A feeding experiment was conducted on gilthead seabream (Sparus aurata) broodstock to investigate the incidence of n-3 highly unsaturated fatty acids (n-3 HUFA) dietary deficiencies on the lipid composition of female liver, gonads and eggs, in relation to spawning quality. Broodstock were fed a control (C) diet or a n-3 HUFA deficient (D) but linolenic acid rich diet. After 20 weeks of feeding, the results showed that levels of total neutral (TNL) and total polar (TPL) lipids of female gonads and eggs were independent of diet. However the fatty acid composition of phosphatidylcholine (PC), phosphatidylethanolamine (PE) and phosphatidylinositol (PI) of female liver, gonads and eggs in the two groups of fish showed marked differences, reflecting the influence of fatty acid levels in the broodstock diets. This influence was even higher in TNL than in the phospholipid classes examined. In fish fed n-3 HUFA deficient diet, fatty acid composition of TNL of female gonads and eggs reflected the diet more than liver. A higher egg production in broodstock fed C diet (1.8% n-3 HUFA in diet) was extended to spawning quality such as percentages of fertilised and hatched eggs.

This study investigated the effect of dietary arachidonic acid (AA) in broodstock of Japanese flounder on subsequent egg and larval quality. Diets with similar proximate composition and n-3 HUFA level, but with different AA levels (0.1%, 0.6% and 1.2% of diet), were fed to the broodstock from 3 months before and during the spawning season. Spawning was observed from March to May. Total egg production over the spawning season was highest in fish fed the 0.6% AA diet and lowest in fish fed the 1.2% AA diet. All parameters measured as egg quality (percentage of buoyant eggs, hatching rate, larval survival and normality of larvae.) were highest in fish fed the 0.6% AA diet. AA content in eggs proportionally increased with the dietary AA level. EPA content of polar lipids of eggs had a negative correlation with the AA level in diets whereas the DHA content was independent of dietary AA. The results of this study indicate that a supplement of AA at 0.6 g/100 g diet improved the reproductive performance of Japanese flounder, but a higher level of AA (1.2 g/100 g diet) negatively affected both egg and larval quality due to a potential inhibitory effect on EPA bioconversion.

Total lipid (TL), lipid classes and fatty acid composition of neutral (NL) and polar (PL) lipids were studied in the gonads, liver and muscle of gilthead seabream males ( Sparus aurata ) fed a control diet (diet C) or an n-3 and n-6 highly unsaturated fatty acids (HUFA)-deficient diet (diet D), at different stages of the reproductive cycle. Between pre-spermatogenesis (November) and spermatogenesis (March), the lipid content was high and particularly rich in cholesterol, phosphatidylcholine and phosphatidylethanolamine in gonads from both dietary groups. At post-spermatogenesis (June), TL and especially PL dramatically decreased in the gonads from both groups. However, at this period diet C fish gonads were richer in triacylglycerides (TAG) than those from diet D fish. The liver lipid contents and particularly TAG were over 200% lower in June than in March for both groups. Nevertheless, the most noteworthy depletion of lipids during this period was achieved by the n-3 HUFA in diet D fish. Conversely, arachidonic acid (20:4n-6) did not decrease in NL or PL from gonads and liver in groups C and D. Muscle lipids from diet C fish were relatively insensitive to seasonal influences. However, in June, the muscle TAG content was significantly reduced in diet D fish.

We investigated the effect of high levels of n−3 highly unsaturated fatty acids (n−3 HUFA) in broodstock diet on egg quality and chemical composition of eggs of Japanese flounder. The broodstock were fed diets containing three levels of n−3 HUFA (2.1%, 4.8% or 6.2%) 2 months before and during the spawning period. No significant difference was found for weight gain of broodstock among the treatments. Egg production was highest in fish fed the highest level of n−3 HUFA. However, egg quality parameters, such as percentage of buoyant eggs, hatching rate and percentage of normal larvae, were significantly higher in the group fed the lowest n−3 HUFA diet. The fatty acid composition of eggs was influenced more markedly in the neutral lipid fraction than in the polar lipid fraction by dietary n−3 HUFA levels. Arachidonic acid (AA; 20:4n−6) and egg quality parameters both decreased with increasing dietary n−3 HUFA levels. The results suggest that a high level of n−3 HUFA in broodstock diet negatively affects egg quality of Japanese flounder.

