Dry mycelium (DM) ofPenicillium chrysogenum and its water extract (DME) were examined for their effects on induced resistance against Verticillium wilt and plant growth of cotton in the greenhouse. Soil application of 0.1–5% DM or 0.5–5% DME provided significant protection against the wilt, relative to the control. As neither DM nor DME inhibited mycelial growth ofVerticillium dahliae in vitro, it is suggested that the disease-controlling effects of DM or DME are attributed to induced resistance. DME (5%), as well as DME treated with chloroform or cold acetone, were as effective as 2% DM in reducing disease severity of Verticillium wilt, implying that the resistance-inducing substance(s) in DM are mostly water-soluble, with neither proteins nor lipids likely to be responsible for the induction of resistance. No significant difference in root colonization withV. dahliae was found between control-inoculated and 2% DM- or 5% DME-inoculated plants. However, colonization of hypocotyls and epicotyls was drastically suppressed by either 2% DM or 5% DME relative to the control. Treatments with 2% DM or 5% DME significantly increased ionically-bound peroxidase (POX) activity in roots, hypocotyls and the second leaf of cotton plants, with the hypocotyls expressing the highest increase. Soil application of DM or DME increased plant height, fresh and dry weight of inoculated and non-inoculated cotton plants, relative to their corresponding controls. It is concluded that DM may be used in cotton crops to promote plant growth and to induce resistance againstV. dahliae. Dry mycelium (DM) of killedPenicillium chrysogenum and its water extract (DME) were used to induce resistance in cotton plants againstFusarium oxysporum f.sp.vasinfectum (Fov). Results showed that the efficacy of either DM or DME in controlling the disease depends on both the concentration and the mode of application. DM amended to the soil at 0.25–2% (w/w) provided 32–75% protection againstFov. Soil drench with 2–5% DME (w/v) and pre-sowing seed soakage with 5–10% DME provided 51–77% and 28–35% protection against the wilt disease, respectively, whereas no protection was obtained with foliar sprays of 1–10% DME. DM and its water extract had no direct antifungal activity on growth ofFov in vitro, suggesting that disease control with DM or DME resulted from the induction of natural defense mechanisms in the cotton plants. Soil drench with 5% DME was as effective as 2% DM powder in inducing resistance againstFov, implying that the resistance-inducing substances were mostly water-soluble. Four cotton cultivars with various genetic resistance levels againstFov were tested at the seedling stage: two resistant ‘Pima’ cultivars and two susceptible ‘Acala’ cultivars. The level of protection achieved in the two susceptible cultivars with DME was equal to, or higher than, that of the two resistant cultivars treated with water. Innate and induced peroxidase activity in cotyledons or hypocotyls and roots coincided with the level of genetic resistance and DME-induced resistance, respectively. Based on our results, an integrated control strategy ofFov with both genetic resistance and induced resistance is suggested.  相似文献

Initial applications of 104 spores g−1 of Pasteuria penetrans, and dried neem cake and leaves at 3 and 2% w:w, respectively, were applied to soil in pots. Juveniles of Meloidogyne javanica were added immediately to the pots (500, 5,000 or 10,000) before planting 6-week-old tomato seedlings. The tomatoes were sampled after 64 days; subsequently a second crop was grown for 59 days and a third crop for 67 days without further applications of P. penetrans and neem. There was significantly less root-galling in the P. penetrans combined with neem cake treatment at the end of the third crop and this treatment also had the greatest effect on the growth of the tomato plants. At the end of the third crop, 30% of the females were infected with P. penetrans in those treatments where spores had been applied at the start of the experiment. The effects of neem leaves and neem cake on the nematode population did not persist through the crop sequences but the potential for combining the amendments with a biological control agent such as P. penetrans is worthy of further evaluation.  相似文献

Experiments were conducted to determine the effectiveness and profitability of the Mi-resistance gene in tomato in suppressing populations of Meloidogyne javanica in a plastic-house with a natural infestation of the nematode. Experiments were also conducted to test for virulence and durability of the resistance. Monika (Mi-gene resistant) and Durinta (susceptible) tomato cultivars were cropped for three consecutive seasons in non-fumigated or in soil fumigated with methyl bromide at 75 g m–2 and at a cost of 2.44 euros m–2. Nematode densities were determined at the beginning and end of each crop. Yield was assessed in eight plants per plot weekly for 6 weeks. The Pf/Pi values were 0.28 and 21.6 after three crops of resistant or susceptible cultivars, respectively. Growth of resistant as opposed to susceptible tomato cultivars in non-fumigated soil increased profits by 30,000 euros ha–1. The resistant Monika in non-fumigated soil yielded similarly (P > 0.05) to the susceptible Durinta in methyl bromide fumigated soil but the resistant tomato provided a benefit of 8800 euros ha–1 over the susceptible one because of the cost of fumigation. Selection for virulence did not occur, although the nematode population subjected to the resistant cultivar for three consecutive seasons produced four times more eggs than the population on the susceptible one. Such a difference was also shown when the resistant cultivar was subjected to high continuous inoculum pressure for 14 weeks. The Mi-resistance gene can be an effective and economic alternative to methyl bromide in plastic-houses infested with root-knot nematodes, but should be used in an integrated management context to preserve its durability and prevent the selection of virulent populations due to variability in isolate reproduction and environmental conditions.  相似文献

