Effect of sex and exogenous porcine somatotropin on longissimus muscle fiber characteristics of growing pigs 总被引:3,自引:0,他引:3
Thirty-seven pigs with an initial live weight of 60 kg were used to investigate the effects of daily exogenous porcine somatotropin (pST) administration at two dose levels (0 and 100 micrograms.kg-1.d-1) for a 31-d period on muscle fiber characteristics and meat tenderness of boars, gilts and barrows. Excipient boars and gilts had more alpha W and fewer alpha R fibers than did those receiving pST. The percentage of muscle fiber type for barrows was not affected by pST treatment. The administration of pST resulted in an increase in muscle fiber size for all three fiber types in all three sexes, but these changes were of greater magnitude in barrows (31.8%) and gilts (27.8%) than in boars (9.3%). Somatotropin negated the intrinsic sex effect differences in fiber area of the pigs. There was no difference in tenderness among excipient boars, barrows and gilts; however, with the inclusion of pST, shear force decreased in boars and gilts and increased in barrows. A high proportion of the pST-treated pigs contained giant fibers in the longissimus muscle. Furthermore, a small proportion of the pST-treated pigs exhibited pale, soft, exudative muscle. Whether the giant fiber anomalies occurred through increased muscle activity or from fibers undergoing degenerative changes was not determined. 相似文献

Thirty two barrows were used in a 2 x 3 factorial arrangement to evaluate the effects of porcine somatotropin (pST) administration (USDA-pGH-B1; 0 and 100 micrograms.kg BW-1.d-1) and feed intake (FI; ad libitum [A], 1.64 [R1:80% of A] and 1.38 [R2:60% of A] kg/d) on longissimus (LM) muscle fiber characteristics of pigs growing from 25 to 55 kg live weight. The administration of pST resulted in an increase (P less than .05) in muscle fiber size (all three fiber types) and shear-force (17.1%). The percentage of muscle fiber types was not affected by pST treatment. Feeding of the diet at restricted levels (R1 and R2) resulted in smaller (P less than .05) beta R and alpha W fibers compared with feeding ad libitum. Pigs with ad libitum access to their diet had fewer alpha R fibers and more alpha W fibers in the LM compared with those fed at R1 and R2 levels. Restricted feed intake (R1 and R2) resulted in lower (P less than .05) shear-force values compared with ad libitum feeding. The present study indicates that pST administration of young barrows results in an increase in muscle hypertrophy. Restricted FI reduces both transformation from alpha R to alpha W fibers and muscle fiber hypertrophy. 相似文献

In pigs, a paternally (pat) imprinted mutation in the IGF-II gene is associated with increased muscle mass and decreased backfat thickness. The aim of this study was to determine whether this mutation influenced cellular, biochemical and metabolic features of skeletal muscle and adipose tissue. Muscle (trapezius) and subcutaneous adipose tissue (SCAT) were collected from pigs (106kg) carrying (Qpat, n=6) or not carrying (qpat, n=7) the mutation. Adipocytes were isolated from those tissues by collagenase treatment. Lipid content and activity of lipogenic enzymes were determined using standard assays. Gene expression levels were determined by real-time PCR. Levels of IGF-II mRNA were higher (P<0.01) in muscle of Qpat than in that of qpat pigs, but they did not differ significantly between the two groups in SCAT. Whereas levels of IGF-I mRNA in muscle were similar in both groups, they were higher (P<0.05) in SCAT of Qpat pigs than in that of qpat pigs. Muscle lipid content and intramuscular adipocyte diameters were not influenced significantly by the IGF-II genotype. In SCAT, the reduction of backfat thickness in Qpat pigs compared with qpat pigs was associated with lower (P<0.05) lipid content and smaller (P<0.05) adipocytes, with no significant genotype-effects on expressions and/or activities of lipogenic enzymes. In summary, our results suggest that the IGF-II mutation altered body composition in pigs by favoring myofiber hypertrophy and repressing adipose cell development in SCAT. 相似文献

