Sphaeropsis sapinea is an important latent pathogen of Pinus spp., outbreaks of which have a considerable impact on plantations. This study considers the population diversity and distribution of S. sapinea in northern Spain at different spatial scales from single plantations to a wide area covered by Pinus radiata trees. Estimation of genotypic diversity is an important component of the analysis of the genetic structure of plant pathogen populations. Ten simple sequence repeat (SSR) markers were used, together with vegetative compatibility tests, to study the genetic diversity among S. sapinea isolates. Polymorphism analysis at SSR loci is a simple and direct approach for estimating the genetic diversity of S. sapinea isolates. From a total of 86 isolates collected from four different areas, 14 microsatellite haplotypes and 13 vegetative compatibility groups (VCGs) were identified. The percentage of maximum genotypic diversity, based on Stoddart and Taylor's index, for microsatellites of the northern Spain population ranged from 14.6% to 38.1% and from 8.0% to 29.4% for VCGs. Analysis of these markers and vegetative compatibility groups confirmed that S. sapinea reproduces mainly asexually due to its reduced genotypic diversity in spatially close populations. Isolates of S. sapinea from northern Spain populations were predominantly monomorphic at the tested SSR loci. Vegetative compatibility groups also indicate a low level of genetic variability in these samples, which appear to be clonal.  相似文献

We compared the water-use characteristics of co-occurring mature Quercus cerris L. and Quercus pubescens Willd. trees growing in resource-limited (mainly water) hilly habitats in Tuscany, Italy. The species differed in their distribution along soil water gradients and in their access to, and use of, water, even though the study year was wetter than average, though with a summer drought. Compared with Q. cerris, Q. pubescens had greater access to soil water (less negative predawn water potentials) and a more conservative water-use strategy based on its relatively low stomatal conductance, high instantaneous water-use efficiency, less negative midday water potential and high soil-to-leaf hydraulic conductance. Quercus cerris had less conservative water-use characteristics than Q. pubescens, exhibiting relatively high stomatal conductance, low instantaneous water-use efficiency, more negative midday water potentials and low soil-to-leaf hydraulic conductance; however, Q. cerris had higher photosynthetic rates than Q. pubescens. Photosynthesis and stomatal conductance were positively correlated in both species. Although a strong correlation between ring widths and precipitation patterns was not found, some dry periods influenced ring-width growth. Quercus pubescens has always grown faster than Q. cerris, probably because of more efficient water use, although stand dynamics (driven by exogenous disturbance factors, including coppicing, browsing and competition) cannot be excluded. Ring-width variability, as well as tree-ring growth in dry years, which should be unaffected by stand dynamics, were higher in Q. pubescens than in Q. cerris. Moreover, Q. pubescens recovered completely after the drought in the seventies, even showing higher tree-ring growth than in the recent past, whereas Q. cerris showed a minor growth decline followed by a recovery to values comparable with those observed before the 1970s drought. Beginning in the early eighties, tree-ring growth decreased in both species, though Q. pubescens showed consistently higher values than Q. cerris. These differences can be explained by differences in water-use efficiency. Despite differences between the species in water use and water status, the results are consistent with the interpretation that both are drought tolerant, but that Q. pubescens is at an advantage on xeric ridges because of its greater ability to access soil water and use it more conservatively compared with Q. cerris.  相似文献

Tests in vitro and in situ showed that both mycelial and conidial suspensions of Acremonium mucronatum were antagonistic for Diplodia mutila, a common pathogen on the apical twigs of declining Quercus cerris, Quercus frainetto, Quercus pubescens and Quercus robur. Antagonism was greater with the mycelium than with the conidial suspensions, and was strongest at a test temperature of 20°C. The results suggest that pathogenicity of D. mutila on declining trees may be significantly reduced by the presence of A. mucronatum.  相似文献

