大气苯酚对蓖麻蚕的影响及其对策   总被引:3,自引:0,他引:3  
蓖麻接触15ul/L大气苯酚2-10h后叶片出现伤斑、失绿、枯焦卷曲等受害症状,接触6h后株高、条重、产叶量以及叶绿素、粗蛋白和可溶性糖含量分别下降9.9%、11.8%、9.5%、34.7%、3.8%和42.8%。全龄蓖麻蚕接触1ul/L大气苯酚或食下苯酚含量7-21mg/kg的污染叶后均影响其虫蛹率、产孵量及孵化率;接触8ul/L以上的大气苯酚或食下苯酚含量45-70mg/kg的污染叶后吞生长发育变慢,生命力和生殖能力下降,茧丝性状刀明显不良,大气苯酚在这些方面的影响比苯酚污染叶大,蓖麻蚕对大气苯酚的抵抗力随龄期增大而显著增强。用5%石灰水浸渍苯酚污染叶后喂蚕,或将苯酚污染叶与蛎污染叶交替喂蚕,可减轻苯酚对蚕的毒害。  相似文献

Antioxidative compounds were isolated from the 50% methanol extract of dried leaves of Celastrus hindsii. Eight phenolic compounds (1-8) were finally obtained by reversed-phase high-performance liquid chromatography, and their structures were elucidated by nuclear magnetic resonance spectrometry and mass spectrometry analyses. They were the five known compounds, rutin (1), kaempferol 3-rutinoside (2), rosmarinic acid (3), lithospermic acid (4), and lithospermic acid B (6), and three novel oligomers of rosmarinic acid, a dimer (5) and two trimers (7 and 8). The major components in the extract were rosmarinic acid (3) and lithospermic acid B (6). These phenolic compounds were shown to have antioxidative activities against the autoxidation of methyl linoleate in bulk phase and the radical-initiated peroxidation of soybean phosphatidylcholine in liposomes. In the liposomal peroxidation, the number of phenolic hydroxyl group in each molecule was correlated with the effectiveness of antioxidative activity.  相似文献

A new secoiridoid compound was isolated from the leaves of Olea europaea. This compound, not previously identified, is the bis methylacetal of oleuropein aglycone, the 3,4-dihydroxyphenylethyl [(2,6-dimethoxy-3-ethylidene)-tetrahydropyran-4-yl]acetate (3,4-DHPEA-DETA), and was found in different olive cultivar phenolic extracts as one of the major secoiridoid components. This compound was shown to be easily transformed in acidic aqueous media into 3,4-DHPEA-EDA, the major polyphenolic compound found in olive oil, and permitted us to increase the yield of 3,4-DHPEA-EDA isolation from the olive leaf extract. The antiradical activity of this new compound, evaluated by scavenging of 2,2-diphenyl-1-picrylhydrazyl radicals, was much higher than the one found for 3,4-DHPEA-EDA or alpha-tocopherol. Results also call to attention the need for a careful identification of compounds by HPLC-MS, usually performed in acidic conditions.  相似文献

5-(4,5-Dihydroxy-2-imino-1-imidazolidinyl)norvaline (1) was identified as the only product of the early reaction of arginine with glyoxal, which was slowly degraded to N(5)-[[(carboxymethyl)amino](imino)methyl]ornithine (3, N(7)-carboxymethylarginine). No other structures could be detected within a range of pH 4-8 and 20-50 degrees C in reaction conditions. The rates of formation for both products increased with pH and temperature. In equilibrium, the vincinal diol groups of 1 were 86% trans configured. The formation of 1 was reversible, as could be shown by cis-trans isomerization of the separated isomers and by regeneration of arginine in the presence of the alpha-dicarbonyl trapping reagents, o-phenylenediamine and aminoguanidine. Both 1 and 3 were converted to 5-(2-imino-5-oxo-1-imidazolidinyl)norvaline (2) only under strong acidic conditions.  相似文献

生态环境对白肋烟上部叶的品质和中性香气成分的影响   总被引:1,自引:0,他引:1  
为了探索生态环境对白肋烟晾制品质的影响,将云南省宾川县和云龙县生产的白肋烟异地互换晾制。调查了白肋烟调制期间的温、湿度,并采用高效液相色谱、气质联用仪测定调制结束后烟叶中的质体色素和中性香气成分的含量。结果表明,两地生态条件存在差异,湿度差异明显,温度差异较小,整个白肋烟晾制期间,云龙县空气相对湿度较宾川县高4.42%~16.54%。宾川县晾房烟叶晾制时间比云龙县晾制的烟叶少9 d。两个生态环境下,烟叶的物理外观和内在品质差异显著。宾川县晾制的云龙县生产的烟叶β-类胡萝卜素的残留量比宾川县生产的烟叶高6.44%,而云龙县晾制的宾川县生产烟叶比云龙县生产烟叶高14.45%。在云龙县晾制的烟叶中性香气成分含量比在宾川县晾制的高,宾川县生产烟叶和云龙县生产烟叶相应高出47.31%和20.37%。同一产地烟叶,烟碱和总糖含量表现为在宾川县晾制的烟叶极显著高于在云龙县晾制的烟叶;而总氮含量均表现为在云龙县晾制的烟叶极显著高于宾川县晾制的烟叶。生态环境与烟叶中的质体色素、中性香气成分、化学成分含量密切相关。因此,在白肋烟生产中,应因地制宜,科学调制白肋烟,提高烟叶质量,增加烟叶香气。  相似文献

