This study compares the aetiology of pansteatitis in Lake Loskop, relative to two other impoundments along the Olifants River. Macroscopic and microscopic pathology, age determination and analysis of stomach content, fatty acids and stable isotopes explain the high prevalence of pansteatitis in Oreochromis mossambicus (Peters) and several other species in Lake Loskop. All the dietary indicator comparisons between pansteatitis‐affected and healthy fish fail to support a systemic cause. Pansteatitis in Lake Loskop was linked to size and weight of O. mossambicus, but not to ontogenic age. Fish in Lake Loskop showed abnormally high omega‐3 to omega‐6 fatty acid ratios normally only found in marine fish with no significant difference in degree of assimilation of these fatty acids between pansteatitis‐affected and healthy fish. This explains the vulnerability to, but not the occurrence of, pansteatitis. As a cause for the pansteatitis, these results point towards sporadic vitamin E‐depleting trigger events, known sporadic fish die‐off occurrences that provide surviving fish with a rich source of rancid fats on which to scavenge. The mechanism ties pansteatitis to eutrophication and trophic cascade effects, the intrinsic drivers of the disease and suggests an adaptive management strategy that might be applied by relevant conservation authorities.  相似文献

Large numbers of adult Nile crocodiles, Crocodylus niloticus (Laurenti), died from pansteatitis during autumn and winter 2008 in the lower Letaba and Olifants River gorge in the Kruger National Park, South Africa. Consequently, the health status of fish from these waters was investigated. The study presents the pathological findings in fish inhabiting these rivers within the boundaries of the Park. Changes typical of steatitis were diagnosed in many of the larger specimens of sharptooth catfish, Clarias gariepinus (Burchell), caught within the Olifants River gorge. These fish carried large amounts of mesenteric fat with characteristic small brown granulomata within the adipose tissue. Necrosis and inflammation of the adipose tissues, with characteristic ceroid accumulation within the resultant granulomata and the associated aggregation of ceroid‐containing macrophages, were demonstrated histologically and were typical of steatitis. Other changes included mild thickening and pallor of the gill tissues and swollen, orange, fatty livers. Focal hepatic lipidosis was demonstrated histologically, and special stains revealed storage of large amounts of iron in the livers. Blood smears revealed chromatin clumping in erythrocyte nuclei and nuclear and cell membrane irregularities. This is the first record of steatitis in wild‐caught C. gariepinus.  相似文献

Pansteatitis is the leading cause for the decline in Nile crocodile populations and the sporadic mortality of fish in the Olifants River System, South Africa. To determine the prevalence of this disease in lentic systems, Mozambique tilapia, Oreochromis mossambicus, were collected from Lake Loskop, Lake Flag Boshielo, Phalaborwa Barrage and Lake Luphephe-Nwanedi. The former three impoundments are located within the main stem of the Olifants River, while the latter, which is geographically isolated and situated in the Limpopo River System, served as a reference site. Mesenteric adipose, liver, serosa of the swim bladder, gill and the skeletal muscle of fish sampled were examined for gross and microscopic evidence of pansteatitis. Microscopically observed changes were used to statistically compare pansteatitis prevalence between samples and sites. Based on histopathological evaluation, the adipose tissue in the liver, swim bladder serosa and coelom from severely debilitated individuals showed the most significant pathological changes. Lesions indicative of steatitis were observed in fish collected from Lake Loskop (75%), Lake Flag Boshielo (22%) and Lake Luphephe-Nwanedi (15%). Further investigation is warranted to understand the pervasiveness and mechanisms driving pathological changes of pansteatitis at Lake Flag Boshielo, Phalaborwa Barrage and Lake Luphephe-Nwanedi.  相似文献

