Twenty four school children of 7–9 years of age were divided into two groups of six boys and six girls each. One group was given a daily supplement of Suji halwa, a sweet snack made with semolina and red palm oil, supplying 2400 µg of -carotene and the second group was the control group which was given 600 µg of oral vitamin A palmitate, for 60 days. Vitamin A status before and after supplementation was assessed by the Modified Relative Dose Response Assay (MRDR). Results indicated that serum vitamin A levels increased from the basal level of 0.86±0.13 µmol/l to 1.89±0.23 µmol/l in the Red Palmoil (RPO) group and from 0.74±0.09 to 1.94±0.21 µmol/l in the control vitamin A group. Dehydroretinol/Retinol (DR/R) ratio decreased from 0.073±0.025 to 0.023±0.004 in the RPO group and from 0.090±0.023 to 0.023±0.004 in the vitamin A group, indicating liver saturation with vitamin A after feeding RPO snacks, comparable to synthetic vitamin A. This study indicates that RPO is an efficient source of -carotene which is found to be bioavailable in all the subjects tested, hence it can be used for supplementary feeding programmes to combat vitamin A deficiency in target population.  相似文献

The content of campesterol, stigmasterol, and β-sitosterol in raw and cooked Big Flint Maize (BFM), Pop Corn Maize (PCM) and Red Maize (RM) were analyzed by using a chemical method and using in vitro digestion of cooked samples. RM contained 15.14, 8.36, and 72.19 mg/100 g campesterol, stigmasterol, and β-sitosterol, respectively, which were higher compared to in BFM and PCM. Samples cooking resulted in 6–48% increase in phytosterol content, while cooked RM had the highest phytosterol content. Using the in vitro digestion method, 44–61% reduction in phytosterol content was observed in contrast to the chemical method. The difference bioaccessible campesterol and β-sitosterol contents among varieties was insignificant, except for stigmasterol in RM. The bioaccessibility of campesterol was 44–56% higher than those of stigmasterol and β-sitosterol, and it was higher in BFM and PCM. In conclusion, phytosterol contents estimated in different maize samples using in vitro digestion can be another determinative for the ultimate content and reveals the necessity to consider with chemical method of determination.  相似文献

The stress relaxation behaviour of barley kernels with differences in β-glucan was studied using the generalized Maxwell model for explaining the differences in quality. The relaxation moduli show that cultivar significantly affected the relaxation times (τ2 and τ3) and compressional viscosity (ηE2 and ηE3) of the 3 Maxwell elements and the pure elastic component (E0). These data were twice as high in cultivars with high β-glucans compared to low β-glucans. Protein and malt extract, as well as β-glucan, showed similar patterns to the corresponding cultivars from the same locations. Malt extract was correlated with both insoluble fibre (−0.71; P < 0.0007) and total fibre (−0.66; P < 0.006). High β-glucan cultivars presented relatively high soluble fibre, wort viscosity and presented higher relaxation times. Soluble fibre and the wort viscosity were correlated with relaxation times τ1, τ2, τ3. Also, β-glucans were correlated with τ3 and viscosity ηE3. The overall importance of the effect of β-glucans on viscoelasticity, malting and food quality as shown in this investigation is in agreement with earlier investigations.  相似文献

Nowadays, consumers demand dietary fibre-enriched products of appropriate taste, texture, smell and appearance. Unfortunately, addition of the dietary fibre supplements to bread significantly reduces its quality which is connected with changes in the structure of gluten proteins. Structural changes as well as changes in the water state of gluten matrix induced by eight dietary fibres were observed by using Fourier transform infrared spectroscopy. To facilitate this the difference spectra were calculated by subtraction of the control (gluten only) infrared spectrum from the spectra of gluten-fibre mixtures. The presence of positive bands at ca. 1597 and 1235 cm−1 indicated aggregation of gluten proteins into hydrogen bonded β-sheets. These β-sheets can be formed by other β-sheets, antiparallel-β-sheets, β-turns and/or α-helices. The aggregation is probably induced by partial dehydration of gluten matrix due to competition for water molecules between gluten proteins and fibre polysaccharides. This assumption is confirmed by the presence of the negative band at 3237 cm−1 and decrease in the intensity of the band at 3051 cm−1. These bands are assigned to the weak and strong H-bonds in the gluten matrix, respectively. The results indicated that both weak and strong H-bonds are necessary to dough formation of adequate rheological properties.  相似文献

Depolymerization of mixed linkage cereal β-glucans has been observed during the mixing and fermentation of multigrain breads. To maintain the bioactivity of β-glucans in bread and other foods, control of β-glucanase activity during production is needed. β-Glucanase activity in wheat and dairy ingredients was determined by adding them to β-glucan solutions and following the change in the molecular weight of β-glucan at 50 °C for 60 min. The activities in six wheat and six dairy ingredients ranged from undetectable to a 98% reduction in molecular weight. Calcium propionate, potassium sorbate, and sodium benzoate are salts of organic acids commonly used as food preservatives. Low concentrations of these additives (0.1–1.0%) were found to have an inhibitory effect on the β-glucanases found in wheat flour and whey protein isolate. The depolymerization rate of β-glucan was reduced by up to 76% by the three additives.  相似文献

