Identifying the Fusarium species cause Fusarium head blight (FHB) and produces mycotoxins in wheat and other cereal is difficult and time consuming because of confusing phenotypic classification systems. In Japan, the F. graminearum complex, F. culmorum, F. avenaceum, and Microdochium nivale predominantly cause FHB. The internal transcribed spacer (ITS) and 5.8S of rDNA, a partial sequence of β-tubulin and mitochondrial cytochrome b (cytb) genes of the four species were PCR-amplified and analyzed. On the basis of the ITS, β-tubulin and cytb sequences, F. avenaceum and M. nivale are distinct from the F. graminearum complex and F. culmorum, whereas the F. graminearum complex is closely related to F. culmorum. Moreover, thiophanate–methyl-resistant isolates of the F. graminearum complex and F. culmorum did not have an amino acid substitution at amino acid codon 198 or 200 of β-tubulin. In contrast, very highly or highly thiophanate–methyl-resistant isolates of M. nivale had Glu (GAG) substituted with Ala (GCG) or Lys (AAG) at codon 198, respectively. The allele-specific PCR assay was used to identify the F. graminearum complex and F. culmorum, and these Fusarium species could be distinguished rapidly.  相似文献

Determination of the Fusarium protein equivalent (FPE) levels in kernels for better characterisation of genotypes showing Fusarium head blight (FHB) resistance, and better detection of susceptibility to kernel infection among genotypes with slight symptom expression was carried out. Twelve wheat cultivars and eight hexaploid winter wheat lines derived from a cross of Triticum aestivum with related species T. macha, T. polonicum, and T. dicoccoides were evaluated for levels of spike and kernel infection, the content of the mycotoxin deoxynivalenol (DON) and FPE in kernels after artificial inoculation with the fungus Fusarium culmorum in the field in 2006–2007. The ELISA immunochemical method was employed for the quantitative analyses of DON and FPE. Three wheat lines had a significantly low infection of spikes and kernels compared to cvs Sumai 3 and Nobeoka Bozu, indicating the presence of specific resistance mechanisms to FHB. The significantly low AUDPC (area under the disease progress curve) and the high level of FPE and DON content in kernels indicated a lack of resistance in one wheat line (crossed with T. polonicum). The results showed highly significant correlations (P < 0.01) between FPE and DON content and between FPE and AUDPC. In addition, correlations between FPE and reductions in yield components were also highly significant. Quantification of Fusarium spp. in wheat kernels can be helpful for evaluating wheat genotypes for their levels of resistance to FHB.  相似文献

Interactions between Barley yellow dwarf virus (BYDV) and Fusarium species causing Fusarium head blight (FHB) in winter wheat cvs Agent (susceptible to FHB) and Petrus (moderately resistant to FHB) were studied over three years (2001–2003) in outdoor pot experiments. FHB developed more rapidly in cv. Agent than in cv. Petrus. The spread of FHB was greater in BYDV-infected plants than in BYDV-free plants. Thousand grain weight (TGW) was reduced more in Fusarium-infected heads of cv. Agent than in cv. Petrus. A highly significant negative correlation was found between disease index and TGW in cv. Agent (r = −0.916), while in cv. Petrus the correlation was less significant (r = −0.765). Virus infection reduced TGW in cv. Petrus more than in cv. Agent. In plants with both infections, TGW reductions in cv. Petrus corresponded to those of BYDV infection, and in cv. Agent TGW was more diminished than in BYDV infection. Effects of different treatments determined over three years on ergosterol contents in grain were generally similar to effects on disease indices. Grain weight per ear and ear weight of the different treatments of both cultivars largely corresponded with the TGW results. Deoxynivalenol (DON) content in grain of cv. Agent infected with Fusarium spp. was 11–25 times higher compared to the corresponding treatments in cv. Petrus. The DON content in grain of plants of the two cultivars infected with both pathogens was higher than that of plants infected only with Fusarium over the three years.  相似文献

