Plasma cholinesterase (pChE) levels and erythrocyte acetylcholinesterase (eAChE) levels were studied in 6 cows before, during and after parturition (Group I), their calves (Group II), 38 cows suffering from parturient paresis (Group III) and 14 newly delivered non-paretic cows (Group IV).The mean of the pChE level in Group I was 1.5 μkat/1 ± 0.20 before parturition and decreased significantly (P ≦ 0.05) to 1.2 ukat/1 ± 0.16 after parturition. The eAChE level was before parturition ≅ 140 ukat/1 and decreased to ≅ 130 μkat/1 4–5 weeks after parturition.At birth the pChE level was 12.8 ukat/1 ± 5.9 in Group II. After 4 weeks the level had decreased to 2.3 ukat/1 ±0.3. In the bull calves the pChE level started to increase when they were 6 weeks old and reached a level of 5.7 μkat/1 ± 0.6 before slaughter at 6 months of age. The heifers did not show this increase. They had a level of around 2 μkat/1 throughout the investigation. The eAChE level at birth was 119 μkat/1 and increased slowly to a level of 145 μkat/1 at 6 months. No differences between the sexes were found.The cows suffering from parturient paresis had a pChE level of 1.80 μkat/1 ± 0.30 before treatment with calcium (Ca). The level decreased significantly (P ≦ 0.001) after Ca-infusion to a level of 1.67 ukat/1 ±0.29. Group IV had a pChE level of 1.65 μkat/1 ± 0.42 at parturition. Two to 4 months later the cows that had recovered from milk fever had a level of 1.61 μkat/1 ± 0.31 and the control cows 1.66 ukat/1 ± 0.48. No differences between the groups were found for the eAChE level.The findings show that parturition influences the pChE level in cows and that sex influences the pChE level in calves between 6 weeks to at least 6 months of age. Furthermore the elevated pChE level found in the cows suffering from parturient paresis before Ca infusion may be a further sign of a disturbance in the cholinergic system with a special preference to the neuromuscular junctions.  相似文献

We studied the relationship of serum prolactin, estradiol-17 beta, and progesterone concentrations to plasma calcium, phosphorus, and free hydroxyproline concentrations, as well as to dry matter intake, in 14 aged dairy cows (mean of 4.5 parities), 7 of which became paretic, from 28 days before to 4 days after calving. Plasma calcium and phosphorus concentrations and dry matter intake decreased more at parturition in paretic cows than in nonparetic cows. Prolactin concentrations were not different between paretic and nonparetic cows, but were variable. Concentrations of estradiol were higher in paretic cows from 15 to 5 days before parturition, whereas hydroxyproline concentration was lower in paretic cows on days 10 through 3 before parturition. Progesterone concentration was lower in paretic cows and decreased earlier at parturition, compared with that in nonparetic cows. The findings suggested that high estradiol concentrations in late pregnancy inhibit bone resorption and predispose aged cows to parturient paresis. The earlier decrease in progesterone concentration at parturition and lower concentrations throughout late pregnancy might have contributed to the greater inappetence in paretic cows at parturition. The importance of prolactin in the pathogenesis of parturient paresis is not clear.  相似文献

Three cows of the Swedish Red and White Breed (SRB) were used in the experiment. Cow no. 1 pregnant for 247 days, was given 20 mg dexamethasone twice with an interval of 48 hrs. Cows nos. 2 and 3, each pregnant for 254 days, received 20 mg of dexamethasone twice with an interval of 24 hrs. The cows delivered normal living calves 153, 138 and 137 hrs., respectively, after the second injection of dexamethasone. Blood samples were drawn from the jugular vein every third hour during the experimental period and the samples were analyzed for estrone, progesterone and 15-keto-13,14-dihydroprostaglandin F2α. Following the dexamethasone injections there was a continuous increase in the blood plasma levels of estrone followed by a sharp decrease in conjunction with parturition. The blood plasma level of progesterone showed a slow but continuous decrease until about 24 hrs. before delivery when a marked drop occurred. The levels of the prostaglandin metabolite increased gradually until about 24 hrs. prior to delivery. This was followed by an abrupt rise, and high levels of the prostaglandin metabolite were recorded for up to four days following parturition. It is concluded that the estrone increase preceded that of the prostaglandin metabolite and that the final drop in the progesterone was synchronous with the final rise of the prostaglandin metabolite level.  相似文献