ABSTRACT: The effect of an n-3 essential fatty acid (EFA) -deficient diet on spawning and on the lipid and fatty acid contents of eggs, semen and livers of rainbow trout Oncorhynchus mykiss was investigated. Fish were split into two groups and fed either of two diets for a period of 4 months prior to the start of the spawning season. The control group was fed a commercial diet, containing n-3 EFA, whereas the experimental group was fed an n-3 EFA-deficient diet. Fish were 3 years old at the time of spawning. Eggs and semen were stripped off five females and five males from each diet group and cross-fertilized. Two of the five males fed the deficient diet showed a lower sperm motility, resulting in slightly lower mean hatching rates when crossed with eggs of either group. Higher lipid contents in the EFA-deficient diet were reflected in the egg and semen lipid contents, whereas the lipid contents of male livers of both diet groups were higher than those of female livers. In livers and eggs, the main polar lipid was phosphatidylcholine accompanied by phosphatidylethanolamine in sperm polar lipids. The non-polar lipids of eggs were mainly triacylglycerols but in livers and semen, free fatty acids and free sterols were more abundant. Essential fatty acids, namely n-3 highly unsaturated fatty acids and linolenic acid, were generally lower in eggs, semen and livers sampled from the EFA-deficient diet group. These results indicate that the fertilization, eyed and hatching rates obtained from crossings with males fed the EFA-deficient diet were slightly lower because of the dietary effect on sperm motility. Moreover, lipids of eggs, semen and livers of male and female rainbow trout were influenced greatly by their dietary availability.

This study was conducted to determine the effects of broodstock diet on flame angelfish (Centropyge loriculus) reproduction and generate baseline spawning performance (mean fecundity, egg fertilization, egg viability) and egg biochemistry data for this species. Sixteen flame angelfish broodstock pairs were maintained on one of three formulated diets or a raw diet (consisting of squid, shrimp, fish eggs and algae) to determine the effect of diet composition on the fecundity, egg fertilization rates and egg viability over a 7‐month period. Although all the diets supported similar mean spawn frequency and fecundity over the duration of the experiment, the mean fertilization rates and egg viability were significantly greater among pairs receiving the raw diet. Egg fatty acid composition was generally reflective of the maternal diet and similar to the values reported for other tropical species. In particular, dietary factors such as n‐3 highly unsaturated fatty acid and ARA level (which were elevated in the raw diet and eggs from fish receiving that treatment) are correlated with egg quality and are therefore worthy of future investigation. Because of the ease of conditioning, adaptability to varied formulated diets and continuous, sustained reproductive output, flame angelfish are an excellent model for examining the factors affecting marine fish egg and larval quality.

The arachidonic acid (20:4n-6,AA) requirements of larval summer flounder weredetermined for the rotifer- and Artemia-feeding stages. Experimental emulsionscontained adequate n-3 highly unsaturated fattyacid (HUFA) ratios and emulsion levels of AAwere set at 0, 3, 6, 9, and 12% (AA0, AA3,AA6, AA9, and AA12). Examination of fatty acidlevels in live feeds and larval tissuesconfirmed the physiological incorporation offatty acids relative to dietary levels. In thefirst experiment, survival, growth, andsalinity tolerance (2-h in 70) were measuredat 18 days after hatch (dah) after feeding thelarvae the various levels of AA. Larvae fedAA6-enriched rotifers were better able tosurvive the salinity tolerance test. AAenrichment up to 12% had no effect on growthand survival. In the second experiment, larvaewere fed AA0- or AA6-enriched rotifers until 23dah, followed by unenriched 24- and 48-h Artemia nauplii until 32 dah. These larvaethen were subdivided and fed AA-enriched Artemia from 33-45 dah. At the end of thisexperiment, larvae fed AA6-enriched rotifershad higher survival, increased growth, andsurvived better in the salinity tolerance test(2-h in 80). The enrichment of Artemiadid not have any effect on these variables. Thus, the provision of AA6-enriched rotifers(10 mg AA g–1 DW) early in larvaldevelopment may serve to enhance larval stresstolerance at the rotifer stage while alsoincreasing larval survival, growth, and stresstolerance later in the Artemia stage.

