Moldy corn in diets of gestating and lactating swine   总被引:1,自引:0,他引:1  
Moldy corn was substituted for normal corn in four dietary treatments to provide 0, 2.1, 3.7 and 4.8 ppm of zearalenone. The diets were fed to gilts from the day after first observed estrus through two gestation-lactation periods. Each diets was fed to 11 gifts initially. Overall reproductive performance appeared to decrease as the level of moldly corn in the diet increased, with fewer sows completing two parities and fewer piglets weaned per litter, especially from sows fed the highest level of moldy corn. Three-week-old gilts that nursed sows fed the highest level of moldy corn had increased weight of reproductive tracts. In 35-d-old gilts, symptoms of estrogenism increased as the level of moldy corn in the diet increased, even though there were no symptoms of estrogenism in the mature gilts or sows fed the same diets. Neither zearalenone nor its metabolites (alpha- and beta-zearalenol) were detected in milk obtained 1 or 14 d postpartum.  相似文献

The main arguments, discussed by different international organizations, which confirm the significance of problems connected with the presence of zearalenone in animal feed materials and their influence on the safety of food materials are shown in this review article. The main world research trends focused on zearalenone, as undesirable substances, are also presented. A variety of Fusarium fungi produce a number of different mycotoxins, for example zearalenone. Is not true that Fusarium fungi are they the most prevalent toxin-producing fungi only in the northern temperate regions, are also commonly found on cereals grown in the temperate regions of America, Europe and Asia. Zearalenone have been shown to cause a variety of toxic effects in both experimental animals and livestock, and have also been suspected of causing toxicity in humans. Zearalenone is a stable compound, both during storage/milling and the processing/cooking of food, and it does not degrade at high temperatures. Studies of metabolism indicate that zearalenone is fairly rapidly absorbed following oral administration, with the formation of alpha- and beta-zearalenol and alpha- and alpha-zearalanol, which are subsequently conjugated with glucuronic acid. This mycotoxin and some of these metabolites have been shown to competitively bind to estrogen receptors in a number of in vitro systems. Binding to specific receptors has been demonstrated in the uterus, mammary gland, liver and hypothalamus of different species. Additionaly alterations of immunological parameters were found at high zearalenone concentrations in vitro. Zearalenone causes alterations in the reproductive tract of laboratory animals and domestic animals. Various estrogenic effects such as decreased fertility, increased embryolethal resorptions, reduced litter size, changed weight of adrenal, thyroid and pituitary glands and change in serum levels of progesterone and estradiol have been observed, and teratogenic effects were found in pigs and sheep. It may be that the safety of zearalenone could be evaluated on the basis of the dose which had no hormonal effects in pigs, the most sensitive species, and a temporary Tolerable Daily Intake for zearalenone of 0.2 microgram/kg of body weight, could be established.  相似文献

The effects of mycotoxin zearalenone and their major metabolites alpha- and beta-zearalenol on spontaneous contractions in isolated lamb uterine smooth muscle were examined. The study was carried out on 20 female prepubertal lambs aged between 45 and 50 days. Myometrial strips were set up in two isolated organ baths (10ml) at 37 degrees C and were exposed to increasing concentrations (10(-11)M-10(-6)M) of these mycoestrogens and results were compared with the effect, at the same concentrations, of natural estrogen 17beta-estradiol. Our findings suggest that mycotoxins and 17beta-estradiol, at nanomolar concentrations, rapidly enhance phasic spontaneous smooth muscle contraction. In particular, zearalenone increases the uterine activity similarly to 17beta-estradiol. On the contrary, its metabolite alpha-zearalenol significantly inhibits myometrial contractility.  相似文献

