Glucose-6-phosphate dehydrogenase (G6PDH), lactate dehydrogenase (LDH) and pyruvate kinase (PK) are key metabolic enzymes. G6PDH has been used as a biomarker of pollution-induced carcinogenesis in fish. LDH has been used as marker of lesions in toxicology and clinical chemistry, and PK catalyses the conversion of phosphoenol pyruvate to pyruvate, with regeneration of ATP. The effect of different concentrations of lead nitrate on the activity of these enzymes in two different early ontogenetic stages (embryonic and free embryonic stage) of the African catfish Clarias gariepinus was investigated. Embryo homogenates were used for measurements of G6PDH, LDH and PK activity spectrophotometrically at 340 nm and 25°C. The ontogenetic variations of the three enzymes during early ontogeny, from the 30 h to the 168 h post-fertilisation stage (PFS) (beginning of exogenous feeding), were studied. There was a significant decrease in activities of all three enzymes from 30 h-PFS to 96 h-PFS, followed by a significant increase in G6PDH and LDH. PK showed insignificant fluctuations in activity. Different patterns of enzyme activities were recorded due to exposure to different lead nitrate concentrations (100 μg/l, 300 μg/l and 500 μg/l). In the pre-hatching stage (30 h-PFS) the activity of the three enzymes increased at exposure to 100 μg/l lead nitrate and then decreased with increasing dose. In the post-hatching stages (48 h-PFS–168 h-PFS) G6PDH activity increased and LDH activity decreased with increasing lead concentrations. Unlike G6PDH and LDH, the PK enzyme fluctuated during the post-hatching stages and did not reveal a specific trend of response (increase or decrease) with increasing lead concentrations. Therefore, the measurement of G6PDH and LDH activities, but not PK activity, could be useful biomarkers of intoxication to reveal the embryotoxic potential of lead nitrate in fish embryos. The post-hatching stages of the African catfish are more sensitive than the pre-hatching stage (30 h-PFS) is, probably due to the protective capacity provided by the hardened chorion. The interaction and the main effects of age and lead doses were found to be highly significant, referring to the great impact of lead on these enzyme systems with increasing early development.  相似文献

Problems of limited number of dry feeds as supplement or replacement of live feeds have led to poor larval nutrition in many species of fish. Therefore, the suitability of co‐feeding 8‐day‐old African catfish (Clarias gariepinus) posthatch larvae using live feed (Artemia salina) and formulated dry diet containing freshwater atyid shrimp (Caridina nilotica) during weaning was investigated. The experiment ended after 21 days of culture and respective groups compared on the basis of growth performance, survival, feed utilization and nutrient utilization. Larvae co‐fed using 50%Artemia and 50% formulated dry diet resulted in significantly (P < 0.05) better growth performance, food gain ratio (FGR), protein efficiency ratio (PER) and productive protein values (PPV) than other treatments. The lowest growth performance occurred in larvae weaned using 100% formulated and commercial dry diets. Better survival of over 90% was obtained in larvae weaned using 50%Artemia and 50% dry diet, while abrupt weaning using 100% dry diets resulted in lower survival (<75%). These results support a recommendation of co‐feeding C. gariepinus larvae using a formulated dry diet containing C. nilotica and 50% live feed when weaning is performed after 8 days posthatching period.  相似文献

Sutchi catfish Pangasianodon hypophthalmus hatch with morphologically immature sensory organs; however, sensory organs develop rapidly with larval growth. Two-day-old larvae commenced ingesting Artemia nauplii. The larvae displayed many taste buds on the barbels, the head surface, and in the buccal cavity. Other sense organs were also well developed at this stage. Feeding experiments revealed that 2-day-old larvae ingested Artemia under both light and dark conditions, moreover, the larvae could ingest frozen dead Artemia. The ingestion rates for 4- and 7-day-old larvae were significantly higher under dark conditions than under light conditions. The rates using frozen dead Artemia were mostly higher than the rates using live Artemia. Therefore, feeding behavior under dark conditions is most likely not mediated by visual or mechanical senses, but rather by chemosensory senses, such as taste buds. Larval fish are vulnerable to predators; thus, if they can search for and eat food at night, they can avoid diurnal predators. The behavior observed here appears to represent their survival strategy. Moreover, these results suggest a new possibility that sutchi catfish larvae can be reared under dark or dim light conditions in order to improve survival and growth rates as in the case of African catfish Clarias gariepinus.  相似文献

