Induction of ovulation for timed artificial insemination (TAI) with the Ovsynch protocol was evaluated in 49 anoestrous and lactating Bos taurus x Bos indicus cows. Palpation per rectum and transrectal ultrasonography were used on Days -30, -20, -10 and 0 (start of treatment) to confirm anoestrus but with the presence of follicles > or = 10 mm, and every other day during treatment to determine ovarian activity. Cows were randomly assigned to: (1) Ovsynch (n = 24; Day 0, 200 microg GnRH; Day 7, 150 microg PGF2alpha; Day 9, 200 microg GnRH + TAI 16 to 20 h later) and (2) control (n = 25; no treatment). Rates of ovulation for the first GnRH injection, detection of a corpus luteum (CL) at PGF2alpha injection, pregnancy and induction of cyclicity were greater (P < 0.05) with Ovsynch. There was no effect of body condition score (P > 0.05). In conclusion, the Ovsynch protocol was not effective in obtaining acceptable pregnancy rate for TAI, but it was effective for induction of cyclicity in anoestrous and lactating Bos taurus x Bos indicus cows under tropical conditions.  相似文献

In this double-blind Study 72 dairy cows were treated with injections of Luprostiol (Reprodin). 32 cows served as control. Gynaecological examinations in 10-day intervals up to day 40 post partum and determination of progesterone levels in milk samples taken in 3-day intervals up to day 73 p. p. were performed. The administration of PGF2 alpha did not improve the early onset of ovarian activity in cows with uncomplicated or complicated delivery. Ovaries without detectable function up to day 73 p. p. only occurred in the placebo group, representing a significant (p < 0.05) difference to the control group. In cows after uncomplicated as well as in animals after complicated delivery involution of the uterus up to day 20 p. p. progressed faster in the verum group compared to the control group. Cervical diameters were highest in the placebo group up to day 20 p. p. Until day 40 p. p. the cervical diameters of the probands equaled those of the control animals. In contrary to cows after uncomplicated delivery the injection of PGF2 alpha after complicated parturition lead to a significant (p < 0.05) reduction of puerperal endometritis. Neither in cows with uncomplicated nor complicated delivery the administration of PGF2 alpha resulted in an improved conception rate. There were no significant positive effects on fertility parameters to be detected in general. Partial results observed were not dose dependent.  相似文献

Twenty-four beef cows were infused continuously for 11 d (d 2 to 13 after parturition) into the descending aorta with either prostaglandin F2 alpha-Tham salt (PGF2 alpha; 33.5 mg/d; n = 12) or with .9% NaCl (saline; n = 12) vehicle. Cows from each infusion treatment then were assigned to three slaughter groups (G). Cows in G1 and G2 were suckled until slaughter on d 15 and 35, respectively, whereas calves in G3 were weaned on d 31 and cows were slaughtered on d 35 after parturition. Nonatretic and atretic (greater than 4 pycnosis) antral follicles (greater than .15 mm) on ovaries ipsilateral to the previously gravid uterine horn were evaluated by histological techniques. Compared with saline infusion, PGF2 alpha increased mean diameters of the largest (F1) follicles on the ipsilateral (d 15 and 35; P less than .07) and contralateral (d 15; P less than .07) ovaries (surface evaluation) as well as mean diameters of the largest (F1, d 35; P less than .002), second- and third-largest (F2, F3 d 15 and 35; P less than .01) nonatretic follicles (histologic evaluation) in suckled cows. Compared with suckled cows slaughtered on d 35, weaning on d 31 increased the number of medium follicles (3 to 5 mm), diameters of the three largest nonatretic follicles, percentage of large nonatretic follicles (greater than 3.67 mm) and mitotic index of class 3 (.68 to 1.57 mm) follicles in saline-infused cows. In contrast, in PGF2 alpha-infused cows, weaning decreased (P less than .05 to P less than .001) all of these responses. Surface and histologic evaluations of follicular activity indicated that postpartum infusion of PGF2 alpha stimulated development of large follicles. In response to weaning, PGF2 alpha infusion favored development of those large follicles toward ovulation or atresia on nonovulated follicles.  相似文献

