几种蛋白质原料体外消化率测定方法的比较   总被引:8,自引:0,他引:8

试验分别采用胃蛋白酶-胰蛋白酶两步法、肉食性鱼类(如鲈鱼)消化道粗酶提取液消化法和草食性鱼类(如草鱼)消化道粗酶提取液消化法测定了酪蛋白、鱼粉、豆粕、菜籽粕、棉籽粕、酵母粉和玉米蛋白粉等7种蛋白质原料的体外消化率。3种测定方法中,鱼粉的消化率差异不显著(P>0.05);豆粕、菜籽粕、酵母粉和玉米蛋白粉用两步法和草鱼消化酶法测定的消化率无显著差异(P>0.05);棉籽粕消化率用两步法测定值高于用消化酶法的测定值,差异极显著(P<0.01);豆粕、菜籽粕、酵母粉和玉米蛋白粉的消化率用两步法比用鲈鱼消化酶测定的值高,差异极显著(P<0.01);草鱼消化酶法和鲈鱼消化酶法对酪蛋白的消化率无显著差异(P>0.05),而对于豆粕、菜籽粕、棉籽粕、酵母粉和玉米蛋白粉草鱼消化酶法测定值高于鲈鱼消化酶法的测定值,差异极显著(P<0.01)。结果表明,胃蛋白酶-胰蛋白酶两步体外消化法在测定鱼粉蛋白质消化率时可以替代鱼类消化液粗酶消化法,对于其它蛋白质原料使用该方法应慎重。  相似文献

青鱼对8种饲料原料中营养物质的表观消化率   总被引:1,自引:0,他引:1  
本试验旨在研究青鱼(Mylopharyngodon piceus)对国产鱼粉、蝇蛆粉、玉米蛋白粉、大豆粕、花生粕、棉籽粕、菜籽粕和米糠8种饲料原料干物质、粗蛋白质、氨基酸、粗脂肪、总磷和总能的表观消化率。试验饲料由70%基础饲料和30%待测饲料原料组成,并以0.1%的三氧化二钇(Y2O3)为外源指示剂。选取平均体重为(60.17±1.28)g的青鱼270尾,随机分成9组,每组3个重复,每个重复10尾鱼。对照组试验鱼饲喂基础饲料,试验组试验鱼分别饲喂1种试验饲料。饲喂1周后采用自排法收集粪便待测。结果表明:8种饲料原料的干物质、粗蛋白质、粗脂肪、总磷、总能以及总氨基酸的表观消化率的范围分别是62.17%~86.47%、83.34%~95.84%、78.93%~100.06%、37.33%~81.99%、66.75%~89.86%、86.33%~96.58%。在8种饲料原料中,玉米蛋白粉的干物质、总磷和总能的表观消化率最高,而大豆粕的粗蛋白质、粗脂肪和总氨基酸的表观消化率最高;棉籽粕的干物质和总能的表观消化率最低,米糠的粗脂肪和总磷的表观消化率最低。各饲料原料中总氨基酸表观消化率与粗蛋白质表观消化率的变化趋势一致,其中以蝇蛆粉最低。由此可见,大豆粕和玉米蛋白粉是青鱼的优质植物蛋白质源,可适量替代鱼粉;蝇蛆粉中粗蛋白质和氨基酸的消化率都较低,在青鱼饲料中的添加量不宜过高;花生粕、棉籽粕和菜籽粕也是较好的植物性蛋白质源,在青鱼饲料中适量添加既有利于饲料的营养平衡,还可降低饲料成本;米糠作为青鱼的能量原料必须保持新鲜,并控制其在饲料中的用量。  相似文献