  • 1. Top predators such as crocodiles often reflect ecosystem degradation. The recent spate of close to 200 Nile crocodile deaths may reflect the ecosystem state of the Olifants–Letaba River system in the Kruger National Park. This paper investigates whether the crocodile deaths were indeed the consequence of a rare and perhaps acute event given the variability in annual population growth rates of crocodilians.
  • 2. Spotlight‐ and helicopter‐based surveys designed to correct for availability and detectability bias were used to estimate population sizes of crocodiles from 2008 to 2010 in and around the Olifants River Gorge, the focal area of crocodile deaths.
  • 3. Correction factors derived from individually observed crocodiles for both spotlight‐ and helicopter‐based counts were lower than those typically used for Nile crocodiles as well as other crocodile species. Even so, corrected spotlight‐ and helicopter‐based estimates were comparable and the number of crocodiles in the focal study area declined significantly from 780 (95% CI: 637–1222) to between 460 (spotlight estimate, 95% CI 375–665) and 505 (aerial estimate, 95% CI: 559–1746) during the period of crocodile deaths. The average annual decline of 35% was at the lower end of the distribution of annual population growth rates across the rivers of Kruger National Park.
  • 4. The crocodile deaths reflect a possible rare event that suggests a degraded crocodile population, possibly the consequences of broad‐scale cascades of environmental deterioration of the Olifants–Letaba River system.

Even so, the potential risk of local demise of the population in the focal study area in the short term may be diminished through evolutionary, demographic and spatial resilience inherent within crocodiles that can accommodate the as yet unknown disease dynamics of pansteatitis. The present study examines the effect of four semi‐purified diets (casein–gelatin based) where the source of fatty acids was free (esterified) oleic acid and linoleic acid (LA) (LOA diet), linseed and olive oil (predominantly LA and linolenic acid) (LO diet), cod liver oil (rich in highly unsaturated fatty acids) (CLO diet), and soybean lecithin (phospholipids; mostly LA) (LE diet) on the growth of juvenile South American catfish (surubim, Pseudoplatystoma fasciatum, Pimelodidae) (0.98 ± 0.04 g individual weight). Fish were fed at a restricted–readjusted feeding rate for 8 wk. At the end of the experiment, LE‐diet‐fed fish grew significantly larger than those of the other three groups (P < 0.05). Considerable cannibalism was observed in all the treatments. It is suggested that the quantitative growth performance may possibly change under other conditions, with less or no cannibalism. Survival did not differ significantly among the fish fed four different diets. Muscle and liver lipid contents did not vary among dietary treatments (P > 0.05), but whole‐body lipid concentrations were affected by dietary treatments. Fish fed LE diet contained significantly lower lipid level than those fed three other diets (P < 0.05). Muscle and liver fatty acid profiles reflected dietary fatty acid composition. Arachidonic acid level was significantly higher in muscle and liver of fish fed LOA and LE diets than in those fed LO and CLO diets. The results suggest that the efficiency of elongation and desaturation of 18C fatty acids depends on the dietary lipid source, and South American catfish has considerable capacity to transform linoleate to arachidonate.  相似文献

Effects of various concentrations of Scutellaria baicalensis (SB) extract in diets on growth, body composition, serum chemistry and disease challenge test of far eastern catfish (Silurus asotus) were determined and compared with a commercially available immune enhancer. Eight experimental diets were prepared in triplicate: control (Con) diet without supplementation of SB and SB‐0.25, SB‐0.5, SB‐1, SB‐2, SB‐3 and SB‐5 diets containing SB at concentrations of 0.25, 0.5, 1, 2, 3 and 5%, respectively. In addition, 0.1% of a commercial immune enhancer product (CP) was also tested. No significant difference in weight gain of fish was found. Feed consumption, feed efficiency ratio and protein retention of fish were not affected by the experimental diets. At the end of the 8‐week feeding trial, 10 externally normal fish from each tank were infected by Vibrio anguillarum or Strepotococcus iniae. Cumulative mortality of fish fed the Con diet was higher than that of fish fed the all other diets in 10 and 25 days after V. anguillarum or S. iniae infection. Results of this study indicated that dietary inclusion of SB extract was effective in improving survival of eastern catfish after V. anguillarum and S. iniae infection, but the various concentrations of SB did not affect fish performance.  相似文献

