Background: Inflammatory airway disease has a high prevalence in horses, but is often a diagnostic challenge. Flowmetric plethysmography and histamine bronchoprovocation (FP/HBP) is a simple and effective tool for diagnosis, but reproducibility of these measurements made over time has not been established.
Hypothesis: We hypothesize that the measurement of airway responsiveness in horses using FP/HBP is consistent over both short and long periods of time.
Animals: Twenty-nine healthy adult horses from 2 university herds.
Methods: In this prospective experimental study, airway responsiveness was determined in each horse at day 0 (baseline [BL]) with FP/ HBP, using PC35 (provocative concentration of histamine needed to increase Δflow by 35%) as a measure of airway responsiveness. Each horse was re-tested 1–4 weeks after BL (short-term [ST]) and again at 3–12 months after BL (long-term [LT]).
Results: In the ST period, 23/27 (85%) of the horses had a PC35 that was within 1 doubling concentration of histamine of their BL value, with a mean change of 0.52 doubling concentrations (95% CI 0.26–0.79, range 0–2.06). For the LT data, 19/26 (73%) of horses were within 1 doubling concentration of their BL value, with a mean change of 0.81 doubling concentrations (95% CI 0.45–1.17, range 0.14–3.10). There was no significant difference in reproducibility between the 2 groups of subjects.
Conclusions and Clinical Importance: Repeated measurements of airway responsiveness obtained with FP/HBP show acceptable reproducibility over time periods up to a year. However, caution must be used when testing horses when ambient air temperature is low.  相似文献

Background

Inflammatory airway disease (IAD) in horses is a widespread, performance‐limiting syndrome believed to develop in response to inhaled irritants in the barn environment.

Objectives

To evaluate changes in bronchoalveolar lavage fluid (BALF) cytology and exposure to particulates, endotoxin, and ammonia during horses'' first month in training.

Animals

Forty‐nine client‐owned 12‐ to 36‐month‐old Thoroughbred horses entering race training.

Methods

In this prospective cohort study, a convenience sample of horses was assigned to be fed hay from a net (n = 16), whereas the remaining horses were fed hay from the ground (n = 33). BALF was collected at enrollment and after 14 and 28 days in training. Respirable particulate, inhalable particulate, respirable endotoxin, and ammonia concentrations were measured at the breathing zone of each horse weekly.

Results

Median respirable particulates were significantly higher when horses were fed from hay nets than when fed hay from the ground (hay net 0.28 mg/m3, no hay net 0.055 mg/m3, P < .001). Likewise, inhalable particulate (hay net 8.3 mg/m3, no hay net 3.3 mg/m3, P = .0064) and respirable endotoxin (hay net 173.4 EU/m3, no hay net 59.2 EU/m3, P = .018) exposures were significantly higher when horses were fed from hay nets. Feeding hay from a net resulted in significantly higher BALF eosinophil proportions over time (P < .001). BALF eosinophils were significantly related to respirable particulate exposure (14 days in training r s = 0.37, P = .012, 28 days in training, r s = 0.38, P = .017).

Conclusions and Clinical Importance

Pulmonary eosinophilic inflammation develops in response to respirable particulate exposure in young Thoroughbreds, indicating a potential hypersensitivity to inhaled particulate allergens.  相似文献

Background: Summer pasture-associated recurrent airway obstruction (SPA-RAO), a seasonal airway obstructive disease of horses, is characterized by clinical exacerbation after exposure to pasture during warm months of the year. Endothelin (ET)-1, potent bronchoconstrictor, mitogen, secretagogue, and proinflammatory mediator, has been implicated in the pathogenesis of asthma and equine heaves.
Hypothesis: Immunoreactive ET-1 concentrations increase during clinical exacerbation and return to basal values during periods of disease remission.
Animals: Twelve horses, 6 affected with SPA-RAO and 6 nonaffected.
Methods: Prospective, observational study. Bronchoalveolar lavage fluid (BALF), arterial and venous plasma samples, and clinical variables were obtained from affected horses during clinical exacerbation and remission. Samples and data of nonaffected horses were collected during the summer and winter on dates similar to affected horses. Immunoreactive ET-1 was determined using a commercial ELISA.
Results: The median and range ET-1 concentrations (pg/ml) in arterial (1.3, 0.7–1.8) and venous (1.3, 1.2–1.7) plasma and in BALF (0.3, 0.2–0.4), and pulmonary epithelial lining fluid (PELF) (25.5, 21–50) were greater in affected horses during clinical exacerbation compared with remission ( P < .01). The concentrations of immunoreactive ET-1 were greater in affected horses during clinical exacerbation compared with nonaffected horses ( P < .05).
Conclusions and Clinical Importance: During clinical exacerbation of SPA-RAO, ET-1 is increased in circulation and pulmonary secretions. Intervention with ET receptor antagonists should provide further information on the role of ET-1 in SPA-RAO.  相似文献

