Abstract— The freshwater prawn Macrobrachiurn rosenbergii is a commercially important culture species in the South Central United States. Two major constraints in the commercial culture of the freshwater prawn in the U.S. are poor survival during live transportation of seed‐stock to growout ponds, and live transportation of pond harvested prawn to distant live markets due to the territorial and cannibalistic nature of prawn. The use of anesthetics could possibly improve transport survival; however, to date anesthetic agents have not been evaluated for use with prawn. Two trials were conducted with juvenile freshwater prawn to compare the efficacy of anesthetics commonly used on fish. The first trial was designed to identify the most promising candidates. In Study 1, tricaine methanesulfonate (MS‐222), 2‐phenoxyethanol, quinaldine sulfate (quinaldine), clove oil, and Aqui‐STM were evaluated at 25 and 100 mg/L for 1 h in three replicate 10‐L glass containers, containing five juvenile prawn each. Relative sedation level was determined every 3 min for 1 h, then recovery time and survival were measured. In Study 1, MS‐222 and 2‐phenoxyethanol were determined to be ineffective on prawn at all rates tested. Based on their performance in Study 1, quinaldine, clove oil, and AquiSTM were evaluated at 100, 200, and 300 mg/L in Study 2. Observations were determined as in Study 1. Clove oil and Aqui‐STM induced anesthesia faster and at lower concentrations than quinaldine. At the highest treatment rate (300 mg/L) prawn suffered 60% mortality in the Aqui‐STM treatment, 13% mortality in the quinaldine treatment, and 0% mortality in the clove oil treatment and control following a 1‐h exposure to these concentrations. Based on these data, Aqui‐STM and clove oil applied at 100 mg/L may be suitable anesthetic treatments for prawn. Additional research is needed to determine optimal time and dose relationships to minimize stress during holding, handling, and transportation of prawn.  相似文献

ABSTRACT

The objective of this study was to determine the microbiological quality of farmed giant freshwater prawn (Macrobrachium rosenbergii) collected from four freshwater farms located in Kerala (India). Microbiological counts on whole, deheaded, and deheaded and deveined prawn as well as on head and intestine were determined. Average counts (log10 cfu g?1) on whole prawn were 6.9 (total mesophilic counts (TPC) at 37°C) and 4.1 (psychrotrophic count at 7°C). The results indicated that the aerobic counts were within the acceptable limit. The levels of enterococci and faecal coliforms in farmed M. rosenbergii were high. There was an increase in the numbers of enterococci and Staphylococcus aureus after deheading. The major groups of bacteria comprising the flora of M. rosenbergii were Aeromonadaceae, Enterobacteriaceae and Pseudomonas. Farmed M. rosenbergii carried pathogenic bacteria such as Enterococcus spp., S. aureus, Aeromonas hydrophila, A. veronii biovar sobria and Clostridium perfringens. The results of the study suggest adoption of good farming and post harvest practices to improve the microbiological quality of farmed freshwater prawn.  相似文献

Giant freshwater prawn (GFP; Macrobrachium rosenbergii) aquaculture has expanded rapidly since 1990. Most local culture industries, however, have developed in an unsystematic way. Fiji has a small culture industry producing the ‘Anuenue’ strain; however, performance of this strain has never been systematically evaluated. Recently, some Fijian farmers have reported declines in stock productivity. The current project evaluated the relative performance of three exotic strains with different genetic backgrounds from Malaysia, Indonesia and Vietnam, against the ‘local’ strain in Fiji in a 4 × 3 replicated pond trial experiment. A total of 5827 prawns were harvested after 143 days growout. Individual growth rate and relative survival of the Fiji strain were not statistically different from any of the introduced strains, but Vietnam strain was superior to that of the Malaysia strain. Genetic diversity showed significant differences in variability among strains, with the Malaysian strain displaying the lowest genetic diversity. Indonesia strain showed that females were reaching maturation earlier than other strains and were smaller in size. This study suggests that Malaysian and Indonesian strains would constitute a poor choice for Fiji, whereas the Vietnam strain consistently performed well on all criteria measured. High variation among replicate ponds within strains unfortunately confounded among‐strain variation.  相似文献