A sensitive bioassay for detection of dietary estrogens in animal feeds   总被引:3,自引:0,他引:3  
Estrogen-responsive proliferation in the MCF-7 cell line was used as a bioassay for detection of dietary estrogens. The bioassay procedure was adapted to screen for estrogenic activity in feedstuffs that have been associated with hyperestrogenism in livestock. Methanolic feed extracts were added to the cell culture medium at microliter/ml concentrations for 4 days, after which the cell proliferation response was measured as DNA content. The half-maximal response for estradiol occurred at 2 pM, or 0.54 pg/ml. For zearalenone, a weaker estrogen, the half-maximal response occurred at approximately 200 pM, or 64 pg/ml. The bioassay was calibrated against a number of known estrogens (estradiol, diethylstilbestrol, zearalenone, zearalanol [cattle implant], beta-zearalenol, zearalane), including the naturally occurring phytoestrogens (formononetin, genistein, daidzein, biochanin A, and coumestrol). The estrogenic activity of feed samples was expressed as equivalents of zearalenone (ppm zearalenone) that would have to be present to equally stimulate proliferation of the MCF-7 cells. The sensitivity of the bioassay was 0.05-0.1 ppm equivalents of zearalenone in feed, well below the threshold level associated with reproductive problems. The feed additive melengestrol acetate (MGA) showed no estrogenic activity in this assay. Estrogenic activity of feed extracts was confirmed by competitive inhibition with the antiestrogens tamoxifen or LY156758 (keoxifene) to show that stimulation of growth by feed extracts was through an estrogenic mechanism. Confirmation of known estrogens was by tandem mass spectroscopy. The assay is a sensitive and reliable screening procedure for detecting estrogenic activity in feedstuffs.  相似文献

We investigated the effects of in vivo exposure to low zearalenone levels on the anti‐Müllerian hormone endocrine levels and the reproductive performance of cattle. Urine and blood samples and reproductive records were collected from two Japanese Black breeding female cattle herds with dietary zearalenone contamination below the threshold levels (<1 ppm) at 30 days after calving. Urinary zearalenone, α‐zearalenol and β‐zearalenol concentrations were measured by chromatography‐tandem mass spectrometry, and serum anti‐Müllerian hormone concentrations were determined along with serum biochemical parameters. Urinary concentrations of α‐zearalenol were significantly higher (p < 0.05) in cattle in Herd 1 than in cattle in Herd 2, reflecting the different amounts of zearalenone in the diet of the two herds. Although the number of 5‐mm and 10‐mm follicles of the herds and their fertility after artificial insemination were similar, the serum anti‐Müllerian hormone concentrations in herds 1 and 2 were 438.9 ± 48.6 pg/ml and 618.9 ± 80.0 pg/ml, respectively, with a trend towards a significant difference (p = 0.053), which may indicate differences in the antral follicle populations between herds. Thus, zearalenone intake from dietary feed, even when below the threshold zearalenone contamination level permitted in Japan, may affect the ovarian antral follicle populations, but not the fertility, of post‐partum cows.  相似文献

Bioactivation of zearalenone by porcine hepatic biotransformation   总被引:1,自引:0,他引:1  
Zearalenone (ZEA) is a resorcylic acid lactone derivative produced by various Fusarium species that are widely found in food and feeds. The structure of zearalenone is flexible enough to allow a conformation able to bind to mammalian oestrogen receptors, where it acts as an agonist. Using oestrogen-dependent Human Breast Cancer (MCF-7) cells, the oestrogenic activity of zearalenone and its derivatives were compared using 17 beta-oestradiol as a positive control. The results obtained demonstrate that the oestrogenic potency of ZEA derivatives could be ranked in the following order: alpha-zearalenol > alpha-zearalanol > zearalenone > beta-zearalenol. Since pigs have been reported to be among the most sensitive animal species, biotransformation studies with pig liver subcellular fractions were conducted. These studies indicated that alpha-zearalenol is the main hepatic metabolite of zearalenone in pigs, and it is assumed that 3 alpha- and 3 beta-hydroxysteroid dehydrogeneases are involved in the hepatic biotransformation, since the formation of alpha-zearalenol and beta-zearalenol could be inhibited by prototypic substrates for either enzyme. The bioactivation of ZEA into the more active alpha-zearalenol seems to provide a possible explanation for the observed high sensitivity of pigs towards feeding-stuffs contaminated with the mycotoxin.  相似文献