The aim of this study was to evaluate equine performance in competitive trotter horses under normal racing conditions. Data of stride length, stride frequency, and mean horse speed were obtained from 50 2-year-old Italian fillies that were winners during the racing and the sprint laps of competition. The enhancement of speed in the sprint lap was obtained by increasing both stride length and stride frequency. Stride frequencies for the racing and the sprint laps were (mean ± SD) 133.63 ± 3.93 and 138.97 ± 6.62 strides per minute, respectively, and stride lengths for the racing and the sprint laps were 7.17 ± 0.17 and 7.30 ± 0.11 m, respectively; mean speed was 48.18 ± 1.09. The usefulness of the kinematic assessments as well as its potential reference for future indicators of performance is discussed in this study.  相似文献

Background

The identification of serum biomarkers of lung inflammation would facilitate the diagnosis of inflammatory airway disease (IAD) in horses.

Hypothesis

Horses with IAD have higher serum concentrations of markers of inflammation compared to controls.

Animals

Twelve horses with IAD and 10 control horses.

Methods

This was a prospective case–control study. Blood and BALF were collected from horses with IAD and controls. Serum concentration of surfactant protein D (SP‐D), haptoglobin, serum amyloid A (SAA) and of the soluble form of triggering receptor expressed on myeloid cells 1 (sTREM‐1) was measured using commercial ELISA tests.

Results

Horses with IAD had higher serum concentration (log‐transformed values) of SP‐D (mean ± SD: 1.773 ± 0.51), haptoglobin (6.657 ± 0.202) and SAA (0.128 ± 0.396) compared to controls (0.942 ± 0.226, 6.38 ± 0.22, −0.398 ± 0.319, respectively; P < .01 for all). Furthermore, the concentrations of SP‐D and haptoglobin combined allowed differentiating the 2 groups (IAD: 8.43 ± 0.564, controls: 7.322 ± 0.249, P < .0001) with a sensitivity and specificity of 100% when a cut‐off of 7.70 (log value) was employed.

Conclusions and Clinical Importance

Surfactant protein D and haptoglobin serum concentrations could be a diagnostic aid in IAD. Further studies are necessary to establish the specificity of our findings before they can be applied in everyday practice.  相似文献

Arabian horses are not only one of the most ancient breeds in the world, but they are also one of the most appreciated racehorse breeds today. The breed generates attention for their phenomenal endurance ability and their capability for gallop racing. Consequently, genetic testing to select the best individuals is attracting ever increasing interests from the Arabian industry. As such, the aim of this study was to further investigate associations between performance and variation at candidate genes suspected of having a key role in Arabian gallop racing performance. Generalized linear models were fit to test associations between eight candidate gene variants and a variety of gallop racing performance traits in a sample of Arabian racehorses (n = 287). Two genes, solute carrier family 16 member 1 (SLC16A1) and acyl-CoA oxidase 1 (ACOX1), were significantly associated with multiple gallop racing performance traits, whereas another gene, actinin alpha 3 (ACTN3) was associated with best race distance. Previously established associations between these three genes and equine metabolism strongly suggest further investigation of these genes, and their relationship with Arabian horse performance is warranted.  相似文献

This report documents the successful conservative medical management of hemothorax in 2 horses. Hemothorax occurred after a lung biopsy procedure (horse 1) and strenuous exercise on a treadmill (horse 2). The horses had tachypnea, tachycardia, nostril flaring, hemoptysis, and pawing. Hemothorax was suspected based upon absence of auscultable ventral lung sounds; development of cool extremities and pale, tacky mucous membranes; the ultrasonographic appearance of moderate to severe amounts of pleural fluid; and a concurrent decrease in hematocrit and total plasma protein. Both horses were treated successfully by intranasal administration of oxygen, intravenous administration of balanced polyionic solutions, and treatment with antibiotics, nonsteroidal anti-inflammatory drugs, and analgesics. In neither case was pleural blood removed. The hemothorax resolved in both horses without lasting abnormalities. Hemothorax does not require drainage for successful resolution.  相似文献

