试验结果表明:A组与B组差异不显著(P〉0.05);A组、B组与C组差异显著(P〈0.05)。

Background: In farm animal practice, determination of blood glucose concentration under field conditions is often necessary. Objective: As there is no portable glucose meter device developed for use in farm animals, the analytical accuracy of a portable glucometer designed for people was evaluated for its use in cattle and sheep. Methods: Blood samples from 90 cattle and 101 sheep were used in the study. Glucose concentration was determined in whole blood immediately after blood collection from the jugular vein with the One Touch Vita portable glucometer and in serum with an enzymatic colorimetric method. The agreement between methods was assessed by Passing and Bablok regression analysis. The precision and the accuracy of the measurements were determined using the concordance correlation coefficient. Results: There was a strong linear relationship between the glucose values obtained using the portable glucometer and those obtained by the bench method in both cattle and sheep. Precision was 95% for cattle and 88% for sheep, whereas accuracy was 92% and 99%, respectively. The mean glucose values obtained using the portable glucometer were significantly lower by 8.3% in cattle and 3.2% in sheep than those determined by the bench method. Conclusion: The One Touch Vita portable glucometer can be used in clinical practice to determine blood glucose concentrations in cattle and sheep, but reference intervals (RI) must be corrected to allow for negative bias. Based on these equations the RI for blood glucose in cattle and sheep using the portable glucometer were corrected to 1.84–4.17 and 2.41–4.35 mmol/L, respectively.

Background

The somatic cell count (SCC) is commonly used to monitor udder health and diagnose subclinical intramammary infection (IMI) in dairy cattle.

Hypothesis

The Somaticell test (ST) 2 and California mastitis test (CMT) are clinically useful cow‐side tests for diagnosing subclinical IMI.

Animals

One hundred and eleven dairy cows at dry‐off and 92 cows within 4–7 days postcalving.

Methods

Quarter foremilk samples were obtained and analyzed with a DeLaval cell counter (DCC, reference method), 1 ST, and CMT. The ST was run in a simulated cow‐side manner using milk at 37°C instead of 0–8°C as recommended by the manufacturer. Test performance for diagnosing IMI (DCC SCC >200,000 cells/mL) was evaluated by calculating the area under the receiver operating characteristic curve (AUC) and the kappa coefficient (κ) at the optimal cut‐point for each test. The effect of milk/reagent temperature also was evaluated.

Results

Compared to the reference method, the ST run in a simulated cow‐side manner had an AUC = 0.68 and κ = 0.24 at dry‐off, and AUC = 0.74 and κ = 0.40 in fresh cows. The CMT performed much better than the ST in diagnosing subclinical IMI with AUC = 0.88 and κ = 0.77 at dry‐off, and AUC = 0.87 and κ = 0.76 in fresh cows. The measured ST value decreased with increasing temperature of the milk/reagent mixture.

Conclusions/Clinical Importance

The ST is optimized for use on milk at 0–8°C and is therefore designed for on‐farm use on refrigerated milk samples. The ST is not suited for use as a cow‐side screening test for IMI because the milk temperature exceeds the recommended range for the test.

Background: Success in management of diabetes mellitus (DM) is defined as improvement of blood glucose concentrations and clinical signs. However, the psychological and social impact of DM and its daily treatment regimen on quality of life (QoL) of both animal and owner is uncertain. Hypothesis/Objectives: To design, validate, and apply a diabetic pet and owner‐centered, individualized measure of impact of DM (DIAQoL‐pet). Animals/Subjects: Two hundred and twenty‐one owners of insulin‐treated diabetic cats were recruited to complete the DIAQoL‐pet. Methods: Discussions and pilot surveys with clinicians and owners of diabetic cats led to the design of 29 specific DM‐associated QoL questions. Owners of diabetic cats completed the finalized survey. Each item was scored according to impact frequency and perceived importance. An item‐weighted impact score (IWIS) for each item was calculated, as was an average‐weighted impact score (AWIS) by averaging all IWISs. Principal component analysis and Cronbach's α calculation assessed the measure's reliability. Two overview questions measured overall QoL and diabetes‐dependent QoL. Results: The DIAQoL‐pet showed high reliability (Cronbach α 0.83). The AWIS was ?1.76 ± 2.4 (mean ± SD). Areas reported as most negatively impacting QoL included: “boarding difficulties” (IWIS ± SD: ?4.67 ± 5.3), “owner wanting more control” (?4.34 ± 4.7), “difficulties leaving cat with friends or family” (?4.21 ± 4.7), “worry” (?4.10 ± 3.9), “worry hypo” (?3.67 ± 3.5), “social life” (?3.48 ± 3.9), “costs” (?3.04 ± 3.8), and “work life” (?3.03 ± 3.7). Forty‐one percent of owners believed their cat's life would be “a little better” without DM. Conclusions and Clinical Importance: The DIAQoL‐pet proved robust and identified specific areas most negatively impacting on diabetic cats and their owners' QoL. This tool warrants further investigation for use in clinical or research settings.
