Ribavirin treatment of cultured potato shoot tips was tested as a means of eradicating potato virus X (PVX) from two potato cultivars. Shoot tips were cultured on liquid medium containing 0, 1, 10 or 100μg/ml ribavirin. Cultures were evaluated periodically for viability, and scored for vigor on a relative growth scale. Developed plantlets were assayed for PVX by transmission tests toGomphrena globosa. Ribavirin treatment was phytotoxic at all tested concentrations, and lethal to all cultivars treated at 100 μg/ml. Treatment also delayed plantlet development by up to 2 months at 1 and 10 μg/ml as compared with nontreated controls. PVX assays indicated that 80 and 83% of the plantlets were free of PVX following treatment with 10 μg/ml for cultivars Russet Burbank and Red McClure, respectively. Five and 6% of the plantlets developed from the 1 μg/ml treatment were PVX-free, whereas 0 and 2% of the controls were PVX-free for the same cultivars. Six to 8 months were required to develop plants from shoot-tip cultures treated with 10 μg/ml ribavirin.  相似文献

A refuge of conventional potato plants adjacent to Colorado potato beetle (CPB),Leptinotarsa decemlineata (Say)-resistant,Bt-potato (transgenic) plants may reduce opportunity for the development of tolerance to the resistant plants. The refuge strategy was developed on the basis of data available for CPB recruitment in conventional potato fields. This study was undertaken to provide information on CPB recruitment inBt-transgenic potato fields. A marking experiment was conducted over the 2000 and 2001 crop seasons to determine the relative contributions of beetle populations from fields 10 m, 175 to 300 m, and 1200 to 1280 m distant to the pattern of CPB recruitment (immigration) in aBt-potato field. Season-long CPB recruitment in theBt-potato field decreased with source distance in a manner similar to that previously reported in conventional potato fields. Although marked beetles from the 10-m field plot contributed more to the recruitment than the more distant fields, they contributed only 3.3% and 6.6% of the total beetle sightings in theBt-potato field. Therefore, results suggest that a larger acreage of conventional potato fields at some distance from the resistant crop could replace the adjacent designated refuge. Seasonally, beetles from the overwintering sites provided the first recruits to the resistant field. As their contribution declined, the overwintered beetles from the refuge were recruited until the beginning of the summer population. These results suggest that locating the resistant fields close to active overwintering sites will improve the probability that the refuge strategy will be effective by ensuring the presence of recruits at the very beginning of the crop season. The substantially lower recruitment level obtained for summer than for overwintered CPB in resistant and conventional potato fields highlights the need to reconsider the applicability of the refuge strategy for the summer population.  相似文献

Reinfection of potato seed stocks with the potato viruses S (PVS) and X (PVX) varied with cultivar, virus, and grower. Rapid recontamination was observed for the cultivars Norgold Russet and Ontario with PVS and for the cultivars LaChipper, Norchip, and Norgold Russet with PVX. Recontamination was low for the cultivars LaChipper and Monona with PVS and for the cultivars Kennebec, Monona, Norland, and Superior with PVX. Survey results suggest that PVS and/or PVX can be eliminated from cultivars which appear to possess field resistance to infection, but that further evaluation of cultivars which are very susceptible to reinfection will be necessary.  相似文献

The ultrastructure of Katahdin tubers infected with potato virus X (PVX) is compared with PVX-free tubers by electron microscopy. Electron-dense globules surrounding the inner periphery of the tonoplast were observed in PVX-infected tubers, while PVX-free tubers did not show such bodies. The systemic insecticides, Dasanit®, carbofuran (Furadan®), phorate (Thimet®), aldicarb (Temik®) and disulfoton (Di-Syston®) were applied in the planting furrow at 2–3 lb actual/acre for potato tuberworm (Phthorimaea operculella (Zeller)) control in Irish potato foliage. None of these compounds gave entirely satisfactory results. Dasanit was the best with slightly over 80% decrease in the number of mines and was followed closely by carbofuran; all treatments needed to be supplemented by foliage sprays for satisfactory tuberworm protection. Azinphosmethyl (Guthion®) gave consistently outstanding results as a foliage spray at 0.5 lb/acre with over 99% tuberworm reduction. Other foliar-applied insecticides giving at least 98% tuberworm control included: Monitor®, monocrotophos (Azodrin®), phosphamidon, Orthene®, Torak®, methomyl (Lannate®) and Dursban® (all at 0.5 lb/acre) and Phosvel® (1 lb/acre).  相似文献

Summary Segregation for extreme resistance to PVY was evaluated in progenies derived from crossing two extremely resistant potato clones with parents differing in resistance. Resistance was evaluated after mechanical inoculation with PVYO and PVYN, and after graft inoculation with PVYO. Biological and serological tests (ELISA) were used for virus detection. The extreme resistance is governed by a single dominant gene, but observed segregations deviated from the expected ratios. Considerable modifying effects were detectable, depending on the potato genotype and virus isolate, for a significant excess of susceptible genotypes was observed in some progenies. Moreover, genotypes with non-parental types of resistance to PVY were observed.  相似文献