Pectinases extract (PE) from the fermentation product ofPenicillium oxalicum BZH-2002 was tested for its ability to induce protection against scab caused byCladosporium cucumerinum on cucumber (Cucumis sativus L.) plants. Seedlings with one true leaf were sprayed with various concentrations of PE (10–200 units ml−1) 3 days before inoculation withC. cucumerinum. Results showed that the induced local protection against the pathogen was dose-dependent when the concentrations of PE were between 20 and 120 units ml−1; systemically induced resistance against the pathogen was not observed. Boiled PE had a slight effect on disease reduction. Commercial pectinases prepared fromAspergillus niger showed lower protection against scab compared with PE when they were used at the same concentration of enzyme activity. No inhibitory activity was observed on conidial germination or germ-tube growth ofC. cucumerinum. PE was further evaluated for its enhancement of defense-related enzymes. Peroxidase (PO), polyphenol oxidase (PPO) and phenylalanine ammonia lyase (PAL) increased in cucumber seedlings after treatment with PE. 1,2-Dehydropyrrolizidine alkaloids (PAs), known to be nematotoxic in vitro, represent a class of secondary plant metabolites from hundreds of plant species worldwide. Pot experiments with the commercially available PA-containing plants Ageratum houstonianum, Borago officinalis, Senecio bicolor, and Symphytum officinalis demonstrate that Meloidogyne hapla is not per se repelled by these plants as all species were infested with nematodes. However, the development of M. hapla juveniles was completely suppressed on A. houstonianum and S. bicolor. Soil in which A. houstonianum and S. bicolor were cultivated and incorporated contained 200–400 times less nematodes than soil treated with Lycopersicon esculentum. Depending on their qualitative composition of PAs at least some of these plants thus appear to be valuable tools for integrated root-knot nematode management.  相似文献

Nematode reproduction on the nematode-susceptible tomato cv. Durinta grafted onto the Mi-resistance gene tomato rootstock SC 6301 was compared to the Mi-resistance gene tomato cv. Monika in a plastic house infested with Meloidogyne javanica. The ungrafted susceptible cv. Durinta was included as a control for reference. Final soil population densities were lower (P ≤ 0.05) on the resistant than susceptible cultivar but intermediate values were recorded on the rootstock SC 6301. The lowest numbers of eggs per gram root were recorded on the resistant cultivar followed by those on the rootstock; in both cases, they were lower (P < 0.05) than on the susceptible control. Cumulative yield (kilogram per square meter) was higher (P < 0.05) on the resistant than susceptible cultivar whether or not it had been grafted. The rootstock SC 6301 provided an intermediate resistance response to M. javanica and was less effective than the resistant cultivar in suppressing nematode populations and plant damage under the experimental conditions of this study.  相似文献