本试验研究了肉仔鸡性别与日龄对菜籽粕、棉籽粕、肉骨粉和羽毛粉的氨基酸消化率的影响。试验中测定了4周龄和6周龄的肉仔鸡公鸡和母鸡对菜籽粕、棉籽粕、肉骨粉和羽毛粉的氨基酸消化率。研究表明 ,除羽毛粉以外 ,4周龄肉仔鸡对其他3种原料的氨基酸真消化率显著高于6周龄肉仔鸡(P<0.01) ,肉仔鸡日龄对羽毛粉的氨基酸真消化率无影响。肉仔鸡性别对棉籽粕的氨基酸真消化率具有极显著影响(P<0.01) ,公鸡对棉籽粕的氨基酸真消化率极显著低于母鸡 ;性别对其它3种原料的氨基酸真消化率无显著影响。肉仔鸡性别与日龄对菜籽粕氨基酸真消化率存在极显著互作(P<0.01) ,对棉籽粕的氨基酸真消化率存在显著互作(P<0.05) ,且各种氨基酸消化率均表现出相同的变化趋势  相似文献

利用异育银鲫(allogynogenetic crucian carp)的肠道为消化酶酶源,采用离体消化率的测定方法,分别测定了米糠、麦芽根、血粉、豆粕、花生粕、花生饼、棉粕、菜粕、玉米蛋白粉、鱼粉的离体消化率。异育银鲫对10种饲料原料的蛋白质消化率分别为:花生饼43.38%、玉米蛋白粉41.27%、米糠39.72%、血粉37.20%、花生粕36.85%、棉粕35.60%、鱼粉33.87%、豆粕32.69%、菜粕32.10%、麦芽根21.28%。试验结果表明:血粉、花生粕、花生饼、棉粕、玉米蛋白粉作为优质的高蛋白饲料原料替代或部分替代鱼粉在异育银鲫饲料配方中使用是可行的。  相似文献

利用体外消化法研究了欧洲鳗粗酶液对鱼粉、发酵豆粕、菜籽粕、玉米蛋白粉、啤酒酵母、蝇蛆粉、膨化大豆、豆粕、花生粕、红虫粉等10种单一蛋白源以及另外5种复合蛋白源酶解7 h的体外消化率以及酶解液中游离氨基酸的生成量,以期筛选出最有可能替代鱼粉的蛋白原料。结果表明:(1)最有望部分或全部取代鱼粉的植物性蛋白源为啤酒酵母、发酵豆粕和花生粕,其干物质和粗蛋白质体外消化率分别为62.75%、55.15%、49.45%和78.50%、70.70%、85.70%;动物性蛋白源中红虫粉和蝇蛆粉的干物质体外消化率与鱼粉差别不大,粗蛋白质消化率红虫粉蝇蛆粉鱼粉。(2)在0~7h,各种蛋白源酶解氨基酸生成量基本呈增加趋势,对植物性蛋白源来说,啤酒酵母氨基酸生成量明显高于其他植物性蛋白源,其次是发酵豆粕、玉米蛋白粉和花生粕,较低的为菜籽粕、膨化大豆、豆粕。对于动物性蛋白源来说,红虫粉体外消化生成氨基酸的量明显高于鱼粉和蝇蛆粉。这一结果说明红虫粉和啤酒酵母是优质的蛋白源,很容易被鳗鱼消化吸收利用。(3)对于混合蛋白源混1、混2、混3、混4、混5来说,其干物质和粗蛋白质的体外消化率分别为:混1混2混5混4混3;混2混1混5混4混3,而原料中粗蛋白含量为混3混4混5,其鱼粉添加量分别为80%、70%、60%,混1、混2均不含鱼粉,这说明鳗鱼对几种植物性蛋白源的粗蛋白体外消化率均高于鱼粉,这也与之前单一蛋白源的粗蛋白体外消化率结果一致。  相似文献

本试验对黄羽和爱拔益加肉种鸡在棉籽粕、菜籽粕、禽副粉3种饲料原料氨基酸消化率进行了比较研究。试验结果表明 ,经内源校正后 ,黄羽肉鸡对棉籽粕、禽副粉的总真氨基酸消化率高于爱拔益加肉鸡 ,对菜籽粕的总真氨基酸消化率和爱拔益加肉鸡十分接近。就单个饲料原料的总真氨基酸消化率而言 ,黄羽肉鸡对禽副粉的消化率最高 ,其次是菜粕和棉粕 ;爱拔益加肉鸡显示相同的规律 ;虽然爱拔益加肉鸡和黄羽肉鸡在谷氨基、酪氨酸、赖氨酸、色氨酸的内源排泄量差异达到显著水平 (P<0.05,P<0.01),两种肉鸡在内源总氨基酸排泄量之间不存在显著差异 (P<0.05)。  相似文献

