The impact of pH changes on microbial biomass carbon (Cmic) and microbial biomass phosphorus (Pmic) were examined for 3 red soils under citrus production with different lengths of cultivation. Soil pH significantly affected Cmic and Pmic. The Cmie and Pmic changes, as a function of soil pH, appeared to follow a normal distribution with the original soil pH value at the apex and as pH increased or decreased compared to the original soil pH, Cmic and Pmic declined. Moreover, there were critical pH values at both extremes (3.0 on the acidic side and 8.0 to 8.5 on the alkaline side), beyond which most of microorganisms could never survive. The effect of pH on Cmic and Pmic was also related to the original soil pH. The higher the original soil pH was, the less Cmic or Pmic were affected by pH change. It is suggested that soil microorganisms that grow in a soil environment with a more neutral soil pH range (i.e. pH 5.5-7.5) may have a greater tolerance to pH changes than those growing in more acidic or more alkaline soil pH conditions.  相似文献

 Microcosms were used to determine the influence of N additions on active bacterial and active fungal biomass, cellulose degradation and lignin degradation at 5, 10 and 15 weeks in soils from blackwater and redwater wetlands in the northern Florida panhandle. Blackwater streams contain a high dissolved organic C concentration which imparts a dark color to the water and contain low concentrations of nutrients. Redwater streams contain high concentrations of suspended clays and inorganic nutrients, such as N and P, compared to blackwater streams. Active bacterial and fungal biomass was determined by direct microscopy; cellulose and lignin degradation were measured radiometrically. The experimental design was a randomized block. Treatments were: soil type (blackwater or redwater forested wetlands) and N additions (soils amended with the equivalent of 0, 200 or 400 kg N ha–1 as NH4NO3). Redwater soils contained higher concentrations of C, total N, P, K, Ca, Mn, Fe, B and Zn than blackwater soils. After N addition and 15 weeks of incubation, the active bacterial biomass in redwater soils was lower than in blackwater soils; the active bacterial biomass in blackwater soils was lower when 400 kg N ha–1, but not when 200 kg N ha–1, was added. The active fungal biomass in blackwater soils was higher when 400 kg N ha–1, but not when 200 kg N ha–1, was added. The active fungal biomass in redwater wetland soils was lower when 200 kg N ha–1, but not when 400 kg N ha–1, was added. Cellulose and lignin degradation was higher in redwater than in blackwater soils. After 10 and 15 weeks of incubation, the addition of 200 or 400 kg N as NH4NO3 ha–1 decreased cellulose and lignin degradation in both wetland soils to similar levels. This study indicated that the addition of N may slow organic matter degradation and nutrient mineralization, thereby creating deficiencies of other plant-essential nutrients in wetland forest soils. Phosphate rock (PR), limestone, coal combustion by-product (CCBP) high in Ca and high organic manures are potential amendments for increasing agricultural production in the acidic soils of the Appalachian region. The objective of this study was to examine effects of PR, CCBP and cellulose addition on soil microbial biomass in an acidic soil based on the measurement of soil microbial biomass P (P mic) and on the mineralization of organic matter. Application of PR alone or in combination with CCBP increased P mic. The P mic was far less when the soil received PR in combination with limestone than with PR application alone or PR in combination with CCBP. Either CCBP or limestone application alone considerably decreased P mic in the soil due to reduced P solubility. Cellulose addition alone did not increase P mic, but P mic was significantly increased when the soil was amended with cellulose in combination with PR. The decomposition of added cellulose was very slow in the soil without PR amendment. However, mineralization of both native organic matter and added cellulose was enhanced by PR application. Mineralization of organic matter was less when the soil was amended with PR in combination with high rates of CCBP (> 2.5%) because PR dissolution varied inversely with amount of CCBP addition. Overall, CCBP had no detrimental effect on soil microbial biomass at low application rates, although, like limestone, CCBP at a high rate may decrease P mic in P-deficient soils through its influence on increased soil pH and decreased P bioavailability in the soil. The microbial biomass C, N and P of soils all over China were determined in this study to study their affecting factors. The results, about 100-417 mg C kg-1 soil, 18-51 mg N kg-1 soil and 4.4-27.3 mg P kg-1 soil, showed the biomass C, N and P in linear relationship with the soil total organic C, toal N and soil organic P. The ratios of C: N and C:P, ranging from 5.6 to 9.6 and from 11.2 to 48.4 respectively, were affected by soil pH, texture, crop rotation, macroclimate, etc. The ratio of C:N in soil biomass increases gradually from the north to the south in China.  相似文献