The prophylactic effect in parturient paresis of an intramuscular injection of 10 million i.u. vitamin D3 one week prior to the expected calving date, was investigated in 84 cows which had previously suffered from parturient paresis. Clinical observations in connection with the development of parturient paresis after injection of vitamin D3 were compared with corresponding observations made on cows within the same population at the previous calving when no vitamin D3 was administered. The incidence of clinical parturient paresis in cows given vitamin D3 was 44.3 per cent. There were no significant differences in the incidence of parturient paresis in cows injected with vitamin D3 during the periods less than 2, 2--4, 5--8 and greater than 8 days before parturition. Average plasma calcium levels (mg/100 ml) before first calcium treatment, results of treatment, and the incidence of retained placenta in vitamin D3 injected cows did not differ significantly from corresponding data for cows with clinical parturient paresis which had not been given vitamin D3 before calving. It is concluded that vitamin D3 does not have any prophylactic effect in parturient paresis in cows in Eastern Norway.  相似文献

Peripheral plasma levels of 15-keto-13,14-dihydro-PGF, progesterone, Cortisol, LH and prolactin were studied in 6 primiparous postpartum dairy cows. The cows were followed by hormone measurements and clinical examinations from parturition until pregnancy was established. Blood was collected 3 times per day. The cervix, uterus and ovaries were examined by rectal palpation at 6–10 days intervals. The cows were observed for signs of oestrus twice daily and were additionally teased with a bull to provoke standing heat.Four cows had a normal parturition and dropped their fetal membranes shortly afterwards. (NR group). The remaining 2 retained their fetal membranes for more than 24 h following parturition (RFM group). One out of 6 cows showed standing oestrus at the first ovulation, 4 animals were in oestrus at the second ovulation and all cows showed signs of oestrus at the third ovulation. Although the length of the first luteal phase varied from 9 to 22 days a corpus luteum was in all cases palpated. The secretion of progesterone during the first luteal phase was terminated by a PGF release.A significant difference in 15-keto-13,14-dihydro-PGF levels between the 2 groups was found on days 0–4 (2.39 vs 6.87 nmol/1 at Ρ < 0.06). Postpartum prostaglandin F release as reflected by the level of 15-keto-13,14-dihydro-PGF lasted shorter in the NR group than in the RFM group (15–17 vs 21 days). Significant positive correlations between 15-keto-13,14-dihydro-PGF and Cortisol as well as between prolactin and Cortisol during the first 24 days postpartum were noted only in cows having normal parturition. The most pronounced daily prolactin variations occurred during the second luteal phase (NR group), when a significant difference between the times 8.00, 12.00 and 15.00 was recorded (14.7, 31.5 and 19.7 μg/l, respectively). Moreover, a partial negative correlation between log value of prolactin and arithmetical value of LH was found in these cows only during the first luteal phase after parturition.  相似文献