Black catfish (Rhamdia quelen) is a species of interest for aquaculture in Brazil, Argentina and Uruguay. A feeding trial was conducted to evaluate the effect of feeding R. quelen larvae on either only an artificial diet or in combination with Artemia nauplii (AN) on larval performance and fatty acid composition. For 12 days, larvae were fed from first feeding (3 days after hatching, TL = 5.88 ± 0.23 mm) with artificial food only or a combination of artificial food and AN (co‐feeding). At the end of the trial, total length of co‐fed larvae was significantly higher than that of larvae fed solely artificial food (P < 0.001). No significant differences were found in survival rates. Co‐feeding microdiet with a small amount of AN significantly affected larval fatty acid composition. Lipid and fatty acid composition of food and larvae revealed the importance of n‐3 fatty acids for growth of black catfish larvae and that, as most freshwater fish, R. quelen larvae can elongate and desaturate linolenic acid to n‐3 highly unsaturated fatty acids. Results suggest that R. quelen larvae can be fed from first feeding on microdiets as unique food source, although better larval performances are obtained by co‐feeding with a small amount of AN.  相似文献

Experimental diets were formulated containing six levels of linolenic acid (0%, 0.5%, 1.0%, 1.5%, 2.0% and 2.5%) and each diet was fed to darkbarbel catfish Pelteobagrus vachelli in triplicate for 84 days. Weight gain, specific growth rate and feed efficiency increased with the increasing levels of dietary linolenic acid, but were not significantly different among diets containing 1.0%, 1.5%, 2.0% and 2.5% linolenic acid. Red blood cell count and haemoglobin were significantly higher in fish fed the 1.0% linolenic acid than other diets. Lysozyme activity and immunoglobulin M content in fish fed 1.0% of linolenic acid were significantly higher compared with the other diets. Antibody titre in fish fed 1.0–2.5% linolenic acids was significantly higher than in fish fed 0 or 0.5% linolenic acid 14 days after Aeromonas hydrophila challenge. This study indicates that linolenic acid is essential to darkbarbel catfish. Two‐straight broken‐line analysis with SGR showed that a dietary level of 1.29% linolenic acid is the minimum required for adequate growth of darkbarbel catfish. Considering other measured indicators of fish performance, a level of 1.0–1.5% linolenic acid in the diet is recommended for the darkbarbel catfish.  相似文献

A major challenge in Clariid catfish seed production is our inability to induce voluntary captive spawning of the brooders. Since voluntary captive spawning had been previously elicited in a Clarias species by simulating natural breeding environment, we speculated that the major obstacle in voluntary spawning might be the absence of an inducing agent that triggers mating response in the brooders. To address the issue, oxytocin was injected to the brooders along with Ovatide (a commercial inducing agent comprising salmon gonadotropin‐releasing hormone analogue and domperidone). Administration of 40 mIU/kg body weight of oxytocin after 12 hr of Ovatide injection resulted in voluntary spawning of the fish (81.5%, i.e. 31 out of 38 trials), with more than 70% hatching rate in simple and low‐cost spawning facilities. Histology of male gonads indicated sustained gamete maturation and the gonads were observed to be visibly turgid in the fish that received both the hormones. In contrast, gamete maturation slackened after 10 hr in the control fish, injected with Ovatide only. Administration of oxytocin along with Ovatide was also observed to induce aggressive sexual behaviour among the males. Neither of the hormones, acting alone, could induce voluntary spawning in the fish or influence sexual behaviour. Our results clearly show that oxytocin plays a significant role in sustaining the male gamete maturation induced by Ovatide and trigger the behavioural mating response, leading to voluntary spawning in C. magur. The results hold potential to make a paradigm shift in the commercial catfish seed production technology.  相似文献

The effect of using a finishing diet containing menhaden fish oil on the fatty acid composition of fingerling channel catfish, Ictalurus punctatus, was evaluated in a 12‐wk growth trial. Three isocaloric, isonitrogenous practical diets with three different sources of lipids (menhaden oil [MO], catfish oil [CO], or beef tallow [BT]) were formulated (35% crude protein). No differences in eicosapentaenoic acid, docosahexaenoic acid, or arachidonic acid were observed to occur in catfish fed MO or CO diets; however, these fatty acids were significantly lower in fish fed BT diet. No differences were observed for unsaturated fatty acid content in channel catfish fed a diet containing MO for 8 or 12 wk. In addition, no differences in production characteristics were observed to occur when catfish were fed diets containing CO, MO, or BT as the dietary lipid source, which indicates that BT, CO, and MO are equally effective as sources of energy. It is apparent from these results that CO may be successfully substituted for MO in formulated diets without adversely affecting n‐3 highly unsaturated fatty acid content in channel catfish.  相似文献