The effect of GnRH pretreatment on estrus detection rate, precision of estrus, and reproductive performance of postpartum beef cows synchronized to estrus using GnRH and PGF2alpha was evaluated. In Exp. 1, Angus cows (n = 87) were randomly assigned by parity, postpartum interval, and body condition score (BCS) to receive either 1) GnRH on d -7 and PGF2alpha on d 0 (GP) or 2) the GP treatment and an additional injection of GnRH on d -16 (GGP). Estrus detection and AI were conducted twice daily from d -3 to d 3. At 72 h after PGF2alpha, all animals not previously detected in estrus were bred by AI and received a concurrent injection of GnRH (TAI). Synchronized pregnancy rates were numerically increased (P = 0.15) in cows treated with GGP (55%) compared with those on the GP treatment (44%). In Exp. 2, 1,276 spring-calving, suckled beef cows in nine herds were randomized to treatments as described for Exp. 1, except that the initial GnRH injection for the GGP treatment was administered on d -14. Herd affected all indicators of reproductive performance (P < 0.05). The percentage of animals detected in estrus prematurely (d -3 to d 0; 7%) was not affected by treatment. Estrus response rate was influenced by postpartum interval (< 60 vs > or = 60; 61 vs 73%; P < 0.01) and a three-way interaction of parity, BCS, and treatment (P < 0.01). Within animals with a BCS > or = 5.5, the GGP treatment tended to increase the detection of estrus in primiparous cows (GP vs GGP; 76 vs 91%; P = 0.11) and decrease detection in multiparous cows (GP vs GGP; 78 vs 72%; P < 0.10). However, because conception rate to TAI in animals with a BCS > or = 5.5 was greater (P < 0.05) in the GGP than in the GP group (28 vs 8%, respectively), this interaction was interpreted to represent a shift in interval to estrus induced by the GGP treatment, rather than a reduction in the synchronization of ovarian function. Conception rates of animals inseminated to an observed estrus did not differ among treatments (P = 0.15). Synchronized pregnancy rate tended (P = 0.06) to be greater in GGP- (53%) than in GP-treated animals (47%). In conclusion, pretreatment with GnRH tended to increase pregnancy rates during a 6-d synchronization period, primarily through enhanced conception rates of cows bred by TAI. In contrast to our hypothesis, GnRH pretreatment did not increase the percentage of animals detected in estrus or the precision of estrus expression.  相似文献

AIM: To compare 2 treatments for anovulatory anoestrus (AA) in postpartum dairy cows. The treatments were combinations of gonadotropin-releasing hormone (GnRH) and prostaglandin F2 (PG) or progesterone (P4) and oestradiol benzoate (ODB). METHODS: Forty AA cows from each of 5 herds were blocked by age (2 or 2 years old) and randomly assigned to 1 of 2 treatments. The first group (GPG) were treated with 250 mug of a GnRH analogue, gonadorelin, followed 7 days later by 15 mg of the PG analogue, luprostiol. Two days later the cows were injected with 250 mug of gonadorelin. Cows were artificially inseminated 16-24 h after the second GnRH injection. The second group (P4+ODB) were treated with an intravaginal P4 releasing device for 6 days, followed 24 h after device removal by injection of 1 mg of ODB. Cows were pregnancy tested 35-40 days after the initial insemination and twice again at 6-8 week intervals thereafter. RESULTS: There was no significant difference between P4+ODB and GPG groups in the percentage of cows submitted for insemination in the first 7 days (94.0% vs 100% for P4+ODB vs GPG, respectively; p>0.3), in conception rate to first insemination within the first 7 days (43.6% vs 35.0% for P4+ODB vs GPG, respectively; p>0.2), in the percentage of cows conceiving in the first 28 days of the breeding period (68.0% vs 58.3%, P4+ODB vs GPG, respectively; p>0.1), or in median interval from the end of treatment to conception (20 vs 21 days; p>0.1). CONCLUSIONS: No differences in the reproductive performance of AA cows treated with either P4+ODB or GPG were detected. However, given the small number of animals enrolled, further data are required before the GPG protocol can be recommended for treatment of AA cows.  相似文献

