The 16-androstene steroids, one of the principal causes of boar taint, are synthesized in the testis by the andien-beta synthase enzyme system. This system has been shown in vitro to involve both cytochrome P450c17 and cytochrome b5. The objective of this work was to investigate the relationship between the levels of cytochrome b5 in the testis, in vitro steroidogenesis, and the accumulation of 16-androstene steroids in the fat of pubertal boars. We found that the in vitro rate of 16-androstene steroidogenesis in testis microsomes was correlated with 16-androstene steroid concentrations in fat (r = .66, P < .01). Western blots were used to determine the amounts of cytochrome b5 and cytochrome P450c17 protein in testis, and two immunoreactive cytochrome b5 proteins of approximately 12 and 16 kDa were found. Levels of cytochrome P450c17 or the high molecular weight cytochrome b5 in testis were not significantly correlated to levels of 16-androstene steroids in fat. However, levels of total cytochrome b5 immunoreactive protein and levels of the low molecular weight immunoreactive cytochrome b5 were correlated to fat 16-androstene steroid concentrations (r = .59, P < .001; r = .72, P = .0001, respectively). Levels of the low molecular weight immunoreactive cytochrome b5 were also correlated to 16-androstene steroid synthesis rates in vitro (r = .62, P < .05). These results indicate that increased levels of a low molecular weight immunoreactive cytochrome b5 protein, and not of cytochrome P450c17, are related to increased testicular 16-androstene steroid production and accumulation in fat. These results support the hypothesis that selection for reduced levels of this low molecular weight immunoreactive cytochrome b5 protein in the testis may result in decreased levels of 16-androstene steroids in fat and reduced boar taint in uncastrated male pigs.  相似文献

Three studies examined the effects of early postnatal treatment with a GnRH agonist on plasma concentrations of testosterone, dehydroepian-drosterone sulfate, 16-androstene steroids in fat and salivary glands, androstenone in fat and plasma, and testicular development of intact male pigs. The first study involved 45 7-d-old pigs assigned to three treatment groups: 1) boars administered 100 microg/kg of Lupron depot, 2) boars administered 200 microg/kg of Lupron depot, and 3) control boars receiving a saline carrier. The second study involved 20 7-d-old pigs assigned to two treatments: daily injection of 200 microL of 0.5 mg/mL Lupron from d 7 to 35 and controls treated with saline. The third study involved a total of 100 animals assigned to 10 groups of 10 based on their age at slaughter. These groups were subdivided into one of two treatments: 1) boars injected with 200 microL of 0.5 mg/mL of Lupron from d 3 to 35 and 2) control boars injected with saline. Testicular steroid hormone concentrations in plasma decreased (P < 0.01) within 7 d of GnRH agonist treatment. Following cessation of treatment, steroid levels increased to control levels and remained constant until the final rise at 5 mo. Plasma testosterone levels in the 100 microg/kg depot treatment group were higher (P < 0.05) than that of the 200 microg/kg and control group at 164 d of age. There were no differences between treatments (P > 0.05) in testicular steroid hormone levels at the end of study 2 or 3. There were no differences (P > 0.05) in concentrations of 16-androstene steroids in salivary glands between any of the treatment groups at market weight in studies 1 and 2. Fat androstenone levels measured in the third study ranged between 0.6 microg/g and 4.2 microg/g at 7 to 28 d of age. Treatment with GnRH agonist decreased plasma steroid levels and testicular development; however, by d 60 testicular size and weight were at control levels and remained similar until 180 d of age. The results of these studies indicate that daily administration of a GnRH agonist significantly decreased testicular development and steroidogenesis only during treatment, but testis growth and steroidogenesis had returned to control levels by 60 d of age in male pigs. Suppression of the early postnatal rise in testicular steroid hormones did not affect growth performance or steroid hormone levels at 5 to 6 mo of age.  相似文献

