdenum concentrations were increased in the kidney of sheep receiving copper and thiomolybdate or thiomolybdate alone. By electronprobe X-ray microanalysis at the pre-haemolytic and haemolytic stages, copper together with small amounts of calcium and chromium were found in lysosomes of liver cells. In addition, in animals killed during haemolysis, high concentrations of iron were found in apical lysosomes of kidney proximal tubule cells. Copper and molybdenum were found in apical lysosomes of kidney tubule cells in animals given copper and thiomolybdate or thiomolybdate alone. The accumulation of copper with molybdenum and sulphur in the lysosomes of thiomolybdate treated copper intoxicated animals was demonstrated for the first time and may indicate the sequestration within lysosomes of a copper-molybdenum-protein complex.  相似文献

The aims of this study were to demonstrate the effects of zinc sulphate administration (2 g/week p.o., 2% solution) on zinc, copper, iron, calcium and magnesium levels in blood serum of 20 pregnant Ivesi sheep during the last 2 month of pregnancy, immediately post partum as well as in their newborn lambs. In these 20 lambs, also total serum protein, gamma globulin and birth weight were determined. The control group consisted of 15 pregnant sheep and their 15 lambs housed under the same conditions. Zinc sulphate administered to sheep caused significant increases in their serum levels of zinc as well as in those of their lambs. The lambs of the zinc supplemented group had also slightly higher birth weights (not significant) and significant higher gamma globulin levels, whereas the total protein values was almost identical in both groups.  相似文献

The effects of different dietary levels of zinc and vitamin A on tissue mineral accumulation and histopathological alterations were examined in laying hens. For this purpose, 130 Hisex brown laying hens aged 56 weeks were divided into two main groups. Each group of hens were fed on diets supplemented with two different levels of vitamin A (0-10,000 IU kg(-1)). Then, laying hens in both of the main groups were subdivided into five treatment groups given different levels of zinc (0, 25, 50, 100, and 200 mg kg(-1)) for 12 weeks. Dietary additions of zinc and/or vitamin A did not significantly affect the zinc concentrations of the pancreas, liver, gizzard, thyroid, and copper concentrations of the pancreas, gizzard, and thyroid. The group fed the highest zinc (200 mg kg(-1)) in the diet had twofold iron concentration in their thyroid tissue compared to the concentration of the control group receiving no extra zinc. Histopathologically, there was slight oedema, degranulation, and vacuolation in exocrine cells and some fibrotic areas in the pancreas and reduced follicular size. Follicular cell hypertrophy and hyperplasia occurred in the thyroids, and gizzard erosions were seen in some cases.  相似文献

Adult and kit male and female natural dark ranch mink (Mustela vison) were fed a conventional diet supplemented with 0, 500, 1,000, or 1,500 ppm zinc, as ZnSO4.7H2O, for 144 days. No marked adverse effects were observed in feed consumption, body weight gains, hematologic parameters, fur quality, or survival. Zinc concentrations in liver, kidney, and pancreas of the mink increased in direct proportion to the zinc content of the diet. Histopathologic examination of the livers, kidneys, and pancreata revealed no lesions indicative of zinc toxicosis. The results indicate that mink can tolerate at least 1,500 ppm dietary zinc, as ZnSO4.7H2O, for several months without apparent adverse effects.  相似文献

Two groups of adult Merino sheep, initially grazed on pasture, were dosed daily with zinc sulphate (1 mg Zn/kg LW) or zinc oxide (15 mg Zn/kg LW) for six and seven weeks, respectively. On the 18th day, both groups were transferred to indoor pens together with unmedicated control sheep, and five days later the feet of all animals were infected artificially with a virulent strain of Bacteroides nodosus. By the fourth week after challenge, 85% or more of the feet challenged had developed advanced footrot, and no significant differences in the incidence and severity of lesions between dosed and un-dosed groups were recorded. Plasma zinc concentrations, monitored throughout the experiment, remained at similar levels in sheep receiving the lower dose rate and in the controls. At the higher dose rate, plasma zinc levels increased till Day 23, but thereafter declined to values comparable to those of undosed animals. At post mortem, no evidence of zinc toxicosis was found, and only the kidneys of sheep receiving the higher dosage showed a significant accumulation of zinc compared with the controls.  相似文献

选用临床检查健康的(26±2)日龄"杜×长×大"三元杂交断奶仔猪100头,按体质量和性别随机分为5组,每组20头,分别饲喂基础日粮、基础日粮+2000mg/kg氧化锌、基础日粮+3000mg/kg氧化锌、基础日粮+250mg/kg蛋氨酸锌、基础日粮+500mg/kg蛋氨酸锌。试验期14d。于断奶后0,7,14d,经前腔静脉采血,用原子吸收光谱仪检测血清中铜、铁、锌水平。试验结束时,每组选5头仔猪放血致死,取心、肝、肾、脑、脾、胸腺组织,测定铜、铁、锌含量。结果显示,仔猪断奶后,血清中铜、锌水平显著或极显著下降(P0.05或P0.01);添加不同锌源和锌水平的高锌日粮能提高断奶仔猪肝、肾、脑、血清锌的含量,显著或极显著降低仔猪血清铜和心、胸腺铜以及脾铁水平(P0.05或P0.01),对血清铁及其他组织铁含量无明显影响。这表明高锌日粮能增加断奶应激仔猪体内锌水平,降低部分组织中铜、铁含量。  相似文献

The copper (Cu), zinc (Zn), and iron (Fe) excreted daily in the bile of three sheep were estimated at 0.20, 0.10 and 0.36 mg respectively. Intravenous administration of tetrathiomolybdate (TM) increased bile Cu excretion and reduced liver Cu concentration. Tetrathiomolybdate had no significant effect on the excretion of Zn and Fe. These findings indicate that at least one of the mechanisms by which TM induces hypocuprosis is by increasing biliary Cu excretion. Hence it has a possible use in the treatment of Cu storage diseases in man and animals.  相似文献