The protective efficacy of a DNA construct containing extra small virus antisense (XSVAS) gene of nodavirus encapsulated with chitosan nanoparticles (NPs) was investigated in giant freshwater prawn Macrobrachium rosenbergii (De Man, 1879). The delivery was carried out using oral and immersion methods. A plasmid concentration of 100 ng μL?1 when conjugated with chitosan NPs was found to be more effective in increasing the survivability of the infected prawn. The particle mean size, zeta potential and loading efficiency percentage were 297 nm, 27 mV and 85%, respectively. The ability of the chitosan to form a complex with the plasmid was studied by agarose gel electrophoresis. The NPs were characterized by atomic force microscopy (AFM). Persistence study showed the presence of the DNA construct up to 30th day post‐treatment. The oral treatment was found to be better than the immersion treatment for delivery of the chitosan‐conjugated DNA construct. This is probably the first report on the delivery of nanoconjugated DNA construct in M. rosenbergii, against nodavirus.  相似文献

Surveys for bacteriological analysis of larval samples to isolate the associated vibrios were carried out during 1985–1992, 2001 and 2002 in three different hatcheries located on the southwest coast of India. Vibrio isolates were examined for their species diversity, virulence based on haemolysis in prawn blood agar, lipolysis, proteolysis and chitinolysis and antibiotic sensitivity. Vibrio cholerae was the predominant species in the apparently healthy larval samples, whereas V. alginolyticus and V. vulnificus dominated during disease and morbidity. No correlation was found between the hydrolytic properties and haemolytic activity of the vibrios associated with the larvae. All isolates were resistant to erythromycin and resistance to oxytetracycline, ampicillin and streptomycin sulphate was prevalent among the larger section of the Vibrio population. This suggested that antibiotic application may not be of much use to protect the larvae from vibriosis. This is the first report on the diversity of Vibrio species associated with Macrobrachium rosenbergii larvae and their virulence characteristics based on haemolysis in prawn blood agar.  相似文献

Nutrition plays an important role in the development of all organisms and in particular that of farmed aquatic species where costs associated with feed can often exceed 60% of total production costs. Crustacean species in addition, have the added metabolic requirement for regular moulting to allow normal growth and this requires large amounts of energy in the form of sugars (glucose). The current study explored the capacity of the giant freshwater prawn to produce endogenous cellulose‐degrading enzymes capable of extracting nutrients (simple sugars) from plant sources in formulated feeds used in the prawn aquaculture industry. We identified a putative cellulase cDNA fragment in the target organism of 1576 base pairs in length of non‐microbial origin that after protein modelling exhibited a TM‐score of 0.916 with a described cellulase reported from another crustacean species. The functional role of cellulase enzymes is to hydrolyse cellulose to glucose and the fragment identified in GFP was highly expressed in the hepatopancreas, the site of primary food digestion and absorption in crustaceans. Hepatopancreatic tissue from Macrobrachium rosenbergii also showed active digestion of cellulose to glucose following an endoglucanase assay. Cellulase gene(s) are present in the genomes of many invertebrate taxa and play an active role in the conversion of cellulose to available energy. Identification and characterization of endogenous cellulase gene(s) in giant freshwater prawn can assist development of the culture industry because the findings confirm that potentially greater levels of low‐cost plant‐material could be included in artificial formulated diets in the future without necessarily compromising individual growth performance. Ultimately, this development may contribute to more efficient, cost‐effective production systems for freshwater prawn culture stocks that meet the animal's basic nutritional requirements and that also support good individual growth rates.  相似文献