The relationship between the metabolism of androsterone and skatole, the major compounds responsible for boar taint, was investigated in F4 Swedish Yorkshire x European Wild Pig intact males. The metabolism of androstenone and skatole were studied in liver microsomes, and the testicular steroid production was measured in testes microsomes. Including androstenone in the assays of skatole metabolism reduced the formation of 6-hydroxyskatole (pro-MII), and three other skatole metabolites (P<.05). The formation of three additional metabolites was not affected. Liver microsomal incubations of androstenone produced two metabolites, I and II. The rate of the formation of metabolite I and the rate of androstenone metabolism were correlated with the rate of skatole metabolism. Liver metabolism of androstenone was not related to levels of androstenone in fat. Testicular synthesis of 16-androstene steroids was correlated with combined synthesis of estrogens and androgens, plasma levels of androstenone, levels of skatole in fat, and skatole metabolism in the liver (P<.05). Plasma levels of estrone sulfate were correlated with levels of skatole in fat and with androstenone levels in fat and plasma and were negatively correlated with synthesis of skatole metabolite F-1 and pro-MII sulfation. These results indicate that the liver metabolism of androstenone and skatole are related. However, it is likely that the relationship between levels of androstenone and skatole in fat is due more to a link between the testicular synthesis of androstenone rather than to the metabolism of androstenone and skatole in the liver. Sex steroids may affect this relationship because of their biosynthesis along with androstenone and possible inhibition of skatole metabolism in the liver.  相似文献

This study describes the age-related variation in boar taint compounds, skatole and androstenone, and testosterone, oestradiol-17 beta (E17 beta), oestrone sulphate (ES), dehydroepiandrosterone sulphate (DHEAS), triiodothyronine (T(3)) and insulin-like growth factor-1 (IGF-1) in six boars. Three pairs of littermates of crossbred entire male pigs (from three Yorkshire x Duroc dams and one Hampshire sire) were included. Blood samples were taken at the age of 9-15 weeks and thereafter at weekly intervals from the age of 20-32 weeks. Plasma concentrations of skatole, androstenone, testosterone, E17 beta, ES, DHEAS, T(3) and IGF-1 were measured. We found that skatole levels in boars increased at the age around puberty after an increase in the levels of testicular steroids. Levels of skatole were not associated with the levels of sex steroids, T(3) and IGF-1. However, the increased level of testicular steroids is probably the underlying factor needed for high skatole levels to occur although the specific mechanism leading to increased skatole levels remains unknown.  相似文献

Concentrations of oestrogens in the blood plasma and seminal plasma of mature boars are high. However, little is known about their concentrations after reaching sexual maturity. The aim of this study was to determine the concentration of oestrogens in blood plasma and seminal plasma of boars during the postpuberal period. Free and conjugated oestrone and oestradiol-17beta were determined by radioimmunoassay in blood from the testicular vein and artery, and peripheral circulation as well as in seminal plasma collected from 18 Polish Landrace boars. The animals were divided into three groups (n = 6) according to age (8, 12 and 16 months, respectively). Oestrone was predominant free and conjugated oestrogen. The highest values of oestrogens were measured in the testicular vein (p < or = 0.05). The concentrations of oestrogens in seminal plasma did not differ from those found in the peripheral circulation. An age-dependent increase in levels of all four oestrogens (p < or = 0.05) was observed. This can be associated with biochemical maturation of the reproductive system during the postpuberal period.  相似文献

Copulation was generally followed by increases in peripheral plasma 5α-androstenone and testosterone levels lasting for periods of about 60 to 100 min. The effect of copulation on the plasma levels of these steroids did, however, vary between boars. In seven out of eight boars the maximum levels of 5α-andro-stenone in the period 60 to 100 min. after copulation were from 114 to 218 % (mean 150 %) of the levels in samples collected before copulation. The corresponding figures for testosterone were from 104 to 283 % (mean 190 %). One boar showed decreasing plasma steroid levels after copulation.The coefficient of correlation between the peripheral plasma levels of 5α-androstenone and testosterone was found to be + 0.61 (n = 2.03).  相似文献