Twenty-four barrows were used to investigate the effects of exogenous porcine somatotropin (pST) administration (0 and 100 micrograms.kg-1.d-1) between 30 and 60 kg on longissimus muscle morphology and meat tenderness of pigs grown to 90 kg. Administration of pST was by daily i.m. injection. Pigs were fed a fortified diet in restricted amounts between 30 and 60 kg and had ad libitum access to this diet from 60 to 90 kg. Excipient pigs had fewer alpha R fibers and more alpha W fibers than pST-treated pigs (P less than .05). Administration of pST increased (P less than .05) muscle fiber area for all three fiber types at both the 60 kg (34%) and 90 kg (29%) slaughter weights. Furthermore, pST administration increased shear-force (30% at 60 kg; 19% at 90 kg) of the longissimus muscle, indicating that pST administration reduced meat tenderness. All the pST-treated pigs exhibited the "giant fiber syndrome" and a high proportion (62%) of pST-treated pigs exhibited pale, soft, exudative muscle. Whether the giant fiber anomaly resulted from increased muscle protein accretion rate or fiber degeneration is unknown. Results indicated that the stimulatory effects of pST on muscle growth are sustained following cessation of hormone treatment. However, the incidence of pale, soft, exudative muscle in 62% of the pST-treated pigs indicates that pST potentially has undesirable side effects. 相似文献

Eighteen steers were used to evaluate the effect of supplemental corn oil level to steers grazing endophyte-free tall fescue on fatty acid composition of LM, stearoyl CoA desaturase (SCD) activity and expression as well as cellularity in s.c. adipose. Corn oil was supplemented (g/kg of BW) at 0 (none), 0.75 (medium), and 1.5 (high). Cottonseed hulls were used as a carrier for the corn oil and were supplemented according to pasture availability (0.7 to 1% of BW). Steers were finished on a rotationally grazed, tall fescue pasture for 116 d. Fatty acid composition of LM, s.c. adipose, and diet was determined by GLC. Total linoleic acid intake increased linearly (P < 0.01) with corn oil supplementation (90.7, 265.1, and 406.7 g in none, medium, and high, respectively). Oil supplementation linearly reduced (P < 0.05) myristic, palmitic, and linolenic acid percentage in LM and s.c. adipose. Vaccenic acid (C18:1 t11; VA) percentage was 46 and 32% greater (linear, P = 0.02; quadratic, P = 0.01) for medium and high, respectively, than none, regardless of tissue. Effect of oil supplementation on CLA cis-9, trans-11 was affected by type of adipose tissue (P < 0.01). In the LM, CLA cis-9, trans-11 isomer was 25% greater for medium than for none and intermediate for high, whereas CLA cis-9, trans-11 CLA isomer was 48 and 33% greater in s.c. adipose tissue for medium and high than for none, respectively. Corn oil linearly increased (P = 0.01) trans-10 octadecenoic acid and CLA trans-10, cis-12; however, values were low (<0.35 and <0.035% of total fatty acids, respectively). Oil supplementation did not change (P > 0.05) the percentage of total SFA, MUFA, or PUFA but linearly increased (P = 0.03) n-6:n-3 ratio from 2.4 to 2.9 in none and high, respectively. Among tissues, total SFA and MUFA were greater in s.c. adipose than LM, whereas total PUFA, n-6, and n-3 fatty acids and the n-6:n-3 ratio were lower. Trans-10 octadecenoic acid, VA, and CLA trans-10, cis-12 were greater (P < 0.01) in s.c. adipose than in LM. Oil supplementation did not alter (P > 0.05) stearoyl CoA desaturase activity or mRNA expression. Corn oil supplementation to grazing steers reduced the percentages of highly atherogenic fatty acids (myristic and palmitic acids) and increased the percentages of antiatherogenic and anticarcinogenic fatty acids (VA and cis-9, trans-11 CLA). 相似文献