Chemical compositions and thermal properties of cultured freshwater prawn meat (FPM) were studied. FPM contained 83.2% protein (dry basis), 62.7% of which was myofibrillar protein. Pepsin-soluble collagen (PSC) and insoluble collagen (ISC) contents were 0.63 and 0.32%, respectively. Both collagens were similar to type V collagen from porcine placenta. Glutamic acid/glutamine, arginine, aspartic acid/?asparagine, and lysine were abundant amino acids in FPM. Glycine, proline, hydroxyproline, and aspartic acid/?asparagine were predominant in both collagens. FPM exhibited thermal transition temperatures (Tmax) of 48.3 and 64.7°C, whereas Tmax of PSC and ISC were 43.0 and 46.0°C, respectively. Textural changes in FPM during post-mortem storage on ice are plausibly dependent upon its compositional and thermal properties.  相似文献

Abstract— Differences in culture duration, metamorphosis rate and the productivity in hatchery culture of M. rosenbergii using a closed system with natural and artificial brackish water were evaluated. Reuse of brackish water in more than one hatchery cycle was also evaluated. Natural and artificial brackish water constituted the two tested treatments, which were distributed in four independent recirculating systems (tank and respective biofilter). Four batches of cultures were conducted and the 2nd and 4th reused the water from the 1st and 3rd. respectively. Mean duration of the hatchery period was 28 d in natural brackish water and 31 d in artificial brackish water. The metamorphosis rate and the average productivity for the natural brackish water treatment were 74% and 60 postlarvael l, respectively, and values obtained with artificial brackish water were 55% and 44 postlarvae/L. The successful hatchery culture of M. rosenbergii in this specific artificial brackish water suggests its potential use in enterprises located far from the coast. Brackish water can be used in two consecutive cultures without a negative effect on productivity.  相似文献

ABSTRACT

In temperate regions, post-larvae freshwater prawn, Macrobrachium rosenbergii, are grown to more advanced sizes in tanks prior to pond stocking. This intermediate stage of culture is referred to as the nursery period. Little research has been conducted on different management practices on juvenile prawn growth and survival during this 30-60 day period. Survival during the nursery stage has been highly variable and may be related to the cannibalistic behavior of juvenile freshwater prawn when cultured at high densities in the nursery. The objective of this study was to evaluate the effect of stocking density, relative to the provision of artificial substrate (number of prawns/m2 of substrate), on growth, survival, and economic variables for freshwater prawn juveniles during nursery production. Post-larvae (0.01%0.00 g, n = 300) were stocked into nine 1900 L tanks, each provided with 20.5 m2 of artificial substrate in the form of horizontal layers of black plastic mesh (10 mm) spaced 5 cm apart. Tanks were randomly assigned one of three prawn densities (215, 430, or 860 post-larvae/m2 of substrate), which equated to 2.3,4.6 and 9.2 prawn/L, respectively. Juvenile prawn were fed a commercial trout diet (42% protein) at a percentage of body weight according to a feed rate table. Water quality was maintained using a flow rate of 8 L/min in each tank from a reservoir pond. Temperature was maintained at approximately 28°C using heat pumps. After 56 days there was no significant difference (P >0.05) in average weight of juvenile prawn stocked at the three densities (0 = 0.58%0.12 g, n = 9). Survival was significantly lower (P <0.05) for prawn stocked at 860 m2 (62%) than in those stocked at 430/m2 (78%) and 215/m2 (94%), which were not statistically different (P >0.05). Even with reduced survival, the highest stocking density produced the greatest number of nursed juveniles based on both tank volume (5.5/l) and surface area (530/m2), at the lowest average cost.  相似文献

The feeding energetics of the prawn Macrobrachium rosenbergii (De Man 1879) were examined with respect to the effect of dietary fibre. Adult animals were fed semi‐purified diets with 0%, 5%, 10% and 15%α‐cellulose to determine the maximum aerobic scope that could be elicited by the increase in metabolism as specific dynamic action (SDA). The excretion rates of ammonia were also measured. Prawns fed all diets had significantly higher respiration rates after feeding due to SDA. Feed intake was not related to metabolism rate. Both oxygen uptake and ammonia excretion were affected by feed intake, dietary cellulose and time after feeding. SDA was related to dietary fibre, indicated by increments varying from 37% to 386%. O:N ratios were influenced by feed intake, dietary cellulose levels and time after feeding. O:N ratios of between 10 and 49 indicate that the prawns predominately metabolize carbohydrate.  相似文献