本试验旨在研究鳙鱼(Aristichthys nobilis)对鱼粉(国产)、干酒糟及其可溶物(DDGS)、菜籽粕、米糠、豆粕、酶解羽毛粉、棉籽粕、小麦麸、玉米蛋白粉和花生粕中干物质、粗蛋白质、粗脂肪、总能以及氨基酸的表观消化率。试验饲料按照基础饲料和待测饲料原料7∶3的比例构成,并添加0.5%的三氧化二钇(Y2O3)作为外源指示剂。挑选初体重为(290.02±2.82)g的鳙鱼,随机分成11组,每组3个重复,每个重复10尾鱼。随机选取1组作为对照组投喂基础饲料,剩余10组为试验组分别投喂试验饲料。投喂1周后通过虹吸法收集粪便。结果表明:1)10种饲料原料中干物质、粗蛋白质、粗脂肪和总能的表观消化率的范围分别为69.9%~85.7%、73.5%~88.3%、81.2%~91.1%和81.9%~86.3%。2)在10种饲料原料中,米糠总能的表观消化率最高,而鱼粉的干物质、粗蛋白质和粗脂肪的表观消化率最高;酶解羽毛粉的干物质、粗脂肪和总能的表观消化率最低,玉米蛋白粉的粗蛋白质的表观消化率最低。3)各饲料原料氨基酸的表观消化率基本维持在65.59%~99.17%,且都以小麦麸较低。由此可见,鳙鱼对鱼粉具有很好的利用效果,其次是豆粕和花生粕,而酶解羽毛粉中粗蛋白质的表观消化率最低,小麦麸各氨基酸的表观消化率均比较低。  相似文献

饲料氨基酸利用率的测定是进行可利用氨基酸营养研究的一项基础性工作。本研究利用去盲肠公鸡,应用Sibbald“TME”方法,测定了玉米、次粉、高粱、菜籽饼、棉籽饼、棉仁粕、胡麻饼、豆饼、豆粕、鱼粉、豌豆蛋白粉、绿豆蛋白粉、蚕豆蛋白粉的氨基酸表观和真消化率。氨基酸平均真消化率超过90%的依次为豆粕、蚕豆蛋白粉、鱼粉、绿豆蛋白粉、豌豆蛋白粉。消化率低于85%、高于80%的依次为高粱、玉米、次粉,消化率低于80%的依次为棉仁粕、豆饼、菜籽饼、胡麻饼、棉籽饼。所测饲料各氨基酸的平均真消化率大于85%的依次为谷氨酸、异亮氨酸、精氨酸、门冬氨酸、亮氨酸、苯丙氨酸、酪氨酸;消化率在80%~85%之间的依次为丝氨酸、缬氨酸、蛋氨酸、丙氨酸;小于80%的依次为苏氨酸、赖氨酸和胱氨酸。研究结果表明,不同饲料和不同氨基酸之间消化率存在很大的差异。内源氨基酸排泄量对低蛋白饲料氨基酸利用率存在较大的影响。适当的加工处理对于提高饼粕类饲料的氨基酸消化率具有重要意义。  相似文献

