In order to study the possibility of gaining commercial benefit from culturing an excess of one sex of the Pacific oyster (Crassostrea gigas), comparative data on the growth rate and condition of male and female oysters are reported. Historically, measurement of sex‐specific growth rate in oysters has been overlooked or confounded by protandric sex. The recent conclusion that the sex ratio of Pacific oysters is predominantly under genetic rather than environmental control introduces the possibility of manipulating sex ratio for commercial gain if they exhibit asynchronous sex‐specific growth rates. Pacific oysters were cultured intertidally in Smoky Bay, south Australia. The observations, made over the 7‐month gametogenic cycle from August to February to ensure no sex reversal, were of growth rates of male and female oysters and ambient chlorophyll a concentrations. Mean shell growth of female oysters was significantly faster than that of males (4.5 ± 3.3 compared with 3.8 ± 3.2 μm day?1 mm?1 total length). Sex‐specific asymmetries in length and weight were generally significant and increased in magnitude during the 7‐month study period, suggesting potential commercial benefits from increasing the proportion of cultured female oysters. The fastest increase in the sex‐specific disparity in growth and condition came after the October chlorophyll a peak, suggesting that females utilize blooms more efficiently than males. Our results compare favourably with methods currently used to increase oyster growth (e.g. triploidy can provide growth gains of 13–51%).  相似文献

Additions of bacteria (strain CA2) as a food supplement to xenic larval cultures of the oyster Crassostrea gigas consistently enhanced growth of larvae during different seasons of the year. Bacterial enhancement of larval growth occurred when either Isochrysis galbana (ISO), I. aff. galbana (T-ISO) or Pseudoisochrysis paradoxa (VA-12) were used as algal foods. Additions of CA2 bacteria at 105 cells ml−1 to cultures of algal-fed larvae increased larval growth, the proportion of larvae that set to produce spat, and the subsequent size of spat. A lower proportion of slow-growing larvae in populations receiving additions of CA2 bacteria compared with populations of larvae fed only algae, suggests a bacterial nutritional contribution to larval growth. Manipulation of bacterial populations present in bivalve larval cultures is a potentially useful strategy for the enhancement of oyster production.  相似文献

The Pacific oyster industry in Australia is derived from importations from Japan in the late 1940s and early 1950s to Tasmania and is almost completely hatchery based. This makes it a good target for developing and deploying genetically improved strains. An allozyme survey comparing hatchery stocks with self‐recruiting Tasmanian stocks and with two collections from Japan found abundant variation and no significant evidence of allele loss. The subsequent selection programme (initiated in the summer of 1996/97) had several strands. We wanted to take advantage of the increased power that marker‐assisted selection could bring and, therefore, needed to develop a linkage map and isolate flanking markers around quantitative trait loci (QTLs). Several types of markers (allozymes, microsatellites and AFLPs) were used, and single‐pair crosses were set up; QTLs have been detected. Conventional selection programmes, one based on mass selection and one on family selection, have been established. Triploid Pacific oysters produced via chemical means have been available for several years, but rates of triploidy achieved by such means are usually less than 100%. In 1999, we will assess whether our tetraploid 2‐year‐old broodstock can be crossed with diploids to give 100% triploid offspring.  相似文献

The robustness of Pacific oyster, Crassostrea gigas (Thunberg), sperm cryopreservation in the context of selective breeding based on family lines was investigated. Irrespective of egg density, high fertilization success was achieved with cryopreserved sperm when sperm:egg ratios of 1000:1 to 10 000:1 were used. Variation among replicate runs on the same oyster batches was minimal, indicating that cryopreservation and larval rearing procedures were repeatable. Twenty independent single male–female crosses were made to assess the utility of cryopreserved sperm in selective breeding. The fertility of unfrozen sperm was generally a poor predictor of cryopreserved sperm fertility. Based on D‐larval yields, 17 of the 20 crosses were likely to yield adequate spat for selective breeding (>105 D‐larvae from 1 million eggs), two were marginal (5 × 104 D‐larvae) and one was inadequate (4 × 103 D‐larvae). An alternative fertilization strategy to improve D‐yield from a given number of sperm was then tested. Fertilizing 10 million eggs at a sperm:egg ratio of 200:1 increased the total D‐yield when compared with fertilizing 1 million eggs at a sperm:egg ratio of 2000:1 for the same male–female pair. We conclude that, despite wide variation in fertility, cryopreserved sperm is useful for family production.  相似文献