Production of freshwater prawns has expanded in the US with some production in >20 states. Two different approaches to reducing production costs have evolved, one based on minimizing input costs and maximizing harvest sizes, the other on maximizing production (kg/ha). Profitability in both is largely dependent on optimizing selling prices by offering product forms which do not face direct import competition (i.e. live, fresh, whole) Prawns have been recognized as one of the most environmentally sustainable aquaculture crops, and attribute which is very appealing in many markets, especially those emphasizing local foods.  相似文献

Effects of harvesting methods (cull and batch) on population structure, growth, and yield of freshwater prawn, Macrobrachiurn rosenbergii, cultured at two densities (5/m2 and 10/m2) were evaluated. Females (67.4%) were more abundant than males (32.6%), but there was no effect of harvesting methods and stocking rates on sex ratio (P > 0.05). Mean survival rates ranged from 78.3 to 92.6%, being highest at a stocking density of 10/m2 and cull-harvested (P < 0.05), but were not different (P > 0.05) among three other treatments. After 196 days of culture, mean weights ranged from 25.3 to 29.9 g and were not significantly different from each other (P > 0.05). Mean weight of males was not hi her than that of f females (P > 0.05), except in one treatment (5/m2 , batch harvest) where males were heavier (P < 0.05). Maximum specific growth rate (SGR) was recorded for prawns cultured at a density of 5/m2 and cull-harvested (0.66%/day), followed by prawns at 10/m2 and cull-harvested (0.58%/day). Minimum SGR was found for prawns cultured at 10/m2 density and balch-harvested (0.43%/day). Maximum yield (2366 kg/ha and 2099 kg/ha) was recorded at the higher density (P < 0.05). Harvesting method had no effect on yield at any density (P > 0.05).  相似文献

ABSTRACT

Dopamine at concentrations of 0.15, 1.5, and 15.0 mg/kg was included in two different formulated diets that were fed to recently metamorphosed postlarvae of the freshwater prawn, Macrobrachium rosenbergii, for 60 days. Dopamine, in the form of dopamine hydrochlo-ride, was incorporated into the formulated diets by either solubilizing it in alcohol, followed by submersion of diet and evaporation, or inclusion in a lipid premix ingredient. The alcohol evaporation method at 0.15, 1.5, and 15.0 mg/kg significantly increased the percentage of individuals without external male characteristics. This research is the first reported use of dietary administration of dopamine to crustaceans, and the first documentation of the apparent dietary control of gonadal development and sex differentiation in crustaceans. Replication of results, a histological examination of a large sample of individuals, and continued monitoring of those individuals fed the dietary dopamine are needed to verify the apparent effect.  相似文献

An experiment was conducted for 60 d in a recirculatory system consisting of 20 glass aquariums (each of size 41 × 41 × 46 cm) to evaluate meat and bone meal (MBM) as partial replacement for fish meal (FM) in Macrobrachium rosenbergii postlarvae (PLs) with a view to develop an inexpensive nursery diet. Three nursery diets were formulated to contain 32% protein and to be as isoenergetic as possible. FM in these diets was progressively replaced with MBM (Diet 1 contained 25% FM and 14% MBM, Diet 2 contained 20% FM and 20% MBM, and Diet 3 contained 15% FM and 26% MBM). Inclusion levels of mustard oilcake and sesame meal in diets were kept fixed at 10%. A commercial shrimp nursery diet (30% protein) from Saudi Bangla Fish Feed Ltd., Mymensingh, was used as the reference diet (Diet 4). Each treatment had three replicates. PLs of M. rosenbergii (26 ± 0.02 mg) were stocked at the rate of 60 PLs (1 PL/L) in each aquarium. At the beginning, PLs were fed three times daily at a rate of 20% of the body weight and it was gradually reduced to 10% at Day 30. The ranges of water quality parameters in the system were as follows: temperature, 27–29 C; dissolved oxygen, 6.5–8.4 mg/L; pH, 6.8–8.3; and total ammonia, 0.01–0.15 mg/L. The result of the study showed that there was no significant difference (P > 0.05) between the weight gains of PLs fed Diet 1 (containing 14% MBM) and Diet 4 (reference diet) but was significantly higher than other dietary groups containing higher levels of MBM. Diets 1 and 4 showed significantly lower (P < 0.05) apparent food conversion ratios (1.74 and 1.73, respectively). The survival (%) of PLs ranged between 62 and 76% with PLs fed Diets 1 and 4 showing significantly higher (P < 0.05) survival. The result of the study demonstrated that the MBM could be included at 14% level in a nursery diet, and a diet containing 25% FM, 14% MBM, 10% sesame meal, and 10% mustard oilcake may be recommended for profitable production of PLs of M. rosenbergii.  相似文献