本试验旨在研究建鲤(Cyprinus carpio var. Jian)对鱼粉、肉骨粉、豆粕、花生粕、棉籽粕、菜籽粕和玉米干酒糟及其可溶物(DDGS)7种常用饲料蛋白质原料中干物质、粗蛋白质、氨基酸、总磷和总能的表观消化率.营养物质表观消化率采用基础饲料(基础饲料Ⅰ、Ⅱ)和试验饲料(按基础饲料和待测原料7︰3的比例配制)测定,以0.5%的三氧化二铬(Cr2O3)为外源指示剂.选用平均体重为(200.0±4.7) g的建鲤240尾,随机分为8组,每组3个重复,每个重复10尾.从8组中随机选取1组为对照组,饲喂基础饲料;其余为试验组,饲喂试验饲料.试验分为2个阶段,第1阶段(1~4周)采用基础饲料Ⅰ,用于干物质、粗蛋白质、氨基酸和总能表观消化率的测定;第2阶段(5~8周)采用基础饲料Ⅱ,用于总磷表观消化率的测定.结果表明,各试验原料的干物质表观消化率在35.44%~71.10%之间,其中花生粕和豆粕的干物质表观消化率(分别为71.10%和65.15%)显著高于其他原料(P<0.05).各试验原料的粗蛋白质表观消化率在60.31%~88.75%之间,其中豆粕的粗蛋白质表观消化率最高,而最低的为玉米DDGS,并且豆粕和花生粕的粗蛋白质表观消化率显著高于其他原料(P<0.05).本研究中总磷的表观消化率相对较低,最高的为玉米DDGS(38.69%),最低的为肉骨粉(6.52%).从总体上来看,鱼粉、豆粕和花生粕的单个氨基酸及总氨基酸的表观消化率均处在较高水平,而肉骨粉和玉米DDGS的单个氨基酸及总氨基酸的表观消化率均处在较低水平.由此可知,对建鲤而言,豆粕和花生粕是较好的植物性蛋白质原料.  相似文献

本研究用去盲肠鸡和回—直肠吻合猪评定了饲用蒸煮血粉的氨基酸真消化率(TAAD),并探讨了添加酸性蛋白酶对蒸者血粉TAAD的调控效应。结果表明,蒸煮血粉平均TAAD在鸡(69.0%)明显低于猪(81.9%),而平均TAAD的酶促消化调控效应在鸡(11.2个百分点)明显高于猪(1.7个百分点)。  相似文献

分别用24只去盲肠和未去盲肠健康公鸡,测定了花生粕、胡麻粕和芝麻粕的氨基酸消化率。结果表明:去盲肠鸡内源氨基酸排泄量显著高于未去盲肠鸡(P<0.05);在花生粕中,胱氨酸、赖氨酸、天冬氨酸和谷氨酸的AAD未去盲肠鸡显著高于去盲肠鸡(P<0.05);在胡麻粕和芝麻粕中,除酪氨酸和脯氨酸外,其它各种氨基酸及16种氨基酸总和的AAAD和TAAD未去盲肠鸡显著高于去盲肠鸡(P<0.05)。鸡盲肠微生物对内源及饲料中未消化蛋白质、多肽及氨基酸具有分解作用,干扰饲料AAAD和TAAD测定。对于不易消化、氨基酸含量相对不平衡的低品质蛋白质饲料干扰尤其严重。在测定的3种饼粕中,花生粕氨基酸消化率最高,胡麻粕次之。过度热处理的芝麻粕氨基酸平均消化率为47.3%,赖氨酸消化率为21.7%。  相似文献

本研究应用排空强饲法及石岐杂公鸡、樱桃谷公鸭分别测定定了5种饲料氨基酸表观消化率和真消化率。结果表明:除玉米氨基酸表观消化率外,其余饲料的大多数氨基酸消化率在鸡和鸭之间都存在着明显差异;在32小时收粪时间里,鸭的内源氨基酸排出量明显比鸡多;内源氨基酸对鸡和鸭的饲料氨基酸真消化率都有影响,影响大小随蛋白质含量高低而异,一般饲料蛋白质含量越低的饲料两者相差越大。而内源氨基酸排出量对鸭饲料,尤其对低蛋白含量饲料的氨基酸真消化率的影响比鸡更明显。  相似文献

