BACKGROUND: There is insufficient evidence in the literature concerning the fluctuations and correlations of serum fat-soluble vitamin concentrations during lactation in dairy cows and the potential effect of age of the animals on these values. OBJECTIVES: The objectives of this study were to determine fluctuations in mean serum concentrations of the fat-soluble vitamins A and E and b-carotene and the correlations among them in clinically healthy dairy cows, from the last month of the dry period until the end of lactation, and to investigate whether age was a factor that affects these serum concentrations. METHODS: Forty-five clinically healthy Holstein cows were assigned to 2 groups based on age. Group A consisted of 23 cows 4 years old (mean +/- SD, 3.24 +/- 0.44 years), and group B consisted of 22 cows >4 years old (6.68 +/- 1.66 years). Blood samples for determination of serum beta-carotene, vitamin A, and vitamin E concentrations were collected from each animal just before the start of study (1 month prior to expected parturition), at parturition, and thereafter, at monthly intervals until the end of lactation. RESULTS: There was a significant positive correlation between beta-carotene, vitamin A, and vitamin E values. Mean serum concentrations of beta-carotene, vitamin A, and vitamin E were significantly higher in younger animals. CONCLUSIONS: To our knowledge, this is the first evidence that age is a factor affecting serum concentrations of fat-soluble vitamins in dairy cows.  相似文献

An oral vitamin E absorption test used in human beings was modified for use in horses. The most appropriate techniques with which to measure gastrointestinal tract absorption of vitamin E (alpha-tocopherol) in horses were developed. Vitamin E was administered orally, and serum values of alpha-tocopherol were measured by use of high-performance liquid chromatography at 0, 3, 6, 9, 12, and 24 hours after vitamin E administration. Variables included comparison of 2 dosages (45 and 90 IU/kg of body weight), routes of administration, and absorption dynamics of 3 preparations of dl-alpha-tocopherol. Absorption of the 2 doses of dl-alpha-tocopherol acetate indicated a dose response; the area under the curve at 24 hours (AUC24) was 4.3 micrograms.h/ml for the 45-IU/kg dose and 32.2 micrograms.h/ml (P less than 0.01) for the 90-IU/kg dose. Maximal absorption was apparent when vitamin E was naturally consumed in grain, compared with administration of identical preparations by stomach tube or paste. In the same horses, dl-alpha-tocopherol and dl-alpha-tocopherol acetate plus polyethylene glycol had statistically similar absorption curves and both had significantly greater AUC24, compared with dl-alpha-tocopherol acetate; values for the 3 compounds were 23.6, 25.8, and 12.6 micrograms.h/ml, respectively. Nutrition most certainly affects health and may play a role in the etiology of growth and reproductive problems in captive cheetah (Acinonyxjubatus) populations. The objective of our research was to examine nutritional differences between two dietary regimens and quantify their physiologic effects on cheetahs held in captivity. Twelve cheetahs were randomly assigned to either a commercial diet (COM) or a supplemented meat diet (SMD) group. These cats were physically examined and had blood samples taken three times over the course of a year. Representative samples of COM and four separate components of the SMD treatment were analyzed over the same time frame for proximate nutrient composition, digestibility, and concentrations of taurine, fat-soluble vitamins, and selected minerals. Concentrations of fat, vitamins A and E, Se, Fe, Cu, Na, and Mn were significantly higher in COM compared with those in SMD samples, with the exception of fat content in turkey. Mg content was lower in COM than in SMD; other nutrients did not differ. Mean concentrations of vitamins A and E in COM were markedly higher than in SMD samples (408,140 vs. 29,696 IU/kg dry matter [DM] and 431 vs. 48 IU/kg DM, respectively) and varied dramatically between sampling periods. Percent crude protein and protein-to-fat ratios were high for SMD compared with either whole prey-based or commercial food preparations. Blood urea nitrogen and serum creatinine levels were above normal reference means for domestic cats. Plasma concentrations of vitamins A, D, and E were significantly higher in COM-fed than in SMD-fed cheetahs. Both plasma retinol and tocopherol levels were almost three times higher in COM-fed cats (1.26 +/- 0.06 vs. 0.53 +/- 0.03 microg/ml and 17.5 +/- 0.7 vs. 6.4 +/- 0.02 microg/ml, respectively) and exceeded the normal ranges expected for domestic felids. Significant differences between male and female cheetahs were found for plasma concentrations of vitamin E, Se, and Fe after allowing for effects of diet and time of collection. Excess fat-soluble dietary vitamins can result in direct toxicities as well as nutrient antagonisms and may be linked to reproductive and health issues in captive cheetahs. The high protein levels found in SMD may be linked to chronic renal disease, which was detected in some of these cheetahs.  相似文献

