Although poultry is recognized as the major source of food-poisoning caused by Salmonella, pork also contributes to human infections. This study was therefore undertaken in order to develop a reliable serological method for the evaluation of the Salmonella status of piglets. A complete ELISA was performed using lipopolysaccharides of Salmonella Typhimurium, Anatum, Hadar and Infantis because these serovars were representative of the serogroups isolated from 30 contaminated fattening farms. S. Enteritidis was also added because of its importance in human infection and to include the O:9 antigen. This method potentially detects 100% of infected pigs. A significant correlation was found between this serological method and the bacteriological data from mesenteric lymph nodes (p = 0.01). In addition, both sensitivity and specificity were high (97% and 94% respectively). The ELISA test was therefore used in a cross-sectional study on 4 farms to evaluate when pigs became contaminated: seropositive pigs were only found for the 20 week old finishing pigs. The antibody response to Salmonella in piglets was also investigated: maternal antibodies persisted until 7 weeks of age and post-Salmonella contamination seroconversion was detected from 8 weeks of age onwards.  相似文献

A national program to reduce Salmonella in pork and pork products should include monitoring and intervention at farm level. To develop an adequate intervention strategy at farm level, risk factors for Salmonella infections in finishing pigs have to be determined. In this study, blood samples were collected randomly at two slaughterhouses from slaughter pigs. Samples were tested by the Dutch Salmonella ELISA, based on the O-antigens 1, 4, 5, 6, 7 and 12, using a cut-off of OD%=10. This ELISA has been calibrated against the Danish ELISA to give comparable results. Workers from herds from which at least forty blood samples had been collected, were asked to participate in a questionnaire. In total, 353 questionnaires were obtained and analysed. Significant risk factors associated with the proportion of seropositive samples were identified by multiple linear logistic regression. The feeding of a complete liquid feed containing fermented by-products and the omission of disinfection after pressure washing a compartment as part of an all-in/all-out procedure, were both associated with a lower Salmonella seroprevalence. A small to moderate herd size (<800 finishing pigs), a previous diagnosis of clinical Salmonella infection in the herd, the use of tylosin as an antimicrobial growth promoter in finishing feed, or herds which had more than 16% of the livers of their pigs condemned at the slaughterhouse as a result of white spots were associated with a higher Salmonella seroprevalence. Hypothetical intervention strategies based on these risk factors can be studied for their effect on the Salmonella seroprevalence and practical applicability in field studies.  相似文献

Pork and pork products are increasingly recognized as an important source of human salmonellosis. In the present study, we describe the characteristics of recent German salmonellosis outbreaks related to pork, and discuss legal efforts for Salmonella control. From 2001 to 2005,five large salmonellosis outbreaks were reported in Germany for which pork was the probable vehicle of infection. Evidence came from epidemiologic studies, microbiologic testing, and trace back investigations. All outbreaks showed disperse spatial distribution, affecting multiple federal states. A full trace back of implicated food items was not possible in three of these outbreaks. In two outbreaks, international trade in pigs appears to have played a role. To prevent further human disease from contaminated pork and pork products, an integrated programme for the reduction of Salmonella on all steps of the production chain is warranted. In addition, close collaboration between epidemiologists and microbiologists from both the human and the veterinary side should be further strengthened.  相似文献

Salmonellae are wide spread in man and animals world wide and are of increasing significance as causative agents of foodborne diseases in man. The European Union, national authorities and the pig industry are therefore more and more interested in the Salmonella status of the pig population. The aim of this study was to estimate the bacteriological prevalence of Salmonella in finishing pig herds, the serogroup and the resistance to antibiotics of the isolated Salmonellae and a preliminary risk analysis of factors associated with infection. For this, 317 finishing pig herds were randomly selected from a database containing 1500 herds in the southern part of the Netherlands. In each herd 24 samples of fresh faeces were collected from two compartments with pigs close to market weight. Per compartment 12 samples of faeces were pooled into one pooled sample. Pooled samples were cultured in duplicate. Salmonella spp. were recovered from 71 out of 306 herds (23%) in which two compartments could be sampled. A total of 108 isolated Salmonella's were serotyped: 71 serogroup B, 3 serogroup C1, 6 serogroup C2, 22 serogroup D1, and 6 isolates neither serogroup B, C or D1. Of a total of 115 Salmonella isolates tested, none were resistant to colistin, enrofloxacin, flumequin or gentamicin. Automated liquid feeding of by-products, and membership of an Integrated Quality Control (IQC) production group were associated with a decreased risk of infection, while use of trough feeding was associated with an increased risk of infection. It is necessary to test these presumed risk factors in intervention studies to evaluate their potency to reduce the Salmonella prevalence in finishing pigs and thereby reduce the risk of Salmonellosis in people consuming pork.  相似文献