The suitability of the freshwater rotifer Brachionus calyciflorus as starting food for the larviculture of African catfish, Clarias gariepinus Burchell, was investigated through studies of growth and fatty acid profiles in relation to different feeding sequences combining live food and dry diet in various proportions and during different mixed feeding periods. The best results for survival were observed when rotifers were supplied during the first week of feeding, i.e. sequences R2 (exclusive supply of rotifers, then dry diet from day 8, onwards) and R5 (50% rotifers+50% dry diet until day 7, then dry diet 100%), reaching 99.2% and 96.3%, respectively. The specific growth rate of larvae was largely dependent on the duration of preliminary feeding with the rotifers. A feeding with rotifers as a unique food source did not produce satisfactory growth during the first week of feeding. A precocious weaning showed that the highest growth rate and protein efficiency ratio (PER) can be obtained by feeding the larvae rotifers in association with a dry diet. The best PER and protein productive value (PPV) were recorded with feeding sequences R2 and R5. On the other hand, the series of polyunsaturated fatty acids was characterized by a relatively constant concentrations, and represented about 11.6% of the total fatty acids in sequence R2 because of the presence of the acids of the linoleic series, which apparently originated from the food. The R5 regime provided larvae with significant amounts of highly unsaturated fatty acids, such as linolenic acid C18:3n-3.  相似文献

In the present study, thiourea-induced thyroid hormone depletion and thyroxine (T4) ‘overdose’ were used as a strategy to understand the influence of thyroid hormones on ovarian recrudescence of juvenile (3-months-old), immature (8-months-old) and adult (1-year-old) air-breathing catfish, Clarias gariepinus. Thiourea-induced thyroid hormone depletion in juvenile catfish impaired ovarian development, but no significant effect was observed in immature catfish and during late stage of ovarian recrudescence of mature catfish. T4 treatment in females undergoing late stages of ovarian recrudescence induced rapid oocyte growth by promoting its early entry into maturational phase as evident from the presence of more number of vitellogenic and post-vitellogenic follicles, decrease in aromatse immunoreactivity and reduced estradiol–17β levels. Hence, thyroid hormones have an important role to play during early stages of ovarian development and vitellogenesis of catfish and also indicating that thyroid has a stage dependent effect on ovary.  相似文献

Rainbow trout (100 g initial weight) were subjected to the combined effect of two culture densities (15 and 40 kg m−3, D15 and D40, respectively) and two dietary energy levels (22 and 27 MJ kg−1 E22 and E27, respectively) during a 75-days experimental period. At the end of the experiment, the growth rate as well as the metabolic and oxidative status of liver and muscle of fish were studied.The results showed that combination of culture density and dietaryenergy level negatively affected growth, cholesterol and LDL plasma levels and oxidative stress in muscle. Higher culture density negatively affected the values of total protein, triglycerides, and HDL in plasma, values of hepatic and muscular metabolic activities pyruvate kinase (PK), citrate synthase (CS), and hydroxiacil-CoA dehydrogenase (HOAD); glutamate pyruvate transaminase (GPT) and glutamate oxaloacetate transaminase (GOT) activities in plasma, liver, and muscle; glucose 6P dehydrogenase (G6PDH) activity in muscle; and oxidative stress in liver.High energy intake, adversely affected the hepatic activity of G6PDH, HOAD, GPT and oxidative stress in muscle.Consequently our results indicate that a combination of high culture density and a high level of dietary energy (27 MJ kg−1 in diet) exert a negative impact on the physiology and consequently on the welfare of the farmed fish.  相似文献

A high rate of sibling cannibalism is one of the principal obstacles in the rearing of larvae and juveniles of the African catfish Clarias gariepinus. This paper examines the underlying behavioural components of agonistic behaviour under culture conditions. Ten day old catfish larvae were stocked at 30 larvae l–1 in three tank designs with different surface areas and equal volumes. Stocking densities were 1.2, 0.6 and 0.3 fish cm–2 bottom surface. Growth did not differ between treatments. The highest mortality over 30 days was recorded at the medium density (0.6 larvae l–1). Fish stocked at this density showed the highest rate of aggression, while there was no difference in aggression between the highest and the lowest stocking densities. The results indicate that stocking density should be at least as high as 1.2 larvae cm–2 bottom surface area to obtain high production at best survival rates.  相似文献