1. Diets with increasing proportions of Fusarium-toxin-contaminated wheat were fed to Pekin ducks for 49 d in order to titrate the lowest effect level. Dietary deoxynivalenol (DON) and zearalenone (ZON) concentrations were successively increased up to 6 to 7 mg/kg and 0.05 to 0.06 mg/kg, respectively. 2. Feed intake, live weight gain and feed to gain ratio were not influenced by dietary treatment. 3. Gross macroscopic inspection of the upper digestive tract did not reveal any signs of irritation, inflammation or other pathological changes. The weight of the bursa of Fabricius, relative to live weight, decreased in a dose-related fashion. Activities of glutamate dehydrogenase and gamma-glutamyl-transferase in serum were either unaffected or inconsistently affected by dietary treatments. 4. Concentrations of DON and of its de-epoxydised metabolite in plasma and bile were lower than the detection limits of 6 and 16 ng/ml, respectively, of the applied high performance liquid chromatography (HPLC) method. 5. ZON or its metabolites were not detectable in plasma and livers (detection limits of the HPLC method were 1, 0.5 and 5 ng/g for ZON, alpha-zearalenol (alpha-ZOL) and beta-zearalenol (beta-ZOL), respectively). Concentrations of ZON, alpha-ZOL and beta-ZOL in bile increased linearly with dietary ZON concentration. The mean proportions of ZON, alpha-ZOL and beta-ZOL of the sum of all three metabolites were 80, 16 and 4%, respectively. 6. Taken together, it can be concluded that dietary DON and ZON concentrations up to 6 and 0.06 mg/kg, respectively, did not adversely affect performance and health of growing Pekin ducks.  相似文献

There is considerable evidence that exogenous estrogenic compounds can have adverse effects on fertility. The main reason cited in literature for hyperestrogenism in pigs is contamination of feedstuffs by the mycotoxin zearalenone (Boehm, 2000), but further estrogenically active substances might also be involved in cases of impaired fertility with symptoms like enlarged, red-coloured vulvae in piglets, irregular estrus cycles and anestrus of sows (Bennetts et al., 1946; Drane et al., 1981). It is well known that soy used in diets for pigs as a main protein source contains phytoestrogens. Amongst them, isoflavones like genistein and daidzein are of particular interest. Aim of this study was to optimize and use an established bioassay (Kluczka, 2003) to determine estrogenic activity in feedstuffs for pigs related to isoflavones and further substances with estrogenic potential. This bioassay is a reporter gene assay based on stably transfected human embryonal kidney cells (HEK 293) that contains either alpha or beta estrogen receptor (alpha- or beta-HEK). The estrogenic activity measured in the luciferase assay was expressed in estradiol-equivalents (EEQ) and the results were compared with the isoflavone content (genistein, daidzein) obtained by chemical analysis using high performance liquid chromatography-Ultraviolet (HPLC-UV). Mean estrogenic activity in diets fed to sows in herds with altered fertility was 275.8 microg EEQ/kg feed in alpha-HEK cells and 295.0 microg EEQ/kg feed in beta-HEK cells. Feedstuffs from herds without any altered fertility showed an average estrogenic activity of 204.9 microg EEQ/kg feed in alpha-HEK and 213.3 microg EEQ/kg feed in beta-HEK. The estrogenic activity was strongly related to the concentration of the isoflavones (alpha-HEK, r(2)=0.9488; beta-HEK, r(2)=0.9427). Clinically relevant zearalenone concentrations (>50-150 microg/kg feed) displayed estrogenic effects in the bioassay that did not differ significantly from those caused by high isoflavone concentration because of the use of soy as protein source.  相似文献

Concentrations of 25, 50, or 100 ppm of 95% purified zearalenone fed to groups of healthy, multiparous sows during preestrus or throughout the gestation period (or both) produced multiple reproductive deficiencies. These reproductive disorders included infertility, constant estrus, pseudopregnancy, diminished fertility, reduced litter size, smaller offspring, malformation, juvenile hyperestrogenism, and probably fetal resorption. Gross and histologic examinations of sows revealed lesions in the reproductive organs. Marked epithelial changes characterized by squamous metaplasia were noticed in the uterus, uterine duct, cervix, vagina, and mammary glands.  相似文献