Influence of ectomycorrhizal mat soils on lignin and cellulose degradation   总被引:3,自引:0,他引:3  
Summary The ectomycorrhizal fungus Hysterangium setchellii (Fisher) forms extensive hyphal mats at the soillitter interface with the roots of the host tree Douglas fir Pseudotsuga menziesii [(Mirb.) Franco]. Microbial biomass, and lignin and cellulose decomposition rates were measured seasonally for 1 year, using 14C techniques in ectomycorrhizal mat soils and adjacent non-mat soils in a second-growth Douglas fir forest. The microbial biomass and cellulose degradation rates were 3–6 times higher in ectomycorrhizal mat soils than in adjacent nonmat soils. Lignin degradation rates were higher in ectomycorrhizal mat soils than adjacent non-mat soils. Our data suggest that the ectomycorrhizal fungus H. setchellii provides a microenvironment with increased microbiological activity which results in faster lignin and cellulose decomposition.  相似文献

The response of the soil microbial biomass to seasonal changes was investigated in the field under pastures. These studies showed that over a 9-month period, microbial biomass carbon, phosphorus and sulphur (biomass C, P, S), and their ratios (C:P, C:S, and P:S) responded differently to changes in soil moisture and to the input of fresh organic materials. From October to December (1993), when plant residues were largely incorporated into the soils, biomass C and S increased by 150–210%. Biomass P did not increase over this time, having decreased by 22–64% over the dry summer (July to September). There was no obvious correlation between biomass C, P, and S and air temperature. The largest amounts of biomass C and P (2100–2300μg and 150–190μgg–1 soil, respectively) were found in those soils receiving farmyard manure (FYM or FYM+NPK) and P fertilizer, whereas the use of ammonium sulphate decreased biomass C and P. The C:P, C:S, and P:S ratios of the biomass varied considerably (9–276:1; 50–149:1; and 0.3–14:1, respectively) with season and fertilizer regime. This reflected the potential for the biomass to release (when ratios were narrow) or to immobilize (wide ratios) P and S at different times of the year. Thus, seasonal responses in biomass C, P, and S are important in controlling the cycling of C, P, and S in pasture and ultimately in regulating plant availability of P and S. The uptake of P in the pasture was well correlated with the sum of P in the biomass and soil available pools. This study was undertaken to investigate the long-term influence of lime application and tillage systems (no-till, ridge-till and chisel plow) on soil microbial biomass C (Cmic) and N (Nmic) and the activities of glycosidases (- and -glucosidases, - and -galactosidases and -glucosaminidase) at their optimal pH values in soils at four agroecosystem sites [Southeast Research Center (SERC), Southwest Research Center (SWRC), Northwest Research Center (NWRC), and Northeast Research Center (NERC)] in Iowa, USA. Results showed that, in general, the Cmic and Nmic values were significantly (P <0.001) and positively correlated with soil pH. Each lime application and tillage system significantly (P <0.001) affected activities of the glycosidases. With the exception of -glucosidase activity, there was no lime×tillage interaction effect. Simple correlation coefficients between the enzyme activities and soil pH values ranged from 0.51 (P <0.05) for the activity of -glucosidase at the NWRC site (surface of the no-till) to 0.98 (P <0.001) at the SWRC site. To assess the sensitivity of the enzymes to changes in soil pH, the linear regression lines were expressed in activity/pH values. In general, their order of sensitivity to changes in soil pH was consistent across the study sites as follow: -glucosidase>-glucosaminidase>-galactosidase>-galactosidase>-glucosidase. Lime application did not significantly affect the specific activities (g p -nitrophenol released kg–1 soil organic C h–1) of the enzymes. Among the glycosidases studied, -glucosidase and -glucosaminidase were the most sensitive to soil management practices. Therefore, the activities of these enzymes may provide reliable long-term monitoring tools as early indicators of changes in soil properties induced by liming and tillage systems.  相似文献