Abstract. A habitat suitability index (HSI) model for suspended tray culture of the Pacific oyster, Crassostrea gigas Thunberg, was constructed from existing information in the literature on oyster-environment relationships. In the model, biophysical data are used to rate aquaculture potential of coastal areas on a scale from 0.0 to 1·0. where 1·0 represents optimal conditions for growth and survival and 0·0 represents totally unsuitable habitat conditions. The model was tested with environmental, growth (increase in shell height) and secondary production (g· 100 oysters−1 day−1) data collected over a 14-month period for two age-classes of oysters transplanted to 10 locations along the coast of British Columbia. Canada. Regressions of HSI values calculated from entire study environmental data against oyster growth and secondary production were highly significant for both age-classes. Environmental data subsets representing short sampling programmers (1–3 months) resulted in HSI values significantly correlated to oyster growth, provided that some sampling occurred during periods of high food availability. HSI modeling techniques have potential application in site selection and coastal management of aquaculture.  相似文献

The Pacific oyster Crassostrea gigas is a representative bivalve mollusc that is widely cultured in the world and is the largest molluscan group cultured in China. In order to assess the feasibility of improving survival of C. gigas through genetic selection, the heritability and genetic correlations for growth and survival traits between different life stages were examined. Genetic parameters were estimated based on intraclass correlations of 49 full‐sib families (29 half‐sib families) in larvae (4 and 20 days after fertilization) and spat (140 days after fertilization) stages. The heritability for growth traits in larvae and spat was 0.30–0.86 and 0.53–0.59, respectively, and varied with ages. The heritability of survival was low in larvae (0.13 ± 0.05 and 0.17 ± 0.04, respectively for 4 and 20 days after fertilization) but medium (0.39 ± 0.07) in spat, suggesting that selection for increasing spat survival was feasible. The genetic correlation between growth traits within age was medium to high and positive (ranging from 0.47 to 0.96, respectively, between shell length (SL) and shell height (SH) at 20 days and between SL and SH at 140 days after fertilization), suggesting that selection to improve single growth trait will cause positive response in another growth traits in C. gigas. The genetic correlations between survival and growth traits at 140 days were low but positive (ranging from 0.23 to 0.27, respectively, between survival and SH and between survival and SL at 140 days after fertilization), suggesting that selection for survival may not have a negative response in growth. Overall, this study suggests that survival traits should be taken as improving target of next selection breeding programme in C. gigas.  相似文献

Abstract. Growth rates, condition indices and diet composition of the Japanese oyster, Crassostrea gigas Thunberg, were studied in two types of ponds which form part of a fish-bivalve integrated culture system. Although abiotic parameters (e.g. temperature, salinity, pH, ammonia, particulate inorganic matter) were similar in the two pond types, oysters supplied with water from a sedimentation pond grew significantly faster and showed better condition indices than the oysters supplied with water from the PVC-lined ponds. It is suggested that the main reasons for the better performance of the oysters supplied with water from sedimentation pond water are: higher algal diversity, additional nutritious Food consisting of attached benthic diatoms and stable algal concentration.  相似文献

Temperature and quality of the available food are important factors that influence the physiology of oysters; however, the combined effects have not been well studied. We evaluated the impacts of the temperature and diet on the growth, survival and biochemical composition in the Pacific oyster Crassostrea gigas spat, cultured in the laboratory for 8 weeks at 23, 26, 29 and 32°C and fed Isochrysis sp.‐Pavlova lutheri (IP) and Dunaliella tertiolecta (Dt). The growth and biochemical composition showed a pattern, which changed in response to rising temperature. The shell length was significantly longer, in spat fed the IP diet, except at 32°C, where both diets produced poor growth results. The survival was <50% after 5 weeks at 32°C, whereas at all other temperatures it was >88%. High temperatures directly increased lipids and saturated fatty acids, while the proteins, carbohydrates and unsaturated fatty acids decreased. High temperatures achieved in the environment, as those reached on clear summer days during low tides, are an important stressor in oyster spat, especially when the quality of the available food is poor.  相似文献