The role of dietary linolenic acid (LN), vitamin E (E) and vitamin C (C) in regulating fish growth and immune response was tested on juvenile darkbarbel catfish Pelteobagrus vachelli. Five dietary combinations were used (?E?LN, +E?LN, ?E+LN, +E+LN and ?C+E+LN; ‘+’ with addition and ‘?’ without addition) in triplicate. Weight gain was highest in the ?E+LN feeding group. Red blood cell in fish fed the +E+LN diet was highest. The haematocrit and haemoglobin of fish fed the ?E+LN diet was lowest. Superoxide dismutase, catalase, glutathione peroxidase and glucose‐6‐phosphate dehydrogenase activities in fish fed the ?E+LN diet were higher than those in fish fed other diets. Malondialdehyde in fish fed the ?C+E+LN diet was highest. Fish fed the +E+LN diet had higher levels of lysozyme activity, serum protein, complements C3 and C4, and immunoglobulin contents than fish fed other diets. Fish fed the +E+LN diet showed lower mortality and higher antibody titre than fish fed other diets after the fish were challenged with Aeromonas hydrophila for 14 day. This study suggests that the growth of darkbabel catfish is improved by increasing dietary linolenic acids. The diets with high linolenic acid, vitamin E and vitamin C can enhance the immune response and resistance in darkbarbel catfish challenged with A. hydrophila.  相似文献

An 8‐week feeding trial was conducted on juvenile beluga sturgeon Huso huso to evaluate the effects of different dietary lipid levels and sources on growth performance, physiological indices, proximate composition and fatty acid (FA) profile. Four practical diets, which had either low level (120 g/kg) of canola oil (LCO) and fish oil (LFO) or high level (240 g/kg) of canola oil (HCO) and fish oil (HFO), were fed to triplicate groups of 25 beluga (mean initial body weight 207 ± 0.5 g). The growth performance of beluga was improved by replacing dietary fish oil with canola oil and increasing dietary lipid level. Except the number of red blood cells, lymphocytes, neutrophils and eosinophils, the rest of haematological factors including the values of haemoglobin, haematocrit, number of white blood cells, mean corpuscular haemoglobin concentration, cholesterol and triglyceride concentrations and the number of basophils and monocytes were not significantly affected by dietary lipid sources or levels. Results showed that both moisture and crude fat of the beluga muscle were affected by dietary lipid. The highest moisture and the lowest fat contents were found in the muscle of beluga fed fish oil (LFO). Moreover, the lowest moisture and the highest fat contents were observed in the muscle of beluga fed canola oil (HCO) (< .05). The FA profile of the beluga muscle was significantly influenced by dietary treatments. The highest monounsaturated fatty acids, total n‐6 fatty acids containing linoleic acid and arachidonic acid, and total unsaturated fatty acids were found in fish fed canola oil (LCO and/or HCO) (< .05). However, n‐3 fatty acids containing linolenic acid, eicosapentaenoic acid and docosahexaenoic acid were not affected by the diet (> .05). FA profile of the beluga muscles reflected the proportions of CO and FO in the diet except that there was a decrease in oleic acid and linolenic acid, but an increase in arachidonic acid (C20:4n‐6), eicosapentaenoic acid and docosahexaenoic acid. The obtained data showed that canola oil is an excellent source of supplemental dietary lipid in a practical fish‐meal‐based diet of beluga sturgeon under the experimental conditions. Moreover, the data demonstrated that increasing dietary lipid up to 240 g/kg in beluga sturgeon resulted to improve growth performance and haematology.  相似文献