Three hull-less barleys, Washonupana (WSNP), Waxbar (WXB), and Bangsa (BGS), were fed to broiler chicks in 21% protein diets containing 0.5% cholesterol in replicate trials. A corn-based diet, with added cholesterol, served as a control. Alternate diets were supplemented with -glucanase (ENZ). -glucan content ranged from 4.9% to 6.1% and soluble dietary fiber (SDF) from 3.6% to 7.5% in the barleys. Data from the two trials were pooled for statistical analysis by the SAS General Linear Models procedure. In body weight gain, chicks fed WSNP — ENZ were lower (P<0.05) than all other treatments. The -glucanase supplement to the WXB and BGS barley tended to improve gains, but the differences were not significant for either barley. Feed to gain ratios were lowest (P<0.0001) for corn fed chicks and lower (P<0.05 toP<0.0001) for those fed the barley+ENZ diets compared to barley –ENZ. Chicks fed barley diets had lower (P<0.05) total serum cholesterol (TSC) and LDL-cholesterol than those fed corn diets, regardless of ENZ supplementation. For chicks on barley –ENZ diets, TSC levels for WSNP, WXB, and BGS were 146, 152, and 142 mg/dl respectively and for chicks on barley +ENZ diets, 218, 200, and 178 mg/dl. LDL-cholesterol levels followed the same trend and there was little difference in serum triglycerides. The BGS+ENZ lowered TSC 30% from the corn control compared to 10.7% and 18% for WSNP+ENZ and WXB+ENZ, suggesting additional hypocholesterolemic factors, possibly tocotrienol and SDF other than 13, 14 -D-glucans.Contribution No. J-2487, Montana Agricultural Experiment Station. The barley mutant TR49 and the cultivar from which it was derived, Troubadour, were grown in Scotland and Spain and harvested in 1991 and 1992. The development of β-glucanase together with the breakdown of β-glucan and the reduction in milling energy were followed during malting. While similar amounts of β-glucan were found in samples from both sites, TR49 showed less response to environmental conditions. Grain from Spain showed higher levels of β-glucanase activity, differences being apparent after steeping and 1–2 days of germination. The milling energy of grain was the same, but, during malting, samples from Spain showed a greater reduction in milling energy than those from Scotland. The most extreme environment in the study was the 1991 season at the Spanish site and was the driest and hottest. But even in the cooler, wetter season of 1992, Spanish grown material showed a greater production of β-glucanase than Scottish grown samples. These findings have important implications for plant breeding as well as the growing and trading of malting barley on a European scale.  相似文献

The polymorphism of barleyalpha-amylases, as revealed by isoelectric-focusing (IEF), was studied in a population of 126 doubled-haploids from a cross between the cultivars Morex and Steptoe. In particular, the pattern was determined for an additional low pI alpha-amylase isozyme detected in cultivar Steptoe. No significant effect of this isozyme onalpha-amylase activity was observed, but a significant effect on β-glucanase activity was detected for two out of four trial locations. Quantitative Trait Loci (QTL) foralpha-amylase and β-glucanase activities were also investigated. The results suggest a potential location for the β-glucanase locusGlb2, on chromosome 1 close to theAmy2locus foralpha-amylase genes.  相似文献

Barley (Hordeum vulgare L.) with high grain β-glucan content in the soluble dietary fiber fraction may be useful as a specialty crop for human food. In contrast, low β-glucan content is desirable for brewing or animal feed. The objective of this study was to evaluate the effect of nitrogen and irrigation on barley grain β-glucan content. Relationships between barley grain β-glucan content and some agronomic traits were examined. Generally, high nitrogen levels increased barley grain β-glucan content in both years. The N2 treatment gave the best grain β-glucan contents. A negative effect of irrigation was observed for barley grain β-glucan content. Increased levels of irrigation tended to decrease grain β-glucan content in barley. Non-irrigated treatments commonly had the greatest values for grain β-glucan content in all treatments. Higher positive correlations between β-glucan and protein content were observed in both years. A negative correlation between β-glucan content and 1000 grain weight was significant in 1999 while a negative correlation between β-glucan content and sieve analysis was significant in 2000. It may be concluded that increased nitrogen is desirable for high grain β-glucan content in barley but not irrigation.  相似文献