抽选2~9胎次的皖南花哺乳母猪6头,每头母猪带仔10~14头,有效乳头数7对。试验母猪日粮含DE11.72MJ/kg和CP12%,取日粮2.5~3.0千克,按12与青绿多汁饲料混合打匀投喂。供试母猪自分娩开始,每5天进行一次泌乳量测定,直到产后泌乳50天。利用线性模型校正不同胎次和带仔数的泌乳量,再采用一元高次方程进行泌乳曲线分析。结果表明,皖南花猪泌乳高峰期在分娩后的第10~15天,其间平均泌乳量为5.66~5.91kg/天,分娩后的第35天泌乳量为3.7kg/天。不同乳头的泌乳量为第1、2和3号乳头最高,平均每个单乳头泌乳380~385g/天,向后乳头的泌乳量呈下降趋势,第7号乳头泌乳量为300.5g/天。不同泌乳阶段表现出明显的节律性,泌乳曲线方程的拟合度均在0.90以上。  相似文献

To study the relationship between on-farm welfare and reproductive performance in the sow, the TGI 35L Animal Needs Index was modified for use in Finnish pig production. The modified index had a maximal total score of 100. It was comprised of six categories: 'locomotion' (maximal score 21 for dry sows and 11 for lactating sows), 'social interaction' (12/8), 'floor quality' (16/9), 'stable climate' (16/21), 'feeding' (16/23) and 'health and stockmanship' (19/29). Index scorings were performed separately in farrowing, breeding and gestation units on 28 representative Finnish sow farms. One-year production parameters were collected. Multiple linear regression was used for statistical analysis. Total ANI-points varied between 36.5-68.0 for lactating and 39.5-86.0 for dry sows. Litter size increased with increasing scores for 'feeding' in the dry sow unit. Controlling for breed, high scores for 'health and stockmanship' and 'floor quality' shortened the reproductive cycle, probably because of good leg health. The number of weaned piglets per sow per year (PSY) increased with increasing scores for dry sow 'health and stockmanship', 'floor quality' and an interaction of 'feeding' in the farrowing and mating units. PSY increased with decreasing scores for farrowing pen 'climate'. High-quality floors and stockmanship appear to correlate positively with reproductive performance in the sow. Effects of a welfare-promoting feeding strategy on reproduction are contradictory.  相似文献

Sixteen primiparous sows were bred and fed either a control ration (n = 8) or a diet containing purified zearalenone (n = 8; 1 mg/kg of body weight) from days 7 to 10 after breeding. On day 7 after breeding, the jugular vein of each sow was cannulated and blood was collected at 20-minute intervals for 4 hours before feeding and 4 hours after feeding. On day 10 after breeding, blood samples were collected from 4 control sows and 4 zearalenone-fed sows at 20-minute intervals for 4 hours before collection of blastocysts. A similar blood sampling schedule was followed for the remaining 4 control and 4 zearalenone sows on day 14 after breeding. On day 10 after breeding, spherical blastocysts were recovered from all control sows and from 3 of 4 zearalenone-treated sows. Average diameter of blastocysts from zearalenone-treated sows were similar to that of control sows. On day 14 after breeding, blastocysts were recovered from all control sows and 3 of 4 zearalenone-treated sows. Blastocysts from the control sows were filamentous, whereas blastocysts from zearalenone-treated sows were fragmented and contained foci of necrosis. Incidence of luteinizing hormone (LH) secretory spikes per sow was less (P less than 0.01) in zearalenone-treated sows (0.25 +/- 0.25/4 h) than control sows (1.75 +/- 0.25/4 h) on day 10 after breeding. In sows, a strong relationship exists between body condition and reproductive efficiency and milk yield. Leptin may act as a metabolic gate which permits the activation of reproductive axis: in the sow, serum concentration of leptin was positively correlated with adiposity at farrowing. An interesting aspect useful to clarify the biology of leptin, was the discovery that the placenta expresses the ob gene, the ob receptor gene and it is a site of leptin production, suggesting a possible role of the hormone in fetal growth; after birth, the placenta functions were taken over from milk, especially to the delivery of maternal hormones and growth factors to the neonate. The exact role of maternal leptin in the physiology of neonatal piglets remains to be determined. Our aim was to evaluate if maternal leptin levels at the beginning of lactation and at weaning could predict the resumption of cycle activity and/or the piglet gain. Thirty-eight Large White × Landrace pregnant sows (16 nulliparous and 22 pluriparous) were used. Blood samples were taken from sows and piglets at d 5 and d 21 after farrowing; in the same days, milk samples were taken after oxytocin injection by means of complete manual milking of all mammary glands of one side. On the basis of the blood leptin at d 5, sows were divided into 3 groups (Low: < 2.3 ng/ml; Medium: 2.3 to 2.6 ng/ml; High: > 2.6 ng/ml). Our results show a correlation at d 5 between backfat thickness and blood leptin (r = 0.342; P < 0.05). The resumption of the cyclic activity was faster in sows with a leptin level at d 5 greater than 2.3 ng/ml (P < 0.01). Milk composition at d 5 and 21 was not affected by parity and leptin. Piglet ADG was significantly (P < 0.05) influenced by sow leptin groups (0.180 kg day− 1 for piglets from Low group and 0.224 for High group). Piglets weaned by High group sows have shown a greater blood leptin content at weaning (P < 0.01) than other groups. In conclusion we have found a significant correlation between leptin and productive and reproductive performances of pigs. This paper underlines the pleiotropic actions exerted by leptin in the productive sow.  相似文献

