The anthelmintic efficacy of ivermectin (administered subcutaneously at 200 micrograms kg-1 body weight) was assessed for control of Thelazia skrjabini in experimentally infected calves. Twenty-four uninfected male Holstein calves, 1-2 weeks old, were artificially infected with Thelazia skrjabini by placing 15 third-stage larvae under the third eyelid of calves. The challenge larvae were recovered from naturally infected face flies, Musca autumnalis. The exposed calves were randomly assigned to either an ivermectin treatment group or a control group within pairs ranked by weight. Equal numbers of calves were housed in each of four indoor fly-free rooms. The calves were treated with ivermectin or saline 35 days post-infection, then slaughtered 14 days later to determine eyeworm numbers. All eyes and associated tissues (including the lacrimal glands and ducts) were removed and examined for total number, species and viability of eyeworms. Thelazia skrjabini was found in the control group of calves only. The efficacy of ivermectin against Thelazia skrjabini was thus shown to be 100%.  相似文献

Persistent anthelmintic activity of ivermectin in cattle   总被引:1,自引:0,他引:1  
Two studies are described which demonstrate the persistent activity of ivermectin injected subcutaneously into cattle at 200 micrograms/kg in preventing the establishment of induced infections with the gastrointestinal parasites Ostertagia ostertagi and Cooperia oncophora and the lungworm Dictyocaulus viviparus. These results indicated a reduction in mean worm count compared with the control group for O ostertagi of more than 99, 45 and 94 per cent with a seven, 14 or 21 day interval between treatment with ivermectin and the administration of infective larvae, respectively, in trial 1 and more than 99, more than 99 and 99 per cent at seven, 10 or 14 days, respectively, in trial 2. Corresponding values against C oncophora were 99, 0 and 45 per cent at seven, 14 and 21 days in trial 1 and more than 99, 84 and 31 per cent at seven, 10 and 14 days in trial 2. Against D viviparus, reduction in counts were more than 99, 98 and more than 99 per cent at seven, 14 and 21 days, respectively, in trial 1 and 100, 100 and 100 per cent at seven, 10 and 14 days, respectively, in trial 2. The relevance of these results to the build-up of infective larvae on pasture and infection in cattle is discussed.  相似文献

The persistent anthelmintic effect of ivermectin as a topical treatment at 500 µg/kg was evaluated against induced infections of Ostertagia ostertagi, Trichostrongylus axei and Dictyocaulus vivparus in calves. The results showed a highly significant (P<0.001) anthelmintic activity for at least 14 days against O. oslertagi and T: axei (>99 per cent efftcacies) and for at least 28 days (98 per cent efficacy) against D. viviparus.  相似文献

Eighteen Chinese cattle were experimentally infected with metacercariae of Fasciola hepatica and randomly assigned to 6 groups. Five groups of cattle were treated with a single oral dose of triclabendazole at a dose rate of 12 mg kg-1. At necropsy, the reduction in fluke burden compared with the untreated group was 85, 99.6, 99.8, 100 and 100% for cattle treated 2, 6, 8, 12 and 16 weeks after infection, respectively. Data are also presented on body weight changes during the experimental period and on serum gamma-GT activity in cattle from selected groups. Triclabendazole is considered to be safer and more efficacious than currently available fasciolicides in China.  相似文献

Excretion of Mycobacterium bovis by experimentally infected cattle   总被引:4,自引:0,他引:4  
Three groups, each of five calves, four to seven months old, were inoculated intranasally with different numbers of Mycobacterium bovis. Infection was established readily in the calves which received an inoculum containing either 10(6) or 10(4) colony forming units (cfu). After every infection there was a lag period during which the organisms could not be isolated from specimens of nasal mucus. All the animals excreted M bovis and the time of commencement, quantity and duration of excretion appeared to be related to the inoculation dose. Excretion continued for many weeks, and for two calves excretion became intermittent over many months. All the calves which were given inocula of 92 cfu failed to develop the disease and no immunological responses were detected; however, M bovis was isolated from nasal secretions from one of these animals 100 days after inoculation.  相似文献

