The aim of this study was to determine effects of konjac glucomannan (KGM) in a high fat corn oil diet on risk factors of colon carcinogenesis, that is, fecal β-glucuronidase, mucinase, and bile acids, and on preventive factors, that is, fecal microflora and cecal short-chain fatty acids (SCFAs). Sprague-Dawley rats (n = 8 animals per group) were fed a normal-fat fiber-free (5% corn oil, w/w) or high-fat (25% corn oil, w/w) diet containing no fiber, KGM (5%, w/w), or inulin (5%, w/w, as a prebiotic control) for 4 weeks. Results indicated that the high-fat fiber-free diet significantly elevated the fecal β-glucuronidase and mucinase activities and total bile acid concentration and decreased cecal SCFA contents, as compared with its normal-fat counterpart. The incorporation of KGM, as well as inulin, into the high-fat fiber-free diet beneficially reduced the fecal β-glucuronidase and mucinase activities and lithocholic acid (secondary bile acid) concentration. Although KGM elevated the daily fecal total bile acid excretion, the change was due to the primary, instead of the secondary, bile acids. In addition, KGM beneficially promoted the daily fecal excretion of bifidobacteria and lactobacilli and cecal SCFA contents, as compared with the high-fat fiber-free diet. Therefore, the present study suggests that KGM potentially attenuated the high fat-induced risk in colon carcinogenesis.  相似文献

An improved method for the measurement of fructans in wheat grains is presented. A mild acid treatment is used for fructan hydrolysis, followed by analysis of the released glucose and fructose with high performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD). Not only the amount of fructose set free from fructans but also the released glucose can be quantified accurately, allowing determination of the average degree of polymerization of fructans (DP(av)). Application of the mild acid treatment to different grain samples demonstrated that a correction should be made for the presence of sucrose and raffinose, but not for stachyose or higher raffinose oligosaccharides. The fructan content and DP(av) of spelt flour, wheat flour, and whole wheat flour were 0.6%, 1.2%, and 1.8% of the total weight and 4, 5, and 6, respectively. Validation experiments demonstrate that the proposed quantification method is accurate and repeatable and that also the DP(av) determination is precise.  相似文献

Experiments were conducted to study the effect of a dietary supplement of dehulled adlay (Coix lachryma-jobi L. var. ma-yuen Stapf) on the culture counts of some important groups of intestinal bacteria and their metabolism in the gastrointestinal (GI) tract of Sprague-Dawley rats. Rats were divided into four groups, and each group was fed a diet containing different levels of dehulled adlay for 30 days as follows: 0% (control), 5%, 20%, and 40%. All animals fed with adlay had normal healthy intestinal walls and no pathogenic signs whatsoever. There were no significant differences in body weight gain or the cecal pH between different groups of rats. Both the 20% and 40% groups had lower culture counts of enterics in their feces than the 5% and control groups, whereas the culture counts of fecal lactic acid bacteria were higher in feces of rats fed with adlay than in the control group. Cecal total short-chain fatty acid (SCFA) content and fecal SCFA were significantly higher in the 20% and 40% groups than in the control and 5% groups. All the adlay-fed rats had a higher fecal butyric acid concentration than the control rats. It is concluded that adlay has a significant influence on the growth of intestinal bacteria, which may ultimately affect the physiology and other functions of GI tracts of rats.  相似文献

Epilactose (4-O-beta-galactopyranosyl-D-mannnose) is a rare disaccharide in cow milk that can be synthesized from lactose by the cellobiose 2-epimerase of Ruminococcus albus. In this study, we examined the biological activities of epilactose using male Wistar-ST rats. The apparent rates of calcium and magnesium absorption of rats fed epilactose and fructooligosaccharide diets were greater than those fed control and lactose diets, accompanied by greater weight gain of the cecal wall and higher levels of short-chain fatty acids and other organic acids. Epilactose also increased the calcium absorption in everted small intestinal sacs. In addition, the levels of plasma total cholesterol and nonhigh-density lipoprotein cholesterol were lower in epilactose-fed rats. These results indicate that epilactose promotes calcium absorption in the small intestine and possibly lowers the risk of arteriosclerosis. Cecal microbes may efficiently utilize epilactose and contribute to these biological activities.  相似文献

