侵染肥城桃的病毒和类病毒的分子检测与鉴定   总被引:1,自引:0,他引:1  
为明确山东肥城桃种植区桃树上主要存在的病毒和类病毒及其发生情况,采集具有花叶、斑驳和皱缩典型症状的肥城桃样品,提取叶片总RNA后,分别选用桃树上已报道的啤酒花矮化类病毒Hopstuntviroid(HSVd)、桃潜隐花叶类病毒Peach latent mosaic viroid(PLMVd)、苹果褪绿叶斑病毒Apple chlorotic leaf spot virus(ACLSV)、樱桃锉叶病毒Cherry rasp leaf virus(CRLV)、桃花叶病毒Peach mosaic virus(PMV)、李属坏死环斑病毒Prunus necrotic ringspot virus(PNRSV)、李痘病毒Plum pox virus(PPV)、李矮缩病毒Prunus dwarf virus(PDV)、樱桃绿环斑驳病毒Cherry green ring mottle virus(CGRMV)、杏假褪绿叶斑病毒Apricot pseudo-chlorotic leaf spot virus(APCLSV)、李树皮坏死茎纹孔伴随病毒Plum bark necrosis stem pitting-associated virus(PBNSPaV)和小樱桃病毒1号Little cherry virus 1(LchV1)的特异性引物进行RT-PCR检测。PCR结果显示仅HSVd、PLMVd、ACLSV、PNRSV和PBNSPaV的扩增产物中得到了预期大小的目的片段,将目的片段克隆测序后,经NCBI BLAST比对发现,山东肥城桃分离物HSVd、PLMVd、ACLSV、PNRSV和PBNSPaV与GenBank已报道分离物序列一致性均达90%以上。表明山东肥城桃已感染HSVd、PLMVd 2种类病毒和ACLSV、PNRSV、PBNSPaV 3种病毒。  相似文献

Farmers' field survey data sets represent extremely valuable information, often having a heterogeneous data format. A large number of methods are available to process this kind of information, which may be combined to address successive, connected hypotheses, with definite objectives. The National Grapevine Trunk Diseases Survey was established in France in order to monitor and analyze the importance and progress of several grapevine diseases, in particular Eutypa dieback and Esca decline. Here we report a first series of analyses pertaining to the years 2003 to 2005 to characterize the incidences of the two diseases, as well as grapevine mortality, in relation to grapevine age, crop management, cultivars, and growing regions. Information representing 256 individual vineyards indicated mean incidences of 2.23 and 3.25% for Eutypa dieback and Esca decline, respectively. A combination of approaches (hierarchical cluster analysis, multiple correspondence analyses, and binomial logistic regressions) indicated distinct patterns of variation in the incidences of the two diseases according to crop management practices; limited linkage of specific crop management practices with diseases, especially Esca decline; a shared contribution of both diseases to mortality; and a weak linkage of the (now banned) arsenite-based pesticides with reduced Esca decline symptoms, but no such association with grapevine mortality. Differences and complementarities of the analytical approaches are discussed, as well as the implications of these analyses on grapevine health.  相似文献

The occurrence of grapevine leafroll-associated virus 1 (GLRaV-1), grapevine leafroll-associated virus 3 (GLRaV-3) and grapevine virus A (GVA) was demonstrated in a viticultural region of northern Italy (Emilia-Romagna) using immunoelectron microscopy. Virus incidence was subsequently assessed using ELISA. A total of 60.6% of the 150 clone selections tested, from 18 local Vitis vinifera cultivars, were found to be infected. ELISA did not reveal the presence of grapevine leafroll-associated virus 2 (GLRaV-2) or grapevine leafroll-associated virus 5 (GLRaV-5). GLRaV-1, GLRaV-3 and GVA were found individually and in various combinations. The most common findings were GLRaV-1 alone (25.3%) and associated with GVA (33%). Serological data confirmed that the majority (91%) of the clones known to be affected by grapevine leafroll (GLR), on its own or in association with rugose wood (RW), contained viruses. On the other hand, where the RW phenomenon was present on its own, only 40% of these clones were ELISA-positive. The implications for the biology of GLR and RW are discussed and the complex aetiology of these grapevine diseases is confirmed.  相似文献

 Zucchini yellow mosaic virus (ZYMV) was detected by RT-PCR from pumpkin (Cucurbita moschata) plant showing yellowing and mosaic symptom from Liaocheng, Shandong Province. The 3'-termial 1 684 bp genomic sequence covered 633 bp of NIb encoding sequence, 840 bp of cp gene and 211 bp of 3'-untranslated region of the isolate ZYMV-Liaocheng was determined. The cp gene of ZYMV-Liaocheng shared identities of 81.4%-98.8% and 89.4%-99.5% at nucleotide and amino acid levels, respectively, with other ZYMV sequences available in the GenBank. Phylogenetic analysis indicated that ZYMV could be clustered to 6 genotypes. ZYMV-Liaocheng belonged to genotypeⅠ, which contained isolates from Asia, Europe and America. Genotypes Ⅲ and Ⅴ were unique and contained only isolates from East Asia. The isolates from East Asia had the highest variability.  相似文献

During the spring of 2001, approximately 10 000 yellow passion flower plants, from two orchards in the county of Livramento de Nossa Senhora, Bahia State, Brazil, exhibited intense yellow mosaic symptoms and drastic reduction of the leaf lamina and plant development. A large population of whiteflies ( Bemisia tabaci ) was also found colonizing the plants. All field samples collected tested positive for Passion fruit woodiness virus in DAS-ELISA. Five out of 20 passion flower plants inoculated with adult whiteflies collected from diseased plants in the field developed symptoms 20–30 days after inoculation. Two of these plants gave a positive reaction in TAS-ELISA using antiserum against a begomovirus. Degenerated PCR primers amplified viral DNA fragments from the DNA-A and DNA-B components of a begomovirus infecting these plants. The fragment corresponding to the core region of the coat protein (DNA-A) was cloned and sequenced. A phylogenetic analysis placed this begomovirus isolated from passion flower in the same clade of the New World begomoviruses as several other species from Brazil. Based on the symptoms induced by this virus alone, the disease was tentatively named passion flower little leaf mosaic.  相似文献