A technique is described in which irrigation of the mantle cavity of oysters, Crassostrea gigas, induces pumping at steady rates. Simultaneous net influx of 10 amino acids (asp, glu, asn, ser, gly, tau, ala, val, phe, lys) is observed by high-performance liquid chromatography (HPLC). Individual amino acids exhibit first-order exponential depletion from an initial medium concentration of 2.0 μM. Influx of radiolabeled alanine corresponds very closely to net entry of alanine as estimated by HPLC. Therefore, net entry of alanine from dilute solution (5 μM) occurs. The entry of alanine is well described by Michaelis-Menten kinetics with a maximum influx (Jmax) of 1.76 μmol g−1 h−1 ±0.028 (SD) and a half-saturation constant (Kt) of 33.70 μM ±1.15 (SD). Amino acids taken up from solution are distributed rapidly to the internal tissues of the animal. This is demonstrated for alanine and glutamate and for the amino acid analogues, cycloleucine and α-amino-n-butyric acid (ABA). HPLC analysis of the free amino acid pools of mantle, gill, adductor muscle and hemolymph of C. gigas demonstrates variability in total concentration and in details of FAA composition.  相似文献

Starting in 1997, mortality outbreaks in cultured oysters Crassostrea gigas have been reported in northwestern México. Previous studies have shown that massive die‐offs result from multi‐factor processes related to reproduction of the species. We studied the reproductive cycle and the condition index of cultured oysters in the coastal lagoon of El Soldado, Sonora, as well as the relationship of the life cycle with environmental parameters. We used oocyte diameter to determine reproductive stages and the condition index to describe the physiological state of oysters. Additionally, the temperature, salinity, seston and chlorophyll a were recorded at the study site. The results showed that C. gigas began accelerated reproductive activity in March under the influence of high temperature and increased concentrations of food. No spawning events were recorded and gametes were reabsorbed within the gonad in September and October. The results showed a period of nutrient storage during autumn–winter and another period of gamete production in spring–summer. A mortality event occurred at the end of winter, tied to significant increases in temperature and availability of food and in accelerated reproductive activity and high condition index. These conditions were very similar to those reported in other countries during summer die‐offs of C. gigas.  相似文献

Natural seedling collection is widely used in the culture of various bivalve species. For successful natural seedling collection, collectors must be installed when larvae appear in the water column at a stage immediately before attachment. Aquaculture farmers generally identify target larvae by morphological features through microscopic examination in a time- and labor-expensive exercise, which also requires a level of expertise to ensure accurate larval identification. We develop a deep-learning-based object-detection technique that ultimately might reduce the time and effort required to accurately identify and count Pacific oyster larvae, render their identification more consistent, and negate the need for expertise. Images of plankton net samples collected in Matsushima and Sendai bays, Japan, were taken using a new photographic device with a CMOS image sensor. Images of oyster larvae identified by an expert were used to create a library of labeled images to train a deep-learning model, which proved to be 82.4% accurate in precision, 90.8% in recall, and 86.4% in F-measure. A further method for estimating larval shell height from the rectangular shape of oyster larval images is also developed. The standardized mean difference in shell height between measurements and estimates is 3.3%. This deep-learning model has the potential to significantly reduce the time and effort required to identify oyster larvae in plankton samples, and thereby costs of this exercise.  相似文献

Abstract. Production of Pacific oysters was studied under pilot-scale conditions in Baynes Sound, British Columbia, using common bottom culture strategies. Four seeding treatments, each with a different seed per cultch density were cultured: wild-caught seed 5mm in shell height at 10 seed per cultch piece and hatchery produced seed 1-2 mm in shell height at densities of 11, 40 and 105 per cultch piece. The cultch material for all treatments was Pacific oyster shell. All seed was reared for approximately 1 year in a seed nursery located at the 2.2-m tidal level then transferred to a 1-m tidal level grow-out plot until harvest 4 years later in May 1988, Clusters of large numbers of oysters were separated and evenly distributed within the plots when the oysters attained a shell height of 60-100 mm. During the first year, growth was slow and mortalities were relatively high. All treatments produced oysters of similar size at harvest. The proportion surviving at harvest was substantially higher for the wild oysters which were initially larger at time of planting. Within the hatchery treatments proportional survival per cultch piece was inversely related to initial density on the shell; however, total production per cultch piece was positively related to initial density. Most efficient use of seed is attained at lower densities per shell; however, most efficient use of cultch and effort to handle cultch is attained at high densities.  相似文献