Sows of differing parities and genetics were used at different locations to determine the effects of feeding added L-carnitine during lactation on sow and litter performance. In Exp. 1, sows (n = 50 PIC C15) were fed a lactation diet (1.0% total lysine, .9% Ca, and .8% P) with or without 50 ppm of added L-carnitine from d 108 of gestation until weaning (d 21). No differences in litter weaning weight, survivability, sow ADFI, or sow weight and last rib fat depth change were observed. Number of pigs born alive in the subsequent farrowing were not different (P>.10). In Exp. 2, parity-three and -four sows (n = 115 Large White cross) were used to determine the effect of feeding 0, 50, 100, or 200 ppm of added L-carnitine during lactation (diet containing .9% total lysine, 1.0% Ca, and .8% P) on sow and litter performance. No improvements in the number of pigs or litter weights at weaning were observed (P>.10). Sows fed added L-carnitine had increased weight loss (linear; P<.04), but no differences (P>.10) were observed in last rib fat depth change or subsequent reproductive performance. In Exp. 3, first-parity sows (n = 107 PIC C15) were fed a diet with or without 50 ppm of added L-carnitine during lactation (diet containing 1.0% total lysine). Sows fed added L-carnitine tended (P<.10) to have fewer stillborn and mummified pigs than controls (.42 vs .81 pigs). No differences were observed for litter weaning weight, survivability, or subsequent farrowing performance. Feeding 50 to 200 ppm of added L-carnitine during lactation had little effect on sow and litter performance.  相似文献

A marker indicative of the fertility potential of replacement gilts early in development would decrease culling rates in the sow herd, improve sow herd reproductive efficiency, and reduce production costs. The objective of this study was to determine if vaginal lipid profiles at 21 d postnatal (PN) could predict sow reproductive performance. Vaginal swabs of the anterior vagina were taken at 21 ± 4 d PN from gilts born on a commercial sow production facility for lipidomic analysis. Animals were followed prospectively for 2 yr and assigned to reproductive performance categories based on the observation of estrus or piglets weaned per sow per year (PSY) across two farrowings. Lipids were extracted from cellular material collected with swabs taken from high fertility (HF; n = 28; ≥26 PSY) and infertile (IF; n = 34; no estrus, no pregnancy) animals, and multiple reaction monitoring profiling was used for lipidome analysis. The relative abundance of arachidonic acid (C20:4) and docosahexaenoic acid (C22:6) was lower (P < 0.05) in IF gilts than HF gilts, whereas the abundance of the free fatty acids such as cerotic (C26:0), ximenic (C26:1), and nonadecanoic (C19:0) acids was greater (P < 0.05) in IF gilts. Additionally, eicosapentaenoic acid (C20:5), a precursor of prostaglandins, was higher (P < 0.05) in IF gilts. The perspective of having a panel of lipids captured with vaginal swabs at weaning that can predict the reproductive efficiency of gilts shows promise and warrants future research in this area.  相似文献