Abstract. Three feeding trials, involving pre-pupal larvae of soldier fly, Hermetia illucens L., grown on poultry manure, were conducted to assess: (1) channel catfish, Ictalurus punctatus (Rafinesque), response to substitution of dried meal larvae for the fish meal component of the catfish diet and (2) if feeding 100% whole or chopped larvae to channel catfish or blue tilapia, Oreochromis aureus (Steindachner), will support normal growth comparable to those fed a commercial diet. Effects on fish quality were also evaluated. Replacement of 10% fish meal with 10% soldier fly larvae resulted in slower growth over a 15-week period for subadult channel catfish grown in cages (trial 1). However, the replacement did not reduce growth rate significantly when channel catfish were grown in culture tanks at a slower growth rate (trial 2). Feeding 100% larvae did not provide sufficient dry matter or protein intake for good growth for either species grown in tanks (trials 2 and 3). Chopping of the larvae improved weight gain and efficiency of the utilization.  相似文献

Direct use of pig wastes as inputs into fish culture systems may be unacceptable or an inferior use of valuable inputs. High value, but non-filter feeding fish, such as African catfish, Clarias gariepinus (Burchell), may be unable to recover nutrients efficiently through the pond food web and require complete diets in intensive culture. Live feeds such as the larval stage of the green blow fly. Lucilia sericata, can be used as intermediate organisms to utilize pig waste and subsequently be fed live as part of a complete ration for catfish raised in cages. The nutrient efficiency of the system is further enhanced by the stocking of phytophagous fish, the Nile tilapia, Oreochromis niloticus (L.), in the pond in which the catfish culture cages are suspended. A model derived from on-farm experimentation is presented that demonstrates system design and nutrient efficiencies. An extrapolated catfish production of 61 year1 using only fly larvae produced from a standing herd of approximately 1000 fattening pigs was demonstrated. The static water pond in which the catfish were cultured ensured that the environmental impact of both pig and catfish systems was minimal compared to conventional production systems.  相似文献

The ontogenetic development of the digestive enzymes amylase, lipase, trypsin, and alkaline phosphatase and the effect of starvation in miiuy croaker Miichthys miiuy larvae were studied. The activities of these enzymes were detected prior to exogenous feeding, but their developmental patterns differed remarkably. Trypsin activity continuously increased from 2 days after hatching (dah), peaked on 20 dah, and decreased to 25 dah at weaning. Alkaline phosphatase activity oscillated at low levels within a small range after the first feeding on 3 dah. In contrast, amylase and lipase activities followed the general developmental pattern that has been characterized in fish larvae, with a succession of increases or decreases. Amylase, lipase, and trypsin activities generally started to increase or decrease at transitions from endogenous to exogenous feeding or diet changes, suggesting that these enzymatic activities can be modulated by feeding modes. The activities of all the enzymes remained stable from 25 dah onwards, coinciding with the formation of gastric glands and pyloric caecum. These results imply that specific activities of these enzymes underwent changes due to morphological and physiological modifications or diet shift during larval development but that they became stable after the development of the digestive organs and associated glands was fully completed and the organs/glands functioned. Trypsin and alkaline phosphatase were more sensitive to starvation than amylase and lipase because delayed feeding up to 2 days after mouth opening was able to adversely affect their activities. Enzyme activities did not significantly differ among feeding groups during endogenous feeding; however, all activities were remarkably reduced when delayed feeding was within 3 days after mouth opening. Initiation of larvae feeding should occur within 2 days after mouth opening so that good growth and survival can be obtained in the culture.  相似文献

Abstract

Three experiments were conducted. In Experiment 1, the acceptance time of pelleted diets sprayed with fresh palm fruit extract (FPFE), commercial palm oil (COM), or a control diet to African catfish, Clarias gariepinus, fingerlings was investigated. In Experiment 2, the effects of five diets on growth and survival of African catfish larvae were determined: (1) bambara nut waste-based (BW) diet; (2) bambara nut waste-based diet with 5% of diet formula of FPFE (BWP); (3) bambara nut waste-based diet with 5% of diet formula of FPFE plus 1.5% of diet formula of Spirulinapowder (BWPS); (4) fish waste-based diet (FWP); and (5) brine shrimp, Artemiasp., nauplii (control). In the BWP, BWPS, and FWP diets, fresh palm fruit extract was sprayed on the diets as a feed attractant. The Spirulinapowder was included in BWPS as an additional attractant. In Experiment 3, the effects of seven diets on growth and survival of the African catfish larvae were investigated: (1) BW; (2) BWP; (3) FWP; (4) a bambara waste-based diet with 5% COM (BWC); (5) a fish waste-based diet with 5% of formula as COM (FWC); (6) a fish waste-based diet with neither FPFE nor COM, and (7) brine shrimp nauplii (control). African catfish finger-lings accepted the pellets containing FPFE in significantly less time (P < 0.01) than they did the other pelleted diets. Inclusion of FPFE as 5% of diet formula significantly (P<0.05) improved the growth and survival of African catfish larvae fed formulated diets. Inclusion of Spirulinaat 1.5% of diet formula had no effect. Brine shrimp was nutritionally superior to the formulated diets for the African catfish larvae during the first 28 days of feeding. The results indicate that FPFE can be added to a diet for African catfish at 5% of diet formula. However, brine shrimp nauplii fed to larvae had the highest percentage survival and highest growth rates of any of the treatments.  相似文献