In Eurasian perch (Perca fluviatilis), the variability in spawning quality is a major limiting factor for successful production, especially when breeders are fed with an artificial diet. The influence of the dietary DHA/EPA/AA ratio on the egg and larval quality and on the fatty acid and lipid class composition of eggs has been investigated in perch broodstock. Two experimental diets (16% lipids) with two different DHA/EPA/AA ratios, D1 (3/2/2) and D2 (23/9/1), were compared with a natural diet consisting of cultured carp juveniles, CC (10/10/1) and with a commercial diet for salmonids, CDS (14/16/1). Percentages of fertilization and hatching were comparable between fish fed D1, D2 and CC, with the highest hatching rate observed for D1 (63.5 ± 3.8%). These diets supported better values than the CDS. Larval survival and TL50 observed after osmotic stress were higher for the D1 group, followed by larvae produced by fish fed D2 and CC. Larvae from fish fed D1, D2 and CC were significantly more robust than larvae from the CDS group. Differences were observed regarding the fatty acid (FA) profile in the eggs, which was related to the dietary FA composition. The results indicate that a ratio of 3/2/2 seemed to be effective for obtaining eggs and larvae of good quality.

Eggs from a F1 cultured broodstock of sole were compared with eggs from wild‐caught breeders throughout one spawning season, to evaluate if egg quality may be affected by culture‐related conditions. Fourteen batches of eggs from cultured broodstock and 17 batches from wild‐caught sole were compared with respect to fatty acid (FA) composition, egg size, fertilization rate and hatching rate. Based on a multivariate analysis of the FA profiles, it was possible to discriminate between culture and wild inheritance. Eggs from cultured broodstock had high levels of C20:1(n‐9), C18:2(n‐6) and C18:3(n‐3), whereas eggs from wild fish had high levels of C16:1(n‐7), C20:4(n‐6) and C20:5(n‐3). Differences in FA profiles were most likely related to dietary differences. Fertilization and hatching rates were generally low and lowest in eggs from cultured broodstock, but not related to FA composition. Larval growth of one batch from each group was compared. Larval growth was not correlated to broodstock origin, FA composition or egg or larval size. However, larval survival was significantly lower for larvae from cultured broodstock.

This study investigated the effect of n‐3 to n‐6 fatty acid ratios in broodstock diets on reproduction performance, fatty acid composition of eggs and gonads of tongue sole Cynoglossus semilaevis. Broodstock were fed five isonitrogenous and isoenergetic diets for 60 days. The supplemented lipids were prepared by a combination of fish oil and soybean oil inclusion FO (fish oil); FSO1 (fish oil: soybean oil = 7:1); FSO2 (fish oil: soybean oil = 2.2:1); FSO3 (fish oil: soybean oil = 1:1); FSO4 (fish oil: soybean oil = 1:4.3) as lipid sources with different n‐3 to n‐6 fatty acid ratios 10.40, 5.21, 2.81, 1.71 and 0.87. Results showed that relative fecundity, fertilization rate and survival rate of larvae at 7 days posthatching were all higher in broodstock fed FSO1 and FSO2 diet and significantly (< 0.05) decreased in groups fed FSO3 and FSO4 diets. The best result in starvation tolerance test was obtained in FSO2 diet. The present study suggests that n‐3 and n‐6 PUFA ratio in broodstock diet has a considerable effect on spawning performance, egg and larval quality for C. semilaevis.