The additivity of true amino acid digestibility (TAAD) and true metabolizable energy (TME) values in corn, soybean meal, and wheat red dog for White Pekin ducks was investigated. Differences between observed values for the complete diets and values predicted from measurements of individual ingredients were used to test additivity. Eight ducks were each assigned to the following dietary treatments: corn, soybean meal (44% CP), wheat red dog (wheat by-product with less than 4% fiber), complete Diet 1 (corn-soybean meal), complete Diet 2 (corn-red dog-soybean meal), and dextrose. Dextrose-fed ducks were used to estimate endogenous losses. The nitrogen-corrected TME (TMEn) in corn, soybean meal, wheat red dog, and two complete diets were 3.411, 2.919, 2.502, 3.148, and 3.111 kcal/g, respectively. In general, the TME and TMEn values observed in the two complete diets were not different (P > 0.05) from predicted values and indicated that the TME and TMEn in corn, soybean meal, and wheat red dog were all additive. The mean TAAD of corn, soybean meal, wheat red dog, and the two complete diets were not different, and were 87.03, 88.15, 90.58, 85.83, and 87.02%, respectively. The differences in TAAD between observed and predicted values were significant (P < 0.05) only for arginine, lysine, and aspartate in complete Diet 1, and for arginine, histidine, lysine, and aspartate in complete Diet 2. These results indicated that TME and TMEn values for corn,soybean meal, and wheat red dog were all additive in the two complete diets, but digestibilities of some amino acids were not additive and demonstrated some associative effects.  相似文献

应用Sibbald“TME”方法,比较了去盲肠和未去盲肠公鸡对饲料氨基酸的消化率。试验测定了10种饲料的去盲肠和未去盲肠公鸡的氨基酸消化率。结果表明,未去盲肠鸡对10种饲料的氨基酸表观和真消化率均高于去盲肠鸡。轻度受热鱼粉、重度受热鱼粉、脱毒棉籽饼、未脱毒棉籽饼和豆饼的表观氨基酸消化率在去盲肠鸡和未去盲肠鸡之间存在显著差异(P<0.05),未受热鱼粉和芝麻饼的表观氨基酸消化率在去盲肠鸡和未去盲肠鸡之间不存在显著差异(P>0.05)。轻度受热鱼粉、重度受热鱼粉、豆饼的真氨基酸消化率在去盲肠鸡和未去盲肠鸡之间存在显著差异(P<0.05);未受热鱼粉、脱毒棉籽饼、未脱毒棉籽饼和芝麻粕的真氨基酸消化率在去盲肠鸡和未去盲肠鸡之间不存在显著差异(P>0.05)。去盲肠鸡内源氨基酸排出量显著高于未去盲肠鸡(P>0.05)。微生物对未消化食糜中的肽类、氨基酸具有降解作用。  相似文献

Eight digestion trials were conducted with pigs fitted with ileal T-cannulas to determine the apparent digestibilities of N and amino acids in common protein feedstuffs. Trial-to-trial variation was minimal as determined by the variation in digestibilities for soybean meal, which was evaluated in each trial. Apparent digestibilities varied widely; ileal N digestibility ranged from 88 to 63%, and ileal lysine digestibility ranged from 93 to 40%. In general, ring-dried blood meal, corn gluten meal, Menhaden fish meal, poultry-by-product meal and extruded whole soybeans were the most digestible, followed by canola meal, sunflower meal, peanut meal and meat and bone meal. Cottonseed meal and feather meal were the least digestible. Mean values for each feedstuff agree well with published data. Variation in digestibilities among samples of the same feedstuff was greater for the meat and bone meals. Regression of ileal essential amino acid digestibilities on ileal and fecal N digestibility indicated that amino acid digestibilities can be predicted more precisely from ileal N digestibility than from fecal N digestibility. However, neither ileal nor fecal N digestibility could be used with a high degree of certainty to predict ileal amino acid digestibilities.  相似文献