Lymphocytes of lambs on a low selenium/vitamin E diet were isolated from peripheral blood, and mitogenic responses to phytohaemagglutinin (PHA) tested in the presence of different doses of sodium selenite and emulsified vitamin E added in vitro. An enhancing effect of selenium was observed at doses of 1 ng/ml or less, and reached a plateau at about 10 ng/ml. Toxic effects were evident beyond 1 micrograms/ml. The stimulatory potential of selenium among lambs was inversely related to their ability to respond to PHA in control cultures but was not related to the blood glutathione peroxidase activity of the animals concerned. Optimal doses of vitamin E added to culture (0.15-1.5 micrograms/ml) elevated responses beyond those seen with selenium, but synergistic effects were not apparent. Similar results were obtained when lymphocytes from deficient, myopathic lambs were cultured with serum from lambs supplemented in vivo, and when PHA responses of untreated and treated lambs were compared. Tests with other phytolectins (concanavalin A and pokeweed mitogen) suggested that the two micronutrients exert a differential influence on lymphocyte sub-populations. It was also concluded that the poor lymphocyte responses seen in myopathic lambs can be readily and rapidly reversed by injection of these nutrients, and that prophylaxis is most effective during the first 6 weeks of life.  相似文献

In a trial conducted in the south of England in January to February 1989, blood samples were obtained from nine dairy herds with more than 30 cases of clinical mastitis/100 cows and from nine herds with less than 30 cases/100 cows during the previous 12 months. Whole blood glutathione peroxidase (GSHPx) activity and plasma vitamin E concentration were determined for 12 cows in each herd. The mean (+/-sd) values for the herds with the lower incidence of mastitis were 7.57 +/- 1.86 micrograms/ml plasma vitamin E and 23.8 +/- 22.8 U/ml rbc GSHPx activity, compared with 7.74 +/- 1.69 micrograms/ml plasma vitamin E and 20.61 +/- 8.8 U/ml rbc GSHPx activity for the herds with the higher incidence of the disease. These values indicate that the vitamin E levels were generally adequate but that some animals and herds had low GSHPx activities, suggesting that their diets may have contained inadequate selenium. The activities of GSHPx and the vitamin E levels in plasma were not significantly different in the two groups of herds, and no relationship was found between the two nutrients and the incidence of clinical mastitis. However, there was a significant negative correlation between the activity of GSHPx and the bulk milk cell counts in the herds with a low incidence of mastitis suggesting that there was an association between the incidence of subclinical mastitis or inflammation and the selenium status of these herds.  相似文献

[目的] 建立同时测定蒙古族传统奶酪中VA、VD2、VD3、α-生育酚、β-生育酚、γ-生育酚、δ-生育酚、VK1共8种脂溶性维生素及天然色素β-胡萝卜素含量的高效液相色谱方法(high performance liquid chromatography,HPLC)。[方法] 选取蒙古族传统奶酪为试样,经无水乙醇提取,酶解后用正己烷萃取,旋蒸浓缩,复溶过滤膜。经Agilent poroshell 120 EC-C18(150 mm×4.6 mm,4 μm)色谱柱分离,以甲醇和水(体积比为95∶5)为流动相,采用FLD和VWD检测器检测,外标法定量。[结果] VA、α-生育酚、β-生育酚、γ-生育酚、δ-生育酚、VK1、β-胡萝卜素在0.1~1.0 μg/mL浓度范围内线性关系良好,VD2、VD3在0.05~1.00 μg/mL浓度范围内线性关系良好,相关系数R2均大于0.998。[结论] 建立的HPLC方法具有简便、快捷、灵敏度高、回收率高、分析时间短等优点,可为蒙古族传统乳制品中多种脂溶性维生素及天然色素的快速检测提供技术支撑。  相似文献