Gonadotrophin releasing hormones have become widely used hormonal compounds in veterinary medicine, particularly with respect to bovine reproduction. The character and physiological actions of gonadotrophin releasing hormone are briefly reviewed and its clinical applications are summarized. The endocrinological research concerned with the use of gonadotrophin releasing hormone in the early postpartum period is discussed. Field trials which have been conducted to assess the effects of postpartum gonadotrophin releasing hormone administration on reproductive performance have varied widely in both design and interpretation of results. These experiments are reviewed, including the clinical trials using normal cows as well as those on cows with retained placenta.  相似文献

This study was designed to evaluate the effects of exogenous prostaglandin F2 alpha (PGF2 alpha) on hormone secretion in cows without a corpus luteum. Blood samples were taken from 10 Friesian dairy cows at frequent intervals from a jugular vein and the caudal vena cava starting between nine and 20 days after parturition. PGF2 alpha (25 mg dinoprost) was injected intramuscularly into five cows after the first eight hours of sampling. Plasma concentrations of 13,14-dihydro 15-keto PGF2 alpha (PGFM) increased rapidly but had returned to baseline by 14 hours after injection. There was no significant effect of the treatment on the time taken by the cows to resume ovarian cycles, and it had no consistent effect on plasma luteinising hormone (LH) patterns; however the amplitude of pulses of LH was temporarily suppressed in two cows and the frequency of pulses of LH was immediately increased in one cow. Treatment with PGF2 alpha had no significant effect on the concentration of oestradiol in blood from the vena cava. It is concluded that any enhancement of the reproductive performance of cows treated with PGF2 alpha after parturition is not due to a direct effect on pituitary-ovarian function.  相似文献

Thirty-six postpartum Holstein cows consisting of eighteen cows that shed the placenta soon after calving (NRP) and eighteen cows that retained placenta greater than 24 h (RP) were used. There were four treatment groups. Group I consisted of 9 NRP cows which received intramuscular injection of sterile saline on day 15 postpartum. The second group consisted of 9 NRPN cows which received 100 micrograms of gonadotrophin releasing hormone on day 15 postpartum (NRPT). The third group consisted of 9 RP cows which received saline on day 15 postpartum (RPN) and the fourth group consisted of 9 RP cows which received 100 micrograms of gonadotrophin releasing hormone on day 15 postpartum (RPT). Blood samples were collected once daily during the first month and once every other day during the second month postpartum. In addition fourteen cows (RPT, n = 7; NRPT, n = 7), were used to study short-term changes in serum luteinizing hormone concentrations following gonadotrophin releasing hormone treatment on day 15 postpartum. Blood samples were obtained from these cows every 15 min during 1 h before and 6 h after gonadotrophin releasing hormone administration. Sera from all samples were assayed for progesterone and luteinizing hormone concentrations. Starting from four days after calving rectal palpations and ultrasound examinations of the ovaries were carried out once every four days until day 28 postpartum in order to monitor ovarian changes. The efficacy of various combinations of melengestrol acetate (MGA), GnRH, and PGF2alpha for the synchronization of estrus in Angus-based beef cattle was compared. Hormones were administered as follows: MGA, 0.5 mg x animal(-1) x d(-1) mixed in a grain carrier; GnRH, 100 microg i.m.; PGF2alpha, 25 mg i.m. In Exp. 1, 2, and 3, cows were randomly assigned to treatments by parity and interval postpartum. The detection of estrus and AI were conducted from d -2 until 72 to 96 h after PGF2alpha, at which time cows not detected to be in estrus received GnRH and fixed-time AI (TAI). Data were analyzed separately for primiparous and multiparous cows. In Exp. 1, cows (n = 799) at three locations received GnRH on d -7 and PGF2alpha on d 0 and either no further treatment (GnRH-PGF) or short-term MGA from d -6 through d -1 (STMGA). Among multiparous cows, conception rate at TAI was greater (P < 0.05) for STMGA (41%, 47/115) than for GnRH-PGF treated cows (26%, 24/92). Across herds and parity, synchronized AI pregnancy rate (SPR) was not affected (P > 0.10) by treatment (GnRH-PGF vs. STMGA; 54%, 210/389 vs. 57%, 228/402). In Exp. 2, cows (n = 484) at three locations received either STMGA or long-term MGA from d -32 through d -19, GnRH on d -7, and PGF2alpha on d 0 (LTMGA). Among primiparous cows, SPR was greater (P < 0.01) in LTMGA (65%, 55/85) than STMGA-treated cows (46%, 40/87). Treatment had no effect (P > 0.10) on SPR among multiparous cows (STMGA vs. LTMGA; 59%, 92/155 vs. 64%, 101/157). In Exp. 3, cows (n = 838) at four locations received the LTMGA treatment and either no further treatment or an additional period of MGA exposure from d -6 through d -1 (L&STMGA). Among primiparous cows, SPR tended to be influenced (P < 0.10) by the herd x treatment interaction and was greater (P < 0.01) among L&STMGA (86%, 19/22) than LTMGA-treated cows (56%, 14/25) at a single location. Among multiparous cows, SPR was lower (P < 0.05) in L&STMGA (46%, 165/358) than LTMGA-treated cows (55%, 184/336). In Exp. 4, Angus heifers (n = 155) received either STMGA or 14 d of MGA (d -32 through d -19) and PGF2alpha on d 0 (MGA-PGF). The detection of estrus and AI were conducted from d -2 to d 6. Interval to estrus was greater (P < 0.05) and estrous response was lower (P < 0.05) in STMGA than MGA-PGF-treated heifers. In conclusion, primiparous cows responded more favorably to longer-duration MGA treatments than did multiparous cows. All protocols achieved sufficient SPR to justify their use for improved reproductive management of postpartum beef cows.  相似文献