Fusarium head blight of wheat, often associated with mycotoxin contamination of food and feed is caused by various Fusarium species. The efficacy of fungicide sprays for the control of the disease and mycotoxins varies from being highly effective to even increasing mycotoxin levels. The potential role of antagonistic fungi in this variability was investigated assessing sensitivity of Fusarium species and saprophytic fungi colonizing wheat kernels to fungicides. Saprophytes were tested for their antagonistic activity to the prevalent Fusarium species Fusarium avenaceum, Fusarium culmorum, Fusarium graminearum, and Fusarium poae. Fungal isolates from mature winter wheat kernels were Alternaria alternata, Arthrinium sp., Aspergillus niger, Epicoccum sp., Microdochium spp., Rhizopus oryzae and Trichoderma sp. In dual culture A. niger, R. oryzae and Trichoderma hamatum were more effective in reducing mycelial growth of Fusarium species than Microdochium majus; A. alternata and Epicoccum sp. were ineffective because of slow growth rates. Saprophytic fungi were sensitive to triazoles; however, prothioconazole and tebuconazole had stronger effects on mycelial growth of Fusarium species. ED50 values also indicated significant differences in the sensitivity of Fusarium species to triazoles (range 0.1–1.7 mg l−1). Azoxystrobin and fluoxastrobin were largely ineffective in inhibiting in vitro growth of Fusarium spp.; sensitivity of the other fungi was generally lower, except for M. majus which was highly sensitive. Due to differences in fungicide sensitivity among Fusarium spp. and ear-colonizing fungi antagonistic to Fusarium spp. fungicides are likely to modify the balance within the mycoflora of wheat ears which may also affect the mycotoxin contamination of grain.  相似文献

The aim of this research was to study levels of resistance to Fusarium basal rot in onion cultivars and related Allium species, by using genetically different Fusarium isolates. In order to select genetically different isolates for disease testing, a collection of 61 Fusarium isolates, 43 of them from onion (Allium cepa), was analysed using amplified fragment length polymorphism (AFLP) markers. Onion isolates were collected in The Netherlands (15 isolates) and Uruguay (9 isolates), and received from other countries and fungal collections (19 isolates). From these isolates, 29 were identified as F. oxysporum, 10 as F. proliferatum, whereas the remaining four isolates belonged to F. avenaceum and F. culmorum. The taxonomic status of the species was confirmed by morphological examination, by DNA sequencing of the elongation factor 1-α gene, and by the use of species-specific primers for Fusarium oxysporum, F. proliferatum, and F. culmorum. Within F. oxysporum, isolates clustered in two clades suggesting different origins of F. oxysporum forms pathogenic to onion. These clades were present in each sampled region. Onion and six related Allium species were screened for resistance to Fusarium basal rot using one F. oxysporum isolate from each clade, and one F. proliferatum isolate. High levels of resistance to each isolate were found in Allium fistulosum and A. schoenoprasum accessions, whereas A. pskemense, A. roylei and A. galanthum showed intermediate levels of resistance. Among five A. cepa cultivars, ‘Rossa Savonese’ was also intermediately resistant. Regarding the current feasibility for introgression, A. fistulosum, A. roylei and A. galanthum were identified as potential sources for the transfer of resistance to Fusarium into onion.  相似文献

Recent research on the epidemiology of Fusarium ear (or head) blight (FEB or FHB) of small-grain cereals is reviewed, focusing on inoculum, infection and disease forecasting. Both conidia and ascospores have been shown to be important for causing FEB. For Fusarium graminearum, propagules from crop debris are the main source of initial inoculum. Inoculum production is critically dependent on rainfall although the precise relationship is not clear. Recent work on understanding the effects of climatic variables on FEB development has been based on field observations. These field-based studies confirmed that warm and moist conditions during anthesis are the key factors for FEB development. Several empirical models were derived from the field data and proposed for use in disease forecasting. However, these models may not be applicable to a broader range of areas because of the limited nature of the field data. Several areas are proposed for future research, focusing on the development of more generally applicable forecasting models and on understanding the relationships between disease severity, fungal biomass and the production of associated mycotoxins.  相似文献