The epidemiology of clinical ketosis, hypocalcaemia and hypomagnesaemia was examined. In addition, the genetic variability of ketosis and parturient paresis was investigated. The data set consisted of the lactation records of 70,775 Finnish Ayrshire dairy cows. Each cow was under observation for 2 days before and for 305 days after calving. Lactation incidence rates (%) were: ketosis 6.0, parturient paresis 3.8, non-parturient paresis 0.6, hypomagnesaemic tetany, outdoor 0.6, and indoor 0.2. These diseases formed 22 % of all first treatments by veterinarians during farm visits. 92 % of the cases of ketosis occurred with 8 weeks of parturition, with the highest occurrence 3–5 weeks after calving. Four % of cases of parturient paresis occurred before, and 45 % within 24 h after calving. When cases were categorized by month of calving the risk of ketosis was higher during indoor feeding (October-April) than during outdoor feeding (May-September). The risk of parturient paresis did not significantly vary with month of calving. The occurrence of ketosis increased with parity up to the 4th and decreased thereafter. The occurrence of parturient paresis increased with parity. Both the increase in herd milk yield and the increase in individual milk yields were positively associated with the occurrence of ketosis and parturient paresis. The cows with a history of the reproductive tract infection had a higher risk of contracting ketosis. Heritability estimates for ketosis in various parity groups were from 1.6 % to 4.1 % on the binomial scale (corresponding to 7.3 %–14.4 % on the normal scale), and for parturient paresis from 3.5 to 10.5% (corresponding from 18.3 % to 27.4 %). The genetic correlation between ketosis and parturient paresis, and these and current milk production for all material were insignificant.  相似文献

The aim of the present study was to examine the effect of the somatic cell count (SCC) in milk on reproductive performance, such as pregnancy status in the prepartum period and ovarian function in the postpartum period, in dairy cows. Blood samples were collected every week from one month prepartum to parturition in order to measure the concentrations of 13,14-dihydro-15-keto-PGF (PGFM), estrone sulfate (E1S) and progesterone. Milk samples were collected three times per week in both the prepartum (for one month before the dry period) and postpartum periods (for 3 months immediately after parturition) to measure the SCC. Progesterone was also determined in the whole milk of postpartum cows to define the day of the first ovulation. In the prepartum period, the maximum SCC negatively correlated with the pregnancy period (r = –0.77), but not the calf birth weight. Positive and negative correlations were observed between the average SCC and PGFM or progesterone concentrations in plasma, respectively (r = 0.84 or –0.92, respectively), at 39 weeks of pregnancy. In the postpartum period, a correlation was observed between the day of the first ovulation and both the average and maximum SCC (r = –0.74 and –0.75, respectively), whereas days open was not related to the SCC. These results suggest that a high SCC in the prepartum period may advance parturition by increasing PGF and decreasing progesterone and that the first ovulation in the postpartum period was affected by a high SCC.  相似文献

To assess endocrine and morphological responses of ovaries to total weaning at parturition, 6 Zebu (Bos indicus) cows 5 years or older were investigated. Following parturition, blood samples were collected daily during the first month and twice weekly thereafter until day 60 to determine concentrations of progesterone (P4) and prostaglandin F metabolite.It took between 25 to 32 days to complete uterine involution.The prostaglandin metabolite remained elevated for a mean period of 14.2 days (range, 4-21) postpartum. Five of the animals resumed cyclicity with a short estrous cycle starting between days 7 to 34 and lasting between 7 and 14 days. No estrous behavior was recorded prior to the short estrous cycles, but subsequent normal-length estrous cycles were all preceded by signs of estrus. In the 1 animal that resumed cyclicity with an estrous cycle of normal length on day 37 (length 20 days), the cycle was preceded by estrous behavior.Progesterone concentrations reached a mean maximum of 4.8 nmol liter−1 during the short estrous cycles, and prostaglandin metabolite concentrations peaked while P4 concentrations were decreasing. P4 concentrations reached a mean maximum of 12.2 nmol liter−1 during the estrous cycles of normal length. The interval from parturition to the first estrous cycle of normal length varied between 16 and 48 days, and the length of the cycle was 18 to 22 days.Starting 2 days postpartum, ovaries from 5 of the cows were scanned by ultrasonography every second day until day 30 postpartum. Medium-sized follicles were detected between days 4 to 7 postpartum in 4 of the scanned cows that later had short estrous cycles. The time between parturition and the appearance of the first dominant follicle was 7.6 days (range 6-10 days). The interval between parturition and the appearance of the first ovulatory-sized follicle was 10.2 days (range 8-13 days). In 3 of the scanned cows this ovulatory-sized follicle ovulated.We conclude that cyclic ovarian activity in Zebu cows can start early in the postpartum period in the absence of offspring, and that short luteal phases, not preceded by estrous behavior, may play an important role in establishing normal postpartum ovarian activity.  相似文献