Galactooligosaccharides (GOS), prebiotic nondigestible oligosaccharides derived from lactose, have the potential for improving mineral balance and bone properties. This study examined the dose-response effect of GOS supplementation on calcium and magnesium absorption, mineral retention, bone properties, and gut microbiota in growing rats. Seventy-five 4-week-old male Sprague-Dawley rats were randomized into one of five treatment groups (n = 15/group) and fed a diet containing 0, 2, 4, 6, or 8% GOS by weight for 8 weeks. Dietary GOS significantly decreased cecal pH and increased cecal wall weight and content weight in a dose-dependent manner (p < 0.0001). Fingerprint patterns of the 16S rRNA gene PCR-DGGE from fecal DNA indicated the variance of bacterial community structure, which was primarily explained by GOS treatments (p = 0.0001). Quantitative PCR of the samples revealed an increase in the relative proportion of bifidobacteria with GOS (p = 0.0001). Net calcium absorption was increased in a dose-response manner (p < 0.01) with GOS supplementation. Dietary GOS also increased (p < 0.02) net magnesium absorption, femur ??Ca uptake, calcium and magnesium retention, and femur and tibia breaking strength. Distal femur total and trabecular volumetric bone mineral density (vBMD) and area and proximal tibia vBMD increased (p < 0.02) with GOS supplementation. Trabecular-rich bones, that is, those that rapidly turn over, were most benefited. Regression modeling showed that GOS benefited calcium and magnesium utilization and vBMD through decreased cecal pH, increased cecal wall and content weight, and increased proportion of bifidobacteria.  相似文献

Agave plants utilize crassulacean acid metabolism (CAM) for CO(2) fixation. Fructans are the principal photosynthetic products generated by agave plants. These carbohydrates are fructose-bound polymers frequently with a single glucose moiety. Agave tequilana Weber var. azul is an economically important CAM species not only because it is the sole plant allowed for tequila production but because it is a potential source of prebiotics. Because of the large amounts of carbohydrates in A. tequilana, in this study the molecular structures of its fructans were determined by fructan derivatization for linkage analysis coupled with gas chromatography-mass spectrometry (GC-MS), nuclear magnetic resonance (NMR), and matrix-assisted laser desorption time-of-flight mass spectrometry (MALDI-TOF-MS). Fructans were extracted from 8-year-old A. tequilana plants. The linkage types present in fructans from A. tequilana were determined by permethylation followed by reductive cleavage, acetylation, and finally GC-MS analysis. Analysis of the degree of polymerization (DP) estimated by (1)H NMR integration and (13)C NMR and confirmed by MALDI-TOF-MS showed a wide DP ranging from 3 to 29 units. All of the analyses performed demonstrated that fructans from A. tequilana consist of a complex mixture of fructooligosaccharides containing principally beta(2 --> 1) linkages, but also beta(2 --> 6) and branch moieties were observed. Finally, it can be stated that fructans from A. tequilana Weber var. azul are not an inulin type as previously thought.  相似文献

The purpose of this study was a comparative examination of the fructan and fructooligosaccharide (FOS) content of different varieties of onions (Allium cepa L. cv. Sturon, Hysam, Durco, Grano de Oro, and Caribo) and the changes produced during their commercial storage. In fresh onions, the Grano de Oro variety presented a remarkably different behavior, showing low contents of total fructans and FOS and high levels of reducing sugars. In the other varieties, Sturon, Hysam, Durco, and Caribo, fructans were the main carbohydrates, the lowest polymerized FOS being the major oligomer. Storage period caused in these varieties important increased levels of free fructose attributed to fructan hydrolysis. Maleic hydrazide treatment had no significant effect in avoiding the hydrolysis of fructans during storage conditions for the Sturon variety. Varieties with >16% dry matter or 15% soluble solids contents could be stored for 6 months at 0 degrees C and 60-65% relative humidity.  相似文献