Severe rot of leaves, peduncles and flowers caused by Gibberella zeae (anamorph: Fusarium graminearum) was found on potted plants of hyacinth (Hyacinthus orientalis), a liliaceous ornamental, in greenhouses in Kagawa Prefecture, Japan, in January 2001. This disease was named “Fusarium rot of hyacinth” as a new disease because only the anamorph, F. graminearum, was identified on the diseased host plant. The authors contributed equally to this work. The fungal isolate and its nucleotide sequence data obtained in this study were deposited in the Genebank, National Institute of Agrobiological Sciences and the DDBJ/EMBL/GenBank databases under the accession numbers MAFF239499 and AB366161, respectively.  相似文献

Details of our long-term research programme concerning the epidemiology of Fusarium spp. and mycotoxin production are summarized. Evaluation of the occurrence of Fusarium spp., mainly on winter wheat (Triticum aestivum), was carried out by investigating Fusarium infection and mycotoxin contamination. Two to 15% of grains were infested during 1995–1998 at three climatologically differing localities of the Rhineland, Germany. Disease progress was accelerated by rainfall during the flowering season. The species most frequently isolated were Fusarium avenaceum, F. poae, F. culmorum and F. graminearum. The mean deoxynivalenol (DON) content varied from 19gkg–1 (1995) to 310gkg–1 (1998) and was not always correlated with disease severity. Organic farming systems showed lower rates of infection with ear blight and lower mycotoxin contamination than conventional farming systems.  相似文献

Fusarium head blight (FHB) is an important fungal disease of wheat. The aim of this research was to determine the diversity of Fusarium species infecting winter wheat ears in East Croatia. In 2008 wheat kernels were obtained from three locations in the eastern part of Croatia (Tovarnik, Osijek, Pozega), and in 2009 from two additional locations (Slavonski Brod, Nova Gradiska). In total, 498 visually diseased kernels were selected for morphological identification of Fusarium spp. The identity of 226 selected isolates was further investigated by molecular techniques. The predominant species on wheat kernels in East Croatia in 2008 were F. graminearum, isolated and confirmed from more than 80% of sampled wheat kernels, followed by F. avenaceum (8%) and F. culmorum (7%). Incidence of F. poae was less than 2%. The most common species identified in 2009 were F. graminearum (50%), F. culmorum (13%), F. avenaceum (12%) and F. poae (7%). This is the first report on the identification of Fusarium species isolated from naturally infected wheat ears in Croatia.  相似文献

Fusarium head blight of wheat is caused by a disease complex comprised of toxigenic pathogens, predominantly Fusarium spp., and a non-toxigenic pathogen Microdochium nivale, which causes symptoms visually indistinguishable from Fusarium and is often included as a causal agent of Fusarium head blight. Four field trials are reported here, including both naturally and artificially inoculated trials in which the effect of fungicide treatments were noted on colonisation by Fusarium and Microdochium, and on the production of deoxynivalenol (DON) mycotoxin. The pathogen populations were analysed with quantitative PCR and samples were tested for the presence of the mycotoxin DON. Application of fungicides to reduce Fusarium head blight gave a differential control of these fungi. Tebuconazole selectively controlled F. culmorum and F. avenaceum and reduced levels of DON, but showed little control of M. nivale. Application of azoxystrobin, however, selectively controlled M. nivale and allowed greater colonisation by toxigenic Fusarium species. This treatment also lead to increased levels of DON detected. nobreak Azoxystrobin application two days post-inoculation increased the production of DON mycotoxin per unit of pathogen in an artificially inoculated field trial. This result indicates the potential risk of increased DON contamination of grain following treatment with azoxystrobin to control head blight in susceptible wheat cultivars. This is the first study to show differential fungicidal control of mixed natural pathogen populations and artificial inoculations in field trials.  相似文献