The aim of the present study was to determine the effects of dietary supplements of vitamins on vitamin status, libido, and semen characteristics in young boars under normal and intensive semen collection. Sixty Landrace, Yorkshire, and Duroc boars were allocated randomly from 6 to 10 mo of age to one of the following diets: 1) basal diet (industry level) for minerals and vitamins (Control, n = 15); 2) basal diet supplemented with vitamin C (ASC, n = 15); 3) basal diet supplemented with fat-soluble vitamins (FSV, n = 15); and 4) basal diet supplemented with water-soluble vitamins (WSV, n = 15). After puberty (approximately 12 mo of age), semen was collected at a regular frequency (three times every 2 wk) for 5 wk. Thereafter, all boars were intensively collected (daily during 2 wk). A recovery period (semen collection three times every 2 wk) followed and lasted for 10 wk. Sperm quality (percentage of motile cells and percentage of morphologically normal cells) and quantity (sperm concentration, semen volume, and total sperm number) were recorded as well as direct and hormone related measurements of boar libido. Blood and seminal plasma samples were taken to monitor vitamin status. High concentrations of B6 (P < 0.05) and folic acid (P < 0.05) were observed in the blood plasma of WSV boars, whereas greater concentrations of vitamin E (P < 0.01) were obtained in FSV boars. In the seminal plasma, folic acid concentrations tended to be greater in WSV boars (P < 0.08). During the intensive collection period, there was a tendency (P < 0.06) for semen production to be greater in WSV boars, the effect being less pronounced (P < 0.10) in FSV boars. During the recovery period, the percentage of motile sperm cells was greater in WSV boars (P < 0.03) and, to a lesser extent, in FSV boars (P < 0.10) compared with Control boars. Sperm morphology and libido were not affected by treatments. These results indicate that the transfer of vitamins from blood to seminal plasma is limited and the dietary supplements of water-soluble and fat-soluble vitamins may increase semen production during intensive semen collection.  相似文献

To study porcine melatonin secretion in a stable environment 3 daytime (10.00-15.00) and 3 nighttime (22.00-03.00) plasma samples were collected by jugular venipuncture from 15 gilts, 16 sows, 3 boars and 48 piglets (24 females and 24 males from 8 litters) and analysed for melatonin content. Nighttime melatonin concentrations were higher than daytime melatonin concentrations (p < 0.001), whereas no effect of sampling order could be discerned. The 3 adult Hampshire boars had higher melatonin concentrations during the day and the night, than the 31 adult Yorkshire females (p < 0.05). There was no clear difference between gilts and sows in plasma melatonin. The gilts from one of the litters had higher plasma melatonin concentrations than the gilts in 3 other litters (p < 0.05). Among the 48 piglets, the increase of nocturnal melatonin secretion differed between litters (p < 0.01), whereas the influence of father was not quite significant (p = 0.12). No difference in daytime melatonin concentrations between litters could be found, and there was no difference in melatonin levels between the male and female piglets. In conclusion, this study demonstrates that domestic pigs express a nocturnal increase of melatonin secretion in a standard stable environment. For some animals the amplitude of nighttime melatonin secretion was very low, although always higher than the daytime base levels. Furthermore, the levels of nighttime melatonin secretion differed between litters, which suggests a genetic background.  相似文献

Boar taint in 500 Swedish Landrace and Yorkshire boar carcasses was tested by smelling fat samples heated on the tip of a soldering iron at 150°C. About 40% of the boars were scored as non-tainted; in another 40%, the judges were unsure whether taint was present or not. Boar taint was judged present in only 20% of the boars.The effects of weight and sex on boar taint were studied in approx. 275 boars, barrows and gilts slaughtered at 70, 90, 110 and 130 kg live weight. The number of boars scored as tainted increased with weight, but weight did not affect the level of taint in all boars. When the taste of boar meat and fat was compared to castrates, no differences were found in 80% of comparisons at 70 or 90 kg.The effect of weight on boar taint was studied in 69 boars reared on ad libitum or restricted feed. Biopsies were taken, by a needle biopsy technique, at 70, 90 and 100 kg live weight. The incidence of boar taint did not increase from 70 to 100 kg when boars were on restricted feed, but the weight effect was significant and linear at the ad libitum feeding level. The cause was probably an interaction between physiological development, age and weight of the boars.  相似文献