Protein-energy malnutrition (PEM) is frequently associated with the occurrence of infection due to a decline in immune function. Here, an experiment was conducted with the objective of enhancing mucosal immunity by administration of Lactobacillus pentosus ONRIC b0240 (b240) in PEM model mice. Three groups of male C3H/HeN mice aged approximately 12 weeks were caged in groups of five or six and received various treatments. The mice were fed 4 (low-protein diet; PEM model), 20 (standard-protein diet), or 40% (high-protein diet) ovalbumin (OVA) with or without 0.05% b240. Five weeks later, all mice were sacrificed, and the organs were extracted for analysis of the immune response. Acute toxicity was not observed in this study. The addition of b240 showed no influence on body weight; however, body weight decreased with increasing protein level. Interestingly, intestinal total IgA was significantly increased (p<0.05) in all test diets with b240. The in vitro study showed that the number of B cells and type 2 helper T (Th2) cells were significantly increased in mouse spleen cells with b240 treatment, whereas no differences were found in the number of Th1 cells. b240 also has the ability to augment IgA and IgG production in mouse Peyer's patch cells. These results suggest that b240 enhances IgA production and helps recover the intestinal immune system in PEM model mice via augmentation of humoral immunity.  相似文献

Seven healthy females and six males consumed daily 256 mg of vitamin C, 271 mg of flavanones (mainly as glycosides), 6 mg of carotenoids (mainly xanthophylls and cryptoxanthins), and 0.16 mg of folate by incorporation of daily three times 236 mL of not from concentrate orange juice (OJ) into their habitual diet. At the end of 3 weeks, mean vitamin C, folate, carotenoid, and flavanone plasma concentrations increased significantly relative to baseline by 59% (p < 0.001), 46% (p = 0.018), and 22% (p < 0.001), and 8-fold (p = 0.045), respectively. Flavanones were excreted in urine 9-fold more at the end of the intervention (p = 0.01) but returned to baseline 2 days after study completion. After the 3 week intervention, plasma concentrations of vitamins A and E did not change. 8-Hydroxydeoxyguanosine in white blood cells declined by 16% (p = 0.38; n = 11), and in individuals with high baseline concentrations by 29% (p = 0.36; n = 7), respectively. Low-density lipoprotein (LDL)-/high-density lipoprotein (HDL)-cholesterol ratios decreased but cholesterol (HDL, LDL, total) and thiobarbituric acid reactive substance plasma concentrations did not change significantly. We conclude from this pilot study that OJ is an excellent food source to enhance circulating concentrations of valuable hydrophilic as well as lipophilic phytochemicals.  相似文献

The objective of this study was to report on the variation of DP 6 isomers (6b, 6c + 6d1 + 6d2), 7a, 8, and 9-12 fructooligosaccharides (FOS) and their hydrolysis parameters [percent hydrolysis, consumption rate, hydrolysis rate constant (k(obsd)), and half-life time (t(1/2))] in onion bulb tissues stored for 6 months at 10, 15, or 20 degrees C. The hydrolysis of DP 6 isomers, 7a, and 8 ranged from 74 to 85%, whereas that of DP 9-12 averaged 86%. The consumption rate of 6b, 6c + 6d1 + 6d2, 7a, 8, and -12 averaged 25, 58, 38, 26, and 48 microg/g of fresh weight per week, respectively. The k(obsd) showed large variation from 53 x 10(-3) week(-1) (lowest value) to 92 x 10(-3) week(-1) (highest value), whereas the half-life t(1/2) of the different FOS ranged between 7.5 and 13.1 weeks. DP 6b isomers increased during the first month, and then the content decreased sharply during the second month at 20 degrees C but remained stable during the last 4 months, whereas at 10 and 15 degrees C, 6b decreased progressively from the first to the six month. In contrast, DP 6c + 6d1 + 6d2 decreased abruptly within the first 3 months at 10, 15, and 20 degrees C; however, during the last 3 months they remained stable, ranging between 0.32 and 0.39. Variation of DP 7a, 8, and 9-12 FOS was close to that of DP 6 FOS isomers. DP 7a increased slightly during the first month, and afterward 7a started decreasing progressively during the last 5 months. DP 8 FOS showed a similar pattern independent of temperature regime: they increased slightly within the first month, but from the second month, DP 8 began decreasing progressively and continued decreasing to the sixth month. DP 9-12 FOS also varied similarly to DP 6c + 6d1 + 6d2. They decreased sharply within the first 2 months, and during the last 4 months, they continued to decrease slowly. Surprisingly, these variations occurred independently of temperature regimes and were affected only by storage duration. It was concluded that highly polymerized FOS are preferably hydrolyzed rather than low DP FOS isomers because they have a relatively high content of fructosyl end chains.  相似文献