Glasshouse studies were undertaken to determine if fungicides used for the control of Fusarium head blight (FHB) result in elevated concentrations of the trichothecene mycotoxin, deoxynivalenol (DON) in harvested wheat grain. Metconazole and azoxystrobin, at double, full, half or quarter the manufacturer's recommended dose rate, were applied to ears of wheat (cv. Cadenza), artificially inoculated with conidia of either Fusarium culmorum or F. graminearum. Metconazole demonstrated high activity against both pathogens, reducing significantly the severity of FHB and the DON concentrations at each of the four dose rates tested when compared to untreated controls. Applications of azoxystrobin significantly reduced FHB and DON compared to unsprayed controls. However, their effectiveness was significantly less than that of metconazole and no dose rate response was observed. Quantification of the amount of trichothecene-producing Fusarium present in harvested grain was determined using a competitive PCR assay based on primers derived from the trichodiene synthase gene (Tri5). Simple linear regression analyses revealed strong relationships between the amount of trichothecene-producing Fusarium present in grain and the DON concentrations (r 2=0.72–0.97). It is concluded that fungicides, applied for the control of FHB, affect DON concentrations indirectly by influencing the amount of trichothecene-producing Fusarium species present in wheat grain. There was no evidence that fungicide applications directly increase the concentration of DON in grain.  相似文献

Four-hundred-sixty-eightFusarium andFusarium-like isolates were obtained from crowns and subcrown internodes of winter wheat grown in Erzurum, Turkey. Of these isolates, 34.8% wereFusarium acuminatum, 32.3% wereF. equiseti, 16.9% wereF. oxysporum, 15.0% wereMicrodochium nivale (formerlyFusarium nivale), 0.6% wereF. tabacinum and 0.4% wereF. solani. In pathogenicity tests on wheat, the highest disease severity was caused by isolates ofM. nivale, whereas isolates ofF. acuminatum, F. equiseti, F. oxysporum andF. solani were slightly virulent; isolates ofF. tabacinum were nonpathogenic. Fusarium head blight (FHB), mainly caused by Fusarium graminearum species complex (FGSC) and also by other species of this genus, is one of the most destructive cereal diseases with high yield losses and mycotoxin contamination worldwide. The aim of this study was to identify Fusarium species, characterize their virulence factors such as trichothecene genotypes and cell wall degrading enzymes (CWDEs), and also investigate virulence of the isolates obtained from wheat plants with FHB symptoms in Golestan province of Iran. Among 41 isolates tested, 24 were F. graminearum sensu stricto (s.s.), six were F. proliferatum, four were F. culmorum, three isolates belonged to each of F. subglutinans and F. meridionale species and one isolate of F. asiaticum was identified. Among Fusarium isolates, the nivalenol (NIV) genotype could be found more frequently, followed by 3-acetyl deoxynivalenol (3-ADON) and 15-acetyl deoxynivalenol (15-ADON) genotypes. Production of trichothecenes in autoclaved rice cultures was analyzed by gas chromatography (GC) and confirmed by GC–MS. The mean levels of NIV, 3-ADON and 15-ADON produced by Fusarium spp. were 824, 665 and 622 μg kg?1, respectively. All Fusarium isolates were capable of producing CWDEs, mainly cellulase and xylanase. Lipase and pectinase activities appeared later and at less quantities. In overall, the isolates FH1 of F. graminearum and FH8 of F. proliferatum showed the maximum activity of CWDEs, which was correlated with high level of their virulence and aggressiveness on wheat. On the other hand, correlation was observed between the level and type of trichothecene produced by each isolate and its virulence on wheat. Virulence of trichothecene producing isolates was higher than that of non-trichothecene producing isolates. Our results suggested that CWDEs and trichothecenes, as virulence factors, have considerable roles on virulence and aggressiveness of the pathogen. This is the first report on the effect of trichothecenes and CWDEs on virulence and aggressiveness of Fusarium spp. associated with FHB disease in wheat growing regions of Iran.  相似文献