Effect of experimentally induced thyroxine overdose on the testis and seminal vesicles was studied in the air-breathing catfish, Clarias gariepinus during the preparatory and the pre-spawning phase. The present study revealed a marked reduction in testosterone level in serum, testis and seminal vesicles (SV). Histological examination showed a considerable reduction in the number of spermatozoa/spermatids in the seminiferous tubular lumen as well as depletion of fluid in the loculi of SV. SDS-PAGE analysis of SV fluid proteins demonstrated a significant decrease in the level of a ~27 kDa protein in thyroxine treated fishes. Juvenile African catfish, Clarias gariepinus (Burchell), were fed low-tocopherol, practical test-diets containing graded levels of α-tocopheryl acetate (0, 80, 200 and 500 mg kg?1 dry feed) at proportionately 0.02 body weight per day for 70 days. After the feeding period, selected fish tissues were assayed for α-tocopherol and thiobarbituric acid reactive substances (TBARS) concentrations, before and after iron-ascorbate stimulated peroxidation. Results show that tissue accretion of α-tocopherol in muscle, liver and blood plasma increased linearly (R2= 0.83, 0.82 and 0.93, respectively) in response to elevated dietary supplementation of α-tocopheryl acetate within the range of doses under study. In muscle and liver this resulted in a significant decrease (P < 0.05) in basal TBARS and TBARS after stimulated peroxidation, indicating a greater stability against oxidation. Growth indices were not significantly affected (P > 0.05) by dietary α-tocopheryl acetate inclusion. Clarias fed the highest α-tocopheryl acetate dose (500 mg kg?1 dry feed) were observed to have significantly lower (P < 0.05) haematocrit (% packed cell volume) than fish fed the basal diet (0 mg kg?1 dry feed). It was concluded that supplemental α-tocopheryl acetate in practical diets, for Clarias gariepinus, was effective in reducing the degree of tissue-lipid peroxidation under conditions of increased oxidative stress. Thus, the post mortem resistance to oxidation achieved would improve the stability of catfish products destined for human consumption.  相似文献

Rates of predation by tadpoles of Bufo regularis on larvae of the African catfish Heterobranchus longifilis were determined under controlled laboratory conditions. Predation rates were high on early stage yolk‐sac and first‐feeding larvae, which have a poorly developed escape ability. A steady decline in predation rates was found with an increase in the size of the larvae. Predation rates were significantly related to prey density. The relationship between larval biomass eaten and biomass available suggested that tadpoles were not satiated when feeding on the largest larvae, probably because of the improved escape ability of these larvae. Prey movement was not found to be an important factor eliciting predation by tadpoles. Starved larvae were more vulnerable to predation because of a poor escape response. Our results indicate that tadpole predation may represent a substantial source of mortality of catfish larvae in earthen primary nursing ponds.  相似文献

Temporary accumulation of ascorbic acid 2-sulfate (AAS) was measured to estimate food intake and gut evacuation in larvae of African catfish. Fish larvae were fed decapsulated cysts of Artemia containing AAS. In a first experiment it was found that no biosynthesis of AAS occurs in the larvae of this species. In a second experiment, the gut contents of the fish larvae fed were calculated as they changed during development. In a third experiment, the gut evacuation rate of fish larvae was determined during continuous and discontinuous feeding regimes in the first five days after the start of exogenous feeding. Food consumption by catfish larvae increased from 46.5% of their body dry weight (BDW) on day 1 after the start of exogenous feeding to 53.8% BDW on day 3. Thereafter, food consumption decreased to 27.8% BDW on day 5. A similar pattern was observed for gut evacuation, which increased during the first days of exogenous feeding and decreased as fish growth continued. The rate of gut evacuation in a continuous feeding regime was significantly higher (P < 0.05) than that under discontinuous feeding. On day 1 post-hatch and 7 h after first food ingestion the fish larvae evacuated 87% of the food in continuous feeding compared with 43% under discontinuous feeding. It was found that gut emptying differs during larval development. Under continuous feeding, on days 1 and 3 post-hatch and 11 h after the first meal 90% of the food was evacuated compared with 71% evacuated on day 5. The advantages and limitations of the AAS method for estimation of food consumption by fish larvae are discussed.  相似文献