Absorption of dietary anthocyanins is limited; however, fecal anthocyanin excretion has been rarely studied. We developed a method for extraction and analysis of fecal anthocyanins. Aqueous methanol (60%) maximized extraction efficiency (approximately 88%). Severe anthocyanin degradation (monitored by high-performance liquid chromatography) was observed in feces stored at -18 degrees C; therefore, storage time should be minimized and lower temperatures used. Fecal and cecal content samples were collected from 32 rats receiving either chokeberry, bilberry, grape-enriched (3.85 g monomeric anthocyanin per kg diet), or control diet for 14 weeks. Fecal anthocyanin concentrations were significantly different among groups (0.7/1.8/2.0 g/kg wet feces, chokeberry/bilberry/grape). Anthocyanin profiles of cecal contents and feces were similar. Losses in the intestinal contents were high for anthocyanin glucosides, moderate for galactosides, and negligible for arabinosides or xylosides. Acylation or diglucosylation enhanced anthocyanin stability. High anthocyanin concentration in the fecal content may favor anthocyanin absorption into the colon epithelial cells, resulting in potential health benefits.  相似文献

This study provides a contribution to hard-type cheese starter culture production through the use of a freeze-dried culture in the ripening of hard-type cheeses. The effect of initial cell concentration, ripening temperature, and cell immobilization of kefir on the degree of openness, mold spoilage, microbial associations, physicochemical characteristics, and aroma-related compounds was studied. Use of kefir starter cultures resulted in cheese with an increased shelf life and resistance to spoilage as compared to control cheeses without kefir inoculants. Furthermore, the freeze-dried kefir culture improved aroma, taste, and texture characteristics while increasing the degree of openness in comparison to traditional hard-type cheese products. The kefir culture resulted in an increase in counts of total aerobic bacteria, yeasts and molds, lactococci, and lactobacilli until the 15th day of ripening. From then on, only lactobacilli counts increased, reaching levels up to 9.17 log CFU/g in cheeses ripened at 5 degrees C using freeze-dried kefir cells immobilized on casein. SPME-GC/MS analysis revealed major differences in volatile composition, especially with regard to alcohols (up to 75%), carbonyl compounds (up to 75%), and esters (up to 64%) between cheeses made with kefir cells and cheeses made without kefir inoculants.  相似文献

  【目的】  磷富集植物对高磷具有良好的耐性是其应用于植物修复的前提。本研究探讨磷富集植物矿山生态型水蓼根系对高磷的耐受能力,为后期利用其修复环境中的过量磷提供一定的理论依据。  【方法】  采用矿山生态型和非矿山生态型水蓼(ME和NME)进行了水培试验,营养液中设置无机磷2、4、8、16 mmol/L 4个高浓度处理,以正常磷0.5 mmol/L浓度为对照,分析对比了两种生态型水蓼根系对高磷的耐受特征。  【结果】  随着磷浓度的增加,ME地上部和根系生物量和磷积累量均先显著增加后降低,在磷浓度4 mmol/L时出现峰值,而NME则大体呈降低趋势。高磷处理ME地上部和根系生物量分别为NME的1.35~2.56和1.18~1.86倍,除磷浓度16 mmol/L处理外,ME地上部和根系磷积累量分别为NME的1.35~2.58和1.36~1.96倍,磷积累能力更强。ME根系质膜开始受到明显损伤的营养液磷浓度是8 mmol/L,而NME是磷浓度4 mmol/L。随着磷浓度的增加,ME根系中H2O2、MDA和NME根系中的MDA含量均表现为先稳定后显著增加,而NME根系中的H2O2含量则持续增加;两种生态型水蓼根系SOD、POD和CAT活性均先显著增高后降低,分别在磷浓度4和2 mmol/L时显著高于对照。与NME相比,ME根系抗氧化酶活性更高,对H2O2和MDA的清除能力更强。两种生态型水蓼根系亚细胞组分磷含量均表现为可溶部分(含液泡)>细胞壁>细胞器。随着磷浓度的增加,两种生态型水蓼各亚细胞组分磷含量均显著增加,且在磷浓度2和4 mmol/L处理下,根系可溶部分(含液泡)磷分配比例明显高于对照,液泡是其磷素储存的重要场所。  【结论】  矿山生态型水蓼对高磷的耐受能力和磷积累能力均强于非矿山生态型水蓼。高磷处理下矿山生态型水蓼存在根系自由基和保护酶动态平衡、细胞壁固持和可溶部分的液泡区隔化现象,这是其根系的重要耐受特征。  相似文献