选择3~4月龄犊牛、后备母牛和成年母牛(产奶后期、产前及产后1月内).测定了血清Se、Cu、Zn、Mn、Fe含量厦谷胱甘肤过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量.结果表明,除Zn含量外,所测元素含量犊牛明显低于后备母牛和成年牛(P<0.05或P<0.01),后备母牛和成年牛之闻差异不显著(P>0.05),血清Se含量在产奶后期和产前明显下降(P<0.05),血清Cu、Mn含量在产后1月内有降低的趋势,但与产前相比均差异不显著(P>0.05);所测血清氧化状态指标与年龄无明显关系,在妊娠末期和泌乳早期发生改变,血清CAT、GSH-Px、SOD活性和MDA含量在产前和产后1月内最高.表明犊牛上述所测指标最低,奶牛产前和泌乳早期血清微量元素含量和氧化状态失衡.  相似文献

Two experiments were conducted to determine if administration of progesterone within a low, subluteal range (0.1-1.0 ng/mL) blocks the luteinizing hormone (LH) surge (experiments 1 and 2) and ovulation (experiment 2) in lactating dairy cows. In experiment 1, progesterone was administered to cycling, lactating dairy cows during the luteal phase of the estrous cycle using a controlled internal drug release (CIDR) device. CIDRs were pre-incubated in other cows for either 0 (CIDR-0), 14 (CIDR-14) or 28 days (CIDR-28). One group of cows received no CIDRs and served as controls. One day after CIDR insertion, luteolysis was induced by two injections of prostaglandin (PG) F(2alpha) (25 mg) at 12 h intervals. Two days after the first injection, estradiol cypionate (ECP; 3 mg) was injected to induce a LH surge. Concentrations of progesterone after luteolysis were 0.11, 0.45, 0.78 and 1.20 ng/mL for cows treated with no CIDR, CIDR-28, CIDR-14, and CIDR-0, respectively. LH surges were detected in 4/4 controls, 4/5 CIDR-28, 2/5 CIDR-14 and 0/5 CIDR-0 cows following ECP. In experiment 2, progesterone was administered to cycling, lactating, Holstein cows during the luteal phase of the estrous cycle as in experiment 1. Luteolysis was induced as in experiment 1. The occurrence of an endogenous LH surge and ovulation were monitored for 7 days. Concentrations of progesterone after luteolysis were 0.13, 0.30, 0.70 and 1.20 ng/mL for cows treated with no CIDR, CIDR-28, CIDR-14 and CIDR-0, respectively. LH surges and ovulation were detected in 5/5 controls, 3/7 CIDR-28, 0/5 CIDR-14 and 0/5 CIDR-0 cows. It was concluded that low concentrations of progesterone can reduce the ability of either endogenous or exogenous estradiol to induce a preovulatory surge of LH and ovulation.  相似文献