Five diets that contained fresh squid meat as the basic constituent and were supplemented with different amounts of highly unsaturated fatty acids (HUFA) and astaxanthin were fed to pond‐reared Penaeus monodon broodstock. Diet A was sole squid meat. Diets B and C were supplemented with astaxanthin 50 and 100 mg kg?1 respectively. Diets D and E were supplemented with HUFA 5 and 10 g kg?1 and astaxanthin 50 mg kg?1 respectively. The result showed that the group fed diet E had the best reproductive performance in all experimental groups. It had a higher proportion of spawns (71.5%), spawning rate (0.047), a shorter latency period (7.7±0.3 d), higher absolute fecundity (× 103) (361.6±5.5) and egg production/female (× 103) (597.0±18.0) than all the other experimental groups. The fatty acid composition in broodstock diets strongly affected the tissue and fecundity of broodstock. Good correlations between the content of 20:4n‐6 in eggs and the fecundity (r2=0.6109) and egg production (r2=0.9876) of broodstock were found. On the other hand, 22:6n‐3 and DHA/EPA ratio was negatively correlated with the fecundity of broodstock (r2=0.5362, 0.8702 respectively). The result also showed that the balance between n‐3 and n‐6 fatty acid families, total polyunsaturated fatty acids and total saturated fatty acid and 20:5n‐3 (EPA) and 22:6n‐3 (DHA) may play vital roles in maturation and reproductive performance of P. monodon broodstock.

The influence of broodstock dietary lipids on egg quality and egg fatty acid composition throughout the spawning season of gilthead seabream was investigated. For this purpose, the fish were fed for 7 months either a control diet (diet C) or a diet deficient in n−3 highly unsaturated fatty acids (n−3 HUFA) but rich in both oleic (18:1n−9) and linolenic (18:3n−3) acids (diet D). Eggs spawned by both groups of fish were sampled at the beginning, middle and end of the spawning season and the fatty acid composition of their neutral (NL) and polar lipids (PL) determined. In the early season, percentages of fertilized and hatched eggs, relative proportions of NL and PL as well as their fatty acid compositions, were not affected by the lipid composition of the broodstock diet. However, the eggs spawned during the middle and late seasons showed marked differences among the two groups of fish, clearly reflecting the influence of dietary fatty acids. This influence was more evident in the neutral lipid fraction than in the polar lipids. No correlation was found between the number of buoyant eggs and eicosapentaenoic (20:5n−3, EPA), docosahexaenoic (22:6n−3, DHA) fatty acids or total n−3 HUFA contents in egg phospholipids. However, a negative correlation was detected when percentages of fertilized eggs were compared with the levels of 18:1n−9, 18:3n−3 and with the ratio 18:1n−9/n−3 HUFA present in the phospholipids. Our results indicate the importance of maintaining not only the level of n−3 HUFA in egg membrane phospholipids, but also the balance between n−3 HUFA and other fatty acids such as 18:1n−9 and 18:3n−3, in order to obtain a high spawning quality.