Forty crossbred wethers (BW = 28.7 kg) were used to evaluate the effects on LM lipid composition of diets containing high and low levels of vitamin A. Four treatments arranged as a 2 x 2 factorial with a completely random design were investigated: backgrounding (BG) and finishing (FN) with no supplemental vitamin A (LL); BG with no supplemental vitamin A and FN with high vitamin A (6,600 IU/kg of diet, as fed) supplementation (LH); BG with high vitamin A supplementation and FN with no vitamin A supplementation (HL); and BG and FN with high vitamin A (HH) supplementation. Diets included cracked corn (62.4%), soybean meal (16.0%), cottonseed hull pellets (14.8%), and supplement (7%), and contained <100 IU of vitamin A/kg (as fed) from carotenes before vitamin A was added. During the BG period (d 1 to 56), feed intake was restricted to achieve 0.22 kg of ADG. During the FN period (d 57 to 112), lambs consumed the same diet ad libitum. Lambs were weighed every 14 d, and blood was sampled every 28 d to evaluate changes in serum fatty acids and vitamin A levels. Lambs were slaughtered after 112 d. Lipid composition was determined for liver and LM. There were no treatment differences (P > 0.05) in feed intake, ADG, or final BW. Carcass weights were not affected by vitamin A treatment (P > 0.20), although backfat thickness tended to be different between HL and LL lambs (0.80 vs. 0.64 cm, respectively; P = 0.08). Carcasses from the HH group had greater (P < 0.05) marbling scores than those from the LL group (514 vs. 459) and had 25.8% more extractable intramuscular lipids (3.88 vs. 3.08% for HH and LL, respectively; P < 0.05); the LH and HL treatments were intermediate. Interestingly, the LL group had the greatest increase in serum fatty acids throughout the experimental period (change of 127 vs. 41 microg/g for LL and HH, respectively; P < 0.01). The degree of saturation of fatty acids was not affected by treatment (P = 0.18) in the serum but was affected in the longissimus thoracis fat. Oleic acid increased and linoleic acid decreased in the longissimus thoracis of HH-treated lambs (P < 0.02). These data suggest that increases in total intramuscular lipids may be achieved with high levels of vitamin A supplementation for 112 d in young lambs. 相似文献

Sera obtained from pigs before and after subtherapeutic levels of ASP250 supplementation (pre and post serum pools) have been subjected to comparative fractionation by using gel filtration and affinity chromatography on immobilized Cibacron Blue F3G-A. Comparable serum fractions obtained from pre- and post-ASP250 blood sera were assayed in muscle cell culture bioassays designed to measure their effect on proliferation. Pre- and post-ASP250 sera were subjected to gel filtration and divided into the following fractions: fraction 1, Kav less than .17; fraction 2, Kav = .17 to .41; fraction 3, Kav = .41 to .59. Post-ASP250 fractions 2 and 3 increased proliferation rate in cultured muscle cells to a greater extent than comparable pre-ASP250 fractions (P less than .001). Chromatography of fraction 3 on immobilized Cibacron Blue F3G-A showed that both pre- and post-ASP250 fraction 3 contained a putative inhibitor of myogenic cell proliferation as well as mitogenic factors. However, negative growth factor activity was greater in pre-ASP250 fraction 3 than in post-ASP250 fraction 3 (P less than .05). Additionally, positive growth factor activity was lower in pre-ASP250 fraction 3 than in post-ASP250 fraction 3 (P less than .05). These data suggest that levels and(or) activities of both positive and negative muscle growth factors in serum may be altered by the addition of antimicrobials to the diets of growing pigs. 相似文献

Forty-eight Yorkshire cross pigs of three stress susceptibility classes (stress positive, stress-carrier, and stress negative) were injected daily with porcine somatotropin (pST; 4 mg/d) or placebo. Each pig was injected in the neck once daily until taken off test, starting when the pigs weighed 59 kg. Porcine somatotropin treatment was terminated at weekly intervals as individual pigs reached 109 kg, but animals continued to be fed for six additional days to allow for required withdrawal time. The effect of pST and stress classification on the sensory, physical, chemical, and processing characteristics of cured semimembranosus (SM) muscle was evaluated. Treatment of animals with pST had no effect on the sensory scores, lipid and protein content, cooking yields, or color values of SM muscle slices. Semimembranosus muscles from stress-positive animals, however, had reduced sensory scores for texture, flavor, and overall palatability. Semimembranosus muscles from stress-positive pigs also had smaller cooking yields and greater Hunter a and b values of processed slices. The greater Hunter a and b values suggested that the color of these slices were redder and yellower than the color of SM muscle slices from negative and carrier animals. Semimembranosus muscles from stress-susceptible animals also had a significantly lower lipid content. Treatment of animals with pST did not significantly alter sensory, chemical, or processing characteristics of SM muscle slices from these animals. 相似文献

Circular RNAs (circRNAs) participated in regulation of lipid metabolism; however, its functional role on castration-induced lipid deposition has not been deeply researched. So in this research, we firstly compared circRNAs expressional differences in subcutaneous adipose tissue between intact and castrated male Huainan pigs. A total of 6116 differentially expressed circRNAs (DECs) were detected between these two groups (|log2foldchange| ≥ 1 and padj ≤ 0.05); GO and KEGG analysis showed that their parent genes were mainly enriched in metabolism-related pathway. And TGF-beta, insulin, AMPK, and MAPK pathways might play vital role in castration-induced lipid deposition. The miRNAs enriched in the constructed circRNA–miRNA network were mainly participated in adipogenesis and lipid metabolism, such as miR-143a-3p, miR-378, and miR-195. And it was verified that testosterone upregulated miR-181a but downregulated circ_0005912 expression in a dose-dependent manner in porcine intramuscular adipocytes, and overexpression of miR-181a inhibited circ_0005912. Taken together, these DECs may participate in the regulation of lipid metabolism after castration by reaction with miRNAs, which indicated the novel role of circRNAs in castration-induced lipid deposition. 相似文献