Four experiment stations (IL, KS, MN, and MO) conducted experiments to determine effects of introducing a CIDR (controlled internal device release) into an ovulation control program for postpartum suckled beef cows. Five hundred sixty cows were assigned randomly to two treatments: 1) 100 microg of GnRH (i.m.) followed in 7 d with 25 mg of PGF2alpha, followed in 48 h by a second injection of GnRH and one fixed-time insemination (Cosynch; n = 287) or 2) Cosynch plus one CIDR during the 7 d between the first injection of GnRH and PGF2alpha (Cosynch+P; n = 273). Cows at three stations were inseminated at the time of the second GnRH injection (n = 462), whereas 98 cows at the fourth station were inseminated 16 to 18 h after that injection. Blood samples were collected at d -17, -7, 0, and 2 relative to PGF2alpha to determine concentrations of progesterone. Ultrasonography was used to monitor follicle diameter on d 2 and to determine the presence of an embryo at 30 to 35 d after insemination. Pregnancy rates were greater (P < 0.05) for Cosynch+P- (58%) than for Cosynch-treated (48%) cows. No station x treatment interaction occurred; however, cows at MO (62%) and KS (60%) had greater (P < 0.05) pregnancy rates than those at IL (47%) and MN (44%). Cows that had follicles > 12 mm on d 2 had greater (P < 0.01) pregnancy rates than those with follicles < or = 12 mm regardless of treatment. Pregnancy rates were similar between Cosynch and Cosynch+P treatments when cycling cows had elevated concentrations of progesterone at d 0, but pregnancy rates were greater (P < 0.05) in the Cosynch+P (79%) than in the Cosynch (43%) treatment when cycling cows had low concentrations of progesterone on d 0 (at PGF2alpha injection). Similarly, among noncycling cows, pregnancy rates were greater (P < 0.05) in the Cosynch+P (59%) treatment than in the Cosynch (39%) treatment. Cows in greater body condition at the onset of the breeding season experienced improved (P < 0.001) overall pregnancy rates. Pregnancy rates for cows that calved > 50 d before the onset of the breeding season were greater (P < 0.01) than those for cows that calved < or = 50 d. Thus, treatment of suckled cows with Cosynch yielded acceptable pregnancy rates, but addition of a CIDR improved pregnancy rates in noncycling cows. Body condition and days postpartum at initiation of the breeding season affected overall efficacy of the Cosynch and Cosynch+P protocols.  相似文献

The effect of early lactation on serum paraoxonase activity was studied on 21 postpartum dairy cows and 19 non-pregnant late lactating dairy cows. A significant decrease of the paraoxonase activity was found in the early postpartum period compared to the late non-pregnant lactation. The serum triglyceride, cholesterol and LDL-cholesterol concentration were also markedly reduced during the postpartum period, while the serum HDL-cholesterol concentration showed no significant change. The results indicate that lower serum paraoxonase activity is associated with lipid metabolic disorders in the early postpartum period. A decreased serum paraoxonase activity may lead to the reduction of the antioxidative capacity and antioxidative protection during the early postpartum period.  相似文献

Objectives were to examine how the conceptus and recombinant bovine interferon-tau (rbIFN-tau) regulate intracellular components of the PGF(2a) synthetic pathway and to determine if arachidonic acid (AA) is limiting in endometrial tissue of pregnant cows. In Experiment 1, uteri were collected from either cyclic or pregnant dairy cows on Day 17 post-estrus. Intercaruncular explants were dissected and incubated for 60 min to quantify PGF(2a) production in response to oxytocin (10(-6) M), A23187 (10(-5) M), melittin (10(-5) M), and phorbol 12, 13 dibutyrate (PDBu, 10(-6) M). Additional explants from the same cows were incubated for 24 h with and without AA. Oxytocin and A23187 did not stimulate PGF(2a) in explants from either cyclic or pregnant cows. Both PDBu, melittin, and A23187 + melittin stimulated PGF(2a) production in explants of cyclic cows, but not in explants of pregnant cows. The addition of AA to explant cultures for 24 hr did not increase PGF(2a) production during a subsequent 60-min incubation. In Experiment 2, explants were collected from cows that received intrauterine infusions of either BSA (1.9 mg/1.2 ml) or rbIFN-tau (0.2 mg rbIFN-tau + 1.7 mg BSA/1.2 ml) twice a day from Days 14 to 17 of the estrous cycle. Treatments of rbIFN-tau attenuated PGF(2a) secretion induced by in vitro PDBu and A23187 treatments. However, rbIFN-tau treatment in vivo had no effect on the in vitro induction of PGF(2a) secretion by melittin. IFN-tau may regulate the PGF(2a) synthetic pathway by reducing activity of PKC or PKC mediated events.  相似文献