A linear relationship was observed between the amount of total respiratory quinones and the microbial biomass measured by a fumigation-extraction method in 15 soil samples regardless of the significant differences in the composition of the quinone profiles, with one exception in a soil amended with a very high application rate of farmyard manure. It is suggested that the amount of total respiratory quinones can be used as an indicator of the microbial biomass in soil.  相似文献

In order to analyze the N mineralization process under shifting cultivation in northern Thailand, labile pools of soil organic matter were studied, which were considered to be the factors contributing to the N mineralization process. Organic C, (organic + NH4 +)-N, and hexose-C were extracted from fresh soils in the surface 0–5 cm layers with a 0.5 M K2S0. solution at 110°C in an autoclave (fraction A) or at room temperature with a reciprocal shaker (fraction B), and analyzed as labile pools of organic matter. In the traditional shifting cultivation system, the content of organic C in fraction A in the fallow fields for 8 to 15 y was 3,710 mg kg-1 while that in the fallow fields for 1 y and 3 to 5 y was 2,640 and 2,600 mg kg-1, respectively. A high correlation was observed between the contents of the labile pool in fraction A and total soil organic matter. The ratio of the pool in fraction A to total soil organic matter apparently remained constant through the input-output balance in the pool. The content of the labile pool in fraction B was the highest among the fields cultivated for 1 y after the slash and burn practice and it decreased in the course of the fallow period. The content of organic C was 548 mg kg-1 in the fields cultivated for 1 y and 235 mg kg-1 in the fallow fields for 8-15 y, respectively. There was a reverse relation between the contents of the pool in fraction B and microbial biomass. Therefore, the origin of the pool in fraction B was attributed to the microbial debris associated mainly with a decrease in the soil moisture content in the dry season. On the other hand, in the relatively intensive cultivation system, there was no significant difference in the contents of the labile pools both in fractions A and B among the land use stages, suggesting that the preservation mechanism of these pools, which was observed in the traditional cultivation system, did not operate well in the intensive system. In alternative farming systems in future, it will be essential to apply organic materials to soils to supply organic matter and to maintain the microbial biomass.  相似文献

Microbial biomass C (MBC) is one of the soil properties used as an indicator for the fertility status of a soil. A study was conducted on a semi-arid Loess Plateau in China. The field was planted with spring wheat and mulched with plastic film for various lengths of time. Our primary objectives were to (i) explore the influence of film mulching on soil MBC and soil fertility, and (ii) seek an effective approach of maintaining and improving sustainability of cropland mulched with plastic film in two growing seasons. Four treatments were tested, non-mulching (M0), mulching for 30 days after sowing (M30), mulching for 60 DAS (M60) and mulching for the whole growing period (Mw). An increasing air temperature with time within the growing season promoted soil MBC in the two growing seasons, but a severe drought led to a lower MBC in 2000 compared with the wet year of 1999. Film mulching promoted MBC significantly in the 2 years, but decreased soil organic carbon (SOC). SOC is very low in the experimental soil, accounting for the higher MBC/SOC ratio compared with ratios reported by others. The SOC is greatly reduced in the non-mulched and the Mw treatments compared to the M30 and M60 treatments. In conclusion, the benefits of film mulching in semi-arid agricultural systems are enormous but realizing their full potential depends on how long the mulching material is maintained during the growing season. In the system tested, it is desirable to mulch the plots for 30-60 DAS in order to enhance microbial biomass and cycling of nutrients and also to provide a more stable soil micro-environment that generates more residues in the rhizosphere.  相似文献

The incorporation of organic amendments from pruning waste into soil may help to mitigate soil degradation and to improve soil fertility in semiarid ecosystems. However, the effects of pruning wastes on the biomass, structure and activity of the soil microbial community are not fully known. In this study, we evaluate the response of the microbial community of a semiarid soil to fresh and composted vegetal wastes that were added as organic amendments at different doses (150 and 300 t ha−1) five years ago. The effects on the soil microbial community were evaluated through a suite of different chemical, microbiological and biochemical indicators, including enzyme activities, community-level physiological profiles (CLPPs) and phospholipid fatty acid analysis (PLFA). Our results evidenced a long-term legacy of the added materials in terms of soil microbial biomass and enzyme activity. For instance, cellulase activity reached 633 μg and 283 μg glucose g−1 h−1 in the soils amended with fresh and composted waste, respectively. Similarly, bacterial biomass reached 116 nmol g−1 in the soil treated with a high dose of fresh waste, while it reached just 66 nmol g−1 in the soil amended with a high dose of composted waste. Organic amendments produced a long-term increase in microbiological activity and a change in the structure of the microbial community, which was largely dependent on the stabilization level of the pruning waste but not on the applied dose. Ultimately, the addition of fresh pruning waste was more effective than the application of composted waste for improving the microbiological soil quality in semiarid soils.  相似文献