The persistent anthelmintic effect of subcutaneously and orally administered ivermectin given at a dose rate of 0.2 mg/kg was evaluated against experimentally induced infections of Haemonchus contortus, Ostertagia spp., Trichostrongylus spp. and Cooperia curticei in sheep. While both ivermectin treatments apparently resulted in some reduction in the establishment of most of the parasite species examined, only for C. curticei were these reductions statistically significant. For C. curticei, statistically significant antiparasitic activity was evident in animals five and ten days after treatment with ivermectin given by injection and five days after treatment with ivermectin given orally. The possible relevance of these results to parasite control and the potential for selecting for anthelmintic resistance are briefly discussed.  相似文献

Calves 1-2 months old were experimentally infected with eggs of Taenia saginata and clinical and haematological deviations, development and distribution of cysticerci and pathological changes were recorded. The calves infected with 5,000, 10,000 or 50,000 eggs showed an increase in pulse and respiratory rates. The animals that received 50,000 eggs had significantly increased pulse (p < 0.05) and respiratory rates (p < 0.005). The symptoms were more severe in young, 30-day-old calves infected with 50,000 eggs. Haemoglobin concentration, haematocrit values and red blood cell count decreased, but white blood cell count increased slightly. Lymphocytes and eosinophils also increased up to 88% and 14% (p < 0.05), respectively. Most of the cysticerci were not fully formed 1 month post-infection, but at 2 months the cysts were fully mature and at 4 months, some cysts had degenerated. There was no uniform pattern of distribution of cysticerci in the body of infected calves, but the most commonly affected sites were masseter and heart muscles, followed by diaphragm, tongue and other skeletal muscles. The maximum concentration of 8-14 cysticerci per 10 g of tissue was recorded in masseter muscles and heart. The affected parts revealed tissue reactions that included pressure atrophy, necrosis and fibrosis. Microscopically, the lesions comprised infiltration with lymphocytes, plasma cells, eosinophils and macrophages, fibrosis, necrosis and calcification. The tissue reaction was severe in calves infected with 50,000 eggs. The severity of clinical signs, haematological and pathological changes depended mostly on the age of the animals and dose of infection.  相似文献

A study was conducted to investigate the persistent nematocidal activity of two avermectins against experimentally-induced infections of Ascaris suum in swine. Seventy-two nematode-free cross-bred pigs of similar bodyweight were randomly allotted to nine treatment groups of eight pigs each. Eight of the groups were treated with injectable solutions containing 300 microg of doramectin/kg (IM) or 300 microg of ivermectin/kg (SC) either 0 (same day), 7, 14, or 21 days prior to an oral challenge of 50000 embryonated A. suum eggs. The ninth group (control) was challenged in parallel without any avermectin treatment. At 41 or 42 days after challenge, pigs were euthanatized and adult and larval stages of A. suum were collected from the gastrointestinal tract of each pig and counted. Both avermectins significantly (P < 0.0002) reduced nematode counts when given on the day of challenge (0 days prior), and the efficacy was 100% and 97.5% for doramectin and ivermectin, respectively. Doramectin given 7 days prior to challenge significantly (P < 0.0001) reduced nematode counts, and the efficacy was 98.4%. For all other avermectin-treatment groups, nematode counts were not significantly reduced compared to those in control pigs. These data indicated that anthelmintic activity of ivermectin against A. suum persisted for less than 7 days and the activity of doramectin persisted for more than 7, but less than 14 days.  相似文献