Crude ethanolic seed extracts ofAnnona muricata, A. squamosa (Annonaceae),Lansium domesticum andSandoricum koetjape (Meliaceae) collected from different locations and years in Maluku, Indonesia, were screened for inhibition of larval growth against the polyphagous lepidopteranSpodoptera litura (Noctuidae). Extracts ofA. squamosa were significantly more active (20-fold) than those ofA. muricata. A. squamosa collected from Namlea yielded the extracts with the greatest inhibitory activity. There were significant differences among locations for bothA. squamosa andA. muricata but not forL. domesticum andS. koetjape. Extracts ofA. squamosa, collected from Namlea, inhibited larval growth in a dose-dependent manner, with a dietary EC50 (effective concentration to inhibit growth by 50% relative to controls) of 191.7 ppm fresh weight. Extracts ofA. squamosa collected from individual trees in Namlea also varied in growth inhibitory effect againstS. litura andTrichoplusia ni larvae. The study deals with the tachinid parasitoids ofAncyrosoma leucogrammes (Gmelin) (Heteroptera: Pentatomidae):Clytiomya dupuisi Kugler,Clytiomya sola (Rondani) andGymnosoma clavatum (Rohdendorf) (Diptera: Tachinidae). All species are recorded for the first time as parasitoids ofA. leucogrammes, andC. dupuisi was reared for the first time from a host. The response of four Mi-resistance gene tomato rootstocks to seven populations of Meloidogyne was determined in pot tests conducted in a glasshouse. Rootstocks PG76 (Solanum lycopersicum × Solanum sp.) and Brigeor (S. lycopersicum × S. habrochaites) and resistant cv. Monika (S. lycopersicum) were assessed against one population of M. arenaria, three of M. incognita, and three of M. javanica. Rootstocks Beaufort and Maxifort were assessed against one population of M. arenaria, two of M. incognita and two of M. javanica. Rootstock PG76 was highly resistant (reproduction index <10%) to all the populations, whereas rootstock Brigeor and cv. Monika were highly to moderate resistant. Rootstocks Beaufort and Maxifort showed reduced resistance or inability to suppress nematode reproduction, and their responses varied according to the population tested. Beaufort and Maxifort were susceptible to the two populations of M. javanica as Maxifort was to one of M. incognita. The reproduction index of the nematode was higher (P < 0.05) on Maxifort than Beaufort for all root-knot nematode populations.  相似文献

During a nematode survey, severe infections of tobacco feeder roots and heavy soil infestations byMeloidogyne incognita race 1 were found in S. Miguel (Azores islands, Portugal). This is the first record ofM. incognita infection of tobacco in Azores. Morphology of various life stages, analysis of the esterase electrophoretic pattern and differential host tests were used for nematode characterization and identification. Nematode-induced mature galls were spherical and/or ellipsoidal and usually contained more than one female, males and egg masses with eggs. Feeding sites were characterized by the development of giant cells that contained granular cytoplasm and many hypertrophied nuclei. Giant cell cytoplasm was aggregated along a thickened cell wall. Vascular tissues within galls appeared disorganized. The relationship between the initial nematode population density and growth of tobacco plants was tested in a glasshouse experiment in which inoculum levels varied from 0 to 512 eggs and juveniles (J2) cm−3 of soil. Seinhorst’s model was fitted to height and top fresh weight data of the inoculated and control plants. Tolerance limits with respect to plant height and fresh top weight of tobacco cv. ‘Erzegovina’ plants toM. incognita race 1 were estimated as 1.25 eggs and J2 cm−3 of soil. Eighteen toxins produced byFusarium species were tested at different concentrations onOrobanche ramosa seeds to evaluate their effectiveness in inhibiting germination. Many of the toxins were active at the highest concentration used. Seven of them,viz. fusarenon X, nivalenol, deoxynivalenol, T-2 toxin, HT-2 toxin, diacetoxyscirpenol and neosolaniol, were highly active at 10μM, causing 100% inhibition of germination. Many of them were still active when assayed at a concentration ten times lower, with T-2, HT-2, nivalenol, neosolaniol and diacetoxyscirpenol still able to cause total inhibition; the last mentioned was very active also at 0.1μM, causing more than 90% inhibition. Essential oils extracted fromEucalyptus blakelyi (1,8-cineole, 77.5%),Melaleuca fulgens (1,8-cineole, 56.9%) and 1,8-cineole were shown to have fumigant toxicity against different development stages ofSitophilus oryzae. The eggs ofS. oryzae were the most tolerant, followed by pupae, larvae and adults in that order.M. fulgens oil,E. blakelyi oil and 1,8-cineole at 100 μl per liter of air gave, respectively, LT50 values of 16.2, 17.4 and 9.1 h for adults, 31.1, 19.3 and 17.5 h for larvae, 55.6, 75.2 and 39.7 h for pupae, and required >7 days for eggs. Tachinid parasitoids ofTraumatocampa ispartaensis Doğanlar & Avcı (Lepidoptera, Thaumetopoeidae), which was found to be a new species, were collected from the cedar forests around Isparta-Kapıdağ. The species found wereBlondelia nigripes (Fall.),Compsilura concinnata (Meig.),Pales processioneae (Ratz.),Phryxe caudata (Rond.),Exorista segregata (Rond.) andCarcelia iliaca (Ratz.). The plasmid pUFZ75 conferred constitutive GFP expression on the bacterial pathogen Xanthomonas euvesicatoria (syn. X. campestris pv. vesicatoria). Colonisation of the tomato phyllosphere and invasion of tomato leaves by X. euvesicatoria was examined using both fluorescence and confocal laser scanning microscopy. Xanthomonas euvesicatoria established a limited population on the tomato leaf surface, primarily occupying the depressions between epidermal cells and around the stomata, prior to invasion of the leaf via the stomata and subsequent growth within the substomatal chamber and the leaf apoplast. Additionally, hrp-gfp fusions were used to report on the temporal and spatial expression of hrp genes during epiphytic colonisation and invasion. Xanthomonas euvesicatoria cells carrying hrpG- and hrpX-gfp reporter constructs were not fluorescent in vitro on non-hrp-inducing LB agar but did exhibit a low level of fluorescence on the leaf surface within 24 h of inoculation, particularly in the vicinity of stomata. Cells carrying hrpG- and hrpX-gfp fusions exhibited high levels of fluorescence 72 h after inoculation in the substomatal chamber and the leaf apoplast. Cells carrying the hrpF-gfp fusion were slightly fluorescent on LB agar and showed no further increase in fluorescence on the leaf surface by 24 h after inoculation, but did show a significant increase in fluorescence 72 h after inoculation in the substomatal chamber and apoplast. The apparent low-level induction of the regulators hrpG and hrpX on the tomato leaf surface may suggest that some of the genes of the X. euvesicatoria HrpG/HrpX regulon are up- or down-regulated prior to invasion of the stomata while still on the leaf surface.  相似文献