The relationships of soil microbial biomass C (Cmic) or N (Nmic) with mean annual precipitation and temperature were studied along a climatic transect in the Mongolian steppe. Soil organic C (Corg) and total N (Nt), respiration rate, Cmic and Nmic at depths of 0–5 and 5–10 cm decreased with increasing aridity. The contents of Corg and Nt in the 0- to 5-cm soil layers decreased linearly with precipitation reduction along the transect. Cmic and Nmic changes with precipitation were not linear, with higher changes between 330 and 128 mm mean annual precipitation. Cmic/Corg and Nmic/Nt increased with increasing aridity. The metabolic quotient qCO2 of 0- to 5-cm soil layers was low between 330 and 273 mm precipitation. The relationship between the qCO2 of the 0- to 5-cm soil layers and the mean annual precipitation was well fitted with a quadratic function y =0.0006x2 –0.40x +86.0, where y is the qCO2 (µmol CO2-C mmol–1 Cmic) and x is the mean annual precipitation (mm). Corg, Nt, Cmic, Nmic and respiration rate decreased exponentially with increasing mean annual temperature in both the 0- to 5- and 5- to 10-cm soil layers, and change rate was lower when the mean annual temperature was higher than 2.6°C. The close relationships of the mean annual precipitation or temperature with soil Corg, Nt, Cmic, Nmic, Cmic/Corg and qCO2 indicate that each parameter can be calculated by determining the other parameters in this specific climatic range.  相似文献

Soil organic matter level, soil microbial biomass C, ninhydrin-N, C mineralization, and dehydrogenase and alkaline phosphatase activity were studied in soils under different crop rotations for 6 years. Inclusion of a green manure crop of Sesbania aculeata in the rotation improved soil organic matter status and led to an increase in soil microbial biomass, soil enzyme activity and soil respiratory activity. Microbial biomass C increased from 192 mg kg–1 soil in a pearl millet-wheat-fallow rotation to 256 mg kg–1 soil in a pearl millet-wheat-green manure rotation. Inclusion of an oilseed crop such as sunflower or mustard led to a decrease in soil microbial biomass, C mineralization and soil enzyme activity. There was a good correlation between microbial biomass C, ninhydrin-N and dehydrogenase activity. The alkaline phosphatase activity of the soil under different crop rotations was little affected. The results indicate the green manuring improved the organic matter status of the soil and soil microbial activity vital for the nutrient turnover and long-term productivity of the soil. Decomposing needles from a Norway spruce forest in southern Sweden were studied for 559 days under laboratory conditions. Falling needles were collected in control (Co) plots and plots that had received 100 kg N ha−1 yr−1 as (NH4)2SO4 for 9 years under field conditions. One of the aims was to determine whether the previously documented low decomposition rate of the N fertilized (NS) needles could be explained by a lower degradation degree of lignin. The lignin content was studied using the alkaline CuO oxidation method, the Klason lignin method and CPMAS 13C NMR spectroscopy. The amounts of cellulose and hemicellulose were also determined.The fertilized needle litters initially decomposed faster than the unfertilized, but later this reaction reversed, so that at the end the mass loss was 45% initial C in the control and 35% initial C in NS. Klason lignin decreased with time in both treatments and overall, the change of Klason lignin mirrored the litter mass loss. No major difference as regards the decomposition of hemicellulose occurred between the treatments, whereas significantly lower concentrations of cellulose were found in NS needles throughout the incubation. The CuO derived compounds (VSC) were somewhat lower in NS needles throughout the decomposition time. Initially, VSC increased slightly in both treatments, which contradicts the Klason lignin data. There was a weak positive relationship (p>0.05) between VSC and Klason lignin. Both vanillyls compounds (V) and cinnamyl compounds (Ci) increased slightly during decomposition, whereas syringyl compounds (S) vanished entirely. The lignin degradation degree, i.e. the acid-to-aldehyde ratio of the vanillyl compounds expressed as (Ac/Al)v, showed no significant effect of treatment. The 13C NMR analyses of the combined samples showed increased content of aromatic C with increasing decomposition time. The carbohydrate content (O-alkyl C) was lower in the fertilized needle litter throughout the incubation time. The alkyl C content tended to increase with decomposition time and N fertilization. The alkyl C/O-alkyl C ratios increased in both treatments during the incubation. The NMR results were not tested statistically.In conclusion, no major difference concerning lignin degradation could be found between the unfertilized and N fertilized needle litter. Thus, the study contradicts the hypothesis that higher amounts of N reduce lignin degradation. The reduced biological activity is probably due to direct N effects on the microorganisms and their decomposing ability.  相似文献

