The studies were carried out on Balb/c mice (5-6 weeks of age) exposed to immunosuppression by a single intraperitoneal dose (125 mg/kg) of hydrocortisone. Prior to hydrocortisone injection the mice were treated with diethyldithiocarbamate (DTC) intra-peritoneally at a dose of 20 mg/kg, five times at 48 h intervals or calf thymus extract (TFX) at a dose of 10 mg/kg, 10 times at 24 h intervals. The two drugs were used per se or in zinc ions interactions, by adding zinc ions (as sulphate salt) to drinking water at a dose of 72 microg/mouse per day. The results obtained in the study show that hydrocortisone injection drastically decreases the number of thymocytes and splenocytes, which is also accompanied by a decreasing weight ratio of the thymus and spleen. The decreasing number of thymic and spleen cells corresponds to a decreasing percentage of CD4+, CD8+ and CD19+ splenocytes and double positive CD4+CD8+ thymocytes. Changes in the number of thymic cells affect their activity, which is expressed in a decreased proliferative response of thymocytes stimulated in vitro with concanavalin A (Con A) and phytohaemagglutinin (PHA). It has also been found that a single hydrocortisone dose decreases interleukin (IL)-1 production by murine intraperitoneal macrophages stimulated in vitro with lipopolysaccharide (LPS) from Escherichia coli. TFX or DTC counteract hydrocortisone-induced immunosuppression, which is expressed in partial normalization of the total number of thymic and spleen cells, accelerated regeneration of the two lymphatic organs, shorter suppressive action of hydrocortisone on the percentage of CD4+, CD8+ splenocytes and double positive (CD4+CD8+) and CD4+ thymocytes. Furthermore, total counteraction against the suppressive action of hydrocortisone to proliferative activity of thymocytes stimulated in vitro with Con A and PHA was observed. TFX administered prior to hydrocortisone injection partially prevented the suppressive action of the drug on IL-1 production by intraperitoneal macrophages, but such an effect was not observed with DTC. The immunorestorative effect of TFX and DTC was augmented by zinc supplementation. The results obtained in the study show that neither TFX nor DTC administration per se and in interaction with zinc supplementation were able to change the suppressive effect of hydrocortisone on the percentage of B splenocytes (CD19+ cells).  相似文献

The immunotropic properties of calf thymus extract (TFX-Jelfa) is connected with the mimic action of the thymus to modulate the differentiation, maturation and function of prothymocytes and mature thymus dependent (T) cells. The studies were carried out on CFW male mice aged 3 months. The animals were infected per os with 200 larvae of Trichinella spiralis. TFX-Jelfa was administered i.p. at a dose of 10 mg/kg seven times at 24 hour intervals prior to infection. The percentage of CD4+ and CD8+ in suspension of splenocytes and mesenteric lymphonode cells by flow cytometry using monoclonal antibodies coupled with fluorescein isothiocyanate (FITC) or phycoerythrin (PE) were determined. At the same time, cryostat preparations, made from jejunum and muscle samples, were examined by the direct immunofluorescence method using FITC-labeled antibody to mouse CD4+ and CD8+. It has been found that infection with T. spiralis in mice decreases the percentage of CD8+ splenocytes, while the percentage of CD8+ mesenteric lymphonode cells does not change. However, in infected mice the percentage of CD4+ spleen cells and mesenteric lymphonode cells is increased. It has been also found that during the course of infection an increase in the number of CD8+ and CD4+ cells in the basal lamina propria of the intestines was observed. In infected mice, CD4+ lymphocytes were visible in the inflammatory infiltrates of the muscle tissue on the 14th day, whereas CD8+ lymphocytes were first observed a week later. Pretreatment with TFX does not change the inhibitory effect of infection on the percentage of CD8+ splenocytes, but potentiates the percentage of CD4+ spleen cells and mesenteric lymphonode cells increased by infection. Furthermore, administration of TFX prior to infection also potentiates the stimulatory effect of T. spiralis on the number of CD8+ and CD4+ in the basal lamina propria of the jejunum, and on the number of CD8+ cells in the inflammatory infiltrates of the muscle tissue.  相似文献