Thirteen primiparous and 41 multiparous Holstein-Friesian cattle were used to study the relationship between maternal plasma progesterone (P4) and estrone sulfate (E1S) concentrations and the prevalence of dystocia. The calvings in 4 heifers and 30 cows were normal (eutocia), while the calvings in 9 heifers and 11 cows were difficult (dystocia). Neither the concentrations of P4 nor E1S were different between the groups with eutocia and dystocia from days 90 to 270 of pregnancy. However, a few days prior to parturition, eutocial cows and heifers showed a sharp decline of plasma P4, while dystocial cattle did not show such a remarkable decline of P4 concentration. Plasma P4 levels in dystocial cows a few days antepartum were significantly higher than in eutocial animals (P<0.05 or P<0.01). Prepartum E1S concentrations were significantly lower (P<0.05) in dystocial than eutocial cattle during the prepartum period from days 6 to 1 in heifers and from days 3 to 1 in cows. These results suggest that insufficient production of E1S and delayed regression of the corpora lutea are possible causes of dystocia in cattle.  相似文献

Progesterone concentrations in heparinized plasma harvested immediately after blood collection were compared with levels obtained after storage of the corresponding whole blood for 2 h, 4 h, 6 h, 1 day, 2 days and 5 days at room temperature and in a refrigerator. The blood was taken during the luteal phase from 4 dogs, 4 horses, 4 pigs and 8 cows. For 4 cows the storage time was extended to 9 and 20 days. No significant effect of whole blood storage time on plasma progesterone concentrations could be shown for dogs or pigs. For the horse a slight but significant decrease was demonstrated when the blood was kept at room temperature. For the cow, however, a dramatic decrease was observed even when the blood was stored in the refrigerator. Following incubation of cow’s blood at room temperature, progesterone levels were close to zero after 1–2 days. By further extending the storage period, a reappearance of assayable progesterone could be elicited. For all species it was found that the storage of plasma at room temperature for 5–9 days did not change the progesterone concentrations.  相似文献

The hypocalcaemia caused by parturition and onset of lactation in high-production dairy cows was mimicked by subjecting cows to starvation periods before and after partus. The changes in plasma calcium, phosphate and magnesium were followed and compared with immunoreactive parathyroid hormone (iPTH) in 2 cows. During the starvation periods before partus, the cows developed hypocalcaemia with no or only small changes in the plasma concentration of magnesium. After the onset of hypocalcaemia, the concentration of iPTH increased on the average 3–4-fold and the raised hormone levels lasted about 24 h after start of refeeding. An increase in plasma phosphate occurred somewhat later than the rise in iPTH and lasted longer. After partus hypocalcaemia developed, together with smaller increases in iPTH concentration (about 2-fold). The post-partum starvation period again resulted in hypocalcaemia and raised iPTH concentrations.In conclusion, starvation and parturition induced inverse changes in plasma calcium and iPTH in dairy cows. The increases in plasma iPTH were reversible and considered secondary to the hypocalcaemia. Through the effect of paratyroid hormone, plasma calcium was normalized and phosphate concentration increased. Therefore, fatal hypocalcaemia which may occur following the course of parturition and onset of lactation is not due to impaired PTH secretion.  相似文献