Effects of temperature on food consumption, growth and oxygen consumption were estimated for the freshwater prawn Macrobrachium rosenbergii postlarvae at 23 °C, 28 °C and 33 °C in the laboratory. The results showed that the animal's initial body weight had a close linear relationship with food consumption and growth. Food consumption increased directly with temperature. Consumption rates (C; mg day?1 ind?1 ) of the 28 °C and 33 °C groups were much higher than that of the 23 °C group (P < 0.001), and the 33 °C group's consumption rate was higher than that of the 28 °C group (P < 0.05). The relationship of food consumption with temperature and initial body weight (W; mg) could be described as: C = 0.0679W + 0.185t? 3.17. Growth increased significantly with increased temperature. The relationship among specific growth rate, temperature and initial body weight was as follows: SGR = ?0.110W + 0.213t + 0.176. However, temperature showed no effect on growth efficiency. Oxygen consumption increased significantly with temperature (P < 0.01). The weight‐specific oxygen consumption rates (mg O2 g?1 h?1) at 23 °C, 28 °C and 33 °C were 0.83, 1.16 and 1.49 mg O2 g?1 h?1 for 61.92 mg M. rosenbergii.  相似文献

Macrobrachium rosenbergii (de Man 1879) juveniles (0.4 g) were cultured in experimental cages (L × W × H: 2.5 × 1 × 1 m) in Laguna de Bay, the largest lake in the Philippines. The following stocking densities at four replicates each were used: 15, 30, 60 and 90 prawns m−2 of cage bottom. The mean sizes at harvest after 5 months of culture ranged from 14.3 g for the highest stocking density to 26.3 g for the lowest. The mean size at harvest, daily growth rate and size class distribution were significantly influenced by stocking density, with those at the lowest stocking density showing significantly better growth and overall proportion of larger prawns. Heterogeneous individual growth (HIG) was fairly evident in all treatments. The percentage of blue‐clawed males was not influenced by treatment but the mean weight was significantly higher in the lower stocking densities. Both the percentage and mean weight of berried females were significantly higher in the lowest stocking density. Survival was the highest in the lower stocking densities (55.3%, 54.0%, 52.7% and 36.9% for 15, 30, 60 and 90 prawns m−2 respectively). Feed conversion ratio (FCR) improved with decreasing stocking density, ranging from 2.1 to 3. As expected, yield per cropping increased with stocking density and ranged from 450 to 1089 g m−2 yr−1 of actual cage area. Production values obtained in the cage cultured M. rosenbergii were comparable to or even higher than those reported from pond culture, given that the stocking densities used in this study were generally higher than in ponds. The results show that the farming of M. rosenbergii in cages in lakes is a viable alternative to pond culture and has the potential of improve aquaculture production in lakeshore fish farming communities.  相似文献