The present study analyzed the effects of prey density, the time of day, and ontogenetic development on the predation of Artemia nauplii by the larvae of the Amazon river prawn, Macrobrachium amazonicum, as well as possible synergy among these factors. Larvae were raised in 120‐L tanks with biological filter systems, and fed on recently hatched Artemia nauplii, using two feeding management protocols: (a) fed once per day at 2000 h (high density – HD) and (b) half of the ration provided at 2000 h, complemented at 0800 h the following day by a replacement of the nauplii consumed up to a maximum of the full ration (low density with replacement – LDWR). Each treatment consisted of six replicates. The consumption of nauplii was estimated prior to the feeding times. Consumption varied according to time of day, ontogenetic development, and feeding protocol. The larvae ingested more nauplii during the daytime at most developmental stages. Ingestion rates were similar during the day under both treatments, but at night the higher density of prey in the HD treatment caused a higher encounter rate and increased ingestion of nauplii by the larvae. Among the performance indicators only survival was greater in HD in comparison with LDWR; productivity and dry weight were similar. The results indicate a circadian trophic rhythm in M. amazonicum, with the encounter rate being an important mechanism for the capture of prey during the night. A second mechanism – probably the visual system – aids the perception of prey during the daytime. Based on these results, we suggest that feeding captive Amazon river prawn larvae only once a day would be appropriate and economically beneficial. Further work is necessary to determine the most effective time that this single feed should be applied.  相似文献

The fillet yield, total protein content and amino acid profile of the African sharptooth catfish Clarias gariepinus (Burchell) was compared with that of its hybrid with the African catfish Heterobranchus longifilis Valenciennes. The flesh of the C. gariepinus × H. longifilis hybrid had a protein content of 18.3%, compared with 18.6% for C. gariepinus. The amino acid profiles were similar for the two groups. The fillet yield of the C. gariepinus females was significantly lower than that of the male and female hybrids and C. gariepinus males. The difference in fillet yield was attributed to the high gonadosomatic index (GSI) of the C. gariepinus females, compared with the other groups. The overall fillet yield was 38.9% for C. gariepinus and 43.8% for the hybrid, which has important implications for African catfish production.  相似文献

为了探究饲料不同脂肪水平对黄颡鱼卵巢脂肪代谢潜在的影响,本研究设计了活体与离体两个实验。活体实验中,分别投喂3组不同脂肪水平的饲料(脂肪含量分别为6.98%、11.34%和15.41%)8周。离体实验中,分离原代黄颡鱼卵母细胞,采用(0、0.2和0.5 mmol/L)3组不同脂肪酸浓度孵育48 h。活体实验结果显示:与6.98%和15.41%脂肪水平组相比,11.34%脂肪水平显著增加黄颡鱼卵巢甘油三酯水平(TG),并上调FAS、G6PD、6PGD和ME的酶活性水平,以及LPL和CD36基因表达水平。与6.98%和11.34%脂肪水平组相比,15.41%脂肪水平组显著升高黄颡鱼的性腺指数(GSI),并上调CPTIA和DNMT3b的基因表达水平,以及PI3KCa启动子-64和-52 CpG位点的甲基化水平,而显著降低PI3KCa的基因表达水平。体外细胞实验显示:与对照组(0)相比,0.5 mmol/L脂肪酸孵育显著增加卵母细胞TG含量,并上调FAS、G6PD和ME酶活水平,以及G6PD和PI3KCa的基因表达水平。同时,与对照组相比,脂肪酸孵育组显著降低CPTIA、ACCb、LPL、CD36、DNMT1以及DNMT3b基因的表达水平。然而,脂肪酸孵育对卵母细胞PI3KCa的启动子甲基化水平没有影响。研究表明,饲料不同脂肪水平影响卵巢TG的合成,可能主要是通过脂肪转运相关基因,并且高脂肪很可能是通过影响黄颡鱼卵巢PI3KCa启动子甲基化水平来影响PI3KCa的表达。而离体条件下,脂肪酸促进卵母细胞TG的合成很可能是通过升高脂肪合成相关基因、降低脂肪分解和转运相关基因来实现,但脂肪酸孵育不通过影响黄颡鱼卵母细胞PI3KCa甲基化水平来影响PI3KCa基因表达。  相似文献