The objectives of the current experiment were to determine whether boars heterozygous for the mutation in skeletal ryanodine receptors (sRyR), known to cause porcine stress syndrome, differed from wild-type boars in hypothalamic-pituitary-adrenal axis (HPA) function. We have examined basal plasma ACTH, cortisol, and corticosteroid-binding globulin (CBG) concentrations; plasma ACTH and cortisol responses to a nose-snare stressor and at slaughter; dexamethasone suppression of plasma ACTH and cortisol concentrations; and glucocorticoid receptor (GR) density in the pituitary gland, hippocampus, hypothalamus, and frontal cortex. We have also examined carcass yields, composition, and meat quality to determine whether differences in HPA activity were accompanied by an increased incidence of meat quality characteristics associated with pale, soft, exudative (PSE) meat. Thirty boars either heterozygous or wild-type (n = 15 per genotype) for mutated sRyR were tested for HPA function at 7 mo of age. Heterozygous boars had lower basal plasma ACTH (P < .05) and cortisol (P < .04) concentrations. Integrated basal plasma ACTH and cortisol levels were also lower (P < .05 and P < .005, respectively). Genotype had no significant effect on basal CBG, stressor-induced (nose snare or slaughter) or dexamethasone suppression of plasma ACTH or cortisol concentrations. No differences in immunoreactive GR levels were found in the pituitary gland or any brain region examined. We did find a significant, negative correlation (r = -.62, P < .02) between peak (0800) basal plasma ACTH concentrations and hippocampal GR levels. The alterations in basal HPA function in heterozygous boars were accompanied by lighter body weights (P < .03), decreased carcass fat depth (P < .04), and increased carcass lean yields (P < .02). There was a higher incidence of meat quality characteristics associated with PSE meat in heterozygous boars indicated by higher carcass temperatures (P < .04) and meat brightness (P < .0001) with lower carcass pH at slaughter (P < .03) and after chilling (P < .003). In conclusion, we have found differences in basal and not stressor-induced HPA function between boars heterozygous and wild-type for mutated sRyR. This altered basal HPA activity was accompanied by an increased incidence of meat quality aspects associated with PSE meat in heterozygous boars.  相似文献

The objective of this study was to evaluate, in the pre-pubescent intact male pig, the relationship between skatole levels and the activity of hepatic cytochrome P4502E1 (CYP2E1), cytochrome P4502A (CYP2A), aldehyde oxidase (AO), and phenol sulfotransferase 1A1 (SULT1A1). The activity of these enzymes has been positively associated with skatole clearance in mature boars. Twenty-four intact male pigs were weaned at 28 days of age and slaughtered 2 weeks postweaning, at which time caecal contents, blood, fat, and liver samples were collected. Caecal contents and fat were analyzed for skatole concentrations, and plasma was analyzed for skatole and steroid hormone (testosterone (T), dehydroepiandrosterone (DHEAS), estrone sulphate (E1S)) concentrations. CYP2A, CYP2E1, and AO, as well as SULT1A1 activities were evaluated in liver samples. Stepwise regression was utilized considering plasma or fat skatole concentration as the dependent variables and hormone concentrations and enzyme activities as independent variables. The activities of the enzymes CYP2A, CYP2E1, and AO and concentrations of the hormones T, DHEAS, or E1S were not correlated with concentrations of skatole in plasma or fat. However, SULT1A1 activity was negatively correlated with plasma (r = − 0.70, P < 0.05) and backfat (r = − 0.41, P < 0.05) skatole concentrations. Furthermore, this correlation was improved in plasma (r = − 0.88, P < 0.05) and fat (r = − 0.63, P < 0.05) when the concentrations of skatole in caecal contents was included as an independent variable in the multiple regression analysis, demonstrating the importance of measuring skatole production in these studies. T, DHEAS, and E1S concentrations in plasma were not correlated with the activity of any of the enzymes evaluated. This study suggests that SULT1A1 is important in the metabolism of skatole in pre-pubescent pigs and the overall metabolism of skatole in the pre-pubescent pig differs from that in the mature boar.  相似文献