Forty-eight helminth-free lambs were divided into eight groups (A-H) of six animals. Groups A-G were infected artificially with 10,000 third stage larvae of Haemonchus contortus and 20,000 third stage larvae of Trichostrongylus colubriformis, whereas group H remained uninfected. Thirty days post-infection the lambs were treated orally with a single dosage of one of the following products: group A with 3 mg/kg body weight (BW) of an aqueous ethanol extract (70%, v/v) of the seeds of Azadirachta indica A. Juss syn. Melia azedarach L. (Meliaceae); group B with 1 g/kg BW of a raw powder of the leaves of Ananas comosus (L.) Merr. (Bromeliaceae); group C with 0.3 mg/kg BW of an aqueous ethanol extract of a 1:1 mixture (g/g) of Vernonia anthelmintica (L.) Willd. (Asteraceae) seeds and Embelia ribes Burm (Myrsinaceae) fruits; group D with 183 mg/kg BW of an aqueous ethanol extract of the whole plants of Fumaria parviflora Lam. (Fumariaceae); group E with 28 mg/kg BW of an aqueous ethanol extract of the seeds of Caesalpinia crista L. (Caesalpiniaceae); group F with 25 mg/kg BW of pyrantel tartrate and group G with 50% ethanol. Group H remained untreated. Only the ethanol extract of F. parviflora caused a strong reduction of the faecal egg counts (100%) and a 78.2 and 88.8% reduction of adult H. contortus and T. colubriformis on day 13 post-treatment. The extract was as effective as the reference compound pyrantel tartrate. Therefore, the ethanol extract itself or single constituents of F. parviflora could be a promising alternative source of anthelmintic for the treatment of gastrointestinal trichostrongylids in small ruminants.  相似文献

A study was conducted to evaluate and compare the efficacy of two injectable formulations of ivermectin (IVM-1 and IVM-2) at a dose rate of 0.3 mg/kg bodyweight versus placebo in the treatment and control of larval and adult stages of Ascaris suum and Oesophagostomum spp. in experimentally infected pigs. Seventy helminth free pigs were allocated on a liveweight basis to 7 groups each comprising 10 pigs (A-G). Group A served as an untreated control group. Groups B and C were used to investigate the efficacy of both formulations against adult stages of A. suum and Oesophagostomum spp., Groups D and E for efficacy against larval stages of A. suum and Groups F and G for efficacy against larval stages of Oesophagostomum spp. Pigs of groups A, B, C, D and E were infected on Day-0 with 1000 infective A. suum eggs each. Infective larvae of Oesophagostomum spp. (10,000/pig) were given on Day-0 to pigs of Groups F and G and on Day-21 to pigs of Groups A, B and C. Treatment was given to pigs of Group A (saline as placebo) on Day-7 and -28, IVM-1 to pigs of Group F on Day-7, pigs of Group D on Day-14 and pigs of Group B on Day-49. IVM-2 was given to pigs of Group G on Day-7, Group E on Day-28 and Group C on Day-49. Pigs of Groups F and G were sacrificed on Day-28, pigs of Groups A, D and E on Day-49 and pigs of Groups B and C on Day-56. Post mortem worm counts showed the following efficacies: (IVM-1) against larval A. suum 100%, against adult A. suum 94.4%, against larval Oesophagostomum spp. 52.0% and against adult Oesophagostomum spp. 83.0%. (IVM-2) against larval A. suum 100%, against adult A. suum 90.3%, against larval Oesophagostomum spp. 94.0% and against adult Oesophagostomum spp. 94.7%.  相似文献

Five of a group of six calves were inoculated with Mycobacterium bovis. Two more uninoculated calves were introduced to the group 84 days later. All the inoculated calves were subsequently shown to be excreting M bovis in nasal mucus. The uninoculated calf in the initial group of six became infected and subsequently excreted M bovis. The two uninoculated calves which were introduced later did not become infected. It was concluded that contact with nasal mucus from the infected cattle resulted in infection of the uninoculated calf and that the density of accommodation of animals excreting M bovis was an important factor in transmission of the disease.  相似文献