The purpose of this investigation was to study the incidence and course of Salmonella infections in finishing pig herds in order to asses the stability of a given Salmonella herd status. Five low- and 7 high-seroprevalence herds were followed for seven sampling rounds. Each round, blood and faecal samples were tested in an indirect ELISA and by bacteriological culturing, respectively. In high-seroprevalence herds a positive Salmonella status was an indication of a long-term problem and the status was relatively stable over time. The herds experiencing clinical salmonellosis were not necessarily the herds with the highest seroprevalence. It is possible to deliver sero-negative finishers to the slaughterhouse, even though these pigs were seropositive as growers. In three out of five low-prevalence herds, major infection incidents occurred, indicating that changes in the Salmonella status should be anticipated. Low-prevalence herds can remain negative over a longer period of time as a result feeding a complete liquid feed containing fermented by-products.  相似文献

The present study was conducted to study the impact of the virulence factors invC and sseD of the two type III secretion systems of Salmonella enterica serovar Typhimurium (S. Typhimurium) on the pathogenesis of the porcine S. Typhimurium DT104 infection. For this purpose, two S. Typhimurium mutant strains with a disrupted invC gene of the Salmonella pathogenicity island 1 or with a disrupted sseD gene of the Salmonella pathogenicity island 2 have been studied in experimental infection of pigs. Twenty-two 7-week-old male hybrid pigs were either infected with one of the mutants or the wild-type S. Typhimurium DT104 strain. Each group was examined for clinical signs, Salmonella shedding rate and the specific antibody response. Survival and replication were evaluated by qualitative and quantitative determination of the colonization rate. The humoral and cellular immune responses were examined using isotype-specific ELISA and quantitative real-time PCR of IL-2, IL-4, IL-10, IL-12 and IFN-gamma. The results proved that both mutants had a lower virulence (with marked differences between both mutants) than the wild-type and that both virulence factors have importance in porcine salmonellosis. Only pigs infected with the wild-type S. Typhimurium DT104 exhibited typical clinical symptoms of salmonellosis like anorexia, vomiting, disturbed demeanour, fever and diarrhoea. Deletion of the invC gene resulted in a significantly reduced colonization rate. Interestingly, the mRNA expression of both type-1 and type-2 cytokines were significantly decreased in pigs infected with either the invC-mutant and the sseD-mutant strain.  相似文献

Salmonella in pork can be combated during pre- or post-harvest. For large slaughterhouses, post-harvest measures like decontamination might be cost-effective while this is less likely with small-to-medium sized slaughterhouses. In this study, pre-harvest measures might be more relevant. We describe an extended surveillance-and-control programme for Salmonella in finisher pigs, which, to establish equivalence to the Swedish control programme, is intended for implementation on the Danish island, Bornholm. The effect of the programme on food safety was estimated by analysing Salmonella data from pig carcasses originating from herds that would have qualified for the programme during 2006-2008. Food safety was interpreted as prevalence of Salmonella on carcasses as well as the estimated number of human cases of salmonellosis related to pork produced within the programme. Data from the Danish Salmonella programme were obtained from Bornholm. We used a simulation model developed to estimate the number of human cases based on the prevalence of Salmonella on carcass swabs. Herds are only accepted in the programme if they have one or less seropositive sample within the previous 6 months. In this way, the Salmonella load is kept to a minimum. The programme is not yet in operation and pigs that qualify for the programme are currently mixed at slaughter with those that do not qualify. Therefore, we had to assess the impact on the carcass prevalence indirectly. The prevalence of Salmonella in carcass swabs among qualifying herds was 0.46% for the 3?years as a whole, with 2006 as the year with highest prevalence. According to the simulation the expected number of human cases relating to pork produced within the programme was below 10. When the programme is in operation, an extra effect of separating pigs within the programme from those outside is expected to lower the prevalence of Salmonella even further.  相似文献

In this paper, we describe the transmission of Classical Swine Fever virus (CSF virus) within herds during the 1997–1998 epidemic in the Netherlands. In seven herds where the infection started among individually housed breeding stock, all breeding pigs had been tested for antibodies to CSF virus shortly before depopulation. Based upon these data, the transmission of CSF virus between pigs was described as exponential growth in time with a parameter r, that was estimated at 0.108 (95% confidence interval (95% CI) 0.060–0.156). The accompanying per-generation transmission (expressed as the basic reproduction ratio, R0) was estimated at 2.9. Based upon this characterisation, a calculation method was derived with which serological findings at depopulation can be used to calculate the period in which the virus was with a certain probability introduced into that breeding stock. This model was used to estimate the period when the virus had been introduced into 34 herds where the infection started in the breeding section. Of these herds, only a single contact with a herd previously infected had been traced. However, in contrast with the seven previously mentioned herds, only a sample of the breeding pigs had been tested before depopulation (as was the common procedure during the epidemic). The observed number of days between the single contact with an infected herd and the day of sampling of these 34 herds fitted well in the model. Thus, we concluded that the model and transmission parameter was in agreement with the transmission between breeding pigs in these herds.