Within a research project on quality of plants for forestation of agricultural lands, we studied the aetiology of a late seedling rot affecting holm oak (Quercus ilex ssp. ballota) in two forest nurseries in southern Spain. Major disease symptoms were foliage wilting and necrosis of feeder roots. Phytophthora cinnamomi, Phytophthora cryptogea and Phytophthora drechsleri were isolated from necrotic roots of holm oaks. Selected isolates of the three Phytophthora species were pathogenic to Quercus ilex ssp. ballota and Quercus suber seedlings in artificial inoculations. Soil flooding conditions were essential for infection and root rot development. There was no host specificity among the species, the isolates of P. cinnamomi being the most virulent in all inoculated plants. In these inoculations, Q. ilex ssp. ballota plants were more susceptible than those of Q. suber. This work is the first report of P. cinnamomi, P. drechsleri and P. cryptogea affecting Q. ilex ssp. ballota in forest nurseries.  相似文献

Chrysoporthe austroafricana is one of the most damaging pathogens of Eucalyptus trees in southern Africa. It also occurs on non‐native Tibouchina granulosa trees and native Syzygium species. Additional isolates of the pathogen from previously unstudied countries in the region have become available from survey studies. The aim of this study was to use VCGs to consider the diversity in populations of isolates collected in various countries in southern Africa (Malawi, Mozambique, Namibia, South Africa and Zambia) and from different hosts. We also wanted to determine whether there are shared VCGs among these countries and hosts in southern Africa and establish a VCG tester strain database. Results showed a high diversity amongst isolates from different countries and hosts, but suggested little movement of VCGs among countries or hosts based on the available isolates. A total of 108 VCG tester strains were identified for southern Africa.  相似文献

The composition of the endophytic fungal assemblages in Quercus cerris, Q. pubescens and Q. robur, three oak species most susceptible to decline, was investigated in Fagarè (Padova), Ulignano (Pisa), and Radda in Chianti (Firenze). The endophytic assemblages were studied as a function of health status of the whole tree and of twigs and leaves. The effect of station altitude was tested on Q. cerris. Samples were gathered from healthy and declining trees. In April (bud opening time) 20 twigs and 10 leaves were collected from each tree. Fungi were isolated from twig segments and leaf portions. A total of 23 fungal genera with 27 different species were isolated, in addition to a few sterile mycelia. Eleven species were found to be common to all three oak species, while other species were found only on one oak species, suggesting host specificity. Among the fungal species isolated, some were known to be pathogenic (Apiognomonia quercina, Colpoma quercinum, Diplodia mutila, Phomopsis quercina). The colonization frequency (CF) of pathogenic species varied between 0.9% for A. quercina in Q. cerris and 60.2% for P. quercina in Q. robur. Analysis of variance showed a statistically significant difference among the oak species tested. The CF was higher in declining trees in comparison with healthy trees, and also in twigs vs. leaves. The CF was found to be higher for Q. cerris trees growing at sea level as compared with those situated at an elevation of 350–400 m. In stands where, on account of particular ecological conditions, pathogenic behaviour is displayed simultaneously by more than one endophytic fungal species, trees accentuate their decline.  相似文献

We studied morphological, biochemical and physiological leaf acclimation to incident Photon-Photosynthetic-Flux-Density (PPFD) in Quercus ilex (holm oak) and Quercus suber (cork oak) at Mediterranean evergreen oak woodlands of southern Portugal. Specific leaf area (SLA) decreased exponentially with increasing PPFD in both species. Q. ilex had lower SLA values than Q. suber. Leaf nitrogen, cellulose and lignin concentration (leaf area-based) scaled positively with PPFD. Maximum rate of carboxylation (Vcmax), capacity for maximum photosynthetic electron transport (Jmax), rate of triose-P utilization (VTPU) and the rate of nonphotorespiratory light respiration (Rd) were also positively correlated with PPFD in both Quercus species, when expressed in leaf area but not on leaf mass basis. Q suber showed to have higher photosynthetic potential (Vcmax, Jmaxm and VTPUm) and a higher nitrogen efficient nitrogen use than Q.ilex. Leaf chlorophyll concentration increased with decreasing PPFD, improving apparent quantum use efficiency (Φ) in both Quercus species. We concluded that, in Q.ilex and Q.suber, leaf structural plasticity is a stronger determinant for leaf acclimation to PPFD than biochemical and physiological plasticity.  相似文献