A comparative study was carried out on the chemical composition of plasma from the cauda epididymidis, semen fractions, and whole semen of boars. A total of 22 boars were used in this study. The boars, which ranged in age from 8 to 14 months, were of Swedish Landrace and Swedish Yorkshire breed. All boars used presented a normal semen picture. A dummy sow and an artificial vagina were employed for semen collection. The semen was collected as whole semen and as semen fractions in 10 nil volumes. The contents of the cauda epididymidis was removed post mortem.The following parameters were investigated: sperm concentration, dry weight of spermatozoa and of seminal plasma, osmotic pressure, sodium, potassium, chloride, inorganic phosphorus, calcium, magnesium, total protein, GOT, GPT, and alkaline phosphatase in seminal plasma. Paper electrophoresis was carried out on seminal plasma. Tlxe results of the analysis are summarized in Tables 1–6.The sperm concentration was approximately 3.2 mill./mm3 in the cauda epididymidis, 1 mill./mm3 in the sperm-richest fraction (II) and 0.25 mill./mm3 in whole semen. The dry weight (expressed in per cent dry matter) of spermatozoa was highest in the cauda epididymidis (25.47 %), showing a tendency to decreasing in semen fractions I—IV and was lowest in whole semen (15.29 %). The per cent dry weight in plasma was higher in the cauda epididymidis (4.56 %) than in semen fraction I (2.20 %). In semen fractions I—IV the per cent dry weight rose from 2.20 (U to 4.51 % and reached the level of approximately 3.80 % in the sperm-free fractions V—VII. The osmotic pressure was significantly higher in the cauda epidi-dymal plasma than in the whole seminal plasma or the seminal plasma fractions. The same phenomenon was observed in a boar where the cauda epididymal content was collected in vivo from a patent established fistula. There appears to be a connection between the per cent dry weight of spermatozoa and the osmotic pressure, which means that the per cent dry weight of the cauda epididymal spermatozoa decreases when mixed with the accessory gland secretions, which have a lower osmotic pressure. The fall in per cent dry weights is thought to be caused by an intake of water.The amount of sodium, chloride and magnesium was higher in ejaculated seminal plasma than in cauda epididymal plasma. The reverse was true for inorganic phosphorus and potassium. Moreover the sperm-free fractions contained more sodium, chlorides and magnesium than the sperm-containing fractions, while the concentration of potassium and inorganic phosphorus was comparatively higher in the sperm-containing fractions. A connection is apparent between sperm concentration and the potassium, inorganic phosphorus and magnesium levels. Statistical analysis of the values of chloride and magnesium revealed significant differences between individual boars for most of the semen fractions.The concentration of plasma proteins in the cauda epididymidis was approximately the same as in whole semen and in the semen fractions except for fraction I, which contained a relatively low concentration. As regards total protein there were significant differences between individual boars in most of the semen fractions as well.The paper electrophoretic pattern of epididymal plasma was different from that of semen plasma. Thus there were three or four distinct components in the cauda epididymidis numbered 1, 2, 3, and 4, and three distinct components in whole seminal plasma numbered 3, 4, and 5, while the sperm-richest semen fractions contained four components (2, 3, 4, and 5) and the others three components, namely 3, 4, and 5.The level of GOT was high in the cautlu cpiflidymill contents (99.1 i. u./ml) compared with that for whole seminal plasma (99.1 i.u/ml). In semen fractions there was a clear positive correlation between the level of GOT and the sperm concentration. The GPT concentration wis as a whole low and. in contrast to GOT. somewhat higher in the sperm-free fractions than in the sperm-containing fractions. The concentration of alkaline phosphatase was very high in cauda epididymal plasma (31,463 i. u./ml) as well as in the sperm-rich fractions (e.g. 7,096 i. u./ml in fraction II). Preliminary investigation has moreover revealed a very low alkaline phosphatase concentration in seminal plasma of vasectomized boars, which condition suggests thai the main origin for alkaline phosphatase in boars is the testis and epididymis.  相似文献