Because of the limited sample size and because it was usually unknown in which specific pen the infection started, we were unable to estimate transmission parameters for weaned piglets and finishing pigs from the data collected during the epidemic. However, from the results of controlled experiments in which R0 was estimated as 81 between weaned piglets and 14 between heavy finishing pigs (Laevens et al., 1998a. Vet. Quart. 20, 41–45; Laevens et al., 1999. Ph.D. Thesis), we constructed a simple model to describe the transmission of CSF virus in compartments (rooms) housing finishing pigs and weaned piglets. From the number of pens per compartment, the number of pigs per pen, the numbers of pigs tested for antibodies to CSF virus and the distribution of the seropositive pigs in the compartment, this model gives again a period in which the virus most probably entered the herd. Using the findings in 41 herds where the infection started in the section of the finishers or weaned piglets of the age of 8 weeks or older, and of which only a single contact with a herd previously infected was known, there was no reason to reject the model. Thus, we concluded that the transmission between weaned piglets and finishing pigs during the epidemic was not significantly different from the transmission observed in the experiments.  相似文献

The aim of this study was to determine the role of maternally derived antibodies (MDA) against an influenza H1N1 virus in the clinical protection of piglets and especially their effect on the development of the active immunity after an infection with a homologous influenza H1N1 virus. Twenty piglets with MDA and 10 piglets without MDA were housed together and inoculated twice with influenza H1N1 virus, at 7 and 15 weeks of age. Nine piglets without MDA were added to these groups at 12 weeks of age to be inoculated at 15 weeks of age only. Clinical signs, body temperature, growth performance, virus excretion, antibody responses, and influenza-specific T-cell response were monitored. It was shown that MDA protect piglets against the clinical consequences of a primary influenza infection, but that this protection is not complete. A short but significant rise in body temperature was observed and growth seemed to be inhibited due to the infection. Piglets with MDA shed virus for a longer period after an infection than piglets without MDA. Piglets with and without MDA were protected against the clinical consequences of a secondary infection. However, both after primary and secondary infection significant differences in immune responses were observed that indicated that pigs with MDA developed a weaker immunity than pigs without MDA. Furthermore, overall growth performances from weaning to slaughter show a trend in favour of pigs without maternal antibodies, compared to pigs with maternal antibodies, mainly caused by a significant better performance in the second half of the finishing period. The results of this study provide us insight in the role of MDA in clinical protection and their influence on active immunity after an influenza virus infection of pigs. Furthermore, it leads us to the discussion about the profitability of massive sow herd vaccinations in an attempt to increase MDA levels in piglets, taking into account the overall performance of these piglets and the possible effects on antigenic drift.  相似文献

Clinical salmonellosis associated with Salmonella is increasingly reported in finishing swine. Since S. Typhimurium is often associated with these episodes and given that this serotype is among the most often reported in humans, we were interested to determine if various tissues and carcasses from animals coming from herds that were clinically affected were more likely to be contaminated by Salmonella compared to carcasses from animals raised in herds without any history of salmonellosis. Carcasses from animals from affected herds were significantly more contaminated by Salmonella while showing increased titers in antibodies directed against this bacterium. At the opposite, caecal contents and mesenteric lymph nodes from both groups of animals were similarly contaminated by Salmonella. In the second part of the study, we studied the persistence of the bacterium in various tissues after an experimental infection with S. Typhimurium. We found that, after the infection, Salmonella persisted for as many as 7 d in many extraintestinal tissues, while it was present in the feces of infected animals for all 14 d of the experiment. These findings indicated that carcasses from animals that experienced salmonellosis during their growth phase are more likely to be contaminated by this bacterium and that precautions must be taken in order to ensure that clinically affected animals should be kept on the farm for at least 7 d before being shipped for slaughter.  相似文献