Engeletin, a flavonoid compound, was isolated from the leaves of Engelhardia roxburghiana for the first time, along with astilbin, another flavonoid. The chemical structures of engeletin and astilbin were confirmed by (1)H and (13)C nuclear magnetic resonance (NMR) and mass spectrometry (MS) spectra, and their anti-inflammatory activities were studied in lipopolysaccharide (LPS)-stimulated mouse J774A.1 macrophage cells. LPS induced the inflammatory state in macrophage cells and increased mRNA expressions of pro-inflammatory cytokines. Engeletin and astilbin exhibited remarkable inhibitory effects on interleukin (IL)-1β and IL-6 mRNA expression. Significant inhibition of LPS-mediated mRNA expressions were also seen in LPS binding toll-like receptor (TLR)-4, pro-inflammatory cytokine tumor necrosis factor (TNF)-α, IL-10, chemoattractant monocyte chemotactic protein (MCP)-1, and cyclooxygenase (COX)-2 genes. The reduced expression of these cytokines may alleviate immune response and reduce inflammatory activation, indicating that engeletin and astilbin may serve as potential anti-inflammatory agents.  相似文献

Patatin has, so far, been considered a homogeneous group of proteins. A comparison of the isoforms in terms of structural properties or stability has not been reported. A method to obtain various isoform fractions as well as a comparison of the physicochemical properties of these pools is presented. Patatin could be separated in four isoform pools, denoted A, B, C, and D, representing 62%, 26%, 5%, and 7% of the total amount of patatin, respectively. These isoforms differed in surface charge, resulting in a different behavior on anion exchange chromatography, isoelectric focusing, native polyacrylamide gel, and capillary electrophoresis. All isoforms of the patatin family contained proteins with two molecular masses of approximately 40.3 and 41.6 kDa, respectively. The size of this difference in the molar mass (1300 Da) is on the order of one carbohydrate moiety. Despite the biochemical differences given above, no variations in the structural properties nor in the thermal conformational stability could be observed using far-ultraviolet circular dichroism, infrared, and fluorescence spectroscopy.  相似文献

Storage of olive (Olea europaea) leaves for 22 h at 37 degrees C in closed plastic bags caused the content of a nonglycosidic secoiridoid, 3,4-dihydroxyphenylethyl 4-formyl-3-formylmethyl-4-hexenoate (3,4-DHPEA-EDA) to rise from 15% to 50% of the phenolic extract with corresponding falls in the content of oleuropein and two oleuropeindials, which were identified as precursors of 3,4-DHPEA-EDA. Pure product was isolated from one set of stored olive leaves in a 0.16% yield. Storage of olive leaves under various conditions showed that the moisture present in closed plastic bags was important for the formation of 3,4-DHPEA-EDA. The time taken to reach the maximum concentration of the product varied widely for different samples of olive leaves, with a shorter time for the sample with lower initial oleuropein content. The oleuropeindial precursors of the product were readily hydrolyzed to carboxylic acid derivatives, which have been identified by NMR. The antiradical activity of 3,4-DHPEA-EDA, evaluated by scavenging of 2,2-diphenyl-1-picrylhydrazyl radicals, was comparable to that of alpha-tocopherol.  相似文献

Two varieties of banana green fruit growing in Guerrero, Mexico, were used for starch isolation. Chemical analysis and physicochemical and functional properties were studied in these starches. The "macho" variety presented higher starch yield than "criollo". In general, chemical compositions in both starches were similar, except in ash content, where the "criollo" variety showed a lower value than "macho". The results of freeze-thaw stability suggested that banana starches cannot be used in frozen products. Both starches presented similar water retention capacity values that increased when temperature increased. Solubility profiles showed that at low temperature "criollo" had lower solubility than "macho", but at higher temperature an inverse behavior was evident; also the solubility increased when temperature increased. Behavior similar to that for solubility was obtained in the swelling test. The banana starch studies indicate the "macho" and "criollo" varieties have different starch structures as evidenced by viscosity.  相似文献