To clarify the differences in susceptibility of six species in the Fagaceae (Quercus crispula, Quercus serrata, Quercus acutissima, Quercus phillyraeoides, Quercus glauca, and Castanopsis cuspidata var. sieboldii) to Raffaelea quercivora, we inoculated this fungus on seven potted 5-year-old seedlings of each species, observed symptom development, and measured xylem pressure potential (XPP) after inoculation. The first death was observed on the 11th day in Q. crispula and on the 56th day in Q. serrata. The number of dead seedlings of Q. crispula and Q. serrata were five and one, respectively, whereas no mortality was observed in the other four species. The XPP of inoculated seedlings in both Q. crispula and Q. serrata decreased after inoculation. In contrast, the XPP of inoculated seedlings of the other four species remained almost the same as in the control seedlings. These results indicate that R. quercivora is pathogenic to Q. crispula and Q. serrata and that the susceptibility of the six Fagaceae species in our study differed among species.  相似文献

In recent years damage to Austrian Castanea sativa populations caused by Cryphonectria parasitica has increased. A total of 34 isolates out of 13 observation plots of this phytopathogen in the south-east of Austria were examined for laccase activity, virulence, number of vegetative compatibility groups and for the existence of hypovirulence associated dsRNA. Furthermore, the applicability of the random amplified polymorphic DNA (RAPD) technique for investigating population structure was tested. Small dsRNA fragments were found in two Austrian strains. The same two strains exhibited reduced virulence. Seven vegetative compatibility groups were found in the areas examined. RAPD analysis proved to be an efficient method for distinguishing between different C. parasitica genotypes.  相似文献

Mass mortality of Fagacean tree species caused by Raffaelea quercivora has occurred widely in Japan. Because conidia or other propagules of the pathogen have not been found in infected trees, pathogen spread is assumed to occur primarily by hyphae. To clarify the relationship between hyphal growth of the pathogen within trees and their vessel arrangements, we examined two native Japanese oaks, Quercus crispula and Quercus glauca, and three exotic American oaks, Quercus coccinea, Quercus palustris and Quercus rubra. Quercus glauca is a radial‐porous species, whereas the other four species have a ring‐porous wood structure. Hyphal growth within inoculated potted living seedlings and in cut, sterilized stem segments of these species was examined microscopically after fungal inoculation. Water conductance in the seedlings was examined using transverse stem sections. The proportion of non‐conductive sapwood in Q. crispula, Q. coccinea and Q. palustris differed between inoculation and control treatment, being much higher in inoculated seedlings. The proportions were positively correlated with the extent of the hyphal growth. In sterilized stem segments, the extent of fungal colonization varied among the foreign ring‐porous species Q. coccinea, Q. palustris and Q. rubra. It is hypothesized that the extent of colonization by R. quercivora reflects the extent of non‐conductive sapwood irrespective of tree species, but is little affected by vessel arrangements.  相似文献

The association between water stress and susceptibility of Quercus cerris to the fungal parasite Hypoxylon mediterraneum was studied in field-grown trees and greenhouse-grown seedlings. Susceptibility of Q. cerris to the fungus, expressed as extension of wood discolorations, increased in field-grown trees during drought and increased in greenhouse-grown seedlings in response to limited water supply. In both seedlings and trees, spread of the fungus in the vascular system was higher when leaf water potentials were low than when leaf water potentials were high, and was significantly correlated to loss of hydraulic conductivity of xylem. The presence of the fungus in the xylem suggests that H. mediterraneum could use embolized vessels to spread in the host.  相似文献

This study was conducted to compare the virulences of various isolates of Raffaelea quercivora—a fungus that causes Japanese oak wilt disease—towards Japanese oak trees. Five isolates were collected from a wide range of Japan and inoculated into Quercus serrata logs and Q. crispula saplings. The tangential length of the discolored sapwood in the Q. serrata logs differed significantly among the isolates. The trend in isolate virulence was similar for the Q. serrata logs and the Q. crispula saplings. This is the first report suggesting that there is variability in virulence among isolates of R. quercivora.  相似文献