Five years of data were collected on 124 Brangus-type cows to evaluate weaning calves at 8.5 or 10.5 mo of age and winter supplementation of the brood cow with 2.25 kg of molasses daily. Cows with calves weaned at 10.5 mg of age gained 14 kg less weight (P less than .01) during the late nursing period than did cows from which calves were earlier weaned. Age of calf at weaning had no effect on cow reproduction, but calves weaned at 10.5 mo were 2 to 3 d younger (P less than .06) and 5.3 kg lighter (P less than .05) at 8.5 mo of age. During the last 2 mo of nursing, calves weaned at 10.5 mo of age gained 37.2 kg and had a 31.9-kg heavier weaning weight than calves weaned at 8.5 mo of age. Molasses supplementation resulted in differences (P less than .01) in cow weight changes. Cows fed molasses had calving percentages 5 to 7 units higher (P less than .30) than cows not fed molasses. Feeding cows molasses for 145 d throughout the calving and breeding season increased calf weaning weight 7.7 (P less than .02) and 11.2 (P less than .03) kg, respectively, at 8.5 and 10.5 mo of age over the nonsupplemented controls.  相似文献

The exposure of pigs to heat stress (HS) appears to damage their intestinal epithelia, affecting the absorption of amino acids (AAs). Arg is involved in the restoration of intestinal epithelial cells but HS reduces Arg intake. The effect of dietary supplementation with Arg on the morphology of intestinal epithelia, AA transporter gene expression, and serum concentration (SC) of free AAs in HS pigs was analyzed. Twenty pigs (25.3 ± 2.4 kg body weight) were randomly assigned to two dietary treatments: Control (0.81% Arg), wheat–soybean meal diet supplemented with l-Lys, l-Thr, dl-Met, and l-Trp, and the experimental diet where 0.16% free l-Arg was supplemented to a similar Control diet (+Arg). All pigs were individually housed and exposed to HS, fed ad libitum with full access to water. The ambient temperature (AT), recorded at 15-min intervals during the 21-d trial, ranged on average from 29.6 to 39.4 °C within the same day. Blood samples were collected on day 18 at 1600 hours (AT peak); serum was separated by centrifugation. At the end of the trial, five pigs per treatment were sacrificed to collect samples of mucosa scratched from each small intestine segment. The expression of AA transporters in intestinal mucosa and the SC of AAs were analyzed. Villi height (VH) was higher (P < 0.01) in the duodenum, jejunum, and ileum but the crypt depth did not differ between the Control and the +Arg pigs. Supplementation of l-Arg increased the mRNA coding for the synthesis of the cationic AA transporter b0,+ (P < 0.01) and the neutral AA transporter B0 (P < 0.05) in the duodenum by approximately 5-fold and 3-fold, respectively, but no effect on mRNA abundance was observed in the jejunum and ileum. The supplementation of l-Arg increased serum Arg, His, Met, Thr, Trp, and urea (P < 0.05) and also tended to increase Val (P < 0.10) but did not affect Ile, Lys, Leu, and Phe. These results indicate that supplementing 0.16% l-Arg to the Control diet may help to improve the function of the small intestine epithelium, by increasing the VH, the abundance of AA transporters, and the SC of most indispensable AAs in pigs exposed to HS conditions. However, the lack of effect of supplemental Arg on both Lys SC and weight gain of pigs suggests that increasing the Lys content in the +Arg diet might be needed to improve the performance of HS pigs.  相似文献