Commercial tomato pulp was air-dried to two final moisture contents in order to obtain intermediate moisture pulp (IMP, 23% moisture) and dried pulp (DP, 9% moisture). IMP and DP were stored at 4, 20, and 37 degrees C for approximately 5 months; periodically samples were analyzed to evaluate heat and oxidative damage by measurement of color changes, total phenolics, rutin, lycopene and furosine concentrations, and antioxidant activity of the lipophilic extract. IMP and DP, despite different drying degree, had similar antioxidant activity; in fact, whereas lycopene was stable to drying treatments, ascorbic acid was totally degraded in both products. During storage, IMP and DP showed different behavior: IMP was more sensitive to degradation than DP, especially with regard to lycopene, rutin, and antioxidant activity degradation and to nonenzymatic browning. Effects of storage temperature varied with regard to different parameters: variations in rutin, furosine, and color indices were higher in products stored at higher temperatures, while lycopene and antioxidant activity of the lipophilic fraction were maximally degraded in products stored at 4 degrees C.  相似文献

The objectives of the current studies included the characterization of the temporal changes in indices of folate status and amino acid concentrations during both folate depletion and repletion phases. In trial 1, a 6 week folate depletion protocol was employed, using 60 weanling rats assigned to receive an amino acid-defined diet with or without 1 mg/kg folic acid. A 4 week folate depletion period was judged to be optimal on the basis of the development of nadirs in both plasma and hepatic folate stores and elevated (>6-fold relative to folate-adequate controls) concentrations of plasma homocysteine and glycine. In trial 2, 54 weanling rats, previously maintained on a folate-devoid diet for 4 weeks, were assigned to receive 0.25 mg/kg folate as either crystalline folic acid or folate from a folate-enriched egg yolk powder. Both forms of folate supported similar rates of gain, increases in plasma and hepatic folate stores, and reductions in plasma glycine concentrations, whereas the folate in egg yolk powder lowered plasma homocysteine concentrations further than the crystalline folic acid (P < 0.05). These data support the use of both plasma glycine and homocysteine as sensitive response criteria for folate status in a rat bioassay of folate depletion and repletion and establish appropriate temporal end-points for such studies.  相似文献

选用16头18月龄、体况良好、体重380±14kg的西门塔尔牛育成母牛,采用完全随机区组设计分为4组,研究日粮添加硫酸铜(0、8、16和24mg铜/kg)对母牛发情周期生殖激素分泌规律的影响。结果表明:与对照组比较,8mg/kg组发情周期促黄体素(LH)、促卵泡素(FSH)、孕酮(P4)、雌二醇(E2)水平显著提高(P<0.05);16mg/kg组有提高趋势,但差异不显著(P>0.05)。添加8mg铜/kg的硫酸铜对母牛发情周期生殖激素分泌有促进作用,加上基础日粮的含铜量,建议日粮以硫酸铜为铜源时铜水平为13.87mg/kg。  相似文献