To compare the distributions of Raffaelea quercivora hyphae within seedlings of a susceptible species, Quercus crispula, and a resistant species, Q. glauca, we examined water conductance at transverse sections. Raffaelea quercivora was inoculated into the stems of seedlings in July (summer) and October (autumn) and observed by light and fluorescence microscopy at 1 and 2 weeks after inoculation. The hyphal distribution patterns expressed as the Iδ index differed between the species at each occasion. The hyphal growth was wider in Q. crispula than in Q. glauca in July inoculation, but did not differ in October inoculation. Non‐conductive sapwood in Q. crispula in transverse section was wider than that in Q. glauca at each occasion. The differences of hyphal growth and hyphal distribution patterns between species as little as 1 week after inoculation reflect differences in host susceptibility to the fungus.  相似文献

Within the scope of a research project on the condition of roots of declining oaks (Quercus robur, Quercus petraea), samples of fine roots and surrounding soil, specimens of stripe cankers on the stem base, and samples of stream water were examined for the presence of Phytophthora species using both baiting methods and selective agar media. At 27 sites in Germany (Bavaria, Rheinland-Pfalz, Schleswig-Holstein), Switzerland, Hungary, Italy and Slovenia the following species were isolated (mainly from soil): Phytophthora citricola, Phytophthora cactorum, Phytophthora cambivora, Phytophthora gon-apodyides, Phytophthora undulata, a species with affinity to Phytophthora drechsleri, and two additional species with close affinity to the Phytophthora cactorum group. Moreover, Pythium group P, Pythium anandrum, Pythium chamaehyphon, and many other Pythium species that have not yet been identified could be recovered. In a soil infestation test most isolates induced dieback of long root tips and necrotic lesions in the root cortex and at the root collar of Quercus robur seedlings. All Phytophthora species tested and Pythium group P caused cortical necrosis after stem inoculation of young Quercus robur trees. It could be shown in vitro that Phytophthora gonapodyides and Pythium group P were able to produce a wilting toxin. Nitrogen input and climatic changes are discussed as predisposing factors for root damage observed in the field.  相似文献

The survival and growth of Quercus germana Schltdl. & Cham., Q. xalapensis Humb. & Bonpl. and Magnolia dealbata Zucc. was evaluated in three treatments: (1) under the canopy of Pinus maximinoi H.E. Moore; (2) under the canopy of Liquidambar macrophylla Oersted, and (3) in open areas. The following hypothesis was presented: P. maximinoi and L. macrophylla canopy facilitate the establishment of Quercus germana, Q. xalapensis, and Magnolia dealbata. After one year, the survival of seedlings and the growth rate in height and basal diameter were significantly different between treatments and species (p < 0.05). The results suggest a facilitation establishment of Magnolia dealbata under the canopy of Pinus and Liquidambar, whereas the tolerance was observed for Quercus germana and Q. xalapensis. The results will allow to identify restoration strategies of the mountain cloud forest in Mexico.Resumen. Se evaluó la supervivencia y crecimiento de Quercus germana, Q. xalapensis y Magnolia dealbata, en tres tratamientos; (1) bajo el dosel de Pinus maximinoi, (2) bajo el dosel de Liquidambar macrophylla y (3) en áreas abiertas. La hipótesis planteada fue; la cobertura de P. maximinoi y L. macrophylla facilitan el establecimiento de Quercus germana, Q. xalapensis y Magnolia dealbata. Después de un año, la sobrevivencia de plántulas, y las tasas de crecimiento en altura y diámetro de la base, fueron significativamente diferentes entre tratamientos y especies (p < 0.05). Los resultados sugieren que el establecimiento de Magnolia dealbata es facilitado bajo el dosel de Pinus y Liquidambar, mientras que para Quercus germana y Q. xalapensis se observó tolerancia. Los resultados permitirán identificar estrategias de rehabilitación del bosque mesófilo de montaña en México.  相似文献

In 2000, one-year-old seedlings of pyrenean oak (Quercus pyrenaica Willd.) and sessile oak (Quercus petraea [Matt.] Liebl) were planted in a thinned and an unthinned plot in a pinewood (Pinus sylvestris), and in a nearby clearing. In summer 2002 and 2003, water relations and gas exchange parameters were measured to address the impact of drought on the seedlings. Chlorophyll a fluorescence was also measured to explore leaf photochemistry and a possible non-stomatal limitation to photosynthesis (A). Reduction in stomatal conductance (g) in response to the decrease of predawn water potential (Ψpd) resulted the main cause affecting net carbon uptake. Water potential at midday (Ψmd) was similar in both species but Quercus petraea was more sensitive to soil water deployment occurred along summer, showing slightly lower Ψpd because worse recover of water potential during night. Rate of photosynthesis was higher in Q.␣pyrenaica probably in relation to its greater leaf mass per area (LMA) and nitrogen content per leaf area (Na). Mortality was highest in the clearing and lowest in the thinned pinewood. Throughout the summer, soil moisture was higher in the thinned area, possibly because of the reduction in tree transpiring surface and interception of rainfall. Accordingly, Ψpd of both species was higher in the thinned site.  相似文献