Zinc is an essential micronutrient and has significant effects on human growth, development, and immune function. Zinc supplementation or deficiency may affect the course of infection. Zinc enhances immune response against a wide range of viral, bacterial, and parasitic pathogens. In the present study, we investigated the effects of zinc sulphate (ZnSO4) supplementation (20 mg/kg/day) during pregnancy in mice, Swiss Webster strain infected by the Y strain of Trypanosoma cruzi. Oral supplementation of zinc sulphate in pregnant and non-pregnant infected animals did not affect the count of blood parasites as well as tissue parasitism in the heart, liver, and spleen. Zinc supplementation did not alter female body weight, the length of fetuses and neonates, placental size/weight and mortality rate. Among zinc supplied animals, no significant plasmatic zinc concentrations were observed. Concerning to tissue zinc concentrations, only the liver displayed enhanced values as compared to other organs. For placental parasitism, zinc supplied group displayed a significant decrease in amastigote burdens (P < 0.05). However due to the reduced number of parasite burdens in placenta of animals supplied with zinc, these data suggest that zinc was partially effective in up-regulating the host’s immune response against parasite, probably attenuating the infection in fetuses.  相似文献

Because of the very wide spectrum of infectious and non-infectious diseases for which preparations from Azadirachta indica are said to be efficacious, it was suspected that a general immunopotentiating ability could be part of the mechanisms by which it ameliorates so many disease conditions. Using the haemolytic plaque technique, an aqueous extract of Azadirachta indica stem bark was shown to enhance the immune response of BALB/C mice to sheep red blood cells in vivo.  相似文献

This study was conducted to evaluate the Salvia officinalis hydroalcoholic extract on fertility capacity and behavioral features in rats exposed to immobilization stress. Male rats were randomly divided into five groups; Control; Stressed rats; and Stressed rats received 50, 100 and/or 200 mg/kg bw S. officinalis hydroalcoholic extract. To induce stress, rats were immobilized for 49 days and received S. officinalis extract orally. On day 56, we analyzed behavioral tests and evaluated reproduction capacity by measuring LH, FSH, and testosterone. Sperm parameters such as motility, viability, and total count were also determined. Bodyweight changes were also calculated on day 56. Male rats from different groups were mated with healthy female rats. Data showed that the use of 100 and 200 mg/kg bw S. officinalis extract in stressed rats increased bodyweight gain and improved behavioral disorders compared to control‐matched groups (p < .05). Besides, administration of 100 and 200 mg/kg bw S. officinalis extract had the potential to improve sperm parameters and fertility capacity in stressed rats (p < .05). Decreased testosterone levels were blunted in the stressed rats that received plant extract coincided with the reduction of LH and FSH compared to control‐matched stressed rats (p < .05). We found neutral effects in stressed rats that received 50 mg/kg bw plant extract. Collectively, the hydroalcoholic extract of S. officinalis could improve the fertility capacity and behavioral features under stressful conditions in a dose‐dependent manner.  相似文献

将45只6周龄体质量(18±2)g SPF级雄性SD小鼠随机分为5组(n=5),常温对照组5只(A组);低温对照组冷刺激4、8h各5只(B组);冷诱导RNA结合蛋白(cold-inducible RNA binding protein,CIRP)过表达组冷刺激4、8h各5只(C组),注射CIRP过表达慢病毒液0.1mL;CIRP干扰组冷刺激4、8h各5只(D组),注射CIRP干扰病毒液0.1mL;阴性对照组冷刺激4、8h各5只(E组),注射空载体病毒液0.1mL。A组置于常温(21±0.1)℃条件下饲养,后4组置于低温(4±0.1)℃环境饲养,分别在4h及8h后通过摘除眼球的方法采取血液。采用ELISA法检测血清中IL-1、IL-2、IL-6、GSH-PX、SOD、T-AOC、MDA、丙酮酸、PFK-1、LDH的含量。结果显示:(1)冷应激处理后,小鼠血清中相关细胞因子水平出现明显变化,人为的致使CIRP在小鼠体内出现不同的表达对血清中相关细胞因子的影响差异显著(P0.05);(2)冷应激处理后,小鼠血清中氧化还原指标GSH-PX、SOD、T-AOC、MDA变化非常明显,人为的致使CIRP在小鼠体内出现不同的表达对血清中相关氧化还原指标的影响差异极显著(P0.01);(3)冷应激处理后,血清当中能量代谢相关指标无明显变化,人为的致使CIRP在小鼠体内出现不同的表达对血清中相关能量代谢指标的影响差异不显著(P0.05)。结果表明:在不影响动物机体能量代谢的情况下,CIRP可以有效地抑制细胞内氧自由基的生成,从而提高小鼠抗氧化能力,以及调节动物机体的相关免疫因子,对小鼠抵抗低温应激起到积极地保护作用。  相似文献