Abstract This study was conducted to compare the fatty acid composition of muscle, liver, and depot fat of common snapping turtles Chelydra serpentina from the wild with those of captive common snapping turtles fed a diet of known fatty acid composition. Total lipid from each tissue was separated by thin-layer chromotography, methylated, and analyzed by gas chromatography/mass spectrometry. The fatty acid composition of total lipid, polar lipid, and nonpolar lipid in muscle and depot fat of wild and captive turtles exhibited greater variability than liver polar lipid. Cultured turtles contained significantly lower levels of linolenic and arachidonic acids in liver polar lipid than wild turtles. Total n-9 fatty acids were significantly higher in whole-lipid and nonpolar fractions of muscle, liver, and depot fat of cultured turtles indicating de novo synthesis. Total polyunsaturated fatty acids were significantly lower in total lipid of muscle tissue from cultured turtles. Total polyunsaturated fatty acids were significantly lower in both depot fat and liver nonpolar lipid of cultured turtles, but did not differ (P > 0.05) in the polar lipid fractions of cultured and wild turtles.  相似文献

Recent studies in terrestrial animals have shown that feeding the oxidized lipids led to a reduction in triacylglycerols (TAG) and total cholesterol (TC) in liver and plasma. However, limited information is available on the effect of oxidized lipids on lipid metabolism in fish. In this study, four diets containing 0 g kg?1 (control: fresh fish oil), 30 g kg?1 (low‐oxidized oil, LOO), 60 g kg?1 (medium‐oxidized oil, MOO) and 90 g kg?1 (high‐oxidized oil, HOO) graded oxidized oil levels with the same dietary lipid level were fed to channel catfish for 86 days. The tissue lipid metabolism and fatty acid composition of the fish were investigated after this period. The results showed that plasma and liver concentrations of TAG and TC decreased with increasing dietary oxidized oil level (< 0.05). Decreasing liver lipoprotein lipase and hepatic lipase activities were observed with increasing dietary oxidized fish oil inclusion (< 0.05). The liver C22:6n?3 concentrations significantly decreased with increasing dietary oxidized oil level (< 0.05), while muscle lipid had a high proportion of polyunsaturated fatty acids. It suggests that the adverse effects of dietary oxidized oil may be induced by inhibiting lipid metabolism enzymes and, consequently, inhibition of cholesterol homoeostasis and fatty acid synthesis.  相似文献

Channel catfish, Ictalurus punctatus, 88.4 ± 2.6 g/fish, were fed a basal diet amended with 4% of three processed menhaden, Brevoortia tyrannus, oils. These were compared with basal diets amended with 4% corn oil or 4% canola oil. Three replicate aquaria of nine fish each were fed assigned diets twice daily. At 6 wk, fish were group weighed, fillets were collected for sensory evaluation, fatty acid analysis by gas chromatography (GC). In a second study, catfish, 118.8 ± 3.2 g/fish, were stocked into fifteen 0.04‐ha earthen ponds and fed once daily for 16 wk one of four diets containing 2 or 4% of either catfish offal oil or refined (RF) menhaden oil. At harvest, fillets were saved for sensory evaluation and fatty acid analysis. Results showed no significant (P > 0.05) differences among treatments for aquarium study and pond study variables such as weight gain, fillet proximate analysis, or pond production. GC analysis showed that levels of omega‐3 (n‐3) highly unsaturated fatty acids (HUFA) in fillet lipid were significantly (P < 0.05) elevated for fish fed menhaden oil diets. Sensory evaluation revealed that fillets from fish fed RF menhaden oil had satisfactory flavor and could be a source of n‐3 HUFA for humans.  相似文献

A 15‐wk study was conducted to evaluate the effect of supplemental menhaden fish oil levels and feeding duration on growth performance and tissue proximate and fatty acid (FA) compositions of juvenile channel catfish, Ictalurus punctatus. Dietary fish oil levels had no effect on final weight gain, feed efficiency, and survival of channel catfish. Tissue lipid contents were directly correlated to dietary lipid levels, while moisture contents were inversely related to dietary lipid levels. Fillet moisture contents progressively decreased, whereas fillet lipid increased with increasing feeding duration. Significant increase in saturated and total n‐3 FAs and decrease in monoenoic and total n‐6 FA in whole body and fillet were observed at each incremental level of dietary fish oil. Percentages of n‐3 and n‐3 highly unsaturated fatty acids in fillet of fish fed the control and 3% fish oil diets decreased with increasing feeding periods, whereas those of fish fed 6 or 9% added fish oil diets remained stable or increased. Ratios of n‐3/n‐6 were statistically comparable throughout the 15‐wk feeding. When expressed in terms of mg/g of fillet, the highest concentration of n‐3 was obtained in fillets of fish fed the 9% added fish oil diet for 15 wk.  相似文献