Background: Equine recurrent airway obstruction (RAO) shares many characteristics with human asthma. In humans, an inverse relationship between susceptibility to asthma and resistance to parasites is suspected. Hypothesis/Objectives: Members of a high‐incidence RAO half‐sibling family (F) shed fewer strongylid eggs compared with RAO‐unaffected pasture mates (PM) and that RAO‐affected horses shed fewer eggs than RAO‐unaffected half‐siblings. Animals: Seventy‐three F and 73 unrelated, age matched PM. Methods: Cases and controls kept under the same management and deworming regime were examined. Each individual was classified as RAO affected or RAO unaffected and fecal samples were collected before and 1–3 weeks and 3 months after deworming. Samples were analyzed by combined sedimentation‐flotation and modified McMaster methods and classified into 3 categories of 0 eggs per gram of feces (EpG), 1–100 EpG, and >100 EpG, respectively. Results: PM compared with RAO‐affected F had a 16.7 (95% confidence interval [CI]: 2.0–136.3) times higher risk for shedding > 100 EpG compared with 0 EpG and a 5.3 (95% CI: 1.0–27.4) times higher risk for shedding >100 EpG compared with 0 EpG. There was no significant effect when RAO‐unaffected F were compared with their PM. RAO‐unaffected compared with RAO‐affected offspring had a 5.8 (95% CI: 0.0–1.0) times higher risk for shedding 1–100 EpG. Age, sex, breed, and sharing pastures with other species had no significant confounding effects. Conclusion and Clinical Importance: RAO is associated with resistance against strongylid parasites in a high‐prevalence family.  相似文献

The objectives of this study were to characterize the inflammatory response after microchip insertion, evaluate pain response and swelling at the microchip insertion site, and measure migration of the microchips. Eighteen mature Quarter Horse mares were separated by expected parturition dates and then assigned randomly to treatment groups. The microchip group (n = 7) had microchips inserted using a sterile needle and syringe; the sham group (n = 7) had a needle inserted but no microchip; and the control group (n = 4) had no insertion. The inflammatory response was measured over a 2-week period by measuring dermal temperature, response to pressure, and swelling at the insertion site and plasma serum amyloid A (SAA). For the migration component of the study, radiographs of the seven microchipped horses were taken over 6 months after insertion. These radiographs allowed measurement between a select vertebral point and the microchip. The microchip and sham insertion did not cause a detectable increase in temperature. Algometer readings, used to quantify pressure necessary to induce a pain threshold response, indicated that microchip insertion area was more sensitive than sham insertion at 2 hours, day 1, and day 3 post insertion. Visible swelling began 2 hours postinsertion and resolved by day 3. SAA concentrations were affected by day following insertion, but not by treatment group. Increases in SAA concentration could not be matched with local insertion reactions. Migration was not detected in any of the horses during the 6 months within a 0.7-cm margin of error.  相似文献

通过对内蒙古马术队123匹马体内寄生虫检查,发现共有6种寄生虫。分别是马副蛔虫、马尖尾线虫、圆线虫、毛线虫、马网尾线虫、马胃蝇寄生于体内,它们的感染率分别为51.22%、27.64%、86.18%、87.80%、70.70%、65.85%。敌百虫按0.05g/kg的剂量给马匹投药驱虫和伊维菌素按0.2mg/kg的剂量给马匹混饲给药驱虫2次,其用敌百虫驱虫后的虫卵减少率仅达到了46%,而用伊维菌素驱虫后的虫卵减少率可达到99%以上,即伊维菌素驱虫效果明显要优于敌百虫驱虫效果。  相似文献

Upper airway pressure was measured during maximal exercise in 10 Thoroughbred racehorses with naturally occurring upper airway obstruction. Left laryngeal hemiplegia and arytenoid chondropathy resulted in substantial increases (30-40 cm H2O) in inspiratory upper airway pressure (Pl), whereas complicated aryepiglottic entrapment and subepiglottic cysts produced only modest increases (15 cm H2O) in Pl. Uncomplicated aryepiglottic entrapment and grade IV pharyngeal lymphoid hyperplasia produced only slight increases (3-5 cm H2O). In general, surgical procedures restored airway pressures to within normal limits. Subtotal arytenoidectomy improved but did not normalize airway pressures in horses with arytenoid chondropathy. Pharyngeal lymphoid hyperplasia appeared to have little effect on upper airway function.  相似文献