Large variations in the level of 5α-androstenone in fat from different boars have been found. No significant difference in the level of 5α-androstenone was detected in fat from boars selected for high rate of gain and low back fat (HP-line), low rate of gain and high degree of fatness (LP-line) and a control group maintained without deliberate selection (GL-line). In boars used in artificial insemination service relatively high levels of 5α-androstenone in fat were observed, and in these animals a significant (P < 0.05) positive regression of 5α-androstenone level on age was found. Positive but non-significant regression coefficients were found between number of services which the boars had performed and level of 5α-androstenone in fat from the same animals.  相似文献

Fasting plasma hormones and metabolites in feral and domestic newborn pigs   总被引:1,自引:0,他引:1  
Newborn Yorkshire and Ossabaw (feral) pigs were examined under thermoneutral conditions to determine whether survival rate during fasting differs between these breeds and whether any blood-borne factors are associated with improved survival. Newborn pigs were removed from the sow before suckling. Body composition was determined on 10 newborn Ossabaw and 12 newborn Yorkshire pigs. Another group of animals (eight Ossabaw, 12 Yorkshire) was fasted for 72 hr, with blood samples drawn at birth and 12 and 24 hr into fasting. Glucose, free fatty acid (FFA), growth hormone (GH), insulin, thyroxine (T4), triiodothyronine (T3), cortisol and glucagon concentrations were measured in plasma of fasted pigs. Concentrations of carcass lipid, dry matter and ash were higher in newborn Ossabaw pigs than in newborn Yorkshire pigs. Survival through 72 hr of fasting was lower among Yorkshire pigs. Yorkshire and Ossabaw pigs had similar concentrations of metabolites and hormones at birth, with the exceptions of lower plasma GH and higher T3 concentrations in Ossabaw pigs. Higher plasma T3 concentrations would indicate a greater potential for fatty acid oxidation. During fasting, Ossabaw pigs had lower plasma GH and T4 concentrations and higher glucagon and FFA concentrations. Increased survival among newborn Ossabaw pigs may have been due to increased availability of FFA during fasting, and to a greater potential for gluconeogenesis through increased oxidation of fatty acids and higher plasma glucagon concentrations. This would suggest that maternal treatments that would increase storage of fat and(or) increase the capacity for oxidation of fat in utero would improve survival of newborn pigs.  相似文献

在不同季节采集56头大白、长白、杜洛克公猪的精液,对精液品质进行了评定和分析。结果表明:在一年内,杜洛克和大白公猪的精液品质均以秋季最好,春季次之,夏季最差,而长白公猪的精液品质以春季最好、夏季最差;精液量有显著(P<0.05)的品种间差异,长白猪、大白猪、杜洛克猪依次降低;精子活率表现为杜洛克猪、大白猪、长白猪依次降低但差异不显著(P>0.05);精子畸形率以长白猪为最高,而在各品种间差异也不显著(P>0.05)。经综合评定,在受试的3个品种公猪中,杜洛克猪的精液品质较好,而且在夏季也优于其他2个猪种。  相似文献

Three normal, sexually mature young boars of Swedish Yorkshire breed were treated with daily intramuscular injections of diethylstilboestrol and/or oestradiolbenzoate for a total period of 13–15 weeks. Whole ejaculates were collected on a dummy sow with an artificial vagina twice a week. The ejaculate was examined as regards semen volume, sperm concentration, sperm motility and sperm morphology. The seminal plasma was analysed for Na, K, Cl, Mg, inorganic phosphate, total protein and fructose. Testicles, epididymides and the accessory sex glands were obtained at slaughter. Material from epididymal segments A, B, Db, Fa and Fb was collected and examined for sperm morphology. Spermatocrit, osmotic pressure and plasma concentrations of Na, K, Gl, total protein and GPC were assessed in material from epididymal segment Fa. The sex glands and the accessory sex glands were examined maeroscopically and microscopically.The semen volume, the sperm concentration and consequently the total sperm count per ejaculate showed a gradual increase during the course of the investigation. This is to be expected as the boars used were comparatively young at the beginning of the experiment. These values were all within normal limits for adolescent boars.Taking all the characteristics examined into consideration no conclusive evidence was found that an exogenic administration of oestrogens to intact boars has any influence on the function of the epididymis and the accessory sex glands.  相似文献