In five trials with 99 suckling or weaned piglets, the effects of the increased adrenocortical function, caused by cold, weaning from the sow or fasting or by stimulation with exogenous adrenocorticotrophic hormone (ACTH) were studied as exerted on vitamin A and vitamin E concentrations in the blood serum. Three hours after an exposure of three-day and four-day piglets to the temperature of 8-12 degrees C, a small drop of the concentrations of both vitamins occurred. In four-week weanling piglets a decrease in vitamin E concentration was observed in two days, the trend being slight in vitamin A concentration. At the same time some sibs were left fasting, which considerably reduced the concentrations of both vitamins. The situation was similar in two hours after ACTH administration to suckling piglets, however the difference was insignificant in vitamin E concentrations. In seventeen hours elapsing from two administrations of the adrenocorticotrophic hormone (ACTH) when the increased secretion of corticosteroids was fading out, only the vitamin A concentration in suckling piglets was found to drop. The response in weanling piglets was negligible. The suppressive effects of stress on vitamin A concentrations were usually observed when the levels of circulating corticosteroids were high or in the period immediately following this status. The changes in nutrition after early weaning exert large negative effects on vitamin E concentrations in the blood serum. The differences in the response of the organism to the two vitamins may be due to various types of transport mechanisms in the blood circulation. Abstract

This study investigated the effects of vitamin E and selenium (Se) injections on concentrations of vitamin E, vitamin A, and Se in blood and milk to predict when injections may improve ewes’ performance. Awassi ewes received 0 (control, 13 ewes) or 15 plus 0.05?mg/kg BW of vitamin E and Se (injected, 15 ewes) at 1 and 4 weeks postpartum. Blood and milk samples were collected weekly for 6 weeks. Injections increased vitamin E, decreased Se, and had no effects on vitamin A levels in blood. Blood vitamin E was correlated positively with vitamin A. Injections increased vitamin E, but had no effects on vitamin A or Se levels in milk. Milk vitamin A was positively correlated with vitamin E and Se levels in the control group. Milk vitamins E and A were positively correlated with their corresponding levels in blood. Milk somatic cell count was negatively correlated with blood vitamin A levels.  相似文献

The dynamics of vitamin E in the blood serum of sows of the Slovak Large White breed was studied from the 10th day of gravidity to the 20th day of lactation. A slight variation was observed in the average values of serum vitamin E from the 10th to the 50th day of gravidity (from 0.134 to 0.122 and 0.129 mg per 100 ml). A rise to the absolutely highest average values of serum concentration of vitamin E, 0.166 mg per 100 ml, was ascertained in the period from the 70th to the 90th day of gravidity. In the subsequent period, i.e. 110th day of gravidity, the value dropped to 0.120 mg per 100 ml, and on the 5th day of lactation the absolutely lowest value (0.095 mg per 100 ml) for the whole study was obtained. On the 20th day of lactation the serum concentration of vitamin E increased on an average to 0.103 mg per 100 ml. These data indicate the critical periods in the levels of vitamin E; these are from the 90th to the 100th day of gravidity and the first days of lactation when the concentration of vitamin E markedly decreases. If vitamin E is not supplemented to the sows during gravidity in sufficient amounts, preclinical and clinical vitamin E deficiency can be expected to occur.  相似文献

Vitamin E (alpha-tocopherol) is an antioxidant vitamin important in protecting unsaturated fatty acids in lipid membranes from peroxidation. Variation in collection, storage, and shipping conditions of samples can potentially lead to breakdown of vitamin E prior to analysis. Therefore, the purposes of this project were 1) to determine the stability of vitamin E in refrigerated and frozen porcine liver and serum and 2) to evaluate the effects of red blood cell (RBC) hemolysis on porcine serum vitamin E concentrations. Porcine liver and nonhemolyzed serum were collected and stored refrigerated or frozen. Samples were analyzed for vitamin E immediately or on days 2, 3, 7, or 14. In addition, porcine RBCs were added to normal serum at concentrations from 1 x 10(6) to 1 X 10(9) RBC/ml and hemolyzed by freeze-thaw prior to analysis for vitamin E or products of lipid peroxidation.  相似文献