Using reversed-phase high-performance liquid chromatography (RP-HPLC), the peanut protein profile was shown to be related to the maturity, drying time, and drying procedure of the peanut. Differences were seen between (a) immature and mature seeds for untreated and windrow-dried peanuts, (b) untreated and windrow-dried peanuts for immature and mature seeds, and (c) windrow- and stackpole-dried peanuts. The most pronounced HPLC peak that increased in size as the peanut matured and decreased in size with longer drying times was isolated and identified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and electrospray ionization mass spectrometry to have a molecular weight of 62 500. Since maturity is related to the sensory quality of peanuts, this protein may be a marker for peanuts that will produce a higher quality flavor when roasted.  相似文献

The methanolic extract of Piper lolot, having shown potent inhibitory activity on platelet aggregation induced by arachidonic acid (AA) and platelet activating factor (PAF), was subjected to activity-guided isolation to yield twelve new amide alkaloids, piperlotine A-L (1-12), along with twenty-nine known compounds. Their structures were elucidated on the basis of spectroscopic analysis. The isolated compounds were tested for their inhibitory activity on the rabbit platelet aggregation. The compounds piperlotine A (1), piperlotine C (3), piperlotine D (4), piperlotine E (5), 3-phenyl-1-(2,4,6-trihydroxyphenyl)propan-1-one (21), 3-(4-methoxyphenyl)-1-(2,4,6-trihydroxyphenyl)propan-1-one (22), 1-trans-cinnamoylpyrrolidine (24), sarmentine (26), pellitorine (27), methyl 3-phenylpropionate (32), and (10S)-10-hydroxypheophorbide a methyl ester (40) showed potent antiplatelet aggregation activity.  相似文献

This paper reports the first isolation from cultures of two endoxylanases secreted by Fusarium graminearum Schwabe [teleomorph Gibberella zeae (Schweinitz) Petch]. When F. graminearum is grown on wheat bran hydrated with a modified synthetic medium, high xylanase activity can be extracted. The two endoxylanases were identified by LC-MS/MS as the products of genes FGSG_6445 (Genbank gene id 2788192 ) (xylanase 1) and FGSG_3624 (GenBank accession no. AJ863566 ) (xylanase 2) with 61 and 51% sequence coverage, respectively. Both enzymes showed a pH optimum at pH 6, with xylanase 1 exhibiting a wider active pH range (5.5-9) than xlylanase 2 (5.5-7.5). Their temperature dependences were similar, >60% between 35 and 60 °C, with optimal temperatures of 45 °C for xylanase 1 and 50 °C for xylanase 2. Kinetic studies found that both enzymes had a lower K(m) for linear beachwood xylan than arabinoxylan. For xylanase 2, the V(max) increased with arabinoxylan, but decreased for xylanase 1.  相似文献

The metabolism of exogenous glucose-14C in the light and the dark was studied in the detached leaves of tomato plants grown with ammonium nitrogen and nitrate nitrogen. In the light, 14CO2, release and incorporation of glucose into insoluble materials were hardly affected at all by the nitrogen sources. Among the soluble labelled amino acids, serine had the greatest amount of label in the ammonium-plants while aspartate had the greatest amount in the nitrate-plants. This aspartate was synthesized from C3-compounds by carboxylation. During dark-light transition, the change in the composition of soluble amino acids was more rapid in the ammonium-plants than in the nitrate-plants. In the dark, 14CO2-release, which was ten times as much as in the light, was larger in the ammonium-plants than in the nitrate-plants; but the synthesis of high molecular compounds from glucose in the ammonium-plants was about half that in the nitrate-plants. So, it is considered that respiration operates sufficiently in the ammonium-plants. The effects of DCMU and a 100% O2 atmosphere on glucose metabolism in both groups of plants were studied and the respiration of leaves in the light was discussed.  相似文献

Two major superoxide dismutases (SODs; SODs I and II) were found in the crude enzyme extract of wheat seedlings after heat treatment, ammonium sulfate fractionation, anionic exchange chromatography, and gel permeation chromatography. The purification fold for SODs I and II were 154 and 98, and the yields were 11 and 2.4%, respectively. SOD I was further characterized. It was found that SOD I from wheat seedlings is a homodimer, with a subunit molecular mass of 23 kDa. Isoelectric focusing electrophoresis (IEF) and zymogram staining results indicated that the isoelectric point of SOD I is 3.95. It belongs to the MnSOD category due to the fact that it was insensitive to KCN or hydrogen peroxide inhibitor. This MnSOD from wheat seedlings was found to be stable over pH 7-9, with an optimum pH of 8, but was sensitive to extreme pH, particularly to acidic pH. It was stable over a wide range of temperatures (5-50 degrees C). Thermal inactivation of wheat seedling MnSOD followed first-order reaction kinetics, and the temperature dependence of rate constants was in agreement with the Arrhenius equation. The activation energy for thermal inactivation of wheat seedling MnSOD in the temperature range of 50-70 degrees C was found to be 150 kJ/mol. HgCl2 and SDS at a concentration of 1.0 mM significantly inhibited enzyme activity. Chemical modification agents, including diethyl pyrocarbonate (2.5 mM) and Woodward's reagent K (50 mM), significantly inhibited the activity of wheat seedling SOD, implying that imidazole groups from histidine and carboxyl groups from aspartic acid and glutamic acid are probably located at or near the active site of the enzyme.  相似文献