The objective of this study was to determine whether a chlamydial strain recovered from growing and finishing swine with conjunctivitis or keratoconjunctivitis could cause the same infections in gnotobiotic pigs. The strain shares biological characteristics with Chlamydia trachomatis. After propagation in Vero cells and preparation of the inoculum (10(7) inclusion-forming units/ml), chlamydial strain H7 was instilled into the ventral conjunctival sac (0.15 ml/sac) of 12 anesthetized 3-day-old gnotobiotic piglets. Four age-matched gnotobiotic piglets were anesthetized and sham infected with uninfected cell culture lysates. None of the principal piglets developed clinical symptoms of conjunctivitis or keratoconjunctivitis. Principal piglets necropsied 7 days postinfection (DPI) had histologic lesions of mild or moderate conjunctivitis; immunohistochemical evaluation revealed chlamydial antigen in conjunctival epithelium. A majority of principal piglets necropsied at 14-28 DPI had histologic lesions of mild conjunctivitis, but chlamydial antigen was not detected by immunohistochemistry. The results indicated that chlamydial strain H7 can cause mild or occasionally moderate conjunctivitis in gnotobiotic pigs, but the conjunctival infection is asymptomatic.  相似文献

试验采用流行病学调查、临床观察和实验室检查对华北地区某疑似伪狂犬病发病猪场进行了诊断。猪场发病情况主要为母猪流产,新生仔猪死亡率较高,肥猪死亡率低。发病或死亡仔猪扁桃体充血出血、溃疡,肺脏出血、水肿,心肌色淡、心包积液,脑膜充血;表现为坏死性扁桃体炎、出血性淋巴结炎、出血性间质性肺炎、间质性心肌炎和非化脓性脑炎。发病猪扁桃体、肺脏、脑组织匀浆上清液接种家兔后出现奇痒等典型症状并很快死亡。发病猪脑组织可检出猪伪狂犬病毒gD基因特异性目的片段。猪场伪狂犬病野毒感染抗体阳性率为63%(63/100)。本试验结果表明,确诊该猪场发生了伪狂犬病。  相似文献

The implementation of Salmonella control programs in the pork production chain demands rapid and cost-effective methods to assess the prevalence of infection in pig herds. The objective of the present study was to develop an in-house enzyme-linked immunosorbent assay (ELISA) based on S. Typhimurium lipopolysaccharides (LPS) to measure the prevalence of infection caused by Salmonella in swine herds. Coating antigen was produced by phenol extraction of S. Typhimurium culture. After standardization of ELISA test conditions, the assay was validated by testing serum samples on different animal categories: pigs orally inoculated with S. Typhimurium and sentinel animals in contact with them, naturally infected animals, colostrum-deprived piglets, and bacterin-immunized pigs. Seroconversion was observed in inoculated pigs (7 days postinfection [DPI]) and in the sentinels (21 DPI). Nonspecific reactions were not detected in the sera of colostrum-deprived animals. Serum samples from animals immunized with Salmonella Agona, Salmonella Derby, Salmonella Panama, and Salmonella Bredeney bacterins showed marked cross-reaction with the LPS from the serovar Typhimurium. Moreover, positive results obtained with the in-house ELISA were associated with Salmonella isolation in 75 infected pig herds. Comparisons with 2 commercial kits showed a linear correlation coefficient of 0.847 between the in-house ELISA and kit A and 0.922 with kit B but a low agreement in the qualitative results. In conclusion, the newly developed in-house ELISA based on S. Typhimurium LPS can be a useful tool to determine the intensity of Salmonella sp. infection in swine herds.  相似文献

One hundred and thirteen finishing pig units and 74 sow units in Catalonia, Spain, were examined to determine the prevalence of salmonella infections and the factors that could be associated with them. Pooled faecal samples were taken from the finishing units, and samples of faeces were collected from individual sows. The Salmonella isolates were serotyped, phage typed and examined for their antimicrobial susceptibility to 18 common antimicrobial drugs. In addition, blood samples from pigs on 141 farms were analysed by ELISA. In both the bacteriological and serological surveys, a questionnaire with 84 questions was completed for each farm. Salmonella species were isolated from 20 per cent of the finishing units and 24 per cent of the sow units; 14 serotypes were detected in the finishing pigs and 11 in the sows. More than 30 per cent of the strains were resistant to tetracycline, sulphonamides, ampicillin or streptomycin, and 69 per cent of the strains were resistant to three or more agents up to 10 compounds. Seventy-seven per cent of the farms had at least one seropositive animal, and 26 per cent of these farms had an individual seroprevalence of 50 per cent or more. The factors associated (P<0.05) with the excretion of Salmonella species in the finishing units were the practice of raising livestock other than pigs (odds ratio [OR]=6.18), the herd size (OR=5.87), and a past history of clinical salmonellosis (OR=4.97). For the sows, the factors associated (P<0.05) with the excretion of Salmonella species were having open-flushed drainage of sewage (OR=34.48), a lack of rodent control measures (OR=0.05) and the number of sows in the unit (OR=9.26). Factors associated with seropositivity in the finishing units were a lack of bird-proof nets (OR=0.30) and the use of water from private wells (OR=3.64).  相似文献