In this study, we measured plasma concentrations of progesterone, pregnenolone, estradiol, estrone, estrone sulfate, and cortisol and analyzed the correlations between these hormones during gestation in 13 Suffolk ewes, the main breed in Japan. Progesterone increased during gestation and decreased a few days before parturition; however, this pattern was different in samples with high progesterone concentrations (P4 spike samples). This P4 spike was associated with a high pregnenolone concentration. Apart from the P4 spike, the progesterone change was similar to that in other sheep breeds. Pregnenolone increased during gestation and decreased after parturition. A significant correlation between progesterone and pregnenolone was observed a few days before parturition. Estrone sulfate and estradiol concentrations increased during gestation, but estrone did not. They increased shortly before parturition, and then decreased immediately after parturition. At parturition, the correlation between estrone and estrone sulfate was significantly stronger. Moreover, a strong correlation between estrone sulfate and estradiol was observed after parturition. Cortisol did not change during gestation and increased shortly before parturition. The results showed steroid hormone dynamics in normal pregnant Suffolk ewes, which were mostly in line with those of other sheep breeds. It should be noted that high progesterone concentrations altered the typical patterns.  相似文献

The plasma concentrations of progesterone and oestradiol-17p during pregnancy and the first 20 days after parturition were estimated in reindeer. The concentration of progesterone in the period 75–25 days prior to parturition was significantly higher than in the period 200– 75 days prior to parturition (P < 0.001). During the last 25 days before parturition the concentration decreased significantly. The concentration of oestradiol-17β was in most cases below 70 pg/ml until 50 days prior to parturition. During the last 25 days of pregnancy there was a significant increase in oestradiol-17β and the ratio progesterone/ oestradiol-17β was markedly lower than in the period 75–25 days before calving.Key words: reindeer, gestation, plasma, progesterone, oestradiol-17β  相似文献

The effect of prolactin suppression during lactation was studied in 6 sows nursing 5–10 piglets. In order to inhibit prolactin secretion a dopamin agonist (bromocriptine, Parlodel® “Sandoz”) was given per os from about 4 weeks after parturition until weaning 9–17 days later. Plasma prolactin and plasma progesterone were determined by radioimmunoassays. In all animals plasma prolactin was suppressed during the bromocriptine treatment. Two sows resumed ovarian activity during treatment as indicated by elevated plasma progesterone, probably as a result of the low plasma prolactin levels. The results suggest that the elevated plasma prolactin levels during lactation is one of the factors responsible for lactational an oestrus in the sow.  相似文献

Seventeen cows divided into 3 groups were studied: Group 1 consisting of newly delivered cows (1–3 days post partum) suffering from parturient paresis; Group 2 comprising newly delivered (1–4 days post parturn) healthy cows and Group 3 consisting of non-pregnant, non-lactating cattle. All animals were given an intravenous standard Ca infusion, and blood samples were obtained before treatment and at intervals during a 3-hour period thereafter. Plasma levels of calcitonin (CT), calcium (Ca) and inorg. phosphate (P) were determined. No significant difference for the mean CT response existed between animals in Groups 1 and 2. Blood levels of inorg. P increased significantly with time in animals of Groups 1 and 2 but not in animals of Group 3.The finding here of a similar CT response after Ca stimulation in both paretic and non-paretic cows demonstrates that the C-cells in both groups of animals have the same capacity to secrete CT.  相似文献

No significant differences in plasma testosterone level were observed between cows carrying a male foetus and cows carrying a female foetus at any ten-day interval from day 35 of gestation until parturition. Reported higher abortion rates for male than for female foetuses would thus appear not to be due to effects of foetal testosterone on the maternal endocrine balance. In spite of a great individual variation in plasma testosterone values at similar stages of gestation, certain trends are evident. From the 35th to the 80th day of gestation the average concentration was 90–100 pg/ml. Later it rose and reached 200 pg/ml on the 180th day, remaining at this level until after partus. During the first day after parturition plasma testosterone fell significantly and stabilized around 120 pg/ml.  相似文献