This review describes the naturally occurring mechanisms in cereals that lead to a reduction of Fusarium trichothecene mycotoxin accumulation in grains. A reduction in mycotoxin contamination in grains could also limit fungal infection, as trichothecenes have been reported to act as virulence factors. The mechanisms explaining the low toxin accumulation trait, generally referred to as type V resistance to Fusarium, can be subdivided into two classes. Class 1 includes mechanisms by which the plants chemically transform the trichothecenes, leading to their degradation or detoxification. Among the detoxification strategies, glycosylation of trichothecenes is a natural process already reported in wheat. According to the structure and the toxicity of trichothecenes, two other detoxification processes, acetylation and de-epoxidation, can be expressed, at least in transgenic plants. Class 2 comprises mechanisms that lead to reduced mycotoxin accumulation by inhibition of their biosynthesis through the action of plant endogenous compounds. These include both grain constitutive compounds and compounds induced in response to pathogen infection. There are already many compounds with antioxidant properties, like phenolic compounds, peptides or carotenoids, and with prooxidant properties, like hydrogen peroxide or linoleic acid-derived hydroperoxides, that have been described as ‘modulators’ of mycotoxin biosynthesis. This review addresses for the first time different studies reporting specific in vitro effects of such compounds on the biosynthesis of Fusarium mycotoxins. A better understanding of the natural processes limiting accumulation of trichothecenes in the plant will open the way to the development of novel breeding varieties with reduced ‘mycotoxin risk’.  相似文献

We investigated incidences of Fusarium head blight (FHB) and concentrations of six mycotoxins (deoxynivalenol, nivalenol, 3-acetyldeoxynivalenol, T-2 toxin, HT-2 toxin and zearalenone) in wheat from 2010 to 2013. Field trials were conducted at the Experimental Station of Cultivar Testing in Chrz?stowo, Poland (53o11’N, 17o35’E). We examined the effects of four agronomic factors, including pre-crop type (corn, sugar beets and wheat), date of sowing (late autumn: November 8–December 9 or spring: March 29–April 19), fungicidal application (untreated or treated with two applications) and cultivar (Monsun, Cytra), on FHB index (FHBi) and mycotoxin levels in order to minimize the risk of wheat grain contamination by mycotoxins via integrated pest management methods. The dominant Fusarium species observed on wheat heads were F. culmorum, F. avenaceum (Gibberella avenacea) and F. graminearum (Gibberella zeae), at 21.1%, 17.2% and 7.1%, respectively. A monthly rainfall sum of 113.9 mm and a relatively low air temperature (monthly average 15.5 °C) resulted in the highest FHBi in untreated wheat (25.1%). Agronomic factors crucial for the FHB incidence were the pre-crop, fungicidal treatments and cultivar selection. In wheat planted after wheat or corn, the FHBi was higher compared with a pre-crop of sugar beet. A double application of fungicides at BBCH 30–32 with prothioconazole and spiroxamine and at a BBCH 65 with fluoxastrobin and prothioconazole effectively reduced the FHBi and mycotoxin concentrations, respectively, in grain. The cultivar ‘Cytra’ had a greater FHBi (10.4%) than ‘Monsun’ (4.6%), and grain infestations by Fusarium species were also greater in ‘Cytra’, at 16.5%, than in ‘Monsun’, at 11.2%. Untreated cv. Cytra grown after corn in spring produced grains with the highest amounts of the mycotoxins, deoxynivalenol, 3-acetyldeoxynivalenol, zearalenone and HT-2 (605, 103, 17.5 and 5.53 μg/kg, respectively). Total mycotoxin levels in wheat were correlated with five determinants: duration of the period between the end of flowering and the beginning of kernel abscission, FHBi, F. culmorum isolation, G. zeae isolation and Fusarium ratio (FR) as a % of total mould isolations. Although, the mean concentration of mycotoxins in grain did not exceed the maximum permissible values for unprocessed wheat our study suggests necessity to monitor and mitigate FHB risk for susceptible cultivars, when wheat spring sowing follows corn or wheat.  相似文献