This study evaluated the effect of supplement of raw potato starch (RPS) on the levels of skatole, androstenone, testosterone and oestrone sulphate in plasma from entire male pigs. The study also evaluated relationships between plasma levels of skatole and testicular steroids at three different live weights (LW) of approximately 90, 100 and 115 kg. A total of 111 entire male pigs of a crossbred (Yorkshire dams×Swedish Landrace sires) were used. Animals were raised either in mixed pens, with females and males, or single-sex pens. Each pen contained seven or nine pigs. The most fast-growing three pigs from the pens with nine pigs were slaughtered when they reached 90 kg LW, and the remaining pigs were slaughtered at 115 kg LW. All pigs were fed the same commercial diet until the average pen weight reached 100 kg. Then, 33 out of 80 remaining pigs received RPS, 0.6 kg per pig and day, for 2 weeks prior to slaughter. Blood samples were taken from the pigs at three occasions: first, the day prior to first slaughter occasion, low-weight group; second, the day prior to change in diet, middle-weight group; and third, the day prior to second slaughter occasion, high-weight group. Plasma was analysed for the levels of skatole, androstenone, testosterone and oestrone sulphate. Fat samples were taken at slaughter and analysed for the levels of skatole and androstenone. The levels of skatole and testicular steroids in plasma were significantly higher in entire male pigs from the high-weight group fed no RPS compared to those from low- and middle-weight groups. The levels of the investigated compounds did not differ between low- and middle-weight groups (P>0.1). The diet with RPS induced a decline in skatole levels in plasma and fat (P<0.001), but not plasma levels of testicular steroids and fat levels of androstenone (P>0.05). Skatole levels were positively correlated to testosterone and oestrone sulphate levels in the middle- and high-weight pigs fed no RPS as well as to testosterone in the low-weight group. In the high-weight group fed RPS, skatole levels were not correlated to any of the analysed compounds. Approximately 26% of the entire male pigs (11 out of 43) from the high-weight group fed no RPS produced skatole levels in fat above 0.20 μg/g, whereas the pigs from the low- and high-weight group fed RPS did not produce skatole levels above 0.20 μg/g in fat. Androstenone levels in fat were high in all groups. In total 47% (52 out of 111) pigs expressed androstenone levels above the rejection levels in fat of 1.0 μg/g and 88% (98 out of 111) had androstenone levels above 0.5 μg/g. It was concluded that a lower slaughter weight and the supplement of raw potato starch to the diet could be used to reduce skatole levels in entire male pigs. Androstenone levels in fat, however, could not be reduced by either a lower weight at slaughter or dietary manipulation.  相似文献

为探讨不同配种方式对二元母猪产仔数的影响,以杜洛克猪、长白猪、大白猪、皮特兰猪四个品种公猪精液采用人工授精的方法对11654头长大二元母猪配种,对所产11654窝总产仔数、窝产活仔数、窝损失等进行统计和分析,分析不同配种模式对二元母猪繁殖性能的影响。结果表明,配种次数、不同品种公猪组合配种对二元母猪总产仔数、产活仔数以及窝损失均无显著影响(P〉0.05),但杜洛克公猪所配二元母猪的总产仔数和窝产仔数均高于大白和长白猪(P〈0.05);采用长×杜×大组合配种的二元母猪总产仔数和窝产活仔数均高于其它三种组合。试验结果表明,在生产中,可在长白、大白、杜洛克猪中任选两个品种,通过二次配种的方式提高母猪的产仔数。  相似文献