The need for supplementation of dairy cow diets with fat-soluble vitamins depends on the amount of vitamins naturally present in the diet, the availability of dietary vitamins, and the vitamin utilization rate of the animal. Fresh forage contains ample amounts of the vitamin A precursor beta-carotene as well as vitamin E. Irrespective of the dietary amount, however, the availability of vitamins A, D, and E, as well as beta-carotene, can be adversely influenced by poor fat digestion, as commonly occurs due to enteric disease in young calves. In addition, high-grain diets appear to increase the amount of ruminal vitamin destruction and may thus increase vitamin requirements. The vitamin utilization rate may be increased by inflammation as well as dietary and environmental factors. The factors influencing vitamin availability and utilization rate should be considered when formulating rations. Because the vitamin requirement is variable, blood concentrations of vitamins should be monitored when conditions such as poor fertility, weak calves, and poor immune response are present.  相似文献

The effect of the oral administration of high dosages of vitamins C and E on the innate immune system of the seabream was investigated. Gilthead seabream (Sparus aurata L.) were fed four different diets for 45 days: a commercial diet as control, a 3g/kg vitamin C-supplemented diet, a 1.2g/kg vitamin E-supplemented diet or a diet containing both vitamin supplements. After 15, 30 and 45 days, serum ascorbic acid and alpha-tocopherol levels, growth, complement titers and head-kidney leucocyte phagocytic and respiratory burst activities were evaluated. The results showed that serum vitamin levels reflected dietary input. Fish fed the vitamin C-supplemented diet showed an enhanced respiratory burst activity, while fish fed the vitamin E-supplemented diet exhibited increased complement and phagocytic activities. All of these positive effects were found in fish fed the joint diet, as well as a synergistically enhanced respiratory burst activity at day 30. The results demonstrate that in vivo vitamins C and E exert a synergistic effect enhancing the respiratory burst activity of seabream phagocytes.  相似文献

The physiological and production effects of feeding additional vitamin E and ruminally protected vitamin C were examined in cattle challenged with bovine herpesvirus 1 (BHV 1). Forty-eight individually penned 6-mo-old Angus and Angus crossbred heifer calves with a mean BW of 151 kg were allocated randomly to four diets in a 2 x 2 factorial arrangement of treatments. Pelleted diets provided either 15 or 185 IU/kg of DM of vitamin E, with or without 3.7 g of ruminally protected vitamin C/kg of DM. Blood samples were taken at start of the experiment and at wk 4, 5, and 6. At the start of wk 5, half of each of the dietary groups was challenged with BHV 1. Feeding additional vitamin E was associated with greater (P < 0.001) mean plasma alpha-tocopherol. In contrast, feeding ruminally protected vitamin C was not associated with greater (P = 0.59) mean plasma ascorbate concentration; however, feeding ruminally protected vitamin C was associated with lower (P = 0.03) mean blood total superoxide dismutase (Cu/Zn SOD and Mn SOD) concentration. Calves fed additional vitamin E had greater (P = 0.05) mean plasma beta-carotene concentrations. There were interactions between dietary intake of vitamins E and C with respect to serum ceruloplasmin concentration (P = 0.01) and G:F (P = 0.05). Bovine herpesvirus 1 challenge was associated with lower white cell count (P = 0.007), lymphocyte count (P < 0.001), and DMI (P = 0.03). Feeding additional vitamin E to calves challenged with BHV 1 was associated with a lower (P = 0.03) serum ceruloplasmin concentration. There was a non-significant trend towards an interaction (P = 0.06) between the feeding of vitamins E and C, with virus-challenged calves fed additional vitamin E alone having greater plasma retinol concentrations. The feeding of vitamins E and/or C in calves challenged with BHV 1 was associated with alterations in the concentrations of other antioxidants. More severe disease may have translated these cellular effects to changes in health and performance.  相似文献