Abstract

The effects of heavy metals (Cu, Pb, and As) accumulated in apple orchard surface soils on the microbial biomass, dehydrogenase activity, and soil respiration were investigated. The largest concentrations of total Cu, Pb, and As found in the soils used were 1,010, 926, and 166 mg kg?1 soil, respectively. The amounts of microbial biomass C and N, expressed on a soil organic C and soil total N basis, respectively, were each negatively correlated with the amounts of total, 0.1 M HCI-extractable, and 0.1 M CaCl2-extractable Cu as logarithmic functions, the correlation coefficient being lowest for the 0.1 M CaCl2extractable Cu. Nevertheless, they were not correlated with the soil pH which was controlling the solubility of Cu in 0.1 M CaCl2. The dehydrogenase activity expressed per unit of soil organic C was also negatively correlated with the amounts of total, 0.1 M HCI-extractable Cu, and 0.1 M CaCl2-extractable Cu as logarithmic functions. However, the correlation coefficient was highest for the 0.1 M CaCl2-extractable Cu. Although the soil respiration per unit of soil total organic C did not show any significant correlations with the total concentrations of heavy metals, it showed negative significant correlations with the amount of 0.1 M HCI-extractable Cu, and to a greater extent, with the amount of 0.1 M CaCl2-extractable Cu. Both the dehydrogenase activity and respiration per unit of soil total organic C increased significantly with increasing soil pH. These results suggested that in apple orchard soils with heavy metal accumulation the microbial biomass was adversely affected by the slightly soluble Cu, whereas the microbial activities by the readily soluble Cu whose amount depended on the soil pH. The respiration per unit of microbial biomass C showed a positive significant correlation with the logarithmic concentration of total Cu. Furthermore, the contribution of fungi to substrate-induced respiration increased with increasing total Cu content in the soils.  相似文献

盆栽和大田试验表明,作物根系显著影响土壤微生物体氮的含量。在田间试验条件下,根际土壤微生物体氮比非根际土壤平均高出N54.7μg/g;盆栽试验中,根际土壤微生物体氮平均含量为N77.1±13.6μg/g,而非根际土壤为N65.2±17.0μg/g,差异达显著水平,根际微生物体氮含量为非际根际土壤的1.10~2.04倍。施肥能明显增加土壤微生物体氮含量,但影响程度因肥料种类而不同。秸秆和富含有机物质的厩肥对土壤微生物体氮的影响远大于化学肥料,而且土壤微生物体氮含量随秸秆施用量增加而增加。在红油土上进行的20年长期田间定位试验结果表明,对不施肥和施氮磷处理,0—20cm土层的微生物体氮分别是N102.2和110.4μg/g;在施氮磷的基础上,每公顷配施新鲜玉米秸秆9375kg、18750kg、37500kg和厩肥37500kg时,相应土层微生物体氮分别是N147.5、163.2、286.4和265.3μg/g。培养条件下,当有效能源物质缺乏时,微生物对NH4+-N的同化固定能力远大于NO3--N,但在加入有效能源物质葡萄糖后,微生物对2种形态氮的固定量大幅度增加,且对2种形态氮的固定量趋于一致。  相似文献