The aim of this study was to evaluate possible effects of improved protein and energy diet upon reproductive outcomes of adult goats under marginal rangeland grazing conditions and exposed to the male effect during the anoestrous-dry season. Adult goats (Nubian × Criollo, nonpregnant, nonlactating, n?=?160, 22° 50' N, 100° 59' W, 1,653?m) were randomly divided in two experimental groups: Control (CG; n?=?80) and Supplemented (SG; n?=?80). Both CG and SG goats were kept together during the day for ranging a semiarid rangeland while separated accordingly at night. SG goats were individually supplemented [400?g?kg(-1)?day(-1)?; 14% CP and 2.92 Mcal DE kg(-1)] during a 30-day period. Thereafter, both the CG [35.5?±?0.94?kg LW, 1.95?±?0.06 U, body condition score (BCS)] and the SG [39.5?±?0.94?kg LW, 2.29?±?0.06 BCS] were exposed to adult males (Nubian, n?=?4, two per group) of proven libido and fertility and kept together at night (1700-0900) during a 42-day experimental breeding period. At the beginning of the breeding period, both LW (P??0.05), respectively. Results suggest that nutritional supplementation and the male effect were able to successfully invoke neurophysiological pathways to increase ovarian activity and to promote a uterine milieu prone to the establishment of pregnancy during the anoestrus season. However, the increased abortion rates observed in the study occurred irrespective of the nutritional regime offered during the premating stage. Therefore, there is a need to align the last third of gestation to the onset of the grazing season in order to increase reproductive and economic efficiency in those rangeland-based marginal goat production systems.  相似文献

Heterophil/lymphocyte (H/L) ratios began to rise 18 hours later, reaching their maximum value (0.62) in 20 hours and returning to pre-stress values after 30 hours. This study investigated the protective effects of probiotic on heat stress‐induced intestinal injury and inflammatory response in broilers. A total of 180 male broilers were randomly allocated to three treatments with four replicates each from 22 to 42 days of age. The broilers were either raised under thermoneutral (TN) conditions (23 ± 1°C) or subjected to cyclic heat stress (28–35–28°C for 12 hr daily). The broilers kept at TN conditions were fed a basal diet, and those exposed to heat stress were fed basal diets supplemented with or without probiotic at a dose of 1.5 × 108 cfu/kg. Compared with the TN group, heat stress decreased (p < .05) the growth performance, reduced (p < .05) villus height and villus height: crypt depth ratio in intestinal mucosa, increased (p < .05) serum levels of D‐lactic acid on day 28 and endotoxin, TNF‐α and IL‐6 on day 42, and decreased (p < .05) serum IL‐10 content on day 42. Dietary supplementation of probiotic reversed (p < .05) all these changes except for the growth performance in heat‐stressed broilers. In conclusion, dietary inclusion of probiotic could improve intestinal morphology and barrier function, alleviate inflammatory response, but exert no ameliorative effect on growth performance of broilers under cyclic heat stress.  相似文献