Selenium and vitamin E concentrations were measured in the blood sera of 287 cows in 91 herds. Herds with chronic mastitis and muscle diseases had significantly lower selenium concentrations (10.4 +/- 5.26 and 11.7 +/- 8.63 micrograms/litre serum, respectively) than healthy control herds (17.7 +/- 15.00 micrograms/litre). The selenium concentrations in herds with low fertility did not differ from control herds. The vitamin E concentrations were similar in all the herds. Supplementation of the mineral mixture with 20 mg sodium selenite and 2 g vitamin E/kg on 15 farms with a deficient selenium supply increased the selenium concentrations in the blood within two months in 12 of the herds. The vitamin E concentrations were hardly influenced by the supplementation.  相似文献

本试验采用5×2完全随机试验设计,5个维生素A(VA)添加水平(0、2500、5000、10000IU/kg和15000IU/kg),2个维生素E(VE)添加水平10mg/kg和100mg/kg,360只1日龄北京鸭随机分为10个处理组,研究不同水平VA、VE对北京鸭生产性能的影响。结果表明,VA的添加可显著提高北京鸭生长前期的平均日增重和平均日采食量(P0.01);对料肉比的影响差异不显著(P0.05),VE水平对北京鸭前期生产性能无显著影响(P0.05);建议北京鸭生长前期VA的适宜添加量为5000~10000IU/kg,VE为10mg/kg。  相似文献

目的:采用微生物法测定婴幼儿配方乳粉中生物素、叶酸和VB12.方法:利用不同乳酸菌对特定维生素具有敏感性的特点,通过测定菌株的生长变化来反映样品中相应微生物的维生素含量,对市售的13种婴幼儿配方乳粉维生素进行测定.结果:微生物法对于生物素和VB12测定数据更稳定,重复性好,生物素相对标准偏差为0.056%~4.158%,VB12的相对标准偏差范围为0.112%~3.848%,而叶酸的检测数据之间波动稍大,相对标准偏差为1.236%~8.332%.结论:本法适合婴幼儿配方乳粉中生物素、叶酸和VB12这3种维生素的分析定量研究.  相似文献

Plasma levels of vitamins A, E, beta carotene, both plasma and erythrocyte glutathione peroxidase (GSHPx), lipid peroxidation (LPO) and reduced glutathione (GSH) were investigated in cattle naturally infected with Theileria annulata and treated with buparvaquone. There were two groups each containing 30 cattle. Naturally infected cattle were used in the second group. Buparvaquone (2.5 mg/kg body weight) was administered to animals in the second group. Blood samples were taken from control animals, and immediately before treatment, and from animals 10 days after the injection of buparvaquone. Detection of the infected animals was carried out by blood smears. Plasma vitamins A, E, beta carotene, both plasma and erythrocyte GSHPx, LPO and GSH levels were determined. The levels of LPO in plasma and erythrocyte samples were significantly (P < 0.05, P < 0.01) higher after treatment than in either control animals or before treatment. Plasma levels of antioxidant vitamins, vitamin E and beta carotene were significantly (P < 0.05, P < 0.01) lower after treatment than in either control animals or before treatment, while the vitamin E level was found to be higher before treatment than in either the control group or animals after treatment (P < 0.05, P < 0.01). The levels of vitamin A in plasma and the activity of GSHPx and GSH in both plasma and erythrocytes in control animals after and before treatment did not differ significantly. In conclusion, we observed that there was a decreased plasma level of vitamin E and beta carotene and an increased level of LPO in cattle treated with buparvaquone. Buparvaquone might function in the treatment of Theileria annulata by forming free radicals.  相似文献