The clustered hrp genes encoding the type III secretion system in the Japanese strains MAFF301237 and MAFF311018 of Xanthomonas oryzae pv. oryzae were sequenced and compared. The strains differ in their pathogenicity, location, and year of isolation. A 30-kbp sequence comprising 29 open reading frames (ORFs) was identical in its structural arrangement in both strains but differed from X. campestris pv. campestris, X. axonopodis pv. citri, and X. axonopodis pv. glycines in certain genes located between the hpaB-hrpF interspace region. The DNA sequence and the putative amino acid sequence in each ORF was also identical in both X. oryzae pv. oryzae strains as were the PIP boxes and the relative sequences. These facts clearly showed that the structure of the hrp gene cluster in X. oryzae pv. oryzae is unique.  相似文献

Spray mixtures consisting of the plant activator BABA (DL-3-aminobutyric acid) and the protectant fungicide mancozeb were significantly more effective than BABA or mancozeb alone in controlling late blight (Phytophthora infestans) in potato and tomato and downy mildew (Pseudoperonospora cubensis) in cucumber. A mixture composed of 5 parts BABA and 1 part mancozeb (w/w, a.i.) exhibited a higher synergy factor than the 1+1 or the 1+5 (BABA + mancozeb) mixtures. No synergistic interaction was detected between BABA plus mancozeb in controlling sporangial or cystospore germination, nor mycelial growth ofP. infestans in vitro. In 2003–2004, anthracnoses of Enkianthus campanulatus and Rhynchosia acuminatifolia were found for the first time in Kanagawa Prefecture and Tokyo in Japan. These pathogens were identified as Colletotrichum gloeosporioides based on their pathogenicity, morphology and ribosomal DNA spacer sequences. Results were presented at the annual meeting of The Phytopathological Society of Japan in 2004.  相似文献

During storage studies on asparagus spears packed in plastic film and held at 15°C, a Penicillium fungus was associated with subsequent spoilage. On the basis of colony characteristics, the isolate was identified as Penicillium hirsutum Diercks. This report is the first of Penicillium hirsutum causing spoilage of asparagus spears in Japan. Inoculation tests showed that among all the Penicillium species tested in this study only the identified fungus was able to infect asparagus spears.  相似文献

Leaves of tomato and barley were inoculated with conidia of Blumeria graminis f. sp. hordei race 1 (R1) or Oidium neolycopersici (KTP-01) to observe cytological responses in search of resistance to powdery mildew. Both conidia formed appressoria at similar rates on tomato or barley leaves, indicating that no resistance was expressed during the prepenetration stage of these fungi. On R1-inoculated tomato leaves, appressoria penetrated the papillae, but subsequent haustorium formation was inhibited by hypersensitive necrosis in the invaded epidermal cells. On the other hand, KTP-01 (pathogenic to tomato leaves) successfully developed functional haustoria in epidermal cells to elongate secondary hyphae, although the hyphal elongation from some conidia was later suppressed by delayed hypersensitive necrosis in some haustorium-harboring epidermal cells. Thus, the present study indicated that the resistance of tomato to powdery mildew fungi was associated with a hypersensitive response in invaded epidermal cells but not the prevention of fungal penetration through host papilla.  相似文献