Although elevation of CO2 has been reported to impact soil microbial functions, little information is available on the spatial and temporal variation of this effect. The objective of this study was to determine the microbial response in a northern Colorado shortgrass steppe to a 5-year elevation of atmospheric CO2 as well as the reversibility of the microbial response during a period of several months after shutting off the CO2 amendment. The experiment was comprised of nine experimental plots: three chambered plots maintained at ambient CO2 levels of 360 μmol mol−1 (ambient treatment), three chambered plots maintained at 720 μmol mol−1 CO2 (elevated treatment) and three unchambered plots of equal ground area used as controls to monitor the chamber effect.Elevated CO2 induced mainly an increase of enzyme activities (protease, xylanase, invertase, alkaline phosphatase, arylsulfatase) in the upper 5 cm of the soil and did not change microbial biomass in the soil profile. Since rhizodeposition and newly formed roots enlarged the pool of easily available substrates mainly in the upper soil layers, enzyme regulation (production and activity) rather than shifts in microbial abundance was the driving factor for higher enzyme activities in the upper soil. Repeated soil sampling during the third to fifth year of the experiment revealed an enhancement of enzyme activities which varied in the range of 20-80%. Discriminant analysis including all microbiological properties revealed that the enzyme pattern in 1999 and 2000 was dominated by the CO2 and chamber effect, while in 2001 the influence of elevated CO2 increased and the chamber effect decreased.Although microbial biomass did not show any response to elevated CO2 during the main experiment, a significant increase of soil microbial N was detected as a post-treatment effect probably due to lower nutrient (nitrogen) competition between microorganisms and plants in this N-limited ecosystem. Whereas most enzyme activities showed a significant post-CO2 effect in spring 2002 (following the conclusion of CO2 enrichment the previous autumn, 2001), selective depletion of substrates is speculated to be the cause for non-significant treatment effects of most enzyme activities later in summer and autumn, 2002. Therefore, additional belowground carbon input mainly entered the fast cycling carbon pool and contributed little to long-term carbon storage in the semi-arid grassland.  相似文献

Tree species differ in their effect on soil development and nutrient cycling. Conversion of beech coppice to pine plantations can alter soil physical and chemical properties, which in turn may have significant impacts on soil microbial biomass C and N (Cmic, Nmic). The major objective of this study was to evaluate soil quality changes associated with the forest conversion in humid NW Turkey. Results from this study showed that levels of soil organic carbon (Corg), total nitrogen (Nt), moisture, Cmic and Nmic under beech coppice were consistently higher but levels of pH, CaCO3 and EC were lower compared to pine plantation. Differences between the forest stands in Cmic and Nmic were mainly related to the size of the Corg stores in soil and to tree species. In addition, high level of CaCO3 is likely to reduce pools of soil organic C and possibly even microbial biomass C and N in pine forests. The average Cmic:Nmic ratios were higher in soils under beech coppice than pine plantation, while Cmic:Corg and Nmic:Nt percentages were similar in both forest types. These results revealed the differences in microbial community structure associated with different tree species and the complex interrelationships between microbial biomass, soil characteristics, litter quantity and quality. Soil microbial organisms are central to carbon (C) and nitrogen (N) transformations in soils, yet not much is known about the stable isotope composition of these essential regulators of element cycles. We investigated the relationship between C and N availability and stable C and N isotope composition of soil microbial biomass across a three million year old semiarid substrate age gradient in northern Arizona. The δ15N of soil microbial biomass was on average 7.2‰ higher than that of soil total N for all substrate ages and 1.6‰ higher than that of extractable N, but not significantly different for the youngest and oldest sites. Microbial 15N enrichment relative to soil extractable and total N was low at the youngest site, increased to a maximum after 55,000 years, and then decreased slightly with age. The degree of 15N enrichment of microbial biomass correlated negatively with the C:N mass ratio of the soil extractable pool. The δ13C signature of soil microbial biomass was 1.4‰ and 4.6‰ enriched relative to that of soil total and extractable pools respectively and showed significant differences between sites. However, microbial 13C enrichment was unrelated to measures of C and N availability. Our results confirm that 15N, but not 13C enrichment of soil microbial biomass reflects changes in C and N availability and N processing during long-term ecosystem development.  相似文献