ABSTRACT: For the development of a stepwise cryopreservation technique for larvae of the Pacific oyster Crassostrea gigas , various conditions were examined. Larvae at 9, 12, 15, 18 and 21 h after insemination were cooled at a rate of −1°C/min (seeding at −8°C for 15 min) and then plunged into liquid nitrogen at −35 or −40°C using 1.5 M dimethyl sulfoxide (DMSO) and 250 mM trehalose as cryoprotectants. Among these larvae, 15 h after insemination (the trochophore stage before formation of the shell gland) showed the highest motility and the best external appearance after thawing. Trochophore larvae were cryopreserved in preservation media containing different dilutions (1/4, 1/6, 1/8, 1/10 and 1/30) of seawater. Larvae preserved in the 1/4 seawater medium showed the highest appearance of shelled larvae 4 days after thawing. Trochophore larvae reared in seawater at 21, 25 or 29°C were cryopreserved for 8 months and then reared at 26°C after thawing. Larvae reared at 25°C showed the highest survival rate and normal larval ratio at day 6 after thawing, although larvae reared at 21°C showed the highest rates until day 4. One larva developed at 25°C succeeded to settle.  相似文献

In order to study individual growth variability and its relationship with survival in juvenile Crassostrea gigas, parental oysters were sampled at four sites located along the French Atlantic coast and bred under controlled hatchery conditions. Four groups of larvae were obtained by crossing five males and five females from each of the four sites, and a fifth group by crossing these 20 males and 20 females together in a pool. Larvae were reared under conditions allowing the maintenance of a maximum variability of size and gave five experimental groups. Oysters were individually monitored for growth and survival from 3 to 10 months after fertilization. The individual growth performances were relatively stable over time and no noticeable compensation for growth occurred. Early growth rate was a very good predictor of size later in life: 66% of variation in the live weight at 10 months could be explained by variation in the initial growth rate calculated between 3 and 4 months. A significant group effect was observed on survival and on growth rate. Mortality mostly occurred between 3 and 5 months and appeared to affect the slow-growing animals more. However, two groups can be distinguished among those which died during the experimental period, one which showed a decrease in weight and the other whose growth was similar to surviving oysters. These results are discussed in the light of usual oyster farming practices and selective breeding. Light spatfalls of the Pacific oyster, Crassostrea gigas, have occurred in the estuaries of the Rivers Exe, Teign and Dart (Devon) and in the Menai Strait (Gwynedd), near to sites of commercial cultivation of this species. Comparisons of the sizes of the naturally settled oysters with hatchery oysters suggests that the spat from the River Teign and Menai Strait settled in 1989 and 1990. The springs and summers of these years were exceptionally warm with mean monthly air temperatures 0.8°-1.3°C above the 1951–1980 average for the period April to August. Although sitings of Pacific oyster spat were made in Loch Sween and Emsworth Harbour over 20 years ago, there is no evidence that self-sustaining populations have developed. The spatfalls which occurred in some coastal waters of Devon and Gwynedd in 1989 and 1990 are unlikely to sustain natural fisheries and are likely to die out as did the Portuguese oysters which occasionally settled in the River Blackwater, Essex.  相似文献

Two trials were carried out in the laboratory in order to assess the effect of microparticulated feed (F) and live (Thalassiosira pseudonana, M) diets on the growth of recently set (396 ± 13 μm shell height) and 2 mm Crassostrea gigas postlarvae. Different proportions of M and F (100:0, 75:25, 50:50; 25:75, 0:100) were delivered in a single dose of 3 h d−1 in trial 1. Dietary M:F proportions of 100:0, 50:50, and 0:100 were delivered as a single pulse of 8 h d−1 (P1) or two pulses of 4 h−1 (P2) in trial 2. Maximal daily M ration was 296 cells μl−1 d−1 (trial 1), 150 M cells μl−1 d−1 (trial 2), or their equivalent F dry weight. Shell height (SH), dry (DW), and organic weight (AFDW) were evaluated weekly. Oysters from trial 1 significantly increased their size after 28 days, and exhibited no significant dietary differences in terms of DW (1.21 ± 0.15 to 2.01 ± 0.28 mg) or AFDW (0.091 ± 0.022 to 0.166 ± 0.029 mg). Newly set postlarvae (trial 2) also exhibited significant growth after 25 days. No dietary differences were observed in trial 2, yet P2 oysters attained significantly higher shell heights (825–912 μm) than P1 oysters (730–766 μm) after 25 d. Pulse effects were marginally not significant in terms of AFDW and growth rate. Together, these findings showed that balanced microfeeds have a practical potential for the culture of early C. gigas postlarvae, when they are delivered in pulse-feeding schemes  相似文献