以贵州省铜仁锦江河漾头河段野生黄颡鱼鱼卵和同一区域内养殖黄颡鱼鱼卵为研究对象,采用常规生化方法测定了野生与养殖黄颡鱼鱼卵氨基酸组成和含量,采用气相色谱技术检测了野生与养殖黄颡鱼鱼卵脂肪酸组成和含量。氨基酸测定结果表明,野生黄颡鱼鱼卵的氨基酸总量、必需氨基酸总量、鲜味氨基酸总量显著低于养殖黄颡鱼鱼卵(P0.05),第一限制性氨基酸均为甲硫氨酸+半胱氨酸,必需氨基酸指数分别为55.60和59.12。脂肪酸测定结果表明,野生黄颡鱼鱼卵的饱和脂肪酸总量极显著高于养殖黄颡鱼鱼卵(P0.01),单不饱和脂肪酸总量二者差异不显著(P0.05),多不饱和脂肪酸总量极显著低于养殖黄颡鱼鱼卵(P0.01),但野生黄颡鱼鱼卵的二十碳五烯酸和二十二碳六烯酸的总量极显著高于养殖黄颡鱼鱼卵(P0.01),野生黄颡鱼鱼卵的n-3系列的多不饱和脂肪酸总量与n-6系列不饱和脂肪酸总量的比值(1.57)低于养殖黄颡鱼鱼卵(1.80)。  相似文献

This study determined the effect of different lipid sources on growth, feed use, lipid metabolism and antioxidant status of grass carp (Ctenopharyngodon idellus). Juvenile fish (56.9 ± 4.7 g) were divided into four triplicate groups and fed diets containing 30 g/Kg of fish oil (FO), olive oil (OO), peanut oil (PO) and linseed oil (LO), respectively, for 60 days. Weight gain and feed conversion ratio were not significantly different between the dietary groups, but we observed changes in the fatty acid composition of muscle and intraperitoneal fat reflecting the fatty acid profile of the dietary lipid source. In the hepatopancreas, the highest mRNA level of fatty acid translocase CD36 (FAT/CD36) and carnitine palmitoyl transferase (CPT‐1A) was both observed in the FO group. In muscle, the expression of FAT/CD36 and CPT‐1A in the LO group was significantly higher than that in other groups, except for CPT‐1A in the PO group. In addition, the lowest and highest content of malondialdehyde in serum was observed in OO and FO groups, respectively. In summary, dietary lipid source altered the fatty acid composition, potential uptake (FAT/CD36) and oxidation (CPT‐1A) of fatty acids, and antioxidant status of grass carp, which should be considered when selecting a lipid source.  相似文献

A feeding trial was conducted to evaluate the potential of dietary supplementation of autolysed brewer's yeast (AY) on African catfish. The catfish (22.5 ± 1.15 g/fish, 20 fish 33 L/tank) were fed with either of diets (390 g/kg crude protein, 140 g/kg lipid) supplemented with 0, 3, 6 or 10 g/kg AY (n = 3). After 49 days of feeding, the final body weight and metabolic growth rate of the catfish fed 3 g/kg AY (3‐AY) diet were higher than those fed the control diet (p < .05). The lowest level (p < .05) of alanine transaminase was detected in the blood of the catfish fed 3‐AY diet. The mid‐intestinal histology of the catfish revealed no significant difference (p > .05) in intestinal perimeter ratio. However, an elevated (p < .05) abundance of goblet cells and intraepithelial leucocytes were found in the intestine of catfish fed 3, 6 and 10 g/kg AY diets, with the highest level of abundance recorded in the mid‐intestine of the catfish fed 3‐AY diet. The results suggest that dietary 3 g/kg autolysed brewer's yeast supplementation improves growth performance of African catfish without deleterious effect on liver functionality and gut morphology.  相似文献