Sixty-one finishing pigs (53.4 kg) were fed a control diet (containing soybean meal) or diets containing 20% intact canola (IC) or 20% ground canola (GC) for 8 wk. Diets were not isocaloric. Daily gain and feed efficiency were not affected by dietary treatment, but pigs fed GC ate less than pigs fed either IC or the control diet. Carcass measurements, obtained on 43 of the pigs, were not affected by diet. For 27 pigs, fatty acid composition of perirenal adipose tissue (PRF), subcutaneous adipose tissue (SCF), and longissimus muscle (LDM) was analyzed. Nine pigs (three per treatment) were randomly selected for fatty acid composition analysis of intramuscular adipose tissue (IMF) and for cholesterol analysis of several tissues. Pigs fed canola had greater (P less than .05) proportions of mono- and polyunsaturated fatty acids and less (P less than .05) saturated fatty acids in PRF and SCF. The differences were more pronounced for PRF than for SCF. In the LDM, pigs fed canola tended to have elevated levels of unsaturated fatty acids at the expense of the saturated fatty acids, but this effect was significant for linolenic acid only. The fatty acid composition of IMF was not affected by diet (P greater than .05). Diet did not alter the cholesterol content of the tissues, but cholesterol in IMF was higher (P less than .05) than in PRF, SCF, and LDM. In conclusion, 20% IC or GC did not alter growth performance or carcass characteristics of pigs. As one of the key points related to meat quality, skeletal muscle fibre type is determined by energy metabolism and genetic factors, but its transformation could be also greatly influenced by many factors. Thymol, the primary effective ingredients of thyme, is well known for its anti-oxidation and anti-inflammatory, while little is known about its effect on skeletal muscle oxidative metabolism and fibre type switch. Therefore, in order to investigate its effects and possibility to be applied in livestock production, 36 150-day-old fattening Pigs were fed with different diet for six-week experiment. As a result, the drip loss ratio of longissimus dorsi (LD) was significantly reduced (p < .05). Oxidative metabolism-related enzyme activity, the mRNA levels and protein expression of COX5B and PGC1α, mRNA level of myosin heavy chain I (MyHC I) and protein level of MyHC IIa were significantly upregulated (p < .05). While compared with control group, the protein expression of MyHC IIb was significantly decreased (p < .05). The result revealed that thymol could promote the oxidative metabolism in the muscle of pigs and improve the meat quality to a certain extent. 相似文献

Because this laboratory has been able to demonstrate only a small and somewhat inconsistent stimulation of glucose metabolism by insulin in porcine adipose tissue in vitro, the tissue was preincubated with insulin to attempt to enhance the hormone effect. Preincubation with or without insulin did not increase insulin stimulation. Furthermore, insulin did not stimulate triacylglycerol biosynthesis. Adrenergic hormones stimulated lipolysis in porcine adipose tissue in vitro. Several analogs of norepinephrine incubated with porcine adipose tissue in vitro did not inhibit glucose incorporation into CO2 or total lipids, in contrast to inhibition observed in adipose tissue from other species. Isoproterenol inhibited glycerol-3-phosphate incorporation into lipids; the maximal inhibition was 50% for the initial stages of the pathway. Palmitate incorporation into lipids also was inhibited 50% by isoproterenol but this may have been an artifact. Preincubation of adipose tissue, with no exogenous hormone, might decrease the concentration of endogenous adrenergic hormones and thus make the tissue more responsive to exogenous adrenergic hormones. Preincubation of porcine adipose tissue did not consistently lower the basal lipolytic rate but enhanced the stimulated lipolytic rate; the mechanism is not known. These experiments provide no evidence that preincubation is beneficial to measurement of lipolysis or glucose metabolism in porcine adipose tissue in vitro. 相似文献