The fructan content of Finnish rye grains (13 samples, seven cultivars, harvested in 1998‐2000) varied at 4.6–6.6 g/100 g (db). Commercial whole grain rye flour and rye flakes had fructan content of 4 g/100 g, light refined rye flour had fructan content of 3 g/100 g, and rye bran had fructan content of 7 g/100 g. Fructan content as high as 23 g/100 g was detected in the water‐extractable concentrate of rye bran. Finnish soft rye bread and rye crisp bread contained 2–3 g of fructan/100 g. According to the suggested new definition of dietary fiber, fructans are also classified as dietary fiber. This means that the dietary fiber content of some cereal foods such as rye products may be increased by as much as 20% due to the presence of fructans in the grain.  相似文献

A simplified method to determine total fructans in food and pet food has been developed and validated. It follows the principle of AOAC method 997.08, i.e., high-performance anion exchange chromatographic (HPAEC) determination of total fructose released from fructans (F(f)) and total glucose released from fructans (G(f)) after enzymatic fructan hydrolysis. Unlike AOAC method 997.08, calculation of total fructans is based on the determination of F(f) alone. This is motivated by the inherent difficulty to accurately determine low amounts of G(f) since many food and pet food products contain other sources of total glucose (e.g., starch and sucrose). In this case, a correction factor g can be used (1.05 by default) to take into account the theoretical contribution of G(f). At levels >5% of total fructans and in commercial fructan ingredients, both F(f) and G(f) can and should be accurately determined; hence, no correction factor g is required. The method is suitable to quantify total fructans in various food and pet food products at concentrations >or=0.2% providing that the product does not contain other significant sources of total fructose such as free fructose or sucrose. Recovery rates in commercial fructan ingredients and in selected food and pet food ranged from 97 to 102%. As part of a measurement uncertainty estimation study, individual contributions to the total uncertainty (u) of the total fructan content were identified and quantified by using the validation data available. As a result, a correlation between the sucrose content and the total uncertainty of the total fructan content was established allowing us to define a limit of quantitation as a function of the sucrose content. One can conclude that this method is limited to food products where the sucrose content does not exceed about three times the total fructan content. Despite this limitation, which is inherent to any total fructan method based on the same approach, this procedure represents an excellent compromise with regard to accuracy, applicability, and convenience.  相似文献

Male adult Wistar rats were randomly divided into six groups in a 2 x 3 factorial design and fed diets containing different levels of Tremella fuciformis Berk (TFB) dietary fiber (0, 5, or 10%) and 1 g of cholesterol/100 g of diet with or without 0.7% Nebacitin for 4 weeks. TFB contained 6.2% soluble dietary fiber and 57.3% insoluble dietary fiber. The results showed that the serum LDL-cholesterol, hepatic total cholesterol, and triglyceride levels were significantly decreased (P < 0.05) in the rats fed diets with TFB content with or without Nebacitin. However, the serum total cholesterol, VLDL-cholesterol, and triglyceride levels were significantly decreased (P < 0.05) by Nebacitin. In feces, the presence of TFB (T5, T10, AT5, and AT10) in the diet significantly increased the total neutral steroids and bile acid excretions and undigested fiber concentrations as compared to T0 or AT0. In the small intestine, the Nebacitin diets increased the weights of both cecum and colon-rectum contents and lowered short-chain fatty acid (SCFA) concentrations of serum and cecal contents more than no Nebacitin diets did. It was suggested that the hypocholesterolemic effect of TFB dietary fiber may be mediated by the increase in fecal neutral steroids and total bile acids excretion and the increase in SCFA productions. The TFB edible mushroom dietary supplement altered the intestinal physiology of the rats.  相似文献

本文以研究日粮能量水平对蛋鸡性成熟过程中卵巢重、E2及P4水平的影响为目的。结果表明:在性成熟过程中,卵巢重和卵巢表面卵泡数随着年龄和能量水平的提高而增多,22周龄时,除低能Ⅱ组外,其他各组卵巢质量和优势卵泡(10mm以上)明显增多,与低能Ⅱ组差异达显著水平;血清E2和P4含量持续上升,22周龄时均达较高水平;高能组E2和P4水平显著高于低能组,到22周龄,对照组E2水平显著高于其他各组,高能组又显著高于低能Ⅱ组;其他4组P4水平显著高于低能Ⅱ组,说明过低的能量水平通过影响E2和P4水平的分泌进而影响卵巢卵泡的发育。  相似文献