Cryphonectria parasitica is not common in the forests of the Emilia Romagna region of Italy on hosts other than chestnut. Three isolates with morphological cultural characteristics of C. parasitica were collected: two from infected bark of Quercits pubescens (EQ1 and EQ2) and one from Carpinus betulus (EC1). Pathogenicity tests on chestnut and crosses in vitro with strains of C. parasitica of different mating types confirmed that EQ1 and EQ2 are C. parasitica isolates, whereas EC1 seems to belong to a different species. Progeny analysis of crosses with the isolate EQ1 and a strain from chestnut showed the formation of new vegetative compatibility groups. Therefore hosts other than chestnut can contribute to increasing pathogen variability through recombination of the v-c genes.  相似文献

The occurrence of chestnut blight (Cryphonectria parasitica) on oaks in mixed chestnut‐oak forests was studied in 2003–2008 in Slovakia. Infections on living Quercus trees were found at four of seven localities. The disease incidence on oaks ranged from 1.3% to 15.8%. The symptoms on infected oaks were similar to those on chestnut, but less conspicuous. Cankers of C. parasitica were found only on Quercus robur and Q. petraea. A total of 22 isolates of C. parasitica, all virulent, were isolated. Each site yielded only a single vc type (EU12 or EU13). Field inoculation experiments on chestnuts with seven strains of C. parasitica from oaks and an isolate from Castanea sativa showed no differences in virulence. On Quercus robur stems, the cankered area was significantly smaller than on C. sativa and the cankers developed very slowly.  相似文献

During spring and summer of 2016, dieback symptoms including blights of leaves, twigs, and acorns were observed on current year shoots of Quercus infectoria in the Zagros oak forests of Iran. The fungus isolated from damaged tissues was identified as Discula quercina based on both morphological characteristics and ITS sequencing. To fulfil the Koch's postulates, a representative isolate was inoculated onto shoots of Q. infectoria and Quercus libani in both laboratory and forest conditions. Responses of the two oak species to inoculation with D. quercina were examined under laboratory and forest conditions in a completely randomized experiment. Discula quercina was clearly involved in oak dieback, and Q. infectoria was more susceptible than Q. libani to damage by the pathogen. This is the first record of the occurrence and pathogenicity of the fungus Discula quercina on Quercus infectoria. The fungus is considered as an emerging pathogen on oak trees in Zagros forests in Iran. Furthermore, the pathogenicity of the Discula quercina on Q. libani under laboratory and forest conditions increases the potential importance of this pathogen in Zagros forests.  相似文献

在北京周边县区、湖北大老岭山区以及陕西安康地区分别调查栗疫病发生情况,采集病原栗疫菌(Cryphonectria parasitica)菌株并进行营养体亲和性试验.结果表明:北京板栗栽培区和陕西安康地区野生栗栗疫病发病率较高,局部地区危害严重;湖北大老岭山区栗疫病发病率低,危害轻微.北京地区菌株群体营养体亲和型多样性指数(Shannon-Wiener's diversity index)极显著低于湖北菌株群体和陕西菌株群体,而陕西与湖北菌株群体营养体亲和型多样性指数差异不显著.随机选取湖北、陕西野生栗栗疫菌部分菌株,利用特异性引物,通过普通PCR和巢氏PCR,对其交配型进行测定,发现陕西与湖北野生栗菌株群体中均存在MAT-1与MAT-2两种交配型的菌株.通过PCR扩增,在两地的野生菌株群体里均发现同时具有两种交配型基因的菌株,其中,陕西群体此类菌株比例较大,湖北菌株比例较小  相似文献