The national control programme for Salmonella in Danish swine herds introduced in 1993 has led to a large decrease in pork-associated human cases of salmonellosis. The pork industry is increasingly focused on the cost-effectiveness of surveillance while maintaining consumer confidence in the pork food supply. Using national control programme data from 2003 and 2004, we developed a zero-inflated binomial model to predict which farms were most at risk of Salmonella. We preferentially sampled these high-risk farms using two sampling schemes based on model predictions resulting from a farm's covariate pattern and its random effect. Zero-inflated binomial modelling allows assessment of similarities and differences between factors that affect herd infection status (introduction), and those that affect the seroprevalence in infected herds (persistence and spread). Both large (producing greater than 5000 pigs per annum), and small herds (producing less than 2000 pigs per annum) were at significantly higher risk for infection and subsequent seroprevalence, when compared with medium sized herds (producing between 2000 and 5000 pigs per annum). When compared with herds being located elsewhere, being located in the south of Jutland significantly decreased the risk of herd infection, but increased the risk of a pig from an infected herd being seropositive. The model suggested that many of the herds where Salmonella was not detected were infected, but at a low prevalence. Using cost and sensitivity, we compared the results of our model based sampling schemes with those under the standard sampling scheme, based on herd size, and the recently introduced risk-based approach. Model-based results were less sensitive but show significant cost savings. Further model refinements, sampling schemes and the methods to evaluate their performance are important areas for future work, and these should continue to occur in direct consultation with Danish authorities.  相似文献

This article describes the assessment of consumer risks of residues of tetracyclines in slaughter pigs in the Netherlands. The assessed risks were toxic and allergic reactions, and the disturbance of the consumers' intestinal flora. Toxic and allergic reactions in humans and animals have only been observed at therapeutic doses, affecting between an estimated 1 in 5,000 and one 1 in 140,000 individuals exposed. Residues of tetracyclines in pigs are closely associated with treatment with injectable formulations. Established Maximum Residue Levels (MRLs) do not reflect actual consumer risks in case a limit is violated incidentally. For example, when the established MRLs for tetracyclines in meat are exceeded with a factor 400, 40,000, and 200,000, respectively, the actual risk of an adverse drug reaction for the consumer following a single consumption of this meat is maximally 1 in 3 million, 1 in 300,000, and 1 in 8,000, respectively. At the current estimated low levels of incidental exposure via pork, the annual risk of negative health effects for a random consumer is estimated at maximally 1 in 33 million. The annual risk that a temporary disturbance of the intestinal flora may also result in a facilitated infection with certain enteropathogens, such as Salmonella spp., is estimated at 1 in 45 million. It is concluded that the current microbiological risks of pork are greater than the risks of residues of tetracyclines as such, and that the control of the microbiological risks of pork should therefore be given first priority.  相似文献

The degree of resistance to Salmonella choleraesuis infection in a reference family purposely bred to map resistance genes was assessed. Aspects of the innate and specific immune system were studied to find a parameter that might predict the resistance of pigs to salmonellosis. The family was bred from commercial full-sister pairs of F1-gilts and four boars. One boar (G398) was identified as breeding susceptible offspring, and one boar (G402) as breeding resistant offspring on the basis of pyrexial responses and numbers of Salmonella in liver and spleen post mortem. The other two boars were classified as 'possible resistant' (Y2008) and 'unknown' (Y6101) respectively. Functional differences in immune cells (neutrophils and lymphocytes) between the offspring of G398 and G402 were detected. The most resistant piglets had a higher number of circulating neutrophils and better polymorphonuclear neutrophils (PMNs) function, but a lower mitogenic response of lymphocytes both pre- and post-infection and a lower antibody response. Between the offspring groups of Y2008 and Y6101 no differences were found in the number of viable Salmonella in liver and spleen at post mortem or in immune cell function, however, the survival rate of these offspring groups was clearly different. Twenty three percent of the Y2008-offspring and 33% of the Y6101-offspring reached the predetermined humane clinical endpoint before the end of the experiment. Our findings suggest a role for several inherited immunological traits, including PMN function and lecithin-induced mitogenic proliferation, which appear to influence resistance to salmonellosis.  相似文献