This study evaluated the nutritional value of dietary n‐3 and n‐6 polyunsaturated fatty acids (PUFA) such as linoleic (LOA) and linolenic (LNA) acids, and highly unsaturated fatty acids (HUFA) such as arachidonic (AA), eicosapentaenoic (EPA), and docosahexaenoic (DHA) acids for juvenile Litopenaeus vannamei, based on their effects on growth, survival, and fatty acid composition of hepatopancreas and muscle tissue. Diets contained 5% total lipid. A basal diet contained palmitic and stearic acids each at 2.5% of diet. Five diets contained 0.5% dry weight of LOA, LNA, AA, EPA, or DHA. An additional diet evaluated HUFA in combination by supplementing at 0.5% of diet, a mixture of n‐3 HUFA. All HUFA showed higher nutritional value than PUFA for shrimp and produced significantly (P < 0.05) higher final weight, weight gain, and total lipid in shrimp muscle. Fatty acid profiles of shrimp tissues reflected the composition of the dietary lipids. In general, saturated fatty acids were more abundant in the neutral factions, while PUFA and HUFA were more abundant in the polar fractions of tissues. Under these experimental conditions, HUFA had much greater nutritional value than PUFA for juvenile L. vannamei; moreover, dietary requirements for PUFA were not demonstrated.

Farmed eels had lower levels of arachidonic acid (20:4 n‐6) (ARA) and higher ratios of eicosapentaenoic acid (20:5 n‐3) (EPA):ARA compared to wild European eels collected from the Baltic Sea and southern Norwegian coast. Eels fed a formulated feed (JD) with a distribution of essential fatty acids (EFA) resembling wild European eel were sampled after 0, 5, 10, 14 and 44 weeks of feeding to examine changes in fatty acid composition (FAC) in ovaries, visceral fat and muscle. The results showed a slow but steady incorporation of EFA. Lipids are incorporated in the oocytes early in oogenesis, and the leading cohort of oocytes is rich in lipid droplets before the onset of vitellogenesis. This indicates that feeding with optimized broodstock feeds should start early to allow the incorporation of EFA in the first cohort of oocytes. At least 14 weeks of feeding is required to change lipid EFA in broodstock eel to resemble EFA in the diet or in wild fish. After 44 weeks of feeding, ARA was significantly higher in the neutral lipids of ovaries (1.9%) compared to visceral fat (1.2%) or muscle (1.0%). EPA:ARA ratios decreased two‐ to threefold in all tissues examined during that time. ARA and docosahexaenoic acid (22:6 n‐3) (DHA) had accumulated in ovarian polar lipids.

Lipid content, lipid class and fatty acid compositions were determined in eggs from wild and captive black sea bass, Centropristis striata L., from northern (New England) and southern (South Carolina) regions to determine the effects of diet on egg composition and fertilization success. The formulated diets fed to the northern captive (NC) fish were higher in total lipids (22%) compared with the cut fish and squid diet fed to the southern fish (SC; 3.3% lipid) and had a higher relative amount of linoleic acid (LA) and lower relative amounts of docosahexaenoic acid (DHA) and arachidonic acid (AA). These dietary differences were broadly reflected in the egg lipid composition. The LA levels were higher in all lipid classes in eggs of NC fish while AA levels were lower. The DHA was higher in the major polar lipids (PL) (phosphatidylcholine and phosphatidylethanolamine) of SC eggs compared with NC. Compared with wild fish, both captive groups produced eggs with PLs richer in LA and lower in DHA and AA. Over all fish groups, fertilization success was directly correlated with levels of DHA and AA in the PLs and was inversely correlated with LA levels.

Total lipid content, total lipid fatty acids, lipid classes and their fatty acid compositions were studied in eggs of gilthead sea bream from two different broodstocks. The two broodstock diets used had the same total lipid content, but differed with respect to their fatty acid levels. The lipid class composition of eggs from different origins was not significantly different. The predominant polar lipids were phosphatidylcholine, phosphatidylethanolamine and sphingomyelin, and the main neutral lipid classes were cholesterol esters and triacylglycerols. The fatty acid composition of total lipids and main lipid classes in the two groups of eggs showed marked differences, reflecting the influence of fatty acid levels in the broodstock diets.