Alpha6beta2 nicotinic acetylcholine receptors (nAChRs) are potential therapeutic targets for the treatment of several neuropsychiatric diseases, including addiction and Parkinson’s disease. Alpha-conotoxin (α-CTx) TxIB is a uniquely selective ligand, which blocks α6/α3β2β3 nAChRs only, but does not block the other subtypes. Therefore, α-CTx TxIB is a valuable therapeutic candidate peptide. Synthesizing enough α-CTx TxIB with high yield production is required for conducting wide-range testing of its potential medicinal applications. The current study optimized the cleavage of synthesized α-CTx TxIB resin-bounded peptide and folding of the cleaved linear peptide. Key parameters influencing cleavage and oxidative folding of α-CTx TxIB were examined, such as buffer, redox agents, pH, salt, co-solvent and temperature. Twelve conditions were used for cleavage optimization. Fifty-four kinds of one-step oxidative solution were used to assess their effects on each α-CTx TxIB isomers’ yield. The result indicated that co-solvent choices were particularly important. Completely oxidative folding of globular isomer was achieved when the NH4HCO3 or Tris-HCl folding buffer at 4 °C contained 40% of co-solvent DMSO, and GSH:GSSG (2:1) or GSH only with pH 8~8.7.  相似文献

Some locally available fruits and vegetables of the arid zone of Rajasthanwere analyzed for their -carotene content; the selected fruits and vegetables were: Dansra (Rhus myserensis), Kachri (Cucumis collosus), fruit Kachri (Cucumis collosus), Kair (Capparisdecidua) and Sanghri (Prosopis cineriria). The -carotene contents of the samples were estimated in fresh and various processed states (blanched, dried and roasted). The results showed the heat lability of -carotene. Among the samples, kair was found to be the richest source of -carotene. Variety, too, had an impact on the content of this fat soluble vitamin.  相似文献

Oat β-glucan has been shown to have cholesterol, insulin and glucose lowering effects, which are related to increased viscosity of the intestinal contents. Some important factors influencing the viscosity are molecular weight, structure and concentration. To study the effect of variety and environment on β-glucan content and molecular weight of β-glucan, four oat varieties were grown in 11 different environments (location × year combinations) in a field experiment. The β-glucan content varied between 2.3 and 3.2% and the average molecular weight of β-glucan between 1.73 and 2.02 × 106 g mol−1. There was a significant difference between varieties and environments for both β-glucan content and molecular weight of β-glucan. The effect of environment was much greater on molecular weight (71%) than on β-glucan content (42%), while the effect of variety was greater on β-glucan content (23%) than on molecular weight (4%). There was also a positive significant correlation between β-glucan content and molecular weight of β-glucan (p < 0.001, r = 0.46). These results show that β-glucan content is a better target for plant breeding than molecular weight. However, both factors are important for the physiological effects of β-glucan, and selection for high β-glucan content will probably also give higher molecular weight.  相似文献

Neem seeds contain many substances with insecticidal properties, the main insecticidal ingredient being azadirachtin A. In developing countries such as Mali, a neem seed water extract is prepared by soaking ground seeds in water for three or seven days. The aim of this study was to check the effectiveness of this extract in terms of azadirachtin A extraction yield and insecticidal activity. The yield of extraction was 0.19 g azadirachtin A/100 g seeds. The concentration of azadirachtin A in the seed extract was approximately 200 mg l−1, eight times higher than the recommended concentration of commercial products (25 mg l−1). A comparison of the extractive capacity of different solvents indicated that the best solvents were water and methanol. The azadirachtin A concentration declined in extracts stored for more than 3 days at a temperatures higher than 30 °C. Bioassays were performed on target insects (the leafhopper Macrosteles quadripunctulatus, the moth Spodoptera littoralis and the tobacco whitefly Bemisia tabaci) in order to compare the insecticidal activity of the neem extract with that of the commercial product Neemazal T/S and of a solution of pure azadirachtin A. The bioassays conducted on the leafhopper and the moth demonstrated that the neem extract at the recommended concentration (25 mg l−1 active ingredient) was as effective as the azadirachtin-based commercial product at the same concentration, while for the control of the whitefly B. tabaci a higher concentration of the water extract was needed.  相似文献

Oat malt is a nutritionally rich ingredient mainly used in a small number of speciality products. The aim of this study was to evaluate the suitability of oat malt in wheat baking. The effect of oat malt on bread and dough properties at levels ranging from 0.5% to 5% was studied and compared with barley and wheat malts. The addition of all malts increased loaf specific volumes. Barley and wheat malts at levels above 2.5% led to a sticky and coarse crumb, but the effect of oat malt on the crumb grain was negligible. Rheological characterisation could not explain the superior baking performance of oat malt, as it increased extensibility and decreased resistance extensively indicating weakening of the extensional properties of the gluten network. The high lipolytic activity may have compensated for the loss of dough strength by improving the surface properties of gas cells. The results show that oat malt can be used in wheat baking to improve the loaf volume and nutritional quality without the detrimental effects associated with the excess amylolytic activity of barley and wheat malts.  相似文献
