通过选择性培养基、PDA—RB平板显色和PDA-Bavendamm平板显色反应从土壤样品中反复筛选获得5株产木质素降解酶能力较强的菌株,经鉴定为白腐菌属。小麦秸秆降解试验显示,菌株P3对秸秆中木质素的降解率最高,培养15d后达到11.47%,其次为菌株P5,降解率为8.12%,为进一步的研究奠定了基础。 相似文献

平菇处理小麦和水稻秸秆对纤维成分和体外瘤胃发酵特征的影响 总被引:1,自引:0,他引:1
本试验旨在研究平菇处理小麦和水稻秸秆对纤维成分和体外瘤胃发酵特征的影响。选用未处理和平菇处理的小麦和水稻秸秆作为发酵底物,通过瘘管采集3只成年湘东黑山羊的瘤胃液,进行体外模拟瘤胃发酵72 h试验。结果表明:平菇处理小麦秸秆显著降低酸性洗涤木质素含量(P0.05),显著提高72 h总产气量、干物质降解率和甲烷产量(P0.05);平菇处理水稻秸秆显著提高酸性洗涤木质素含量和氢气产量及含量(P0.05),显著降低半纤维素含量、72 h总产气量、中性洗涤纤维降解率、挥发性脂肪酸浓度和甲烷产量及含量(P0.05);此外,平菇处理小麦和水稻秸秆显著降低乙酸比例(P0.05),显著提高丙酸比例(P0.05)。由此得出,平菇处理小麦秸秆可降低木质素含量,有助于瘤胃纤维降解和丙酸生成;平菇处理水稻秸秆会增加木质素沉积,抑制瘤胃纤维降解以及挥发性脂肪酸和甲烷生成,造成氢气累积和丙酸生成。 相似文献

木质素是仅次于纤维素的一种最丰富可再生的纤维资源,它的降解在碳素循环及植物纤维素的有效利用中占有非常重要的地位。自然界中,木质素的完全降解是真菌、细菌、放线菌及相应微生物群落共同作用的结果,其中真菌起着主导作用,降解木质素的真菌主要有3类:白腐菌、褐腐菌和软腐菌,其中白腐菌降解木质素的能力相对较强,而且分泌胞外氧化酶降解木质素时不产生色素,因此应用最广,研究也最深入。本文介绍了白腐真菌降解木质素的主要酶系及生理生化特性,并对白腐真菌对木质素的降解机理及木质素降解酶系的分子生物学研究进展进行论述。 相似文献

Background: The present work investigated the influence of lignin content and composition in the fungal treatment of lignocellulosic biomass in order to improve rumen degradability. Wheat straw and wood chips,differing in lignin composition, were treated with Lentinula edodes for 0, 2, 4, 8 and 12 wk and the changes occurring during fungal degradation were analyzed using pyrolysis-gas chromatography-mass spectrometry and detergent fiber analysis.Results: L. edodes preferentially degraded lignin, with only limited cellulose degradation, in wheat straw and wood chips, leaving a substrate enriched in cellulose. Syringyl(S)-lignin units were preferentially degraded than guaiacyl(G)-lignin units, resulting in a decreased S/G ratio. A decreasing S/G ratio(wheat straw: r =-0.72, wood chips: r =-0.75) and selective lignin degradation(wheat straw: r =-0.69, wood chips: r =-0.88) were correlated with in vitro gas production(IVGP), a good indicator for rumen degradability.Conclusions: L. edodes treatment increased the IVGP of wheat straw and wood chips. Effects on IVGP were similar for wheat straw and wood chips indicating that lignin content and 3D-structure of cell walls influence in vitro rumen degradability more than lignin composition. 相似文献

试验旨在研究温度对侧耳菌处理玉米秸秆饲料营养价值及纤维分解酶活性的影响。在16,20和24℃温度条件下,采用侧耳菌处理玉米秸秆饲料,测定处理秸秆纤维素、半纤维和木质素含量、纤维素和木质素分解酶活性及秸秆养分瘤胃消失率。结果表明,在3个温度条件下,处理秸秆的纤维素、半纤维素和木质素含量均随时间的延长而降低。16℃组3种纤维含量降低速度(P<0.05)、酶活性(P<0.01)及干物质瘤胃消失率(DMD48)(P<0.05)显著低于20和24℃组,而20℃组与24℃组以上指标没有显著差异,这2组秸秆DMD48随着时间延长而增加,21 d后趋于稳定。结果显示温度显著影响侧耳菌处理秸秆饲料的纤维含量和营养价值,16℃处理秸秆显著低于20~24℃,而20℃与24℃处理秸秆饲料的纤维含量和营养价值差异不显著。 相似文献

试验依据国家饲料卫生标准(GB13078—2001)中的检测方法对平菇不同生长阶段培养基的基质组分进行了检测和安全性评定。通过对平菇不同生长阶段培养基(以棉籽壳和玉米芯为主)基质组分的检测及对基质饲料价值的评定,结果发现,第4潮菌糠用作饲料营养价值较高且生产成本较低。此时粗蛋白质含量为10.10%,较初始培养基提高了59.31%,而粗纤维含量降低了29.57%,木质素、纤维素和半纤维素的降解率分别提高到63.62%、46.60%和77.06%;并依据国家饲料卫生标准要求对第4潮菌糠中的游离棉酚、黄曲霉毒素B1和重金属含量等指标进行了检测,其中游离棉酚的含量仅为67.90 mg/kg,未测出黄曲霉毒素B1的含量(<10 μg/kg),铅、氟、铬、镉、汞的含量均符合国家饲料安全标准。因此,用第4潮菌糠制备反刍动物粗饲料可满足要求,切实可行。 相似文献

Degradation of wheat straw and alkaline hydrogen peroxide-treated wheat straw by Ruminococcus albus 8 and Ruminococcus flavefaciens FD-1 总被引:3,自引:0,他引:3
Degradation of wheat straw (WS) and alkaline hydrogen peroxide (AHP)-treated wheat straw (AHPWS) by Ruminococcus albus 8 and Ruminococcus flavefaciens FD-1 was determined by measuring the growth (OD600) of each bacterium and determining DM disappearance (DMD) of the substrate. Complex medium and defined medium with or without the addition of phenylpropanoic acid (PPA) and phenylacetic acid (PAA) were used. Tubes were incubated at 39 degrees C for 8 d. Both OD600 and DMD indicated that AHPWS was degraded to a much greater extent by either bacterium (R. flavefaciens FD-1, 60.8 +/- 1.8% and R. albus 8, 42.3 +/- 3.5%) vs untreated WS (R. flavefaciens FD-1, 16.5 +/- 1.8% and R. albus 8, 8.6 +/- 6%) in the complex medium. Most degradation occurred between d 1 and 4. With the complex medium, addition of PPA and PAA did not stimulate degradation by either bacterium. When the defined medium was used, the addition of PPA and PAA enhanced (P less than .05) degradation of AHPWS (39.6 +/- 2.6%) vs AHPWS with no added PPA and PAA (24.9 +/- 7.6%) by R. albus 8. There was no synergistic effect on degradation when the two species were co-cultured with either WS or AHPWS as the substrate. No effect of PPA and PAA on disappearance of AHPWS was observed for R. flavefaciens FD-1 or when the two bacteria were grown together. Dry matter disappearance analysis showed that R. flavefaciens FD-1 degraded AHPWS more rapidly (6.1 mg/d) than R. albus 8 did (4.2 mg/d) in complex medium.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献