This study was conducted to determine the optimum phospholipids: highly unsaturated fatty acids (PL/HUFA) ratio in maturation diets for female Chinese mitten crab, Eriocheir sinensis broodstock. Five isolipidtic and isonitrogenous diets were formulated to contain different amounts of pork lard, fish oil and soy lecithin with resulting PL/HUFA levels (%dry weight) of 1.4/0.3 (Diet 1), 1.6/2.5 (Diet 2), 2.3/2.0 (Diet 3), 3.8/1.5 (Diet 4) and 5.1/0.8 (Diet 5). Diet 1 lacked supplemental fish oil and soy lecithin and served as the control. Each of the five formulated diets was fed to a total of 150 female crabs for a period of 7 months. All diet treatments were triplicated with 50 crabs stocked in a pond as a replicate. After 6 months, male crabs were introduced into the ponds where females were kept, mating, spawning and egg hatching (in seawater) occurred in the following month. The nutritional value of various formulated diets was assessed based on survival, gonadosomatic index (GSI), egg production per female, fecundity (eggs/g female weight) of the females fed those diets and egg and larval (newly hatched zoea I larvae) quality.Females fed Diet 1 had the lowest GSI, egg production per female and fecundity while no significant difference were found for survival, hepatosomatic index (HSI) and the percentage of female spawned among the diet treatments (> 0.05). Although there were no significant differences in egg diameter, egg wet or dry weights among all treatments (> 0.05), the highest and the second highest proportions of essential fatty acids (EFA), i.e. 20:5n-3 (EPA), 22:6n-3 (DHA) and HUFA were found in the eggs produced by the females fed Diet 3 and Diet 2, respectively. Statistical analysis showed that EPA, DHA and HUFA in eggs produced by females fed Diet 3 were significantly higher than those from the other treatments (P < 0.05). Meanwhile, zoea I larvae from crabs fed Diet 3 had significantly larger carapace length than those from the other four treatments (< 0.05). The zoea I larvae from crabs fed Diet 2 and Diet 3 also showed generally better tolerance to starvation and osmotic shock. In conclusion, our results indicated that diet included levels of PL/HUFA around 1.6/2.5 or 2.3/2.0 (% dry weight) fed female E. sinensis broodstock supported increased fecundity and elevated level of HUFA in egg, which in turn resulted in improved overall quality of newly hatched larvae.

Atlantic salmon fry (4 g) were fed for 4 months on semi-synthetic diets containing fatty acid methyl esters of either 18:2 n-6, 18:3 n-3 or a mixture of equal amounts of 20:5 n-3 and 22:6 n-3. The different amounts of polyunsaturated fatty acids added were 0, 0.1, 0.2, 0.5, 1 and 2% (by dry weight). Increasing levels of dietary n-3 fatty acids up to 1% gave faster growth rates in salmon fry, and fish fed the mixture of 20:5 n-3 and 22:6 n-3 seemed to grow faster than fish fed only 18:3 n-3. No significant effect on growth rate was seen when the dietary level of 18:2 n-6 was increased. Dietary inclusions of n-3 fatty acids reduced the mortality of salmon, while dietary 18:2 n-6 had no such beneficial effects.
The dietary treatments caused substantial changes in the fatty acid composition of blood and liver phospholipids (PL), whereas the total lipid fraction of the carcass was less affected. Increasing doses of 18:2 n-6 in the diet resulted in an increased percentage of 20:4 n-6 in liver and blood PLs, while the percentage of 20:3 n-9 decreased. The percentage of 18:2 n-6 also increased in liver, blood and carcass. Dietary 18:3 n-3 resulted in increased percentages of 18:3 n-3 and 20:5 n-3 in liver PLs, while the percentage of 20:3 n-9 decreased. There was, however, no significant increase in the percentage of 22:6 n-3. Dietary 18:3 n-3 produced no significant changes in the composition of blood fatty acids, but increased the percentage of 18:3 n-3 in the carcass. The dietary combination of the n-3 fatty acids 20:5 and 22:6 resulted in an increased percentage of 22:6 n-3 in blood and liver lipids and a decreased percentage of 20:3 n-9, but there were no changes in the percentage of 20:5 n-3.