The concentration of total iodine in colostrum and normal milk of cows and sows has been determined using a Technicon Autoanalyzer. In cows as well as in sows a lowering of the level of iodine in milk was observed during the first few days after parturition. At the first sampling within 8 hrs. after parturition the concentration of iodine in colostrum of cows from 2 herds was on average 3.4 and 2.4 μg/100 ml, respectively. Corresponding value for colostrum of the sows was 67 μg/100 ml. Based on informations about the composition of the food and daily food consumption it could be estimated that 0.5–1 and 20–45 % of the daily intake of iodine were secreted per 1 milk or colostrum of cows and sows, respectively. It can be concluded that the mammary gland of the sow has a considerably higher ability to concentrate iodine than that of the cow. Furthermore the concentration mechanism is more efficient immediately after parturition than at later stages of lactation.  相似文献

The blood levels of calcium, inorganic phosphorus, magnesium, glucose and NEFA were studied in cows at the time around partus. Eight of 16 cows developed hypocalcaemic paresis. Besides hypocalcaemia the paretic cows showed lower levels of inorganic phosphorus and higher levels of glucose and NEFA than non-paretic cows 24 hrs. post partum. It is known that lipolysis is associated with uptake of calcium in adipose tissue. The calcium content in perirenal adipose tissue was however lower in paretic cows than in non-paretic parturient cows and lactating cows slaughtered 3–5 months after calving. The calcium content in omental adipose tissue was about the same in all 3 groups. Despite increased lipolysis the calcium content in adipose tissue is thus not increased in cows suffering from parturient paresis.  相似文献

The goal of this study was to investigate the efficacy of calcium chloride, sodium phosphate or a combination of these two substances administered orally immediately postpartum for the prevention of parturient paresis in cows. Thirty-two cows that had had parturient paresis at the previous calving, and in which serum biochemistry had shown hypocalcaemia and hypophosphataemia, were used in the study. The cows were transferred to the Department of Farm Animals, University of Zurich, five days before their expected due dates. On a randomized trial, the cows were given calcium chloride, sodium phosphate, both substances or no treatment (controls) via a stomach tube immediately postpartum and 12 hours later. The cows were monitored for 96 hours during which time blood was collected on a regular basis for the determination of total calcium, ionized calcium, inorganic phosphorus and magnesium concentrations. Of the 32 cows treated, 19 (59%) had parturient paresis and 13 (41%) did not. The incidence of parturient paresis did not differ significantly among the groups although there was a tendency for a lower incidence in cows treated with both calcium chloride and sodium phosphate. The various treatments had no apparent effect on serum calcium concentration. The concentration of inorganic phosphorus increased significantly in cows treated with sodium phosphate compared with the controls. The results of this study showed that cows treated with both calcium chloride and sodium phosphate orally tended to have a lower incidence of parturient paresis. Further investigation into multiple administrations of oral calcium chloride and sodium phosphate, started before parturition, for the prevention of parturient paresis is required.  相似文献

The effect of an acid or alkali diet was investigated in 14 mature dairy cows during the last 28 days of pregnancy. The acid diet reduced the incidence of parturient hypocalcaemia compared with the alkali diet and was associated with higher blood ionised calcium and plasma chloride concentrations and lower blood pH and acid-base excess before parturition. Plasma 1,25-dihydroxyvitamin D concentrations before parturition were increased by the acid diet but the concentrations of parathyroid hormone and intact 1–86 parathyroid-hormone-related peptide were unaffected. The estimated fractional calcium absorption and calcium mobilisation rate during an ethylenediamine tetraacetic acid infusion were increased by the acid diet 14 days before parturition. Cortical bone remodelling occurred in all the animals during late pregnancy but was particularly evident in the cows given the acid diet. The data suggest that an acid diet ameliorates parturient hypocalcaemia by enhancing calcium mobilisation before parturition by increasing calcium absorption and bone resorption, these increases possibly being mediated by increases in the plasma concentration of 1,25-dihydroxyvitamin D.  相似文献