A dose response study was carried out with piglets to examine the effects of increasing amounts of Fusarium toxins in the diet on performance, clinical serum characteristics, organ weights and residues of zearalenone (ZON) and deoxynivalenol (DON) and their metabolites in body fluids and tissues. For this purpose, Fusarium toxin contaminated maize (1.2 mg ZON and 8.6 mg DON per kg maize) was incorporated into a maize based diet for piglets at 0, 6, 12.5, 25 and 50% at the expense of control maize. The experimental diets were tested on 100 female piglets allotted to 20 boxes (five animals per box) covering a body weight range of 12.4 +/- 2.2 kg to 32.5 +/- 5.6 kg. Voluntary feed intake and, consequently, body weight gain of the animals receiving the highest proportion of Fusarium toxin contaminated maize were significantly decreased while the feed conversion ratio was not affected by the treatment. The mean weight of the uterus related to the body weight of the animals of the same group was increased by almost 100% as compared to the control. For this group, significantly decreased values of total serum protein were determined, while the serum activity of the liver enzyme glutamate dehydrogenase and the serum concentration of the follicle stimulating hormone were decreased for all treatment groups receiving 6% contaminated maize or more in the diet. Serum concentrations of immuneglobulins were not consistently altered by the treatment. Corresponding to the dietary exposure, increasing concentrations of ZON and alpha-zearalenol were detected in the bile fluid, liver and in pooled urine samples. The metabolite beta-zearalenol was detected only in bile fluid. The total concentration of ZON plus its metabolites in bile fluid correlated well with the diet contamination (r = 0.844). DON was found in serum, bile fluid and pooled urine samples while de-epoxy-DON was detected only in urine. The serum concentration of DON correlated well with the respective toxin intake 3-4 h prior to slaughtering (r = 0.957). For all mentioned analyses of residues it has to be noted that toxin residues were detectable even if negligible concentrations were present in the diet.  相似文献

1. Diets with increasing proportions of Fusarium toxin-contaminated wheat (0, 170, 340 and 510 g CW/kg) were fed to male turkeys (BUT Big 6) from d 21 to d 56 of age. Each diet was tested with or without a non-starch-polysaccharide (NSP) hydrolysing enzyme preparation. Dietary deoxynivalenol (DON) and zearalenone (ZON) concentrations were successively increased up to approximately 5.4 and 0.04 mg/kg, respectively. 2. Weight gain decreased slightly with increasing proportions of CW, by 1.6, 0.7 and 3.6%, whereas other performance parameters remained unaffected. NSP enzyme supplements to the diets had no influence. 3. The weight of the emptied jejunum plus ileum, relative to live weight, decreased in a dose-related fashion whereby the NSP enzyme exerted an additional weight-decreasing effect. A similar weight-decreasing NSP enzyme effect was noted for heart weights. Activity of glutamate dehydrogenase in serum was significantly increased in groups fed the diets with the highest CW proportion, whereas gamma-glutamyl-transferase remained unaltered. 4. Viscosity in the small intestine was significantly reduced by supplementing the diets with the NSP enzyme. This effect successively decreased with increasing proportions of the CW. 5. Concentrations of DON and of its de-epoxidised metabolite de-epoxy-DON in plasma, liver and breast meat were lower than the detection limits of 2 ng/ml (plasma) and 4 ng/g, respectively, of the applied HPLC method. DON concentration in bile reached up to 13 to 23 ng/ml whereas de-epoxy-DON concentration was lower than 4 ng/ml. 6. ZON or its metabolites were not detectable in plasma, liver or breast meat (detection limits of the HPLC method were 1, 0.5 and 5 ng/g for ZON, alpha-zearalenol (ZOL) and beta-ZOL, respectively). Concentrations of ZON and alpha-ZOL in bile increased with dietary ZON concentration. The mean proportions of ZON, alpha-ZOL and beta-ZOL of the sum of all three metabolites were 19, 77 and 4%, respectively.  相似文献

2012年4月,山东某规模猪场暴发疾病,母猪受胎率、产仔数、健仔数等繁殖性能指标显著下降,猪群表现出不同程度的霉菌毒素中毒症状。经初步调查,发现该猪场所饲喂的饲料被霉菌毒素污染,进一步采取原料玉米及饲料样品通过高效液相色谱法鉴定霉菌毒素种类及含量,结果显示,原料玉米中赤霉烯酮、呕吐毒素及烟曲霉毒素含量均超标。经更换饲料和采用脱霉剂后,母猪繁殖性能指标显著恢复。  相似文献