The mechanism by which Fusarium diseases of cymbidium plants are suppressed by a weakly virulent strain HPF-1 of Fusarium sp. was studied. Strain HPF-1 produced microscopic, necrotic local lesions on cymbidium leaves, causing minor damage to palisade tissues at the infection sites. This weakly virulent strain remained near the site of infection and did not develop further. It systemically and nonselectively suppressed some diseases of cymbidium such as yellow spot of leaves caused by Fusarium proliferatum and F. fractiflexum, bulb and root rot caused by F. oxysporum, and dry rot of bulbs and roots caused by F. solani. Because endogenous salicylic acid levels increased in cymbidium leaves inoculated with strain HPF-1, the mechanism of disease suppression is thought to be systemic acquired resistance.  相似文献

The effect of temperature on the in vitro growth rates and pathogenicity of a European Fusarium collection consisting of isolates of Fusarium graminearum, F. culmorum, F. avenaceum, F. poae and Microdochium nivale was examined. Irrespective of geographic origin, the optimum temperature for the growth of F. graminearum, F. culmorum and F. poae was 25 °C, while that for F. avenaceum and M. nivale was 20 °C. In general, the growth rates of F. graminearum, F. culmorum and F. poae increased between 10 and 25 °C and those of F. avenaceum and M. nivale increased between 10 and 20 °C. Pathogenicity tests were carried out by examining the effect of the five species on the in vitro coleoptile growth rate of wheat seedlings (cv. Falstaff). Irrespective of geographic origin, the temperature at which F. avenaceum, F. culmorum and F. graminearum caused the greatest retardation in coleoptile growth ranges 20–25 °C (>89.3% reduction), whilst for F. poae and M. nivale it was 10–15 °C (>45.6% retardation), relative to uninoculated control seedlings. In general, F. culmorum and F. graminearum were the most pathogenic of the five species, causing at least a 69% reduction in coleoptile growth at 10, 15, 20 and 25 °C. General linear model analysis (GLIM) showed that species accounted for 51.3–63.4% of the variation in isolate growth and from 19.5% to 44.3% of the variation in in vitro pathogenicity. Country of origin contributed from 22.6% to 51.9% to growth rate variation and from 0.73% to 7.61% to pathogenicity variation. The only significant correlation between in vitro growth and pathogenicity was that observed for M. nivale at 15 °C (r = -0.803, P < 0.05).  相似文献

Fusarium head blight (scab) epidemics of wheat occurred in uawy (Northern Poland) during 1998 and in Wielkopolska (West) and in Southern regions of Poland in 1999. Four species were identified in wheat heads with scab symptoms: Fusarium culmorum, Fusarium graminearum, Fusarium avenaceum and Microdochium nivale. A significant increase in the frequency of F. graminearum (between 23% and 38%), was observed, compared to about 10% during the previous decade. The mycotoxins deoxynivalenol (DON), nivalenol (NIV) and moniliformin (MON) in amounts up to 24.3, 14.2 and 1.72mgkg–1respectively, were identified in kernels samples.  相似文献

This work presents an analysis of the relationship between components of partial disease resistance (PDR) detected using in vitro detached leaf and seed germination assays, inoculated with Microdochium majus, and Fusarium head blight (FHB) resistance to Fusarium graminearum assessed using point inoculation, termed Type II resistance. Relationships between in vitro-determined PDR components and FHB resistance using techniques which inoculate the wheat spike uniformly, termed Type I resistance (incidence and severity), have been reported previously. In this study shorter incubation periods, longer latent periods and shorter lesion lengths in the detached leaf assay and higher germination rates in the seed germination assay were related to greater FHB resistance measured by single point inoculation (Type II), collectively explaining 54% of the variation. Overall the relationships observed for Type II FHB resistance were similar to previous findings for Type I resistances. However, the relative magnitude of effects of the individual PDR components determined in vitro varied between FHB disease resistance parameters. Resistance in seed germination and latent period in the detached leaf assay were more strongly related to resistance assessed by point inoculation (Type II) and severity-Type I as opposed to incubation period which was most strongly related to disease incidence-Type I. The results provide evidence that individual components of partial disease resistance differentially affect aspects of FHB disease progression in the wheat spike. This work supports the view that the current model of types of resistance is an oversimplification of the interacting mechanisms underlying expression of FHB resistance.  相似文献