Extraction, purification, and identification procedures were developed for the chemical investigation of molybdenum (Mo) in freeze-dried aerial portions of alfalfa (Medicago sativa L.) collected from a reclaimed tailings pond at the Highland Valley Copper mine in British Columbia. The purification procedures were guided by ICP and colorimetric analyses. The methods included development of an efficient aqueous extraction protocol, sample cleanup by partitioning against n-butanol, and filtration through diatomaceous earth. Further purification was achieved by anion-exchange chromatography, elution with aqueous NaCl, and desalting by gel filtration. Final purification of the Mo-containing fraction was carried out using preparative anion-exchange HPLC. Molybdenum was found to be present in its purified form as the molybdate anion (MoO(4)(2-)) primarily on the basis of multinuclear ((1)H, (13)C, and (95)Mo) NMR studies.  相似文献

Besides the already known stilbenes trans-resveratrol as well as isomeric piceids seven novel stilbene derivatives have been isolated from a commercial Riesling wine. The newly identified compounds included the monostilbene 2,4,6-trihydroxyphenanthrene-2-O-glucoside, as well as two isomeric resveratrol-2-C-glucosides. In addition, four dimeric stilbenes, i.e., cis- and trans-epsilon-viniferin diglucoside as well as pallidol glucoside and pallidol diglucoside, have also been obtained for the first time from Riesling wine.  相似文献

Previous work has shown that red wines, grape juices, and other grape products cause endothelium-dependent relaxation (EDR) of blood vessels in vitro by increasing nitric oxide production. In this paper we describe the isolation and characterization of some of the compounds responsible for EDR activity. Concord grape seeds were extracted with methanol and the compounds were separated by Toyopearl TSK HW-40S chromatography. Resulting fractions (primarily phenolic acids, catechins, and proanthocyanidins) were further separated semipreparatively by reversed-phase HPLC, and peaks were collected and bioassayed for EDR activity using the rat aorta preparation. EDR-active compounds were subsequently characterized by HPLC retention times and electrospray-ion-trap mass spectrometry. The compounds exhibiting the most EDR activity were proanthocyanidin trimers, tetramers, pentamers, and polymers and their gallates, as well as a dimer gallate (EC50 values in the range of 0.6-2.5 microg catechin equivalents/mL). These compounds should be useful for in vitro and in vivo studies, particularly as they relate to improvement of cardiovascular function.  相似文献

多环芳烃(Polycyclic Aromatic Hydrocarbons,PAHs)是土壤中一种典型的持久性有机污染物。典型寒区东北地区因农业投入品不合理使用、污灌等造成农田土壤中含有大量致癌、致畸与致突变的多环芳烃,针对寒区气候特征致使农田中微生物降解多环芳烃效果不佳,难以改善农田土壤环境并降低食品风险的关键问题,基于目前常用商品化的降解微生物多来自于温暖地区,且难以适应寒区气候的特性,该研究以PAHs典型污染物-菲(PHE)为研究底物,驯化温度15 ℃,筛选分离出适应寒区低温环境的7株菌。经鉴定及降解性能研究,筛选了3株在温度20 ℃、接种量5%、pH值为8、底物浓度500 mg/L,以及外源物质腐殖酸的促进的条件下PHE降解率达到80%的高效降解耐冷菌。以上3株菌两两之间与三者组合均无拮抗关系。对菌株进行碳源的广谱性分析,菌株对2-5环多环芳烃降解率可达15%~85%之间,在将菌株应用至20 ℃土壤环境时,60 d可降解土壤中75%的PHE。该耐冷菌群适应条件符合寒区农田土壤实际环境,研究结果对黑土地区土壤多环芳烃污染的微生物修复提供了一定的基础资料。  相似文献

Alkali soils are poor in sulphur-oxidizing bacteria. By appropriate enrichment, two bacterial strains resembling Thiobacillus thiooxidans and T. novellus have been isolated from an alkali soil. The strain of T. novellus possesses characteristics which make it suitable in reclamation of alkali soils by sulphur amendment.  相似文献