Quarter milk samples were taken from 48 clinically healthy, pregnant Finnish Ayrshire and Friesian dairy cows at the last milking before drying-off, and 2 and 6 days later to determine the lactoferrin and citrate concentrations in the udder secretion. The mean lactoferrin concentration in the milk increased from 5.29 mg ml(-1) on the last day of drying-off process to 8.09 and 11.26 mg ml(-1), 2 and 6 days later, respectively. Citrate concentration decreased from 1.85 mg ml(-1) to 1.54 and 1.09 mg ml(-1), respectively. Median molar ratio (citrate to native-Lf) decreased from 153 to 86 and further to 44. Lf and citrate concentrations in milk varied greatly among cows. Sampling time had a statistically significant effect on lactoferrin and citrate concentrations in milk while the breed and the parity of the cows had no effect on either of these variables.  相似文献

We investigated whether CIDR-based ovulation-synchronization protocols inhibit secretion of prostaglandin (PG) F2alpha from the uterus in the following luteal phase in non-cycling beef cows. Ten early (a month) postpartum non-cycling Japanese Black beef cows were treated with (1) Ovsynch (GnRH analogue on Day 0, PGF2alpha analogue on Day 7, and GnRH analogue on Day 9; n=3), (2) Ovsynch+CIDR (Ovsynch protocol plus a CIDR for 7 days from Day 0; n=4), or (3) estradiol benzoate (EB) Ovsynch+CIDR (EB on Day 0 in lieu of the first GnRH treatment followed by the Ovsynch+CIDR protocol; n=3). An oxytocin challenge was administered on Day 24 to examine uterine PGF2alpha secretion. Plasma concentrations of 13,14-dihydro-15-keto- PGF2alpha were lower at 30-120 min after oxytocin administration in the Ovsynch+CIDR group and 75 min after administration in the EB Ovsynch+CIDR group than in the Ovsynch group (P<0.05). Plasma progesterone concentrations were higher from Days 1 to 7 in the Ovsynch+CIDR group and from Days 1 to 5 in the EB Ovsynch+CIDR group than in the Ovsynch group (P<0.05). The progesterone concentrations were higher on Days 27 and 29 in both CIDR-treated groups than in the Ovsynch group (P<0.05). In conclusion, in non-cycling beef cows, CIDR-based ovulation-synchronization protocols inhibit uterine PGF2alpha secretion in the following luteal phase and prevent premature luteolysis as is seen with the Ovsynch protocol.  相似文献

OBJECTIVE: To compare calving-to-conception intervals among cows classified as nonlame, moderately lame, or lame during the prebreeding postpartum period and to examine the relationship between severity of lameness and time to conception in cows that were classified as lame. DESIGN: Longitudinal study. ANIMALS: 499 Holstein cows. PROCEDURE: Cows in the prebreeding postpartum period were classified as nonlame, moderately lame, or lame by use of a 6-point locomotion scoring system. Time to conception (days) was compared among cows. A low, medium, or high cumulative locomotion score was assigned to lame cows, and time to conception among those cows was compared. Cows classified as lame were examined on a tilt table for diagnosis and treatment of lameness. RESULTS: 154 (31%), 214 (43%), and 131 (26%) cows were classified as nonlame, moderately lame, and lame, respectively. Most cows classified as lame had laminitis (54%) or disorders of the claw (33%). Median time to conception was 36 to 50 days longer in lame cows than in nonlame cows. Among lame cows, the median time to conception was 66 days longer in cows with high cumulative locomotion scores than in cows with low scores. CONCLUSION AND CLINICAL RELEVANCE: Nonlame cows became pregnant more quickly than lame cows. Lame cows with low cumulative locomotion scores during the prebreeding postpartum period became pregnant sooner than lame cows with high scores. Early diagnosis and intervention may mitigate the effects of lameness and improve reproductive performance in lame dairy cows.  相似文献