The homoarginine technique has been suggested as a means to determine true ileal amino acid digestibilities in nonruminant animals fed protein-containing diets. Conditions for guanidinating lysine to homoarginine in barley and canola meal and the effect of this process on nutrient composition and ileal digestibilities in the resulting material were investigated. Conditions tested were methylisourea concentration (0.4, 0.5, or 0.6 M) and reaction time (4 or 6 d) at pH 10.5. Using 0.4 methylisourea M solution for 4 or 6 d gave guanidination rates of 72.5 and 78.5% for barley and 72.3 and 75.2% for canola meal, respectively. Using 0.5 M gave 88.0 and 84.6% guanidination rates in barley and canola meal, respectively, after a 6-d reaction time. Under these conditions, guanidination did not change the nutrient composition of barley (P > 0.10), whereas it increased CP (38.4 vs 49.0%), crude fiber (10.2 vs 16.0%), acid detergent fiber (30.0 vs 43.4%) and neutral detergent fiber (29.8 vs 49.4%) levels in canola meal (P < 0.05). Four 33.6-kg barrows fitted with a simple T-cannula at the terminal ileum were fed a 16% CP unguanidinated barley and canola meal-based diet for four consecutive 14-d periods. Ileal digesta were collected continuously for 24 h on d 12 and 14 to determine apparent nutrient digestibilities. On the morning of d 14, pigs were fed a diet in which half of the barley and canola meal was replaced with guanidinated material for determining true ileal amino acid digestibilities. Digesta samples were pooled by pig and by 24-h period to give 16 observations per diet. Apparent ileal digestibilities of DM, CP, and AA in the unguanidinated and guanidinated barley-canola meal diet were similar (P > 0.10) despite the changes observed in canola meal. Apparent ileal lysine digestibility was 73.9 and 74.5% in the unguanidinated and guanidinated diet, respectively. The true ileal lysine digestibility was 88.1%. The present results show that guanidination does not interfere with digestion and further support the use of the homoarginine method for determining true ileal amino acid digestibilities in pigs fed practical diets. A methylisourea solution of 0.5 M and a 6-d reaction time are recommended for converting lysine to homoarginine in barley and canola meal.  相似文献

本研究以去盲肠鸡和改良的“TME”法测定了发酵血粉和蒸煮血粉的氨基酸利用率。结果表明,除蛋氨酸外,鸡对发酵血粉的氨基酸利用率显著高于蒸煮血粉(P<0.05)。发酵血粉是一种值得开发利用和推广的蛋白质饲料。  相似文献

1. The apparent ileal and excreta digestibilities of amino acids in 15 samples representing 12 food ingredients were determined using 5-week-old male broiler chickens. The ingredients included 3 samples of cereals (wheat, maize and sorghum), 6 samples of plant protein meals (soyabean meal, cottonseed meal, canola meal and sunflower meal) and 6 samples of animal protein meals (meat meal, meat-and-bone meal, feather meal and fish meal). 2. The test ingredients were incorporated as the sole source of dietary protein in assay diets. Each diet was offered ad libitum to 3 pens (4 birds/pen) from d 35 to d 42 post-hatching. Total collection of excreta was carried out during the last 4 d. All birds were killed on d 42 and the contents of the lower half of the ileum were collected. Apparent ileal and excreta amino acid digestibilities were calculated using acid-insoluble ash as the indigestible marker. 3. The influence of site of measurement was found to vary among food ingredients, among samples within an ingredient and among different amino acids within an ingredient. Ileal amino acid digestibility values were similar in some ingredients, but significantly lower or higher in others than the corresponding excreta values. 4. Average ileal and excreta amino acid digestibilities in sorghum and maize were similar, but significant differences were observed for individual amino acids. In contrast, ileal amino acid digestibility values were higher than the corresponding excreta digestibility values in wheat. 5. The average ileal and excreta digestibilites of amino acids in the 3 soyabean meal samples were similar although small, but significant differences were noted for individual amino acids. Site of measurement had no effect on the digestibility of amino acids in canola meal. Digestibilities of valine, isoleucine, phenylalanine, histidine, glutamic acid, alanine and tyrosine in sunflower meal and those of valine, methionine, isoleucine, leucine, lysine, glutamic acid and alanine in cottonseed meal were lower by excreta analysis. 6. Digestibilities in animal protein meals, with the exception of blood meal and fish meal, were consistently higher by excreta analysis. Ileal-excreta differences in individual amino acid digestibilities were more evident in feather meal, meat meal and meat-and-bone meal. 7. Threonine and valine were the indispensable amino acids that were more frequently influenced by the site of measurement. Of the dispensable amino acids, aspartic acid, serine, glutamic acid and alanine were the most affected. 8. Differences determined between ileal and excreta digestibilities in the present study clearly demonstrate that amino acid metabolism by hindgut microflora in chickens may be substantial and that digestibilities measured in the terminal ileum are more accurate measures of amino acid availability than those measured in the excreta.  相似文献