Sows subjected to prenatal stress have been found to produce offspring that have altered responses to stress. Our objective was to determine if exposing a sow to stress would alter the response of the offspring to lipopolysaccharide (LPS) at 2 mo of age or their response to mixing stress at 4 mo of age. Sow treatments consisted of intravenous injections of ACTH (1 IU/kg of BW), exposure to rough handling for a 10-min duration (rough), or no treatment (control) once per week from d 42 to 77 of gestation. At 2 mo of age, pigs from each treatment, 1 per litter (n = 21, 17, and 15 for the ACTH, rough, and control treatments, respectively), were challenged with 2 μg of LPS/kg of BW or saline, or served as a noninjected control. Their behavioral response to a human approach test and salivary cortisol were measured. At 4 mo of age, 1 pig from each treatment (n = 14, 14, and 15 for the ACTH, rough, and control treatments, respectively) was taken from its home pen and placed in a pen of unfamiliar pigs. At this time, a punch biopsy wound (6 × 6 mm) was created to measure the ability of the pig to heal the wound. At this same time, each pig received a 1-mL intramuscular injection of 20% ovine red blood cells (oRBC), and then a second injection of oRBC at 21 d postmixing. Blood samples were collected 3 times per week for 2 wk and then once a week for 4 more weeks. Blood samples were analyzed for cortisol, porcine corticosteroid-binding globulin, antibody response to oRBC, and nitric oxide production by macrophages. Behavior was recorded during the first 5 d after mixing. All pigs in the LPS challenge responded with characteristic sickness behavior; however, pigs in the rough treatment showed less sickness behavior than those in the other 2 treatments (P < 0.05). Maternal stress treatment did not affect (P < 0.43) salivary cortisol. Pigs from all treatments responded similarly to mixing stress with regard to cortisol, porcine corticosteroid-binding globulin, antibody titers, nitric oxide production, and hematology measures, and all pigs experienced the same amount of aggression in response to mixing. Without altering peripheral measures of stress responsivity, prenatal stress enhanced the ability of pigs to cope with a simulated immune challenge, which could prove to be an adaptation to challenging environments.  相似文献

采用单因子试验设计。选择 96头 2 8日龄大×长×北三元杂交断奶仔猪 ,随机分成 3组 ,研究饲料中添加不同剂量 (0、5 0、10 0mg/kg)植物性抗应激复合添加剂 (即植物提取物 ,以酸枣仁皂甙元为主 )对仔猪生产性能的影响。试验期 2 8d。试验结果表明 :饲料中添加植物提取物 5 0mg/kg和 10 0mg/kg ,日增重提高 7.1%~ 9.2 %(P <0 .0 5 )、采食量增加 3.5 %~ 4 .9%(P <0 .0 5 ) ,饲料转化率改善 (F/G) 3.2 %~ 3.7%(P <0 .0 5 ) ,仔猪腹泻率明显降低 ,皮毛光泽 ,行为安祥。  相似文献

目的:探讨黑种草籽乙酸乙酯提取物和乙醇提取物对成纤维细胞炎症模型作用及保护大肠杆菌感染小鼠的作用机制.方法:将两种提取物按不同浓度添加到细胞炎症模型中,观察细胞形态;将两种提取物均按1 g/mL浓度腹腔注射给炎症模型小鼠,观察小鼠临床症状,ELISA检测血清白细胞介素-10(IL-10)的表达水平.结果:0.1 g/mL乙酸乙酯提取物和0.4 g/mL乙醇提取物均可引起细胞病变,细胞固缩脱壁或裂解破碎;乙醇提取物在其安全浓度0.1-0.2 g/mL、乙酸乙酯提取物在其安全浓度0.025-0.05 g/mL均对细胞炎症模型有抗炎作用.与对照组比较,模型组小鼠血清IL-10显著升高(P<0.05);两种提取物治疗组IL-10水平均较模型组低,与对照组比较无显著差异,两种提取物(1 g/mL)均有降低炎症小鼠血清IL-10水平的趋势,对小鼠大肠杆菌的感染有保护作用,且乙醇提取物较乙酸乙酯提取物有效.结论:黑种草籽乙酸乙酯提取物和乙醇提取物在安全浓度内均对细胞炎症模型有抗炎作用;两种提取物(1 g/mL)均可降低炎症小鼠血清IL-10水平,且对小鼠大肠杆菌感染有保护作用,乙醇提取物较乙酸乙酯提取物好.  相似文献