This study compared the adipogenic potential of porcine stromal-vascular (S-V) cells from semitendinosus muscles and s.c. adipose tissue using thiazolidinediones. Stromal-vascular cells were obtained from s.c. adipose tissue and both semitendinosus muscles from 5- to 7-d-old pigs after collagenase digestion. Preadipocyte recruitment was measured using immunohistological evaluation for AD-3, a preadipocyte antibody. Ciglitazone increased the number of preadipocytes in adipose tissue but not semitendinosus muscle S-V cell cultures, whereas 10 microM troglitazone increased preadipocyte abundance in both adipose and muscle S-V cultures by approximately 3-fold (P < 0.05). Increasing troglitazone doses did not further increase preadipocyte number. Increases in preadipocytes were paralleled by increases in CCAAT/enhancer-binding protein alpha (C/EBPalpha) and peroxisome proliferator-activated receptor gamma (PPARgamma) positive cells in adipose tissue S-V cultures, whereas PPARgamma-reactive but not C/EBPalpha-reactive cells were increased in muscle S-V cultures treated with 10 microM troglitazone. Additionally, troglitazone treatment did not increase lipid content in s.c. adipose tissue or muscle S-V cell cultures. Cells plated on laminin-precoated culture dishes were used to determine whether troglitazone influenced adipogenesis or myogenesis in cocultures from muscle S-V cells. There was no effect on the number of myotubes or the average number of nuclei per myotube, suggesting myogenesis was not impaired by troglitazone treatment. These results suggest that regulation of intramuscular adipogenesis differs from that of subcutaneous adipogenesis. 相似文献

Vitamin A (retinoic acid) is known to be an adipogenic factor influencing both in vitro and in vivo cell development. This study aimed to determine its effect on lamb adipose tissue development during the early phase of postnatal development until 100 d of age. Male lambs (n = 24) of the Rasa Aragonesa breed were used. At birth, lambs were assigned to 1 of 2 experimental groups: 1) the control (C) group, which received feed without vitamin A supplementation, and 2) the vitamin A (V) group, which received a supplement of 500,000 IU/animal twice per week from birth to slaughter. The effect of vitamin A supplementation was studied at 16.8 +/- 0.35 kg of BW (58 +/- 0.7 d of age) and at 27.8 +/- 0.78 kg of BW (101 +/- 6.5 d of age). The variables of lamb growth, carcass, LM area, and lipid content were analyzed. To study adipose tissue development, the amount of adipose tissue accumulated, the size and number of adipocytes, and lipogenic enzyme activities (glycerol 3-phosphate dehydrogenase, fatty acid synthase, and glucose 6-phosphate dehydrogenase) of the omental, perirenal, and s.c. depots were quantified. Results showed that vitamin A supplementation had no influence on growth, carcass variables, LM area, and lipid content during lamb growth but that the number of adipocytes in the perirenal depot was 30% greater in lambs of the V group (P < 0.05) and that these lambs had smaller adipocytes in the omental and perirenal depots (P = 0.06) at 28 kg of BW (101 d of age). These results suggest that the intake of this level of vitamin A during the whole period of growth of the lambs influenced the processes of hyperplasia and hypertrophy in the different adipose depots, depending on their degree of maturity. 相似文献

Effect of porcine somatotropin on in vivo glucose kinetics and lipogenesis in growing pigs. 总被引:7,自引:0,他引:7
Crossbred barrows were used for in vivo studies investigating hormonal regulation of lipogenesis. The first experiment examined an in vivo method for determining rates of lipogenesis. Three barrows were infused with [U-14C]glucose and incorporation of radioactivity into triglycerides was determined in up to five biopsies of subcutaneous adipose tissue obtained over 7 h. Incorporation was linear after blood glucose specific radioactivity had reached a plateau and was constant over the entire infusion. For the second experiment, eight pigs (71 +/- 2.5 kg) were allocated to one of two treatments involving daily injections of excipient (control) or porcine somatotropin (pST; 120 micrograms/kg of BW). On d 10, beginning 15 h after injection, glucose incorporation into adipose tissue lipid was determined under both basal and hyperinsulinemic/euglycemic conditions. Basal glucose incorporation into lipid, particularly fatty acids, was markedly reduced (greater than 90%) during pST treatment. Although glucose incorporation was increased to a similar extent in both groups by hyperinsulinemia, the pST-treated pigs still exhibited markedly lower rates. Based on kinetic data, the decrease in lipid accretion of pST-treated pigs was primarily the result of a decrease in the rate of de novo synthesis. Furthermore, the reductions in glucose incorporation into fatty acids, glucose irreversible loss rate, and feed intake that occur with pST treatment were quantitatively similar. 相似文献