At the Institute of Animal Nutrition of the Federal Agricultural Research Centre (FAL) in Braunschweig a feeding trial with 30 first lactating cows (German Holstein) was realised in 2004 for a complete lactation. All animals received a diet based on maize- and grass-silage ad libitum at the rate of 60:40 (DM-base) and concentrate depending on their milk yield.The animals were fed with energy and protein as recommended (GfE 2001) and were kept in loose housing. After calving they were split randomly into two feeding groups with 15 animals each. Group 1 was fed a concentrate according to the recommendations of the GfE (2001), whereas group 2 was offered the same concentrate with roughly the double amount of calcium, magnesium, phosphorus, copper, zinc, beta-carotene and vitamins A and E. To study the influence of the different concentrates on the serum concentrations of minerals and vitamins, blood, feed and milk samples were regularly taken and analysed in the course of lactation. Significantly higher blood concentrations were detected for phosphorus, zinc and vitamin E in group 2, but therefore a direct influence of the feeding on the blood concentrations was detected only for zinc and vitamin E. Over the whole lactation the average milk yield of group 1 was 2.6 kg/day higher and the average milk fat content was reduced about 0.6 percent points compared to group 2.  相似文献

OBJECTIVE: To examine the effects of injectable vitamins A, D and E at feedlot entry on health and growth rate and the effects of injectable vitamin C at the time of treatment for bovine respiratory disease (BRD) on cattle health. DESIGN: Systematic allocation of 2465 cattle at feedlot entry to: a commercial vitamin A, D and E preparation at the label dose rate; commercial vitamin A, D and E at twice the label dose rate; a formulation with no vitamin D, a lower concentration of vitamin A and a higher concentration of vitamin E; and the oil-based carrier alone at volumes corresponding to the above treatments. Comparisons of growth rate, disease and mortality were made between the groups at the conclusion of the feeding period. In a separate experiment, 176 cattle were alternately administered injectable vitamin C at the time of treatment for BRD, or were not injected with vitamin C, and mortality was compared between the groups. RESULTS: There were no differences between cattle administered vitamin A, D and E at feedlot entry and the controls in growth rate (P=0.11), all diseases (P=0.99), BRD (P=0.60) or mortalities (P=0.95). Cattle treated with the higher vitamin E and lower vitamin A preparation had a higher (P=0.02) incidence of anorexia than the other groups. Fewer cattle treated with 5 g of vitamin C by intramuscular injection at the time of treatment with antibiotics for BRD subsequently died (P=0.04). CONCLUSIONS: The routine injection of cattle with vitamins A, D and E at feedlot entry is unlikely to result in improvements in health and growth rate where cattle are provided with these vitamins in their diets at concentrations equal to the National Research Council recommendations. Mortality rate in cattle diagnosed with BRD may be reduced by intramuscular injection of vitamin C at the time of treatment with antibiotics.  相似文献

The aim of the study was to investigate the ability of vitamin C to increase the antioxidative and immunomodulating potential in healthy dogs. Fifteen dogs were tested for the effects of orally administered vitamin E (60 mg dl‐α tocopheryl acetate) in combination with vitamin C (0, 30 or 60 mg ascorbic acid crystalline). Three treatments (0, 30, 60 mg vitamin C) were tested in a 3 × 3 cross‐over study in three periods of 36 days. Pre‐prandial blood samples were taken for analysis of vitamins C, E, A, retinyl palmitate and stearate, antioxidant status [Thiobarbituric acid reactive substances (TBARS) and uric acid], biochemical and haematological analysis. Subpopulations of lymphocytes, mitogen‐induced peripheral blood mononuclear cell proliferation (PBMC) and serum IgA and IgG concentrations were determined. There was a trend (p = 0.056) for an increased plasma vitamin C concentration by vitamin C supplementation. There was no evidence that dietary treatment altered neither the other plasma vitamin concentrations nor TBARS and uric acid concentrations nor the subpopulations of the lymphocytes except for the number of CD4+ lymphocytes that increased with vitamin C supplementation. There was no effect of vitamin C on serum IgA and IgG concentration. A significant time × treatment interaction was demonstrated on PBMC’s to pokeweed, with an increase observed by 30 mg vitamin C supplementation but a decrease by 60 mg vitamin C supplementation. There was no clear evidence for an effect of dietary vitamin C on antioxidative capacity in healthy dogs fed a diet with vitamin E concentrations well above the recommendations. Yet, a limited number of immunological parameters were slightly affected.  相似文献