 从新疆加工番茄上分离到病毒分离物XJ26-4,对其基因组DNA-A 全序列测定表明,XJ26-4 DNA-A 全长2 737 个核苷酸(GenBank 登录号:FN985163),具有典型的双生病毒基因组特征。进一步序列比较发现,XJ26-4 DNA-A 与中国番茄黄化曲叶病毒(Tomato yellow leaf curl China virus, TYLCCNV)各分离物的同源性最高,达到91. 2% ~ 99. 5% ,而与其他双生病毒的序列相似性均在79. 5% 以下,表明XJ26-4 是TYLCCNV 的一个分离物。这是首次明确新疆加工番茄受到粉虱传双生病毒的侵染。  相似文献

Eutypa lata is a vascular pathogen of woody plants. In the present study we (i) determined which component(s) of the cell wall polymers were degraded in naturally infected grapevines and in artificially inoculated grape wood blocks; (ii) compared the pattern of wood decay in the tolerant grape cv. Merlot versus the susceptible cv. Cabernet Sauvignon; and (iii) identified secondary metabolites and hydrolytic enzymes expressed by E. lata during wood degradation. Biochemical analyses and a cytochemical study indicated that glucose-rich polymers were primary targets of E. lata. Structural glucose and xylose of the hemicellulose fraction of the plant cell wall and starch were depleted in infected woods identically in both cultivars. Moreover, the more tolerant cv. Merlot always had more lignin in the wood than the susceptible cv. Cabernet Sauvignon, indicating that this polymer may play a role in disease resistance. In vitro assays demonstrated the production by E. lata of oxidases, glycosidases and starch degrading enzymes. Phytotoxic secondary metabolites were also produced but our data suggest that they may bind to the wood. Finally, we demonstrated that free glucose in liquid cultures repressed primary but not secondary metabolism.  相似文献

杂草是菜豆金色花叶病毒属病毒的重要中间寄主,常富集多种该属病毒及其卫星病毒。2014年,在云南红河常见杂草苣荬菜Sonchus arvensis上出现了疑似菜豆金色花叶病毒属病毒病症状。利用克隆、测序和生物信息学分析技术对其所含病毒进行分离鉴定,结果从1株病样中共获得了两条菜豆金色花叶病毒属病毒全序列、两条β卫星全序列和一条α卫星全序列。序列分析显示,两条菜豆金色花叶病毒属病毒全序列与中国胜红蓟黄脉病毒相似性最高,分别为99%和96%,确定为中国胜红蓟黄脉病毒的分离物。两条β卫星全序列与赛葵黄脉β卫星相似性最高,为97%,确定为赛葵黄脉β卫星的一个分离物。α卫星全序列与中国番茄黄化曲叶α卫星相似性最高,为86.3%,是中国番茄黄化曲叶α卫星的一个分离物。这是菜豆金色花叶病毒属病毒病害复合体在中国侵染苣荬菜的首次报道。  相似文献

A survey for the incidence of Hop stunt viroid (HSVd) infection in China was conducted using dot-blot hybridization. Out of 553 tested samples, 127 samples of stone fruits (apricot, peach, plum and almond), grapevine and hop were positive for HSVd, giving a mean infection rate of 23?%. Phylogenetic analysis revealed that most of the HSVd variants isolated from stone fruits, grapevine and hop were clustered into known hop and plum groups. However, two grapevine variants, HSVd.g50 and HSVd.g57, could not be clustered into any known groups, indicating a previously unknown phylogenetic group of HSVd isolates. HSVd.g38 was the single grapevine variant that clustered with the plum group isolates, supporting cross transmission between grapevine and stone fruits and the heterogeneity of grapevine isolates.  相似文献

Resistance of Botrytis cinerea to benzimidazoles is present in Italy in almost all vineyards monitored, while resistance to dicarboximides has been detected in 36, 27 and 49% of tested vineyards during 1981, 1982 and 1983, respectively. In 1981 less than 1% of tested bunches carried dicarboximide-resistant conidia. This value increased slowly with time: in 1983 4.9% of the tested bunches carried dicarboximide-resistant conidia. Furthermore, only a low percentage (< 10%) of the conidia obtained from each bunch were resistant. Therefore, the present situation of resistance to dicarboximides on grapevine in Italy is certainly not serious and differs from that reported in other countries or on other crops. As a consequence, dicarboximides are still performing well, even in vineyards where resistance has been detected at a low level. No failure of grey mould control has been reported on grapevine in Italy up to now.  相似文献

Three experimental trials were carried out in Northern Italy during 1985 and 1986 in order to control grey mould of grapevine (Botrytis cinerea) by using isolates of Trichoderma spp. resistant to several fungicides commonly sprayed against grapevine pathogens, alone or in alternation with benzimidazoles or dicarboximides, in vineyards where fungicide-resistant strains of B. cinerea are frequent. The antagonists alone partially controlled the pathogen on cv. Moscato ?Asti. In one case, the integration of chemical and biological control measures showed slightly better results than for the fungicide alone (for benomyl but not for vinclozolin), but further trials are needed to investigate the full potential for using fungicide-resistant Trichoderma in alternation with fungicides. Trichoderma spp. performed very poorly on cv. Barbera.  相似文献