The aim of this investigation was to determine the efficacy of milbemycin oxime in preventing the oesophageal encapsulation of Spirocerca lupi, following the experimental infection of dogs. Two studies were conducted which involved a total of 21 purpose-bred Beagles. Each dog was infected with approximately 40, third stage infective S. lupi larvae. The larvae were dissected from scarabaeid beetles that had been collected from areas endemic for spirocercosis. In the first study, milbemycin oxime (minimum dose 0.5mg/kg body weight) was administered to seven dogs on day 30 post-infection. Seven other dogs served as untreated controls. In the second study, milbemycin oxime (also at a minimum dose of 0.5mg/kg body weight) was administered to four of seven infected dogs on day 28 post-infection. Treatment was repeated at 14- or 28-day intervals. All of the dogs, from both studies, were euthanized 168 or 169 days after infection. All S. lupi were recovered, and lesions in the thoracic aorta and oesophagus were described and quantified. A single treatment with milbemycin oxime was 79.8% effective in preventing the establishment of S. lupi in the oesophagus. This treatment significantly (p<0.05) reduced both the number of S. lupi within the oesophagus and the size of the oesophageal nodules. The efficacy of anthelmintic treatment was increased to 100% when repeat doses of milbemycin oxime were administered at 14- or 28-day intervals. These repeat treatments completely prevented the establishment of S. lupi within the oesophagus and thereby averted the development of oesophageal nodules. As expected, none of the treatment protocols reduced S. lupi related damage within the aorta because the administration of milbemycin oxime only began after the larvae had completed their first stage of migration.  相似文献

The suppression of nematode egg output in faeces was measured in ewes treated just before lambing with either oxfendazole or ivermectin by oral drench or with ivermectin by subcutaneous injection. Ivermectin and oxfendazole given orally were similarly effective, whereas ivermectin given by subcutaneous injection extended the period of suppressed egg output by about one week. The more persistent anthelmintic effect of ivermectin given subcutaneously was probably due to its extended half-life in the plasma of treated sheep. Plasma pepsinogen activity was less in the sheep given anthelmintic than in the untreated controls. Ivermectin caused a significantly greater reduction in pepsinogen activity than oxfendazole and was more effective when given subcutaneously than when given orally.  相似文献

The concentrations of ivermectin in the gastrointestinal tract of sheep and cattle were determined after subcutaneous administration of ivermectin. Ivermectin was not detected (limit of detection 1 ng/ml) in abomasal and ruminal fluids either after a normal therapeutic dose of 200 micrograms/kg or even at an increased dose of 2000 micrograms/kg. It was also not detected in abomasal and ruminal fluids of a sheep infected with the abomasal parasite Ostertagia circumcincta. However, ivermectin was detectable at similar concentrations in abomasal mucus and in small intestinal mucus. Excretion of ivermectin was high in bile but the concentrations in small intestinal mucus, distal and proximal to the bile duct opening, were similar. It is hypothesized that the low efficacy of ivermectin against small intestinal nematodes compared with abomasal nematodes is not due to differences in ivermectin concentrations in the predilection sites but is probably due to tachyphylaxis in the nematodes allowing the small intestinal nematodes to re-establish before they have left their predilection site. Ivermectin was excreted in the milk of ewes at concentrations similar to those in plasma. Lambs suckling ivermectin-treated ewes received about 4% of a normal therapeutic dose (200 micrograms/kg) via the milk.  相似文献

The anthelmintic activity of cambendazole 31% paste was evaluated in a controlled trial against naturally acquired gastro-intestinal parasites of the horse. Four groups of five adult horses were treated at dose rates of 0, 15, 20 and 25 mg/kg. The three dose levels of cambendazole tested were efficacious against the large strongyles Strongylus edentatus and S. vulgaris, the stomach worm Trichostrongylus axei, the pinworms Oxyuris equi and Probstmayria vivipara and all the species of small strongyles present. No efficacy was demonstrated against the tapeworm Anoplocephala perfoliata or the horse bots Gasterophilus intestinalis or G. nasalis  相似文献

A 'dipstick' immunoassay for bovine cysticercosis, using an antigen isolated from Taenia hydatigena cyst fluid, was evaluated in cattle experimentally infected with Taenia saginata. The assay correctly identified six out of seven infected cattle, including an animal in which only 12 living cysticerci were found. Cattle became seropositive as early as 3 weeks post-infection. A false-negative reaction was found for one very lightly infected animal, from which only four living cysticerci were recovered at necropsy. The assay was also used to detect circulating antibodies in experimentally infected cattle before and after therapeutic treatment with anthelminthics. The results suggest that praziquantel-treated animals gradually revert to being seronegative after the cysticerci are killed.  相似文献