Relationship between free oestrone and boar taint compounds in adipose tissue were studied in two groups of entire male pigs of different breeds. Group A consisted of 33 entire crossbred male pigs (dam Yorkshire and sire backcross Yorkshire x Wild Boar, generation seven). Group B consisted of 194 entire male pigs of crossbreeds between Swedish Hampshire (H) and Finnish Landrace (L), LH x H, H x LH, LH x LH (dam x sire). The measurements of free oestrone in adipose tissue were performed with a new method developed and validated in our laboratories. The standard curve was linear for concentrations of free oestrone ranging from 0.13 to 5.10 ng/g. The method exhibited parallelism of results between serial dilutions and a mean recovery of 97 +/- 13.7%. Intra-assay variations for samples with concentrations of free oestrone from 0.67 to 2.08 ng/g were from 9.23 to 11.94%. Inter-assay variations for the samples with concentrations of free oestrone from 0.89 to 2.96 ng/g were from 3.78 to 10.11%. The levels of free oestrone in fat from group A were well correlated with fat levels of androstenone (r = 0.66; p < 0.001) and levels of oestrone sulphate in peripheral plasma collected at the same time as the fat (r = 0.74, p < 0.001). The levels of free oestrone in fat from group B were significantly correlated to fat levels of androstenone (r = 0.68, p < 0.001) and skatole (r = 0.29, p < 0.001). In group B, age-related differences in fat levels of free oestrone, androstenone and skatole were studied. Free oestrone and skatole levels increased simultaneously at the age of 22 week (p < 0.05 for both), and androstenone levels increased at the age of 26 week (p < 0.05). It was suggested that the levels of free oestrone in adipose tissue might be used for the evaluation of hormonal status of male pigs as an alternative to plasma levels of testicular hormones. The levels of free oestrone might be involved in the regulation of skatole levels in fat as indicated by both the simultaneous increases in skatole and free oestrone levels in fat and positive correlation between skatole and free oestrone.  相似文献

An excessive accumulation of androstenone in pig adipose tissue is a major contributor to the phenomenon of boar taint. Androstenone deposition is dependent on the rate of androstenone biosynthesis in testis and androstenone degradation in liver. The aim of the current study was to examine the possibility of the existence of breed-specific mechanisms controlling androstenone accumulation in pig adipose tissue. The specific objective was to investigate the expression of some of the key enzymes involved in testicular and hepatic androstenone metabolism in pigs of 2 breeds by using animals with high and low androstenone concentrations within each breed. The study was conducted with Norwegian Landrace (N. Landrace) and Duroc boars. The mean androstenone values for the low- and high-androstenone groups were 0.1 +/- 0.01 microg/g and 7.58 +/- 0.68 microg/g for N. Landrace boars, and 0.22 +/- 0.04 microg/g and 13.55 +/- 1.14 microg/g for Duroc boars. The enzymes investigated were 3beta-hydroxysteroid dehydrogenase (3beta-HSD), cytochrome P450-c17, and sulfotransferase 2B1 (SULT2B1). Expression of cytochrome P450-c17 in liver and testis did not differ between animals with high and low androstenone concentrations in either the N. Landrace or Duroc breed. Expression of hepatic 3beta-HSD, which catalyzes the first stage of androstenone degradation, was decreased in high-androstenone N. Landrace boars (P < 0.01), but not in high-androstenone Duroc boars. In contrast, the expression of hepatic SULT2B1, which catalyzes the second stage of steroid catabolism, was decreased in high-androstenone Duroc animals (P < 0.05), but not in high-androstenone N. Landrace animals. Sulfotransferase 2B1 was also inhibited in testis of high-androstenone pigs of both breeds compared with low-androstenone animals. We report breed differences in expression of the androstenone-metabolizing enzymes 3beta-HSD and SULT2B1 in the liver of high- and low-androstenone pigs. It is suggested that accumulation of androstenone in adipose tissue of N. Landrace boars might be related to a low rate of hepatic androstenone degradation in metabolic stage I, whereas the high androstenone concentration in Duroc boars might be related to a low rate of androstenone metabolism in metabolic stage II.  相似文献