The effects and interactions of added artificial substrate with different stocking densities of prawns were evaluated. Juvenile prawns (0.2 ± 0.1 g) were stocked into 12 0.04-ha ponds at 60,000ha and 120,000/ha with and without substrate. Added substrate consisted of horizontal plastic mesh and strips of oyster netting sutlicient to increase available surface area 80%. There were no statistically significant ( P > 0.05) interactions between stocking density and presence of added substrate, allowing these main effects to be compared separately. Increasing stocking density produced a significant increase ( P < 0.01) in total production but a significant decrease ( P < 0.01) in average prawn size and production ( P < 0.05) of market size prawns (>20 g, -27%; > 30 g, -56%). Added substrate also produced a significant increase ( P < 0.05) in total production (18%). However, average prawn size was not decreased and production of marketable shrimp was increased (>20 g, 25%; >30 g, 19%). Feed conversion ratios were significantly decreased (P < 0.01) by the presence of substrate. Increased stocking density significantly increased ( P < 0.05) the percentage of males which were small males (SM) and decreased ( P < 0.01) the percentage of orange claw males (OC), but had no impact ( P > 0.05) on numbers of reproductive (RF) and virgin (VF) female morphotypes. Increased stocking density also produced a significant decrease ( P < 0.05) in average size of OC, RF, and VF morphotypes while the addition of substrate had no statistically significant impact (P > 0.05) on the number or size of different morphotypes. The mathematical relationship between available surface area and average prawn size should be determined to produce recommended inclusion rates for added substrate based on desired levels of total production, stocking rates, and optimum market sizes.  相似文献

Meat yield of 233 individuals of giant freshwater prawn, Macrobrachium rosenbergii (de Man, 1879), and their morphometric relationships were examined. Morphometric parameters of different length and weight measurements were recorded for both the sexes separately and subjected to analysis. The meat yield of prawns was size and sex dependent with females showing significantly greater (P < 0.05) tail meat yield than males. The tail meat as a body weight percentage ranges from 26.12 to 40.75 and 32.97 to 42.11 in males and females, respectively. Much of the differences in yield are related to the size and weight of the head and claws. The head weight of males was observed to be higher compared to females in all the size classes. Regression slopes of total length versus body weight and total length versus head weight were significantly higher than critical isometric value indicating high positive allometric growth in both the sexes, with males showing significantly higher (P < 0.05) slope than females. The relationship parameters of total length versus tail weight and total length versus tail meat showed a positive allometric growth in females, whereas it is isometry and negative allometry in males. The morphometric relationship of tail length versus all weight measurements showed positive allometry and negative allometry in females and males, respectively.  相似文献

Biochemical assays and substrate SDS-PAGE were conducted to partially characterize and identify various types of proteases present in the digestive tract of PL15 giant freshwater prawn ( Macrobrachium rosenbergii ). Casein hydrolytic assay of the enzyme extracts showed major proteolytic activities at pH 3.0, 6.0 and 9.0, while assay of preincubated enzyme extracts with phenylmethylsulphonyl fluoride (PMSF), a serine protease inhibitor produced a 33.17% reduction in alkaline protease activity. When specific inhibitors tosyl-lysine chloromethyl ketone and tosyl-phenylalanine chloromethyl ketone were used, they resulted in a reduction in activity of proteases in the enzyme extracts by 82.41% and 55.03%, respectively, confirming the presence of trypsin and chymotrypsin, while ethylenediamine tetraacetic acid produced protease activity reduction in 33.92% showing the presence of metalloproteases in the digestive tract of the prawn. Further characterization of the alkaline proteases using SDS-PAGE technique, after incubating the extract in the presence or absence of specific inhibitors, produced six bands corresponding to molecular masses of between 13.48 and 136.1 kDa; two trypsin bands of 13.48 and 36.4 kDa, three chymotrypsin bands in the range of 23.0–73.4 kDa and one for metalloprotease of 136.1 kDa, all of which were identified from a zymogram. This study suggests that protein digestion in M. rosenbergii is initiated by an acid protease followed by a combination of action of alkaline proteases: trypsin, chymotrypsin and metalloproteases.  相似文献
